安南龟冬眠前后酯酶同工酶的比较研究   总被引:2,自引:0,他引:2  
应用垂直平板聚丙烯酰胺凝胶电泳技术,对安南龟冬眠前后心、肝、脾、肺、肾、胃、骨骼肌7种器官组织进行了酯酶同工酶的检测.结果表明,安南龟不同器官组织的酯酶同工酶酶谱有明显差异,反映了酯酶同工酶的器官组织特异性;同一器官组织在冬眠前后其酶谱也出现明显的变化,反映了安南龟冬眠前后代谢过程和代谢水平的不同.  相似文献

本文报道了用改良z.I. Ogita的小型薄层板聚丙烯酰胺凝胶电泳,分析达乌尔黄鼠10种组织的乳酸脱氢酶(LDH)同工酶,发现了15条亚带。并用尿素抑制与热变性实验初步确定这些亚带分属于5种LDH同工酶。同时测定了LDH同工酶总活性和5种LDH的相对比例,观察和分析了黄鼠在冬眠期及其冬眠前后LDH同工酶的变化。这些变化可能反映了异温动物(冬眠动物)的LDH同工酶基因的表达有不同于恒温动物的某些特点。  相似文献

本研究以鲫鱼和红龙腈金鱼为材料,对肾脏、心脏、肝脏、脾脏、脑、眼、卵和肌肉等8种组织器官分别进行了乳酸脱氢酶(LDH)同工酶的分析比较。结果表明,不同组织中LDH的同工酶谱带是不相同的,存在着组织特异性。但鲫鱼与红龙腈金鱼同一组织器官的LDH的同工酶谱带则基本相同,这在一定程度上为金鱼是由鲫鱼进化而来的推论提供了依据。  相似文献

利用聚丙烯酰胺凝胶电泳方法对家鸽不同组织的酯酶同工酶、乳酸脱氢酶同工酶的酶谱进行了观察和试验分析。计算了家鸽8种组织酯酶同工酶各自酶带的Rf值。显示出家鸽8种组织的酯酶同工酶酶带和7种组织的乳酸脱氢酶同工酶酶谱分布特征具有明显的组织特异性。初步分析了基因位点与酶谱带型之间的相互关系,对于鸟类的生化分析和遗传研究有参考作用。  相似文献

利用水平板淀粉凝胶电泳法和盘状聚丙烯酰胺凝胶电泳法,结合酶的染色法研究了小麦种子萌发后的胚、幼根和幼苗中过氧化物酶等6种同工酶及胚乳中淀粉酶同工酶的变化;测定了幼根和幼苗中过氧化物酶和吲乙酸氧化酶的活性。过氧化物酶和淀粉酶的同工酶数目随萌发大量增加。6-磷酸葡萄糖脱氢酶、谷氨酸脱氢酶、异柠檬酸脱氢酶和酯酶在萌发初期的胚中就有不少同工酶带和较强的活性。过氧化物酶、吲乙酸氧化酶和谷氨酸脱氢酶在根中比在苗中活性大得多,相反,过氧化氢酶和6-磷酸葡萄糖脱氢酶在苗中比在根中活性大得多,酯酶在苗中比在根中活性稍大,只有异柠檬酸脱氢酶在根和苗中无甚差异。同工酶带的数目及其相对强度的不同可能与器官差异有关。此研究有助于进一步探讨不同组织和器官在分化时具有不同的代谢特点。  相似文献

用垂直的淀粉凝胶电泳方法分析了胚胎发育阶段(0—105小时)和成体6种组织中的乳酸脱氢酶(LDH)、苹果酸脱氢酶(MDH)、谷氨酸脱氢酶(GDH)、葡萄糖—6—磷酸脱氢酶(G6PD)、醇脱氢酶(ADH)、异柠檬酸脱氢酶(IDH)、酯酶(EST)和碱性磷酸酶(AKP)等8种同工酶系统的酶带。共约有23个基因座位在其胚胎发育期和成体组织中表达。所分析的大多数同工酶在团头鲂个体发生过程中表达的情况大致可分为3种类型:①胚胎发育期间持续存在的同工酶类,它们在成体组织中无特异性分布;②到胚胎发育后期才开始表达的同工酶类,它们往往和特定的组织或器官的形态发生或机能分化紧密相关,并在成体组织中呈特异性分布;③在成体组织中都没有被发现而只在胚胎发育期所特有的同工酶。团头鲂的MDH同工酶类之间以及它们和G6PD同工酶活性变化之间存在着相关性,二者可能存在共同的调控机制。与许多其他鱼类不同,团头鲂的GDH同工酶在整个胚胎发育过程中都有活性,但在其成体组织中无特异性分布。基因调控系统的时空精确性是保证团头鲂胚胎发育正常代谢活动的必要条件。  相似文献

目的研究乳酸脱氢同工酶在3种东方田鼠不同器官的分布.方法以大鼠和小鼠作对照,用聚丙烯酰胺凝胶电泳对3个不同地区的东方田鼠(Microtusfortis)血清、红细胞、肝脏、肾脏、肺中乳酸脱氢酶(LDH)同工酶进行分析.结果东方田鼠血浆中只有LDH5,血清中湖南洞庭湖地区、宁夏青铜峡地区的东方田鼠也只有LDH5,而东北地区的东方田鼠则含有LDH1和LDH5,3个地区的东方田鼠肾脏和肺中都有5种LDH同工酶,肝脏中除东北地区的东方田鼠含5种LDH同工酶外,其余两种东方田鼠均以LDH5为主(LDH5占95%以上).结论东北地区东方田鼠乳酸脱氢酶同工酶谱与其他两地的东方田鼠不相同.  相似文献

用聚丙烯酰胺凝胶等电聚焦电泳分析了大林姬鼠(Apodemus peninsulae)心、肝、脾、肾和腿肌的 α-酯酶,β-酯酶和苹果酸脱氢酶同工酶。结果表明3种同工酶的活性在5种器官组织中均有明显差异,其中以肝组织的酯酶活性最高,不同器官组织的酶谱也有明显差别,如脾的β一酯酶仅有B区带,同一器官组织通常以α-酯酶活性高于β-酯酶。苹果酸脱氢酶在碱性溶液中染色,肝组织有明显的AB医。心肌与腿肌的苹果酸脱氢酶活性略高于其他组织。  相似文献

In some ant colonies founded by multiple queens, genetically unrelated queens cooperate to establish the colony. To test whether founding queens differentially adjust the amount of food exchange to nestmate and non-nestmate Polyrhachis moesta (Emery) queens before and after hibernation, food exchange in founding queens with different nutritional conditions was investigated. The proportion of non-nestmate queens showing food-begging behavior before hibernation was higher than that of nestmate queens. The number of begging behaviors was greater after hibernation than before hibernation, both between nestmates and between non-nestmates. Weight loss in fed queens performing food exchange with nestmate queens was significantly greater after hibernation than before hibernation. These results suggest that founding queens can discriminate between nestmate and non-nestmate founding queens and change the level of cooperation with founding queens before and after hibernation.  相似文献

Testis size and spermatogenesis were monitored serially in individual golden-mantled ground squirrels before, during, and after the hibernation season. During hibernation, animals spent 81% of days in torpor at body temperatures of 3-4 degrees C. Torpor bouts of 6 days duration were interspersed with brief arousals from torpor during which animals were normothermic. In the 5 mo between December (when animals entered hibernation) and April (when torpor was spontaneously terminated), the estimated mass of testes increased gradually from 500 to 1100 mg, but spermatogenesis did not advance beyond pachytene spermatocytes, which were present before hibernation began. In contrast, during the month after torpor was terminated, testes increased rapidly to 3500 mg and after 31 days, spermatozoa were found in the epididymides. We suggest that the limited testis growth that occurred during the hibernation season was restricted to intervals during which squirrels were aroused from torpor. The major portion of gonadal growth and spermatogenesis in the laboratory, and presumably in the field, occurs after ground squirrels have regained the normothermic state. Since males are reproductively mature when first trapped in spring, these findings suggest that males are normothermic for several weeks before they emerge from their hibernacula in the spring.  相似文献

In some hibernating species, an extended euthermic period before spring emergence has been reported during which testicular maturation occurred. In this study, we investigated whether male European ground squirrels Spermophilus citellus (Linnaeus, 1766) develop scrotal testes before or after the termination of hibernation. The course of testis development and testosterone concentrations were examined in young-of-year male ground squirrels (n = 4) before, during, and after their first hibernation. In the environmental chamber, all animals showed regular hibernation patterns with decreasing torpor bout lengths prior to the end of hibernation. Four weeks before hibernation ended, testosterone levels were elevated during spontaneous arousals. Testosterone concentrations peaked during the first 2 months post heterothermy and decreased thereafter. In 2 experimental males, testes descended from the inguinal region into the scrotum shortly before the end of heterothermy. The state of testis development and testosterone secretion corresponded to that found in semi-free-living males at spring emergence. Testis size increased in active animals to maximum widths during weeks 3 and 4 post heterothermy. Scrotal skin pigmentation was absent until heterothermy ended and developed during the first month thereafter. The data suggest that male S. citellus do not need a pre-emergence euthermic period for reproductive development.  相似文献

用植入式半导体温度记录元件iButton 记录了达乌尔黄鼠冬眠季节及其前后的体温,分析了其冬眠模式和体温调节特点。结果显示:1)实验室条件下,达乌尔黄鼠冬眠季节长短的个体差异较大,可以分成深冬眠型、
少冬眠型和不冬眠型三种类型;2)达乌尔黄鼠在冬季表现出深冬眠阵(最低体温Tbm in <20℃ ,冬眠阵的持续时间BD >24 h)、短冬眠阵(Tbmin < 20℃ , BD≤24h)和日眠阵(Tbmin ≥20℃ , BD≤24 h)3 种类型,最低体温分别
为2.54℃ ± 0.35℃ 、10.05℃ ± 1.97℃ 和23.09℃ ± 0.40℃ ,彼此之间差异显著。日眠阵阵间产热阶段的最高体温为38.09℃ ±0.17℃ ,高于深冬眠阵(37.31℃ ±0.15℃ )和短冬眠阵(37.22℃ ±0.31℃ ); 3)深冬眠阵和日
眠阵中最低体温均与环境温度显著相关,冬眠过程中的最低体温为-2.43℃ ;4)深冬眠过程中,多数个体可以短时(≤3 h)耐受- 2℃ ~ 0℃ 的低温,激醒或继续维持深冬眠,无致死效应,但长时间(15 h)或过度低温
(- 5℃ 以下)的条件下,深冬眠的达乌尔黄鼠被激醒(70% )或死亡(30% ),不能持续冬眠; 5)入眠前10 d体温日波动幅度显著增加,高于出眠后的日体温波动,且多数个体入眠前出现体温的“试降”。表明,冬眠前

We investigated sperm storage by females of a predatory bug, Nabis rugosus, before and after hibernation and with regard to nutritional status. In the field, females had more sperm available before than after hibernation. Food-deprived females maintained fewer sperm than fed females before but not after hibernation. However, after hibernation food-deprived females suppressed egg production rather than decreasing sperm-storage efficiency. Because mated females did not exhibit increased overwinter survival relative to virgin females there is a low likelihood of direct benefits in the form of ejaculate-derived nutrients. If nutrition-dependent sperm storage by females is widespread in the animal kingdom, our findings may have important implications for the understanding of postcopulatory sexual selection and the evolution of food gifts.  相似文献

Long periods of inactivity in most mammals result in bone loss that may not be completely recoverable during an individual's lifetime regardless of future activity. Prolonged inactivity is normal during hibernation, but it remains uncertain whether hibernating mammals suffer decreased bone properties after hibernation that affects survival. We test the hypothesis that relative cortical area (CA), apparent density, bone area fraction (B.Ar/T.Ar), and moments of inertia do not differ between museum samples of woodchucks (Marmota monax) collected before and after hibernation. We used peripheral quantitative computed tomography to examine bone geometry in the femur, tibia, humerus and mandible. We see little evidence for changes in bone measures with hibernation supporting our hypothesis. In fact, when including subadults to increase sample sizes and controlling age statistically, we observed a trend toward increased bone properties following hibernation. Diaphyses were significantly denser in the humerus, femur, and tibia after hibernation, and relative mandibular cortical area was significantly larger. Similarly, relative mechanical indices were significantly larger in the mandible after hibernation. Although tests of individual measures in many cases were not significantly different prehibernation versus posthibernation, the overall pattern of average increase posthibernation was significant for relative CA and densities as well as relative diaphyseal mechanical indices when examining outcomes collectively. The exception to this pattern was a reduction in metaphyseal trabecular bone following hibernation. Individually, only humeral B.Ar/T.Ar was significantly reduced, but the average reduction in trabecular measures post‐hibernation was significant when examined collectively. Because the sample included subadults, we suggest that much of the increased bone relates to their continued growth during hibernation. Our results indicate that woodchucks are more similar to large hibernators that maintain skeletal integrity compared to smaller‐bodied hibernators that may lose bone. 贮脂类动物在冬眠前大量积累脂肪来准备冬眠,并在入眠时迅速降低体温和代谢率。为探究入眠准备期达乌尔黄鼠体温、代谢率、呼吸商及能量代谢的变化,将其入眠准备期分为育肥期、体重高峰期、育肥后期和冬眠前的试降期,使用植入式半导体温度记录元件iButton、开放式代谢仪和改进的代谢笼,监测其体温、代谢率及呼吸商和能量摄入的变化。结果显示:(1)达乌尔黄鼠体温在冬眠前13 - 34 d 开始下降,远早于冬眠但晚于体重高峰期;体重高峰期体温有降低的趋势,持续时间为1 - 3 d;育肥后期体温显著下降,体温日波动幅度增加。(2)体重高峰期的静止代谢率高于育肥期,育肥后期有降低的趋势,试降期最低。(3)呼吸商在体重高峰期先升高,之后迅速衰减;入眠准备期的能量摄入在体重达高峰期前达到最大值。结果表明,达乌尔黄鼠在入眠准备期,其体温和代谢率已开始降低,能源物质已开始转变;体重高峰期可能是达乌尔黄鼠入眠的一个转折点或启动入眠的开关。  相似文献

In mammals, the circadian master clock generates daily rhythms of body temperature (T(b)) that act to entrain rhythms in peripheral circadian oscillators. The persistence and function of circadian rhythms during mammalian hibernation is contentious, and the factors that contribute to the reestablishment of rhythms after hibernation are unclear. We collected regular measures of core T(b) (every 34 min) and ambient light conditions (every 30 s) before, during, and following hibernation in free-living male arctic ground squirrels. Free-running circadian T(b) rhythms at euthermic levels of T(b) persisted for up to 10 d in constant darkness after animals became sequestered in their hibernacula in fall. During steady state torpor, T(b) was constant and arrhythmic for up to 13 d (within the 0.19°C resolution of loggers). In spring, males ended heterothermy but remained in their burrows at euthermic levels of T(b) for 22-26 d; patterns of T(b) were arrhythmic for the first 10 d of euthermia. One of four squirrels exhibited a significant free-running T(b) rhythm (τ = 22.1 h) before emergence; this squirrel had been briefly exposed to low-amplitude light before emergence. In all animals, diurnal T(b) rhythms were immediately reestablished coincident with emergence to the surface and the resumption of surface activity. Our results support the hypothesis that clock function is inhibited during hibernation and reactivated by exposure to light, although resumption of extended surface activity does not appear to be necessary to reinitiate T(b) cycles.  相似文献
