The purpose of this report is to demonstrate that chorionic gonadotropin (CG) interacts nonspecifically with lactate dehydrogenase changing its catalytic activity. The binding in vitro of CG (5 mg/ml) to the enzyme is accompanied by conformational changes of its protein and kinetic characteristics. It may be possible that chorionic gonadotropin exercises a controlling influence on lactate dehydrogenase activity in the cell. 相似文献

The purpose of the study is evaluation of radioprotective effectiveness of indometofen at its prophylactic administration in conditions of acute irradiation. Evaluation of radioprotective efficiency was performed by studying the 30-day survival rate, life expectancy, structure of deathly irradiated mice, and bone marrow hemopoiesis using methods of endogenous and exogenous colony formation. The prophylactic application of indometofen at doses 30 mg/kg for 5 days before irradiation has been observed to protect mice against radiation death induced by gamma or X-ray exposures at doses LD(50-70/30), increasing their survival rate by 16-44%, and reduce severity of post radiation disorders of bone marrow hemopoiesis. 相似文献

In dogs, degenerative joint diseases (DJD) have been shown to be associated with increased lactate dehydrogenase (LDH) activity
in the synovial fluid. The goal of this study was to examine healthy and degenerative stifle joints in order to clarify the
origin of LDH in synovial fluid. In order to assess the distribution of LDH, cartilage samples from healthy and degenerative
knee joints were investigated by means of light and transmission electron microscopy in conjunction with immunolabeling and
enzyme cytochemistry. Morphological analysis confirmed DJD. All techniques used corroborated the presence of LDH in chondrocytes
and in the interterritorial matrix of healthy and degenerative stifle joints. Although enzymatic activity of LDH was clearly
demonstrated in the territorial matrix by means of the tetrazolium–formazan reaction, immunolabeling for LDH was missing in
this region. With respect to the distribution of LDH in the interterritorial matrix, a striking decrease from superficial
to deeper layers was present in healthy dogs but was missing in affected joints. These results support the contention that
LDH in synovial fluid of degenerative joints originates from cartilage. Therefore, we suggest that (1) LDH is transferred
from chondrocytes to ECM in both healthy dogs and dogs with degenerative joint disease and that (2) in degenerative joints,
LDH is released from chondrocytes and the ECM into synovial fluid through abrasion of cartilage as well as through enhanced
diffusion as a result of increased water content and degradation of collagen. 相似文献

L-Lactate dehydrogenase (LD) catalyzes the interconversion of pyruvate and lactate. Using a spectrophotometric assay to determine LD activity, incubation of rabbit, porcine, and bovine LD-1 and LD-5 isozymes with the protease subtilisin (Carlsberg) gave first-order degradation kinetics. Degradation half-lives were significantly lower for the LD-5 isozymes from the three species when incubated with subtilisin at temperatures from 4 degrees C to 25 degrees C. The energy involved in the degradation process, however, was not different. The activation energy for the conversion of pyruvate to lactate by LD-1 at pH 7.4 was significantly higher than that for LD-5 for all three species examined (P less than 0.005). Thermocalorimetry showed that the LD-1 isozymes have both a higher mean temperature of denaturation and a higher heat uptake during the denaturation process than corresponding LD-5 forms. The results suggest that the LD-5 isozymes in the species studied are more metabolically efficient, whereas the LD-1 forms have greater structural stability. 相似文献

Lactate dehydrogenase activities were measured along two sinusoidal paths (1) between small portal tracts and central veins and (2) between regions of adjoining septal branches and central veins in the livers of male Wistar rats, using a Lowry technique. The established profiles of enzyme activity provide further support of functional heterogeneity of liver sinusoids and their abutting hepatocytes related to morphological differences of the sinusoidal bed. Within the hepatocytes a pronounced heterogeneity in enzyme activity was recorded surrounding small portal tracts and central veins. The lowest values of activity were determined in those cells located in close proximity to the vessels, which emphasizes their exceptional morphological and functional position. 相似文献

The influence of seasonal variations in the temperature of the environment on the kinetic parameters of LDH have been followed in the shrimp Palaemon serratus. The results obtained show that this system compensates partially the effects of the temperature on its activity. The complex pattern of the monthly variations of Km values along the year can be resolved into a 2 plateaux system corresponding to the extreme temperature of the environment. 相似文献

An electrophoretically homogeneous lactate dehydrogenase was isolated from soybean seedlings, the specific activity of which was approximately 1800 times higher than the crude extract. From the dependence of the rate of reaction catalyzed by lactate dehydrogenase on substrate concentration, Michaelis constants and Hill coefficients were determined for four natural substrates,i.e. lactate, pyruvate, NAD and NADH. The enzyme from soy-bean plants is non-competitively inhibited by oxalate and mesoxalate,i.e. by the compounds analogous to the substrate. At pyruvate concentrations above 0.8 mM, the rate of reaction catalyzed by lactate dehydrogenase from soy-bean plants decreases, fructose diphosphate and ATP function as inhibitors as well. The inhibition by ATP is pH dependent, which seems to be of importance for the regulation of enzyme activityin vivo. 相似文献

The content of six lactate dehydrogenase isoenzymes in testes of rats exposed to electromagnetic field of 3-cm wavelength range was studied. The changes in their percent contents were found to be inhomogeneous compared with control. It is assumed that electromagnetic radiation affects the organs of the human urinogenital system. The results can be used for estimating the safety of persons professionally exposed to electromagnetic radiation of the industrial frequency range and in the therapy of diseases of the urinogenital system. 相似文献
