Summary. The capability of ficin, a cystine protease, to form peptide bonds was investigated using several types of N-Boc-amino acid phenyl and naphthyl esters as acyl donor components. Enzyme-catalyzed peptide synthesis was carried out under optimized reaction conditions of pH, acyl acceptor concentration and selection of the best yield organic solvent. It used a condensation of N-Boc-Ala-OpGu and Ala-pNA as a model reaction. The products were obtained in 72–96% yield using 10 different substrates, within a few minutes of reaction time.  Two Exobasidium species causing Exobasidium leaf blister on Rhododendron spp. are described. An Exobasidium leaf blister on Rhododendron yedoense var. yedoense f. yedoense has been recognized in Hokkaido Prefecture, Japan, since the first report was issued in 1950. The causal fungus is identified with Exobasidium dubium from the morphology of its hymenial structure and mode of germination of the basidiospores. Another Exobasidium leaf blister on Rhododendron dauricum has been observed in Hokkaido Prefecture, Japan. In comparison with morphology based on hymenial structure and mode of germination of the basidiospores of the 100 validly described taxa, this fungus differs from those known taxa in the size of basidia and basidiospores, the numbers of sterigmata and septa of basidiospores, and the mode of germination of basidiospores. Thus, a new species, Exobasidium miyabei, is established and illustrated. Summary Correlations between the presence of surface coat and immobilization antigen of Paramecium tetraurelia were studied. Supravital, partial removal of the surface coat resulted in accelerated response of monobacterially and axenically grown cells to the homologous antiserum. Ciliates pretreated with trypsin or pronase (0.5mg/ml for 45 min at 0–4° C) were immobilized approximately twice as fast as untreated control cells. The probable localization of at least part, of the immobilization antigen within the surface coat of P. tetraurelia is discussed.The author thanks Prof. S. Dryl (Department of Cell Biology, M. The cuttlefish, Sepia officinalis, provides a fascinating opportunity to investigate the mechanisms of camouflage as it rapidly changes its body patterns in response to the visual environment. We investigated how edge information determines camouflage responses through the use of spatially high-pass filtered 'objects' and of isolated edges. We then investigated how the body pattern responds to objects defined by texture (second-order information) compared with those defined by luminance. We found that (i) edge information alone is sufficient to elicit the body pattern known as Disruptive, which is the camouflage response given when a whole object is present, and furthermore, isolated edges cause the same response; and (ii) cuttlefish can distinguish and respond to objects of the same mean luminance as the background. These observations emphasize the importance of discrete objects (bounded by edges) in the cuttlefish's choice of camouflage, and more generally imply that figure-ground segregation by cuttlefish is similar to that in vertebrates, as might be predicted by their need to produce effective camouflage against vertebrate predators.  相似文献

This paper proposes a new scheduling policy for cluster-based servers called DAS (Deferred Assignment Scheduling). The main idea in DAS is to defer scheduling as much as possible in order to make better use of the accumulated information on job sizes. In broad outline, DAS operates as follows: (1) incoming jobs are held by the dispatcher in a buffer; (2) the dispatcher monitors the number of jobs being processed by each server; (3) when the number of jobs at a server queue drops below a prescribed threshold, the dispatcher sends to it the shortest job in its buffer. To gauge the efficacy of DAS, the paper presents simulation studies, using various data traces. The studies collected response times and slowdowns for two cluster configurations under multi-threaded and multi-process back-end server architectures. The experimental results show that in both architectures, DAS outperforms the Round-Robin policy in all traffic regimes, and the JSQ (Join Shortest Queue) policy in medium and heavy traffic regimes. Victoria Ungureanu (ACM) is a visiting researcher at DIMACS. She has a Ph.D. in Computer Science from Rutgers University. Benjamin Melamed is a Professor II at the Rutgers Business School- Newark and New Brunswick, Department of MSIS. Melamed received a B.Sc. degree in Mathematics and Statistics from Tel Aviv University in 1972, and a M.S. and Ph.D. degrees in Computer Science from the University of Michigan in 1973 and 1976, respectively. He was awarded an AT&T Fellow in 1988 and an IEEE Fellow in 1994. He became an IFIP WG7.3 member in 1997, and was elected to Beta Gamma Sigma in 1998. Michael N. Katehakis is Professor of Management Science in the Department of Management Science and Information Systems, at Rutgers. He studied at the University of Athens, Diploma (1974) in Mathematics, at the University of South Florida, M.A. (1978) in Statistics, and at Columbia University, Ph.D. (1980) in Operations Research. He won the 1992 Wolfowitz Prize (with Govindarajulu Z.) Phillip G. Bradford (ACM) is on the faculty in Computer Science Department at the University of Alabama. He earned his Ph.D. at Indiana University in Bloomington, his MS at The University of Kansas and his BS at Rutgers University.  相似文献

This paper presents three theorems on the peak drug levels that result from injection into a linear pharmacokinetic system. As a preliminary, the “rate of injection” is defined in terms of time expansion or time contraction of the injection function (input). The first theorem then states that the peak drug level will not be greater when the rate of injection is slow than when it is fast, if the impulse response is unimodal. The second theorem sets limits for the time of the maximum drug level, in relation to the time of the maximum of the (unimodal) impulse response and the duration of the input. The third theorem defines conditions which assure a definitely lower peak drug level if the rate of injection is slower. A graphical method is suggested for determining the times and magnitudes of the peak drug levels that result from constant infusions of a fixed dose at different rates. An example is provided to show that if the impulse response is multimodal then the peak drug level may sometimes increase with a decrease in the injection rate. PURPOSE: Does a physiologically plausible model of the retinal ganglion cell (RGC) receptive field (RF) predict the spatial tuning properties of the Hermann Grid Illusion (HGI)? METHODS: The spatial tuning of a single intersection HGI was measured psychophysically in normal observers using a nulling technique at different vertical grid line luminances. We used a model based upon a standard RGC RF, balanced to produce zero response under uniform illumination, to predict the response of the model cell to the equivalent range of stimulus conditions when placed in either the 'street' or the 'intersection' of a single element of a Hermann grid. We determined the equivalent of the nulling luminance required to balance these responses and minimise the HGI. RESULTS: The model and the psychophysical data demonstrated broad spatial tuning with similarly shaped tuning profiles and similar strengths of illusion. The line width at the peak of the model tuning function was around twice the model RGC RF centre size. Modelling the psychophysical functions gave RF centre sizes smaller than expected from human anatomical evidence but similar to that suggested by primate physiological evidence. In the model and psychophysically the strength of the illusion varied with the luminance of the vertical grid line when HGI strength was expressed as a Michelson nulling contrast, but this effect was smaller when HGI strength was expressed as a nulling luminance. CONCLUSIONS: The shape, width, height and position of the spatial tuning function of the HGI can be well modelled by a RGC RF based model. The broad tuning of these functions does not appear to require a broad range of different cell sizes either in the retina or later in the visual pathway.  相似文献

The preceding paper (Thorn, 1981) has shown that in a linear pharmacokinetic system with a multimodal impulse response the peak drug level may sometimes be smaller with slower rates of injection. This paper presents two theorems on this paradoxical injection rate effect where the injection is a constant infusion of finite duration. The first theorem establishes a graphical method for determining whether a given impulse response will give a paradoxical injection rate effect; and the second establishes that the maximum paradoxical increase in peak drug level is by a factor of two. It is further shown that in order to approach this maximum paradoxical increase the impulse response must contain two isolated, sharp, narrow pulses of approximately equal area. Some examples of bimodal arterial dye-dilution curves from the literature are discussed as impulse responses; and there is also a discussion of the behavior of drug level maxima and minima at different injection rates.  The life cycle of a giant carnivorous caddisfly, Himalopsyche japonica (Morton), was studied in two mountain streams in Nagano Prefecture, Central Japan. Field surveys and rearing experiments in the laboratory were conducted from October 1997 to September 2001. The life cycle of H. japonica was estimated to be a complex univoltine cycle that partly includes bivoltine populations. The adults had a long flight period, from April to September, with three distinct peaks of emergence. First to third instar larvae were collected from June to February, and the last (fifth) instar larvae and pupae appeared throughout the year. In autumn, the larvae belonging to all instars were found, and younger ones overwintered in the fourth instar stage and others in the fifth instar stage. On the other hand, fifth instar larvae and pupae ceased developing in autumn even though the water temperature was higher than the developmental zero temperature. The overwintered pupae emerged as adults in April, and the overwintered fifth instar larvae pupated in May and emerged in June. The larvae which overwintered in the fourth instar stage probably emerged after June. More than 100 strains ofLemna paucicostata Hegelm. collected from various localities in Japan were classified into 4 types according to the criterion of Yukawa and Takimoto (1976). N-1 type strains are widely distributed in Hokkaido, Honshu, Shikoku and Kyushu, N-2 type strains only in the area on the Japan Sea side of north Japan, K type strain only at the campus of Kyoto University, and S type strains in southern Japan. Numbers of chromosomes in somatic cells of N-1, N-2, K and S type strains were 2n=66–84, 40, 50 and 40, respectively. In N-2 and S type strains, all chromosomes (20 bivalent chromosomes) were lined up on the equatorial plate at metaphase of the first meiotic division (MI). However, in N-1 type strains, several chromosomes, and in the K type strain, more chromosomes were away from the equatorial plate at MI. Pollen fertility was 60–70%, higher than 90%, 0% and higher than 90%, in N-1, N-2, K and S type strains, respectively.  相似文献

Extracellular cAMP induces chemotaxis and cell aggregation in dictyostelium discoideum cells. cAMP added to a cell suspension is rapidly hydrolyzed (half-life of 10 s) and induces a rapid increase of intracellular cGMP levels, which reach a peak at 10 s and recover prestimulated levels at about 30 s. This recovery is not due to removal of the stimulus because the nonhydrolyzable analogue adenosine 3’,5’-monophosphorothioate-Sp- stereoisomer (cAMPS) induced a comparable cGMP response, which peaked at 10 s, even at subsaturating cAMPS concentrations. When cells were stimulated twice with the same cAMP concentration at a 30-s interval, only the first stimulus produced a cGMP response. Cells did respond to the second stimulus when the concentration of the second stimulus was higher than that of the first stimulus. By increasing the interval between two identical stimuli, the response to the second stimulus gradually increased. Recovery from the first stimulus showed first-order kinetics with a half-life of 1-2 min. The stimulation period was shortened by adding phosphodieterase to the cell suspension. The cGMP response was unaltered if the half-life of cAMP was reduced to 2 S. The peak of the transient cGMP accumulation still appeared at 10 s even when the half- life of cAMP was 0.4 s; however, the height of the cGMP peak was reduced. The cGMP response at 10 s after stimulation was diminished by 50 percent when the half-life of 10(-7) M cAMP was 0.5 s or when the half-life of 10(-8) M cAMP was 3.0 s. These results show that the cAMP signal is transduced to two opposing processes: excitation and adaptation. Within 10 s after addition of cAMP to a cell suspension the level of adaptation reaches the level of excitation, which causes the extinction of the transduction of the signal. Deadaptation starts as soon as the signal is removed, and it has first-order kinetics with a half-life of 1-2 min.  相似文献

Disjunct geographic distributions of boreal plant species at the southern edges of their ranges are expected to result in low genetic diversity and high genetic differentiation in the disjunct populations. This prediction was tested in a riparian willow, Salix arbutifolia, distributed in the northeastern Eurasian continent and the Sakhalin, Hokkaido, and Honshu Islands, using chloroplast DNA haplotypes and nuclear microsatellite genotypes. Hokkaido and Honshu populations shared a chloroplast haplotype identical to a closely related species, S. cardiophylla. This haplotype was divergent from haplotypes in the Eurasian continent (Primorsky) and the Sakhalin Island. In the nuclear microsatellites, most Hokkaido populations were genetically closer to Primorsky populations than to Sakhalin populations in spite of the geographical vicinity between Sakhalin and Hokkaido. The unexpected genetic divergence between Sakhalin and Hokkaido implies a complicated history of migration and colonization. The most peripheral populations in Honshu had the lowest genetic diversity and were most differentiated from the others. Thus, low genetic diversity and high genetic differentiation at the range periphery suggest substantial effects of genetic drift on genetic structure in the disjunct populations of Salix arbutifolia at the southern edge of its range.  相似文献

Bird colour vision is mediated by single cones, while double cones and rods mediate luminance vision in bright and dim light, respectively. In daylight conditions, birds use colour vision to discriminate large objects such as fruit and plumage patches, and luminance vision to detect fine spatial detail and motion. However, decreasing light intensity favours achromatic mechanisms and eventually, in dim light, luminance vision outperforms colour vision in all visual tasks. We have used behavioural tests in budgerigars (Melopsittacus undulatus) to investigate how single cones, double cones and rods contribute to spectral sensitivity for large (3.4°) static monochromatic stimuli at light intensities ranging from 0.08 to 63.5 cd/m2. We found no influences of rods at any intensity level. Single cones dominate the spectral sensitivity function at intensities above 1.1 cd/m2, as predicted by a receptor noise-limited colour discrimination model. Below 1.1 cd/m2, spectral sensitivity is lower than expected at all wavelengths except 575 nm, which corresponds to double cone function. We suggest that luminance vision mediated by double cones restores visual sensitivity when single cone sensitivity quickly decreases at light intensities close to the absolute threshold of colour vision.  相似文献

Two international meetings on ecological engineering, with a focus on riparian buffer zones, served as the source for selected papers in this special issue: (1) an International Workshop on Efficiency of Purification Processes in Riparian Buffer Zones: Their Design and Planning in Agricultural Watersheds, jointly organised by Hokkaido University, Japan, the National Agricultural Research Center for Hokkaido Region, Japan, Civil Engineering Research Institute of Hokkaido, Japan, and the Institute of Geography, University of Tartu, Estonia, and held from 5 to 9 November 2001 in Kushiro City, Hokkaido, Japan; and (2) an International Conference on Ecological Engineering for Landscape Services and Products, jointly organised by the International Ecological Engineering Society (IEES) and Lincoln University, Christchurch, New Zealand, and held from 25 to 29 November 2001 in Christchurch, New Zealand. At these two meetings, altogether 94 oral presentations (17 from invited speakers) and 15 posters by representatives from 21 countries were presented. The editorial paper highlights trends in investigation of the purification processes in riparian buffer zones as well as planning, design and management aspects of riparian buffers regarding the wide spectrum of their ecological functions; it characterises the two international meetings which served as sources for the selected papers and briefly explains the main aspects of these papers.  相似文献

A mathematical model is examined of interaction between cones and horizontal L-type cells (HC) in the carp retina using data from intracellular recording of HC spectral response. This model, describing the operation of ionic channels at the membrane of cones and HC, is based on a numerical analog solution to Hodgkin-Huxley differential equations [11] and enables predictions of spectral response levels in HC to be made as a function of time.M. The on- and off-discharges of three types of locust visual interneurones were recorded in response to luminous discs ranging in subtense from 0.4 to 25°. For the DCMD neurone there was a clear reciprocal relationship between ON and OFF, with large on- and small off-responses at 0.4°, and the reverse at 25° disc subtense. Most of the changeover from almost pure ON to almost pure OFF discharges occurred in the subtense range up to 2°, i.e. within the acceptance angle of one ommatidium or the angle between adjacent ommatidia. This behaviour was not found at low luminance levels, where ON and OFF followed parallel courses. These findings suggest the existence of strong inhibition, augmenting rapidly with increase in target subtense, as also with luminance as shown in separate experiments. For the DCMD the off-response had the characteristics of a simple rebound excitation, following a short period of light exposure.The other visual interneurones (M and S) behaved differently. They did not show reciprocity of ON and OFF, but instead their responses followed parallel courses with increase in subtense, resembling those of the DCMD at low luminance. Nonetheless, like the DCMD, they did show peaking of the on-response at certain subtenses and luminance, although at much higher levels. The inhibitory activity defining the responses of M and S neurones appeared much less effective than that associated with the DCMD. This could be due to greater complexity in their central connexions, of which nothing is known.  相似文献

An element-arrangement pattern is composed of two types of elements arranged differently in different regions of a pattern. Rapid texture segregation depends on spontaneously discriminating the difference in the arrangement of the elements. Five experiments investigated the perceived segregation of patterns composed of two types of squares arranged in vertical stripes in the top and bottom regions and in a checkerboard arrangement in the middle region. The squares were either equal in luminance and different in hue or equal in hue and different in luminance. The rated similarities of the two hues in a pattern failed to predict perceived segregation. For a given background luminance, the perceived segregation was predicted by the square-root of the sum of the squares of the differences in the outputs of the L - M + S and L + M - S opponent channels, where L, M, and S were the cone contrasts of the long-, medium-, and short-wavelength receptors. The perceived similarity of the two hues in a pattern was not affected by the background luminance but was a function of cone excitations instead. For patterns differing in hue and equal in luminance, perceived segregation was an inverse function of the background luminance. A white background decreased the perceived segregation, but a black background did not. The effect of background luminance was not on the discrimination of the individual hues. The two hues making up a texture pattern were clearly distinguishable on a white background. A white background interfered with the discrimination of the vertical and diagonal columns of squares that distinguished the texture regions. For patterns differing in luminance and equal in hue, black and white backgrounds decreased the perceived segregation. The results indicate that adapting to an achromatic luminance distant from the luminance of the squares increased the Weber threshold for discriminating luminance differences, but did not increase the Weber threshold for discriminating hue differences. The experiments also revealed that luminance was the primary factor affecting perceived segregation and that perceived brightness is secondary. The results are consistent with the hypothesis that perceived segregation in element-arrangement patterns is primarily a function of the differences in the outputs of relatively early filtering mechanisms that encode pattern differences prior to the specification of the element shapes and their properties.  相似文献
