The efficacy of the pupal parasitoid Spalangia cameroni Perkins as a biological control agent was tested against house flies Musca domestica Linnaeus and stable flies Stomoxys calcitrans (Linnaeus) in one dairy cattle and two pig installations in Denmark. Weekly releases of S. cameroni from April through to September-October 1999 and 2000 resulted in significant suppressions of house fly populations to below nuisance level, whereas no effect on stable flies was found. Parasitism was significantly higher in the release years compared to the control years, but was below 25% averaged over the fly season for each farm. A statistical model based on a functional relationship between the innate capacity of increase of the two fly species and three explanatory variables (air temperature, fly density and parasitism) provided a fairly good fit to data with the abundances of house flies and stable flies explained mostly by temperature, but intra- and interspecific competition, and parasitism had a significant effect as well. Overall, the model was capable of explaining 14% and 6.6% of the total variation in data for house fly and stable fly, respectively. Spalangia cameroni was the predominant parasitoid to emerge from exposed house fly pupae, but from mid summer onwards Muscidifurax raptor Girault & Sanders (Hymenoptera: Pteromalidae) was also quite common. Cryopreservation of Musca domestica (Diptera: Muscidae) embryos 总被引:1,自引:0,他引:1
Prior studies on cryopreserving embryos of several non-drosophilid flies established that two Drosophila melanogaster embryo cryopreservation protocols were not directly suitable for use with these species. This paper describes our work on developing a protocol for cryopreservation of embryos of the housefly, Musca domestica. Significant progress was made when permeabilization of the vitelline membrane was optimized, a vitrification solution containing ethylene glycol, polyethylene glycol, and trehalose was formulated, and when cooling and recovery of the cryopreservation protocol included a step which passed the embryos through liquid nitrogen vapor. More than 70% of housefly embryos withstand treatments of dechorionation, permeabilization, loading with cryoprotectant, and dehydration in vitrification solution, but the cooling, warming, and poststorage rearing steps still cause a considerable reduction in survival. About 53% of the vitrified M. domestica embryos hatched into larvae. Relative to the percentage of the control adult emergence, about 13% of the embryos stored in liquid nitrogen developed into fertile adults. Hatching of the F(1) progeny of adults having been cryopreserved as embryos was similar to control levels. 相似文献

The toxicity of seven insecticides was evaluated against unparasitized Musca domestica L. pupae and pupae parasitized by Muscidifurax raptor Girault & Sanders or Spalangia cameroni Perkins, two important biological control agents. Only pyrethrins + piperonyl butoxide (Pyrenone) was less toxic to M. raptor compared with house flies. Conversely, all of the insecticides except crotoxyphos were less toxic to S. cameroni compared with house flies. A plateau in the tetrachlorvinphos bioassay line for S. cameroni suggested that this colony had approximately 45% resistant individuals. The selectivity observed between immature stages of house flies and M. raptor or S. cameroni is different from that reported against adult stages of these same species, suggesting that selectivity of an insecticide varies considerably between different life stages. 相似文献

In many taxa, females signal during courtship when they are receptive. However, just because a female signals does not mean that the male responds to the signal. This study examines female signaling of receptivity (readiness to copulate) and male response in the parasitoid wasp Spalangia endius Walker. Females folded their antennae against their heads when they were receptive, and antennal folding has been shown to be effective in eliciting male copulation attempts in a confamilial. However, male S. endius did not respond to antennal folding: males did not contact the female's antennae during courtship, and how quickly a male attempted copulation was independent of whether or not the female had antennae. Males courted from on top of the female's abdomen and appeared to detect receptivity directly from the female's abdomen rising as her genital orifice opened. On females whose abdomens did not rise, initiation of male copulation attempts were delayed but not eliminated. Based on its current lack of function as a receptivity signal and on comparisons to published reports of mating behavior in confamilials, we hypothesize that female antennal folding at receptivity is a vestigial trait in S. endius. 相似文献

Adult house flies, Musca domestica L., of four ages, <1, 3, 7, and 14 day post-eclosion, were exposed to three strains of Beauveria bassiana (P89, L90 and 447). Flies were exposed to moistened filter paper treated with either a low (1.57×104 conidia/cm2) or high (1.57×105 conidia/cm2) concentration of each fungal strain for 6 h. Strain 447 was superior to the two house fly-derived B. bassiana strains in inducing host infection and mortality. Significant spikes in infection and mortality occurred as early as 5 days post-exposure with higher concentration exposures acting more quickly. Few differences were observed in either infection or mortality among the four fly age classes. On Day 10 post-exposure, 77% of the high-concentration, 447-exposed flies were infected, compared with only 24% of the flies from the P89 low-concentration exposure. Potential applications of these results in integrated house fly management programs are discussed. 相似文献

Nasonia vitripennis (Walker) (Hymenoptera: Pteromalidae) is now emerging as genetic model organism for development and genetics research. As a parasitic insect, the egg, larval, pupal, and early adult developmental stages of N. vitripennis occur within the enclosed fly pupae, which differ a lot from the life cycle of other insects that undergo complete metamorphosis. Previous report on the life table of N. vitripennis was based on females only. In this study, the two-sex life table approach was used to examine the parasitic efficiency of N. vitripennis within the pupae of Musca domestica L. (Diptera: Muscidae), and the influence of low temperature and the endosymbiotic bacteria Wolbachia on wasp population growth were investigated. Wolbachia could improve the fecundity of N. vitripennis and prolong the host life history. We propose the preservation of M. domestica pupae at 4 °C for 15 days is suitable in practical use. Age-stage, two-sex life table analysis revealed the stage structure and variability of N. vitripennis population growth, and also provide useful information about the effects of Wolbachia on its reproduction and parasitism of M. domestica. 相似文献

The toxicity of spinosad was determined in one susceptible and five insecticide-resistant laboratory strains of house fly, Musca domestica L. Spinosad was relatively slow-acting, but highly toxic to house flies. In a feeding bioassay, spinosad LC50 at 72 h was 0.51 microg of spinosad per gram of sugar, making it 6.3- and 3.5-fold more toxic to house flies compared with azamethiphos and methomyl, respectively. In topical application bioassay, the LD50 at 48 h of spinosad in susceptible house flies was 40 ng per 20 mg of house fly, making spinosad less toxic than the pyrethroid bioresmethrin synergized by piperonyl butoxide and the organophosphate dimethoate. The insecticide-resistant laboratory strains had resistance factors to spinosad at LC50 in feeding bioassay from 1.5 to 5.5 and at LD50 in topical application bioassay from 2.5 to 4.7, indicating that in house fly cross-resistance to the major insecticide classes will not initially be of major concern for the use of spinosad for house fly control. The toxicity of spinosad was also evaluated against 31 field populations of house flies collected from livestock farms across Denmark. The field populations were 2.2- to 7.5-fold resistant to spinosad at 72 h in feeding bioassay, but based on steep slopes in the bioassay and the limited variation of spinosad toxicity against the various field populations, we consider the field populations to be spinosad-susceptible. We propose a diagnostic dose of 12 microg of spinosad per gram of sugar in feeding bioassay with impregnated sugar for determination of resistant house flies, which is 10x the LC95 of the susceptible strain WHO and approximately = 2x the LD95 of the field populations. Spinosad showed no substantial cross-resistance to the pyrethroid bioresmethrin synergized by piperonyl butoxide, the anticholinesterases dimethoate, azamethiphos, methomyl, and spinosad in house fly field populations. 相似文献

Adult cabbage maggots ( Delia radicum L.) were exposed to dry conidia of isolates of several hyphomycetous fungi by placing them in a centrifuge tube containing conidia, then releasing them into small screened plastic cages. Mortality was assessed after 48, 120 and 160 h. A Beauveria bassiana isolate (P89 from Musca domestica ) caused the highest mortality after 48 h, resulting in 100% mortality and 100% infection. Isolate L90 ( B. bassiana ) and one Metarhizium anisopliae isolate (ARSEF 2521) also caused fatal infection in more than 50% of the flies. To investigate exchange of inoculum, flies were placed in a small container with a dry powder formulation containing B. bassiana (Mycotrol ® ) on the bottom. The flies were removed to small screened cages containing untreated flies. This experiment confirmed the ability of flies to pass inoculum to other flies. In a similar experiment, one treated fly was placed in each cage with one untreated fly. When each fly died, one untreated fly was added to each cage after the dead fly was removed. This study showed that fly to fly transfer of fatal doses of inoculum was possible for a series of at least six flies. When female flies were exposed to the inoculum, then transferred to small cages containing males and an oviposition substrate, no eggs were laid. Further studies are being conducted to develop a system where flies attracted to a trap will be inoculated with the fungus and spread it to a field population. 相似文献

An investigation was undertaken to record the influence of host age on the reproductive performance of Nesolynx thymus (Hymenoptera:Eulophidae). This is an indigenous, gregarious, ecto-pupal parasitoid of certain dipteran insects, including the tachinid fly, Exorista bombycis (Louis) which is a well-known endo-larval parasitoid of the silkworm, Bombyx mori. Each gravid N. thymus female was allowed to parasitise 1–4-day-old puparia of Musca domestica L. for 2 days at a parasitoid–host ratio of 1:20. The parasitised host puparia were observed for progeny recovery, sex ratio, female longevity and fitness (adult size). In addition, reproductive performance of the parasitoid progeny was assessed by allowing its females to parasitise for 2 days, 3-day-old puparia of E. bombycis at a parasitoid–host ratio of 1:4. There was a significant negative correlation between host age and parasitisation rate, parasitoid developmental duration, sex ratio and female longevity while the correlation was positive between host age and parasitoid recovery per host puparium. Similarly, negative correlation was observed between host age and morphometric parameters (body length, head width and wing span of male and female and length and width of female abdomen) of the progeny adults. Host age did not have any impact on the reproductive performance of progeny adults when allowed to parasitise puparia of E. bombycis. 相似文献

We determined the toxicity of the two IGRs, diflubenzuron and cyromazine, in this survey of resistance in Danish field populations of Musca domestica (L.). We observed resistance toward diflubenzuron and for the first time in Denmark and we found field populations with some resistance to cyromazine. Eleven of the twenty-one field populations had larvae surviving a diagnostic dose of 1.6 times of susceptible LC95 of diflubenzuron and two of these populations had larvae surviving 6.1 times of LC95. Eight of the twenty-one field populations had larvae surviving 2.2 times of susceptible LC95 of cyromazine and one population had larvae surviving 4.4 times of LC95. A fivefold cyromazine resistant strain was established after selection with cyromazine. It was 3-, 5-, and 90-fold resistant to diflubenzuron, triflumuron, and methoprene, respectively. Two diflubenzuron resistant strains (120- and 86-fold, respectively) were established. They showed a high level of resistance to triflumuron (1000- and 200-fold, respectively), and moderate resistance to methoprene (73- and 50-fold, respectively). Both were susceptible to cyromazine. This study shows that by applying the recommendations of previous resistance risk assessments, severe control failures and detrimental development of a high level of resistance have been avoided. In this study we examined the relationship between clutch size and parasitoid development of Muscidifurax raptorellus (Hymenoptera: Pteromalidae), a gregarious idiobiont attacking pupae of the housefly, Musca domestica (Diptera: Muscidae). Host quality was controlled in the experiments by presenting female parasitoids with hosts of similar size and age. This is the first study to monitor the development of a gregarious idiobiont parasitoid throughout the course of parasitism. Most female wasps laid clutches of one to four eggs per host, although some hosts contained eight or more parasitoid larvae. In both sexes, parasitoids completed development more rapidly, but emerging adult wasp size decreased as parasitoid load increased. Furthermore, the size variability of eclosing parasitoid siblings of the same sex increased with clutch size. Irrespective of clutch size, parasitoids began feeding and growing rapidly soon after eclosion from the egg and this continued until pupation. However, parasitoids in hosts containing five or more parasitoid larvae pupated one day earlier than hosts containing one to four larvae. The results are discussed in relation to adaptive patterns of host utilization by gregarious idiobiont and koinobiont parasitoids. 相似文献

【目的】从家蝇Musca domestica中克隆一种C-型凝集素(C-type lectin)基因,并分析其在家蝇免疫过程中的功能。【方法】根据家蝇转录组信息,克隆了一条C型凝集素基因,将其命名为Mdlectin-C1。构建Mdlectin-C1的原核表达载体,在大肠杆菌Escherichia coli中表达并纯化r Mdlectin-C1蛋白,制备多克隆抗体。利用实时定量PCR(quantitative real-time PCR,qRT-PCR)及Western blot技术对家蝇不同发育阶段和细菌感染前后Mdlectin-C1的表达水平进行检测。运用RNAi技术敲低家蝇1龄幼虫Mdlectin-C1基因表达,然后进行细菌刺激并观察幼虫存活率变化。【结果】Mdlectin-C1 c DNA包含一个546 bp完整开放阅读框,编码181个氨基酸残基,所推导多肽N端包含由21个氨基酸残基组成的信号肽,成熟肽中含有一个保守的碳水化合物识别结构域(carbohydrate-recognition domain,CRD)。qRT-PCR及Western blot结果显示,家蝇从1龄幼虫到蛹的发育过程中,Mdlectin-C1表达量逐步上升,蛹期达到最大值。在革兰氏阴性菌大肠杆菌E.coli和革兰氏阳性菌金黄色葡萄球菌Staphylococcus aureus刺激后,家蝇2龄幼虫Mdlectin-C1均上调表达,并分别在36和3 h时达到最高值。利用RNAi技术成功敲低Mdlectin-C1表达。对Mdlectin-C1基因敲低的1龄幼虫进行细菌刺激,敲低实验组幼虫存活率显著低于正常对照组。【结论】Mdlectin-C1可能参与家蝇抗菌免疫应答,并在此过程中具有重要作用。 相似文献

【目的】探究香樟Cinnamomum caphora果实精油、天竺桂Cinnamomum pedunculatum叶精油和龙柏Sabina chinensis var.chinensis叶精油对家蝇Musca domestica成虫的熏蒸活性,比较这3种植物精油以及混配精油对家蝇的毒力效果,为植物源灭蝇剂的开发提供理论依据。【方法】利用水蒸汽蒸馏法提取植物精油,采取三角瓶密闭熏蒸法测试植物精油及其混剂对家蝇5日龄成虫的熏蒸毒性和击倒作用。【结果】3种植物精油对家蝇的LC50值(致死中浓)分别为5.28,16.86和14.54μg/cm3。LC90熏蒸剂量下,对家蝇的KT50值(击倒中时)分别为12.03,16.56和13.37 min。天竺桂叶精油和龙柏叶精油混配后对家蝇有增效作用,二者的LC50值配比为9∶1时,增效作用极为显著,共毒系数高达367.95。【结论】香樟果实精油对家蝇成虫有很好的熏杀和击倒作用,龙柏叶精油次之,天竺桂叶精油效果最差。天竺桂叶精油和龙柏叶精油混配后对其对家蝇成虫的熏蒸毒性有增效作用。3种植物精油具备开发环保灭蝇剂的潜力,可深入研究。 相似文献

Total lipid, fatty acid and sterol composition of larvae and adults of Musca domestica was investigated before and after feeding on sucrose syrup or on the same syrup containing 1% lead nitrate. The effects of sucrose and of lead ions were found to be different. In larvae sucrose diet inhibited the fatty acid elongation and stimulated the first stages of their unsaturation. A significant increase of phytosterol concentrations was obtained. These changes increased the cell membrane permeability. The addition of lead caused a decrease of the fatty acid unsaturation, which decreased the cell membrane permeability. In adults the sucrose diet had no effect on the lipid and sterol composition, while the addition of lead decreased the cholesterol concentration. The composition of lipids and sterols also depends on the diet of larvae before pupation. The data obtained suggested that changes in lipid and sterol composition, which control the permeability of the cell membrane, might be an adaptive response of the organism to the changes of the environment. 相似文献

Abstract. Field investigations on Musca domestica houseflies in Rhodesia demonstrate a greater degree of contact than previously reported between domestic M. d. curviforceps and feral M. d. calleva at their habitat interface. There was no evidence of any seasonal disappearance of hybrid flies; stable, intermediate, largely domestic populations are a feature of areas of sympatry. The existence of almost pure calleva feral populations in these areas was verified, though a slight degree of introgression with curviforceps was demonstrated for the first time. Gene-flow between the subspecies was studied and a new explanation for the one-way gene-flow, based on habitat choice of hybrid flies, is advanced.

The ratio, intended as a size-adjusted measure of frons width, is not independent of size, and frons width and head width (shown to be a good measure of size) are at best weakly correlated. It was confirmed that the frons width of male calleva × curviforceps hybrids is stabilized at the fronJ width characteristic of curviforceps although laboratory breeding results indicate that intermediate frons widths are to be expected. The proximal mechanisms leading to monandry have been little studied in most insect orders, including Hymenoptera. In the parasitoid wasp Spalangia endius, mated females are less attractive (less often mounted) than virgins and are unreceptive (unlikely to allow copulation). Which aspects of mating are responsible was tested by observing male responses toward females whose mating had been interrupted at various stages. All females were allowed to receive precopulatory courtship and to open their genital aperture to copulate. Then some were interrupted before copulation, some after copulation but before postcopulatory courtship, and some were allowed to complete postcopulatory courtship. Females that had copulated were not less attractive than females that had not. In contrast, females that had received postcopulatory courtship were clearly both less attractive and less receptive. Thus, postcopulatory courtship functions as extended mate guarding, by making the female less attractive and less receptive to subsequent males even after the original male is no longer present. The effect of postcopulatory courtship on female attractiveness was persistent but imperfect: when males were presented sequentially to mated females, most but not all males retreated without mounting, and a female could repulse more than twenty males in succession. 相似文献
