Chitin, a homopolymer of β-1-4-linked N-acetylglucosamine synthesized by chitin synthase A (Chs-A), is organized in the procuticle of the postembryonic cuticle or exoskeleton, which is composed of laminae stacked parallel to the cell surface to give stability and integrity to the underlying insect epidermal and other tissues. Our previous work has revealed an important role for two proteins from Tribolium castaneum named Knickkopf (TcKnk) and Retroactive (TcRtv) in postembryonic cuticular chitin maintenance. TcKnk and TcRtv were shown to be required for protection and organization of newly synthesized procuticular chitin. To study the functions of TcKnk and TcRtv in serosal and larval cuticles produced during embryogenesis in T. castaneum, dsRNAs specific for these two genes were injected into two week-old adult females. The effects of dsRNA treatment on ovarial integrity, oviposition, egg hatching and adult survival were determined. Insects treated with dsRNA for chitin synthase-A (TcChs-A) and tryptophan oxygenase (TcVer) were used as positive and negative controls for these experiments, respectively. Like TcChs-A RNAi, injection of dsRNA for TcKnk or TcRtv into adult females exhibited no adult lethality and oviposition was normal. However, a vast majority of the embryos did not hatch. The remaining (∼10%) of the embryos hatched into first instar larvae that died without molting to the second instar. Chitin content analysis following TcKnk and TcRtv parental RNAi revealed approximately 50% reduction in chitin content of eggs in comparison with control TcVer RNAi, whereas TcChs-A dsRNA-treatment led to >90% loss of chitin. Furthermore, transmission electron microscopic (TEM) analysis of serosal cuticle from TcChs-A, TcKnk and TcRtv dsRNA-treated insects revealed a complete absence of laminar organization of serosal (and larval) procuticle in comparison with TcVer dsRNA-treated controls, which exhibited normal laminar organization of procuticular chitin. The results of this study demonstrate that in addition to their essential roles in maintenance and organization of chitin in epidermal cuticle in larval and later stages of insect development, TcKnk and TcRtv also are required for egg hatch, chitin maintenance and laminar organization of both serosal and larval cuticle during embryonic development of T. castaneum.  相似文献

Precise epithelial tube diameters rely on coordinated cell shape changes and apical membrane enlargement during tube growth. Uniform tube expansion in the developing Drosophila trachea requires the assembly of a transient intraluminal chitin matrix, where chitin forms a broad cable that expands in accordance with lumen diameter growth. Like the chitinous procuticle, the tracheal luminal chitin cable displays a filamentous structure that presumably is important for matrix function. Here, we show that knickkopf (knk) and retroactive (rtv) are two new tube expansion mutants that fail to form filamentous chitin structures, both in the tracheal and cuticular chitin matrices. Mutations in knk and rtv are known to disrupt the embryonic cuticle, and our combined genetic analysis and chemical chitin inhibition experiments support the argument that Knk and Rtv specifically assist in chitin function. We show that Knk is an apical GPI-linked protein that acts at the plasma membrane. Subcellular mislocalization of Knk in previously identified tube expansion mutants that disrupt septate junction (SJ) proteins, further suggest that SJs promote chitinous matrix organization and uniform tube expansion by supporting polarized epithelial protein localization. We propose a model in which Knk and the predicted chitin-binding protein Rtv form membrane complexes essential for epithelial tubulogenesis and cuticle formation through their specific role in directing chitin filament assembly.  相似文献

Summary

The integument of the leech Hirudo medicinalis is mainly composed of a single layer of cuticle-secreting epidermal cells. The cuticle is made up of collagen fibers which support a layer of membrane-bound epicuticular projections.

Shedding of the old cuticle is preceded by the formation of a new cuticle. The epicuticular projections are the first to develop: they originate from the tips of numerous microvilli of the epidermal cells. As soon as it appears, the newly-formed collagen layer is firmly attached to the epidermal cells by numerous hemidesmo-somes, whereas the old cuticle is no longer connected with the epidermal surface. The epidermal cells exhibit marked characteristics of secretory activity during the laying down of the new cuticle.

The observations are discussed in connexion with recent findings of high ecdysteroid levels in leeches at the beginning of the molting cycle.  相似文献

The insect cuticle is non-cellular matrix secreted from a monolayer of epidermal cells. After abrasion of the larval cuticle of the silkworm, Bombyx mori, a protein with molecular mass of 135 kDa is newly detected in the cuticle. Mass spectrometric analysis of the tryptic fragments from this protein revealed that the 135-kDa protein is encoded by the Cb10 gene. In the predicted amino acid sequence of Cb10, three repeated motifs with [YxGGFGGppG(L/V)L] sequence are found in the C-terminal region. In addition to the repeated motifs, Cb10 has seventeen CxxxxC motifs randomly distributed throughout the polypeptide chain and serine rich region at the N-terminal region. The Cb10 gene is strongly expressed in epidermal cells after pupal ecdysis, and its expression in the larval epidermal cells is induced not only by cuticular abrasion, but also by bacterial infection. These expression patterns suggest some specific roles of this protein in pupal cuticle formation and defense reactions.  相似文献

Incorporation of tritiated leucine, tyrosine and glucosamine into the integument of larval Drosophila melanogaster was followed by electron-microscope autoradiography. Tritiated leucine, tyrosine, and glucosamine were incorporated into the endocuticle by apposition, giving rise to a distinct band of label in the endocuticle at a level which depended on the time between labelling and fixation. The labelled amino acids, but not glucosamine, were also detected in the epicuticle and both above and below the distinct labelled band in the endocuticle. The results indicate that the epicuticle grows within the third instar by intussusception of new materials which are transported from the epidermal cells through the endocuticle to the epicuticle. Breakdown of cuticle which was radioactively labelled by feeding larvae tritiated precursors was also followed by autoradiography. The results indicate that the breakdown products from the old cuticle may be reutilized in the synthesis of new cuticle.  相似文献

Plant epidermal cells dedicate more than half of their lipid metabolism to the synthesis of cuticular lipids, which seal and protect the plant shoot. The cuticle is made up of a cutin polymer and waxes, diverse hydrophobic compounds including very-long-chain fatty acids and their derivatives. How such hydrophobic compounds are exported to the cuticle, especially through the hydrophilic plant cell wall, is not known. By performing a reverse genetic screen, we have identified LTPG, a glycosylphosphatidylinositol-anchored lipid transfer protein that is highly expressed in the epidermis during cuticle biosynthesis in Arabidopsis thaliana inflorescence stems. Mutant plant lines with decreased LTPG expression had reduced wax load on the stem surface, showing that LTPG is involved either directly or indirectly in cuticular lipid deposition. In vitro 2-p-toluidinonaphthalene-6-sulfonate assays showed that recombinant LTPG has the capacity to bind to this lipid probe. LTPG was primarily localized to the plasma membrane on all faces of stem epidermal cells in the growing regions of inflorescence stems where wax is actively secreted. These data suggest that LTPG may function as a component of the cuticular lipid export machinery.  相似文献

The integument and podia of the sea cucumber Thyone briareus were examined by bright field and electron microscopy. The epidermal surface was found to be covered by an acellular, PAS positive cuticle which appeared to be secreted by the underlying epidermal cells. Although the superficial portion of the cuticle contains numerous fine filaments, their ultrastructure bears no resemblance to collagen fibers. The epidermal cells are widely spaced and have long apical processes that extend along the under surface of the cuticle forming a contiguous epithelium. The apical expansions of the epidermal cells are attached to one another by means of septate desmosomes which may run continuously around all epidermal cells. Special attachment structures within these apical expansions appear to bind the cuticle to the dermis. The epidermal cells and their apical expansions are separated from the dermis by an 800 Å thick basement membrane. Granule containing cells in the upper dermis send processes up to the cuticle where they are bound to the typical epidermal cells by septate desmosomes. The abundant membrane bound granules of the cells enter villous-like processes which pass through the cuticle. The function of these cells may be to produce an adhesive material on the podia or they may be pigment cells. The thick dermis consists of a superficial zone, containing largely ground substance; a middle or laminated zone containing laminae of collagen fibers arranged in an orthogonal fashion; and a hypodermis consisting largely of ground substance and reticular fibers. Fibroblasts are abundant in the superficial dermis and between the collagen laminae. Wandering coelomocytes, or morula cells, accumulate between the collagen laminae and in the hypodermis. They may also become an integral part of the epidermis by forming septate desmosomes with epidermal cells. Morula cells contain highly specialized spherules whose tinctorial properties and electron microscopic appearance suggest that they contain protein and mucopolysaccharide.  相似文献

To evaluate the ecdysteroid-like mode of action of tebufenozide (RH-5992), the effects on the fine structure of the integument in last- and third-instar larvae of the beet armyworm, Spodoptera exigua, and on cuticle formation in cultured imaginal wing discs, were studied. After 3 h of treatment with tebufenozide, the first signs of a normal moult were observed in treated larvae. A few hours later, ecdysial space formation and secretion of a new epicuticle were started. Furthermore, the new cuticle was incomplete in treated larvae; the new procuticle was absent or contained only a very low number of lamellae. In addition, epidermal cells showed many vacuoles and symptoms of degeneration with increase in time. Only a few lamellae of the old procuticle were digested, and normal ecdysis was inhibited which led to the presence of a double cuticle within 24–48 h after treatment. Similarly, cultured discs were stimulated to deposit a new cuticle within 12 h after cultivation in a medium containing tebufenozide. Our observations in treated S. exigua larvae on the one hand and in imaginal discs cultured with tebufenozide on the other hand are indicative of a hyperecdysteroid action, and confirm that the moult accelerating mode of action of tebufenozide resulted in a forced, untimely synthesis of cuticle by activation of epidermal or epithelial cells, and that its ecdysis inhibitory activity is mediated by its effect on post-apolysis processes. © 1996 Wiley-Liss, Inc.  相似文献

Summary The sensilla are associated with 6 enveloping cells. The innermost enveloping cell (e 1) secretes the dendritic sheath (=thecogen cell). All other enveloping cells are involved in the formation of the outer cuticular apparatus in secreting the cuticle of a definite region of the new hair shaft.The development of the new sensilla begins when an exuvial space expands between old cuticle and epithelium. The newly forming hair shafts lie folded back in an invagination of the epidermal tissue. Only a distal shaft part projects into the free exuvial space. The cuticle of the distal and middle shaft region is secreted by the three middle enveloping cells (e 2–e 4) (=trichogen cells), which are arranged around the dendritic sheath.The wall of the cylinder, in which the distal shaft is situated, is formed by the cuticle of the future proximal shaft region. It is secreted by the outer enveloping cells (e 5 and e 6). Furthermore, both enveloping cells form the hair socket (=trichogen-tormogen cells).The outer dendritic segments encased within a dendritic sheath run up through the newly formed hair shaft and continue to the old cuticular apparatus. The connection between sensory cells and old hair shaft is maintained until ecdysis. On ecdysis the old cuticle is shed and the newly formed shaft of the sensillum is everted like the invaginated finger of a glove. The dendritic sheath and the outer dendritic segments break off at the tip of the new hair shaft. Morphologically this moulting process ensures that the sensitivity of the receptors is maintained until ecdysis.The internal organization of the sensory cells shows no striking changes during the moulting cycle. An increased number of vesicles is accumulated distally within the inner dendritic segments and distributed throughout the outer segments of the dendrites. The cytoplasmic feature of the enveloping cells indicates that synthesis and release of substances for the cuticular apparatus of the new sensillum take place.  相似文献

It was previously established that Autographa nigrisigna loopers form cuticular cysts at the dorsal site of the 9th (penultimate) abdominal segment after parasitization by the solitary endoparasitoid Campoletis chlorideae and get rid of the parasitoid egg with the old cuticle at ecdysis. The cuticular cyst consists of a space between the old cuticle and new cuticle formed by the epidermis to enclose the parasitoid egg. The fact that A. nigrisigna loopers exclude the oviposited egg from the hemocoel using a cuticular cyst raises the question how the parasitoid egg passes through the epidermis. To exclude the endoparasitoid eggs from the hemocoel, the epidermis is required to move the location of the parasitoid egg. In the current study, we investigated the morphological process of cuticular cyst formation. First, the oviposited egg drifted to the 9th abdominal segment located at the open end of the dorsal vessel as a result of force generated by the hemolymph current from the oviposition site, and formed contacts with the integument containing the fat body (FB). The epidermis, in contact with the egg, then began to move along with the basement membrane formed on the surface of the FB, and settled under the egg, thus altering its location. This inversion was duplicated in vitro using integument from the 9th abdominal segment when parasitoid eggs were inserted between the epidermis and FB. When the integument, without the FB, was incubated on an agar plate, the epidermal cells migrated on the plate. Integument without eggs showed no signs of migration from their original sites. When the actin polymerization inhibitor latrunculin B was added to the cultures, the epidermal cells remained in their original location.  相似文献

Molting fluid accumulates between the old and new cuticles during periodical ecdysis in Ecdysozoa. Natural defects in insect ecdysis are frequently associated with melanization (an immunity response) occurring primarily in molting fluids, suggesting that molting fluid may impact immunity as well as affect ecdysis. To address this hypothesis, proteomic analysis of molting fluids from Bombyx mori during three different types of ecdysis was performed. Many proteins were newly identified, including immunity-related proteins, in each molting fluid. Molting fluids inhibited the growth of bacteria in vitro. The entomopathogenic fungi Beauveria bassiana, which can escape immune responses in feeding larvae, is quickly recognized by larvae during ecdysis, followed by melanization in molting fluid and old cuticle. Fungal conidia germination was delayed, and no hyphae were detected in the hemocoels of pharate instar insects. Molting fluids protect the delicate pharate instar insects with extremely thin cuticles against microorganisms. To explore the function of molting fluids in ecdysis regulation, based on protein similarity, 32 genes were selected for analysis in ecdysis regulation through RNAi in Tribolium castaneum, a model commonly used to study integument development because RNAi is difficult to achieve in B. mori. We identified 24 molting proteins that affected ecdysis after knockdown, with different physiological functions, including old cuticle protein recycling, molting fluid pressure balance, detoxification, and signal detection and transfer of molting fluids. We report that insects secrete molting fluid for protection and regulation of ecdysis, which indicates a way to develop new pesticides through interrupting insect ecdysis in the future.  相似文献

The insect cuticle plays important roles in numerous physiological functions to protect the body from invasion of pathogens, physical injury and dehydration. In this report, we conducted a comprehensive genome-wide search for genes encoding proteins with peritrophin A-type (ChtBD2) chitin-binding domain (CBD) in the silkworm, Bombyx mori. One of these genes, which encodes the cuticle protein BmCBP1, was additionally cloned, and its expression and location during the process of development and molting in B. mori were investigated. In total, 46 protein-coding genes were identified in the silkworm genome, including those encoding 15 cuticle proteins analogous to peritrophins with one CBD (CPAP1s), nine cuticle proteins analogous to peritrophins with three CBD (CPAP3s), 15 peritrophic membrane proteins (PMPs), four chitinases, and three chitin deacetylases, which contained at least one ChtBD2 domain. Microarray analysis indicated that CPAP-encoding genes were widely expressed in various tissues, whereas PMP genes were highly expressed in the midgut. Quantitative polymerase chain reaction and western blotting showed that the cuticle protein BmCBP1 was highly expressed in the epidermis and head, particularly during molting and metamorphosis. An immunofluorescence study revealed that chitin co-localized with BmCBP1 at the epidermal surface during molting. Additionally, BmCBP1 was notably up-regulated by 20-hydroxyecdysone treatment. These results provide a genome-level view of the chitin-binding protein in silkworm and suggest that BmCBP1 participates in the formation of the new cuticle during molting.  相似文献

为了探明20-羟基蜕皮甾酮对昆虫蜕皮过程中体壁的表皮层、 皮细胞及其细胞器的具体影响过程, 本研究利用透射电镜技术研究了20-羟基蜕皮甾酮对舞毒蛾Lymantria dispar (Linnaeus)5龄幼虫体壁超微结构的变化。结果表明, 用高浓度20-羟基蜕皮甾酮溶液浸过的白桦叶片饲喂幼虫, 处理6 h, 摄入约400 μg 20-羟基蜕皮甾酮后, 幼虫停止取食; 处理12 h时表皮细胞顶膜上的微绒毛减少, 在皮细胞与旧表皮之间形成蜕皮间隙, 旧头壳从幼虫头部脱离; 处理24 h时蜕皮间隙继续增大, 旧表皮与皮细胞进一步分离, 新表皮质层开始形成; 处理36 h时皮细胞顶膜形成较短的微绒毛, 胞质区域出现数量较多的电子疏松泡, 新表皮由上表皮、 外表皮及8层左右内表皮片层组成; 处理48 h时顶膜与内表皮界限模糊, 内表皮继续合成至16层左右; 72 h时细胞内出现大面积电子疏松泡, 内表皮合成至20层左右。 处理96 h时, 与对照组相比, 皮细胞细胞器较少, 核仁周围出现小部分空白区域, 胞质区域内含物减少; 虫体发黑缩小, 即将死亡; 内表皮层数仍旧保持20层左右。对照组幼虫6-96 h虫体活跃, 正常取食, 外部观察及透射电镜结果均未显现蜕皮现象; 表皮层由上表皮、 外表皮及内表皮组成; 皮细胞顶膜微绒毛密度高; 表皮细胞分泌活动旺盛, 胞质区域细胞界限明显, 内含物丰富; 细胞器典型而且活跃; 内表皮片层随时间不断增加至50层左右。结果提示, 外源20-羟基蜕皮甾酮能够导致舞毒蛾5龄幼虫的致死性蜕皮。  相似文献

The cheirolepidiaceous conifer Pseudofrenelopsis dalatzensis from the Lower Cretaceous Dalazi Formation, Jilin Province, Northeast China is studied in detail based on abundant material. The gross morphology of vegetative shoots shows a greater variation than previously known. Additional information of microstructure is that the adaxial cuticle from the free part of leaves is also papillate as the abaxial cuticle. The excellent preservation of cuticles allows for the first time in this family to four distinct types of cuticle in ultrastructural level: subsidiary cells, guard cells, epidermal cells with and without papillae. Based on 30 measurements the statistic approach on each type of cell cuticle, i.e. not only the total thickness of the cuticle but also details and proportions of all different layers. A key is provided using a combination of 7 significant ultrastructural characters. These results seem to be very promising for the identity of each taxon within the Cheirolepidiaceae family. The cuticular ultrastructure of this family is characterized by having a cuticle proper A composed of A1 wavy polylamellate layer and A2 granular layer, above the cuticular layer B with B1 fibrillate and B2 granular layers. The importance of ultrastructural characters at different levels of taxonomy and palaeoecology is also discussed.  相似文献

The entomogenous fungusMetarhizium anisopliae attacks a broad range of insects, including the agricultural pestsGalleria mellonella (the Greater Wax Moth) andTrichoplusia ni (the Cabbage Looper). Five strains ofM. anisopliae from widely divergent isolation sources were culturedin vitro on media containing gelatin, glucose plus nitrate, or purified cuticle fromG. mellonella orT. ni larvae. The production of extracellular enzymes such as proteases, chitinases, and esterase was compared. A great deal of natural strain variability was found in enzyme patterns. The highest levels of proteases and endochitinase were produced in cuticle-grown cultures. Three of five strains produced exceptionally high levels of chymoelastase (47,000 to 98,000 IU/mg protein) on cuticle. Surprisingly, the highest levels ofN-acetyl glucosaminidase were produced in gelatin-grown cultures. Most strains produced esterase under all growth conditions. The source of insect cuticle did not strongly influence the production of enzymes.  相似文献

The onset of molting in all stages of Hemicycliophora arenaria was preceded by the appearance of numerous, discrete globular structures which were termed "molting bodies" because they were present in the hypodermis only during the production of the new cuticle. In all parasitic stages the molt commenced with the separation of the cuticle from the hypodermis from which the new sheath and cuticle were differentiated. Following completion of the new sheath and cuticle most of the old outer covering was apparently absorbed before ecdysis. Electronmicrographs of body wall cross sections in molting L4 male specimens revealed the final molt to be a double molt in which an additional sixth cuticle was produced. Since both a new sheath and cuticle were produced during the molt of each stage, the sheath must be considered as an integral part of the cuticle and not as a residual cuticle or the result of an incomplete additional molt. Molting in Aphelenchus avenae and Hirschmanniella gracilis was less complex and "molting bodies" were not observed. After cuticle separation the hypodermis gave rise to a new trilaminate zone, the future cortex, and (later) the matrix and striated basal layers.  相似文献

Summary The integument of the woodlouse,Oniscus asellus, consists of a two-layered epicuticle, a largely lamellate procuticle — itself divided into two regions (pre-and postecdysial cuticles), and the epidermis. At the initiation of new cuticle production the epidermal cells become vacuolated and retract away from the cuticle. Apolysis occurs immediately after the cessation of postecdysial cuticle production. The formation of the epicuticle is unique among the arthropods since material aggregates along the distal epidermal membrane. By indenting, doubling back on itself, and incorporating septa, the epicuticle forms surface structures such as plaques and tricorns.The innervation, and so the receptive function of the tricorns is confirmed, but since there is no connection between the old and new receptors during premoult, sensory information from these exoreceptors must be severely curtailed. This may explain the biphasic moult in all isopods since it ensures that only half the body experiences this sensory deprivation at any one time. In terrestrial species there is the additional advantage of restricting the area of permeable new cuticle. The frequency of moulting may be due to the need to renew disrupted receptor surfaces.Tricorns do not appear to be the mechanoreceptors involved in the marked thigmotactic response of woodlice since they do not have the typical internal structure of such receptors; rather, the dendrite —which extends into the lumen of the tricorn —is protected from deformation by the previously unreported combination of a dendritic sheath and a cuticular tube. The modality of tricorns is possibly one of hygro-perception. One of the behavioural responses of woodlice to desiccation is aggregation. The numerical distribution of tricorns over the body surface is admirably suited to assist in the formation and maintenance of such aggregates during desiccation and to their observed dispersal when the relative humidity rises.  相似文献
