Interactive computerized morphometry was used for the quantitative study of the terminal airway branches (alveolar ducts) that followed the last bronchioles in three human acini. Two normal adult human lungs from the autopsy service were fixed by instillation and serial sections were prepared; three tissues blocks showing a central bronchiole were selected. Primary and secondary alveolar walls were traced and the following parameters were measured: volume, surface area (of primary and secondary septa), curvature (in equivalent radius) for branches of individual generations, and cumulative values starting with the first alveolar duct and moving peripherally. Although branching was dichotomous, we noticed considerable asymmetry in the pattern of branching and number of side branches. The branching trees of alveolar ducts that we studied comprised 6,7, and 10 generations. The average volume of ducts was 0.04-0.13 mm3, the surface area of primary walls ranged from 0.3616 to 0.7931 mm2 and of secondary septal walls from 0.0100 to 0.0647 mm2. The equivalent radius of curvature was between 22.7 and 38.1 microns. Cumulative increases of volume and surface area revealed similarity in the first five generations. Secondary walls represented only 4% (or 8% if 2 sides are considered) of the primary surface area, strengthening the view that alveoli are incompletely developed side chambers secondary to the alveolar ducts.  相似文献

A three-compartment model predicting the recovery of aerosol boli (i.e., the ratio of the number of particles expired to the number inspired) as a function of breath-holding time and bolus penetration was fitted to experimental data measured in nine isolated dog lungs. For each lung, the diameters of alveoli and alveolar ducts, as well as the volume fractions of alveoli, alveolar ducts, and airways, were determined as parameters providing the best fit. Parameter values were alveolar diameter = 0.116 +/- 0.007 (SE) mm, alveolar duct diameter = 0.284 +/- 0.015 mm, total alveolar volume/total lung capacity (TLC) = 0.68 +/- 0.02, total alveolar duct volume/TLC = 0.24 +/- 0.02, and total airway volume/TLC = 0.09 +/- 0.01. These values agreed with published values for linear dimensions and volumetric fractions in the canine lung. The mean alveolar diameter determined by the model in the nine lungs agreed closely with a mean value of 0.115 +/- 0.002 mm determined by morphometric analysis of photographs of the subpleural alveoli in the same lungs. The procedure of fitting the model to experimental data appears to have promise as a noninvasive probe of the lung periphery. However, aerosol-derived dimensions were more variable than morphometric ones, possibly because of interlung differences in aerosol distribution not accounted for in the model.  相似文献

Despite decades of research into the mechanisms of lung inflation and deflation, there is little consensus about whether lung inflation occurs due to the recruitment of new alveoli or by changes in the size and/or shape of alveoli and alveolar ducts. In this study we use in vivo (3)He lung morphometry via MRI to measure the average alveolar depth and alveolar duct radius at three levels of inspiration in five healthy human subjects and calculate the average alveolar volume, surface area, and the total number of alveoli at each level of inflation. Our results indicate that during a 143 ± 18% increase in lung gas volume, the average alveolar depth decreases 21 ±5%, the average alveolar duct radius increases 7 ± 3%, and the total number of alveoli increases by 96 ± 9% (results are means ± SD between subjects; P < 0.001, P < 0.01, and P < 0.00001, respectively, via paired t-tests). Thus our results indicate that in healthy human subjects the lung inflates primarily by alveolar recruitment and, to a lesser extent, by anisotropic expansion of alveolar ducts.  相似文献

The microvascularization of the bile duct was studied in 20 adult guinea pigs (Cavia porcellus) using microvascular corrosion casts in the scanning electron microscope. The main supplying and draining vessels are located in the adventitial layer; from there they form the subepithelial capillary network. The microangioarchitecture of the common bile duct is reminiscent of that of the cystic and hepatic ducts. The diameter of these ducts is 1,000 microns on average. The microvascular arrangement in the bile duct suggests a probable involvement of the capillaries in the absorption of bile hormones.  相似文献

The geometry and morphometry of intraacinar airways in rat and rabbit lungs were studied from silicone rubber casts. Acini, defined as the complex of alveolated airways distal to the "terminal" bronchiole, were trimmed off the bronchial tree. In both species, the acinar volume followed a log-normal distribution over a range in size of one order of magnitude. At an inflation level of 60% total lung capacity, their mean volume was 1.86 mm3 in the rat and 3.46 mm3 in the rabbit. On a representative sample of acini of different volumes, the branching pattern was characterized as irregular dichotomy, and the segment length and inner and outer diameters were measured. The average acinus had a mean of six generations in the rat and seven in the rabbit. Both showed a decrease in segment length and inner diameter with each generation. The mean longitudinal pathway length--that is, the distance from the initial acinar segment to the terminal sacs--was found to depend on the cube root of the acinar volume in both species. It was calculated at 1.46 and 1.95 mm for rat and rabbit, respectively.  相似文献

OBJECTIVE--To determine the efficacy of peroral electrohydraulic lithotripsy performed with an extra large duodenoscope (outside diameter 14.8 mm) and a choledochoscope with a diameter of 4.1 mm (Olympus "mother and baby" endoscope system) in the removal of very large stones from the common bile duct. DESIGN--Prospective study of patients with giant stones in the common bile duct that were resistant to extraction by conventional means. SETTING--Endoscopy unit at a university hospital. PATIENTS--Four women and one man aged 48-82 (mean 66.4 years) with a total of nine stones in their common bile ducts ranging from 2.2 to 3.6 cm in diameter. INTERVENTIONS--Peroral electrohydraulic lithotripsy was performed after intravenous sedation and under antibiotic cover. Two endoscopists took part in each procedure, coordination being achieved by means of a video monitor. The procedures were performed with a Lithotron EL-23 lithotripter and a 3 French lithotripsy probe inserted through the choledochoscope under direct vision. MAIN OUTCOME MEASURE--Complete clearance of the common bile duct confirmed by occlusion cholangiography. RESULTS--All nine stones (mean minimal diameter 2.6 cm; mean maximal diameter 3.1 cm) were successfully fragmented by electrohydraulic lithotripsy, allowing subsequent extraction with the aid of endoscopy and clearance of the common bile duct. A median of three (range two to five) sessions of endoscopic retrograde cholangiopancreatography were required to achieve complete clearance of the ducts. Patients stayed a median of eight days in hospital after lithotripsy (range eight to 14). There were no complications. CONCLUSION--Peroral electrohydraulic lithotripsy offers a safe and effective alternative for the management of patients with large stones in the common bile duct.  相似文献

We tested the hypothesis that economy and efficiency are independent of length in intact cardiac muscle over its normal working range. We measured force, force-time integral, force-length area, and myocardial oxygen consumption in eight isometrically contracting rabbit right ventricular papillary muscles. 2,3-Butanedione monoxime was used to partition nonbasal oxygen consumption into tension-independent and tension-dependent components. Developed force, force-time integral, and force-length area increased by factors of 2.4, 2.7, and 4.8, respectively, as muscle length was increased from 90% to 100% maximal length, whereas tension-dependent oxygen consumption increased only 1.6-fold. Economy (the ratio of force-time integral to tension-dependent oxygen consumption) increased significantly with muscle length, as did contractile efficiency, the ratio of force-length area to tension-dependent oxygen consumption. The average force-length area-nonbasal oxygen consumption intercept was more than the twice tension-independent oxygen consumption. We conclude that economy and efficiency increase with length in rabbit myocardium. This conclusion is consistent with published data in isolated rabbit and dog hearts but at odds with studies in skinned myocardium.  相似文献

The tropical arboreal environment is a mechanically complex and varied habitat. Arboreal inhabitants must adapt to changes in the compliance and stability of supports when moving around trees. Because the orangutan is the largest habitual arboreal inhabitant, it is unusually susceptible to branch compliance and stability and therefore represents a unique animal model to help investigate how animals cope with the mechanical heterogeneity of the tropical canopy. The aim of this study was to investigate how changes in compliance and time of oscillation of branches are related to easily observable traits of arboreal supports. This should help predict how supports react mechanically to the weight and mass of a moving orangutan, and suggest how orangutans themselves predict branch properties. We measured the compliance and time of oscillation of branches from 11 tree species frequented by orangutans in the rainforest of Sumatra. Branches were pulled at several points along their length using a force balance at the end of a stiff rope, and the local diameter of the branch and the distance to its base and tip were measured. Compliance was negatively associated with both local diameter and length to the tip of the branch, and positively, if weakly, associated with length from the trunk. However, branch diameter not only predicted compliance best, but would also be easiest for an orangutan to observe. In contrast, oscillation times of branches were largely unaffected by local diameter, and only significantly increased at diameters below 2 cm. The results of this study validate previous field research, which related locomotory modes to local branch diameter, while suggesting how arboreal animals themselves sense their mechanical environment.  相似文献

Abstract The ultrastructure of unicellular accessory glands (= prostate glands) and external male ducts of the cestode Cylindrotaenia hickmaniare described. Accessory glands open into the lumen of the external common sperm duct (= external vas deferens). The gland cells contain abundant endoplasmic reticulum, Golgi bodies and secretory bodies, and have elongate necks that pierce the apical cytoplasm of the duct. Cell contact with the apical cytoplasm of the sperm duct is mediated by septate desmosomes. Accessory glands secrete spherical particles, with a diameter of approximately 70 nm, that adhere to spermatozoa. The roles of these accessory glands may relate to activity of the sperm or development of the female system after insemination. Paired sperm ducts arise from testes, and unite to form a common sperm duct. Each duct consists of a tubular anucleate cytoplasmic region which is supported by nucleated cytons that lie sunken in the parenchyma. The apical cytoplasm of the paired sperm ducts (= vasa efferentia) possesses apical microvilli and abundant mitochondria, but few other cytoplasmic features. The apical cytoplasm of the common sperm duct possesses sparse apical microvilli and numerous electronlucent vesicles. The male gonoducts form an elongate syncytium which is markedly polarized along the length of the ducts. The ducts also display apical–basal polarity in that sunken nucleated cytons support the apical cytoplasm which in turn has distinct basal and apical domains.  相似文献

The secretory endpieces of the rabbit submandibular gland are unusual in that they consist of seromucous acini (not demilunes) that empty into serous tubules that in turn drain into intercalated ducts. Seromucous granules consist of a moderately dense spherule in a fibrillogranular matrix. Serous granules contain a feltwork of filaments, which are liberated as a tangled skein during exocytosis. Peculiar granulated cells that have secretory granules of complex morphology are present at each end of the serous tubules. Intercalated ducts are, cytologically speaking, relatively simple, but the duct cells may contain a few oblong secretory granules. Striated ducts are typical in structure, although postfixation with ferrocyanide-reduced osmium reveals significant amounts of glycogen in the basal processes. Modified mitochondria are present in striated duct cells, but their frequency varies from rabbit to rabbit. Such mitochondria contain either an array of parallel, rigid cristae linked by intermembranous bridges, or a bundle of helical filaments within an expanded crista. Interspersed with the striated duct cells, especially near the duct origin, are some highly vacuolated cells with sparse mitochondria. Excretory ducts consisting of stratified columnar (sometimes pseudostratified) epithelium often show bleb formation of the luminal surface of the tall cells.  相似文献

Mesonephric and paramesonephric ducts develop in different ways in male and female fetuses. We have analyzed the changes in the expression of cytokeratin and vimentin type of intermediate filaments and desmosomal plaque proteins in progressing and regressing genital ducts of rat fetuses. The concomitant changes in the basement membranes were detected by laminin antibody. Epithelial cells of the indifferent (Day 15) male and female mesonephric and paramesonephric ducts contained faint vimentin positivity which, however, later disappeared. Indifferent mesonephric duct epithelium stained strongly for cytokeratin, whereas in the corresponding paramesonephric duct only a weak and spotty positivity was seen. Immunocytochemical localization of cytokeratin filaments and desmosomal plaque proteins correlated with the ultrastructural differences in the apical junctional complexes of the mesonephric and paramesonephric ducts. Regardless of the ongoing regression of the male paramesonephric duct, cytokeratin positivity increased in the disorganizing epithelium; the most weak and a granular immunoreaction was seen in the cells found in the intensively vimentin-positive periductal mesenchyme. In the regressing female mesonephric duct cytokeratin positivity was lost before the final dissolution of the basement membrane. Immunoblotting analysis of cytokeratin and vimentin polypeptides of the individual genital ducts were in agreement with the immunocytochemical results obtained in 15- and 16-day-old fetuses. The results suggest that the expression of vimentin type intermediate filaments is an indication of the mesothelial origin of the genital ducts. The increase in cytokeratin positivity of the regressing paramesonephric duct epithelium suggests that the degenerative changes are initiated by the mesenchyme. Cytokeratin-positive cells found in the periductal mesenchyme of the male paramesonephric duct may be epithelial cells transforming into mesenchyme. The results emphasize a close relationship between the changes of the intermediate filament system and extracellular matrix upon differentiation of the fetal genital ducts.  相似文献

We used pharmacological methods to test the hypothesis that female Dermacentor variabilis salivary ducts dilated when dopamine-stimulated and that dilation was nitric oxide-mediated. Stimulation with dopamine resulted in an increased diameter (19.7%) compared to unstimulated ducts (P<0.005). Pretreatment with L-NAME, an inhibitor of nitric oxide synthase, or cytochalasin D abolished the dilation. Addition of L-arginine to L-NAME-treated ducts partially restored the ability to dilate. A cuticular coil composed of a series of concentric rings ran the length of the duct adjacent to the epithelial cell layer. In stimulated ducts, the center-to-center periodicity of these rings increased from 0.59 μm in unfed ducts to 1.0 μm from partially fed ducts (P<0.05). When the ducts from partially fed females were stimulated with dopamine, the periodicity increased further to 1.75 μm (P<0.05), suggesting the coils moved further apart in response to stimulation. Prominent folds lining the lumen of unstimulated ducts were less pronounced in stimulated preparations, suggesting that the cuticle stretches, thereby increasing lumen size. Actin was localized in epithelial cells as a honeycomb pattern that we suggest links the epithelial cells to the rings. Together, these data support the following hypothesis: stimulated ducts dilated during fluid production; dilation involved an actin-based system, and was mediated by nitric oxide. Dilation of the duct may enhance its role as a reservoir for saliva produced by the acini during the period between imbibition and salivation.  相似文献

To determine alveolar pressure-volume relationships, alveolar three-dimensional reconstructions were prepared from lungs fixed by vascular perfusion at various points on the pressure-volume curve. Lungs from male Sprague-Dawley rats were fixed by perfusion through the pulmonary artery following a pressure-volume maneuver to the desired pressure point on either the inflation or deflation curve. Tissue samples from lungs were serially sectioned for determination of the volume fraction of alveoli and alveolar ducts and reconstruction of alveoli. Alveoli from lungs fixed at 5 cmH2O on the deflation curve (approximating functional residual volume) had a volume of 173 X 10(3) microns3, a surface area of 11,529 microns2, a mouth opening diameter of 72.7 microns, and a mean caliper diameter of 91.8 micron (SE). Alveolar shape changes during deflation from total lung capacity to residual volume was first (30 to 10 cmH2O) associated with little change in the diameter of the alveoli (102.7 +/- 2.4 to 100.3 +/- 3.3 microns). In the range overlapping normal breathing (10 to 0 cmH2O) there was a substantial decrease in diameter (100.3 +/- 3.3 to 43.3 +/- 2.3 microns). These measurements and others made on the relative changes in the dimensions of the alveolus suggest that the elastic network, particularly around the alveolar ducts, are predominant in determining lung behavior near the volume expansion limits of the lung while the elastic and surface tension properties of the alveoli are predominant in the volume range around functional residual capacity.  相似文献

Transdiaphragmatic pressure is a result of both tension in the muscles of the diaphragm and curvature of the muscles. As lung volume increases, the pressure-generating capability of the diaphragm decreases. Whether decrease in curvature contributes to the loss in transdiaphragmatic pressure and, if so, under what conditions it contributes are unknown. Here we report data on muscle length and curvature in the supine dog. Radiopaque markers were attached along muscle bundles in the midcostal region of the diaphragm in six beagle dogs of approximately 8 kg, and marker locations were obtained from biplanar images at functional residual capacity (FRC), during spontaneous inspiratory efforts against a closed airway at lung volumes from FRC to total lung capacity, and during bilateral maximal phrenic nerve stimulation at the same lung volumes. Muscle length and curvature were obtained from these data. During spontaneous inspiratory efforts, muscle shortened by 15-40% of length at FRC, but curvature remained unchanged. During phrenic nerve stimulation, muscle shortened by 30 to nearly 50%, and, for shortening exceeding 52%, curvature appeared to decrease sharply. We conclude that diaphragm curvature is nearly constant during spontaneous breathing maneuvers in normal animals. However, we speculate that it is possible, if lung compliance were increased and the chest wall and the diameter of the diaphragm ring of insertion were enlarged, as in the case of chronic obstructive pulmonary disease, that decrease in diaphragm curvature could contribute to loss of diaphragm function.  相似文献

The dorsal foot skin supplied by the arteria dorsalis pedis the dorsal venous arch, the peroneal sensory nerves and the musculus extensor digitorum brevis is a very good myocutaneous flap. The material on which the study was carried out, consisted of 20 feet from standard cadavers, injected with a mixture of terebenthene and minium through the arteria tibialis anterior. The m. extensor digitorum brevis is 6.1 cm long, 1.7 cm wide, 3.9 mm thick. It is mainly supplied by the a. dorsalis pedis and its branches: the a. tarsea dorsalis (constant) and the a. metatarsea dorsalis (12 of 20 specimens). The average diameter of the a. dorsalis pedis at the upper limit of the m. extensor retinaculum was 2.14 mm and this was chosen as the most proximal limit of the dorsalis pedis flap. The a. tarsea dorsalis was present in all the specimens, with a diameter of 0.95 mm at its origin and a length of 35 mm. On average, this artery divided into four branches to the m. dorsalis pedis. The a. metatarsea dorsalis was present in 12 of 20 specimens, with an average diameter of 0.53 mm and a length of 22 mm. On average, this artery divided into three branches to the m. dorsalis pedis. We drew three lines in the proximal, middle and distal third of each flap design and calculated the sum of arterial branch sections with our lines. We think this provides a reasonable indication of the comparative richness of the cutaneous blood supply in the flap. The mean number of cutaneous branches was 10 in the proximal third, 6.7 in the middle third (13 if branches supplying the m. extensor pedis brevis are included) and 5 in the distal third. The myocutaneous dorsalis pedis arterialized flap can be safely used as an island flap to cover the ankle or heel and as a free flap for palm defects.  相似文献

从我国6个凤丹(Paeonia ostii T.Hong et J.X.Zhang)中心产区,选取15个群体共398个单株,对20个表型性状进行调查和分析。结果显示,枝条数、地径、株高、茎长、冠幅长、冠幅宽、成花枝、芽位数、叶长和叶宽等10个表型性状受植株株龄的影响显著,随着株龄增加,除叶长和叶宽的平均值呈递减趋势外,其他8个性状的平均值都呈现递增趋势。凤丹群体间的表型分化系数为0~50.14%,平均值为27.62%,变化幅度最大的是二年生枝长,最小的为心皮数和二年生枝径。对当年生枝长、芽位高、当年生枝径、顶生小叶长、二年生枝径、顶生小叶宽、复叶数、心皮数、二年生枝长和叶柄长等不受株龄影响的10个表型性状进行主成分分析,结果显示前6个性状是影响表型差异的主要性状。各表型性状间存在一定的相关性,聚类分析结果将受株龄影响的10个性状划分为4组。研究结果表明凤丹表型性状在群体间和群体内的变异非常丰富,可为遗传改良和品种选育储备丰富的植物材料。  相似文献

We used Monte Carlo simulations to investigate the conformational and thermodynamic properties of DNA molecules with physiological levels of supercoiling. Three parameters determine the properties of DNA in this model: Kuhn statistical length, torsional rigidity and effective double-helix diameter. The chains in the simulation resemble strongly those observed by electron microscopy and have the conformation of an interwound superhelix whose axis is often branched. We compared the geometry of simulated chains with that determined experimentally by electron microscopy and by topological methods. We found a very close agreement between the Monte Carlo and experimental values for writhe, superhelix axis length and the number of superhelical turns. The computed number of superhelix branches was found to be dependent on superhelix density, DNA chain length and double-helix diameter. We investigated the thermodynamics of supercoiling and found that at low superhelix density the entropic contribution to superhelix free energy is negligible, whereas at high superhelix density, the entropic and enthalpic contributions are nearly equal. We calculated the effect of supercoiling on the spatial distribution of DNA segments. The probability that a pair of DNA sites separated along the chain contour by at least 50 nm are juxtaposed is about two orders of magnitude greater in supercoiled DNA than in relaxed DNA. This increase in the effective local concentration of DNA is not strongly dependent on the contour separation between the sites. We discuss the implications of this enhancement of site juxtaposition by supercoiling in the context of protein-DNA interactions involving multiple DNA-binding sites.  相似文献

The effect of age on lung structure in male BALB/cNNia inbred mice   总被引:5,自引:0,他引:5  
Morphometric examination using light, scanning electron, and transmission electron microscopy was performed on the lungs from 32 inbred male BALB/ cNNia mice between 38 days and 28 months of age. Between 38 days and 9 months of age the changes were primarily those of growth. Alveolar multiplication and total elastic-fiber length appeared complete by 38 days of age. The major increase in the number of interalveolar pores occurred by 68 days, but there was a significant further increase from 68 days to 9 months of age. At 9 months, approximately 10% of the alveolar wall was formed by pores. Alveolar ducts, the cylindrical core of air central to alveolar mouths, increased more in diameter than length. Between 9 and 28 months the changes were attributed to aging and were different from those reported in humans and other species. Lung volume, alveolar surface area, and total volume of alveolar wall increased with age; there was no change in mean linear intercept and volume proportion of alveolar and alveolar duct air. Total area of pores increased with age, but their number and area fraction of the alveolar wall did not change. No transmission electron microscopic changes were seen in the alveolar walls. We speculate that the morphometric differences between our animals and those studied in other reports may reflect the fact that our animals were specific-pathogen-free animals and kept under protected circumstances.  相似文献
