Although the development of the soybean ovule has been fairlywell studied, knowledge of the sequence of events in the seedcoat during the first 3 weeks after flowering is incomplete.The goal of the present study was to document, using light microscopy,the early development of the soybean seed coat with respectto changes in structure and histochemistry. At anthesis, theseed coat consists of an outer layer of cuboidal epidermal cellssurrounding several layers of undifferentiated parenchyma (whichtogether constitute the outer integument), and an inner layerof cuboidal endothelial cells (the inner integument). At 3 dpost anthesis (dpa), the inner integument has expanded to includethree to five layers of relatively large cells with thick, heavily-stainingcell walls immediately adjacent to the endothelium. By 18 dpa,the outer integument has developed into a complex of tissuescomprised of an inner layer of thick-walled parenchyma, an outerlayer of thin-walled parenchyma containing vascular tissue whichhas grown down from the lateral vascular bundles in the hilumregion, a hypodermis of hourglass cells, and palisade layer(epidermis). The thick-walled parenchyma of the inner integumenthas become completely stretched and compressed, leaving a single,deeply staining wall layer directly above the endothelium. At21 dpa, the outermost cells of the endosperm have begun to compressthe endothelium. At 45 dpa (physiological maturity) the seedcoat retains only the palisade layer, hourglass cells, and afew layers of thin-walled parenchyma. The innermost layer ofthe endosperm, the aleurone layer, adheres to the inside ofthe seed coat. This knowledge will be invaluable in future studiesof manipulation of gene expression in the seed coat to modifyseed or seed coat characteristics. Ovular development of Magnolia grandiflora was examined to determine the morphology of the outer integument. At the time the ovular primordium begins incurving, the outer integument is initiated subdermally, and later the inner integument arises from the dermal layer. Whereas the inner integument is annular, the outer integument is formed as a semiannular rim interrupted on the concave side of the funiculus. Later the outer integument becomes a hood-shaped envelope. The obturator is formed as a transversely elongate outgrowth on the concave side of the funiculus and intervenes in the gap of the outer integument. During further development the inner integument produces several distal lobes, and the outer integument becomes bilobed. The exostome is a transverse slit with a middle notch, formed by the outer integument and the obturator. Presumed wide occurrence of the hood-shaped outer integument in primitive families suggests that it is a primitive state. The outer integument is compared with the ovuliferous sporophylls of the glossopterid and caytonialean seed ferns.  相似文献

Floral phenology and ovular development ofLiriodendron tulipifera are described. The ovule primordia are initiated in December, followed by prominent development in March, and the ovules are mature in May. The inner integument is formed as an annular rim on the incurving ovule primordia, but the outer integument develops as a semi-annular rim interrupted on the concave side of the funicle. Later, an outgrowth, which is interpreted here as an obturator, arises on the concave side of the funicle. The funicular outgrowth arises far from the inner integument, while the outer integument is close to the inner. The outer integument and the funicular outgrowth together form an envelope complex. Later the outer integument produces two distal lobes, which disappear at maturity. Mature ovules of the threeMagnolia species examined have similar lobes. It is suggested that the hood-shaped outer integument is primitive in angiosperms.  相似文献

In the ovules of Vanilla (Vanilla planifolia Andr.) before fertilization, outer integument surrounded the lower part of ovule. Uranin got into ovule through funiculus, forming, the first center of fluorescence at the chalaza zone of ovule. Then uranin was transported to micropyle end along inner integument, forming the second center of fluorescence at micropyle end of inner integument. Soon, fluorescence appeared in the egg apparatua. After fertilization, the outer integument ovule extended upward, forming micropyle ogerber with inner integument. After getting into ovule through funiculus, uranin spreads to- ward several directions: l. transported to outer integument at the entrance of micropyle; 2. transported downward to chalaza zone along outer integument at the side of funiculus; 3. extended from chalaza zone to the inside and to the outer integument at the side far from funiculus The ovules of Vanilla had no vascular bundles. On transporting in inner integument, however, the cells in inner layer next to the embryo sac appeared to be the major passage. In mature embryo sac, there was cuticle between inner integument and embryo sac at the half of micropyle end. But between embryo sac at the half of chalaza end and nucellus, cuticle was absent. Nutrient could get into embryo sac from chalaza end undoubtedly. As egg apparatus showed the fluorescence after formation of fluorescence center of inner integument at micropylar end, the possibility that nutrient got into embryo sac from micropyle could not be excluded.  相似文献

The embryological characteristics and ovular integument development of the dioecious species Woonyoungia septentrionalis (Dandy) Law (Magnoliaceae), which are poorly understood, were investigated under laser scanning confocal microscope (LSCM) and light microscope (LM). The embryological characteristics conform to most of the previously studied species in Magnoliaceae. The anther has 4 microsporangia, and the anther wall develops according to the dicotyledonous type. Cytokinesis at meiosis of the microspore mother cells follows a modified simultaneous type, giving rise to isobilateral or decussate tetrads, and a cell plate is absent, but a membrane was observed. Mature pollen grains are 2‐cellular and have high germination rates. The ovule is anatropous, crassinucellate and bitegmic, and meiotic result in linear tetrads of megaspores, the one at the chalazal end functions directly as an embryo‐sac cell. The development of the embryo sac is of the Polygonum‐type and endosperm formation is of the nuclear type. The outer integument of the ovule differentiates into an outer fleshy and an inner stony layer while the inner integument is reduced to a tanniniferous layer. The normal embryological development, high germination rates of pollen and high seed set indicate that the primary reason for the decline of the species is not to be found in these developmental processes.  相似文献

TOMLINSON, P. B., TAKASO, T. & RATTENBURY, J. A., 1989. Cone and ovule ontogeny in Phyllocladus (Podocarpaceae). Cones are borne directly on phylloclades, usually in the position of basal segments or as segment appendages. Each cone consists of a series of spirally arranged bracts, of which the middle bracts each subtend a single, sessile ovule. There is no ovuliferous scale. Ovules arise as ovoid outgrowths; integument development involves periclinal divisions of hypodermal cells with the integument becoming bilobed and extended laterally. The mature ovule is flask-shaped. The integument includes an extensive middle region bounded by an inner and outer epidermis; the outer hypodermis is differentiated as two contrasted cell layers. An aril differentiates late by periclinal divisions of the outer hypodermal cells at the base of the ovule. The three outermost layers of the integument become differentiated in the mature seed as an epidermis, with thick, cutinized outer tangential walls, an outer hypodermal tanniniferous layer and a sclerotic inner layer. Each ovule is vascularized by two strands that diverge from the axial bundles delimiting the gap left by the departing bract trace.  相似文献

The development of the floral bud, especially the ovule and seed coat, of Sinomanglietia glauca was observed. Floral buds were covered by eight to nine hypsophyll pieces. The hypsophyll nearest the tepal was closed completely and characterized by two arrays of densely stained cells with dense cytoplasm, which split longitudinally at flowering. The perianth consisted of 16 tepals arranged in three whorls. The gynoecium was composed of numerous apocarpous carpels; the ovule was anatropous with two integuments. Embryogenesis was of the Polygonum type, and the endosperm was nuclear. The inner integument degenerated during seed development. The seed of S. glauca had an endotestal seed coat comprised of a sclerotic layer derived from the inner adaxial epidermis of the outer integument and a sarcotesta derived mainly from the middle cells between the inner and outer epidermis of the outer integument. The embryo developed normally, so embryogenesis is not the cause of difficult regeneration.  相似文献

Illus. 1961.—The seed coat of Musa balbisiana Colla consists of a relatively thick outer integument and a 2–cell-layered inner integument. The entire seed coat is sclerified, but routine tests for lignin are negative. Within the outer integument there is a zone of unusual sclereids tentatively termed “multiluminate.” Between the inner integument and the remnants of the nucellus is a cuticle 10–12 μ thick. The micropylar plug and collar are typical of the genus. The chalazal mass is an annular region of gelatinous cells. The mature embryo is comprised of a massive cotyledon, an epicotyl with 1 leaf primordium, a primary root primordium, and several adventitious root primordia. Procambium is well developed, but no mature vascular elements are present in the embryo.  相似文献

Bambusa tulda and Thyrsostachys siamensis resemble each other in having an obovate ovary which is hairy and thickened along the apex, a pseudo-crassinucellate ovule with a wide region of attachment, poorly-developed and ephemeral outer integument, an inner integument which fails to grow beyond the nucellus, 'Polygonum' type of embryo sac ontogeny, parallel orientation of embryo sac to the long axis of the ovule, multiple antipodals which retain apical position in the embryo sac even during post-fertilization phase of development, an ephemeral nucellus, relatively small bambusoid embryos, and many-layered and apically thickened pericarp. However, they differ from each other in their gynoecial structure, the extent of the development of the outer integument, organization of megaspore tetrads and development-stage-related behaviour of the inner integument in the fertilized ovules. These taxa also differ from other members of the subfamily Bambusoideae in the structure of the mature ovule, endosperm and pericarp.  相似文献

Cells of the unicellular cyanobacteriumGloeothece sp. PCC 6909 are surrounded by an inner (enclosing 1–2 cells) and an outer (enclosing cell groups) sheath. Using conventional Epon-embedding in combination with ruthenium-red staining, the inner and outer sheaths appeared similar and displayed multiple bands of electron-dense subunits. However, embedding in Nanoplast resin to avoid shrinkage led to the detection of two distinct zones (inner and outer zone) each with several distinct layers. The zone delimited by the electron-dense thick inner sheath layer, and the zone enclosed by the thin electron-dense outer sheath layer, are composed of a homogeneous material of little electron-contrast. Whereas the outer zone appears to be of even contrast, the inner zone is characterized by a distinct electron-transparent layer. Element distribution analysis revealed that the electron-transparent layer contained relatively large amounts of sulfur, carbon, and oxygen but only little nitrogen.Inner and outer sheath fractions were isolated by differential mechanical cell breakage and centrifugation. The outer sheath fraction was less hydrated than the inner one. The two fractions differed little in their contents of uronic acids, carbohydrate and protein, although the outer sheath fraction contained less sulfate. A soluble polysaccharide with a chemical composition similar to that of inner and outer sheath fractions was also obtained from the culture supernatant.  相似文献

MANNING, J. C. & BRITS, G. J., 1993. Seed coat development in Leucospermum cordifolium (Knight) Fourcade (Proteaceae) and a clarification of the seed covering structures in Proteaceae . The development of the seed coat and pericarp is studied in Leucospermum cordifolium from ovule to mature seed. The ovule and seed are characterized by a tegmic pachychalaza. The pericarp is adnate to the integuments from anthesis and remains unthickened to maturity. The outer integument forms the seed coat and the seed is endotestal: the outer epidermis becomes tanniniferous and the inner epidermis develops into a crystalliferous palisade. The inner integument degenerates at an early stage. Examination of the literature reveals that the crystal palisade layer of the outer integument has been erroneously assumed to constitute an endocarp. This finding indicates that a re-interpretation of all published information on the seed coat in indehiscent Proteaceae is necessary before any speculations on the phylogenetic significance of the seed coat can be entertained.  相似文献

The micropyle and the integuments of sugar beet (Beta vulgaris) ovules have been investigated by light and electron microscopy during differentiation and maturation of the ovule. The micropyle itself is formed by the inner integument which is surrounded by the outer integument at its base. The micropyle containts a fibrillar PAS+ substance and is often covered by a thin sheet or hymen. Both integuments are cuticle-covered thin sheets, each 2-few cell layers in thickness. In the outer integument an increase in starch accumulation occurs during ovule maturation and probably functions as nutrient storage for embryo development. The inner epidermis of the inner integument differentiates as the most conspicuous cell layer of the beet ovule. During growth and maturation of the ovule a system of small perinuclear vacuoles containing dense material increases steadily in these cells. At maturity this system fills up more than half of each cell and very dense material has accumulated in each vacuole. This vacuole content is highly refractive and contains tannins and/or polyphenols.  相似文献

Illus. 1960.–The integuments of Abutilon theophrasti Medic. undergo a rapid increase in size, predominantly by anticlinal cell divisions during the first 3 days after fertilization. Within 7 days, the outer epidermis of the inner integument becomes thick walled. At maturity this compact, lignified, and cutinized palisade layer accounts for more than half the thickness of the seed coat. During early growth, the palisade cells form a continuous layer in the micropylar region. In the chalazal region the palisade layer is discontinuous in a slit-shaped region, 60 × 740 microns. The shape of this discontinuity constitutes a major difference between dormant-seeded Abutilon and non-dormant Gossypium seeds. Exterior to the palisade layer is the outer integument which consists of a small-celled layer and a large-celled layer sparsely covered with unicellular, lignified hairs. Interior to the palisade is the thick mesophyll of the inner integument which is largely digested during seed growth and leaves only 2 pigmented cell layers in most regions at maturity. The inner epidermis is small-celled, pigmented and cutinized and adheres tightly to the endosperm. Seed coat impermeability increases with seed maturity. Even immature seeds will germinate, if scarified, indicating a lack of embryo dormancy.  相似文献

This study has shown for the first time that the middle layer (or layers) of the outer integument is (are) of subdermal derivation in at least some taxa of the Cruciferae. The outer integument is initiated in the Cruciferae in three different ways, viz. subdermally (Brassica, Sinapis) , partly subdermally and partly dermally (Lunaria) , or completely dermally (Capsella). These differences in initiation are reflected in the structure of the mature testa. The inner integument is completely of dermal derivation and originally two cell-layers thick, but may become more than two-layered during the ovule and seed maturation by periclinal divisions of the inner cell layer. The consequences of the ontogeny of the integuments for the terminology and interpretation of the mature testa is discussed.  相似文献

Summary The ultrastructure and development of the amphiesma of the dinoflagellateGlenodinium foliaceum was studied using conventional electron microscopy and immunocytochemistry. Ecdysis (shedding of the flagella, the outer two membranes of the cell, and the thecal plates) was induced by centrifugation. The cells were resuspended and the thickening of the pellicle and the development of the new thecal vesicles and plates was studied over a 9 h period. After ecdysis, the thin pellicle which underlay the thecal plates in the motile cells thickens to form a complex structure of four distinct layers: an outer layer of randomly oriented fibrils, a 50 nm layer of fibrils oriented perpendicular to the dense layer, the dense layer which has a trilaminate structure, and a wide inner homogeneous layer. The new thecal vesicles form in these pelliculate cells by the migration of electron translucent amphisomal vesicles over the layer of peripheral microtubules to a position directly under the plasmalemma. The thecal vesicles then flatten and elongate. A discontinuous pellicular layer appears within them. Subsequently, the thecal vesicles widen and are filled with a fibrillogranular substance overlying the pelliculate layer. The thecal plates form on top of this fibrillogranular material. By this time, most cells have escaped from the pellicle and are motile. At first, the outer thecal vesicle membrane is continuous with the inner thecal vesicle membrane at the sutures, but when this connection is broken, the dense pelliculate layers become continuous across the suture as does the inner thecal vesicle membrane. At ecdysis, this membrane becomes the new plasmalemma of the cell. Cells at each stage of pellicle thickening and thecal development were labelled with a polydonal antiserum raised against the 70 kDa epiplasmic protein ofEuglena acus. This antiserum labelled both the thecal plates of the motile cells and the inner homogeneous layer of the pellicle of ecdysed non-motile cells. The characteristic Mesozoic pollen genus Eucommiidites is described from pollen organs and seeds recovered in Cretaceous strata of North America and Europe. The pollen organs are from the lowermost Upper Cretaceous (Cenomanian) of Texas and are referred to Erdtmanitheca texensis gen. et sp. nov. They are spherical heads, composed of numerous, densely crowded, radiating pollen sacs that contain abundant well-preserved pollen. Combined LM, SEM and TEM investigations show that the pollen grains each have a distinct distal colpus flanked by two lateral colpi in an equatorial position. Pollen wall ultrastructure is gymnospermous with a thick lamellate inner layer (endexine) and an outer layer (ektexine) composed of a granular inner part and a homogeneous outer part. The endexine is thickened in the region of the colpi. Small seeds from the Lower Cretaceous (upper Berriasian to Valanginian) of Bornholm, Denmark contain abundant Eucommiidites pollen in their micropyles. The seeds are referred to Erdtmanispermum balticum gen. et sp. nov. They are ovoid, and weakly triangular in transverse section and gradually taper at the apex into an elongated tube. The megaspore membrane is granular and well developed, and apparently surrounded by three separate tissues interpreted as nucellus, a thin inner integument and a sclerified outer envelope. Eucommiidites pollen in the micropyles of the seeds has a laminated endexine and an ektexine comprising two homogeneous parts separated by a granular layer. Réévaluation of other seeds known to contain Eucommiidites pollen indicates that they share significant similarities with Erdlmanispermum and that they may have been produced by closely related plants. Comparison of “Eucommiidites plants” with other seed plants suggests that they are probably most closely related to the anthophytes comprising Bennettitales, Pentoxylales, Gnetales and angiosperms.  相似文献

Two homologues of INNER NO OUTER (INO) in Nymphaea alba and N. colorata (Nymphaeaceae) were isolated and the expression pattern of the N. alba INO homologue NaINO was examined by in situ hybridization. The INO homologues obtained have a portion similar to INO in the predicted amino acid sequences between the conserved zinc finger-like and YABBY domains. In an in situ hybridization analysis, NaINO is expressed in the outer epidermis of the outer integument, inner integument, and the tip of the nucellus. The pattern observed in the outer integument is very similar to that of Arabidopsis thaliana, while the expression in the inner integument and nucellus is not observed in A. thaliana.Edited by G. The tribeDrypeteae, whose traditional assignment inPhyllanthoideae ofEuphorbiaceae is now doubtful, is studied embryologically on the basis of a literature survey and examination of six additional species in two of the four constituent genera.Drypeteae are characterized by having several embryological features that are unknown in otherPhyllanthoideae, such as a two- or three-celled ovule archesporium; a thin, two cell-layered parietal layer in the nucellus; no nucellar beak or cap; an early disintegrating nucellar tissue; thick, multiplicative, inner and outer integuments; an endothelium; a few discrete vascular bundles in the outer integument; and a fibrous exotegmen (or its derived state). EmbryologicallyDrypeteae do not fit within thePhyllanthoideae and, as available nucleotide sequence data from therbcL gene suggest, are rather placed nearErythroxylaceae, Rhizophoraceae, Chrysobalanaceae, andLinaceae. Drypeteae share with those families a combination of the fibrous exotegmen, the endothelium, and the thick, multiplicative inner integument.  相似文献
