A submicroscopic structure was studied of protoplasts of five different yeast species multiplying by budding, formation of cross septum and by a division typical for apiculate yeasts. The protoplasts retain their species specificity. Most considerable changes typical for the conversion of a cell to protoplast are found in membrane cell systems. The reduction of membranes of the endoplasmic reticulum is particularly striking. Both membrane units are frequently separated from each other by lenticular pseudovacuoles. Mitochondria in protoplasts are swollen and their number is reduced approximately two-fold. Defects are often observed in a nuclear membrane. The perinuclear space is usually extended by lenticular pseudovacuoles. A large number of vacuoles is observed in the basic protoplast cytoplasm. The surface of the protoplasts of all species studied is formed only by a cytoplasmic membrane. A partially digested original cell wall often adheres to protoplasts ofSchizosaccharomyces pombe.  相似文献

A large field wind tunnel was used to compare four types of CO2-baited mosquito traps. This study compared the plume structure and concentration of CO2 emitted by each trap, flow of suction into the trapping systems, flow of CO2 being released, trap shape and configuration, differences in visual appearance, and differences in temperature and humidity of emissions at the source of CO2 compared to ambient air. The structure of the CO2 plumes emitted by each trap differed considerably. All four plumes were turbulent, causing the concentration of CO2 within several metres of the source to attenuate to between 375 and 875 p.p.m. The Encephalitis Virus Surveillance (EVS) trap emitted concentrations of CO2 exceeding 20,000 p.p.m., the detection limits of our equipment, whereas the Mosquito Magnet Freedom (MMF), Mosquito Magnet Liberty (MML) and Mosquito Magnet X (MMX) traps released CO2 at peaks of about 3500, 7200 and 8700 p.p.m., respectively. The MMX trap produced the greatest air velocity at both the suction inlet and CO2 outlet, followed by the MMF, MML and the EVS traps, respectively.  相似文献

Irradiation of human lymphocytes by x-rays has been seen, in past studies, to produce increasing frequencies of chromosome aberrations at lower x-ray energies. However, in one earlier irradiation experiment with chromium x-rays, the relative biological effectiveness (RBE) did not appear to be larger than that of hard x-rays, especially at higher doses. A possible reason for this unexpected result may have been the irradiation and culture conditions. We have, therefore, in the present study used a technique that has been developed in our laboratory to ensure uniformity of irradiation within lymphocytes and to avoid artefacts due to the cell cycle kinetics. Monolayers of 3-h-stimulated lymphocytes were exposed to 5.4 keV x-rays. A linear-quadratic dose-response was found for dicentrics. The comparison to an earlier finding with 220 kV x-rays shows the expected result of the RBE of the 5.4 keV x-rays to be above that of 220 kV x-rays. The three‐dimensional distribution of the specific absorption rate of energy (SAR) in phantom models was analysed to detect clusters of mobile phones producing similar spatial deposition of energy in the head. The clusters' characteristics were described from the phones external features, frequency band and communication protocol. Compliance measurements with phones in cheek and tilt positions, and on the left and right side of a physical phantom were used. Phones used the Personal Digital Cellular (PDC), Code division multiple access One (CdmaOne), Global System for Mobile Communications (GSM) and Nordic Mobile Telephony (NMT) communication systems, in the 800, 900, 1500 and 1800 MHz bands. Each phone's measurements were summarised by the half‐ellipsoid in which the SAR values were above half the maximum value. Cluster analysis used the Partitioning Around Medoids algorithm. The dissimilarity measure was based on the overlap of the ellipsoids, and the Manhattan distance was used for robustness analysis. Within the 800 MHz frequency band, and in part within the 900 MHz and the 1800 MHz frequency bands, weak clustering was obtained for the handset shape (bar phone, flip with top and flip with central antennas), but only in specific positions (tilt or cheek). On measurements of 120 phones, the three‐dimensional distribution of SAR in phantom models did not appear to be related to particular external phone characteristics or measurement characteristics, which could be used for refining the assessment of exposure to radiofrequency energy within the brain in epidemiological studies such as the Interphone. Experiments were designed to investigate the cytogenetic activity of some commonly used phenolic antioxidants, namely BHA, BHT, BMP and PG, all used widely to preserve and stablize food products rich in fats and oils. Barley and onion were used as test systems. The treatments reduced seed germination and induced a significant amount of seedling injury. Only PMP treatments produced a moderate amount of chromosomal aberrations while others failed to do so. Being strong antioxidants it was expected that these compounds would reduce the cytogenetic damage caused by the radiations. Contrary to expectations, however, a significant increase was observed in the radiation damage measured as seed and seedling lethality and chromosomal aberrations. Chemical post-treatments also increased the radiation damage. The damage was, however, reduced with ATP post-irradiation treatment.  相似文献

A method is described that takes advantage of the intermittency ("blinking") in the fluorescence of quantum dots (QDs) to measure absolute positions of closely spaced QDs. The concept is that even if two QDs are separated by only tens of nanometers, the position of each QD is resolvable if the point spread function of each can be imaged independently of the other. In the case of QDs, this is possible if each QD separately blinks completely on and off during a time-lapse sequence. To demonstrate the principle of this method, time-lapse sequences of single blinking QDs were acquired and the centroids of the point spread functions determined. Images of the blinking QDs were then overlapped in software, pixel by pixel, generating a range of submicroscopic distances between QD pairs. Methods were developed for analyzing the overlapped time sequences of the QD pairs so that the positions of the QDs and the distances between them could be determined without prior knowledge of the single QD positions. We subsequently used this method to measure the end-to-end length of a 122-basepair double-stranded DNA fragment.  相似文献

The Rubinstein-Taybi syndrome (RTS) is a well-defined complex of congenital malformations characterized by facial abnormalities, broad thumbs and big toes, and mental retardation. The breakpoint of two distinct reciprocal translocations occurring in patients with a clinical diagnosis of RTS was located to the same interval on chromosome 16, between the cosmids N2 and RT1, in band 16p13.3. By using two-color fluorescence in situ hybridization, the signal from RT1 was found to be missing from one chromosome 16 in 6 of 24 patients with RTS. The parents of five of these patients did not show a deletion of RT1, indicating a de novo rearrangement. RTS is caused by submicroscopic interstitial deletions within 16pl3.3 in approximately 25% of the patients. The detection of microdeletions will allow the objective confirmation of the clinical diagnosis in new patients and provides an excellent tool for the isolation of the gene causally related to the syndrome.  相似文献
