The polymorphous trichome apices of Oscillatoria redekei are described with respect to their shape and size. Breakage of trichomes may occur either trans- or intercellularly. The development and localization of intercellular trichome breakage were observed. Transmission electron microscopy shows that the fine structure of cell walls resembles that of Pseudanabaena galeata. Longitudinal walls consist of three or four layers and lack a sheath, while the cross walls are composed of three layers with a dominating L2-layer. In contrast to P. galeata, the centroplasm and most of the thylakoidal region of adjacent cells of O. redekei are connected by the cross walls forming obtuse angles at the site of the longitudinal walls. The main characteristics of O. redekei and of Pseudanabaena species are discussed.  相似文献

Trichomes originate from the epidermal cells of nearly all terrestrial plants, which are specialized unicellular or multicellular structures. Although the molecular mechanism regulating unicellular trichome formation has been extensively characterized, most of the genes essential for multicellular trichome formation remain unknown. In this study, we identified an associated locus on the long arm of chromosome 10 using a genome‐wide association study (GWAS) on type‐I trichomes of 180 diverse Solanum lycopersicum (tomato) accessions. Using map‐based cloning we then cloned the key gene controlling the initiation of this type of trichome, named Hair (H), which encodes a single C2H2 zinc‐finger protein. Transgenic experiments showed that hair‐absent phenotype is caused by the deletion of the entire coding region of H. We identified three alleles of H containing several missense mutations and a nucleotide deletion, which result in amino acid substitutions and a reading frame shift, respectively. In addition, knockdown of H or Woolly (Wo) represses the formation of type‐I trichomes, suggesting that both regulators may function as a heterodimer. Direct protein–protein interaction between them was further detected through pull‐down and yeast two‐hybrid assays. In addition, ectopic expression of H in Nicotiana tabacum (tobacco) and expression of its homologs from Capsicum annuum (pepper) and tobacco in tomato can trigger trichome formation. Taken together, these findings suggest that the H gene may be functionally conserved in multicellular trichome formation in Solanaceae species.  相似文献

Trichomes are storage compartments for specialized metabolites in many plant species. In trichome, plant primary metabolism is significantly changed, providing substrates for downstream secondary metabolism. However, little is known of how plants coordinate trichome formation and primary metabolism regulation. In this report, tomato (Solanum lycopersicum) trichome regulator SlMIXTA‐like is indicated as a metabolic regulation gene by mGWAS analysis. Overexpression of SlMIXTA‐like in tomato fruit enhances trichome formation. In addition, SlMIXTA‐like can directly bind to the promoter region of gene encoding 3‐deoxy‐7‐phosphoheptulonate synthase (SlDAHPS) to activate its expression. Induction of SlDAHPS expression enhances shikimate pathway activities and provides substrates for downstream secondary metabolism. Our data provide direct evidence that trichome regulator can directly manipulate primary metabolism, in which way plants can coordinate metabolic regulation and the formation of storage compartments for specialized metabolites. The newly identified SlMIXTA‐like can be used for future metabolic engineering.  相似文献

Summary Trichomes of Microcoleus vaginatus, a motile blue-green alga of the family Oscillatoriaceae, were studied by light and electron microscopy in an effort to determine the sites of trichome breakage during production of hormogonia.According to the evidence presented herein, transcellular breakage of trichomes is the only mechanism of hormogonium production in M. vaginatus. Tearing of the murein sacculus appears to be necessary and sufficient for transcellular breakage to ensue. As Fuhs and earlier investigators have correctly claimed, this process always involves the death of the cell whose wall is torn.When trichomes of M. vaginatus break across cells to produce hormogonia, the murein sacculus usually tears along a circumferential set of junctional pores. This particular mechanism of trichome breakage is not universal among members of the family Oscillatoriaceae.This report is based on a thesis submitted in partial fulfillment of the requirements for a Ph. The collagen protein family is diverse and its membership is continually expanding as new collagen‐like molecules are identified. Identification of collagen in unicellular eukaryotes and prokaryotes has opened discussion on the function of these collagens and their role in the emergence of multicellularity. The previous identification of a collagen gene in Trichodesmium erythraeum raises the question of function of this structural protein in a prokaryote. In this study, we show that this gene is expressed during all phases of growth, indicating that it may be required for all phases of growth. Using immunofluorescence techniques, we demonstrate that the collagen‐like protein is localized in a specific manner between adjacent cells along the trichome of T. erythraeum. Trichomes treated with the enzyme collagenase exhibited fragmentation, supporting our immunofluorescence localization data that this collagen‐like protein is found between adjacent cells. Our data strongly suggest that the collagen‐like protein found in T. erythraeum functions to maintain the structural integrity of the trichome through the adhesion of adjacent cells.  相似文献

The leaves of 52 species of Lithocarpus in China were studied. The adaxial leaf epidermis was investigated by light microscopy. Epidermal cells of the adaxial surface were classified into three types on the basis of the outline of their anticlinal walls, i.e. sinuate, straight and curved. The abaxial leaf epidermis was investigated by light microscopy and scanning electron microscopy. The following types of trichome were observed: appressed parallel tuft, stellate, fused stellate, papillae, stipitate fasciculate, solitary unicellular, appressed laterally attached unicellular, curly thin‐walled unicellular, bulbous and thin‐walled peltate. The fused stellate, appressed laterally attached unicellular and curly thin‐walled unicellular trichomes were reported in Lithocarpus for the first time. The appressed parallel tuft trichome, which is recognized as a salient characteristic of Lithocarpus, was not found in 15 species. A cladistic analysis was performed on the basis of the leaf epidermal features. According to the leaf epidermal features and several morphological characteristics, 26 of the 52 species could be divided into seven groups. Similar groups can be found in Barnett's and Camus' systems. The trichomes of four genera in Fagaceae are listed and compared. Lithocarpus had 14 types of trichome, 11 of which were identical to types found in Quercus, more than in Castanopsis and Cyclobalanopsis. In the near future, grasses must provide most of the biomass for the production of renewable fuels. However, grass cell walls are characterized by a large quantity of hydroxycinnamic acids such as ferulic and p‐coumaric acids, which are thought to reduce the biomass saccharification. Ferulic acid (FA) binds to lignin, polysaccharides and structural proteins of grass cell walls cross‐linking these components. A controlled reduction of FA level or of FA cross‐linkages in plants of industrial interest can improve the production of cellulosic ethanol. Here, we review the biosynthesis and roles of FA in cell wall architecture and in grass biomass recalcitrance to enzyme hydrolysis.  相似文献

Chlorella microalgae are increasingly used for various purposes such as fatty acid production, wastewater processing, or as health‐promoting food supplements. A mass spectrometry‐based survey of N‐glycan structures of strain collection specimens and 80 commercial Chlorella products revealed a hitherto unseen intragenus diversity of N‐glycan structures. Differing numbers of methyl groups, pentoses, deoxyhexoses, and N‐acetylglucosamine culminated in c. 100 different glycan masses. Thirteen clearly discernible glycan‐type groups were identified. Unexpected features included the occurrence of arabinose, of different and rare types of monosaccharide methylation (e.g. 4‐O‐methyl‐N‐acetylglucosamine), and substitution of the second N‐acetylglucosamine. Analysis of barcode ITS1–5.8S–ITS2 rDNA sequences established a phylogenetic tree that essentially went hand in hand with the grouping obtained by glycan patterns. This brief prelude to microalgal N‐glycans revealed a fabulous wealth of undescribed structural features that finely differentiated Chlorella‐like microalgae, which are notoriously poor in morphological attributes. In light of the almost identical N‐glycan structural features that exist within vertebrates or land plants, the herein discovered diversity is astonishing and argues for a selection pressure only explicable by a fundamental functional role of these glycans.  相似文献

The photophobic responses in the Cyanobacterium Anabaena variabilis which belongs to the Nostocaceae have been studied with aid of a population method as well as by single trichome observations. In white light experiments both step-up and step-down photophobic responses were observed. The wavelength dependence was examined at a constant fluence rate. The photophobically active light is absorbed by the photosynthetic pigments, mainly by the phycobiliproteins and chlorohyll a. Above 690 nm only negative reactions were observed, i.e. the trichomes left the light trap. In white light experiments DCMU strongly inhibited the photophobic responses, whereas photokinesis was not affected to the same extent indicating that the reaction is coupled with the non cyclic photosynthetic electron transport. DBMIB impaired the photophobic behaviour only slightly. It seems that the photophobic responses of A. variabilis are controlled by a similar mechanism as in Phormidium uncinatum (Oscillatoriaceae) although the two families and, hence, the two species differ in their movement mechanism as well as in their photoactic behaviour.  相似文献

Thick glistening cell walls occur in sieve tubes of all major land plant taxa. Historically, these ‘nacreous walls’ have been considered a diagnostic feature of sieve elements; they represent a conundrum, though, in the context of the widely accepted pressure–flow theory as they severely constrict sieve tubes. We employed the cucurbit Gerrardanthus macrorhizus as a model to study nacreous walls in sieve elements by standard and in situ confocal microscopy and electron microscopy, focusing on changes in functional sieve tubes that occur when prepared for microscopic observation. Over 90% of sieve elements in tissue sections processed for microscopy by standard methods exhibit nacreous walls. Sieve elements in whole, live plants that were actively transporting as shown by phloem‐mobile tracers, lacked nacreous walls and exhibited open lumina of circular cross‐sections instead, an appropriate structure for Münch‐type mass flow of the cell contents. Puncturing of transporting sieve elements with micropipettes triggered the rapid (<1 min) development of nacreous walls that occluded the cell lumen almost completely. We conclude that nacreous walls are preparation artefacts rather than structural features of transporting sieve elements. Nacreous walls in land plants resemble the reversibly swellable walls found in various algae, suggesting that they may function in turgor buffering, the amelioration of osmotic stress, wounding‐induced sieve tube occlusion, and possibly local defence responses of the phloem.  相似文献

Museum collections are essential for reconstructing and understanding past biodiversity. Many museum specimens are, however, challenging to identify. Museum samples may be incomplete, have an unusual morphology, or represent juvenile individuals, all of which complicate accurate identification. In some cases, inaccurate identification can lead to false biogeographic reconstructions with cascading impacts on paleontological and paleoecological research. Here, we analyzed an unusual Equid mandible found in the Far North of the Taymyr peninsula that was identified morphologically as Equus hemionus, an ancestor of present‐day Asiatic wild asses. If correct, this identification represents the only finding of a putative Late Pleistocene hemione in the Arctic region, and is therefore critical to understanding wild ass evolution and paleoecology. To confirm the accuracy of this specimen's taxonomic assignment, we used ancient DNA and mitochondrial hybridization capture to identify and place this specimen in the larger equid phylogeny. We find that the specimen is actually a member of E. caballus, the ancestor of domestic horses. Our study demonstrates the utility of ancient DNA to validate morphological identification, in particular of incomplete, otherwise problematic, or taxonomically unusual museum specimens.  相似文献

The silverleaf whitefly (SLW), Bemisia tabaci biotype B, is considered one of the most serious pests in cotton worldwide. Its control is based on the use of pesticides, which are well‐known for their harmful effects on non‐target organisms and the environment. Here, we examined resistance of several cotton genotypes in terms of antixenosis and antibiosis against SLW. We also investigated the mechanisms of cotton genotypes involved in antixenosis against SLW by correlating with leaf trichome density and leaf colour parameters. In choice tests, we selected resistant cotton genotypes, Gossypium hirsutum palmeri, Aubum 56‐7, G. hirsutum morrilli, Mocó, Nu‐16 (B2 B3 B6)‐ 78/658, Paymaster 53‐816 and ‘IAC 23’, based on SLW oviposition and host preference. No‐choice tests with these genotypes confirmed antixenosis effect on SLW; however, no antibiosis resistance was detected in terms of developmental period and adult emergence rate. We found SLW oviposition preference was positively correlated with trichome density in the abaxial leaf surface, while adult host preference was mostly explained by high levels of green intensity of adaxial surface. Nevertheless, both SLW oviposition and host preference were negatively correlated with the lightness level of adaxial leaf surface. Probably, the underlying mechanism of cotton resistance is based on more than just these two parameters, but our study will contribute to selecting resistant cotton genotypes and helps to understand the mechanisms involved in cotton resistance against SLW. To our best knowledge, this is the first work showing that cotton leaf colour parameters are related to resistance against SLW.  相似文献

Exceptionally preserved specimens of Ernietta in a shallow‐marine gutter cast from southern Namibia reveal that all previously figured specimens of this iconic Ediacaran megafossil are incomplete, representing only the base of a larger and more complex organism. The complete organism is interpreted as comprising a buried, sand‐filled anchor exhibiting the classical Ernietta morphology that passes distally into a trunk that is crowned by two facing fans that extended into the overlying water column. All parts of Ernietta, from the base of its buried anchor to the tip of its fans, appear to have been composed of a palisade of tubular elements that have been variably preserved. Similarity of tubule morphology despite the inherent difference in function between these constructions supports the view that these tubes were integral to all anatomical parts and functions of Ernietta. This style of architecture, construction and function is unique to the Erniettomorpha, supporting the view that it represents an extinct Ediacaran clade in the early evolution of multi‐cellular life.  相似文献

Morphological and habitat features have been recorded on punchcards for three genera in the Oscillatoriaceae and two in the Nostocaceae. As a result it is concluded that generic divisions in the Oscillatoriaceae are arbitrary and artificial, but that in the Nostocaceae, Cylindrospermum represents a grouping of distinct forms to a greater degree than does the amorphous genus Anabaena.

Some morphological comparisons between the two groups are attempted and detectable associations between morphological and habitat features are suggested : together these may contribute to an eventual separation of taxa on a more logical basis than at present.  相似文献
