Summary A theoretical model describing the genetic aspect of the transition from traditional to modern animal husbandry is presented. Traditional races are characterized by high tolerance to harsh environments but a low rate of response to increased management inputs. Modern, artificially-selected breeds are efficient convertors of management inputs to higher production but have a low resistance to harsh environments. Thus, under lowinput traditional husbandry, the traditional races are best adapted, while under modern, high-input husbandry, modern breeds are most productive, and in the intermediate zone, hybrids between the two races are capable of closing the profit gap in the shift from traditional to modern husbandry. The domesticated European, and the Chinese Big-belly races of the common carp were tested under many environmental treatments involving variation in density, polyculture, aeration, feeding and fertilization. The Big-belly showed, as expected, high resistance to the poor treatments but low response to environmental improvement. The European breeds performed best in the higher half of the environmental range and their response rates were highest. The F1 hybrids between the two races excelled in the lower third of the range, exhibiting, there, a high heterosis but only an intermediate rate of response. It was concluded that successful changes from one aquaculture system to another, and particularly the change from traditional to modern husbandry, require a simultaneous search for the most efficient genotype × environment combination and, for each level of modernization of traditional fish farming, the most effective genotype must be identified and utilized. The transition from traditional to modern animal husbandry, including fish farming, is best quantified by the levels of invested inputs, other than labour, that induce higher production of the individual animals. The major management inputs of modern fresh water fish farming are expensive feeding, veterinary care, control of predators, organic and chemical fertilizers that enrich the production of natural fish food, water circulation and aeration. Since all these inputs are rather expensive, the fish have to pay for them by increased production, i.e., faster growth rate. Thus, the sina qua non of such a transition is the availability of animal stocks capable of converting increased inputs into economically attractive increased yields. We are all aware of the very great physiological plasticity of farm animals. In the case of the European carp, for example, the same genetic stocks, raised under high stocking density and low feeding level may gain an average weight of 10 to 20 g per fish in a whole year, while under low density and abundant feeding, they may gain over 2 kg in the same period. Such physiological responsiveness may give the wrong impression that all that is needed for the transition to more modern husbandry are improved environmental circumstances. The object of this paper is to point out that the proper choice and changeover of genotypes is equally important for the succesfull implementation of the usually gradual process of fish farming modernization. This demonstration will be based on results of experiments with the European and Chinese races of the common carp, and their F1 hybrids.  相似文献

Summary During the disintegration of isolated chloroplasts, the declining capacity for evolving oxygen in the light becomes temporarily color sensitive. The decline of Hill reaction rates follows a different time course if measured in either blue (380<<500 nm) or red light (>600 nm). In this study, we investigate the effects of endothelin-1 (ET-1) and insulin on the cellular activity of protein kinase FA/glycogen synthase kinase-3 (kinase FA/GSK-3) in rat adipocytes. The cellular activity of kinase FA/GSK-3 is inhibited to 50% of control within 30 min when cells are treated with 1 nM ET-1 at 37°C; in addition, significant inhibition to 60% of control is observed at as low as 1 pM ET-1. Conversely, ET-1 at concentrations up to 1 nM has no direct effect on purified kinase FA/GSK-3 in vitro. Immunoblotting analysis further reveals that the protein level of this kinase is not significantly changed when treated with 1 nM ET-1 for 30 min. Similar to ET-1, insulin as low as 10 nM can also induce inactivation of kinase FA/GSK-3 to 50% of control in adipocytes when processed under identical conditions. Most importantly, when treated with both insulin and ET-1, the activity of kinase FA/GSK-3 can be decreased only to 50% of control. Taken together, the results provide initial evidence that ET-1 and insulin may regulate this important multisubstrate/multifunctional protein kinase in a common signaling pathway in cells.  相似文献

Summary We have synthesized 2(3)-O-(glycyl)-adenosine-5-(O-methylphos-phate), an analogue of the 3-terminus of aminoacylated tRNA. A 0.4M solution of this compound maintained at pH 8.2, yields 5.5% of diglycine and 11.5% of diketopiperazine, in addition to the hydrolysis products glycine and adenosine-5-(O-methylphosphate). Under the same conditions, glycine ethyl ester reacts much more slowly, but ultimately gives similar yields of diglycine and diketopiperazine.The aminolysis of 2(3)-O-(glycyl)-adenosine-5-(O-methylphosphate) by free glycine is relatively inefficient, but serine reacts 20 times more rapidly and yields up to 50% of N-glycylserine. Summary The relationship between the arrangement of cell events occurring in cambium in a definite configuration and the grain pattern of wood was investigated. Taking into consideration the growth activity of fusiform cell ends, a model of a migrating morphogenetic wave determining an event configuration was made. Waves of length =1 m for the periods T=2 years and T=3 years and waves of lengths =l m and =0.04 m for the period T=10 years were considered. On the model, events from successive annual rings, conventionally comprising 10 cell layers each, were summed. In this way, event maps were obtained. For wave =4 mm, the domain pattern on the modelled map was compatible with the grain pattern. The domain pattern for the wave =1 m was impossible to recreate because the wave migrated too fast. In this case, the pattern of event configuration, incompatible with the grain pattern, formed microareas, which were not domains.  相似文献

The conserved residue Thr42 of -subunit of the chloroplast ATP synthase of maize (Zea mays L.) was substituted with Cys, Arg, and Ile, respectively, through site-directed mutagenesis. The over-expressed and refolded -proteins were purified by chromatography on DEAE-cellulose and FPLC on mono-Q column, which were as biologically active (inhibiting Ca2+-ATPase activity and blocking proton gate) as the native subunit isolated from chloroplasts. The T42C and T42R showed higher inhibitory activities on the soluble CF1(–) Ca2+-ATPase than the WT. The T42I inhibited the Ca2+-ATPase activity of soluble CF1 and restored photophosphorylation activity of membrane-bound CF1 deficient in the most efficiently. Far-ultraviolet CD spectra showed that the portions of -helix and -sheet structures of the three mutants were somewhat different from WT. Thus the conserved residue Thr42 may be important for maintaining the structure and function of the -subunit and the basic functions of the -subunit as far as an inhibitor of Ca2+-ATPase and the proton gate are related.  相似文献

Summary In the progeny of a hybrid between monotelosomic line 3B of Chinese Spring wheat and Chinese Spring — Aegilops longissima ditelosomic addition line G a cytologically stable strain was selected consisting of 20 wheat chromosome pairs, one pair of telosomic chromosome 3BL and one pair of telosomic longissima chromosome G. Inoculating Chinese Spring — Aegilops longissima addition and substitution lines with ten different powdery mildew isolates, partial resistance was observed. The infection grade as well as the number of spores/cm2 leaf area were significantly reduced.  相似文献

Total cellular creatine content is an important bioenergetic parameter in skeletal muscle. To understand its regulation we investigated creatine transport and accumulation in the G8 cultured skeletal myoblast line. Like other cell types, these contain a creatine transporter, whose activity, measured using a radiolabelling technique, was saturable (Km = 110 ± 25 M) and largely dependent on extracellular [Na+]. To study sustained influences on steady state creatine concentration we measured total cellular creatine content using a fluorimetric method in 48 h incubations. We found that the total cellular creatine content was relatively independent of extracellular creatine concentration, consistent with high affinity sodium-dependent uptake balanced by slow passive efflux. Accordingly, in creatine-free incubations net creatine efflux was slow ( 5 ± 1 % of basal creatine content per day over 6 days), while creatine content in 48 h incubations was reduced by 28 ± 13% of control by the Na+,K+-ATPase inhibitor ouabain. Creatine accumulation after 48 h was stimulated by treatment with the mixed - and -adrenergic agonist noradrenaline, the -adrenergic agonist isoproterenol, the 2-agonist clenbuterol and the cAMP analogue N6,2-O-dibutyryladenosine 3,5-cyclic monophosphate, but was unaffected by the 1 adrenergic agonist methoxamine. The noradrenaline enhancement of creatine accumulation at 48 h was inhibited by the mixed - and -antagonist labetalol and by the -antagonist propranolol, but was unaffected by the 2 antagonist phentolamine; greater inhibition was caused by the 2 antagonist butoxamine than the 1 antagonist atenolol. Creatine accumulation at 48 h was increased to 230 ± 6% of control by insulin and by 140 ± 13% by IGF-I (both at 3 nM). Creatine accumulation at 48 h was also increased to 280 ± 40% of control by 3,3,5-triiodothyronine (at 70 M) and to 220 ± 35% of control by amylin (60 nM). As 3,3,5-triiodothyronine, amylin and isoproterenol all stimulate the Na+,K+-ATPase, we suggest that they stimulate Na+-creatine cotransport indirectly by increasing the transmembrane [Na+] concentration gradient and membrane potential.Abbreviations IGF-I insulin-like growth factor I - IGF-II insulin-like growth factor II - T3 3,3,5-triiodothyronine - CGRP calcitonin gene-related peptide  相似文献

Summary The construction and molecular analysis of four recombinant clones — pTa1, pTa2, pTa7, and pTa8 — is described. The four clones contain different highly repeated sequences of genomic DNA from Triticum aestivum variety Chinese Spring. The wheat specificity has been determined by colony and dot blot hybridization in comparison with total rye DNA (Secale cereale variety Petka). The four clones with a variable degree of specificity were compared by sequence analysis after the recloning of wheat DNA inserts into M13 mp8. Within the sequencing data a tendency can be observed that those repeated sequences which show the highest degree of species specificity contain a significantly increased amount of GC residues.  相似文献

Fry of the European and Chinese races of the common carp and interracial crossbred fry were grown together in a series of ponds for 28 days after hatching. The degree of crowding and the amount of available natural food were intentionally varied in these experimental ponds. This resulted in a large variation in mean growth between ponds, ranging from 0·1 to 5·2 g per fish during the 28 days growth period. Under conditions of very slow growth, resulting from severe crowding and lack of natural food, fry of the Chinese carp grew several times faster than European carp. At lower densities and when natural food was more abundant, resulting in a faster rate of growth, this difference between the two races disappeared. Crossbred fry between the Chinese and European carp grew faster than either of their parents under all conditions tested.  相似文献

Summary The fluorescent intercalation complex of ethidium bromide (ETB) with DNA was used as a probe to compare the effects of various radicals with respect to impairment of the DNA base-pair region.OH radicals inhibit up to 0.7 dye intercalations perOH at low salt concentration, and for various oxidizing species the effect decreases in the orderOH > Br 2 > N 3 > $$ " align="middle" border="0"> (SCN) 2 . DNA impairment by theOH product of Met-Gly is comparable to that of N 3 , but no effect was found due to the interaction between DNA and Lys-Tyr-Lys phenoxyl radicals. Pond culture of brackishwater prawns and shrimps has recently generated much interest in Kenya. The mangrove areas are the target zones for the construction of these ponds. With the increasing awareness of the unique ecological role played by the mangroves, there is an urgent need to stop the conversion of mangrove swamps into aquaculture ponds. To develop pond aquaculture without destroying the mangroves, a shift from tide-fed to pump-fed pond systems is recommended in order to divert the farming from the mangroves to higher grounds. Mangrove-friendly mariculture practices like pen, cage and raft culture are discussed. Methods of efficiently utilising the already destroyed mangrove areas are considered.  相似文献

Summary We describe here the screening of a small group of apparently healthy individuals belonging to the tribal communities of Koya Dora and Konda Reddi. A remarkably high incidence of deletion and nondeletion + thalassemia mutants has been found with allele frequencies and distributions characteristic to each tribe. We have confirmed the strict relationship between Hb S levels and the number of globin genes in double heterozygotes for the S gene and thalassemia. In this population sample we did not find either heterozygous carriers of 0 thalassemia (deletion of both alpha genes in cis) or individuals showing hemolytic anemia due to inactivation of three -globin genes (Hb H disease). Selection by malaria is most probably responsible for the prevalence of the various + thalassemia haplotypes among the two tribal populations of Andhra Pradesh.  相似文献

The effect of different carbon and nitrogen compounds on growth and sporulation ofC. microcephala (Wallr.)Tul. causing ergot disease of Bajra has been studied. Nine different sources of carbon were used but cane sugar was found to be the best source for both, growth and sportulation of the fungus. Glucose, sucrose and maltose gave good growth but fair sporulation. Lactose and sorbitaol proved to be the poor sources. However, fungus failed to utilize starch, dextrin and mannitol.Nineteen nitrogen compounds were tried for the growth and sporulation of the fungus. Best growth and sporulation were supported by peptone and glycine. L-asparagine, DL-valine, Urea, magnesium nitrate and L-proline supported good growth and fair sporulation except DL-valine where it was excellent. Poor growth was obtained on L-isoleucin, ammonium sulphate, potassium nitrate,-alanine, ammonium chloride, DL-aspartic acid and DL-methionine. Fungus failed to utilize thio-urea.  相似文献

The therapeutic effect of granulocyte colony-stimulating factor (G-CSF) against intramuscular infection withPseudomonas aeruginosa in cyclophosphamide (CY)-treated mice was analyzed by measuring plasma levels of amyloid P-component (APC) and proinflammatory cytokine levels. CY (100mg/kg) treatment of mice significantly suppressed plasma concentrations of APC and tumor-necrosis factor- (TNF-) following infection withP. aeruginosa, in associated with enhanced susceptibility of the treated mice to this bacterium. A 4-day treatment of CY-treated mice with recombinant human G-CSF (rhG-CSF) increased resistance of CY-treated mice, together with the marked restoration of APC and TNF- productions. The capacity to produce interleukin 1- and TNF- of peritoneal macrophages and also that to produce IL-6 of spleen cells were significantly enhanced by thein vivo administration of rhG-CSF in CY-treated mice. These results indicate that G-CSF may increase the functions of monocytes/macrophages directly or indirectlyin vivo. Summary The visual pigment of the American lobster,Homarus americanus, has been studied in individual isolated rhabdoms by microspectrophotometry. Lobster rhodopsin has max at 515 nm and is converted by light to a stable metarhodopsin with max at 490 nm. These figures are in good agreement with corresponding values obtained by Wald and Hubbard (1957) in digitonin extracts. Photoregeneration of rhodopsin to metarhodopsin is also observed. The absorbance spectrum of lobster metarhodopsin is invariant with pH in the range 5.4–9, indicating that even after isomerization of the chromophore fromcis totrans, the binding site of the chromophore remains sequestered from the solvent environment. Total axial density of the lobster rhabdom to unpolarized light is about 0.7.As described for several other Crustacea, aldehyde fixation renders the metarhodopsin susceptible to photobleaching, a process that is faster at alkaline than at neutral or acid pH. Small amounts of a photoproduct with max at 370 nm are occasionally seen. A slower dark bleaching of lobster rhabdoms (1/2–2 h) also occurs, frequently through intermediates with absorption similar to metarhodopsin.The molar extinction coefficient of metarhodopsin is about 1.2 times greater than that of rhodopsin, each measured at their respective max. Isomerization of the chromophore fromcis totrans is accompanied by a change in the orientation of the absorption vector of about 3°. The absorption vector of metarhodopsin is either tilted more steeply into the membrane or is less tightly oriented with respect to the microvillar axes.When living lobsters are kept at room temperature, light adaptation does not result in an accumulation of metarhodopsin. At 4 °C, however, the same adapting lights cause a reduction of rhodopsin and an increase in metarhodopsin. There is thus a temperature-sensitive regeneration mechanism that supplements photoregeneration. Following 1 ms, 0.1 joule xenon flashes that convert about 70% of the rhodopsin to metarhodopsin in vivo, dark regeneration occurs in the living eye with half-times of about 25 and 55 min at 22 °C and 15 °C respectively.This work was supported by USPHS research grant EY 00222 to Yale University. Summary Thirteen spontaneously occurring chlorophyll deficient phenotypes have been described and their genetic basis was established. Ten of these — white, white tipped green, patchy white, white virescent, white striping 1, white striping 2, white striping 4, fine striping, chlorina and yellow virescent showed monogenic recessive inheritance and the remaining three — yellow striping, yellow green and light green seedling phenotypes showed digenic recessive inheritance. The genes for (i) white tipped green (wr) and yellow virescent (yv) and (ii) patchy white (pw) and white striping 1 (wst 1) showed independent assortment. Further, the genes for white (w), white tipped green (wr) and yellow virescent (yv) were inherited independently of the gene for hairy leaf margin (Hm).In the mutants — white tipped green, patchy white, white striping 1, white striping 2, fine striping, chlorina, yellow virescent, yellow striping, yellow green and light green phenotypes total quantity of chlorophyll was significantly less than that in the corresponding controls, while in white virescent there was no reduction in the mature stage. For nine of the mutants the quantity of chlorophyll was also estimated in F1's (mutant x control green). In F1's of six of the mutants — white tip, patchy white, chlorina, yellow virescent, fine striping and yellow striping the quantity of chlorophyll was almost equal to the wild type. In the F1's of three of the mutants — white striping 1, white striping 2 and light green an intermediate value between the mutant and wild types was observed. In yellow virescent retarded synthesis of chlorophyll, particularly chlorophyll a was observed in the juvenile stage. Reduced quantity of chlorophyll was associated with defective chloroplasts. In the mutants — white tipped green, white virescent, fine striping, chlorina, yellow striping, yellow green and light green defective plastids were also observed. In patchy white secondary destruction of chlorophylls and the presence of defective plastids were found to be associated with reduced chlorophyll quantity at maturity.Paper chromatographic studies of leaf flavonoids revealed some variation between the inbreds, but there were three common spots, 7, 8 and 9, except for PDP in which the spot 8 was absent. Chlorophyll deficient mutants differed from their respective controls in the absence of one or more of the spots present in the controls and in the presence of new spots in some of the mutants.Most of the chlorophyll mutants showed higher survival rate in the Kharif season than in Rabi season which was attributed to the higher mean day temperature and longer day light period in the Kharif season than in Rabi season.  相似文献

The diatom Cymbellonitzschia diluviana Hustedt is an important indicator species in interglacial deposits but is rarely reported from contemporary sites. The presence of large populations in Lough Neagh, N. Ireland, made possible a study of its ecology and auxosporulation. The apical length of cells in the lake was usually between 7 and 25 µm, but in culture there was a broader range between 5 and 42 µm. Amongst smaller cells (< 11 µm), some (3 %) were found with raphes on the dorsal (i.e. convex side) rather than usual ventral side. At cell division, both daughter cells had hantzschioid symmetry. C. diluviana is found in alkaline waters (pH 7.6–9.0) but not usually where there is significant calcite deposition. Its present rarity is probably because its preferred habitat is now less common. In L. Neagh, C. diluviana was found on exposed rocky and sandy shores, but was particularly abundant in shifting sand bars. This highly abrasive and erosive environment requires cells to have good adhesion and the ability to withstand extended periods of burial. C. diluviana is essentially a ruderal species, adapted to habitats with high disturbance.  相似文献
