Cell proliferation was examined during early embryogenesis of the newt ( Triturus pyrrhogaster ) by various methods. After the two-cell stage, at 23°C, the blastomere (cell) number per whole embryo increased logarithmically until the mid-blastula stage (for about 19 hr) and the rate of increase slowed down in and after the late blastula stage. On the other hand, the synchronous cleavage of the blastomeres at the animal pole continued for 18 hr until the twelfth cleavage (mid-blastula) and the transition from synchronous to asynchronous division occurred abruptly at and after the thirteenth cell division (late blastula). The study also showed that the presumptive neuro-ectoderm consisted mainly of cells of the fifteenth generation (G-15) at the onset of gastrulation (pigment stage).
The present study suggested that the number of ectodermal cells of the early gastrula (stage 12a) nearly doubled during gastrulation at the presumptive neuro-ectoderm. This means that most of the ectodermal cells are in G-16 at the end of gastrulation. On the other hand, both mitotic activity and the rate of cell increase gradually diminished during gastrulation in the ectoderms of both the presumptive neural and epidermal regions, and there are evidently significant differences in both activities between the neuro-ectoderm and the epidermal ectoderm after stage 13b: the epidermal ectoderm showed greater decrease in the rate of both mitotic activity and cell proliferation than the neuro-ectoderm.
These facts suggested that, whether the ectodermal cells will differentiate into neural cells or epidermal cells is determined during G-15 or G-16 in normal primary induction.  相似文献

Summary A polyclonal antibody (SP-2) has been produced, which recognizes antigens expressed in epidermal cells of Pleurodeles waltlii embryos. The antigens appear first at the end of gastrulation in the external surface of the embryo and are selectively expressed in ectodermally derived epidermal structures. Ectodermal commitment was investigated using cell cultures and blastocoel graft experiments. The four animal blastomeres of the 8-cell stage as well as the animal cap explants of the early gastrula stage cultured in vitro differentiate into epidermis, and SP-2 antigens are expressed. The expression of SP-2-defined antigens is inhibited both in vivo and in vitro by the inductive interaction of chordomesoderm. Once dissociated, ectodermal cells do not react with SP-2. Conversely, the aggregation of ectodermal cells may restore the expression of SP-2 antigens. Transplantation of animal cap explants or isolated ectodermal cells into the blastocoel of a host embryo at the early gastrula stage shows that only cells integrated into the epidermis express the marker antigens. When vegetal cells were dissociated from donor embryos before the mid-blastula stage and implanted into the blastocoel of host embryos at the early gastrula stage, their progeny were found in all germ layers, cells that were found in the host epidermis were stained with SP-2, whereas those contributing to mesoderm and endoderm were not. Thus the acquisition of cell polarity in epidermal differentiation and the organization of cells into epithelial structures are essential for SP-2-defined antigen expression.  相似文献

The tetraspanin family of four-pass transmembrane proteins has been implicated in fundamental biological processes, including cell adhesion, migration, and proliferation. Tetraspanins interact with various transmembrane proteins, establishing a network of large multimolecular complexes that allows specific lateral secondary interactions. Here we report the identification and functional characterization of Xenopus Tetraspanin-1 (xTspan-1). At gastrula and neurula, xTspan-1 is expressed in the dorsal ectoderm and neural plate, respectively, and in the hatching gland, cement gland, and posterior neural tube at tailbud stages. The expression of xTspan-1 in the early embryo is negatively regulated by bone morphogenetic protein (BMP) and stimulated by Notch signals. Microinjection of xTspan-1 mRNA interfered with gastrulation movements and reduced ectodermal cell adhesion in a cadherin-dependent manner. Morpholino knock-down of endogenous xTspan-1 protein revealed a requirement of xTspan-1 for gastrulation movements and primary neurogenesis. Our data suggest that xTspan-1 could act as a molecular link between BMP signalling and the regulation of cellular interactions that are required for gastrulation movements and neural differentiation in the early Xenopus embryo.  相似文献

Two-dimensional gel electrophoresis was used to analyze protein synthesis in relation to neural and epidermal differentiation in Cynops pyrrhogaster embryo. Various regions of embryos at different developmental stages, from late morula to early neurula stages, were excised, radiolabelled with 35S-methionine, and the pattern of protein synthesis were compared. The following four types of protein spots were observed: (1) six proteins synthesized characteristically in the epidermal region of the embryo after gastrulation, (2) two proteins synthesized in both epidermal and endodermal regions, but not in other regions, after gastrulation, (3) a protein first detected at early blastula stage, of which expression was nearly constant in presumptive epidermis region but declined in the other regions, (4) the candidate for neural plate specific protein synthesized at a very high level in ectoderm explants treated with concanavalin A, a substance which evokes neural induction.  相似文献

Inductive interactions between different cell layers have an extremely important role in early embryogenesis. One of the most intensively studied and best characterised of these is the induction of neural tissue from ectodermal cells by the dorsal mesoderm. The competence of ectodermal cells to respond to neural induction varies according to dorsal-ventral position; with dorsal ectoderm (much of which forms the neural plate) having a far higher competence. Here we show that overexpression of the nucleotide exchange factor lfc increases ectodermal competence for neural induction as well as the amount of neural tissue in the whole embryo. Lfc is expressed pan ectodermally soon after gastrulation and may respond to an early determinant of dorsal ectoderm.  相似文献

The cement gland is an ectodermal organ in the head of frog embryos, lying anterior to any neural tissue. As analyzed by specific RNA expression, cement gland, like neural tissue, was induced by the dorsal mesoderm. Interestingly, mesoderm with the highest cement gland-inducing potential lay posterior to the ectoderm fated to form this organ, indicating that its induction occurred at a distance from the inducer source. Cement gland induction first occurred during early gastrulation. However, most initially induced cells did not contribute to the mature cement gland, but instead formed part of the neural plate. This change in fate could be reconstituted in vitro. These results suggest that determination of part of the anteroposterior axis occurs progressively, where future neural ectoderm is first induced to a cement glandlike state. As gastrulation proceeds, further induction by mesoderm may override this state, which persists only in the extreme anterior of the embryo.  相似文献

In vertebrates, neural induction occurs during gastrulation when ectodermal cells choose between two fates, neural and epidermal. In Xenopus, neural induction has been regarded as a default pathway as it occurs, in dorsal ectoderm, when ventralizing signals (mainly Bone Morphogenesis Proteins, BMPs, potent epidermal inducers) are inhibited by dorsalizing signals, including factors such as noggin, chordin, and follistatin. However, our previous studies demonstrated that an instructive signal triggered by the activation of L-type voltage-sensitive calcium channels, resulting in a transient increase in intracellular free calcium, appears to be a necessary and sufficient requirement to induce the competent ectoderm toward the neural pathway. Here we further explore the relationship between the Ca2+ transient signals observed and the expression of early neural genes. We have performed a subtractive approach to identify the genes which are transcribed early after the calcium signal and involved in neural determination. We have analyzed a candidate gene (xMLP) which encodes a MARCKS-like protein, a substrate for PKC. We show that this gene is activated by a calcium transient signals and induced by noggin overexpression. xMLP is expressed at the right time in presumptive neural territories. The putative role of xMLP in the process of neural induction is discussed.  相似文献

In an attempt to define the pattern of developmental expression of AP-2rep and AP-2 in Xenopus embryos, we cloned a Xenopus AP-2rep cDNA. The AP-2rep message was localized in the organizer region at the gastrula stage whereas AP-2 was expressed ventro-laterally in the animal hemisphere. Later, AP-2rep was expressed in the entire neural tissue at the neurula stage while AP-2 was predominantly expressed in the cranial neural crest areas. The endogenous expression of AP-2 in the neural crest area was diminished by ectopic injection of AP-2rep RNA, suggesting a role for AP-2rep in the differentiation of neural tissues by restricting the expression of AP-2 in the Xenopus embryo.  相似文献

We report on the temporal and spatial expression pattern of two novel genes of the Xenopus fork head/winged helix family, xFoxB2 and xFoxI1c. xFoxB2 is activated at the late blastula stage and first expressed within the dorsolateral ectoderm except for the organiser territory. During gastrulation, xFoxB2 is found in two ectodermal stripes adjacent to the dorsal midline. Expression is completely down-regulated during neurulation. However, two distinct sets of cells expressing xFoxB2 re-appear in the rhombencephalon of swimming tadpoles. xFoxI1c is initially expressed at the early neurula stage in an epidermal ring around the neural field. Subsequent expression is found to be increased, and is exclusively localised to placodal precursor cells. The placodal expression remains until stage 40, when it is restricted to a distinct region in the lateral body wall behind the gills.  相似文献
