Mucosal cells of the small intestine obtained from rats deprived of vitamin D or given excessive amounts of the vitamin accumulated significantly more calcium than did cells from control animals. 2. Mucosal cells from vitamin D-deficient rats released less calcium than did cells from normal or hypervitaminotic D animals. 3. Studies in vivo showed that the transfer of (45)Ca from the intestine to the blood was delayed in vitamin D deficiency, but was accelerated in hypervitaminosis D. 4. The findings support the thesis that vitamin D is involved in the release of calcium rather than in its uptake by mucosal cells. 5. Further evidence is presented suggesting that uptake of calcium by intestinal mucosal cells at 0 degrees is primarily passive, whereas at 38 degrees uptake and release are effected by an active process that depends on energy derived from glycolytic activity. 相似文献

The effect of various periods of pretreatment in CaSO4 solutions (aging) on the absorption of Na and K by bean stem slices was investigated. Freshly sliced tissue absorbed Na over the entire range of concentrations studied (0.02-50 mm). Potassium absorption by fresh tissue was nil at concentrations below 0.5 mm but at higher concentrations was similar to that of Na. When tissue was aged by aerating slices for 20 hr in 0.5 mm CaSO4, K absorption was substantial over the entire range (0.01-50 mm), with evidence of a dual mechanism of absorption, whereas Na absorption was nil at concentrations below 0.2 mm. The formation of K-absorbing capacity with aging, and the loss of Na-absorbing capacity at low concentrations, were temperature-dependent and did not result from significant changes in rates of efflux of either ion. The absorption of Na by fresh tissue and K by aged tissue was sensitive to antimetabolites, with K uptake the more sensitive. Benzyladenine, an analog of kinetin, suppressed the formation of K-absorbing capability in aged tissue but did not prevent the loss of Naabsorbing capacity. Possible mechanisms for this alteration in ion-specificity of transport mechanisms are discussed. 相似文献

Polyphenolic compound chlorogenic acid (CGA) known to be much contained in coffee beans was found to have a regressive effect on induced aberrant crypt foci (ACF) as well as on development of ACF in azoxymethane (AOM)-induced colorectal carcinogenesis in rats. Rice germ and gamma-aminobutyric acid-enriched defatted rice germ inhibited AOM-induced ACF formation and colorectal carcinogenesis in rats. Ferulic acid (FA) also known to be contained in coffee beans and rice prevented AOM-induced ACF formation and intestinal carcinogenesis in rats. Both of food factors, coffee and rice may be of benefit to prevention of human colorectal cancers. 相似文献

This paper describes experiments on Cl transport into the roots, stem and leaves of bean plants, the roots of which have been exposed to lipids in the root solution. Monoand digalactose diglyceride strongly increased Cl transport into all plant parts, probably by transport of the glycolipids further into the plant. Phosphatidyl choline increased Cl absorption by the roots, but transport into the stem and leaves was not affected. This phospholipid was only absorbed by the root tissue. 32P-glycerophosphoryl choline added to the root solution was readily transported and esterified as phospholipid in all plant parts. This chemical did increase Cl uptake by the roots but Cl accumulation in the leaves was reduced by as much as 40%. Phosphatidyl glycerol, phosphatidyl inositol, and sulfolipid increased Cl transport into roots, stem, and leaves, and a high mobility of 32P-phosphatidyl glycerol was demonstrated. Generally no significant effect of the above lipids on Na transport in beans and cotton was noted except that monogalactose diglyceride did increase Na transport in cotton. 相似文献

Previous studies have shown that the tetrathiomolybdate ion [MoS42?] is a potent antagonist of Cu metabolism. Effects of orally administered MoS42? on the absorption and tissue distribution of 64Cu in rats have now been investigated. Four or 12 mg Mo/kg diet, when given as MoS42?, strongly inhibited 64Cu absorption and modified the fate of absorbed Cu, decreasing hepatic and renal uptake but increasing plasma retention of 64Cu. These effects were not induced by equivalent dietary concentrations of Mo as MoS42? or when S2? was given as CaS. Clinical and biochemical effects induced by orally administered MoS42? were abolished by increasing dietary concentrations of Cu. Such treatment also inhibited the absorption and tissue retention of 99Mo derived from 99MoS42?. Intraperitoneal administration of Cu ameliorated clinical effects attributable to MoS42? but neither inhibited 99Mo absorption nor the appearance of systemic defects in Cu metabolism. Since the absorption of MoS42? (or its derivatives) from the gastrointestinal tract is inhibited by Cu, it is evident that the site of its action as an antagonist influencing either the absorption or the subsequent metabolic fate of Cu depends upon the ratio Cu/MoS42? in the diet. 相似文献

- The mechanism of transport of Krebs cycle intermediates, phosphateand sulfurcontaining compounds across the membrane of purifiedbean mitochondria was investigated by directly measuring dieexchange between intramitochondrial labelled substrates andexternal anions and by testing die inhibitor sensitivity ofdiese transport processes.
- The exchange between intramitochondrialphosphate and externalphosphate or sulfite is insensitive toN-ediylmaleimide or butylmalonatewhen either is added alone,but is completely inhibited by N-ethylmaleimideplus butylmalonateor by mersalyl. Internal phosphate is exchangedwith malate,succinate, oxaloacetate, sulfate and thiosulfate;these reactionsare inhibited by butylmalonate but not affectedby N-ethylmaleimide.
- Internal sulfate is exchanged with malate, malonate, succinate,phosphate and sulfite in a butylmalonate- and mersalyl-sensitivereaction. Also the exchanges of malonate with phosphate, sulfateand sulfite are inhibited by butylmalonate and mersalyl. During germination and early growth of the castor bean (Ricinus communis) nitrogenous constituents from the endosperm are transferred via the cotyledons to the growing embryo. Exudate collected from the cut hypocotyl of 4-day seedlings contained 120 millimolar soluble amino nitrogen and glutamine was the predominant amino acid present, comprising 35 to 40% of the total amino nitrogen. To determine the nature of nitrogen transfer, the endosperm and hypocotyl were removed and glutamine uptake by the excised cotyledons was investigated. Uptake was linear for at least 2 hours and the cotyledons actively accumulated glutamine against a concentration gradient. The uptake was sensitive to respiratory inhibitors and uncouplers and efflux of glutamine from the excised cotyledons was negligible. Transport was specific for the l-isomer. Other neutral amino acids were transported at similar rates to glutamine. Except for histidine, the acidic and basic amino acids were transported at lower rates than the neutral amino acids. For glutamine transport, the K(m) was 11 to 12 millimolar and the V(max) was 60 to 70 micromoles per gram fresh weight per hour. Glutamine uptake was diminished in the presence of other amino acids and the extent of inhibition was greatest for those amino acids which were themselves rapidly transported into the cotyledons. The transport of amino acids, on a per seedling basis, was greatest for cotyledons from 4-to 6-day seedlings, when transfer of nitrogen from the endosperm is also maximal. It is concluded that the castor bean cotyledons are highly active absorptive organs transporting both sucrose and amino acids from the surrounding endosperm at high rates. 相似文献

The intraperitoneal administration of insulin has been recommended because it was found to effectively control the plasma glucose level. Several authors have suggested that intraperitoneal insulin administration may be more "physiological" and therefore preferable because the insulin is absorbed into the portal venous system without, however, identifying the exact pathways. The possibility that insulin is absorbed through the surface of the liver was investigated in rats. The results show that insulin is absorbed rapidly by this route, but the effect on glucose modulation is similar to that of insulin given by other routes. In contrast, the effect on glucose modulation was delayed following insulin administration into the lower peritoneal cavity with exclusion of the liver. 相似文献

Broad bean rhizosphere fungi differently affected plant growth.Fusarium moniliforme, Penicillium martensii, Rhizopus stolonifer, andCladosporium sphaerospermum stimulated both root and shoot growth.Aspergillus niger andAlternaria tenuis have an inhibitory effect on plant growth. Summary The fungal flora of the rhizosphere of three varieties of broad bean and cotton was studied by the dilution-plate technique. The numbers of fungi were higher in the rhizosphere than in the non-rhizosphere soil. Plant type and age, and soil type have a significant influence of the nature and numbers of fungal flora associated with plant roots.Cladosporium was relatively more abundant in the rhizosphere of broad bean varieties, whilePenicillium was found to constitute a high percentage of fungi found in the rhizosphere of cotton varieties. Summary Seed and root exudates from three varieties of both broad bean and cotton grown aseptically were analyzed for amino acids and sugars. Broad bean varieties were found to excrete considerable amounts of amino compounds with 15 different amino acids, while cotton varieties excreted less consisting of 12 amino acids. Five sugars were identified in the seed and root exudates from broad bean varieties, while three were present in the seed and root exudates from cotton varieties. Expression of alternative nitrogenases in Azotobacter vinelandii is repressed by molybdenum. Two strains with Tn5 insertion mutations showed alternative nitrogenase-dependent diazotrophic growth in the presence of Mo. The mutations were in a region which contained four open reading frames (ORFs 1–4). The genetic structure and predicted products of ORFs 2, 3 and 4 are typical of the membrane-associated elements of the ATP-binding cassette (ABC) superfamily of transport systems. The products of 0RF3 and 0RF4 are homologous with the products of the Escherichia coli genes chlD and the partially sequenced chlJ, respectively, both of which are implicated in molybdenum transport. ORF1, which is in the relative position of bacterial permease genes commonly specifying periplasmic binding proteins, encodes a 29 kDa protein with a novel primary structure. It lacks a potential signal sequence, and its C-terminal half consists of a tandem repeat of a segment which is homologous with the Mr 7 kDa molybdenum-pterin binding protein Mop from Clostridium pasteurianum. This suggests that a substituted pterin may be involved in the initial capture or early metabolism of molybdenum. 相似文献
