Water‐soluble graphene oxide (GO) with a two‐dimensional layered nanostructure was synthesized and used as a quencher to construct a highly sensitive and selective fluorescence resonance energy transfer (FRET) aptasensor for sensing Immunoglobulin E (IgE). The fluorescein isothiocyanate (FITC)‐labeled aptamer could be adsorbed stably onto the surface of GO via π → π stacking interaction, which led to the occurrence of FRET from FITC to GO, and the fluorescence of FITC‐labeled aptamer was quenched by GO via energy transfer. In the presence of IgE, the fluorescence was recovered due to a higher affinity between the aptamer and IgE compared with interactions between GO and the aptamer, leading to a high signal‐to‐background ratio. The fluorescence intensity of the aptamer increased in proportion to the amount of IgE in the sample,so that IgE could be detected with a linear range of 60–225 pM and a detection limit of 22 pM. The assay was highly selective because the aptamer was unaffected by the presence of immunoglobulin G (IgG), human serum albumin (HSA) and bovine serum albumin (BSA). A new fluorimetric aptasensor was designed for the determination of adenosine triphosphate (ATP) based on magnetic nanoparticles (MNPs) and carbon dots (CDs). In this analytical strategy, an ATP aptamer was conjugated on MNPs and a complementary strand of the aptamer (CS) was labeled with CDs. The aptamer and its CS were hybridized to form a double helical structure. The hybridized aptamers could be used for the specific recognition of ATP in a biological complex matrix using a strong magnetic field to remove the interfering effect. In the absence of ATP, no CDs–CS could be released into the solution and this resulted in a weak fluorescence signal. In the presence of ATP, the target binds to its aptamer and causes the dissociation of the double helical structure and liberation of the CS, such that a strong fluorescence signal was generated. The increased fluorescence signal was proportional to ATP concentration. The limit of detection was estimated to be 1.0 pmol L–1 with a dynamic range of 3.0 pmol L–1 to 5.0 nmol L–1. The specific aptasensor was applied to detect ATP in human serum samples with satisfactory results. Moreover, molecular dynamic simulation (MDS) studies were used to analyze interactions of the ATP molecule with the aptamer.  相似文献

Water‐soluble CdS quantum dots (QDs) were prepared using mercaptoacetic acid (TGA) as the stabilizer in an aqueous system. A fluorescence resonance energy transfer (FRET) system was constructed between water‐soluble CdS QDs (donor) and Eosin Y (acceptor). Several factors that impacted the fluorescence spectra of the FRET system, such as pH (3.05–10.10), concentration of Eosin Y (2–80 mg/L) and concentration of CdS QDs (2–80 mg/L), were investigated and refined. Donor‐to‐acceptor ratios, the energy transfer efficiency (E) and the distance (r) between CdS QDs and Eosin Y were obtained. The results showed that a FRET system could be established between water‐soluble CdS QDs and Eosin Y at pH 5.0; donor‐to‐acceptor ratios demonstrated a 1: 8 proportion of complexes; the energy transfer efficiency (E) and the distance (r) between the QDs and Eosin Y were 20.07% and 4.36 nm,respectively. Water safety is one of the most pervasive problems afflicting people throughout the world. Microcystin, a hepatotoxin produced by cyanobacteria, poses a growing and serious threat of water safety. According to World Health Organization (WHO), the limit of content of microcystin-LR (MC-LR) in drinking water is as low as 1μg/L; it is thus necessary to explore a sensitive method for the trace detection of microcystins (MCs). Based on the observation of gold nanoparticles (Au NPs) induced graphene oxide (GO) fluorescence quenching, a reliable biosensor was developed here for microcystins detection. MCs could be attached on Au NPs through the interaction with single strand-DNA (ss-DNA) modified on Au NPs, which formed Au-DNA-MCs complexes. These MCs in the complexes could be immunologically recognized by the antibodies adsorbed on GO sheets, as a result, Au NPs were close enough to quench the photoluminescence of GO by the fluorescence resonance energy transfer (FRET). The fluorescence intensity decreased with the increase of MCs as more Au NPs linked onto GO surface. The limit of detection was 0.5 and 0.3μg/L for microcystin-LR and microcystin-RR (MC-RR), respectively, which satisfies the strictest standard of WHO. Well defined results were also obtained in natural lake water and the specificity experiment. The antibody used here could recognize Adda group, the conservative part of MCs, which allowed the biosensor to detect both single toxin and the total content of MCs existing in the water sample.  相似文献

A novel sensing system based on the near infrared (NIR) fluorescence resonance energy transfer (FRET) between Mn:CdTe quantum dots (Qdots) and Au nanorods (AuNRs) was established for the detection of human IgG. The NIR-emitting Qdots linked with goat anti-human IgG (Mn:CdTe-Ab1) and AuNRs linked with rabbit anti-human IgG (AuNRs-Ab2) acted as fluorescence donors and acceptors, respectively. FRET occurred by human IgG with the specific antigen–antibody interaction. And human IgG was detected based on the modulation in FRET efficiency. The calibration graph was linear over the range of 0.05–2.5 μM of human IgG under optimal conditions. The proposed sensing system can decrease the interference of biomolecules in NIR region and increase FRET efficiency in optimizing the spectral overlap of AuNRs with Mn:CdTe Qdots. This method has great potential for multiplex assay with different donor–acceptor pairs.  相似文献

A novel and sensitive method for the determination of ceftazidime and cefepime in an active pharmaceutical ingredient (API) has been developed based on the fluorescence quenching of poly(ethylene glycol) (PEG)2000‐capped carbon quantum dots (CQDs) prepared using a chemical oxidation method. The quenching of fluorescence intensity is proportional to the concentration of ceftazidime and cefepime over the range of 0.33–3.30 and 0.24–2.40 µg/mL, respectively. The mode of interaction between PEG2000‐capped CQDs and ceftazidime/cefepime in aqueous solutions was investigated using a fluorescence, UV/Vis and Fourier transform infrared spectrometry (FTIR) at physiological pH. UV/Vis and FTIR spectra demonstrated that ground state compounds were formed through hydrophobic interaction the fluorescence quenching of CQDs caused by ceftazidime and cefepime. We have developed a sensitive, one-step, homogeneous open sandwich fluoroimmunoassay (OsFIA) based on fluorescence resonance energy transfer (FRET) and luminescent semiconductor quantum dots (QDs). In this FRET assay, estrogen receptor beta (ER-beta) antigen was incubated with QD-labeled anti-ER-beta monoclonal antibody and Alexa Fluor (AF)-labeled anti-ER polyclonal antibody for 30 min, followed by FRET measurement. The dye separation distance was estimated between 80 and 90 A. The current method is rapid, simple, and highly sensitive, and it did not require the bound/free reagent separation steps and solid-phase carriers. A concentration as low as 0.05 nM (2.65 ng/ml) receptor was detected with linearity. In addition, the assay was performed with commercial antibodies. This assay provides a convenient alternative to conventional, laborious sandwich immunoassays.  相似文献

Sensitive detection of doxorubicin (DOX) is critical for clinical theranostics. A novel ratiometric fluorescence strategy based on the inner filter effect (IFE) has been established for the sensitive detection of DOX by designing a ratiometric fluorescence probe. In the presence of DOX, the fluorescence intensity of copper nanoclusters (CuNCs) at 485 nm decreases, and the fluorescence intensity of carbon dots at 560 nm increases. Therefore, DOX can be quantitatively detected by measuring the ratio of the fluorescence intensities at 560 and 485 nm (F560/F485). The F560/F485 ratio exhibits a linear correlation with the DOX concentration in the range from 1.0 × 10−8 M to 1.0 × 10−4 M with the detection limit of 3.7 nM. Furthermore, this method was also successfully applied to the analysis of DOX in human plasma samples, affording an effective platform for drug safety management.  相似文献

Metal atom doping, an easy and convenient method, can optimize and tune the physical–chemical properties and photometrics of carbon dots (CDs). However, there are few reports on the preparation of metal‐decorated CDs that give red emission and a high photoluminescence quantum yield (PLQY). Here, we demonstrate a zinc (existing in human body) ion‐doping strategy to observably enhance the PLQY and lengthen the CD emission wavelength. The prepared Zn/ZnO‐decorated CDs (Zn‐CDs) produced red fluorescence (623 nm) with a superior PLQY of 40.3%. Through a series of analyses, Zn‐CDs were confirmed to contain an oxidation state and reduction state of Zn doping into the internal defects and surface of Zn‐CDs. More valuably, the Zn‐CDs had excellent chemical stability, photo‐stability, long‐term storage stability, and high biocompatibility, and therefore could be used as a robust red fluorescence probe for high‐quality cellular imaging.  相似文献

The as‐prepared graphene oxide (GO) exhibited a strong catalytic effect on reduction of HAuCl4 by trisodium citrate to form gold nanoplasmons (AuNPs) with a strong surface‐enhanced Raman scattering (SERS) effect at 1615 cm?1 in the presence of molecular probe Victoria blue 4R (VB4r). SERS intensity increased with nanocatalyst GO concentration due to the formation of more AuNP substrates. The aptamer (Apt) of Hg2+ can bind to GO to form Apt–GO complexes, which can strongly inhibit nanocatalysis. When target Hg2+ is present, the formed stable Hg2+–Apt complexes are separated from the GO surface, which leads to GO catalysis recovery. The enhanced SERS signal was linear to Hg2+ concentration in the range 0.25–10 nmol/L, with a detection limit of 0.08 nmol/L Hg2+. Thus, a new gold nanoplasmon molecular spectral analysis platform was established for detecting Hg2+, based on Apt regulation of GO nanocatalysis.  相似文献

Endocrine‐disrupting chemicals are a major public health problem throughout the world. In the human body, these compounds functionalize the same as sexual hormones, inducing precocious puberty, gynecomastia, etc. To help prevent this occurrence, a simple detection system is needed. In this study, a nonylphenol ethoxylate (NPE)‐specific aptamer was selected by reduced graphene oxide‐systematic evolution of ligands by exponential enrichment. A random ssDNA library was incubated with rGO for adsorption, followed by elution with the target molecule. As a result of screening, a DNA aptamer was found that specifically bounds to the target with high binding affinity (Kd = 100.9 ± 13.2 nM) and had a low limit of detection (LOD = 696 pM). Furthermore, this NPE‐binding aptamer bounds selectively to the target. Characterization of the aptamer was confirmed by measuring the fluorescence signal recovery from rGO. In addition, detection of NPE was performed with several water samples, and the detection accuracy was 100 ± 10%. From these results, we expect that this aptamer could be applied to an on‐site detection system for NPE in industrial sites or domestic fields.  相似文献

In this work, a turn‐off photoluminescent sensing proof‐of‐concept based on blue luminescent graphene quantum dots (GQDs) as the fluorescent probe was developed. For that purpose, GQDs optical response was related with the catalytic enzymatic activity of alkaline phosphatase (ALP), in the presence of hydroquinone diphosphate (HQDP). The hydrolysis of HQDP by ALP generated hydroquinone (HQ). The oxidation of HQ, enzymatically produced, to p‐benzoquinone (BQ) resulted in the quenching of GQDs fluorescence (FL). Therefore, the developed luminescent sensing mechanism allowed the FL quenching with ALP activity to be related and thus quantified the concentration of ALP down to 0.5 nM of enzyme. This innovative design principle appears as a promising tool for the development of enzymatic sensors based on ALP labeling with fluorescent detection or even for direct ALP luminescent quantification in an easy, fast and sensitive manner.  相似文献

The development of a rapid, sensitive, and straightforward detection method of prostate‐specific antigen (PSA) is indispensable for the early diagnosis of prostate cancer (PCa). This work relates an electrochemical method using functionalized single‐stranded DNA aptamer to diagnose PCa and benign prostate hyperplasia. The sensing platform relies on PSA recognition by aptamer/Au/GO‐nanohybrid‐modified glassy carbon electrode. Besides ferrocyanide TiO2/carbon quantum dots (CQDs) probe is used to investigate the effect of nanoparticle‐containing electrolyte. Optimization of incubation time of aptamer/Au/GO‐nanohybrid and volume fraction of nafion were done using Design Expert 10 software reporting 42.4 h and 0.095% V/V, respectively. In ferrocyanide medium, PSA detection as low as 3, 2.96, and 0.85 ng mL−1 was achieved with a dynamic range from 0.5 to 7 ng ml−1, in accord with clinical values, using cyclic voltammetry, square wave voltammetry, and electrochemical impedance spectroscopy, respectively. Moreover, this sensor exhibited conspicuous performance in TiO2/CQDs‐containing medium with different pH values of 5.4 and 8 to distinguish total PSA and free PSA, resulting in very low limit of detections, 0.028, and 0.007 ng ml−1, respectively. The results manifested the proposed system as a forthcoming sensor in a clinical and point of care analysis of PSA.  相似文献

The development of advanced methods for accurately monitoring cell apoptosis has extensive significance in the diagnostic and pharmaceutical fields. In this study, we developed a rapid, sensitive and selective approach for the detection of cell apoptosis by combining the site‐specific recognition and cleavage of the DEVD–peptide with quantum dots (QDs)‐based fluorescence resonance energy transfer (FRET). Firstly, biotin‐peptide was conjugated on the surface of AuNPs to form AuNPs‐pep through the formation of an Au‐S bond. Then, AuNPs–pep–QDs nanoprobe was obtained through the connection between AuNPs–pep and QDs. FRET is on and the fluorescence of QDs is quenched at this point. The evidence of UV–vis spectra, transmission electron microscopy (TEM), and Fourier transform infrared (FT‐IR) spectroscopy revealed that the connection was successful. Upon the addition of apoptosis cell lysis solution, peptide was cleaved by caspase‐3, and AuNPs was dissociated from the QDs. At this time, FRET is off, and thus the QDs fluorescence was recovered. The experimental conditions were optimized in terms of ratio of peptide to AuNPs, buffer solution, and the temperature of conjugation and enzyme reaction. The biosensor was successfully applied to distinguishing apoptosis cells and normal cells within 2 h. This study demonstrated that the biosensor could be utilized to evaluate anticancer drugs.  相似文献

When excited at 435 nm, tetra‐sulfonate zinc phthalocyanine (ZnPcS4) emitted dual fluorescence at 495 and 702 nm. The abnormal fluorescence at 495 nm was experimentally studied and analyzed in detail for the first time. The abnormal fluorescence at 495 nm was deduced to originate from triplet–triplet (T–T) energy transfer of excited phthalocyanine (3*ZnPcS4). Furthermore, graphene quantum dots (GQDs) enhanced the 495 nm fluorescence quantum yield (Q) of ZnPcS4. The fluorescence properties of ZnPcS4–GQDs conjugate were retained in a cellular environment. Based on the fluorescence of ZnPcS4–GQDs conjugate, we designed and prepared an Apt29/thrombin/Apt15 sandwich thrombin sensor with high specificity and affinity. This cost‐saving, simple operational sensing strategy can be extended to use in sensing/imaging of other biomolecules.  相似文献

In this paper, based on the fluorescence of carbon quantum dots (CQDs) quenched by mercury ions (Hg2+) and the nonresponse of Hg2+ to rhodamine B fluorescence, a dual emission ratio fluorescence sensor was constructed to realize the quantitative detection of Hg2+. Under excitation at 365 nm, the fluorescence spectrum showed double emission peaks at 437 nm and 590 nm, corresponding to the fluorescence emissions of CQDs and rhodamine B, respectively. This method quantitatively detected Hg2+ based on the linear relationship between the ratio of the intensities of the two emission peaks F437/F590 and the concentration of Hg2+. The detection range was 10–70 nM, and the limit of detection (S/N = 3) was 3.3 nM. In addition, this method could also realize the qualitative and semiquantitative detection of Hg2+ according to the fluorescence colour change of the probe under ultraviolet light. After various evaluations, the method could be successfully applied to the quantitative and visual detection of Hg2+ in tap water, and demonstrated excellent selectivity, anti-interference performance, and repeatability of the method.  相似文献

Dual emission carbon dots have a high potential for use as fluorescence‐based sensors with higher selectivity and sensitivity. This study demonstrated the possibility of conversion of a biological molecular system with a single emission peak to a double emission carbon dots system. This report is the first to describe the synthesis of dual emission carbon dots by tuning the electronic environment of a conjugated system. Here we prepared carbon dots from a natural extract, from which carotenoids were used as a new source for carbon dots. Formation of the carbon dots was confirmed by images obtained under a transmission electron microscope as well as from a dynamic light scattering study. The prepared carbon dots system was characterized and its optical property was monitored. The study showed that, after irradiation with microwaves, the fluorescence intensity of the whole system changed, without any change in the original peak position of the carotenoid but with the appearance of an additional peak. A Fourier transform infrared study confirmed breaking of the conjugated system. When using ethylene glycol as a surface passivating agent added to these carotenoid carbon dots, the dual emission spectra became more distinct.  相似文献

In this work, we proposed a novel biosensor to homogeneously detect concanavalin A (ConA) using pyrene-conjugated maltose assembled graphene based on fluorescence resonance energy transfer (FRET). Maltose-grafted-aminopyrene (Mal-Apy) was synthesized and characterized by mass spectra, UV-vis and fluorescence spectra. The Mal-Apy was further employed for fluorescence switch and ConA recognition. When Mal-Apy was self-assembled on the surface of graphene by means of π-stacking interaction, its fluorescence was adequately quenched because the graphene acted as a "nanoquencher" of the pyrene rings due to FRET. As a result, in the presence of ConA, competitive binding of ConA with glucose destroyed the π-stacking interaction between the pyrene and graphene, thereby causing the fluorescence recovery. This method was demonstrated the selective sensing of ConA, and the linear range is 2.0 × 10?2 to 1.0 μM with the linear equation y=1.029x + 0.284 (R = 0.996). The limit of detection for ConA was low to 0.8 nM, and the detection of ConA could be performed in 5 min, indicating that this method could be used for fast, sensitive, and selective sensing of ConA. Such data suggests that the graphene FRET platform is a great potential application for protein-carbohydrate studies, and would be widely applied in drug screening, bimolecular recognition and disease diagnosis.  相似文献

Urethane acrylate (UA) was used to prepare carbon quantum dots (C‐dots) luminescent membranes and the resultants were examined with FT‐IR, mechanical strength, scanning electron microscope (SEM) and quantum yields (QYs). FT‐IR results showed the polyurethane acrylate (PUA) prepolymer –C = C‐vibration at 1101 cm?1 disappeared but there was strong vibration at1687cm?1which was contributed from the–C = O groups in cross‐linking PUA. Mechanical strength results showed that the different quantity of C‐dots loadings and UV‐curing time affect the strength. SEM observations on the cross‐sections of the membranes are uniform and have no structural defects, which prove that the C‐dots are compatible with the water‐soluble PUA resin. The C‐dot loading was increased from 0 to 1 g, the maximum tensile stress was nearly 2.67 MPa, but the tensile strain was decreased from 23.4% to 15.1% and 7.2% respectively. QYs results showed that the C‐dots in the membrane were stable after 120 h continuous irradiation. Therefore, the C‐dots photoluminescent film is the promising material for the flexible devices in the future applications.  相似文献

A novel and sensitive chemiluminescence (CL) procedure based on the synergetic catalytic effects of gold nanoclusters (Au NCs) and graphene quantum dots (GQDs) was developed for the reliable measurement of cimetidine (CM). The initial experiments showed that the KMnO4‐based oxidation of alkaline rhodamine B (RhoB) generated a very weak CL emission, which was intensively enhanced in the simultaneous presence of Au NCs and GQDs. CL intermediates can be adsorbed and gathered on the surface of Au NCs, becoming more stable. GQDs participate in the energy transferring processes and facilitate them. These improving effects were simultaneously obtained by adding both Au NCs and GQDs into the RhoB‐KMnO4 reaction. Consequently, the increasing effect of the Au NCs/GQDs mixture was more than that of pure Au NCs or GQDs, and a new nano‐assisted powerful CL system was achieved. Furthermore, a marked quenching in the emission of the introduced CL system was observed in the presence of CM, so the system was examined to design a sensitive sensor for CM. After optimization of influencing parameters, the linear lessening in CL emission intensity of KMnO4‐RhoB‐Au NCs/GQDs was verified for CM concentrations in the range 0.8–200 ng ml?1. The limit of detection (3Sb/m) was 0.3 ng ml?1. Despite being a simple CL method, good sensitivity was obtained for CM detection with reliable results for CM determination in human urine samples.  相似文献
