Some recommendations in the Guide for the Care and Use of Laboratory Animals (the Guide) are based on best professional judgment. Our current efforts are directed toward replacement with data-driven standards. We demonstrated earlier that young adult C57BL/6J mice could be housed with half the floor space recommended in the Guide without discernable negative effects. This report extends that work by examining optimal housing densities for young adult male and female BALB/cJ, NOD/LtJ, and FVB/NJ mice. These 8-week studies were initiated with 3-week-old BALB/cJ and NOD/LtJ mice and 3- to 5-week-old FVB/NJ mice housed in three cage types. We adjusted the number of mice per cage to house them with the floor space recommended in the Guide (approximately 12 in2 [ca. 77 cm2] per mouse) down to 5.6 in2 [ca. 36 cm2] per mouse. Early-onset aggression occurred among FVB/NJ male mice housed at all densities in cages having 51.7 in2 (ca. 333 cm2) or 112.9 in2 (ca. 728 cm2) of space. FVB/NJ male mice housed in shoebox (67.6 in2 [ca. 436 cm2]) cages did not exhibit aggression until the fifth week. Urinary testosterone output was density-dependent only for BALB/cJ male mice in shoebox cages (output decreased with increasing density) and FVB/NJ male mice. We conclude that all but FVB/NJ male mice can be housed with half the floor space specified in the Guide. The aggression noted for male FVB/NJ mice may have been due to their age span, although this did not impact negatively on the female FVB/NJ mice. 相似文献

The recommendations are based on best professional judgment rather than experimental data. Current efforts are directed toward replacing these guidelines with data-driven, species-appropriate standards. Our studies were undertaken to determine the optimum housing density for C57BL/6J mice, the most commonly used inbred mouse strain. Four-week-old mice were housed for 8 weeks at four densities (the recommended approximately 12 in2 [ca. 77.4 cm2]/mouse down to 5.6 in2 [ca. 36.1 cm2]/mouse) in three cage types with various amounts of floor space. Housing density did not affect a variety of physiologic parameters but did affect certain micro-environmental parameters, although these remained within accepted ranges. A second study was undertaken housing C57BL/6J mice with as little as 3.2 in2/mouse (ca. 20.6 cm2). The major effect was elevated ammonia concentrations that exceeded limits acceptable in the workplace at increased housing densities; however, the nasal passages and eyeballs of the mice remained microscopically normal. On the basis of these results, we conclude that C57BL/6J mice as large as 29 g may be housed with 5.6 in2 of floor space per mouse. This area is approximately half the floor space recommended in the Guide. The role of the Guide is to ensure that laboratory animals are well treated and housed in a species-appropriate manner. Our data suggest that current policies could be altered in order to provide the optimal habitation conditions matched to this species' social needs. 相似文献

after adult emergence (n = 82), 61.0% of them did not guard females. Mate guarding with physical contact occurred more frequently when males had previously
experienced a high density of potential rivals than when they had not, but the difference in behavior between the two groups
of males was marginally not significant. Nevertheless, these results suggest overall that T. kanzawai males change mate guarding behavior in response to previously experienced density. 相似文献

The effects of castration of both resident and intruder rats on territorial aggressive behavior were studied. The results suggest that residence in a home cage is more important than gonadal status in determining the outcome of an aggressive encounter. Resident rats were more likely to be dominant especially if they were intact. Intact residents directed less aggressive behavior toward castrated intruders than toward intact intruders. Intruder rats generally showed low levels of aggressive behavior and were only dominant when the resident had been castrated. Thus, the aggressive behavior of a male rat depends upon both his gonadal status and that of his opponent. 相似文献

Changes in larval density and movement behavior of a commensal midge, Nanocladius (Plecopteracoluthus) sp. #4, were monitored for 26 weeks in recirculating laboratory streams. Adults were captured at emergence, sexed, and weighed to assess the effect of larval density and movement behavior on emergence success and adult size. The density of midges on hosts declined with time and coincided with a springtime increase in larval movement frequency. Midges residing on hosts with high spring densities emerged significantly less than midges on hosts with low densities. Resident midge density on hosts did not influence the likelihood of successful colonization by commensals and colonizers showed no preference for initial attachment site on hosts. However, colonizing midges emerged significantly less than resident midges. Similarly, successful emergers changed tube positions significantly less often as larvae relative to non-emergers. There was no difference in adult body weight of resident midges and colonists/movers, but adult males which emerged from commensal-laden (high density) hosts were significantly smaller than males from low density hosts. These data indicate larval density and movement behavior may have strong fitness consequences for commensal midges. 相似文献

Effect of cage population density on plasma corticosterone and peripheral lymphocyte populations of laboratory mice 总被引:3,自引:0,他引:3
The effect of different population densities of mice per cage on plasma corticosterone, peripheral lymphocytes and specific lymphocyte subpopulations was investigated. The animals were housed in groups of 2, 4 or 8 mice per cage and the blood samples were taken from each animal of these groups on days one, 7 and 14. A significant elevation (P less than 0.05) in plasma corticosterone concentration was observed in the group of 8 mice per cage on days one and 7 as compared with those of 2 or 4 mice per cage. The number of peripheral lymphocytes was significantly decreased in the groups of 2 (P less than 0.01) and 8 (P less than 0.05) mice per cage as compared with the group of 4 mice per cage on day one. A significantly decreased number of lymphocytes (P less than 0.01) in the group of 8 mice per cage continued to day 7. There were no significant differences in specific lymphocyte subpopulations observed among these groups. The results of this study suggest that a population density of 4 mice per cage induced minimal stress compared to that induced by the population densities of 2 or 8 mice per cage. Since stress is known to induce alteration in a variety of biological functions, the population density of mice per cage should be considered in the interpretation of research data. 相似文献

Dopamine (DA) in diencephalon was elevated in high density cages. 2. Peck order rank was assigned on the basis of feather distribution on head, neck and back and lesions on head and back. 3. Upper strata peck order rank was associated with significantly lower levels of brain regional NE and DA compared to lower strata peck order rank. 4. Animals reared at 18 degrees C exhibit enhanced innervation of brown adipose tissue (BAT) and greater cold tolerance as adults, yet gain more weight when fed an enriched diet compared with rats reared at 30 degrees C. To explore this paradox, sympathoadrenal activity was examined using techniques of [(3)H]norepinephrine ([(3)H]NE) turnover and urinary catecholamine excretion in male and female rats reared until 2 mo of age at 18 or 30 degrees C. Gene expression in BAT was also analyzed for several sympathetically related proteins. Although [(3)H]NE turnover in heart did not differ between groups, [(3)H]NE turnover in BAT was consistently elevated in the 18 degrees C-reared animals, even 2 mo after removal from the cool environment. Gene expression for uncoupling proteins 1 and 3, GLUT-4, leptin, and the alpha(1A)-adrenergic receptor was more abundant in BAT and the increase in epinephrine excretion with fasting suppressed in 18 degrees C-reared animals. These studies demonstrate that obesity consequent to exposure to 18 degrees C in early life occurs despite tonic elevation of sympathetic input to BAT. Diminished adrenal epinephrine responsiveness to fasting may play a contributory role. 相似文献

Rearing animals in small litters induces a permanent increase in body weight and body fat. To determine whether changes in sympathoadrenal activity contribute to this effect, litter size was adjusted the day after birth and maintained until weaning at 21 days. Sympathetic nervous system (SNS) activity was measured in adult animals using [(3)H]norepinephrine ([(3)H]NE) turnover in peripheral tissues. Although litter size was without effect on [(3)H]NE turnover in chow-fed animals, acceleration of [(3)H]NE turnover by dietary sucrose was completely abolished in heart and attenuated in interscapular brown adipose tissue and kidney of rats reared in small litters. Body and epididymal fat-pad weights were heavier in rats reared in small litters; however, weight gain in response to dietary enrichment with sucrose did not differ as a function of litter size. Thus litter size alters dietary activation of the SNS, and this effect presumably reflects changes in central nervous system regulation. 相似文献

Behavioral phenotypes of knockout mice are often interpreted as the effects of the absence of the gene product on adult behavior, yet behavioral differences among genotypes may be exaggerated or blurred by the postnatal environment. For example, mice develop in litters of varying sex ratios and genotypes, and it is possible that some of these behavioral differences may result from the composition of the litter. To determine whether these factors might play a role in the development of the behavioral characteristics that have become diagnostic of the knockout, offspring of parents heterozygous for a null mutation of estrogen receptor alpha (ERKO) were sexed and genotyped within 2 days of birth. Litters were then reconstituted, forming same-sex litters of equal numbers of ERKO and wild-type (WT) individuals that were tested in a standard resident-intruder paradigm. In this manner the effect of genotype would be evident without the potential confound of the presence of the opposite sex in the litter. Behavioral differences between the genotypes were more sharply defined than reported previously. ERKO females displayed only aggressive behavior whereas their WT littermates displayed only mounting behavior; both aggression and mounting behavior were greatly reduced in ERKO males. These data suggest that the postnatal environment such as litter composition may influence the development of sociosexual behaviors in ERKO mice. 相似文献

Exercise through perching has been suggested as a way to reduce the impact of leg problems in broiler chickens. It is possible that higher stocking densities may motivate birds to perch more, and perhaps reduce some of the detrimental effects to broiler health seen with high stocking densities. The goal of this research was to investigate the effects of density (10, 15, and 20birds/m(2)) and perch design on the frequency of perch utilization. Mixed sex broilers were assigned to 36 pens in a four perch treatmentxthree density factorial with three replications for each treatment/density combination in a randomized complete block design. Pens were assigned to one of four perch treatments: control (no perches), horizontal (three horizontal perches), angled (three 10 degrees angled perches), or mixed angle (one horizontal, one 10 degrees angled, and one 20 degrees angled perch), at each of the three densities. The results show that although the frequency of perch use was low, (2.6%+/-0.15), significant preferences were clear. Perching frequency was significantly higher when birds were raised at stocking densities of 15 (2.88+/-0.303) or 20birds/m(2) (2.76+/-0.211), as opposed to 10birds/m(2) (2.10+/-0.209) as indicated by a marginally significant main effect (p<0.0597), and a significant linear effect (P<0.0427). The birds used the horizontal perches the most, followed by 10 degrees angled and mixed angle perch treatments with decreasing frequency. An effect of age was found, in that perching increased during the first 4 weeks, and dropped off significantly at the end of the rearing period for all perch treatments. In general, perches closest to the hallway or outside of the house were used more than perches in the middle of the pen. Preferential use of the highest section of the 10 degrees angled perches was also found. The applied perch treatments or densities did not significantly affect final body weight or feed conversion. Percent mortality due to heat stress showed a significant increase with density, while mortality not caused by heat stress did not change with perch treatment or density. 相似文献

After injury, axons of the adult mammalian brain and spinal cord exhibit little regeneration. It has been suggested that axon growth inhibitors, such as myelin-derived Nogo, prevent CNS axon repair. To investigate this hypothesis, we analyzed mice with a nogo mutation that eliminates Nogo-A/B expression. These mice are viable and exhibit normal locomotion. Corticospinal tract tracing reveals no abnormality in uninjured nogo-A/B(-/-) mice. After spinal cord injury, corticospinal axons of young adult nogo-A/B(-/-) mice sprout extensively rostral to a transection. Numerous fibers regenerate into distal cord segments of nogo-A/B(-/-) mice. Recovery of locomotor function is improved in these mice. Thus, Nogo-A plays a role in restricting axonal sprouting in the young adult CNS after injury. 相似文献

Perinatal development represents a critical period in the life of an individual. A common cause of poor development is that which comes from undernutrition or malnutrition. In particular, protein deprivation during development has been shown to have deep deleterious effects on brain's growth and plasticity. Early‐life stress has also been linked with an increased risk to develop different psychopathologies later in life. We have previously shown that perinatal protein malnutrition in mice leads to the appearance of anxiety‐related behaviors in the adulthood. We also found evidence that the female offspring was more susceptible to the development of depression‐related behaviors. In the present work, we further investigated this behavior together with its molecular bases. We focused our study on the hippocampus, as it is a structure involved in coping with stressful situations. We found an increase in immobility time in the forced swimming test in perinatally malnourished females, and an alteration in the expression of genes related with neuroplasticity, early growth response 1, calcineurin and c‐fos. We also found that perinatal malnutrition causes a reduction in the number of neurons in the hippocampus. This reduction, together with altered gene expression, could be related to the increment in immobility time observed in the forced swimming test. 相似文献

Summary Young adult female mice were given a total of 90 intraperitoneal injections each of (methyl-3H)thymidine (3HTdR) at intervals of 8 h over 30 days to establish renewal rates of fibroblasts in various locations. Radioautographs prepared from punch biopsy material of the ears, taken repeatedly during the labeling procedure, revealed an approximately linear increase of labeling indices of dermal fibroblasts with time. Labeling indices of fibroblasts at the end of repetitive injections of 3HTdR differed depending on the site and/or type of connective tissues examined. Low values were obtained for fibroblasts in the leptomeninx (3.9%), the tracheal wall (4.4%), the achilles tendon (5.8%) and the dermis of the ear (6.5%), while higher values were registered for fibroblasts located in the lower half of the abdominal dermis (12.3%), peritendinous sheaths (12.6%), the interstitial connective tissue of the thigh muscles (12.9%), the submucosa of the colon (23%), the fibrous capsule of the adrenals (25.7%), and the upper half of the abdominal dermis (26%). These regional differences, with the exception of the skin and possibly of the tracheal wall, did not correlate with local temperature. Possible additional factors influencing fibroblast renewal rates may include the type of connective tissue, the degree of vascularization, mechanical stress and hormonal action. Estimated turnover times, based on the assumptions of a DNA synthesis time of 6 h or more and a linear increase of labeling indices as a function of time of repetitive labeling, range from about 90 to more than 700 days.
