Phlomisxmargaritae is a rare and sterile hybrid taxon foundin a single endangered population in southern Spain. It waspreviously described as a morphologically intermediate hybrid,putatively between Phlomis purpurea and P. composita. The presentstudy used allozymes as molecular markers to assess the hybrididentity ofP.xmargaritae . Ten putative loci from seven enzymesystems were resolved: five were monomorphic and fixed acrossall taxa studied and the rest (Aat-1, Aat-2, 6-Pgdh-2, Pgi andPgm) were polymorphic in at least one taxon. The two parentaltaxa are fixed for different alleles at 6-Pgdh-2 and show distinctallelic frequency differences for four other loci. Phlomisxmargaritaedisplays fixed heterozygous phenotypes for four of the fivepolymorphic loci, these being composed of combinations of thealleles found in the parental taxa. No unique alleles were detectedin P.xmargaritae. We conclude that this taxon is of hybrid originand confirm the identity of the parental taxa involved. It isfurther suggested that this population is composed of individualsthat are recent F1hybrids that have not undergone backcrossingor introgression. 为了调查D10S1248、D2S441、D1S1677、D9S1122、D10S1435等5个miniSTR(mini short tandem repeats)基因座在西北汉族人群中的遗传多态性、遗传稳定性及其在陈旧降解检材中的法医学应用价值, 文章采用荧光PCR和基因分型技术对西北汉族154份无关个体血样、10个家系血液样本及10份陈旧降解检材进行片段长度分析。在西北汉族人群中, 5个miniSTR基因座分别检测出了8、7、7、6、7个等位基因, 等位基因频率分布符合Hardy-Weinberg平衡定律, 杂合度(Heterozygosity, H)为0.662~0.792, 个人识别率(Power of discrimination, PD)为0.869~0.915, 非父排除率(Power of exclusion, PE)为0.382~0.585, 多态信息含量(Polymorphism informa-tion content, PIC)为0.650~0.750。家系和陈旧降解检材的研究表明, 5个miniSTR基因座具有高度的遗传稳定性, 可对陈旧降解检材DNA进行有效的分型。5个miniSTR基因座适合作为西北汉族人群的遗传标记, 用于陈旧降解检材的法医学个人识别和亲权鉴定案件中。  相似文献

Aquatic snails from south western Zimbabwe belonging to theBulinus trunscatus/tropicus complex vary widely in shell formsuggesting that more than one taxon could be present. This possibilitywas investigated by making observations on snail samples from13 populations from the Plumtree area, in respect of allozymevariation (5 polymorphic loci), shell morphology (9 variables),copulatory organ and chromosome number. Comparative data wereobtained from snails from north western Zimbabwe identifieddefinitely as B. tropicus. Analysis of the genetic structurerevealed a high degree of polymorphism (P) ranging from 0.29–0.80among populations from Plumtree and expected heterozygosity(He) from 0.02–0.22. No enzymatic diagnostic loci werefound which could differentiate the different morphs or populationsand discriminant function analysis on the morphological datashowed an overlap of morphs among populations. Snails analyzedfor chromosome number were all diploid (2n = 36). Snails exposedto Schistosoma haematobium mira-cidia were all refractory. Thisinformation supports the view of a single species, B. tropicus,which is differentiated due to migration barriers and whereenvironmental variables might be implicated in the morphometricdivergence. Near-isogenic lines have been produced in pea to examine theeffects of genes at the r and rb loci on the growth and developmentof the seed. The rb alleles were found to have an effect onthe growth of the testa as well as on that of the embryo incontrast to those at the r locus which only affect the latter.The relationship between the fresh and dry weights of the embryosof rb isoline and the double mutant differed from that of theround-seeded wild-type line. To estimate the geographical extent of introgression, we studiedthe genetic structure of sympatric and allopatric populationsof hybridizing freshwater snail species Viviparus ater and V.contectus in central Europe. Six allozyme loci which were variablein Lake Garda, Italy in a previous study (five nearly diagnosticloci between the two species and one highly polymorphic locusin V. contectus) were analyzed from ten sympatric locationsand four allopatric populations each for the two species. Presumablyintrogressed genes (low allele frequencies) were found fromat least one locus in seven out of the ten sympatric sites.These seven sites covered most of northern Italy. The data indicatethat introgression has occurred from Viviparus contectus toV. ater and vice versa. Therefore, there is a possibility ofwidespread introgression or mosaic zones in nature. However,we cannot rule out that the observed patterns are due to theshared ancestry. V. ater possessed low genetic variation (thejackknifed mean of Wright's FST±S.E. over four loci was0.041±0.004). On the other hand, V. contectus showedhigh genetic differentiation (the jackknifed mean of FST±S.E. over six loci was 0.546±0.166). Although introgressionmay have caused evolutionary changes in V. ater and V. contectus,it was not strong enough to level out the genetic differencesbetween the two species, which may have originated from isolationamong populations in V. contectus and a past bottleneck eventin V. ater. Genetic variation in allozymes of six species of Saco-glossa(Mollusca, Opisthobranchia) from Denmark was studied. Nine enzymesrepresenting 11 loci gave reactions that could be used to calculateallele frequencies. Elysia viridis, Limapontia capitata, L.de-pressa, Ercolania nigra, Alderia modesta and Calliopaea oophagaall showed very great variation both within and between species.Levels of polymorphism varied between 0.5 and 0.8, and levelsof observed heterozygosity were between 0.18 and 0.42. Interspecificidentity of alleles in polymorphic loci was only found betweenthe two species of Limapontia. Even in these two species geneticidentity (I) was very low (0.07) for congeneric species. Samples of the Antarctic limpet, Nacella concinna (Strebel,1908), were collected from four sites in the South Orkney Islandsand from Stromness in South Georgia during 1988. At three SouthOrkney sites, both littoral and sub-littoral samples of limpetswere taken. Shell dimensions were measured and littoral andsub-littoral limpets were shown to have significantly differentshell shapes. Foot and digestive gland tissues were subjectedto electrophoresis and five polymorphic loci {Es-1, lcd-1, lcd-2,Gpi, Got-1 and Pgm-1) and two monomorphic loci (Es-2 and Got-2)were scored. Genetic identities between littoral and sub-littoralforms and also between sites were calculated. At the loci scored,the littoral and sub-littoral forms were virtually identicaland this suggests that the shell shape differences between theforms are the result of environmentally induced phenotypic plasticity.However, significant genetic differences and some morphologicaldifferences were apparent between the sample of limpets fromSouth Georgia and all other sites. Genetic identity values suggestedthat the South Georgia and South Orkney N. concinna are geographicallyseparated populations of a single species rather than distinctsub-species. Three species of the genus Lymnaea (Gastropoda, Pulmonata, Basommatophora)were examined for genetic variation at 11 enzymatic loci insamples collected in Brittany (North-Western France). Variabilitywas greatest in L. percgra, but the limited area of samplingdid not allow specific inferences. The UPGMA clustering of unbiasedgenetic distances (Nei, 1978) isolated first L. auriculariasamples, and secondly the single L. stagnalis sample from theL. peregra set. This scheme of relationships was opposed tothe identification of alleles shared by the three species atthe studied loci Analysis of L. peregra population structure by F-statistics(Weir & Cockerham, 1984) suggested high inbreeding and raisedthe possible role of self-fertilization. The contradiction betweenhigh variability and high homozygosity is discussed. Furthermore,population differentiation was rather weak (although statisticallysignificant), despite some remarkable differences among loci.Nm estimates using Wright's (1951) and Slatkin's (1985) methodsrevealed a gene flow unable to prevent the effect of geneticdrift among populations. The unique flavour of a tomato fruit is the sum of a complexinteraction among sugars, acids, and a large set of volatilecompounds. While it is generally acknowledged that the flavourof commercially produced tomatoes is inferior, the biochemicaland genetic complexity of the trait has made breeding for improvedflavour extremely difficult. The volatiles, in particular, presenta major challenge for flavour improvement, being generated froma diverse set of lipid, amino acid, and carotenoid precursors.Very few genes controlling their biosynthesis have been identified.New quantitative trait loci (QTLs) that affect the volatileemissions of red-ripe fruits are described here. A populationof introgression lines derived from a cross between the cultivatedtomato Solanum lycopersicum and its wild relative, S. habrochaites,was characterized over multiple seasons and locations. A totalof 30 QTLs affecting the emission of one or more volatiles weremapped. The data from this mapping project, combined with previouslycollected data on an IL population derived from a cross betweenS. lycopersicum and S. pennellii populations, were used to constructa correlational database. A metabolite tree derived from thesedata provides new insights into the pathways for the synthesisof several of these volatiles. One QTL is a novel locus affectingfruit carotenoid content on chromosome 2. Volatile emissionsfrom this and other lines indicate that the linear and cyclicapocarotenoid volatiles are probably derived from separate carotenoidpools. 运用16种酶蛋白编码的23个遗传座位对突尼斯非洲跳鼠(Jaculus jaculus)和埃及跳鼠(J. orientalis)自然群体的遗传变异和分化进行了电泳分析。结果表明,与其他啮齿动物等哺乳动物的相关数据比较,发现这两个种群体的遗传变异水平较低。非洲跳鼠群体的观测杂合度 (Hobs) 为0.08—0.19,多态座位百分比(P)为26.2%—45.2%,每个座位的平均等位基因数(A)为1.1—1.4;埃及跳鼠的Hobs为0.10—0.15,P为29.3%—44.1%,A为1.1—1.7。两个种群体各自的遗传分化程度较低(非洲跳鼠和埃及跳鼠的Fst分别为0.0017和0.0019)。而两个种群体间的Fst为0.607(P<0.05),表明两个种之间高度的遗传分化。本研究支持这两个种分类地位的合法性,并强调了地理因素(环境类型和生物气候阶段)对两个种遗传结构的影响。  相似文献

Past findings have established how the faster growth, greaterreproductive output and/or longer survival that are associatedwith heterosis and genomic diversity measured as multi-locusheterozygosity stem from slower intensities with which proteinsare renewed and replaced (=protein turnover). Slower turnoverresults in lower energy requirements and reduced metabolic sensitivityto environmental change, representing a mechanistic basis forevolutionary consequences of genetic polymorphism. To determinethe genetic and functional basis of differences in whole-bodyprotein turnover, we have begun to resolve different proteolyticpathways, searching for genetic polymorphisms with a directeffect upon proteolysis, and assessing the metabolic and physiologicalconsequences of those genetic influences in the mussel Mytilusedulis. Our recent work has established the physiological importanceof lysosomal enzymes under normal conditions of basal proteolysis,and shown that associated effects on energy flux may vary accordingto functional differences between separate enzymes. Data arepresented here which compare metabolic consequences of polymorphismin the lysosomal aminopeptidases Lap-1 and Lap-2. Findings establishthat metabolic and phenotypic effects of genetic polymorphismresult directly from genetic variation at the loci coding forthese peptidases, rather than from linked loci. They also illustratethe complexity of interrelations that ultimately influence theevolutionary consequences of genomic diversity, including associatedinfluences of both Lap-1 and Lap-2 on energy requirements andanimal condition. We impress that energy requirements for proteinturnover may represent a functional basis for epistasis, includingassociations whereby advantages of genetic polymorphism aregreatest at loci that code for enzymes acting in both proteincatabolism and energy provision.  相似文献

Oncomelania lindoensis from Lake Lindu, Sulawesi, was characterizedfor genetic variation at 21 allozyme loci and compared withO. hupensis (China) and O. quadrasi (Philippines). Geneticdistances and interpopulation patterns of allele-sharing pointto a closer relationship between Sulawesi and the Philippines(Nei's unbiased genetic distances (D) averaged 0.50) than betweenSulawesi and China (D= 0.79). These data, coupled with a considerationof the geographic distribution of the genus, support the hypothesisthat the Sulawesi Oncomelaniaoriginated by avian-facilitated colonizationfrom the Philippines about two million years ago. Oncomelania from Sulawesi were originally described as subspecificallydistinct: Oncomelania hupensis lindoensis. However, the allopatricdistribution, unique alleles at five loci, and significant geneticdistances from congeners in Mindanao and elsewhere in the Philippinessuggest that this taxon should be distinguished as a full specieswithin the Oncomelania hupensis species group, namely: O. lindoensisDavis & Carney 1973. Comparison with published data on variationwithin quadrasi and in three Chinese subspecies of hupensisshowed that D values increase with taxonomic level in this speciesgroup. D averaged 0.15 (0–0.26) within Chinese subspeciesand 0.04 (0–0.13) within the Philippines, but was 0.30(0.20–0.45) between Chinese subspecies, and 0.48–0.80between the three species (hupensis, quadrasi and lindoensis).The genotypic cluster species concept and these multilocus geneticdistances can be used to help define species and subspeciesin these medically important snails. Mutant genes at two loci, r and rb, known to encode genes affectingthe starch biosynthetic pathway, were studied for their effecton the structure and gelatinization of pea seed starches. Comparisonswere made using starches from four lines {RRRbRb, rrRbRb, RRrbrb,and rrrbrb), near-isogenic except for genes at these two loci.All the starches had C-type X-ray diffraction patterns, butdifferent contents of ‘A’ and ‘B’ polymorphs.The presence of a mutation at either locus increased the ‘B’polymorph content in the starches, although the influence ofthe r mutation was much greater than that of rb. Differenceswere discovered in the crystalline stucture of the rrRbRb starchwhich correlated with a high content of amorphous phase as wellas with the changes in amylopectin structure. In addition, changesin the crystalline structure of this sample correlated witha lack of co-operative transition during starch gelatinizationin excess water. The RRrbrb starch had a greatly increased enthalpyof gelatinization in excess water compared with the wild-typestarch. It is proposed that this effect is connected with specificcharge interactions between the molecules in the starch granule.The rrrbrb starch had parameters of crystalline structure andgelatinization which reflected the different influences of thetwo genes. With regard to gelatinization, this starch had relativelywide co-operative transition and low enthalpy and a very highpeak temperature of transition. Four Littorina species from the Pacific (L. sitkana, L. brevicula,L. mandshurica, L. squalida) and four from the Atlantic (L.mariae, L. obtusata, L. saxatilis and L. littorea) were comparedelectrophoretically using 17 loci. Analysis of genetic identities showed that there were threegroups. The first group comprises L. sitkana, L. mariae, L.obtusata and L. saxatilis The second group consists of L. breviculaand L. mandshurica and the third one includes L. squalida andL. littorea. In addition to the known loci, If, e, sn and hr, a fifth locus,veg, is shown to control flowering in peas. Regardless of thegenotype for the other flowering genes, plants homozygous forthe gene veg did not initiate flower buds under a wide rangeof photoperiod and temperature regimes, including those normallyhighly promotory in peas. Treatment with various plant growthsubstances and grafting to stocks known to promote floweringalso failed to cause initiation. Gene veg prevented expressionof allelic differences at the If locus but segregation for allelesat the sn and hr loci was clearly visible by examination ofseveral vegetative characteristics. For example, sn hr veg andSn hr veg plants showed an opening of the apical bud, productionof lateral branches, and a reduction in growth rate, leafletsize, internode length and stem thickness at approx the sametime as sn hr Veg and Sn hr Veg plants carrying the Lf allelecommenced fruit production, respectively. The graft-transmissibleinhibitor controlled by gene Sn is therefore not specific forthe transition from vegetative to reproductive growth. Geneveg allows the processes leading to apical and foliar senescenceto be examined independently of any effect of flowering andfruiting. We found that gene Sn influenced the total numberof leaves expanded in veg plants but not the time of shoot senescence,which, in plants without flowers and fruits appeared to resultfrom failure of the root system. Local variation in ecological and genetic traits was analysedin the perennial herb Arabis stelleri var. japonica in a sanddune area in western Japan. Plants of this species are establishedin three contiguous habitats (subpopulations): sand dune, forestand an ecotonal zone between both habitats. Morphological, ecologicaland genetic traits of A. stelleri var. japonica plants wereanalysed to discover the extent of microdifferentiation amongthese subpopulations. All plants in nine quadrats were markedand followed during 2 years. The ecotonal zone presented thehighest number of mature individuals per quadrat. Plants mayproduce the same number of shoots per plant but rosette sizewas larger in plants from sand dune and forest subpopulationsthan the ecotone. Plants in the sand dune showed the highestnumber of large capsules per plant. Mortality of seedlings andmature plants was slightly higher in the ecotone than in theother two subpopulations. Sand dune plants had the highest variationamong quadrats in almost all traits measured. In the electrophoreticanalyses, 21 loci were detected on twelve enzyme systems. Fifteenloci were monomorphic and six loci had 1·2 alleles perloci. Genetic distances and genetic identity Nei's values werevery similar among subpopulations (means I = 0·994, D= 0·006). The total genetic diversity (HT) was 0·340and the within-population component (Hs) was 0·333 indicatinglow genetic differentiation among subpopulations. Gene flowvaried between genes but most of the values were high (meanNm = 188·3). Subpopulations of A. stelleri showed differentiationin morphological and ecological traits and low genetic differentiationin the loci studied. Ataxia telangiectasia (AT) is an autosomal recessive diseaseof unknown etiology associated with cerebellar ataxia, telangiectasia,immune dysfunction, higher cancer risk, genomic instabilityand hypersensitivity to ionizing radiation. The major AT loci,AT-A and AT-C, are shown to be closely linked at chromosome11q22–q23. The most recent genetic linkage mapping andlinkage disequilibrium analysis have localized the major ATloci to a sequence of approximately 850 kb between the markersD11S1819 and D11S1818. The isolation of yeast artificial chromosomesspanning the AT region is an essential step to identify thegene or genes responsible for the mutation(s). We isolated atotal of 20 YAC clones from three independent YAC libraries,using sequence tagged sites mapped in the AT region as primersfor PCR-based YAC screening. The PCR assay for the presenceor absence of 16 different DNA markers allowed us to constructand to order four YAC contigs at the AT region. One of the contigswhich consists of the 10 YAC clones, covers about 2 Mb of DNAat the boundary between Giemsa-positive band 11q22.3 and Giemsa-negativeband 11q23.1 and includes the entire region of the major ATlocus between D11S1819 and D11S1818. Thus, the YAC contigs willfacilitate the positional cloning approach for searching transcribedsequences from the defined genomic region.  相似文献

The effects of mutant alleles at the r and rb loci on starchgrain size and the levels of starch and amylose in developingpea (Pisum sativum L.) seeds have been examined. Four lines,near-isogenic except for genes at these loci, have been usedto show that both mutations reduce levels of starch throughoutembryo development and reduce levels still further when combinedin the ‘double mutant’. The reduction in starchcontent was due, at least in part, to a reduction in starchgranule size. Although the proportion of starch in mature embryoswas similar in the rrRbRb and RRrbrb lines, the starch contentdiffered between these two lines during development, as a percentageof embryo dry weight. This difference was due to a reductionin the absolute growth of the embryo caused by the rb mutation.Lines homozygous for the mutant r allele with either wild-type(RbRb) or mutant (rbrb) alleles at the rb locus contained increasedproportions of amylose in their starch throughout development,due to a reduced production of amylopectin. The presence ofthe rb mutation, however, also reduced the amount of amylosein relation to the reduction in total starch levels. Mutantalleles at both loci also reduced starch levels in the testaduring development, the reduction due to rb being more extreme.Reciprocal crosses showed a maternal effect of the rb mutationon final seed size and on the absolute amount of starch in theembryo. Variation at the leucine aminopeptidase (Lap), glucose phosphateisomerase (Gpi) and tetrazolium oxidase (To) loci was investigatedin samples of three populations, Al-Mayana (MAY), Shigita (SH)and Mina Salman (MS), of Pinctada radiata from pearl oysterbeds around Bahrain. The To locus was monomor-phic. SignificantLap and Gpi heterozygote deficiencies were evident and it issuggested that these were generated by selection. The MS population,to the East of Bahrain, differed significantly in Gpi allelefrequencies from both Northern populations (MAY, SH) and Nei'sgenetic identity indicates a close relationship between theNorthern populations. Measurements of shell morphometrics were used both as ratiosof one dimension to another, and as regressions of one dimensionon another to examine relatedness between populations. Boththese mor-phometric approaches gave different results from eachother and also differed from the electrophoretic data. It isconcluded that estimates of relatedness in pearl oysters basedon electrophoretic data will be more reliable than those basedon shell shape. SYNOPSIS. Colonies of the compound tunicate Botryllus show thecapacity for self—nonself discrimination by fusion betweenseparated pieces of the same colony and rejection between piecesof unrelated colonies. We have found that genes controllingthis colony specificity are similar to those which cause transplantrejection in the vertebrates. Like the loci within the vertebratemajor histocompatibility complex (MHC), Botryllus fusibility(or histocompatibility) genes are highly polymorphic. In Botryllus,the histocompatibility complex also controls self—sterility,and limits cross—fertilization between colonies sharinghistocompatibility alleles. The mouse MHC, the H-2 region, islinked to loci which also affect the frequencies of allelesat H-2 loci in mouse populations. Thus both systems containcharacters which could act to promote the heterozygous conditionat the linked histocompatibility loci. We suggest that suchlinked characters are responsible for the evolution of allogeneicpolymorphism in vertebrates (however currently maintained),and that tunicate fusibility loci may be the evolutionary precursorsof vertebrate MHC genes.  相似文献
