Hormone secretion processes remain part of the biological mystery as they are highly regulated and individual specific. When hormone trajectories from multiple subjects are under investigation, both population-average mechanism and subject-specific deviations are of great interest. In particular, statistical methodologies that enable us not only to identify surge times and surge magnitudes but also to make inference on these biological features is in need. In this paper, we propose a local kernel smoothing method to perform the analysis of multiple hormone curves using the nonparametric mixed-effects model. We develop a local quadratic mixed-effects (LQME) fitting procedure that detects local maxima of the population-average profile curve and the individual profile curves. Related statistical inference is established to carry out a hypothesis test for the local surge and to construct a confidence interval for a detected surge feature. This method is illustrated by simulation studies and a reproductive hormone data analysis.  相似文献

SYNOPSIS. Fundamentally different mechanisms regulate femalesexual behavior in the ovoviviparous guppy and the oviparousgoldfish. In the female guppy, ovarian estrogen evidently synchronizescycles of sexual receptivity with endogenous cycles of ovarianmaturation and also increases female attractivity at the timeof maximum receptivity by stimulating the release of a sexualpheromone. In the goldfish, it appears that prostaglandin, releasedfrom the ovary or oviduct in conjunction with ovulation andthe presence of ovulated eggs, acts on the brain to stimulatespawning behavior. In contrast to the situation in the guppy,steroid treatments alone (in the absence of ovulated eggs) failto stimulate spawning behavior in the goldfish. It isproposedthat endocrine mechanisms regulating female sexual behaviorin the teleosts and in other vertebrates are less related tophylogeny than to the mode of reproduction employed. In thegoldfish and several other externally fertilizing teleosts,where sexual behavior involves oviposition, female sexual behaviorapparently is synchronized with ovulation by mechanisms whichrespond to elevated plasma prostaglandin as an indicator ofthe presence of ovulated eggs. In internally fertilizing species(guppy, reptiles, birds, mammals), where sexual behavior andfertilizationare temporally dissociated, female sexual behavior is synchronizedwith ovulation by mechanisms which anticipate either an imminentspontaneous ovulation, or the potential for reflex ovulation,by responding to increases in plasma estrogen associated withfolliculardevelopment.  相似文献

Physiological and behavioural parameters associated with reproductive effort and success were investigated in female European ground squirrels Spermophilus citellus . The proportion of reproductive (lactating) females in the study population was over 90% and was not related to age. Timing of oestrus and ovulation was found to be affected by the female's emergence date and condition. Females with low emergence mass showed delayed oestrus. Differences in ovulation dates were shown to affect reproductive output in terms of litter size and sex ratio. Early litters were larger and male biased. X-ray techniques were used to determine intrauterine litter size in individual females. The results indicated that litter size and sex ratio were fixed prenatally. Lactation costs were reflected in the intensity of mass loss and duration of lactation. Mass loss varied with litter size, in that females with large litters showed a more rapid loss than others. The second parental investment parameter, lactation duration, varied among individual females and was dependent on the timing of reproduction and litter size (except yearlings). Early born litters, which were, in most cases, larger than later ones, were nursed longer. Prolonged lactation periods affected female condition in that they started prehibernation fattening later and entered hibernation with a lower mass than individuals that had shorter lactation periods. Yearling females probably could not afford the energetic costs of long lactation, independent of their offspring number. These results indicated that females with higher reproductive output and higher investment were unable to compensate these costs before hibernation. Consequences for these individuals could therefore be lower over-winter survival or a delayed oestrus in the following season.  相似文献

目的探讨促性腺激素释放激素类似物(GnRH-A)对动物生殖功能调节的效果和作用机制。方法 24只日本大耳白兔分为四组,分别在实验Ⅰ组(EG-I)、实验Ⅱ组(EG-II)和实验III(EG-III)组兔的颈背侧注射1.0 mL(100、100和50μg/mL)GnRH-A抗原,实验II组和实验III组于第3周以原剂量加强注射一次,用ELISA法测定血清GnRH抗体效价、促卵泡刺激素(FSH)和促黄体生成素(LH)含量。结果注射GnRH-A后10 d实验组兔均出现GnRH抗体,而对照组未检测到;EG-I在第30天达到高峰,而EG-II和EG-III于40~50 d至峰值,但在实验结束时(70 d)实验组均高于对照组,40~70 d时EG-II显著高于EG-I和EG-III。30~50 d时EG-II的LH明显高于EG-I和EG-III及对照组。EG-II和EG-III的FSH浓度在40 d达到峰值,但EG-II高于EG-I、对照组及EG-III,EG-I和对照组无显著差异。结论兔体内注射GnRH-A可以明显提高GnRH抗体效价,增强LH和FSH的合成与分泌,加强注射效果更明显,且与注射剂量相关,持续时间为40 d左右。  相似文献

In most livebearing fish, the gravid spot is an excellent marker to identify brooding females, however its use to predict progress of embryonic development, brood size, timing of parturition and overall reproductive potential of populations remain unexplored. Therefore, to understand these relationships, this study quantified visual attributes (intensity and size) of the gravid spot in relation to key internal development in Gambusia holbrooki. Observations show that the colour of the gravid spot arises from progressive melanisation on the surface of the ovarian sac at its hind margin, rather than melanisation of the developing embryos or the skin of the brooding mother. More importantly, the gravid spot intensity and size were closely linked with both developmental stages and clutch size, suggesting their reliable use as external surrogates of key internal developmental in the species. Using predictive consistency of the gravid spot, we also determined the effect of rearing temperature (23°C and 25°C) on gestation period and parturition behaviour. The results show that gestation period was significantly reduced (F = 364.58; df = 1,48; P˃0.05) at 25°C. However there was no significant difference in average number of fry parturated in the two temperature groups (P<0.05), reaffirming that gravid spot intensity is a reliable predictor of reproductive output. The parturition in the species occurred predominantly in the morning and in contrast to earlier reports, tails of the fry emerged first with a few exceptions of head-first, twin and premature births. This study demonstrates utility of the gravid spot for downstream reproductive investigations in a live-bearing fish both in the field and laboratory. The reproducibility of the relationships (intensity with both developmental stage and clutch size), imply that they are also relevant to wild populations that experience varying temperature climes and stressors, significant deviations of which may serve as indicators of environmental health and climate variability.  相似文献

Paliperidone (PPD) is the most recent second-generation atypical antipsychotic approved for the treatment of schizophrenia. An immediate release dose causes extrapyramidal side effects. In this work, a novel nanolipomer carrier system for PPD with enhanced intestinal permeability and sustained release properties has been developed and optimized. PPD was successfully encapsulated into a lipomer consisting of a specific combination of biocompatible materials including poly-ε-caprolactone as a polymeric core, Lipoid S75, and Gelucire® 50/13 as a lipid shell and polyvinyl alcohol as a stabilizing agent. The lipomer system was characterized by dynamic light scattering, TEM, DSC, and FTIR. An optimized lipomer formulation possessed a particle size of 168 nm, PDI of 0.2, zeta potential of ?23 mV and an encapsulation efficiency of 87.27%?±?0.098. Stability in simulated gastrointestinal fluids investigated in terms of particle size, zeta potential, and encapsulation efficiency measurements ensured the integrity of the nanoparticles upon oral administration. PPD-loaded nanolipomers demonstrated a superior sustained release behavior up to 24 h and better ex vivo intestinal permeation for PPD compared to the corresponding polymeric and solid lipid nanoparticles and drug suspension. The in vitro hemocompatibility test on red blood cells revealed no hemolytic effect of PPD-loaded lipomers which reflects its safety. The elaborated nanohybrid carrier system represents a promising candidate for enhancing the absorption of PPD providing a 2.6-fold increase in the intestinal permeation flux compared to the drug suspension while maintaining a sustained release behavior. It is a convenient alternative to the commercially available dosage form of PPD.  相似文献

蜥蜴的雌性繁殖特征对理解两性异形的进化原因起着重要作用。于2011年4月在安徽滁州采集宁波滑蜥(Scincella modesta),定量研究该种形态特征的两性异形和雌性繁殖特征,检验成体形态特征两性异形与雌性繁殖的相关性。研究共采集43条(17♀♀,26♂♂)宁波滑蜥,雄性和雌性个体的最大体长分别为47.4 mm和46.6 mm。雌雄两性在体长和头宽上没有差异,而在腹长和头长上差异显著,雄性有较大的头长,雌性有较大的腹长。宁波滑蜥年产单窝卵。窝卵数和窝卵重与雌体体长及腹长呈正相关,卵重与雌体体长无相关性。窝卵数及卵重的变异系数分别为0.20和0.12。卵长径与窝卵数呈负相关,而卵短径与窝卵数无关。雌体主要通过增加窝卵数来增加繁殖输出。这些结果表明,宁波滑蜥是雌雄个体大小同形的两性异形模式,性选择使得雄性有着较大的头长,以具有较高的交配成功率,生育力选择使得雌性有着较大的腹长,以具有较大的生育力和繁殖输出。  相似文献

There is compelling evidence on a global scale for compromised growth and reproduction, altered development, and abnormal behaviour in feral fish that can be correlated or in some cases causally linked with exposure to endocrine disrupting chemicals (EDCs). Attributing cause and effect relationships for EDCs is a specific challenge for studies with feral fish as many factors including food availability, disease, competition and loss of habitat also affect reproduction and development. Even in cases where there are physiological responses of fish exposed to EDCs (e.g., changes in reproductive hormone titres, vitellogenin levels), the utility of these measures in extrapolating to whole animal reproductive or developmental outcomes is often limited. Although fish differ from other vertebrates in certain aspects of their endocrinology, there is little evidence that fish are more sensitive to the effects of EDCs. Therefore, to address why endocrine disruption seems so widespread in fish, it is necessary to consider aspects of fish physiology and their environment that may increase their exposure to EDCs. Dependence on aquatic respiration, strategies for iono-osmotic regulation, and maternal transfer of contaminants to eggs creates additional avenues by which fish are exposed to EDCs. This paper provides an overview of responses observed in feral fish populations that have been attributed to EDCs and illustrates many of the factors that need consideration in evaluating the risks posed by these chemicals.  相似文献

Study on the state of bighead carp Aristichthys nobilis females' central and peripheral of hypothesis gonadotropocytes at light and electron microscopy levels after stimulation of ripening a chorionic gonadotropin revealed that the amount of secretory material diminishes in both types of cells. Intensive excretion of both secretory granules and content of cisterns of the granular endoplasmic reticulum take place. Formation by the hypophysis of an ovulatory wave of the gonadotropin in the females with different fecundity differs due to the change in proportions of processes of mero- and holocrine secretion.  相似文献

In seasonal breeding vertebrates, hormone regulation of catecholamines, which include dopamine and noradrenaline, may function, in part, to modulate behavioral responses to conspecific vocalizations. However, natural seasonal changes in catecholamine innervation of auditory nuclei is largely unexplored, especially in the peripheral auditory system, where encoding of social acoustic stimuli is initiated. The plainfin midshipman fish, Porichthys notatus, has proven to be an excellent model to explore mechanisms underlying seasonal peripheral auditory plasticity related to reproductive social behavior. Recently, we demonstrated robust catecholaminergic (CA) innervation throughout the auditory system in midshipman. Most notably, dopaminergic neurons in the diencephalon have widespread projections to auditory circuitry including direct innervation of the saccule, the main endorgan of hearing, and the cholinergic octavolateralis efferent nucleus (OE) which also projects to the inner ear. Here, we tested the hypothesis that gravid, reproductive summer females show differential CA innervation of the auditory system compared to non-reproductive winter females. We utilized quantitative immunofluorescence to measure tyrosine hydroxylase immunoreactive (TH-ir) fiber density throughout central auditory nuclei and the sensory epithelium of the saccule. Reproductive females exhibited greater density of TH-ir innervation in two forebrain areas including the auditory thalamus and greater density of TH-ir on somata and dendrites of the OE. In contrast, non-reproductive females had greater numbers of TH-ir terminals in the saccule and greater TH-ir fiber density in a region of the auditory hindbrain as well as greater numbers of TH-ir neurons in the preoptic area. These data provide evidence that catecholamines may function, in part, to seasonally modulate the sensitivity of the inner ear and, in turn, the appropriate behavioral response to reproductive acoustic signals.  相似文献

The aim of this work was to study the potential of delivering clindamycin phosphate, as an efficient antibiotic drug, into a more absorbed, elastic ultradeformable form, transfersomes (TRSs). These vesicles showed an enhanced penetration through ex vivo permeation characters. TRSs were prepared using thin-film hydration method. Furthermore, they were evaluated for their entrapment efficiency, size, zeta potential, and morphology. Also, the prepared TRSs were converted into suitable gel formulation using carbopol 934 and were evaluated for their gel characteristics like pH, viscosity, spreadability, homogeneity, skin irritation, in vitro release, stability, and ex vivo permeation studies in rats. TRSs were efficiently formulated in a stable bilayer vesicle structure. Furthermore, clindamycin phosphate showed higher entrapment efficiency within the TRSs reaching about 93.3%?±?0.8 and has a uniform particle size. Moreover, the TRSs surface had a high negative charge which indicated the stability of the produced vesicles and resistance of aggregation. Clindamycin phosphate showed a significantly higher in vitro release (p?<?0.05; ANOVA/Tukey) compared with the control carbopol gel. Furthermore, the transfersomal gel showed a significantly higher (p?<?0.05; ANOVA/Tukey) cumulative amount of drug permeation and flux than both the transfersomal suspension and the control carbopol gel. In conclusion, the produced results suggest that TRS-loaded clindamycin are promising carriers for enhanced dermal delivery of clindamycin phosphate.  相似文献

The female reproductive tract (FRT) includes the oviducts (fallopian tubes), uterus, cervix and vagina. A layer of columnar epithelium separates the endocervix and uterus from the outside environment, while the vagina is lined with stratified squamous epithelium. The mucosa of the FRT is exposed to antigens originating from microflora, and occasionally from infectious microorganisms. Whether epithelial cells (ECs) of the FRT take up (sample) the lumen antigens is not known. To address this question, we examined the uptake of 20–40 nm nanoparticles (NPs) applied vaginally to mice which were not treated with hormones, epithelial disruptors, or adjuvants. We found that 20 and 40 nm NPs are quickly internalized by ECs of the upper FRT and within one hour could be observed in the lymphatic ducts that drain the FRT, as well as in the ileac lymph nodes (ILNs) and the mesenteric lymph nodes (MLNs). Chicken ovalbumin (Ova) conjugated to 20 nm NPs (NP-Ova) when administered vaginally reaches the internal milieu in an immunologically relevant form; thus vaginal immunization of mice with NP-Ova induces systemic IgG to Ova antigen. Most importantly, vaginal immunization primes the intestinal mucosa for secretion of sIgA. Sub-cutaneous (s.c) boosting immunization with Ova in complete Freund''s adjuvant (CFA) further elevates the systemic (IgG1 and IgG2c) as well as mucosal (IgG1 and sIgA) antibody titers. These findings suggest that the modes of antigen uptake at mucosal surfaces and pathways of antigen transport are more complex than previously appreciated.  相似文献

Maturational delay of young female mice as the result of exposureto grouped female odors and reproductive inhibition as the resultof exposure to isolated adult females have both been observed(Drickamer, 1974; Skryja, 1978). Each has the potential to reducethe growth rate of populations. Reductions in a female's reproductionfacilitated by social stimulation from other females, whileeffective in reducing population growth, may in the case, ofmaturational delay and reproductive inhibition be an epiphenomenonor exadaptation of selection for improved relative reproductivesuccess in the females possessing these abilities. The ultimateoutcome of these selective processes may be the buffering ofpopulation numbers, but the selective forces may operate tomaximize a female's relative reproductive success. A females'relative reproductive success can be maximized by either increasingher own reproduction or decreasing the reproductive output ofother females. A body of evidence exists to suggest that inPeromyscus mamculatus and Peromyscus leucopus, females are physiologicallyconstrained and unable to increase their own reproduction. Ifthe assumption of physiological restraint is correct, then themost effective way for females to maximize their relative reproductivesuccess is to reduce the reproductive output of their competitors.In this paper, maturational delay and reproductive inhibitionas they affect both the adult female and young females are discussed.Examination of these effects reveals that while they can beeffective in population regulation, their main function andthe selective process that produced them is at the level ofindividual reproduction.  相似文献

Neurobiology of Stress-Induced Reproductive Dysfunction in Female Macaques   总被引:1,自引:0,他引:1  
It is now well accepted that stress can precipitate mental and physical illness. However, it is becoming clear that given the same stress, some individuals are very vulnerable and will succumb to illness while others are more resilient and cope effectively, rather than becoming ill. This difference between individuals is called stress sensitivity. Stress sensitivity of an individual appears to be influenced by genetically inherited factors, early life (even prenatal) stress, and by the presence or absence of factors that provide protection from stress. In comparison to other stress-related diseases, the concept of sensitivity versus resilience to stress-induced reproductive dysfunction has received relatively little attention. The studies presented herein were undertaken to begin to identify stable characteristics and the neural underpinnings of individuals with sensitivity to stress-induced reproductive dysfunction. Female cynomolgus macaques with normal menstrual cycles either stop ovulating (stress sensitive) or to continue to ovulate (stress resilient) upon exposure to a combined metabolic and psychosocial stress. However, even in the absence of stress, the stress-sensitive animals have lower secretion of the ovarian steroids, estrogen and progesterone, have higher heart rates, have lower serotonin function, have fewer serotonin neurons and lower expression of pivotal serotonin-related genes, have lower expression of 5HT2A and 2C genes in the hypothalamus, have higher gene expression of GAD67 and CRH in the hypothalamus, and have reduced gonadotropin-releasing hormone transport to the anterior pituitary. Altogether, the results suggest that the neurobiology of reproductive circuits in stress-sensitive individuals is compromised. We speculate that with the application of stress, the dysfunction of these neural systems becomes exacerbated and reproductive function ceases.  相似文献

本研究以2004年3月2004年5月以卧龙中国保护大熊猫研究中心的11只3.5~17岁、体重约在80~115kg的成年雌性大熊猫为研究对象,采用全事件记录法对雌性大熊猫的整个发情周期的行为和生殖系统外观变化进行详细记录,并于2004年3月9日~5月10日测定了8只雌性大熊猫尿中雌二醇浓度;统计分析了1991~2004年间的雌性大熊猫阴道上皮细胞角化率。结果表明,除了大熊猫帼帼在2004年1月28日开始有发情行为表现外,其他10只均在3~5月期间发情,发情行为无异常现象,其中5只完成了配种;尿中雌二醇浓度均在发情高潮期的当天或头天达到峰值,并于发情后期迅速降至发情前水平;阴道上皮细胞角化率变幅在39.0%~97.0%之间,也表现为在发情高潮期增加并达到最大值,此值介于75%~90%被认为是最佳配种时期。  相似文献

测定了黄颡鱼成体的体长、头长、头宽、头高、吻长、眼径、眼间距、眼后头长、体高、尾柄高、背鳍基前距、背鳍基长、尾柄长、腹鳍基前距、背鳍脂鳍间距、腹鳍臀鳍间距、尾鳍长、体重、去内脏体重等形态指标以及雌体的怀卵数量。雌性成体的体长显著小于雄性成体。其它局部特征皆与体长呈正相关,回归剩余值的t—检验表明,雌性成体的眼径、头高、体高、腹鳍基前距、体重显著大于雄性成体,其它局部形态特征不存在显著的两性差异。黄颡鱼雌体通过个体大小的增加和腹部形态的改变增加腹腔容量,增加繁殖输出。  相似文献

Somatolactin, a pituitary hormone belonging to the growth hormone/prolactin family, is produced in the intermediate lobe of teleost pituitary. To date, the functions of this new hormone and the target tissues are unknown. ASolea senegalensissomatolactin (ssSL) cDNA has previously been cloned and isolated. Here we have inserted this cDNA into a pET-3a plasmid in order to produce recombinant ssSL inE. coliBL21 (DE3) cells. The protein induced was isolated from inclusion bodies by a solubilization–renaturation procedure originally developed to generate native disulfide bonds, to get putative active proteins. The recombinant somatolactin was further purified to homogeneity by gel filtration on FPLC. The estimated molecular weight of 26 kDa by sodium dodecyl sulfate–polyacrylamide gel electrophoresis agrees well with the molecular mass calculated from the translated cDNA sequence and with native somatolactin (SL). The recombinant protein showed electrophoretic mobility identical to that of one of the native forms of SL secretedin vitroby cultured pituitaries from sole. Another native SL expressed inS. senegalensisrepresented a glycosylated modified hormone as shown byN-glycosidase treatment. Further, recombinant SL was recognized by an anti-native SL antibody and used to generate polyclonal sera reactive with the native pituitary hormone. To date, this represents the first recombinant SL protein isolated in sufficient quantities for biophysical and biochemical investigation and for studies on its physiological actions.  相似文献

Atrazine (ATR) blunts the hormone‐induced luteinizing hormone (LH) surge, when administered by gavage (50–100 mg/kg/day for 4 days), in ovariectomized rats. In this study, we determined if comparable doses delivered either by gavage (bolus dose) or distributed in diet would reduce the LH surge and subsequently affect fertility in the intact female rat. ATR was administered daily to intact female Sprague‐Dawley (SD) or Long Evans (LE) rats by gavage (0, 0.75 1.5, 3, 6, 10, 12, 50, or 100 mg/kg/day) or diet (0, 30, 100, 160, 500, 660, or 1460 ppm) during one complete 4‐day estrous cycle, starting on day of estrus. Estrous status, corpora lutea, ova, and LH plasma concentrations were evaluated. A second cohort of animals was mated on the fourth treatment day. Fertility metrics were assessed on gestational day 20. A higher portion of LE rats had asynchronous estrous cycles when compared to SD rats both during pretreatment and in response to ATR (≥50 mg/kg). In contrast, bolus doses of ATR (≥50 mg/kg) inhibited the peak and area under the curve for the preovulatory LH surge in SD but not LE animals. Likewise, only bolus‐treated SD, not LE, rats displayed reduced mean number of corpora lutea and ova. There were no effects of ATR administered by gavage on mating, gravid number, or fetus number. Dietary administration had no effect on any reproductive parameter measured. These findings indicate that short duration, high‐bolus doses of ATR can inhibit the LH surge and reduce the number of follicles ovulated; however, dietary administration has no effect on any endocrine or reproductive outcomes  相似文献
