Inflorescences of Tradescantia clone 4430 were exposed to different concentrations of radon (Rn) gas (0.85, 12.10, 36.50 and 98.16 kBq/m3) from plants placed in an acrylic chamber that received radon from a container with pitchblende (containing uranium mineral). The exposure time was 24 h, afterwards the plants were left for 6 h in water and constantly aerated. Positive control plants were irradiated with gamma rays (0.8 Gy) and negative control plants received ambient air only (the background measurement had a mean of 0.38 kBq/m3). Micronuclei (MCN) in the tetrads induced by alpha particles emitted from Rn were tabulated and a linear concentration response was obtained. The potency of radon to induce MCN from the slope of the regression line was 0.13 MCN/kBq/m3 of alpha-radiation. Radon could reach the anthers by diffusion through the aerial spaces within the buds.  相似文献

通过对蜡梅小孢子发生和花粉形成的研究,结果表明:蜡梅幼小花药中的多列孢原细胞经造孢细胞发良为小孢子母细胞。减数分裂为同时型。四分体呈四面体型排列,同时观察了小孢子在发育过程中液泡的动态变化。成熟花粉为2-细胞型。花药壁的发育为双子叶型。花药壁由5-6层细胞组成,腺质绒毡层。花粉具有异型性现象。  相似文献

西瓜(Citrullus lanatus)小孢子囊的孢原细胞出现在雄花原基出现后4—6天,孢原细胞数目推测只有一列;初生造孢细胞经过2—3次分裂,形成次生造孢细胞。开花前7—8天,小孢子囊发育健全,小孢子母细胞进入减数分裂期。同一花药不同花粉囊相同一药室,花粉母细胞减数分裂和小孢子的发育,并不是高度同步的。绒毡层为异型细胞,腺质绒毡层。雄配子体的发育开始于开花前6—7天,充分成熟的西瓜花粉已分裂为三细胞花粉。  相似文献

用石蜡切片法观察了七叶树花药的发育过程.结果表明:(1)雄蕊花药四室,花药壁完全分化时,从外到内依次是表皮、药室内壁、中层和绒毡层,花药壁发育为基本型.表皮细胞1层,发育过程中始终存在;药室内壁在花药成熟时形成带状纤维层加厚;幼小花药壁的中层3~4层细胞,在花药发育成熟时退化消失;绒毡层1层细胞,发育类型为分泌型,小孢子母细胞减数分裂时绒毡层开始退化解体,花药成熟完全消失,仅剩1层绒毡层膜.每一花药中有多列雄性孢原细胞,发生于幼小花药表皮下方;(2)小孢子母细胞减数分裂为同时型,四分体多呈正四面体排列;减数分裂过程中,小孢子母细胞外方被胼胝质壁所包被,小孢子形成后胼胝质壁逐渐消失.成熟花粉二细胞型,外形呈圆三角状,具三孔沟.  相似文献

首次报道了木莲属胚胎学特性,研究了红花木莲Manglietia insignis(Wall.)Bl.的小孢子发生、雄配子体的发育以及花粉萌发情况,红花木莲花药具有4个小孢子囊。花药壁5-6层,腺质绒毡层具有2-层细胞,小孢减数分裂后胞质分裂为修性同时型;四个分体排列方式为交叉型或左右对称型,偶为直线型;成熟花粉为二细胞型花粉粒,同时报道了化粉木莲在小孢子发生过程中的特有的现象,花粉萌发率为14.7%-25.3%。联系红花木莲花粉粒数目和胚珠数目进行探讨,认为雄配子体发育不是影响红花木莲结籽率低的主要因素。  相似文献

减数分裂前,莴苣花药中的钙颗粒很少。减数分裂后,花药绒毡层细胞中的钙颗粒明显增加。同时在花药药室基质中也出现许多细小的钙颗粒。刚从四分体中释放出的小孢子内钙颗粒很少。伴随着花粉外壁物质在小孢子表面的沉积,钙颗粒开始积累在花粉壁部位。随后。小孢子中开始出现钙颗粒。当小孢子开始形成液泡后,钙颗粒向其中聚集,伴随着小液泡融合成大液泡。体积较大的钙颗粒主要集中在液泡中,而细胞质基质中的钙颗粒很少。随着二胞花粉中的大液泡消失,花粉细胞质中的钙颗粒变得很少。在以后的发育中,只有花粉壁中积累较多的钙颗粒。在莴苣花药发育过程中,钙与绒毡层细胞的退化和小孢子液泡形成以及二胞花粉中大液泡的消失有关。而花粉外壁表面积累丰富的钙与以后花粉的萌发有关。  相似文献

Investigations of the growth of anthers and ontogeny of pollen grains of Oryza sativa (rice) IR-30 were undertaken for the purpose of 1) providing a set of growth measurements and 2) describing stable cytological features of anther and pollen development. Correlations exist between elongation of the floret and growth parameters of the anther such as its length, width, fresh and dry weights and cytological stage of pollen development. In the early ontogeny of the anther, hypodermal archesporial initials divide periclinally to form primary parietal cells and primary sporogenous cells. Each of the latter divides twice mitotically to generate four microspore mother cells, which undergo meiosis. The anther wall is formed by anticlinal and periclinal divisions of the primary parietal cells as well as of cells surrounding the primary sporogenous cells. Subsequent cytological features in the development of anther and pollen grains of rice have much in common with anther and pollen developmental biology of other members of Gramineae.  相似文献

The stage of pollen development at the time of anther culture is an important factor in the production of haploids. The objectives of the current study were to develop a staining procedure for peanut (Arachis hypogaea L., ssp. hypogaea) microspores, to describe and document the stages of microsporogenesis in peanut, and to confirm a previous report concerning correlations of peanut floral bud shape with stage of microspore development. A staining procedure using propionic carmine provided adequate staining of pollen mother cells, microspores, and pollen. Pollen mother cells and microspores could easily be differentiated by their size and cell wall structure. Plants grown in a controlled environment were found to have highly synchronized microspore development, both within an anther and among anthers contained in the same bud. In addition, floral bud shape was confirmed as a reliable indicator of anther stage in peanuts.  相似文献

Published data concerning the effects of indoor radon exposure on the frequency of chromosome aberrations in peripheral lymphocytes of residents are contradictory. Possible reasons for this may be the low radon concentration in dwellings and/or the limited number of investigated persons. We therefore studied the relationship of domestic radon exposure and the occurrence of chromosome aberrations in peripheral lymphocytes in 61 persons living in houses with radon concentrations from 80 up to 13,000 Bq/m3. We analyzed 60,000 cells from fluorescence plus Giemsa (FPG)-stained slides. It could be clearly demonstrated that in groups of persons living in dwellings with indoor radon concentrations >200 Bq/m3 the number of cells containing dicentrics and/or centric rings (C(dic + cr)) (2.45 +/- 0.50 x 10(-3)) was significantly increased (p < 0.05) in comparison to the control level (1.03 +/- 0.15 x 10(-3)). However, there was no difference in the mean frequency of C(dic + cr) between the groups living in dwellings with higher radon concentrations. Using the fluorescence in situ hybridization (FISH) technique for the detection of translocations, we analyzed 23,315 cells in 16 persons of the highest exposed group (>5,000 Bq/m3). The observed frequency of translocations was 3.9 +/- 0.64 x 10(-3). In comparison to the control group (2.02 +/- 0.18 x 10(-3)), there was a slight but not statistically significant increase in the exposed group (P = 0.055). If, however, the age of the examined persons is taken into account, the values are significantly increased (P < 0.05) in the exposed persons older than 40 years in comparison to the age-matched controls. Since most of the translocations were found in stable cells, it is concluded that translocations are also induced in blood-forming tissue and are transmitted to peripheral blood.  相似文献

In flowering plants, pollen formation depends on the differentiation and interaction of two cell types in the anther: the reproductive cells, called microsporocytes, and somatic cells that form the tapetum. Previously, we cloned a pollen specific gene, zm401, from a cDNA library generated from the mature pollen of Zea mays. Expression of partial cDNA of zm401 in maize and ectopic expression of zm401 in tobacco suggested it may play a role in anther development. Here we present the expression and functional characterization of this pollen specific gene in maize. Zm401 is expressed primarily in the anthers (tapetal cells as well as microspores) in a developmentally regulated manner. That is, it is expressed from floret forming stage, increasing in concentration up to mature pollen. Knockdown of zm401 significantly affected the expression of ZmMADS2, MZm3-3, and ZmC5, critical genes for pollen development; led to aberrant development of the microspore and tapetum, and finally male-sterility. Zm401 possesses highly conserved sequences and evolutionary conserved stable RNA secondary structure in monocotyledon. These data show that zm401 could be one of the key growth regulators in anther development, and functions as a short-open reading-frame mRNA (sORF mRNA) and/or noncoding RNA (ncRNA).  相似文献

植物雄性不育既是研究植物生殖生物学重要的植物学性状也是研究作物杂种优势利用重要的农艺性状,在遗传和分子生物学中具有重要地位。以模式植物拟南芥和水稻为主,对植物雄性不育的控制基因和相关分子机理已有众多进展,按照花药发育时期和雄性败育的表现形式可以归纳为减数分裂异常、胼胝质代谢异常、绒毡层发育异常、花粉壁发育异常、花药开裂异常,以及其它类型的雄性不育。在不育相关基因中,导致胼胝质代谢异常、绒毡层发育异常和花粉壁发育异常的基因往往表现一因多效,一个相关基因的突变会产生复合表型。关于植物雄性不育相关基因的研究表明,雄性器官和小孢子形成过程中的任何相关基因的改变,均可导致雄性不育的产生。本文总结了植物核基因雄性不育的研究进展,以期促进不同物种间雄性不育基因的比较分析,使植物雄性不育研究更加深入。  相似文献

采用石蜡切片技术对百合科植物山麦冬大小孢子发生及雌雄配子体发育进行了观察研究。结果表明:(1)山麦冬花药具有4个花粉囊,花药壁的发育方式为基本型,花药壁完全分化时由表皮、药室内壁、中层及绒毡层组成。(2)绒毡层发育类型为分泌型,到四分体孢子彼此分离形成单细胞花粉阶段,绒毡层细胞开始解体退化,花粉成熟时绒毡层细胞完全消失;花粉母细胞减数分裂为连续型,四分体为左右对称形排列,成熟花粉为3-细胞花粉,单萌发沟。(3)子房3室,每室2枚胚珠,胚珠倒生型,双珠被,薄珠心,雌性孢原细胞不经过平周分裂而直接发育而成大孢子母细胞。(4)减数分裂后四分体大孢子呈线型或T型排列,合点端大孢子分化为功能大孢子,胚囊发育为蓼型;花粉母细胞减数分裂过程中,二分体、四分体细胞外方被胼胝质壁所包被,小孢子形成后胼胝质壁逐渐消失。该研究结果丰富了百合科植物生殖生物学研究的内容,也为探讨百合科植物的系统学研究提供了参考。  相似文献

苍术属(Atractylodes DC.)是菊科菜蓟族(Cynareae)刺苞亚族(Carlininae O. Hoffm.)的一个东亚特有属,世界上仅有7种,其中我国有5种。该研究以关苍术为材料,采用石蜡切片法比较研究了两性花和雌花的花药及雄配子体发育进程,并进一步探讨了其雌花产生花药退化的时期及原因。结果表明:(1)关苍术小孢子发育与花蕾长度间存在相关性,当花蕾长度在5 mm时进入花粉母细胞时期,花药壁已分化,在7~9 mm时处于四分体时期,大于11 mm时开始进入花粉粒时期。(2)关苍术花药5个,花粉囊4个,减数分裂属同时型,四分体以正四面体为主,属3-细胞型,萌发沟3个。(3)关苍术花粉囊壁发育属双子叶型,从外层的表皮、药室内壁,到内层的中层和绒毡层均由一层细胞构成,关苍术绒毡层为腺质绒毡层。(4)关苍术雌花花药退化发生在花药发育早期至四分体时期,表现为花药发育早期畸形、药壁分化异常、小孢子母细胞发育停滞在前期、绒毡层增生4个原因。该研究结果为苍术属植物的系统发育、物种形成和进化提供胚胎学依据。  相似文献

以不同发育时期的凤仙花花药为实验材料,采用组织化学方法,对花药发育中的结构变化及多糖和脂滴物质分布进行观察。结果表明:(1)凤仙花的花药壁由6层细胞组成,包括1层表皮细胞,2层药室内壁细胞,2层中层细胞和1层绒毡层细胞。其中绒毡层细胞的形态不明显,很难与造孢细胞区分,且在小孢子母细胞时期退化。(2)在小孢子母细胞中出现了一些淀粉粒,但减数分裂后,早期小孢子中的淀粉粒消失,又出现了一些小的脂滴;随着花粉的发育,小孢子形成大液泡,晚期小孢子中的脂滴也消失;小孢子分裂形成二胞花粉后,营养细胞中的大液泡降解、消失,二胞花粉中又开始积累淀粉;接近开花时,成熟花粉中充满细胞质,其中包含了较多的淀粉粒和脂滴。(3)在凤仙花的花药发育中,绒毡层细胞很早退化,为小孢子母细胞和四分体小孢子提供了营养物质;其后的中层细胞退化则为后期花粉发育提供了营养物质。  相似文献

一品红花药来源于雄蕊原基,花药由表皮(1层)、药室内壁(1层)、中层(1层)、绒毡层(1层)及造胞细胞组成,花药四室,药壁发育为双子叶型。小孢子发生和雄配子体发育是经由小孢子母细胞减数分裂形成四分体,该四分体胞质分裂为同时型,四分体排列为四面体型,小孢子再经有丝分裂形成2-核花粉。花药壁层的变化是表皮在花药成熟期消失,中层在四分体时消失,药室内壁在花药成熟期形成柱状纤维层。绒毡层在单核小孢子期径向伸长,有双核或多核,另外有的绒毡层细胞形成横隔或类胎座;进入2-核花粉期,绒毡层细胞分泌颗粒物进入药室,为非典型腺质绒毡层;进入成熟期绒毡层消失。同时观察到花药发育异常现象。  相似文献

Summary Studies of the chromosomal composition of pollen plants regenerated from the F1 of hybrids produced from Triticum-Agropyron intermediate type and common wheat demonstrated that various gametic types of the F1 could be fully expressed at the whole plant level via anther culture. The observed frequency of each of the eight types of pollen plants (based on their chromosome numbers) was in good agreement with the theoretical probabilities as shown by X2 analysis. Comparative studies of the chromosome composition of somatic cells and pollen mother cells (PMC's) of selected pollen plants permitted classification of the plants into four distinct classes. The majority of these regenerated pollen plants had identical chromosome numbers in both root tip cells and PMC's. An alien disomic addition line, which was cytologically stable for two generations, was obtained directly from anther culture. Moreover, the addition line exhibits resistance to stripe rust disease, a trait which is conferred by the Agropyron chromosome. We suggest that anther culture techniques provide a unique and expeditious route for the introduction of alien genes or chromosomes into wheat cultivars.  相似文献

兰科植物的有性生殖特殊,每朵花只有1个花药,且花粉有聚集成块发育的特征。为了揭示铁皮石斛花粉块的发育特征,该研究以野生铁皮石斛不同时期的花药为材料,采用半薄切片和植物组织化学方法对其发育过程进行解剖学观察分析,并对成熟花粉块进行离体培养,观察花粉管的萌发状况。结果表明:(1)铁皮石斛花药壁由1层表皮细胞,2层药室内壁细胞,1层中层细胞和1层绒毡层细胞组成。开花时,绒毡层细胞退化,中层细胞没有退化,药室内壁细胞则形成纤维状细胞壁;药室中的小孢子母细胞没有明显的胼胝质壁结构。(2)小孢子发生属同时型,减数分裂后四分体小孢子不分散,以四合花粉状态发育,并进一步连接形成花粉块。(3)在小孢子发育中,孢粉素覆盖在整个花粉块表面形成花粉外壁,但花粉块内部的花粉没有花粉外壁结构;在花粉块表面的花粉外壁上未见花粉萌发孔。(4)在花粉离体萌发实验中,具有花粉外壁的花粉块表面花粉未见萌发,仅由花粉块内部的花粉萌发出花粉管。  相似文献

This paper deals with the electron microscopic observations of anther tissues and microsporogenesis of Tai Gu male sterile wheat plant, which is a new species initially discovered in Tai Gu nE Shansi province, China. The results are summarized as follows: 1. There was a tendency of early development of tapetum cells in Tai Gu sterile wheat plant, i.e. the tapetum cells seemed to have developed rapidly toward maturation between the period of pollen mother cell formation and prophase I of reduction division, which is followed by degeneration. Its ultrustructure is somewhat similar to the highly developed tapetum cells of fertile plant at the time of development from tetrad to early microspore stage. A significant character of the nltrastructure of the highly developed mature tapetum cells was the appearance of many sharply-defined endoplasmic reticulum, which demonstrated in a parallelly arranged structure in section. Phagocytic vacuoles were often surrounded by the endoplasmic reticulum. 2. At the prophase I of reduction division, the ultrastructure of pollen mother cells in sterile plant began to show some disparities from that of fertile pIant. Among which there were some concentric circle-shaped or bundle-shaped endoplasmic reticula and many ribosome aggregation in the pollen mother ceils of sterile plant. Perhaps, these features might be relevant to the break down of microtrabecular system. In addition, some large or small dark osmiophilic particles were visible in the cells. From all the feature observed, the fact that activities of endoplasmic reticulum in the anther of Tai Gu wheat is relation to protein synthesis warrants our attention.  相似文献

利用常规石蜡切片技术对柠条锦鸡儿小孢子发生及雄配子体发育的过程进行了观察,为柠条锦鸡儿生殖生物学提供基础资料。结果表明:(1)柠条锦鸡儿雄蕊花药4室,花药壁完全分化时,由外到内依次是表皮、药室内壁、中层和绒毡层,花药壁发育为基本型;表皮细胞1层,发育过程中始终存在;药室内壁在花药成熟时形成带状纤维层加厚;幼小花药壁的中层1~2层细胞,在花药发育成熟时退化消失;绒毡层1层细胞,腺质绒毡层,花药成熟时消失。(2)小孢子母细胞减数分裂过程中的胞质分裂为同时型,产生四面体型和左右对称型小孢子。(3)成熟花粉粒为二细胞型,扫描电镜下观察其成熟花粉粒为圆球形,外壁近光滑。(4)花粉母细胞分裂后形成的四分体小孢子中出现多核仁现象,核仁数在2~6个范围变化,推测这可能和末期Ⅱ核仁融合的不彻底有关。研究发现,柠条锦鸡儿小孢子发生和雄配子发育过程没有发现异常现象。  相似文献
