昆虫谷胱甘肽S-转移酶分离纯化的新方法   总被引:4,自引:0,他引:4  
谷胱甘肽S-转移酶(glutathioneS-transferases,GST)是一类具有多种生理功能的同功酶.从蜡螟幼虫(Galeriamelonela)的提取液中分离纯化谷胱甘肽S-转移酶的基本方法如下:首先将冷冻的蜡螟幼虫在磷酸缓冲液中匀桨,经10000g和100000g分级离心;取上清液通过QAE-SephadexA-25离子交换柱层析除去部分色素和杂蛋白;然后采用谷胱甘肽-琼脂糖凝胶亲和层析(GSH-QT4),四溴酚酞二磺酸盐-琼脂糖凝胶亲和层析(BSP-QT4),铜离子-琼脂糖凝胶螯合层析(Cu2+-QT4)及PBE94-Sepharose(PBE94)聚焦层析等层析技术进一步分离纯化.将上述方法获得的色谱峰以CDNB和DCNB为底物检测生物活性.具有生物活性部分的蛋白质,通过SDS-PAGE测定其分子量.实验结果表明,采用GSH-QT4亲和层析法获得的活性峰,在SDS-PAGE图谱上呈现出两条带,分子量为24kD,24.5kD左右;Cu2+-QT6螯合层析法分离的活性峰,呈现出一条带,分子量为24kD左右;PBE94-聚焦层析法分离获得三个活性峰:第一色谱峰,呈现出一条带,分子量为23kD左右  相似文献

从皖南尖吻蝮蛇(Agkistrodonacutus)毒液中经DEAE-Sepharose和SephacrylS-200两步凝胶柱层析首次纯化出一种中分子量出血毒素(简称AaHⅣ).经SDS-PAGE和等电聚焦凝胶电泳测定其分子量为44kD,等电点为pH5.0.从500mg粗毒中可获得20mgAaHⅣ纯品.AaHⅣ有较强的出血活性,最小出血剂量(MHD)为0.4μg.  相似文献

采用经典的蛋白质色谱技术,纯化沙冬青( Ammopiptanthus mongolicus (Maxim .) Chengf.) 叶片中的抗冻蛋白质,然后经非变性PAGE胶分离、回收,得到具有热滞活性的两条带:B1 和B3。前者在8 g/L时热滞活性为0.46 ℃,并在SDS_PAGE胶上分离出两条带( 分子量为67 kD和21 kD) ;后者在10 g/L时热滞活性为0.45 ℃,在SDS_PAGE胶上只有1 条带( 分子量为39.8 kD)。B1 和B3 均不能被Shiff 试剂染色,也不具有典型糖蛋白的紫外吸收特征,因而可能不是糖蛋白  相似文献

用Kunkel突变法,将单链尿激酶型纤溶酶原激活剂(scu-PA)cDNA基因中编码Pro155—Lys158的片段定点突变,并将此突变的scu-PA(tscu-PA)的cDNA克隆到表达载体pCM-β-neo中,与pCM-dhfr共转染CHO/DHFR-细胞.获得的稳定表达株在无血清培养基中24h的表达量为620IU/106细胞.经锌离子螯合Sepharose亲和层析得到tscu-PA纯品.SDS-PAGE显示tscu-PA分子量为53kD左右,与预期的结果相符.tscu-PA是由凝血酶激活而不是由纤溶酶激活,但激活后也能转变为双链分子(tcu-PA).tscu-PA仍保持了scu-PA的血纤维蛋白亲和性.酶动力学研究表明,激活后的tscu-PA水解S2444的Km和Kcat值与高分子量尿激酶(HUK)相似.体外溶栓实验结果表明,tscu-PA可以选择性地溶解富含凝血酶的血凝块,对贫凝血酶的血凝块作用不大  相似文献

前曾发现在日本血吸虫虫卵中,存在一种新的能与亲和素专一性结合的蛋白质,称为亲和素结合蛋白(ABP)。在分离纯化ABP的基础上,用SDS-PAGE及SephadexG-150分别测定了ABP的分子量,并做了糖蛋白染色及等电,大测定。实验结果表明ABP是一个分子量为65kD的碱性糖蛋白,由数个相同的分子量为12.7kD的亚单位组成。SDS、β-巯基乙醇处理的ABP仍能与亲和素结合,提示ABP的亚单位能与亲和素结合。氨基酸修饰剂NAI、DTNB、NEM及NBS对ABP与亲和素的结合有不同程度的影响,而HNBB、TNBS及IAA对ABP与亲和素的结合无影响,提示ABP分子中氨基酸残基Tyr、Cyr是ABP与亲和素结合所必需的,可能参与结合位点的形成。另外,测得ABP与亲和素结合的结合常数为1.4×10~(-7)mol/L。  相似文献

用正常人胎肺细胞体外培养,从其培养液中分离纤溶酶原活化物(PA),通过CM-SephadexC-50层析,硫酸铵沉淀和Fibrin-Sepharose,Lysine-Sepharose亲和层析及SephadexG-50凝胶过滤等步骤,从10.5l条件培养液中分离纯化得到两种类型的纤溶酶原活化物,t-PA90μg,u-PA800μg.在还原条件下SDS-PAGE均显示单带,分子量t-PA为72kD,u-PA为54kD,纤溶比活分别为156000IU/mg蛋白和106000IU/mg蛋白.  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
