Characterizing denitrification rates in aquatic ecosystems is essential to understanding how systems may respond to increased nutrient loading. Thus, it is important to ensure the precision and accuracy of the methods employed for measuring denitrification rates. The acetylene (C2H2) inhibition method is a simple technique for estimating denitrification. However, potential problems, such as inhibition of nitrification and incomplete inhibition of nitrous oxide reduction, may influence rate estimates. Recently, membrane inlet mass spectrometry (MIMS) has been used to measure denitrification in aquatic systems. Comparable results were obtained with MIMS and C2H2 inhibition methods when chloramphenicol was added to C2H2 inhibition assay mixtures to inhibit new synthesis of denitrifying enzymes. Dissolved-oxygen profiles indicated that surface layers of sediment cores subjected to the MIMS flowthrough incubation remained oxic whereas cores incubated using the C2H2 inhibition methods did not. Analysis of the microbial assemblages before and after incubations indicated significant changes in the sediment surface populations during the long flowthrough incubation for MIMS analysis but not during the shorter incubation used for the C2H2 inhibition method. However, bacterial community changes were also small in MIMS cores at the oxygen transition zone where denitrification occurs. The C2H2 inhibition method with chloramphenicol addition, conducted over short incubation intervals, provides a cost-effective method for estimating denitrification, and rate estimates are comparable to those obtained by the MIMS method.  相似文献

A method for estimating denitrification and nitrogen fixation simultaneously in coastal sediments was developed. An isotope-pairing technique was applied to dissolved gas measurements with a membrane inlet mass spectrometer (MIMS). The relative fluxes of three N2 gas species (28N2, 29N2, and 30N2) were monitored during incubation experiments after the addition of 15NO3. Formulas were developed to estimate the production (denitrification) and consumption (N2 fixation) of N2 gas from the fluxes of the different isotopic forms of N2. Proportions of the three isotopic forms produced from 15NO3 and 14NO3 agreed with expectations in a sediment slurry incubation experiment designed to optimize conditions for denitrification. Nitrogen fixation rates from an algal mat measured with intact sediment cores ranged from 32 to 390 μg-atoms of N m−2 h−1. They were enhanced by light and organic matter enrichment. In this environment of high nitrogen fixation, low N2 production rates due to denitrification could be separated from high N2 consumption rates due to nitrogen fixation. Denitrification and nitrogen fixation rates were estimated in April 2000 on sediments from a Texas sea grass bed (Laguna Madre). Denitrification rates (average, 20 μg-atoms of N m−2 h−1) were lower than nitrogen fixation rates (average, 60 μg-atoms of N m−2 h−1). The developed method benefits from simple and accurate dissolved-gas measurement by the MIMS system. By adding the N2 isotope capability, it was possible to do isotope-pairing experiments with the MIMS system.  相似文献

The seasonal variation and depth distribution of the capacity for denitrification and dissimilatory NO3 reduction to NH4+ (NO3 ammonification) were studied in the upper 4 cm of the sediment of Norsminde Fjord estuary, Denmark. A combination of C2H2 inhibition and 15N isotope techniques was used in intact sediment cores in short-term incubations (maximum, 4 h). The denitrification capacity exhibited two maxima, one in the spring and one in the fall, whereas the capacity for NO3 ammonification was maximal in the late summer, when sediments were progressively reduced. The denitrification capacity was always highest in the uppermost 1 cm of the sediment and declined with depth. The NO3 ammonification was usually higher with depth, but the maximum activity in late summer was observed within the upper 1 cm. The capacity for NO3 incorporation into organic material was investigated on two occasions in intact sediment cores and accounted for less than 5% of the total NO3 reduction. Denitrification accounted for between 13 and 51% of the total NO3 reduction, and NH4+ production accounted for between 4 and 21%, depending on initial rates during the time courses. Changes of the rates during the incubation were observed in the late summer, which reflected synthesis of denitrifying enzymes. This time lag was eliminated in experiments with mixed sediment because of preincubation with NO3 and alterations of the near-environmental conditions. The initial rates obtained in intact sediment cores therefore reflect the preexisting enzyme content of the sediment.  相似文献

Measurements of denitrification using the acetylene inhibition,15N isotope tracer, and N2 flux methods were carried out concurrently using sediment cores from Vilhelmsborg sø, Denmark, in an attempt to clarify some of the limitations of each technique. Three experimental treatments of overlying water were used: control, nitrate enriched, and ammonia enriched water. The N2 flux and15N tracer experiments showed high rates of coupled nitrification/denitrification in the sediments. The acetylene inhibition method did not capture any coupled nitrification/denitrification. This could be explained by acetylene inhibition of nitrification. A combined15N tracer/acetylene inhibition experiment demonstrated that acetylene inhibition of N2O reduction was incomplete and the method, therefore, only measured approximately 50% of the denitrification due to nitrate from the overlying water. Similar rates of denitrification due to nitrate in the overlying water were measured by the N2 flux method and the acetylene inhibition method, after correcting for the 50% efficiency of acetylene inhibition. Rates of denitrification due to nitrate from the overlying water measured by the15N tracer method, however, were only approximately 35% or less of those measured by the acetylene inhibition or N2 flux methods.  相似文献

A mixed beech and spruce forest soil was incubated under potential denitrification assay (PDA) condition with 10% acetylene (C2H2) in the headspace of soil slurry bottles. Nitrous oxide (N2O) concentration in the headspace, as well as nitrate, nitrite and ammonium concentrations in the soil slurries were monitored during the incubation. Results show that nitrate disappearance rate was higher than N2O production rate with C2H2 blockage during the incubation. Sum of nitrate, nitrite, and N2O with C2H2 blockage could not recover the original soil nitrate content, showing an N imbalance in such a closed incubation system. Changes in nitrite and ammonium concentration during the incubation could not account for the observed faster nitrate disappearance rate and the N imbalance. Non-determined nitric oxide (NO) and N2 production could be the major cause, and the associated mechanisms could vary for different treatments. Commonly applied PDA measurement likely underestimates the nitrate removal capacity of a system. Incubation time and organic matter/nitrate ratio are the most critical factors to consider using C2H2 inhibition technique to quantify denitrification. By comparing the treatments with and without an antibiotic, the results suggest that microbial N uptake probably played a minor role in N balance, and other denitrifying enzymes but nitrate reductase could be substantially synthesized during the incubation.  相似文献

Four methods of monitoring the anaerobic digestion process were studied at pilot scale. The methods employed were Micro Gas Chromatography (μ-GC) and Membrane Inlet Mass Spectrometry (MIMS) for measurements in the gas phase, Near Infrared Spectroscopy (NIRS) and pH in the liquid phase. Micro Gas Chromatography accurately measured H2, CH4, H2S, N2 and O2 in the headspace whereas the MIMS accurately measured CH4, CO2, H2S, reduced organic sulfur compounds and p-cresol, also in the headspace. In the liquid phase, NIRS was found to be suitable for estimating the concentrations of acetate, propionate and total volatile fatty acids (VFA) but the error of prediction was too large for accurate quantification. Both the μ-GC and NIRS were low maintenance methods whereas the MIMS required frequent cleaning and background measurements.  相似文献

A method has been developed for measurement of denitrification activity in sediments by application of the acetylene inhibition technique. Acetylene-saturated water was injected, at close intervals, into sediment cores which were then incubated for a few hours at the in situ temperature. Frozen segments of the cores were assayed for accumulation of N2O by a combined gas extraction and detection system. The segments were thawed under a stream of helium from which N2O (and other gases) was collected in a liquid N2 trap, and the quantity of N2O was measured by gas chromatography. The maximum rate of denitrification in a coastal marine sediment was 35 nmol of N per cm3 of sediment per day at 2.5°C, and the rate of denitrification for the total sediment was 0.99 nmol of N per m2 per day.  相似文献

Microzonation of denitrification was studied in stream sediments by a combined O2 and N2O microsensor technique. O2 and N2O concentration profiles were recorded simultaneously in intact sediment cores in which C2H2 was added to inhibit N2O reduction in denitrification. The N2O profiles were used to obtain high-resolution profiles of denitrification activity and NO3 distribution in the sediments. O2 penetrated about 1 mm into the dark-incubated sediments, and denitrification was largely restricted to a thin anoxic layer immediately below that. With 115 μM NO3 in the water phase, denitrification was limited to a narrow zone from 0.7 to 1.4 mm in depth, and total activity was 34 nmol of N cm−2 h−1. With 1,250 μM NO3 in the water, the denitrification zone was extended to a layer from 0.9 to 4.8 mm in depth, and total activity increased to 124 nmol of N cm−2 h−1. Within most of the activity zone, denitrification was not dependent on the NO3 concentration and the apparent Km for NO3 was less than 10 μM. Denitrification was the only NO3-consuming process in the dark-incubated stream sediment. Even in the presence of C2H2, a significant N2O reduction (up to 30% of the total N2O production) occurred in the reduced, NO3-free layers below the denitrification zone. This effect must be corrected for during use of the conventional C2H2 inhibition technique.  相似文献

Denitrifying activity in a sediment from the freshwater part of a polluted estuary in northwest Europe was quantified using two independent approaches. High-resolution N2O microprofiles were recorded in sediment cores to which acetylene was added to the overlying water and injected laterally into the sediment. The vertical distribution of the rate of denitrification supported by nitrate uptake from the overlying water was then derived from the time series N2O concentration profiles. The rates obtained for the core incubations were compared to the rates predicted by a forward reactive transport model, which included rate expression for denitrification calibrated with potential rate measurements obtained in flowthrough reactors containing undisturbed, 1-cm-thick sediment slices. The two approaches yielded comparable rate profiles, with a near-surface, 2- to 3-mm narrow zone of denitrification and maximum in situ rates on the order of 200 to 300 nmol cm−3 h−1. The maximum in situ rates were about twofold lower than the maximum potential rate for the 0- to 1-cm depth interval of the sediment, indicating that in situ denitrification was nitrate limited. The experimentally and model-derived rates of denitrification implied that there was nitrate uptake by the sediment at a rate that was on the order of 50 (± 10) nmol cm−2 h−1, which agreed well with direct nitrate flux measurements for core incubations. Reactive transport model calculations showed that benthic uptake of nitrate at the site is particularly sensitive to the nitrate concentration in the overlying water and the maximum potential rate of denitrification in the sediment.  相似文献

Denitrification and total N losses were quantified from an irrigated field cropped to maize and wheat, each receiving urea at 100 kg N ha-1. During the maize growing season (60 days), the denitrification loss measured directly by acetylene inhibition-soil cover method amounted 2.72 kg N ha-1 whereas total N loss measured by 15N balance was 39 kg ha-1. Most (87%) of the denitrification loss under maize occurred during the first two irrigation cycles. During the wheat growing season (150 days), the denitrification loss directly measured by acetylene inhibition-soil cover and acetylene inhibition-soil core methods was 1.14 and 3.39 kg N ha-1, respectively in contrast to 33 kg N ha-1 loss measured by 15N balance. Most (70-88%) of the denitrification loss under wheat occurred during the first three irrigation cycles. Soil moisture and NO 3 - -N were the major factors limiting denitrification under both crops. Higher N losses measured by 15N balance than C2H2 inhibition method were perhaps due to underestimation of denitrification by C2H2 inhibition method and losses other than denitrification, most probably NH3 volatilization.  相似文献

The source of N2O produced in soil is often uncertain because denitrification and nitrification can occur simultaneously in the same soil aggregate. A technique which exploits the differential sensitivity of these processes to C2H2 inhibition is proposed for distinguishing among gaseous N losses from soils. Denitrification N2O was estimated from 24-h laboratory incubations in which nitrification was inhibited by 10-Pa C2H2. Nitrification N2O was estimated from the difference between N2O production under no C2H2 and that determined for denitrification. Denitrification N2 was estimated from the difference between N2O production under 10-kPa C2H2 and that under 10 Pa. Laboratory estimates of N2O production were significantly correlated with in situ N2O diffusion measurements made during a 10-month period in two forested watersheds. Nitrous oxide production from nitrification was most important on well-drained sites of a disturbed watershed where ambient NO3 was high. In contrast, denitrification N2O was most important on poorly drained sites near the stream of the same watershed. Distinction between N2O production from nitrification and denitrification was corroborated by correlations between denitrification N2O and water-filled pore space and between nitrification N2O and ambient NO3. This technique permits qualitative study of environmental parameters that regulate gaseous N losses via denitrification and nitrification.  相似文献

Electron transport system (ETS) activity, CO2 evolution, O2 consumption, N2-fixation (C2H2 reduction) and methanogenesis were appropriately measured in aerobic and anaerobically incubated sediment at 4, 10 and 20 ° C to better characterize these activities under different incubation conditions. ETS activity was always higher in the aerobically incubated sediment at all three incubation temperatures, whereas (C2H2 reduction was always greater in the anaerobic sediment. Carbon dioxide evolution was detected only in the aerobic sediment at 10 and 20 ° C but not at 4 ° C. Methane evolution in anaerobic sediment increased gradually with an increase in the incubation temperature.  相似文献

The acetylene block technique was employed to study denitrification in intertidal estuarine sediments. Addition of nitrate to sediment slurries stimulated denitrification. During the dry season, sediment-slurry denitrification rates displayed Michaelis-Menten kinetics, and ambient NO3 + NO2 concentrations (≤26 μM) were below the apparent Km (50 μM) for nitrate. During the rainy season, when ambient NO3 + NO2 concentrations were higher (37 to 89 μM), an accurate estimate of the Km could not be obtained. Endogenous denitrification activity was confined to the upper 3 cm of the sediment column. However, the addition of nitrate to deeper sediments demonstrated immediate N2O production, and potential activity existed at all depths sampled (the deepest was 15 cm). Loss of N2O in the presence of C2H2 was sometimes observed during these short-term sediment incubations. Experiments with sediment slurries and washed cell suspensions of a marine pseudomonad confirmed that this N2O loss was caused by incomplete blockage of N2O reductase by C2H2 at low nitrate concentrations. Areal estimates of denitrification (in the absence of added nitrate) ranged from 0.8 to 1.2 μmol of N2 m−2 h−1 (for undisturbed sediments) to 17 to 280 μmol of N2 m−2 h−1 (for shaken sediment slurries).  相似文献

Denitrification in aquatic sediments was measured by an N2O reductase assay. Sediments consumed small added quantities of N2O over short periods (a few hours). In experiments with sediment slurries, N2O reductase activity was inhibited by O2, C2H2, heat treatment, and by high levels of nitrate (1 mM) or sulfide (10 mM). However, ambient levels of nitrate (<100 μM) did not influence activity, and moderate levels (about 150 μM) induced only a short lag before reductase activity began. Moderate levels of sulfide (<1 mM) had no effect on N2O reductase activity. Nitrous oxide reductase displayed Michaelis-Menten kinetics in sediments from freshwater (Km = 2.17 μM), estuarine (Km = 14.5 μM), and alkaline-saline (Km = 501 μM) environments. An in situ assay was devised in which a solution of N2O was injected into sealed glass cores containing intact sediment. Two estimates of net rates of denitrification in San Francisco Bay under approximated in situ conditions were 0.009 and 0.041 mmol of N2O per m2 per h. Addition of chlorate to inhibit denitrification in these intact-core experiments (to estimate gross rates of N2O consumption) resulted in approximately a 14% upward revision of estimates of net rates. These results were comparable to an in situ estimate of 0.022 mmol of N2O per m2 per h made with the acetylene block assay.  相似文献

In this review of sediment denitrification in estuaries and coastal ecosystems, we examine current denitrification measurement methodologies and the dominant biogeochemical controls on denitrification rates in coastal sediments. Integrated estimates of denitrification in coastal ecosystems are confounded by methodological difficulties, a lack of systematic understanding of the effects of changing environmental conditions, and inadequate attention to spatial and temporal variability to provide both seasonal and annual rates. Recent improvements in measurement techniques involving 15 N techniques and direct N2 concentration changes appear to provide realistic rates of sediment denitrification. Controlling factors in coastal systems include concentrations of water column NO 3 , overall rates of sediment carbon metabolism, overlying water oxygen concentrations, the depth of oxygen penetration, and the presence/absence of aquatic vegetation and macrofauna. In systems experiencing environmental change, either degradation or improvement, the importance of denitrification can change. With the eutrophication of the Chesapeake Bay, the overall rates of denitrification relative to N loading terms have decreased, with factors such as loss of benthic habitat via anoxia and loss of submerged aquatic vegetation driving such effects.  相似文献

Estimates of denitrification are one of the key uncertainties in the terrestrial nitrogen (N) cycle, primarily because reliable measurements of this highly variable process—especially the production of its terminal product (N2)—are difficult to obtain. We evaluated the ability of gas-flow soil core and 15N tracer methods to provide reliable estimates of denitrification in forest soils. Our objectives were to: (1) describe and present typical results from new gas-flow soil core and in situ 15N tracer methods for measuring denitrification, (2) discuss factors that affect the relevance of these methods to actual in situ denitrification, and (3) compare denitrification estimates produced by the two methods for a series of sites in a northern hardwood forest ecosystem. Both methods were able to measure accumulations of N2 over relatively short (2–5 h) incubations of either unamended or tracer-amended intact soils. Denitrification rates measured by the direct flux soil core method were very sensitive to incubation oxygen (O2) concentration and decreased with increased O2 levels. Denitrification rates measured by the in situ 15N tracer method were very sensitive to the 15N content of the nitrate (NO3 ?) pool undergoing denitrification, which limits the applicability of this method for quantifying denitrification in N-poor ecosystems. While its ability to provide accurate estimates of denitrification was limited, the 15N tracer method provided estimates of the short-term abiotic and biotic transformations of atmospheric N deposition to gas. Furthermore, results suggest that denitrification is higher and that N2O:N2 ratios are lower (<0.02) than previously thought in the northern hardwood forest and that short-term abiotic and biotic transformations of atmospheric N deposition to gas are significant in this ecosystem.  相似文献

Membrane inlet mass spectrometry (MIMS) uses diffusion across a permeable membrane to detect in solution uncharged molecules of small molecular weight. We point out here the application of MIMS to determine catalytic properties of decarboxylases using as an example catalysis by oxalate decarboxylase (OxDC) from Bacillus subtilis. The decarboxylase activity generates carbon dioxide and formate from the nonoxidative reaction but is accompanied by a concomitant oxidase activity that consumes oxalate and oxygen and generates CO2 and hydrogen peroxide. The application of MIMS in measuring catalysis by OxDC involves the real-time and continuous detection of oxygen and product CO2 from the ion currents of their respective mass peaks. Steady-state catalytic constants for the decarboxylase activity obtained by measuring product CO2 using MIMS are comparable to those acquired by the traditional endpoint assay based on the coupled reaction with formate dehydrogenase, and measuring consumption of O2 using MIMS also estimates the oxidase activity. The use of isotope-labeled substrate (13C2-enriched oxalate) in MIMS provides a method to characterize the catalytic reaction in cell suspensions by detecting the mass peak for product 13CO2 (m/z 45), avoiding inaccuracies due to endogenous 12CO2.  相似文献

Acetylene disappeared from the gas phase of anaerobically incubated estuarine sediment slurries, and loss was accompanied by increased levels of carbon dioxide. Acetylene loss was inhibited by chloramphenicol, air, and autoclaving. Addition of 14C2H2 to slurries resulted in the formation of 14CO2 and the transient appearance of 14C-soluble intermediates, of which acetate was a major component. Acetylene oxidation stimulated sulfate reduction; however, sulfate reduction was not required for the loss of C2H2 to occur. Enrichment cultures were obtained which grew anaerobically at the expense of C2H2.  相似文献

An acetylene inhibition method was satisfactorily used for the in situ measurement of denitrification in two sediment-water systems incubated for not more than 22 h. In the presence of added nitrate, denitrification acted as a source of nitrous oxide in a drainage pond, but acted as a sink in its absence. The averaged rates of nitrous oxide accumulation with nitrate enrichment in the absence and presence of acetylene were 0.15 and 0.30 mg of N m−2h−1, respectively. Acetylene reduction at an average rate of 0.07 mmol of C2H4 formed m−2h−1 was simultaneously measured in the absence of added nitrate. In a small eutrophic lake where nitrogen was nonlimiting, the in situ rates of sediment denitrification were 0.09 and 0.11 mg of N m−2h−1 in the presence and absence of macrophytes, respectively, and no acetylene reduction activity was found.  相似文献
