Architectural and histometric features for the objective grading of prostate adenocarcinoma in histologic specimens were analyzed in five cases each of Gleason primary grades 2, 3A, 3B, 3C, 4A and 4B, selected as "typical" for the histopathologic images. Tissue sections from the selected cases were stained by the Feulgen method. Fifteen fields for each grade, for a total of 4,430 glands, were digitized by a video-based microphotometer at low resolution (pixel spacing of 2 microns). Outer and inner outlines of the glandular epithelium were traced manually using a mouse. For each field the number of glands, the gland area, the lumen area, the area of the glandular epithelium and the cribriformity factor were computed. The gland area and its variance proved to be useful indicators for lower-grade lesions, whereas the variance of cribriformity resulted in an excellent grading indicator in the Gleason 3-4 range when cribriform glands were present.  相似文献

The proportion of stromal fat cells to parenchymal cells in 100 normal parathyroid glands was determined by the image analyzing computer technique. The parathyroid glands were resected at the time of thyroidectomy in 86 patients with thyroid tumors. None of the patients had any evidence of parathyroid dysfunction preoperatively. In the histologic sections of the parathyroid glands, the average percentage of stromal fat cell content was 38%. The percentage of stromal fat cells was correlated with the age and the body constitution of the patients, but the percentages of fat cells varied widely among glands in the given age and body constitution ranges. It was therefore not possible to discriminate a normal parathyroid gland from an abnormal gland solely on the basis of microscopic determination of stromal fat cell content.  相似文献

The reproductive tract of B. truncatus was investigated histologically in order to study possible effects of castration upon the accessory sex glands. In the female part of the reproductive tract—subdivided into albumen gland, oviduct, muciparous gland, oothecal gland, uterus, vagina and bursa copulatrix—13 histochemically different secretory cell types were distinguished. The majority produce different types of (acid) mucopolysaccharides. The roles of the various parts of the female tract in the production of an egg mass were elucidated by comparing the histochemistry of the egg mass to that of the female tract; the abundance and location of the cell types were also taken into account for this purpose.

The male part appeared to contain 12 histochemically different secretory cell types. These produce mainly (phospho lipoproteins together with some polysaccharides and neutral lipids.

Castration causes an acceleration of the growth of the snails. The volumes of female (albumen gland) and male (prostate gland) accessory sex glands were measured on histological sections. It appeared that growth of the albumen gland is not arrested by castration. This was not established beyond doubt for the prostate gland. The results suggest that the stimulating effects of the dorsal body hormone on the growth and synthetic capacity of the female accessory sex glands—such effects have been established for Lymnaea stagnalis—are not exerted via the ovotestis in B. truncatus.  相似文献

The testes, prostate and vesicular glands of 10 bull calves were examined by ultrasonography every 2 wk from 2 to 46 wk of age, at which time the scrotal circumference (SC) of all the calves had reached pubertal size (28 cms). Computer-assisted image intensity analysis (numerical pixel values) was conducted. Blood samples were collected every other week from 2 to 46 wk of age. Testicular diameter increased in a linear manner from 2 to 46 wk of age, but the diameter measured in a transverse plane (caudal) was greater between 10 and 34 wk of age than when measured in a longitudinal (lateral) plane (P<0.05). Growth of the prostate and vesicular glands, based on dimensions, was linear, but vesicular gland length increased more rapidly after 32 wk of age (P<0.05). Image intensity of the vesicular glands and prostate declined from birth or 8 wk of age, respectively, to 14 wk of age, increased to 18 wk and then declined to a nadir at 30 wk, followed by a rapid increase to 34 wk of age for the vesicular glands and to 46 wk of age for the prostate (P<0.05). Image intensity of the testes showed an early increase to 6 to 8 wk of age and a subsequent increase from about 20 wk of age to 46 wk of age, with an inflection at 30 wk of age (P<0.05). There was a transient increase in mean serum concentrations of LH between 6 and 20 wk of age (P<0.05), and LH concentrations appeared to increase again after 36 wk of age (P>0.05). Mean serum concentrations of FSH declined with age (P<0.05). Mean serum concentrations of testosterone increased after 32 wk of age (P<0.05) In summary, numerical pixel values comprising the ultrasound images of the developing testes, prostate and vesicular glands revealed a complex development pattern that may reflect important details of developmental stages.  相似文献

Thorough examinations of the reproductive system of boars are generally not performed on normal boars to be used for breeding; only boars with problems undergo a form of a breeding soundness examination. In order for veterinarians to identify pathological conditions, the normal architecture of the accessory sex glands needs to be described. The purpose of this study was to use B-mode ultrasonography to describe the accessory sex glands in the boar and to see if transrectal ultrasonography would be a viable option in which to obtain this data. Initially, cross-sectional saline bath examinations of accessory sex glands were performed on crossbred boar reproductive tracts (n = 4) using B-mode ultrasonography equipped with a 5 MHz dual frequency linear array transducer. In situ examinations were also performed on terminal line crossbred boars (n = 16) ranging in age from 10 to 23 months old using the same ultrasound methodology; four boars were under general anesthesia and the remaining 12 were standing in crates. Eight boars were abstinent for 2 days and the other eight had ejaculates collected 2 h prior to examination. The paired bulbourethral glands are best described as a long oval gland with a uniformly echogenic appearance with a large anechoic space in the center of the gland extending most of its length. The walls of the vesicular glands were found to be thin, with the parenchyma having multiple small echolucent areas that appeared to merge and form a central canal. The prostate gland was best identified as a pecan-sized gland with a uniform echogenic appearance. Visualization of the prostate gland was accomplished with more proficiency using the saline bath ultrasonography as compared to in situ examinations. All of the accessory sex glands could be examined using both methodologies of ultrasonographic examination with a 5 MHz frequency linear array transducer. It was determined that each accessory sex gland could be recognized, and differences between ejaculated and nonejaculated boars could be identified. The results of this study demonstrate that transrectal ultrasonography can be used as a diagnostic aid in assessing the accessory sex glands of boars.  相似文献

Synopsis The histochemical localization of carbohydrates, ribonucleoproteins (RNA), lipids, some hydrolytic enzymes, succinate and lactate dehydrogenase and acetylcholinesterase were investigated in the prostate, urethral and bulbourethral glands of the camel. These glands probably secrete carbohydrate-protein complexes. In the bulbourethral glands, they are sulphated mucopolysaccharides. RNA was seen in the cytoplasm of the prostate and urethral glands. Neutral lipids were cytoplasmic and present in moderate amounts in the prostate and urethral glands and in traces, in the bulbourethral gland. Acid phosphatase-containing granules were abundant in the prostate, moderate in the urethral glands and in traces in the bulbourethral glands. Alkaline phosphatase was observed in the apical cytoplasm of the prostate and bulbourethral glands and in the ducts of the urethral glands. ATPase and adenosine 5-monophosphatase were seen in the basal laminae and interstitial tissue. In the urethral glands, adenosine 5-monophosphatase was distributed diffusely in the cytoplasm. Succinate dehydrogenase was seen in the urethral and bulbourethral glands. Varying degrees of lactate dehydrogenase activity was observed in all the glands. Acetylcholinesterase was confined to neural elements. The pars disseminata and the urethral glands were considered as two distinct glandular zones along the pelvic urethra. The significance of these histochemical results is discussed.  相似文献

Three of 6 female long-clawed shrews, Sorex unguiculatus Dobson, 1890, collected on the island of Hokkaido, Japan, were found to have unsporulated oocysts and sexual stages (both macro- and microgamonts) in varying stages of development of an unidentified coccidium in both lactating and nonlactating mammary glands. Gamonts developed in the alveoli of the mammary glands, and oocysts were found in the lactiferous ducts and in pools of milk. Mature macrogamonts were 11.9 x 15.2 microm (10-14 x 14-20 microm), whereas completely developed microgamonts with many gametes were 14.8 x 16.8 microm (10-18 x 13-20 microm). Oocysts in tissue sections were 19.5 x 13.8 microm and had a smooth outer wall that was <1 microm thick. Little histopathology was associated with the infections. Infected cells were enlarged and appeared cloudy, and in some areas there was leucocytic infiltration by macrophages, small and large lymphocytes, neutrophils, and eosinophils. No basophil was seen. We also found sections of a nematode, probably a Mammanidula sp., in sections of an active mammary gland in 1 of the shrews not infected with the coccidium.  相似文献

Epidermal growth factor (EGF) stimulates the growth of many tissues and inhibits stimulated gastric acid secretion. Its primary tissue of origin in man is still unknown. We used polyclonal anti-human EGF sera in the peroxidase-antiperoxidase immunocytochemical staining technique to identify immunoreactive human EGF (ihEGF) in tissue sections from 29 subjects ranging from fetuses to 63 years in age. In addition to acinar cells in the submandibular salivary glands and cells of Brunner's duodenal glands, previously reported to contain ihEGF, we found ihEGF in most anterior pituitary glycopeptide hormone-secreting cells, in gastric and pyloric gland cells of the stomach, and in bone marrow cells that resembled mononuclear phagocytes in subjects of all ages. The eccrine sweat glands in the skin of adults also contained ihEGF. Cells containing ihEGF were found singly or in clusters in the trachea of the fetus only. No fetal pancreatic islet cells stained, but occasional cells in neonates and a majority of islet cells in older subjects contained ihEGF; there was no constant association with insulin, glucagon, or somatostatin. Only the lactating breast contained ihEGF. In adults, outer adrenomedullary cells contained ihEGF. Intense immunostaining was observed in the renal medulla, apparently limited to the extracellular area between the renal tubules, and increased with age; the cortex was devoid of ihEGF. No ihEGF was detected in posterior pituitary gland, thyroid gland, heart, lung, or liver at any age. An adult prostate contained ihEGF only in an area of local injury, and some primordial follicles from the ovary of a newborn appeared to contain ihEGF. Thus, many tissues appear to synthesize hEGF, which may exert exocrine, endocrine, or paracrine functions in different tissues and at different ages.  相似文献

The cloacal glands of the male marbled newt Triturus marmoratus marmoratus were studied during winter and summer by histochemical and quantitative histologic methods. Four types of glands were distinguished: pelvic, dorsal, ventral, and Kingsbury's glands. The pelvic and dorsal glands have an eosinophilic epithelium and secrete neutral mucins. The ventral and Kingsbury's glands have a basophilic epithelium and secrete acid mucins. The lectin-histochemical characterization of the carbohydrates secreted by the four gland types revealed that the secretion of both the pelvic and Kingsbury's glands contain β-GalNAc in the peripheral region of the oligosaccharide, and that the dorsal glands secrete a glycoprotein with α-GalNAc. The ventral gland sections did not react to any of the lectins used here. The quantitative study revealed that the cloaca undergoes seasonal variations in volume, being significantly larger in winter than in summer. The total volume occupied by both the pelvic and ventral glands, as well as their tubular diameter, are also significantly greater in winter, while these parameters do not vary in dorsal and Kingsbury's glands. Summary Ornithine decarboxylase was localized immunohistochemically in paraffin sections of prostate glands of the rat using Sternberger's PAP technique. Two types of immunoreactive cells were detected in the gland epithelium: strongly positive ones and weakly positive ones. Unreactive cells were also present. The functional role of the enzyme in the secretory process of the glands is discussed in the light of these findings.  相似文献

In order to investigate the effects of chronic lead intoxication on sexual accessory glands adult male Wistar rats were poisoned by ad libitum ingestion of lead acetate at concentrations of 0.5 g/l and 1.0 g/l for 90 d. Blood lead exhibited a significant increase in both treated groups. A decrease in hematocrit and hemoglobin, besides a rise in glycemic levels, confirmed lead intoxication. No sign of lesion was detected upon histologic examination of prostate, seminal vesicle, and coagulating gland. A morphometric technique revealed that, in this early phase of intoxication, no alteration occurred in the prostate's and seminal vesicle's epithelial height. However, a significant decrease in fructose content was observed in both lead treated groups. The results, revealing a precocious envolvement of male sexual accessory glands in lead intoxication, were related to disturbance in plasma testosterone level and also probably in androgen receptors.  相似文献

OBJECTIVE: To compare the morphology of different parenchymal cell types in healthy human parathyroid glands (HPGs) in cytologic smears with their structure on histologic sections and to establish criteria for their recognition in smears. STUDY DESIGN: The HPGs of 47 subjects (27 females and 20 males) were incidentally removed during surgery on the thyroid gland. The tissue of glands with a normal macroscopic and microscopic appearance was analyzed in cytologic smears and on histologic sections. RESULTS: In cytologic smears (as well on histologic sections), dark and light chief cells predominated. Dark chief cells, which were more numerous than light ones, had a smaller nucleus without a visible nucleolus. The nucleus of light chief cells was larger with 1 or 2 nucleoli visible. The cytoplasm of both types of chief cells was poorly defined, grey-blue and often vacuolated. In the smears, the cytoplasm of oxyphilic cells was dense, gray-rose and well defined. In dark oxyphilic cells, the nuclei were smaller and without a visible nucleolus. Light oxyphilic cells had a larger nucleus and visible nucleolus. CONCLUSION: The identification and distribution of 4 parenchymal cell subtypes in the smears of pathologically altered HPGs may yield insights into the possible role of these cells in a specific disease.  相似文献

The aim of this study was to elucidate the distribution and morphological changes of tight junctions during secretion in parotid gland acinar cells. Localization of tight junction-associated polypeptide ZO-1, and of tight junction transmembrane protein Occludin, was examined in rat parotid gland by immunofluorescence and immunogold labelling of ultrathin sections. Adult male Sprague-Dawley rats were intraperitoneally injected with IPR and, after 10 and 30 minutes, parotid glands were extirpated. In control specimens, positive immunoreaction for ZO-1 and Occludin was observed on the adluminal side between adjacent cells in the form of narrow elongated profiles corresponding to intercellular canaliculi. After IPR injection, canaliculi became dilated and fluorescence was no longer seen as a continuous line but appeared as an aggregation of separate bright particles. ZO-1 was more widely distributed and was recognized in other areas of the cytoplasm as well. Concurrently, omega-shaped concavities, marked by actin fluorescence, appeared along the intercellular canaliculi. We concluded that, during exocytosis, the selective permeability barrier to the paracellular pathway, based on tight junctions, becomes more leaky, owing to segregation of Occludin caused by intracellular ZO-1 distributional changes associated with actin filaments.  相似文献

睾丸切除后,家猫前列腺背叶、腹叶及尿道球腺内的金属硫蛋白(metalothionein,MT)分别下降至正常家猫的212%(P<001)、884%(P>005)和185%(P<001),而在腹叶影响较小。睾丸切除后注射芝麻油,前列腺背叶及尿道球腺MT均未得到恢复。但若在睾丸切除后连续3d注射10μg/kgbw睾酮,两者依次恢复至693%和594%。随睾酮注射剂量增加(5、10、15、20、25μg/kgbw),血浆睾酮的浓度、前列腺背叶及尿道球腺MT含量增高。血浆睾酮与前列腺背叶及尿道球腺MT呈正相关(P<001)。这些结果表明,睾酮诱导前列腺背叶及尿道球腺MT,其最适剂量为20μg/kgbw。  相似文献

OBJECTIVE: To objectively characterize the architectural spectrum of Gleason pattern 3 prostate cancer (PCA) in a biologically meaningful manner. STUDY DESIGN: We define an objective architectural feature of PCA, "pinch point density" (PPD), and explore its relationship to proliferative index (PI). A pinch point (PP) is a site where the epithelium of two neighboring glandular structures is contiguous in one histologic section but not in an adjacent serial section. Seventeen radical prostatectomy specimens with areas of pure Gleason pattern 3 were studied. PPD was measured with computer aid using digital images of serial sections. PI was measured by computer-aided counting of Ki-67-positive cells. RESULTS: PPD correlated inversely with PI (PPD vs. log [PI], P < .004). Characteristics not significantly correlated with PI included total number of malignant glands, PP per gland and total number of malignant cells. Subjectively, tumors with high PPD and low PI tended to contain a larger number of smaller glands as compared to tumors with low PPD and high PI. This impression was confirmed analytically. CONCLUSION: PPD is an objective architectural feature of possible biologic significance. This is an early step toward identifying objective features of growth pattern in Gleason pattern 3 PCA that may be clinically meaningful.  相似文献

In rabbit stomach, gastric lipase activity level was found to increase from birth to 30 days old (weaning), and then decreased. In contrast, pepsin activity only appeared between 30 to 45 days old, and increased till to the adult level. It was observed that maturation of gastric glands in cardial mucosa was a downward elongation process from the mitotic cell pool. These mitotic cells were always found in the neck of the gastric glands, corresponding to the bottom of the gland at 6 days old and to the mid-zone of the gland in adult. Location of rabbit gastric lipase (RGL) cells in cardial glands varied with age and was found along the pit of the gastric glands at 6 days old. The extent of this cellular location decreased with age, whereas a second RGL cell zone appeared below the mitotic cell area at 18 and 30 days old. At 45 days old, the pepsinogen cells appeared in the bottom of the gland, and consequently the RGL cells were located in the mid-zone of the gastric glands, between mitotic cells (neck of the gland) and pepsinogen cells (lower part of the gland). Ultrastructural study of cardial gastric glands revealed different morphologies of the secretion granules in the cells along the gastric glands. In 6-day-old rabbits, secretory granules were found uniformly electron dense in the bottom of the glands and were RGL-labeled by the immunogold technique. In the medium part of the glands, granules appeared biphasic, with a clear and a dense part, and RGL labeling was confined to the electron-dense part.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

We used light and transmission electron microscopy to examine the morphology of the accessory glands of immature and mature adult males of Apis mellifera L. We also made an electrophoretic analysis of the protein content of the mature gland. The glands of the immature male actively secrete a mucous substance that can be seen in the lumen of the gland of the mature male. This secretion stains with mercury bromophenol blue and with periodic acid-Schiff reaction, which stain glyconjugates. The protein content was higher in the lumen secretion than in the gland wall extracts. The electrophoresis patterns of the wall extracts were different from those of the secretion found in the gland lumen.  相似文献

Annexins (ANXs) represent a family of calcium and phospholipid binding proteins that are involved in several physiological processes e.g. signal transduction, cellular differentiation and proliferation. Since they are known to be dysregulated in a variety of cancers we investigated the immunolocalization of ANXs in whole prostate sections containing benign prostatic epithelium (BPE), benign prostatic hyperplasia (BPH), prostatic intraepithelial neoplasia (PIN) and prostate cancer (PCa) in order to evaluate their possible role during tumorigenesis. Samples were obtained from 28 patients undergoing radical prostatectomy. Gross sections of whole prostates were examined immunohistochemically for the distribution of ANX I, II, IV and VII. In BPE all ANXs were localized to the cell membranes and the cytoplasm of all gland cells. In BPH the immunoreactivity of ANX I and II was restricted to the basal cells of glands and expression pattern of ANX IV and VII was similar to BPE. In PIN only basal cells expressed ANX II. In PCa ANX II immunoreactivity was absent and weak ANX I and ANX IV immunoreactivity was restricted to the cytoplasm of tumor cells. ANX VII immunoreactivity was seen in some but not all tumor cells. Since ANX IV and VII expression did not show significant changes in PCa compared to non-neoplastic tissue and PIN an essential role during prostate tumourigenesis seems unlikely. In contrast, as progression from PIN to PCa is characterized by a reduction of ANX I and II this suggests that downregulation of these proteins could represent an important event in prostate carcinogenesis.  相似文献

Summary The effect of copulation on the rat coagulating gland (anterior prostate) was studied. At 4 to 6 h after the beginning of copulation the coagulating glands of rats that had produced copulatory plugs were nearly empty of secretion. Ultrastructurally, the coagulating gland has large cisternae of rough endoplasmic reticulum (RER) and few condensing vacuoles or secretion granules. After copulation the number of secretion granules and the frequency of their expulsion into the lumen increased. Also in the lumen were fragmentation vesicles (50–100 nm diameter) that were bounded by a unit membrane and appeared to arise from microvilli. At 4, 6, and 7h after the beginning of copulation there was an increase in apical blebbing. Blebbing was found in both perfusion and immersion-fixed tissue. Also, after copulation there was an increase in light cells that were characterized by reduced RER cisternae, an electron lucent cytoplasm, and atrophic Golgi apparatus. The luminal ground substance, secretion granules, and some Golgi elements, contained polysaccharides as seen with the periodic acid-thiocarbohydrazide-silver proteinate method.  相似文献

Summary With in vitro autoradiography, specific receptors for atrial natriuretic peptide (ANP) were localized in fetal rat kidney and adrenal glands. Receptors were present over renal vesicles, in the primitive renal medulla and throughout the adrenal gland as early as 16 days gestation. By 20 days gestation, several layers of developing renal corpuscles were present and ANP receptors were localized over developing glomeruli in each layer. Larger accumulations occurred over the juxtamedullary glomeruli. In the medulla, the receptors were localized in a reticular pattern near the pelivis. With emulsion coated sections, ANP receptors in developing renal corpuscles were seen primarily over the lower curve of S-shaped vesicles and around the periphery of the more mature corpuscles. In the renal medulla, receptors were localized over the interstitial cells. In the 16-day-old adrenal gland, ANP receptors were present throughout the cortical area but at 20 days gestation and 1 day postpartum receptors appeared more numerous in the peripheral region. these data suggest that ANP has important developmental effects in the kidney and adrenal gland and may be involved in regulation of body fluid homeostasis in the late gestation rat fetus.  相似文献
