Summary Maximum volumetric productivities of biomass (1.40 gl–1h–1) and lactic acid (8.93 gl–1h–1) for a continuous culture ofLactobacillus delbreuckii occurred between dilution rates 0.35h–1 and 0.40h–1. All major nutrients were in excess in these cultures. Glucose utilisation was complete at dilution rates of 0.1h–1 and lower. Product and biomass yields were constant in the dilution rate range studied (0.05h–1 to 0.50h–1).  相似文献

Summary Batch and continuous two-stage cultures have been conducted in order to determine the effect of yeast extract (YE) on the homolactic fermentation of whey permeate byLactobacillus helveticus. Supplementation with YE had a significant effet on lactic acid concentration, volumetric productivity, and substrate conversion, but not on lactic acid yield. Volumetric productivity in the first stage increased from 2 to 9 g l–1 per hour by increasing the YE concentration from 1.5 to 25 g l–1 At the same time conversion improved from 22% to 93% at a dilution rate of 0.2 h–1. The second stage demonstrated the effect of YE at a lower dilution rate (0.14 h–1. A high system conversion (97%) and a high final lactic acid concentration (40 g l–1) were achieved with 10 g l–1 YE.  相似文献

Summary A new method of d(–)lactic acid production based on the aggregated form of growth of Bacillus laevolacticus in continuous culture in an anaerobic gaslift reactor is presented. With glucose as the substrate a bacterial dry weight of 25 g·1–1 and a lactic acid production rate of 13 g·1–1·h–1 was attained. In conventional glucose-limited chemostat cultures elevated levels of lactic acid stimulated specific lactic acid production while the formation rates of other end-products remained unaffected. In glucose-limited aggregated cultures lactic acid positively influenced the aggregation of cells, improving the volumetric production rate. It is concluded that lactic acid itself is a positive effector in the optimisation of lactic acid production with aggregated cultures of B. laevolacticus.Offprint requests to: J. P. de Boer  相似文献

Summary The influence of different operational parameters, such as the dilution rate (D) and the bleeding rate (B), in the production of a flocculent strain ofLactobacillus plantarum was studied. The effect of the dilution rate was demonstrated to be related to the lactic acid concentration inside the reactor. The effect of the bleeding rate was shown to be critical in the stabilization of the operation (due to a better pH control). It also allowed a continuous recovery of cells outside the reactor. Summary The ability of Lactobacillus acidophilus to aggregate, to produce lactic acid for a long term continuous fermentation process and to exist as aggregate cell cultures in a gas-lift reactor under aerobic conditions was studied. The main product of fermentation was lactic acid and only the traces of other end-products were determined. The highest fermentation efficiency of lactic acid was 98.6% and the highest productivity was 9.6 g.l–1.h–1 of lactic acid.  相似文献

The outdoor production of Monodus subterraneus wasstudied in bubble column and helical reactors, mainly analysing the influenceofdilution rate, air flow rate and solar irradiance on growth rate andbiochemicalcomposition. Photoinhibition and photo-oxidation phenomena were also analysed.The cultures were stressed at high solar irradiance and dissolved oxygenconcentrations. A clear relationship between stress of the cultures and thefluorescence from PSII measurements was observed, the Fv/Fm ratio being lowerinthe helical reactor than in the bubble column. Growth rate and biomassproductivity were both a function of the average irradiance and the Fv/Fmratio;maximum values of 0.040 h–1 and 0.54 gL–1 d–1 were measured. The influenceofphotoinhibition and average irradiance was modelled, the model also fitting theexperimental data reported by another author. The chlorophyll contenthyperbolically decreased, whereas the carotenoid content decreased linearlywiththe average irradiance. The higher the dilution rate the higher the protein andcarbohydrate content of the biomass, and the lower the lipid content. Theeicosapentaenoic acid (EPA) content ranged from 2.3 to 3.2% d.wt, the higherthe dilution rate, the lower EPA content, although the higher the EPAproportion. Maximum EPA productivity was only 9 mg L–1d–1, due to the stress to which the cultures wereexposed.  相似文献

Summary Growth of Propionibacterium acidi-propionici was studied on lactose as substrate and in acid whey permeate in a three-electrode poised-potential system with cobalt sepulchrate as artificial electron donor. In batch culture experiments in a stirred-tank reactor the substrate was fermented completely to propionic acid up to 6.5 g 1–1 lactose in a supplemented whey permeate medium. No acetic acid was produced during the growth of P. acidi-propionici. An electron flow of 80–100 mA was obtained and the electron balance was 101%. In continuously growing cultures with 3 g 1–1 of lactose as the substrate, propionate was formed as the only fermentation product up to a dilution rate (D) of 0.04 h–1. With D>0.04 h–1 the bacteria immobilized on the working electrode surface. It was examined whether an electron transfer occurred between the platinum working electrode and the immobilized cells. Summary In a mixed culture of Saccharomycopsis fibuliger Y76 and Streptococcus lactis 65.1 on starch the main interactions are commensalism and competition. Oxygen-limited batch and continuous cultures of S. fibuliger showed accumulation of sugars. When oxygen was used as an external regulatory parameter in the mixed culture lactic acid, acetic acid and formic acid were formed. Ethanol produced by S. lactis was most likely assimilated by S. fibuliger. Continuous mixed cultures were stable under conditions of oxygen limitation at the dilution rates tested (0.10 h–1 and 0.15 h–1). Conversion yields of 35 to 40% were obtained but may be improved.  相似文献

Lactic acid was added to batch very high gravity (VHG) fermentations and to continuous VHG fermentations equilibrated to steady state with Saccharomyces cerevisiae. A 53% reduction in colony-forming units (CFU) ml–1 of S. cerevisiae was observed in continuous fermentation at an undissociated lactic acid concentration of 3.44% w/v; and greater than 99.9% reduction was evident at 5.35% w/v lactic acid. The differences in yeast cell number in these fermentations were not due to pH, since batch fermentations over a pH range of 2.5–5.0 did not lead to changes in growth rate. Similar fermentations performed in batch showed that growth inhibition with added lactic acid was nearly identical. This indicates that the apparent high resistance of S. cerevisiae to lactic acid in continuous VHG fermentations is not a function of culture mode. Although the total amount of ethanol decreased from 48.7 g l–1 to 14.5 g l–1 when 4.74% w/v undissociated lactic acid was added, the specific ethanol productivity increased ca. 3.2-fold (from 7.42×10–7 g to 24.0×10–7 g ethanol CFU–1 h–1), which indicated that lactic acid stress improved the ethanol production of each surviving cell. In multistage continuous fermentations, lactic acid was not responsible for the 83% (CFU ml–1) reduction in viable S. cerevisiae yeasts when Lactobacillus paracasei was introduced to the system at a controlled pH of 6.0. The competition for trace nutrients in those fermentations and not lactic acid produced by L. paracasei likely caused the yeast inhibition.  相似文献

Summary Continuous and batch cultures of Lactobacillus helveticus operated under different conditions were studied with respect to the limitation of growth and lactic acid production by increasing undissociated lactic acid and hydrogen ion concentrations, respectively. In a single-stage continuous culture without pH control a final pH of 3.8 and 65 mm undissociated lactic acid was obtained. In two-stage continuous cultures provided with different growth media and run at different pH values, 65–70 mm free acid was obtained in the second stage. Further batch-culture experiments showed growth limitation at 60–70 mm lactic acid. After growth ceased, production of lactate continued until a lactic acid concentration of about 100 mm was reached; obviously an uncoupling of growth and acid production had occurred. Examining the effect of different concentrations of either lactic acid or hydrochloric acid, added to growing batch cultures of L. helveticus, it was shown that the undissociated lactic acid concentration was responsible for growth limitation and lactic acid production in this organism, whereas the pH value had only an indirect effect.  相似文献

In aerobic enrichment experiments with a chemostat, using phosphate-limited lactate medium, aSpirillum sp. predominated at the lower range of dilution rates. At the higher dilution rates an (chemoorganotrophic) unidentified rod-shaped bacterium came to the fore. The same result was obtained in competition experiments with pure cultures of the two bacteria. Growth parameters were: Rod, max=0.48 hr–1,k s(PO4 3–)=6.6×10–N M;Spirillum, max=0.24 hr–1· ks(PO4 3–) =2.7×10–8 M. TheSpirillum grew faster than the rod at low dilution rates, not only under phosphate-limitation but also in K+-,Mg2+-, NH4 +-, aspartate-, succinate-, and lactate-limited cultures. Both organisms showed little substrate specificity and could utilize a similar range of carbon and energy sources. The results support the view that part of the diversity among bacteria in the natural environment is based on selection toward substrate concentration. Another set of competition experiments was carried out with pure cultures of two marine obligately chemolithotrophic colorless sulfur bacteria,Thiobacillus thioparus andThiomicrospira pelophila. Tms. pelophila outgrewT. thioparus at low dilution rates under iron limitation, while the reverse was true at high dilution rates. It is concluded that the relatively fast growth ofTms. pelophila at low iron concentration may explain its higher sulfide tolerance. Organisms showing a selection advantage at very low concentrations of limiting substrates appear to have a relatively high surface to volume ratio.  相似文献

Summary Pseudomonas paucimobilis was isolated from a consortium which was capable of degrading dicamba (3,6-dichloro-2-methoxybenzoic acid) as the sole source of carbon. The degradation of dicamba byP. paucimobilis and the consortium was examined over a range of substrate concentration, temperature, and pH. In the concentration range of 100–2000 mg dicamba L–1 (0.5–9.0 mM), the degradation was accompanied by a stoichiometric release of 2 mol of Cl per mol of dicamba degraded. The cultures had an optimum pH 6.5–7.0 for dicamba degradation. Growth studies at 10°C, 20°C, and 30°C yielded activation energy values in the range of 19–36 kcal mol–1 and an average Q10 value of 4.0. Compared with the pure cultureP. paucimobilis, the consortium was more active at the lower temperature.  相似文献

Lactose of sweet whey permeate was converted into sodium lactate byLactobacillus helveticus. To increase the, productivity of the lactic acid fermentation and to reduce the amounts of effluents, the bioreactor was coupled with an ultrafiltration module and an electrodialysis unit. Without the electrodialyzer, with total cell recycling and at a dilution rate of 0.88 h–1, a cellular concentration of 64 gl–1 and a productivity of 22 gl–1 h–1 were obtained. When the electrodialysis unit is coupled, the outlet concentration of lactate was stabilized at 85±5 gl–1.  相似文献

Summary Growth and metabolite formation were studied as a function of oxygen feed rate, in glucose-limited chemostat cultures of Hanseniaspora uvarum K5 at a dilution rate of 0.26 h–1. Alcoholic fermentation occured at an oxygen feed rate of 80 mmol.l–1.h–1 . Below this value, pyruvate decarboxylase and alcohol dehydrogenase were present at high levels. In contrast, activities of oxidative metabolism enzymes, pyruvate dehydrogenase, aldehyde dehydrogenase and acetyl-CoA synthetase, decreased.  相似文献

Summary A high performance fermentation process for the continuous production of citric acid from sugarcane molasses by using the combination of submerged calcium alginate-immobilized and surface-stabilized cultures of Aspergillus niger KCU 520 in a continuous flow horizontal bioreactor is described. The citric acid productivity was dependent on the dilution rate with an optimum value of 0.015/h. Presaturation of fermentation medium with sterile air, in addition to surface aeration, before feeding into the bioreactor enhanced the citric acid productivity. The highest productivity, citric acid product concentration and yield obtained were 1.7 kg M–3h–1, 110kg M–3 and 91% respectively. The cultures were continuously used for 30 days without any apparent loss in citric acid productivity.  相似文献

H2 photoproduction by Rhodobacter sphaeroides GL-1 immobilised on polyurethane foam in a continuous flow photobioreactor was shown to occur for prolonged periods. Under optimal conditions (300 W m–2; dilution rate 0.023 h–1) the rate of H2 production was 0.21 ml h–1 ml–1 foam matrix with an efficiency for lactic acid to H2 conversion of 86%. The effect of the dilution rate on biomass and product synthesis in fermentations of glucose, fructose and a commercial mixture of fructooligosaccharides (FOS) by Bifidobacterium longum ATCC 15707 was studied. Kinetic parameters (maximum specific growth rate, Monod constant, maintenance, and yield coefficients) in the mathematical model of the fermentation were estimated from experimental data. In the FOS mixture fermentations, approximately 12% of the total reducing sugars (mainly fructose) in the feed were not metabolized by the bacterium. In fermentations of fructose and the FOS mixture, biomass concentration increased as the dilution rate increased and, once maximum values were reached [3.90 (D=0.20 h–1) and 2.54 g l–1 (D=0.15 h–1), respectively], decreased rapidly as the culture was washed out. Formic acid was detected at low dilution rates in glucose and fructose fermentations. The main products in fermentations of the three carbon sources were lactic and acetic acids. Average values of the molar ratio between acetic and lactic acids of 1.18, 1.21 and 0.83 mol mol–1 were obtained in glucose, fructose and FOS mixture fermentations, respectively. In batch fermentations carried out without pH control this molar ratio was lower than 1.5 only when fructose was used as the carbon source.  相似文献

Summary The effect of various culture conditions on growth kinetics of a Schizosaccharomyces pombe strain were investigated in order to facilitate the potential use of this organism in enology. The effect of ethanol was characterized as a 80 g·l–1 critical concentration. An optimal pH range of 3.5–4.0 was found. Growth was slightly affected by malic acid at pH 3.0 but was shown to decrease drastically at pH 5.5. Aerated cultures had increased rates of growth and malate degradation than those grown under anaerobiosis, although the specific rate of malate degradation remained fairly constant. Taking into account biomass synthesis data it would seem likely that under anaerobiosis malate can replenish the Krebs' cycle for amphibolic precursor synthesis and hence avoid potential growth limitation. Summary The cell-associated and exocellular hemicellulolytic polysaccharide depolymerase and glycoside hydrolase activity ofBacillus macerans NCDO 1764 was monitored over a range of anaerobic growth conditions in batch and continuous culture. The enzymes were detectable throughout the complete growth cycle in batch culture reaching and maintaining maximum levels in the stationary phase. In continuous culture enzyme activity was largely independent of growth rate (D=0.025–0.1 h-1) although the activity was reduced at higher dilution rates (0.125–0.15 h-1). Although activity was detectable over a wide pH range (pH 5.5–7.5) it was pH dependent, and maximum activities of both the cell-associated and exocellular enzymes were measured in cultures maintained at pH 6.5–7.0±0.1.The principal metabolites formed anaerobically from xylose byB. macerans in batch and continuous culture were acetic acid, formic acid and ethanol which represented 95–99% of the products formed. Smaller amounts of acetone,d,l-lactic acid and succinic acid were formed together with traces of butyric acid (<5 nmol/ml) and isovaleric acid (<25 nmol/ml). The proportions of the metabolites produced varied with growth conditions and were influenced by the pH of the culture and the rate and stage of growth of the microorganism.  相似文献

With a cell concentration of 125 g dry biomass 1–1 and a dilution rate of 0.1 h–1,Propionibacterium acidipropionici produces 30 g propionic acid 1–1 from sugar with a productivity of 3 g 1–1 h–1. The yield of propionic acid is approx. 0.36–0.45 g propionic acid g–1 sucrose and is independent of the dilution rate and cell concentration. Acetic acid is an unwanted by-product in the production of propionic acid. The concentration of acetic acid only increases slightly when the cell concentration is increased. A two-stage fermentation process was developed for the conversion of sugar or molasses of various types to propionic acid and vitamin B12. By fermentation of blackstrap molasses (from sugar beet and sugar cane) in the first fermentation stage 17.7 g propionic acid 1–1 with a yield of 0.5 g propionic acid g–1 carbohydrate was produced with a dilution rate of 0.25 h–1. In the second stage 49 mg vitamin B12 1–1 was produced at a dilution rate of 0.03 h–1.  相似文献
