Coastal salt marshes are sensitive to global climate change and may play an important role in mitigating global warming. To evaluate the impacts of Spartina alterniflora invasion on global warming potential (GWP) in Chinese coastal areas, we measured CH4 and N2O fluxes and soil organic carbon sequestration rates along a transect of coastal wetlands in Jiangsu province, China, including open water; bare tidal flat; and invasive S. alterniflora, native Suaeda salsa, and Phragmites australis marshes. Annual CH4 emissions were estimated as 2.81, 4.16, 4.88, 10.79, and 16.98 kg CH4 ha?1 for open water, bare tidal flat, and P. australis, S. salsa, and S. alterniflora marshes, respectively, indicating that S. alterniflora invasion increased CH4 emissions by 57–505%. In contrast, negative N2O fluxes were found to be significantly and negatively correlated (< 0.001) with net ecosystem CO2 exchange during the growing season in S. alterniflora and P. australis marshes. Annual N2O emissions were 0.24, 0.38, and 0.56 kg N2O ha?1 in open water, bare tidal flat and S. salsa marsh, respectively, compared with ‐0.51 kg N2O ha?1 for S. alterniflora marsh and ?0.25 kg N2O ha?1 for P. australis marsh. The carbon sequestration rate of S. alterniflora marsh amounted to 3.16 Mg C ha?1 yr?1 in the top 100 cm soil profile, a value that was 2.63‐ to 8.78‐fold higher than in native plant marshes. The estimated GWP was 1.78, ?0.60, ?4.09, and ?1.14 Mg CO2eq ha?1 yr?1 in open water, bare tidal flat, P. australis marsh and S. salsa marsh, respectively, but dropped to ?11.30 Mg CO2eq ha?1 yr?1 in S. alterniflora marsh. Our results indicate that although S. alterniflora invasion stimulates CH4 emissions, it can efficiently mitigate increases in atmospheric CO2 and N2O along the coast of China.  相似文献

Commercial cultivation of Spirulina sp. is highly popular due to the presence of high amount of C‐phycocyanin (C‐PC ) and other valuable chemicals like carotenoids and γ‐linolenic acid. In this study, the pH and the concentrations of nitrogen and carbon source were manipulated to achieve improved cell growth and C‐PC production in NaCl‐tolerant mutant of Spirulina platensis . In this study, highest C‐PC (147 mg · L?1) and biomass (2.83 g · L?1) production was achieved when a NaCl‐tolerant mutant of S. platensis was cultivated in a nitrate and bicarbonate sufficient medium (40 and 60 mM, respectively) at pH 9.0 under phototrophic conditions. Kinetic study of wildtype S. platensis and its NaCl‐tolerant mutant was also done to determine optimum nitrate concentrations for maximum growth and C‐PC production. Kinetic parameter of inhibition (Haldane model) was fitted to the relationship between specific growth rate and substrate concentration obtained from the growth curves. Results showed that the maximum specific growth rate (μmax) for NaCl‐tolerant mutant increased by 17.94% as compared to its wildtype counterpart, with a slight increase in half‐saturation constant (Ks), indicating that this strain could grow well at high concentration of NaNO3. C‐PC production rate (Cmax) in mutant cells increased by 12.2% at almost half the value of Ks as compared to its wildtype counterpart. Moreover, the inhibition constant (Ki) value was 207.85% higher in NaCl‐tolerant mutant as compared to its wildtype strain, suggesting its ability to produce C‐PC even at high concentrations of NaNO3.  相似文献

The greenhouse gas (GHG) balance of European grasslands (EU‐28 plus Norway and Switzerland), including CO2, CH4 and N2O, is estimated using the new process‐based biogeochemical model ORCHIDEE‐GM over the period 1961–2010. The model includes the following: (1) a mechanistic representation of the spatial distribution of management practice; (2) management intensity, going from intensively to extensively managed; (3) gridded simulation of the carbon balance at ecosystem and farm scale; and (4) gridded simulation of N2O and CH4 emissions by fertilized grassland soils and livestock. The external drivers of the model are changing animal numbers, nitrogen fertilization and deposition, land‐use change, and variable CO2 and climate. The carbon balance of European grassland (NBP) is estimated to be a net sink of 15 ± 7 g C m?2 year?1 during 1961–2010, equivalent to a 50‐year continental cumulative soil carbon sequestration of 1.0 ± 0.4 Pg C. At the farm scale, which includes both ecosystem CO2 fluxes and CO2 emissions from the digestion of harvested forage, the net C balance is roughly halved, down to a small sink, or nearly neutral flux of 8 g C m?2 year?1. Adding CH4 and N2O emissions to net ecosystem exchange to define the ecosystem‐scale GHG balance, we found that grasslands remain a net GHG sink of 19 ± 10 g C‐CO2 equiv. m?2 year?1, because the CO2 sink offsets N2O and grazing animal CH4 emissions. However, when considering the farm scale, the GHG balance (NGB) becomes a net GHG source of ?50 g C‐CO2 equiv. m?2 year?1. ORCHIDEE‐GM simulated an increase in European grassland NBP during the last five decades. This enhanced NBP reflects the combination of a positive trend of net primary production due to CO2, climate and nitrogen fertilization and the diminishing requirement for grass forage due to the Europe‐wide reduction in livestock numbers.  相似文献

Bioenergy crop cultivation on former peat extraction areas is a potential after‐use option that provides a source of renewable energy while mitigating climate change through enhanced carbon (C) sequestration. This study investigated the full C and greenhouse gas (GHG) balances of fertilized (RCG‐F) and nonfertilized (RCG‐C) reed canary grass (RCG; Phalaris arundinacea) cultivation compared to bare peat (BP) soil within an abandoned peat extraction area in western Estonia during a dry year. Vegetation sampling, static chamber and lysimeter measurements were carried out to estimate above‐ and belowground biomass production and allocation, fluxes of carbon dioxide (CO2), methane (CH4) and nitrous oxide (N2O) in cultivated strips and drainage ditches as well as the dissolved organic carbon (DOC) export, respectively. Heterotrophic respiration was determined from vegetation‐free trenched plots. Fertilization increased the above‐ to belowground biomass production ratio and the autotrophic to heterotrophic respiration ratio. The full C balance (incl. CO2, CH4 and DOC fluxes from strips and ditches) was 96, 215 and 180 g C m?2 yr?1 in RCG‐F, RCG‐C and BP, respectively, suggesting that all treatments acted as C sources during the dry year. The C balance was driven by variations in the net CO2 exchange, whereas the combined contribution of CH4 and DOC fluxes was <5%. The GHG balances were 3.6, 7.9 and 6.6 t CO2 eq ha?1 yr?1 in RCG‐F, RCG‐C and BP, respectively. The CO2 exchange was also the dominant component of the GHG balance, while the contributions of CH4 and N2O were <1% and 1–6%, respectively. Overall, this study suggests that maximizing plant growth and the associated CO2 uptake through adequate water and nutrient supply is a key prerequisite for ensuring sustainable high yields and climate benefits in RCG cultivations established on organic soils following drainage and peat extraction.  相似文献

Sclerotinia stem rot caused by Sclerotinia sclerotiorum is one of the most important diseases in oilseed rape‐growing areas of China. To determine the frequency of resistance of field isolates of S. sclerotiorum to carbendazim and dimethachlone, a total of 556 isolates from 10 different regions of Henan Province were obtained between 2015 and 2016. The frequency of isolates with a high‐resistance phenotype and a moderate‐resistance phenotype to carbendazim was 69.2% and 10.8%, respectively. However, S. sclerotiorum isolates resistant to dimethachlone were not detected. The baseline sensitivity of S. sclerotiorum to dimethachlone was distributed as a unimodal curve with a mean EC50 value of 0.39 ± 0.09 μg ml?1 for the inhibition of mycelial growth. Four dimethachlone‐resistant mutants were obtained from 20 wild‐type isolates induced by exposure to increasing concentrations of the fungicide in vitro. The mutants showed high levels of resistance to dimethachlone, with resistance factors that ranged from 179 to 323. Positive cross‐resistance occurred between dimethachlone and procymidone, iprodione, and fludioxonil; however, no cross‐resistance was observed for carbendazim and boscalid. The fitness of the dimethachlone‐resistant mutants was significantly lower than that of the wild‐type isolates, as measured by mycelial growth, hyphal dry weight, sclerotium number and dry weight, and pathogenicity. Additionally, based on osmotic tests, the inhibition of mycelial growth caused by NaCl applied at different concentrations was significantly higher for the dimethachlone‐resistant mutants than for their wild‐type parents.  相似文献

Recent morphology-based investigations of freshwater Arctic diatoms suggest that many species remain to be discovered, reflecting a unique polar flora. During a survey of the freshwater diatom flora of northern Billefjorden, including the Petuniabukta fjord region (Spitsbergen), several morphodemes belonging to the Achnanthidium minutissimum species complex were recorded. Molecular phylogenies based on rbcL, 28S and 18S sequences, including single cells from Canada and strains from Marion Island (sub-Antarctica) and GenBank revealed the presence of 12 distinct A. minutissimum complex lineages, of which three contained strains from Spitsbergen. One Arctic lineage is described as a new species. Achnanthidium digitatum sp. nov. is morphologically characterized by narrow, linear to only slightly lanceolate valves and usually two areolae per stria. The two remaining Arctic lineages are in need of a more complete morphological and molecular comparison with other representatives of the A. minutissimum complex to clarify their taxonomic identity. It is argued that implementation of molecular data in the taxonomy of Achnanthidium will be essential to solve the taxonomic problems associated with this group, eventually resulting in a better understanding of the biogeography and niche differentiation of different species belonging to the A. minutissimum complex. Ideally, this should be based on more variable genes than the currently widely used 18S, which does not have a species level resolution in the A. minutissimum complex.  相似文献

Wetland‐adapted trees are known to transport soil‐produced methane (CH4), an important greenhouse gas to the atmosphere, yet seasonal variations and controls on the magnitude of tree‐mediated CH4 emissions remain unknown for mature forests. We examined the spatial and temporal variability in stem CH4 emissions in situ and their controls in two wetland‐adapted tree species (Alnus glutinosa and Betula pubescens) located in a temperate forested wetland. Soil and herbaceous plant‐mediated CH4 emissions from hollows and hummocks also were measured, thus enabling an estimate of contributions from each pathway to total ecosystem flux. Stem CH4 emissions varied significantly between the two tree species, with Alnus glutinosa displaying minimal seasonal variations, while substantial seasonal variations were observed in Betula pubescens. Trees from each species emitted similar quantities of CH4 from their stems regardless of whether they were situated in hollows or hummocks. Soil temperature and pore‐water CH4 concentrations best explained annual variability in stem emissions, while wood‐specific density and pore‐water CH4 concentrations best accounted for between‐species variations in stem CH4 emission. Our study demonstrates that tree‐mediated CH4 emissions contribute up to 27% of seasonal ecosystem CH4 flux in temperate forested wetland, with the largest relative contributions occurring in spring and winter. Tree‐mediated CH4 emissions currently are not included in trace gas budgets of forested wetland. Further work is required to quantify and integrate this transport pathway into CH4 inventories and process‐based models.  相似文献

The goal of this study was to investigate whether chilling tolerance of C4 photosynthesis in Miscanthus can be transferred to sugarcane by hybridization. Net leaf CO2 uptake (Asat) and the maximum operating efficiency of photosystem II (ФPSII) were measured in warm conditions (25 °C/20 °C), and then during and following a chilling treatment of 10 °C/5 °C for 11 day in controlled environment chambers. Two of three hybrids (miscanes), ‘US 84‐1058’ and ‘US 87‐1019’, did not differ significantly from the chilling tolerant M. ×giganteus ‘Illinois’ (Mxg), for Asat, and ΦPSII measured during chilling. For Mxg grown at 10 °C/5 °C for 11 days, Asat was 4.4 μmol m?2 s?1, while for miscane ‘US 84‐1058’ and ‘US 87‐1019’, Asat was 5.7 and 3.5 μmol m?2 s?1, respectively. Miscanes ‘US 84‐1058’ and ‘US 87‐1019’ and Mxg had significantly higher rates of Asat during chilling than three tested sugarcanes. A third miscane showed lower rates than Mxg during chilling, but recovered to higher rates than sugarcane upon return to warm conditions. Chilling tolerance of ‘US 84‐1058’ was further confirmed under autumn field conditions in southern Illinois. The selected chilling tolerant miscanes have particular value for biomass feedstock and biofuel production and at the same time they can be a starting point for extending sugarcane's range to colder climates.  相似文献

Boreal peatlands contain approximately 500 Pg carbon (C) in the soil, emit globally significant quantities of methane (CH4), and are highly sensitive to climate change. Warming associated with global climate change is likely to increase the rate of the temperature‐sensitive processes that decompose stored organic carbon and release carbon dioxide (CO2) and CH4. Variation in the temperature sensitivity of CO2 and CH4 production and increased peat aerobicity due to enhanced growing‐season evapotranspiration may alter the nature of peatland trace gas emission. As CH4 is a powerful greenhouse gas with 34 times the warming potential of CO2, it is critical to understand how factors associated with global change will influence surface CO2 and CH4 fluxes. Here, we leverage the Spruce and Peatland Responses Under Changing Environments (SPRUCE) climate change manipulation experiment to understand the impact of a 0–9°C gradient in deep belowground warming (“Deep Peat Heat”, DPH) on peat surface CO2 and CH4 fluxes. We find that DPH treatments increased both CO2 and CH4 emission. Methane production was more sensitive to warming than CO2 production, decreasing the C‐CO2:C‐CH4 of the respired carbon. Methane production is dominated by hydrogenotrophic methanogenesis but deep peat warming increased the δ13C of CH4 suggesting an increasing contribution of acetoclastic methanogenesis to total CH4 production with warming. Although the total quantity of C emitted from the SPRUCE Bog as CH4 is <2%, CH4 represents >50% of seasonal C emissions in the highest‐warming treatments when adjusted for CO2 equivalents on a 100‐year timescale. These results suggest that warming in boreal regions may increase CH4 emissions from peatlands and result in a positive feedback to ongoing warming.  相似文献

Aliphatic C–H bonds are one of the major organic signatures detected in Proterozoic organic microfossils, and their origin is a topic of interest. To investigate the influence of the presence of silica on the thermal alteration of aliphatic C–H bonds in prokaryotic cells during diagenesis, cyanobacteria Synechocystis sp. PCC6803 were heated at temperatures of 250–450°C. Changes in the infrared (IR) signals were monitored by micro‐Fourier transform infrared (FTIR) spectroscopy. Micro‐FTIR shows that absorbances at 2,925 cm?1 band (aliphatic CH2) and 2,960 cm?1 band (aliphatic CH3) decrease during heating, indicating loss of the C–H bonds, which was delayed by the presence of silica. A theoretical approach using solid‐state kinetics indicates that the most probable process for the aliphatic C–H decrease is three‐dimensional diffusion of alteration products under both non‐embedded and silica‐embedded conditions. The extrapolation of the experimental results obtained at 250–450°C to lower temperatures implies that the rate constant for CH3 (kCH3) is similar to or lower than that for CH2 (kCH2; i.e., CH3 decreases at a similar rate or more slowly than CH2). The peak height ratio of 2,960 cm?1 band (CH3)/2,925 cm?1 band (CH2; R3/2 values) either increased or remained constant during the heating. These results reveal that the presence of silica does affect the decreasing rate of the aliphatic C–H bonds in cyanobacteria during thermal maturation, but that it does not significantly decrease the R3/2 values. Meanwhile, studies of microfossils suggest that the R3/2 values of Proterozoic prokaryotic fossils from the Bitter Springs Group and Gunflint Formation have decreased during fossilization, which is inconsistent with the prediction from our experimental results that R3/2 values did not decrease after silicification. Some process other than thermal degradation, possibly preservation of specific classes of biomolecules with low R3/2 values, might have occurred during fossilization.  相似文献

Periphyton dominated by the cellulose‐rich filamentous green alga Cladophora forms conspicuous growths along rocky marine and freshwater shorelines worldwide, providing habitat for diverse epibionts. Bacterial epibionts have been inferred to display diverse functions of biogeochemical significance: N‐fixation and other redox reactions, phosphorus accumulation, and organic degradation. Here, we report taxonomic diversity of eukaryotic and prokaryotic epibionts and diversity of genes associated with materials cycling in a Cladophora metagenome sampled from Lake Mendota, Dane Co., WI, USA, during the growing season of 2012. A total of 1,060 distinct 16S, 173 18S, and 351 28S rRNA operational taxonomic units, from which >220 genera or species of bacteria (~60), protists (~80), fungi (6), and microscopic metazoa (~80), were distinguished with the use of reference databases. We inferred the presence of several algal taxa generally associated with marine systems and detected Jaoa, a freshwater periphytic ulvophyte previously thought endemic to China. We identified six distinct nifH gene sequences marking nitrogen fixation, >25 bacterial and eukaryotic cellulases relevant to sedimentary C‐cycling and technological applications, and genes encoding enzymes in aerobic and anaerobic pathways for vitamin B12 biosynthesis. These results emphasize the importance of Cladophora in providing habitat for microscopic metazoa, fungi, protists, and bacteria that are often inconspicuous, yet play important roles in ecosystem biogeochemistry.  相似文献

High‐temperature tolerance in plants is important in a warming world, with extreme heat waves predicted to increase in frequency and duration, potentially leading to lethal heating of leaves. Global patterns of high‐temperature tolerance are documented in animals, but generally not in plants, limiting our ability to assess risks associated with climate warming. To assess whether there are global patterns in high‐temperature tolerance of leaf metabolism, we quantified Tcrit (high temperature where minimal chlorophyll a fluorescence rises rapidly and thus photosystem II is disrupted) and Tmax (temperature where leaf respiration in darkness is maximal, beyond which respiratory function rapidly declines) in upper canopy leaves of 218 plant species spanning seven biomes. Mean site‐based Tcrit values ranged from 41.5 °C in the Alaskan arctic to 50.8 °C in lowland tropical rainforests of Peruvian Amazon. For Tmax, the equivalent values were 51.0 and 60.6 °C in the Arctic and Amazon, respectively. Tcrit and Tmax followed similar biogeographic patterns, increasing linearly (?8 °C) from polar to equatorial regions. Such increases in high‐temperature tolerance are much less than expected based on the 20 °C span in high‐temperature extremes across the globe. Moreover, with only modest high‐temperature tolerance despite high summer temperature extremes, species in mid‐latitude (~20–50°) regions have the narrowest thermal safety margins in upper canopy leaves; these regions are at the greatest risk of damage due to extreme heat‐wave events, especially under conditions when leaf temperatures are further elevated by a lack of transpirational cooling. Using predicted heat‐wave events for 2050 and accounting for possible thermal acclimation of Tcrit and Tmax, we also found that these safety margins could shrink in a warmer world, as rising temperatures are likely to exceed thermal tolerance limits. Thus, increasing numbers of species in many biomes may be at risk as heat‐wave events become more severe with climate change.  相似文献

Lakes are a major component of boreal landscapes, and whereas lake CO2 emissions are recognized as a major component of regional C budgets, there is still much uncertainty associated to lake CH4 fluxes. Here, we present a large‐scale study of the magnitude and regulation of boreal lake summer diffusive CH4 fluxes, and their contribution to total lake carbon (C) emissions, based on in situ measurements of concentration and fluxes of CH4 and CO2 in 224 lakes across a wide range of lake type and environmental gradients in Québec. The diffusive CH4 flux was highly variable (mean 11.6 ± 26.4 SD mg m?2 d?1), and it was positively correlated with temperature and lake nutrient status, and negatively correlated with lake area and colored dissolved organic matter (CDOM). The relationship between CH4 and CO2 concentrations fluxes was weak, suggesting major differences in their respective sources and/or regulation. For example, increasing water temperature leads to higher CH4 flux but does not significantly affect CO2 flux, whereas increasing CDOM concentration leads to higher CO2 flux but lower CH4 flux. CH4 contributed to 8 ± 23% to the total lake C emissions (CH4 + CO2), but 18 ± 25% to the total flux in terms of atmospheric warming potential, expressed as CO2‐equivalents. The incorporation of ebullition and plant‐mediated CH4 fluxes would further increase the importance of lake CH4. The average Q10 of CH4 flux was 3.7, once other covarying factors were accounted for, but this apparent Q10 varied with lake morphometry and was higher for shallow lakes. We conclude that global climate change and the resulting shifts in temperature will strongly influence lake CH4 fluxes across the boreal biome, but these climate effects may be altered by regional patterns in lake morphometry, nutrient status, and browning.  相似文献

Controls on the fate of ~277 Pg of soil organic carbon (C) stored in permafrost peatland soils remain poorly understood despite the potential for a significant positive feedback to climate change. Our objective was to quantify the temperature, moisture, organic matter, and microbial controls on soil organic carbon (SOC) losses following permafrost thaw in peat soils across Alaska. We compared the carbon dioxide (CO2) and methane (CH4) emissions from peat samples collected at active layer and permafrost depths when incubated aerobically and anaerobically at ?5, ?0.5, +4, and +20 °C. Temperature had a strong, positive effect on C emissions; global warming potential (GWP) was >3× larger at 20 °C than at 4 °C. Anaerobic conditions significantly reduced CO2 emissions and GWP by 47% at 20 °C but did not have a significant effect at ?0.5 °C. Net anaerobic CH4 production over 30 days was 7.1 ± 2.8 μg CH4‐C gC?1 at 20 °C. Cumulative CO2 emissions were related to organic matter chemistry and best predicted by the relative abundance of polysaccharides and proteins (R2 = 0.81) in SOC. Carbon emissions (CO2‐C + CH4‐C) from the active layer depth peat ranged from 77% larger to not significantly different than permafrost depths and varied depending on the peat type and peat decomposition stage rather than thermal state. Potential SOC losses with warming depend not only on the magnitude of temperature increase and hydrology but also organic matter quality, permafrost history, and vegetation dynamics, which will ultimately determine net radiative forcing due to permafrost thaw.  相似文献

The present paper describes environmental and seasonal‐related chemical composition variations, vasorelaxant and angiotensin I‐converting enzyme (ACE) activities of essential oil from aerial parts of Seseli pallasii Besser . The composition was analyzed by GC and GC/MS. Monoterpenes were found to be the most abundant chemical class with α ‐pinene (42.7 – 48.2%) as the most prevalent component. Seseli pallasi essential oil relaxed isolated endothelium‐intact mesenteric arteries rings precontracted with phenylephrine with IC 50 = 3.10 nl/ml (IC 50 = 2.70 μg/ml). Also, S. pallasii essential oil was found to exhibit a dose‐dependent ACE inhibitory activity with an IC 50 value of 0.33 mg/ml. In silico evaluation of ACE inhibitory activity of the individual components showed that spathulenol exhibited the best binding affinity with ACE, and the lowest binding energy of ?7.5 kcal/mol. The results suggested that combination of vasorelaxing and ACE inhibitory effects of the analyzed S. pallasii essential oil might have the potential therapeutic significance in hypertension.  相似文献

As a controversial strategy to mitigate global warming, biochar application into soil highlights the need for life cycle assessment before large‐scale practice. This study focused on the effect of biochar on carbon footprint of rice production. A field experiment was performed with three treatments: no residue amendment (Control), 6 t ha?1 yr?1 corn straw (CS) amendment, and 2.4 t ha?1 yr?1 corn straw‐derived biochar amendment (CBC). Carbon footprint was calculated by considering carbon source processes (pyrolysis energy cost, fertilizer and pesticide input, farmwork, and soil greenhouse gas emissions) and carbon sink processes (soil carbon increment and energy offset from pyrolytic gas). On average over three consecutive rice‐growing cycles from year 2011 to 2013, the CS treatment had a much higher carbon intensity of rice (0.68 kg CO2‐C equivalent (CO2‐Ce) kg?1 grain) than that of Control (0.24 kg CO2‐Ckg?1 grain), resulting from large soil CH4 emissions. Biochar amendment significantly increased soil carbon pool and showed no significant effect on soil total N2O and CH4 emissions relative to Control; however, due to a variation in net electric energy input of biochar production based on different pyrolysis settings, carbon intensity of rice under CBC treatment ranged from 0.04 to 0.44 kg CO2‐Ckg?1 grain. The results indicated that biochar strategy had the potential to significantly reduce the carbon footprint of crop production, but the energy‐efficient pyrolysis technique does matter.  相似文献

The gene AAE3 was recently identified to encode an oxalyl‐CoA synthetase (OCS) in Arabidopsis that catalyses the conversion of oxalate and CoA into oxalyl‐CoA. It will be particularly important to characterise the homologous gene in rice since rice is not only a monocotyledonous model plant, but also a staple food crop.
  • Various enzymatic and biological methods have been used to characterise the homologous gene.
  • We first defined that AAE3 in the rice genome (OsAAE3) also encodes an OCS enzyme. Its Km for oxalate is 1.73 ± 0.12 mm , and Vm is 6824.9 ± 410.29 U·min?1·mg protein?1. Chemical modification and site‐directed mutagenesis analyses identified thiols as the active site residues for rice OCS catalysis, suggesting that the enzyme might be regulated by redox state. Subcellular localisation assay showed that the enzyme is located in the cytosol and predominantly distributed in leaf epidermal cells. As expected, oxalate levels increased when OCS was suppressed in RNAi transgenic plants. More interestingly, OCS‐suppressed plants were more susceptible to bacterial blight but more resistant to Al toxicity.
  • The results demonstrate that the OsAAE3‐encoded protein also acts as an OCS in rice, and may play different roles in coping with stresses. These molecular, enzymatic and functional data provide first‐hand information to further clarify the function and mechanism of OCS in rice plants.

Arctic lakes are a significant source of the greenhouse gas methane (CH4), but the role that methane oxidizing bacteria (methanotrophs) play in limiting the overall CH4 flux is poorly understood. Here, we used stable isotope probing (SIP) techniques to identify the metabolically active aerobic methanotrophs in upper sediments (0–1 cm) from an arctic lake in northern Alaska sampled during ice‐free summer conditions. The highest CH4 oxidation potential was observed in the upper sediment (0–1 cm depth) with 1.59 µmol g wet weight?1 day?1 compared with the deeper sediment samples (1–3 cm, 3–5 cm and 5–10 cm), which exhibited CH4 oxidation potentials below 0.4 µmol g wet weight?1 day?1. Both type I and type II methanotrophs were directly detected in the upper sediment total communities using targeted primer sets based on 16S rRNA genes. Sequencing of 16S rRNA genes and functional genes (pmoA and mxaF) in the 13C‐DNA from the upper sediment indicated that type I methanotrophs, mainly Methylobacter, Methylosoma, Methylomonas and Methylovulum miyakonense, dominated the assimilation of CH4. Methylotrophs, including the genera Methylophilus and/or Methylotenera, were also abundant in the 13C‐DNA. Our results show that a diverse microbial consortium acquired carbon from CH4 in the sediments of this arctic lake.  相似文献
