Ilex asprella (Hook. et Arn.) Champ. ex Benth is one of the most important traditional Chinese medicines in southern China. The seeds of Ilex asprella usually have extremely low germination due to their dormancy characteristics, which severely impacts the efficiency of seedling raising and increases labor costs. In this study, to improve the seed germination of I. asprella, the effects of germination substrate, hormone, winnowing, and stratification treatments on the seed germination of I. asprella were investigated. The results of the germination matrix showed that the highest germination percentage of 45.2% was achieved under the 20°C/10°C day/night temperature and vermiculite germination medium conditions. The results of hormone treatments revealed that 100–400 mg/L of gibberellin (GA) and 50–100 mg/L of salicylic acid (SA) were found to be effective in releasing the dormancy of I. asprella seeds. Moreover, winnowing could effectively eliminate unsaturated seeds and impurities, thus improving the seed germination of I. asprella. Furthermore, warm temperature (15°C) stratification could expand the temperature range of I. asprella’s seed germination, which was beneficial for seed germination of I. asprella and for seed nursery at room temperature in production practice. The present study obtained a method to break dormancy and increase seed germination in I. asprella, thereby forming a groundwork for improving the efficiency of large-scale planting of I. asprella.  相似文献

Seeds with a water‐impermeable seed coat and a physiologically dormant embryo are classified as having combinational dormancy. Seeds of Sicyos angulatus (burcucumber) have been clearly shown to have a water‐impermeable seed coat (physical dormancy [PY]). The primary aim of the present study was to confirm (or not) that physiological dormancy (PD) is also present in seeds of S. angulatus. The highest germination of scarified fresh (38%) and 3‐month dry‐stored (36%) seeds occurred at 35/20°C. The rate (speed) of germination was faster in scarified dry‐stored seeds than in scarified fresh seeds. Removal of the seed coat, but leaving the membrane surrounding the embryo intact, increased germination of both fresh and dry‐stored seeds to > 85% at 35/20°C. Germination (80–100%) of excised embryos (both seed coat and membrane removed) occurred at 15/6, 25/15 and 35/20°C and reached 95–100% after 4 days of incubation at 25/15 and 35/20°C. Dry storage (after‐ripening) caused an increase in the germination percentage of scarified and of decoated seeds at 25/15°C and in both germination percentage and rate of excised embryos at 15/6°C. Eight weeks of cold stratification resulted in a significant increase in the germination of scarified seeds at 25/15 and 35/20°C and of decoated seeds at 15/6 and 25/15°C. Based on the results of our study and on information reported in the literature, we conclude that seeds of S. angulatus not only have PY, but also non‐deep PD, that is, combinational dormancy (PY + PD).  相似文献

It is not known how embryos of seeds of the Pinaceae protrude from their enclosing tissues to complete germination. Prior to protrusion of the radicle there is an increase in endo-β-1,4-mannanase (EC 3.2.1.78) activity associated with weakening of the micropylar megagametophyte/nucellus from seeds of white spruce ( Picea glauca [Moench.] Voss). Mannanase activity is present as three isoforms (pI values 5.0, 4.8, 4.7) in both the embryo and surrounding structures (megagametophyte and nucellus) prior to and during imbibition. Activity of all the isoforms increases in the chalazal and micropylar megagametophyte during germination. Activity then declines after the testa splits, typically 1 day prior to radicle protrusion, due partially to its leaching from the seed into the surrounding water. Activity increases in the cotyledons and axis as the embryo commences elongation. Seeds from dormant seedlots exhibit a lower germination percentage, relative to seeds from nondormant seedlots, and the force necessary for the embryo to puncture the surrounding structures tends to be greater. Although similar mannanase activities are present in unimbibed seeds of dormant and nondormant seedlots, during germination, enzyme activity in seeds of dormant seedlots is lower. Moist chilling alleviates dormancy in the seeds of the Pinaceae and, during 3 weeks of this treatment, mannanase activity slowly increases. After 3 weeks of moist chilling and regardless of whether the seedlot was dormant or not prior to moist chilling, the force necessary to puncture the micropylar megagametophyte and nucellus is lower, and the speed of germination greater. Seeds from previously dormant seedlots also complete germination to a greater percentage, relative to unchilled seeds from dormant seedlots. Upon transfer to 25°C, mannanase activity in moist-chilled seeds decreases during germination of all seedlots regardless of their previous dormancy status.  相似文献

Echium plantagineum (Boraginaceae) is native of the Mediterranean regions, has been introduced and become widespread within the American continent (North to South), South Africa, New Zealand and Australia. This plant has seed dormancy, aggressively spreads to infest vast areas of predominantly agricultural land and is considered a toxic weed to livestock. The objectives of this research were: (i) to study the morpho-anatomy of the diaspores of E. plantagineum; (ii) to identify the pathway of water uptake; and (iii) to characterize the germination and the seedling. The morpho-anatomical studies were carried out analyzing semi-permanent slides of transverse and longitudinal sections of the fruits, seeds and seedlings. Histological, histochemical and conventional staining techniques by using stereoscopic, optical and scanning electron microscopy were applied. In the diaspores, the water uptake pathway was determined by fast green staining, germination tests were performed and the morpho-anatomy of seedlings was analyzed. The diaspores showed acrescent calyx surrounding the fruit, composed by 4 rough tuberculate mericarpids. Each indehiscent mericarpid encloses an exalbuminous seed with a thin coat and a spatulate embryo with folded cotyledons. Water uptake took place through the vascular trace “protuberance” in the cicatrix of the mericarpid, which triggers the germination process. Germination was epigeal, the cotyledons were photosynthetically active at the emergence, and a rudimentary developed gemmula was observed. Germination percentage was 66.5%. The macrosclereids of the pericarp functions as an obstacle to water uptake, which is overcome when the mericarpid is detached from the gynobase, and the protuberance that acts as a water uptake path is exposed, leading to germination. The morpho-anatomical characteristics of diaspore, explain the successful dispersal for this aggressive weed, and provide important information in relation to the necessary care for its control.  相似文献

Yellow-cedar (Chamaecyparis nootkatensis [D. Don] Spach) seeds exhibit prolonged coat-imposed dormancy following their dispersal from the parent plant. Analyses were undertaken using S-(+)-[(3)H] abscisic acid (ABA) to monitor the capacity of embryos to metabolize ABA following their isolation from seeds subjected to various dormancy-breaking and control treatments. Radiolabelled phaseic acid (PA) and dihydrophaseic acid (DPA) were detected in embryos and, to a greater extent in the surrounding media, by 48 h regardless of whether the embryos had been excised from seed previously subjected to only a 3 d soak or to a full dormancy-breaking treatment. Of the two enantiomers of ABA, only the natural S-(+)-ABA effectively inhibited germination of isolated embryos. A metabolism-resistant synthetic ABA analogue S-[8',8',8',9',9',9']-hexadeuteroabscisic acid, S-(+)-d6-ABA, consistently slowed the germination rate of excised embryos to a greater extent than that caused by natural S-(+)-ABA. The deuterium-labelled ring methyl groups of the analogue made it more resistant to oxidation by yellow-cedar embryos and thus rendered the analogue more persistent and possessing greater activity. With increasing time of exposure to moist chilling, yellow-cedar embryos became increasingly insensitive to both ABA and to the analogue. Subjecting seed to chemical treatments (GA(3) in combination with 1-propanol) prior to moist chilling strongly enhanced the germinability of whole seeds. This treatment also had a relatively greater impact on ABA metabolism than did moist chilling alone, as indicated by a greater capacity of S-(+)-d6-ABA to inhibit the germination of embryos as compared to S-(+)-ABA. Moist chilling was most critical for reduced ABA sensitivity of embryos. A change in the embryo's ability to metabolize ABA and reduced embryo sensitivity to ABA are two factors associated with dormancy termination of whole seeds of yellow cedar; a change in only one of these factors is insufficient to elicit high germinability.  相似文献

We previously reported that an apparent water potential disequilibrium is maintained late in muskmelon (Cucumis melo L.) seed development between the embryo and the surrounding fruit tissue (mesocarp). To further investigate the basis of this phenomenon, the permeability characteristics of the tissues surrounding muskmelon embryos (the mucilaginous endocarp, the testa, a 2- to 4-cell-layered perisperm and a single cell layer of endosperm) were examined from 20 to 65 days after anthesis (DAA). Water passes readily through the perisperm envelope (endosperm + perisperm), testa, and endocarp at all stages of development. Electrolyte leakage (conductivity of imbibition solutions) of individual intact seeds, decoated seeds (testa removed), and embryos (testa and perisperm envelope removed) was measured during imbibition of freshly harvested seeds. The testa accounted for up to 80% of the total electrolyte leakage. Leakage from decoated seeds fell by 8- to 10-fold between 25 and 45 DAA. Presence of the perisperm envelope prior to 40 DAA had little effect on leakage, while in more mature seeds, it reduced leakage by 2- to 3-fold. In mature seeds, freezing, soaking in methanol, autoclaving, accelerated aging, and other treatments which killed the embryos had little effect on leakage of intact or decoated seeds, but caused osmotic swelling of the perisperm envelope due to the leakage of solutes from the embryo into the space between the embryo and perisperm. The semipermeability of the perisperm envelope of mature seeds did not depend upon cellular viability or lipid membrane integrity. After maximum seed dry weight is attained (35-40 DAA), the perisperm envelope prevents the diffusion of solutes, but not of water, between the embryo and the surrounding testa, endocarp, and mesocarp tissue.  相似文献

Summary

Seeds of 32 montane species were collected throughout northern Scotland. Germination responses, particularly the effects of chilling on breaking dormancy, were examined using a serial test incorporating single- and double-chilling treatments, alternating 20°C/10°C incubation temperatures, gibberellic acid and, for some species, nicking of the seed coat. Where practicable, any ungerminated seeds were ultimately dissected to assess their viability. Only three species had an absolute requirement for pre-chilling before seeds germinated. A further eight species required chilling to break the dormancy of 15% or more seeds; otherwise chilling generally increased the extent and rate of germination. Unusually, chilling induced dormancy in the seeds of four species whereas warm conditions induced dormancy which was not broken by subsequent chilling amongst seeds of Draba incana. Germinability and germination rates for nine species were regressed on the altitude, latitude and oceanicity of the plants' origin to assess the relative effects of chilling. These three environmental factors accounted for up to 98% of the variability in the germination parameters but few regressions attained statistical significance. Broad patterns suggest that chilling had a decreasing effect on both the extent and, rate of germination as the altitude of the seed source increased. A similar pattern, but only for germinability, was seen with respect to latitude.  相似文献

Summary Dormancy of intact sycamore (Acer pseudoplatanus) seeds was broken by chilling (5°C) for several weeks in moist conditions. Treatment of unchilled seeds with kinetin induced some germination, but gibberellin was ineffective. This stimulation by kinetin was not suppressed by the added presence of abscisic acid during incubation. The chilling requirement of intact seeds was eliminated by removal of the testa, and the naked embryos developed with no morphological abnormalities. During early growth of isolated embryos in the light, two distinct developmental processes were recognised. One involved initial elongation of the radicle accompanied by geotropic curvature and was stimulated by kinetin but not by gibberellin, while the other involved unrolling of the cotyledons, which was accelerated by gibberellin but much less by kinetin. Abscisic acid strongly suppressed both developmental processes when applied alone, inhibited cotyledon expansion in the presence of gibberellin, but failed to overcome the promotory effects of kinetin on radicle growth. Experiments with CCC indicated that under natural conditions the unrolling of the cotyledons is dependent upon endogenous gibberellin. Radicle growth of isolated embryos was unimpaired by incubation in the dark, but cotyledon expansion of water incubated embryos was poor, and although it was accelerated by gibberellin, the responses in all treatments were slower than in the corresponding light grown samples. It is suggested that endogenous cytokinins are primary factors in the initiation of radicle growth, while gibberellins are important in cotyledon expansion. Abscisic acid appears to have an inhibitory role in both processes, and the interactions of these regulators in the control of germination and development are discussed.

The dormancy breaking and storage behavior of Garcinia cowa Roxb. seeds were investigated. The seeds of G. cowa had 8-11 months dormancy in their natural habitat. Seeds were matured and dispersed at the end of the rainy season (mid-late August to late September) and were scatter-hoarded by rodents as food for winter after the seeds had fallen to the ground. Seedlings often emerged in the forest during the rainy season (May to August) the following year. Intact seeds of G. cowa failed to germinate after being sown at 30 ℃ for 120 d and the mean germination time (MGT) of seeds cultured in a shade (50% sunlight)nursery was 252 d. The most effective method of breaking dormancy was to remove the seed coat totally, which reduced the MGT to 13 d at 30 ℃. Germination was also promoted by partial removal of the seed coat (excising the hilum and exposing the radicle) and chemical scarification (immersion in 1% H2O2 for 1 d).Unscarified seeds take up water rapidly in the first 96 h, but water was absorbed by the outside seed coat, without penetrating through it. The moisture content (MC) of G. cowa seeds was high (50% in fresh weight)at shedding. The seeds could tolerate desiccation to some extent, until the MC reached approximately 40%;below that, the viability decreases rapidly and all seeds died at approximately 17% of MC. Seed viability decreased rapidly when seeds were chilled at 4 ℃; germination was 2% after storage for 1 week. Even stored at 10 ℃, seeds began to be damaged after 4 weeks. Seed storage for 1 yr revealed that in both dry (relative humidity (35 ± 5)%) and moist (wet sand) storage conditions, seed viability declined, but germination percentages for seeds stored under moist conditions are better than for seed stored under dry conditions.Because of their low tolerance to desiccation, marked chilling sensitivity and relatively short lifespan, G.cowa seeds should be classified into the tropical recalcitrant category. The ecological implications of dormant recalcitrant seeds and cues on storing recalcitrant seeds were discussed.  相似文献

Dormant seeds of 18 species from 9 families covering a diverse range of seed dormancy syndromes and life histories from the southwest Australian biodiversity hotspot were assessed for germinability following storage at 15–25°C for 36 months. A total of 10 species with physical dormancy (PY) and 8 with either physiological dormancy (PD) or morphophysiological dormancy (MPD) were assessed as part of the study. Prior to storage, germination from dormant seeds was 1–27%, rising to 41–100% following specific dormancy‐breaking treatments. When seed dormancy was removed prior to storage for 36 months seeds from all species were found to maintain a nondormant state and germinate to a similar level to that observed at the beginning of the experiment (44–100%). Likewise, seeds that did not receive a prestorage dormancy‐breaking treatment maintained a dormant state (0–50% germination) and subsequently responded well to a dormancy‐breaking treatment immediately prior to germination assessment (49–99%). There were minimal differences in response to dormancy‐breaking treatments before and after 36 months storage (average 4–6% difference) and in the germination responses observed between both storage environments assessed (15°C/15% eRH or 15–25°C air dried). Based on these findings, storing seeds in a nondormant state does not alter germinability and this approach provides significant benefits to current seed‐based restoration programs through reduction of double handling and improved seed use efficiency.  相似文献

The initiation of radicle growth during seed germination may be driven by solute accumulation and increased turgor pressure, by cell wall relaxation, or by weakening of tissues surrounding the embryo. To investigate these possibilities, imbibition kinetics, water contents, and water (Ψ) and solute (ψs) potentials of intact muskmelon (Cucumis melo L.) seeds, decoated seeds (testa removed, but a thin perisperm/endosperm envelope remains around the embryo), and isolated cotyledons and embryonic axes were measured. Cotyledons and embryonic axes excised and imbibed as isolated tissues attained water contents 25 and 50% greater, respectively, than the same tissues hydrated within intact seeds. The effect of the testa and perisperm on embryo water content was due to mechanical restriction of embryo swelling and not to impermeability to water. The Ψ and ψs of embryo tissues were measured by psychrometry after excision from imbibed intact seeds. For intact or decoated seeds and excised cotyledons, Ψ values were >−0.2 MPa just prior to radicle emergence. The Ψ of excised embryonic axes, however, averaged only −0.6 MPa over the same period. The embryonic axis apparently is mechanically constrained within the testa/perisperm, increasing its total pressure potential until axis Ψ is in equilibrium with cotyledon Ψ, but reducing its water content and resulting in a low Ψ when the constraint is removed. There was no evidence of decreasing ψs or increasing turgor pressure (Ψ-ψs) prior to radicle growth for either intact seeds or excised tissues. Given the low relative water content of the axes within intact seeds, cell wall relaxation would be ineffective in creating a Ψ gradient for water uptake. Rather, axis growth may be initiated by weakening of the perisperm, thus releasing the external pressure and creating a Ψ gradient for water uptake into the axis. The perisperm envelope contains a cap of small, thin-walled endosperm cells adjacent to the radicle tip. We hypothesize that weakening or separation of cells in this region could initiate radicle expansion.  相似文献

Seed Coat Dormancy in Two Species of Grevillea(Proteaceae)   总被引:3,自引:0,他引:3
The role played by the seed coat in seed dormancy of Grevillealinearifolia(Cav.) Druce and G. wilsonii(A. Cunn.) was testedby a series of manipulations in which the seed coat was dissectedand removed, dissected and returned to the decoated seed, ordissected, removed and given a heat shock, and returned to thedecoated seed. Germination of intact seeds of both species wasalso examined after exposure to heat shock, smoke, or heat shockand smoke combined. Water permeability of the seed coat wasinvestigated by examining imbibition. For intact seeds, virtuallyno germination occurred under any treatment (G. wilsonii), orgermination was increased by exposure to either heat or smoke(G. linearifolia). Removal of the seed coat led to germinationof all decoated seeds for G. linearifolia, or a proportion ofdecoated seeds for G. wilsonii. Inclusion of smoked water inthe incubation medium led to a higher proportion of decoatedseeds germinating for G. wilsonii. Returning the seed coat, either with or without heat shock to the seed coat, did notsignificantly affect germination in either species. Seed coatswere permeable to water in both species. For the two Grevilleaspecies, there were different dormancy mechanisms that werecontrolled by the seed coat (G. linearifolia) or by both theseed coat and embryo (G. wilsonii). An unusual case of epigeal cryptocotylar germination found in Rollinia salicifolia Schltdl. (Annonaceae) is described and is discussed in relation to the terminology used for germination studies. The terms cryptocotylar and phanerocotylar are used to describe the permanence of cotyledons within the seed coat or endocarp; the terms epigeal and hypogeal are used to refer the position of cotyledons in relation to ground level. The terms to describe position about ground level and permanence of cotyledons within the seed coat or endocarp are not used simultaneously in many papers, and some authors consider the terms cryptocotylar and hypogeal to be synonyms. Cases of epigeal cryptocotylar germination seem to be scarce in the literature. The morphological characters shown by R. salicifolia seeds and seedlings − a non-chlorophyllous embryo, abundant endosperm, thick woody seed coat, thickened hypocotyl and non-photosynthetic haustorial cotyledons − are closely related to its epigeal cryptocotylar germination. This is the first record of this type of germination in Argentina.  © 2004 The Linnean Society of London, Botanical Journal of the Linnean Society , 2004, 146 , 53–56.  相似文献

In the present study, the germination characteristics of three endemic species from Turkey, Tripleurospermum pichleri (Boiss.) Bornm., Cirsium leucopsis D.C and Senecio olympicus Boiss. (Asteraceae), were investigated. Germination was studied for fresh seeds, for seeds subjected to short‐time chilling (15 days, moist +4°C), to GA3 (100, 150 and 250 ppm) and a combination of chilling and GA3; in all cases seeds were incubated either at 20/10°C day/night with light daytime or at 20°C in darkness with daily short‐time dim light (DSDL). In C. leucopsis seeds, all GA3 treatments enhanced the final germination percentages. The mean germination time (MGT) of C. leucopsis was lower under DSDL than with photoperiod. The chilling treatment with GA3 in DSDL significantly increased germination in S. olympicus seeds (from 45 to 87%). Germination increased to 55% in T. pichleri by chilling under photoperiod compared with 32% by chilling followed by DSDL. In conclusion, these three co‐existing endemic Asteraceae species have different germination behaviours; something that should be taken into account for ex situ propagation. However, an efficient way to germinate all species is to use 250 ppm GA3 and 20/10°C with photoperiod.  相似文献

Aristolochiaceae have been described as having seeds with underdeveloped embryos and morphological or morphophysiological dormancy. Aristolochia galeata is a native climber found in the Cerrado biome, associated with road and gallery forest edges. The aims of this study were to investigate: embryo growth rate, morphology and seed germination parameters under different treatments. Embryos were excised to obtain embryo length at four stages: initial, seeds after coat rupture, radicle tip protrusion and cotyledon emergence from the seed coat. Germination tests were conducted at 30 °C under three nitrate concentrations (1, 10 and 20 mM), fluctuating temperature (27/20 °C) and light and dark conditions. We found that seeds have underdeveloped embryos, which take about 301 ± 178 h (±SD) to achieve seed coat rupture, another 205 ± 126 h to reach radicle protrusion and 176 ± 76 h more to the final stage of cotyledon emergence. Germinability was above 52% in all treatments, except in the dark (15%). For all treatments, average germination time was above 290 ± 123 h. Potassium nitrate increased germinability to >87%. No particular treatment was required for embryo development, but seeds in the population that continued to germinate after 1 month were probably in various states of non-deep, simple morphophysiological dormancy. Increased germinability in nitrate treatments and light requirement for germination could prevent germination under unsuitable environmental conditions and be a strategy to increase seedling establishment in the cerrado.  相似文献

Arisaema dracontium (green dragon) is a perennial herb that is widely distributed in eastern North America. However, in Canada, at the northern edge of its distribution, the species is designated as “vulnerable” with respect to conservation status. In natural populations, seedlings are uncommon; the present study was undertaken in order to characterize seed and seedling properties in green dragon. Seeds were sampled from five sites, ranging from Ontario at the northern limit of the distribution range, to Louisiana in the south. Seed germinability ranged from 25 to 55%, depending upon source. Experiments indicated that neither the hard seed coat nor a water-soluble exudate from the seed was responsible for inducing or maintaining dormancy. Patterns of seed germination appear to reflect general climatic conditions at the sites where seeds had originated. Cold stratification at 3°C produced significantly greater relative germinability in all seed collections except the most southerly one, from Baton Rouge. These seeds also had a slower overall speed of germination. In contrast, germination of seeds from the most northerly site was promoted by cold stratification and occurred over a relatively brief period. Germination in alternating light and dark conditions decreased the speed of germination compared to germination in the dark, however exposure to light changed the phenology of germination by promoting development of adventitious roots and primary leaves in these seedlings.  相似文献

Summary One of the foremost technical issues addressed in reintroduction and restoration projects is the feasibility of establishing living plants. To advance the recovery process, the germination requirements of 201 threatened Western Australian seed‐bearing taxa representing a range of life forms, families and ecophysiological characteristics were studied. Procedures used to stimulate germination in otherwise dormant seed involved pretreatment using thermal shock, scarification, seed coat removal, soaking in an aqueous smoke solution and/or additions of the growth hormone gibberellic acid (GA3). Sixty‐one taxa germinated under the basic trial conditions of light (12‐ h photoperiod), temperature (constant 15°C) and moisture, without additional pretreatments. These taxa were generally those with canopy‐stored seeds in the families Proteaceae and Casuarinaceae, and small‐seeded taxa in Myrtaceae. Taxa with soil‐stored seeds required single or multiple cues to stimulate germination. Seeds in the families Fabaceae and Mimosaceae were dependent on cracking of the seed coat, mechanically through nicking of the testa or through thermal shock, as were several non‐leguminous species of the Sterculiaceae and Liliaceae. Complete or partial removal of seed coats, in conjunction with GA3 enhanced germination percentage in some taxa of the Myoporaceae, Lamiaceae and Myrtaceae. Application of GA3 also enhanced germination percentage in members of the Epacridaceae. Several taxa previously stimulated by aqueous smoke solutions were equally responsive to additions of GA3 after complete seed coat removal. In general, species with seed weights greater than 10 mg germinated better under a range of conditions than those with lighter seeds. There was no difference in germinability between resprouter and seeder species, and no obvious relationship between seed weight and germination rate. In the light of previous studies these results indicate that the relationship between germination requirements and ecophysiological characteristics is similar for both threatened and common species. These data will enable better prediction of likely dormancy breaking cues for other related species and will greatly assist the process of recovery and restoration work for mining operations and community bushland regeneration as well as single species reintroductions.  相似文献
