The results of comparative analysis of gene and structural mutations found in peripheral blood lymphocytes of inhabitants of Orel district areas contaminated with radionuclides as a result of Chernobyl accident are presented. The average level of 137Cs contamination in those areas ranged about 22-113 kBq/m2. In the study group was found the enhanced frequency of somatic cells with gene and structural mutations compared with laboratory control level by synchronous applying a T-cell receptor (TCR) loci mutation assay and cytogenetic analysis of unstable aberrations. The case-control comparison was carried out using the measured mutation frequencies and cases of various thyroid gland sickness recognized by ultrasonic examination. The cytogenetic assay did not show the statistical difference between healthy group and subjects with thyroid gland sickness. The average frequency of TCR loci mutation cells in the subjects with thyroid gland sickness was found to be statistically higher comparing with healthy persons. This finding was true for each study region and for Orel district in total. The subgroup of subject exposed in utero in 1986, soon after accident was analyzed. Both cytogenetic and TCR loci mutation assays shown enhancement of average mutation frequency in somatic cells in the subjects of this subgroup with thyroid gland sickness comparing with healthy persons.  相似文献

The frequency of lymphocytes with mutations at genes of T-cell receptor (TCR) is determined in 165 persons exposed to ionizing radiation 16-40 years after the influence. Depending on a type of irradiation and time, elapsed from the moment of exposure, all inspected persons were divided into 3 groups: 1) the analysis performed 16-40 years after acute irradiation; 2) the analysis--9-13 years after acute irradiation; 3) the analysis--9-13 years after prolonged irradiation. The persons with elevated frequencies of TCR-mutant cells were detected in all three groups. In the first group they have compounded 36% of persons inspected, in the second--25%, in the third--15%. The percentage of persons with elevated frequencies of mutant cells in all groups was significantly higher than in control groups. Taking into account fast enough elimination of radiation-induced TCR-mutant cells it is possible to suspect, that the cause of elevated frequency of mutant cells in the part of the persons can be genome instability.  相似文献

Results of multiyear cytogenetic study of children and teenagers living in areas, radioactive by contaminated after Chernobyl accident, were adduced. Mean density of 137Cs contamination in two compared living areas were 111 and 200 kBq/m2 and mean external doses accumulated for 1986-2001 were 6.7 and 11.4 mGy correspondingly. Averaged thyroid doses receives by subjects of all age groups in the second area were approximately 1.5 times higher than in the first area; in the youngest group (0-1 year) the doses were 114.3 and 174.3 mGy. During 17 years cytogenetic investigation approximately from 30% to 60% of examined persons were observed the increased level of chromosome aberrations in lymphocytes of peripheral blood. Average frequency of unstable aberrations (acentrics, dicentrics and centric rings) constituted about 0.4 per 100 cells (0.22 per 100 cells in controls) during all period of observation. Level of marker aberrations (dicentrics and centric rings) was increased almost all times of study and varied within 0.04-0.19 per 100 cell (0.03 in control group). The parallel investigation of frequency of stable aberrations by FISH method showed up their level about 3 times exceeding observed dicentrics level. Comparably higher indexes of cytogenetic disturbances were revealed in group exposed in utero during period of accident.  相似文献

A complex genetic examination of children which belong to two cohorts and their parents were carried out. The first cohort included children and constantly living on territories contaminated with radionuclides (Novozybkov district, Bryansk region). They were subdivided in groups according to the ontogenetic age periods of development of their parents at the time of the Chernobyl accident. In the children born in 1986-1995 the level of aberrant genomes is significantly higher as compared to the control (p < 0.001). In children born in 1998-2002 the differences are insignificant (p > 0.05). The frequency of aberrant genomes had a tendency to decrease with the period of time between the birth date of a child and the moment of the accident. Analysis of the results of cytogenetic investigation for the same living on territories with different densities of radioactive contamination (zone I-- 627-688 kBq/m2, 137Cs and zone II-- 135-402 kBq/m2, 137Cs) revealed insignificant differences in the spectrum and average frequencies of chromosome aberrations. The second cohort included children born in 1987-1991 and 1993-2002 from irradiated fathers (Chernobyl clean-up workers) and unirradiated mothers living on territories without radionuclide contamination. These children also displayed increased frequencies of aberrant genomes as compared to the control (p < 0.001). The analysis of the dynamics years of birth of cytogenetic disturbances in the same cohorts of children showed the average frequencies of aberrant genomes remain higher than the control level. In most of the children of both cohorts the repair synthesis of genome DNA by gamma- and UV-radiation is reduced as compared to one in the children from the control group.  相似文献

Frequency of lymphocytes mutant at T-cell receptor (TCR) loci was defined in 42 workers of nuclear chemical plants. In 11 persons mainly exposed to external radiation the mean frequency of TCR-mutant lymphocytes was statistically significant by higher compared with control group of unexposed donors: 9.1 x 10(-4) vs 3.5 x 10(-4) correspondently (p < 0.01). Frequency of TCR-mutant lymphocytes did not correlate neither the frequency of structural mutations non doses of external exposure. In group of workers exposed to combined external and internal radiation (n = 31) the average frequency of TCR-mutant lymphocytes was higher compared with control level: 8.9 x 10(-4) vs 3.5 x 10(-4) correspondently (p < 0.01). Correlations between the frequency of TCR-mutant cells and Pu content in organism (r = 0.5; p = 0.005) and between the frequency of chromosome aberration of unstable and stable types (r = 0.5; p = 0.002 and r = 0.6; p = 0.036, correspondently) were set. Comparison of results of analysis of structural and gene mutations allows us to supose that in case of external exposure the observed disturbances can result from genome instability in remote period after irradiation. In case of combined exposure the genetic changes were possibly caused by the constant action of alpha-radiation from Pu containing in the body.  相似文献

The purpose of this work was the study of possible relationship between intracellular NO level and somatic mutagenesis after irradiation with low doses. The level of NO in peripheral blood lymphocytes and frequency of the TCR-mutant cells were assessed by flow cytometry in 64 workers of atomic industry with mean dose (+/- SE) 114.9 +/- 10.8 MSV, accumulated within 21.4 +/- 1.1 years, and 66 age- matched control donors. The mean frequency of the TCR-mutant cells in this groups was (6.1 +/- 1.0) x 10(-4) and (4.1 +/- 0.2) x 10(-4) respectively (p = 0.06). 14% of workers of atomic industry had the TCR-mutant cell frequencies exceeding the 95% confidence interval in control donors. It was found the positive correlation between the intracellular NO level and the TCR-mutant frequency (R = 0.36, p < 0.01). The mean level of NO in individuals with the elevated TCR-mutant frequency was significantly higher than in others: 1619 +/- 57 vs 1340 +/- 40 relative units (p = 0.01). The results suggest that nitric oxide may come into elevating frequency of the mutant cells in some proportion of individuals exposed to low doses of ionizing radiation not excepting formation of genome instability.  相似文献

The paper deals with a comparative study of 137Cs contamination in forest, old arable and cultivated soils of Tula Region. Initial interception of Chernobyl derived 137Cs is higher in forest ecosystems: oak-forest > birch-forest > pine-forest > agricultural ecosystems. Vertical migration of 137Cs in deeper layers of soils was intensive in agricultural ecosystems: cultivated soils > old arable soils > birch-forest soils > oak-forest soils > pine-forest soils. In study have been evaluated spatial variability of 137Cs in soil and asymmetrical distribution, that is a skew to the right. Spatial heterogeneity of 137Cs in agricultural soils is much lower than in forest soils. For cultivated soil are determined the rate of resuspension, which equal to 6.1 x 10(-4) day(-1). For forest soils are described the 137Cs concentration in litter of different ecosystems. The role of main accumulation and barrier of 137Cs retain higher layers of soils (horizon A1(A1E) in forest, horizon Ap in agricultural ecosystems) in long-term forecast after Chernobyl accident.  相似文献

Eighteen Ukrainian evacuees from the Chernobyl exclusive zone, twenty one inhabitants of radioactively contaminated areas of Belarus and twelve control donors age-matched to the exposed persons were investigated 14-15 years after the Chernobyl accident for chromosomal aberration yields detected in blood lymphocytes by fluorescence in situ hybridisation technique. Unstable aberration yields measured in both Chernobyl cohorts were close to the background frequencies. Positive age-dependence trends in control donors were determined for the all type stable aberration levels. In evacuees the tendency for diminishing the difference between them and controls for stable aberration levels with persons' age increasing was found. The total stable chromosome exchange yields in evacuees 46-55 years old and inhabitants of areas with low contamination level didn't exceed the control values, but for younger evacuees and inhabitants of sufficiently contaminated regions the statistical increase above the age relevant background meanings was detected for this end-point. The advantages of using the FISH-detectable stable aberrations and particularly the total level of stable chromosome exchanges as the end-points for retrospective biological indication of past radiation exposure in Chernobyl cohorts were discussed.  相似文献

The nuclear reactor accident, which occurred on 26 April 1986 at Chernobyl, has been one of the greatest ecological disasters in human history. In our study we discussed the most recent data on the accident, and the natural and synthetic sources of radiation. According to the recent data, the air at Chernobyl had been contaminated with about 5300 PBq radionuclide activity (excluding rare gases), including 1760 PBq (131)I and 85 PBq (137)Cs. The highest radiation received by the liquidators (0.8-16 Gy), lower doses were received by the population which was evacuated or inhabited the contaminated areas (in which the level of (137)Cs activity deposited in the earth was 37 kBq/m(2)). In the European countries the highest mean radiation dose per year for the whole body in the first year after the accident was in Bulgaria (760 microSv), Austria (670 microSv) and Greece (590 microSv), while the lowest radiation dose was observed in Portugal (1.8 microSv) and Spain (4.2 microSv). In Poland the mean effective equivalent dose resulting from Chernobyl accident was 932 microSv and is close to the limited dose permitted in Poland, equalling 1 mSv/year. The highest radiation dose to thyroid was received by inhabitants of the states previously known as Bielskopodlaskie, Nowosadeckie and the north-east region of Poland. Lowest dose was received by inhabitants of the areas previously known as Slupski and Rzeszowski.  相似文献

On the basis of the metaphase analysis results, the peculiarities of dynamics of the genome mutation frequency (polyploid cells) were studied in bone marrow of bank voles inhabiting the areas with different radiocontamination level due to the Chernobyl accident (8-1526 kBq/m2 for 137Cs) in 1986-1991. Unexpectedly high frequencies of polypoid cells exceeding the pre-accidental level by a factor of 10(1)-10(3) were recorded in all populations studied. Relationship between the frequency of the parameter studied and the concentration of radionuclides incorporated in animal carcasses was proved. A statistically significant rise in the frequency of genome mutations with the time was revealed up to 1991, i.e. approximately to 12th post-accidental animal generation.  相似文献

Studies were carried out on wood samples collected in October 1997 from breast height of Scots pine trees (Pinus sylvestris) from site located 5 km south from the Chernobyl nuclear power plant. The radioactive contamination at the site was 3.7x10(5) kBq m(-2). These samples of secondary wood were used as an archive of information about the dynamics of a meristematic tissue cambium affected by ionising radiation from the Chernobyl reactor accident. The results show that frequency of the cambial cells events like anticlinal divisions, intrusive growth and cells elimination, was after the Chernobyl accident, about three times higher in comparison to preceding years. The most interesting finding was that after irradiation the length of tracheids increased. This increase is interpreted as an effect of intracambial competition among cells in the initial layer.  相似文献

Phytoplankton of reservoir R-11 was investigated at vegetative seasons of 2007 and 2008. R-11 is a locking reservoir in the cascade of Mayak PA reservoirs. The specific activity in water of R-11 ranged from 0.9 to 1.8 kBq/dm for 90Sr and from 0.98 to 3.97 kBq/dm3 for 137Cs; in bottom sediments it ranged from 20 to 400 kBq/kg for 90Sr and from 0.35 to 220 kBq/kg for 137Cs. Concentration of SO4- exceeded the maximum permissible concentration for fishing reservoirs in 5-7 times. The content of 90Sr in the phytoplankton has made 400 kBq/kg (for dry weight), and 137Cs--2.20 kBq/kg (for dry weight). The absorbed dose of irradiation for the phytoplankton calculated under the content of 137Cs and 90Sr in water and in phytoplankton (the external irradiation from activity of bottom sediments was not considered) has made 901 mGy per year. Altogether the phytoplankton of reservoir R-11 included 107 species. Such species of Cyanobacteriae as Oscillatoria agardhii Gom. was the dominant in phytoplanktonic community. Another Cyanobacteriae, Lyngbya limnetica Lemm. and Aphanizomenonflos-aquae (L.) Ralfs. acted the most important part in a complex of species-subdominants. According to quantitative development of the phytoplankton the reservoir R-11 can be classified as P-mezosaprobe waters.  相似文献

The decision of protein problem on contaminated by radionuclides territories as a result of the accident on Chernobyl NPP is not possible without increase of the sowing areas legumes crops (peas, lupine, soybean). Introduction in agricultural manufacture of grades with a low level of accumulation radionuclides (peas--Aist, Polesskay, Gomelskay Agat; soybean--Severnaya zvezda, Mageva) will allow to receive agricultural production adequate "Republican allowable levels of the contents of 137Cs and 90Sr in agricultural raw material and forages" on arable lands at their higher density of radioactive contamination: 137Cs--1300-1480 kBq/m2 (35.0-40.0 Ci/km2): 90Sr--18.5-37.0 kBq/m2 (0.50-1.00 Ci/km2).  相似文献

This study considers the exposure of the population of the most contaminated Gomel and Mogilev Oblasts in Belarus to prolonged sources of irradiation resulting from the Chernobyl accident. Dose reconstruction methods were developed and applied in this study to estimate the red bone-marrow doses (RBMs) from (i) external irradiation from gamma-emitting radionuclides deposited on the ground and (ii) 134Cs, 137Cs and 90Sr ingestion with locally produced foodstuffs. The mean population-weighted RBM doses accumulated during 35 years after the Chernobyl accident were 12 and 5.7 mGy for adult residents in Gomel and Mogilev Oblasts, respectively, while doses for youngest age groups were 20–40% lower. The highest mean area-specific RBM doses for adults accumulated in 1986–2021 were 63, 56 and 46 mGy in Narovlya, Vetka and Korma raions in Gomel Oblast, respectively. For most areas, external irradiation was the predominant pathway of exposure (60–70% from the total dose), except for areas with an extremely high aggregated 137Cs soil to cow’s milk transfer coefficient (≥?5.0 Bq L?1 per kBq m?2), where the contribution of 134Cs and 137Cs ingestion to the total RBM dose was more than 70%. The contribution of 90Sr intake to the total RBM dose did not exceed 4% for adults and 10% for newborns in most raion in Gomel and Mogilev Oblasts. The validity of the doses estimated in this study was assessed by comparison with doses obtained from measurements by thermoluminescence dosimeters and whole-body counters done in 1987–2015. The methodology developed in this study can be used to calculate doses to target organs other than RBM such as thyroid and breast doses. The age-dependent and population-weighted doses estimated in this study are useful for ecological epidemiological studies, for projection of radiation risk, and for justification of analytical epidemiological studies in populations exposed to Chernobyl fallout.

The western Bryansk region in south-western Russia was highly contaminated with 137Cs and 134Cs due to the Chernobyl accident in 1986. In 1990, a joint Nordic-Russian project was initiated in order to make measurements and estimates of the absorbed doses to selected groups of inhabitants in this area. The participating individuals were living in small villages with contamination levels between 0.9 and 2.7 MBq m–2. Only some villages had been decontaminated. Both schoolchildren and adults participated in the study and the number of persons was between 100 and 130 each year, residing in 5 villages. Every year in September–October, from 1990 to 1998, we performed individual measurements of external absorbed doses, assessed with thermoluminescent (TL) dosemeters (LiF). The mean effective dose per year from external irradiation due to the Chernobyl accident of the inhabitants in the villages ranged between 0.8 and 2.9 mSv during the study period and decreased with an apparent half-time of 3.7–8.2 years, depending on village and group. The highest individual doses within one village were, on average higher by a factor of 3 than the mean value for that village. Under the conservative assumption of a decrease rate in the external effective dose of 2% per year after 1998, individuals in the most highly exposed village are assumed to receive a life-time effective dose of about 75 mSv (between 1986 and 2056) from external exposure to caesium radionuclides. Cytogenetic monitoring was carried out on a group of children from Gomel (Belarus), one of the areas most severely affected by radioactive contamination following the accident at the Chernobyl nuclear power plant, in 1986. We performed the micronucleus test (MN) in binucleated lymphocytes of 42 children (mean age: 11+/-2.34 years), 16 of whom were affected by thyroid gland tumor. Thirty healthy children living in Pisa (mean age: 14.96+/-2.17 years) were enrolled in the study as controls. Thyroid tumor affected children living in Gomel showed a statistically significant increase in the frequency of micronucleated cells as compared with the healthy children from Pisa. Moreover, a significant correlation was found between MN frequency and both the presence of tumor and higher 137Cs contamination. In addition, higher 137Cs contamination was more frequently observed in tumor affected children. These results suggest that the increased MN frequency is attributable more to 137Cs contamination rather than to the presence of the tumor itself.  相似文献

Cytogenetic analysis using the fluorescence in situ hybridisation (FISH) technique was performed late time after the Chernobyl accident in groups of liquidators, evacuees from 30 km exclusive zone, residents of radioactively contaminated areas and control donors age-matched to exposed persons. Stable and unstable chromosome type exchanges were recorded using a hybrid conventional-PAINT nomenclature. The mean yield of stable chromosome exchanges in liquidators did not correlate with registered radiation doses but had a clear negative dependence on the duration of liquidators' staying in Chernobyl zone, that was in a good agreement with early data based on conventional dicentrics plus rings analysis. The overspontaneous excess for stable chromosome exchange level appeared to be higher in evacuees 16-40 years old than that of senior persons, whereas no age-dependent difference occurred for initially induced dicentrics plus rings yields in this cohort. The stable chromosome exchange yield, as well as combined yield of dicentrics plus rings and potentially unstable incomplete translocations in residents of radioactively contaminated areas showed a reasonable positive correlation with levels of 137Cs contamination. The observed yields of stable chromosome exchanges in all three exposed groups appeared to be somewhat lower than those of expected from unstable exchange-based doses which were referred to an in vitro dose response of stable exchanges outcome in human lymphocytes. Thus, FISH analysis can be successfully applied for qualitative cytogenetic indication of past and chronic radiation exposure to low doses but further refinement of FISH-based system for quantitative dose assessment is still required. Some practical approaches of solving this task are discussed.  相似文献

Cesium-137 (137Cs) is one of the most important nuclear fission elements that contaminated the environment after the explosion of the Chernobyl nuclear power plant in Ukraine (1986). The aim of the study was to compare the efficacy of two chelating agent, Prussian blue and apple-pectin on 137cesium decorporation in rats. Rats were intravenously injected with a solution of 137cesium (5 kBq per rat). Chelating agents, Prussian blue or apple-pectin were given immediately after Cs contamination and during 11 days by addition of each chelating agent in drinking water at a concentration corresponding to 400 mg kg(-1) day(-1). Efficiency was evaluated 11 days after contamination (at the end of treatment) through their ability to promote Cs excretion and to reduce the radionuclide accumulation in some retention compartments (blood, liver, kidneys, spleen, skeleton and in the remaining carcass). In these conditions after treatment with Prussian blue a fivefold increase in fecal excretion of Cs was observed and was associated with a reduction in the radionuclide retention in the main organs measured. In contrast, no significant differences were observed between untreated rats and rats treated with apple-pectin. These observations were discussed in terms of ability of pectins to bind Cs and compared to recently published results obtained after treatment of Cs-contaminated children with this chelate.  相似文献

The cytogenetic examination of a group of people, self-residing in ChNPP Exclusion Zone with soil contamination density 137Cs 74-477 kBq/m2, 90Sr 33-289 kBq/m2, 238, 239 + 240Pu 1.5-10.0 kBq/m2, was conducted in 1998-1999 and also 2001. It is shown, that after 12-15 years of the accident the level of chromosome damages in Zone self-residents' lymphocytes detected by a routine analysis is higher then at the residents of control Yagotin district, Kiev region and comes to a plateau. Lymphocytes with multiple chromosome damages are detected. The probability of its transuranium elements induction which are present in the Zone is discussed at present.  相似文献

The genetic effect of combined exposure to incorporated 137Cs and external gamma-irradiation was studied in germ cells of male mice. The activity of incorporated 137Cs was 3.7 x 10(4) Bq/g. The dose of external gamma-irradiation was 1.5 Gy. In the case of combined exposure, mice were treated with a cesium solution immediately after irradiation. The genetic effect was estimated by the frequency of dominant lethal mutations (DLM) induced at different stages of spermatogenesis. Upon combined exposure to external and internal irradiation, the frequency of DLM in premeiotic cells significantly exceeded the total frequency of DLM induced by separate exposure to external and internal irradiation; i.e., a marked synergistic interaction of external and internal irradiation was observed.  相似文献
