金黄色葡萄球菌是医院和社区获得性感染最常见的病原菌之一,而且可形成生物膜,从而导致生物膜相关疾病的产生。患者一旦发生金黄色葡萄球菌生物膜感染,难以彻底治愈。深入研究金黄色葡萄球菌生物膜形成的分子机制和调控网络,对寻找有效的防治、治疗药物和手段具有重要意义。我们就金黄色葡萄球菌生物膜形成过程和调控机制的研究近况做一综述。  相似文献

目的 构建一种简便易行的新生隐球菌生物膜感染动物模型.方法 采用大鼠皮下置管法构建新生隐球菌生物膜感染动物模型,并使用电子扫描显微镜观察新生隐球菌体内形成生物膜的结构;采用MTT法对获得的体内生物膜结构进行活性的检测.结果 成功构建了新生隐球菌生物膜大鼠感染模型;MTT法检测体内生物膜活性表明,随着体内培养时间的延长,生物膜活性增强,与直接镜检和电镜观察结果一致.结论 新生隐球菌生物膜大鼠模型简单易行,便于操作,对于研究新生隐球菌生物膜体内活性的研究具有一定的实际应用价值.  相似文献

目的构建新生隐球菌的PMT4基因缺陷株及新生隐球菌生物膜的体内、外模型;研究PMT4基因对生物膜形成的影响。方法采用PCR介导的长侧翼同源重组的方法敲除新生隐球菌H99的PMT4基因;采用基础培养基96孔板培养的方法建立生物膜体外模型;兔中心静脉插管、管内放置聚苯乙烯薄膜的方法建立生物膜动物模型;用倒置显微镜、共聚集激光扫描显微镜、MTT、CFU计数等方法研究PMT4缺陷株与野生株生物膜的异同。结果新生隐球菌在体内、外模型中均能形成生物膜;PMT4缺陷株与野生株生物膜在生物量和结构方面存在明显差异。结论本实验的生物膜动物模型可行;PMT4基因缺陷可造成隐球菌生物膜代谢活性下降,并形成假菌丝样结构。  相似文献

ica位点编码的胞外多糖(PIA/PNAG)对理解葡萄球菌生物膜相关感染病理学方面具有重要的意义.关于ica位点与PIA/PNAG之间如何调节的研究还不全面,另外一种独立于ica的生物膜形成机制存在于表皮葡萄球菌和金黄色葡萄球菌中;细胞表面相关蛋白也能调节生物膜的形成,这些发现为探究它们在生物膜形成机制的潜在作用提供了重要基础.  相似文献

目的:探讨DTPA对人工关节假体(prosthetic joints infection,PJI)感染后表皮葡萄球菌生物膜形成及所需基因聚集相关蛋白(AAP)基因表达的影响。方法:构建新西兰大白兔假体感染表皮葡萄球菌生物膜模型,随机分为实验组(n=25)和对照组(n=25),对照组只注射细菌,实验组在注入细菌的同时,注射DTPA,分别于注射后第2、4、6、8、16天灌洗、收集膝关节液,采用RT-PCR方法检测不同时间点目的基因AAP基因表达的变化。对不同时间点的假体感染动物行膝关节假体扫描电镜和假体周围组织学观察。结果:DTPA可以显著抑制AAP基因的表达,扫描电镜观察第2、4、6、8、16天实验组较对照组假体表面粘附表皮葡萄球菌明显减少(P<0.05),生物膜明显受抑制。实验组假体周围组织炎性反应较对照组明显减轻。结论:DTPA可以抑制表皮葡萄球菌生物膜形成中相关蛋白AAP基因的表达,可能抑制生物膜的形成。  相似文献

生物膜是指由黏附于生命体或非生命体表面的微生物和其胞外聚合物所构成的复杂、多维的空间结构,在自然界中普遍存在。微生物在人类体内许多部位均可形成生物膜结构,如肺、心脏瓣膜、泌尿生殖道、肝胆系统、耳、鼻、皮肤等。越来越多的感染性疾病患者或动物模型体内均可检测出生物膜,如囊性纤维化肺炎、慢性中耳炎、细菌性心内膜炎、皮肤外伤后慢性感染、慢性鼻窦炎等。生物膜的形成与感染性疾病的关系及其在疾病的发生、发展、转归过程中的作用成为近年来研究的热点。目前体内生物膜检测技术主要有扫描电镜、激光共聚焦显微镜、荧光原位杂交技术等。就临床常见感染性疾病与生物膜的关系及体内生物膜检测方法展开综述。  相似文献

连翘苷和黄芩苷对表皮葡萄球菌生物膜抑制作用的研究   总被引:3,自引:0,他引:3  
目的通过中药有效成分连翘苷和黄芩苷分别对表皮葡萄球菌生物膜抑制作用的研究,为表皮葡萄球菌生物膜引起的相关感染提供新的治疗途径。方法体外构建表皮葡萄球菌生物膜,XTT减低法评价连翘苷、黄芩苷对表皮葡萄球菌初始黏附及生物膜内细菌代谢的影响,显微镜下观察用药后表皮葡萄球菌生物膜形态和结构改变。结果连翘苷和黄芩苷对表皮葡萄球菌生物膜的早期黏附均无抑制作用;连翘苷对表皮葡萄球菌生物膜菌的SMIC50为31.25μg/ml,而黄芩苷对表皮葡萄球菌生物膜菌的代谢无影响;在显微镜下观察,连翘苷使部分表皮葡萄球菌被膜的形态发生改变,而黄芩苷对其形态影响不显著。结论连翘苷对表皮葡萄球菌生物膜的初始黏附阶段无抑制作用,对生物膜菌的代谢和生物膜形态均有显著影响;黄芩苷对表皮葡萄球菌生物膜无显著作用。  相似文献

希瓦氏菌(Shewanella spp.)是海产品中常见的优势腐败菌,易在食品加工设备表面形成生物被膜而难以清除。生物被膜的存在不仅会造成食品的持续污染和交叉污染,也会影响加工设备的使用,从而对国民健康和经济发展造成威胁。目前,针对希瓦氏菌生物被膜的研究主要集中在表型研究上,对其生物被膜形成分子机制的研究尚处于起步阶段。总结希瓦氏菌生物被膜的形成过程,重点论述希瓦氏菌生物被膜的形成机制并对希瓦氏菌生物被膜控制方法进行简要概括,展望未来的研究方向,以期为希瓦氏菌生物被膜的深入研究提供参考。  相似文献

【目的】筛选能有效抑制单核细胞增生李斯特菌(Listeria monocytogenes,LM)形成生物被膜的乳酸菌,分析其活性成分并进行功能表征。【方法】采用结晶紫染色法筛选抑制LM形成生物被膜的不同乳酸菌提取物;通过酸中和、蛋白酶处理及热处理,推测抑制生物被膜活性物质以胞外多糖(extracellular crude polysaccharide,ECP)为主;乙醇沉淀法提取目标乳酸菌分离株胞外粗多糖,分析其抑制生物被膜形成活性和对LM生长的影响;运用激光共聚焦扫描显微镜(laser confocal scanning microscopy,LCSM)和扫描电子显微镜(scanning electron microscopy,SEM)观察胞外粗多糖对生物被膜细胞形态和结构的影响。【结果】发酵乳杆菌CSC-19发酵上清液对1516-2LM生物被膜的抑制率为81.7%;经热和蛋白酶处理后,发酵上清抑制生物被膜形成的活性未发生显著变化(P>0.05),表明发酵上清液中抑制生物被膜形成的物质可能为胞外多糖;在不抑制LM生长的条件下所提取的胞外粗多糖抑制生物被膜形成能力具有浓度依赖性。激光共聚焦扫描显微镜和扫描电子显微镜结果显示,胞外粗多糖显著抑制了生物被膜的形成能力,生物被膜三维、有组织的蜂窝状结构被破坏,仅有少量的粘附细胞分散于细胞爬片表面。【结论】发酵乳杆菌CSC-19胞外粗多糖能有效抑制LM生物被膜的形成,有望应用于高效防控该菌污染食品。  相似文献

Conventional antibiotics might fail in the treatment of biofilm-associated infections causing infection recurrence and chronicity. The search for antimicrobial peptides has been performed with the aim to discover novel anti-infective agents active on pathogens in both planktonic and biofilm associated forms. The fragment 9–19 of human thymosin β4 was studied through 1 μs MD simulation. Two main conformations of the peptide were detected, both constituted by a central hydrophobic core and by the presence of peripheral charged residues suggesting a possible mechanism of interaction with two models of biological membranes, related to eukaryotic or bacterial membrane respectively. In addition, the peptide was chemically synthesized and its antimicrobial activity was tested in vitro against planktonic and biofilm form of a group of reference strains of Staphylococcus spp. and one P. aeruginosa strain. The human thymosin β4 fragment EIEKFDKSKLK showed antibacterial activity against staphylococcal strains and Pseudomonas aeruginosa ATCC 15442 at concentrations from 12.5 to 6.2 mg/ml and inhibited biofilm formation at sub-inhibitory concentrations (3.1–0.75 mg/ml). The activity of the fragment in inhibiting biofilm formation, could be due to the conformations highlighted by the MD simulations, suggesting its interaction with the bacterial membrane. Human thymosin β4 fragment can be considered a promising lead compound to develop novel synthetic or recombinant derivatives with improved pharmaceutical potential.  相似文献

葡萄球菌是临床常见致病菌及食源性致病菌,可在食品原料加工、包装及运输过程中污染食品,引起人体多种严重感染,其耐药性的不断增强对公共卫生安全产生了重大的威胁。葡萄球菌中cfr (chloramphenicol-florfenicol resistance)基因编码的甲基转移酶,可引起细菌核糖体RNA的甲基化,从而阻碍或减弱多种化学结构不同的抗生素与肽基转移酶活性中心(peptidyl transferase center,PTC)的结合,导致葡萄球菌多重耐药表型的出现。噁唑烷酮类药物−利奈唑胺是继万古霉素后治疗耐药革兰氏阳性菌所致感染的最后一道防线,cfr基因的出现大大加速了利奈唑胺耐药性的传播。cfr基因广泛分布于多种致病性葡萄球菌中,cfr基因与各类型可转移元件(质粒、转座子和整合相关元件等)密切关联的遗传环境是其广泛传播的结构基础。在cfr基因水平传播的过程中,食源性致病葡萄球菌作为中间者扮演着重要的角色。本文就近年来国内外对致病性葡萄球菌中cfr基因的分布状况、耐药机制、遗传环境、传播机制等进行综述,以期为防控致病性葡萄球菌的传播提供参考,以遏制多重耐药菌的进一步传播。  相似文献

【背景】小菌落变异株(smallcolonyvariant,SCV)是一种具有独特的表型及致病特征且生长缓慢的细菌亚群,而国内鲜有关于食源性沙门菌SCV的研究报道。【目的】为食源性沙门菌的防治及动物性食品安全提供实验数据。【方法】使用氨基糖苷类抗生素对羊源胆汁中分离的沙门菌进行实验室诱导得到SCV,然后分别对野生株和诱导株的菌落形态、生长、生化特性、营养缺陷型检测、运动性、耐药性检测及耐药基因、毒力基因、生物被膜形成能力进行比较和分析。【结果】经卡那霉素诱导获得一株血红素依赖型沙门菌SCV,与野生株相比,诱导株生长缓慢,低于野生株84%,不利用柠檬酸盐,溶血能力增强40%,对磺胺类和氨基糖苷类药物的耐受性增强,生物被膜形成能力减弱45%,运动能力减弱78%。【结论】沙门菌SCV的生长和生理生化特性与野生株相比有显著差异,使得沙门菌SCV的分离鉴定尤为困难;并且SCV的致病性与耐药性等方面的变化可能给沙门菌病的防治带来更大挑战,其机制还有待深入研究。  相似文献

Quorum sensing (QS) regulates virulence and biofilm formation in Pseudomonas aeruginosa and other medically relevant bacteria. Human paraoxonases (hPONs) are a family of closely related enzymes with multiple functions, including inactivation of the QS signal molecule in P. aeruginosa. However, there is no direct evidence to show the functions of hPONs on biofilm formation and antibiotic resistance in P. aeruginosa. In the present study, hPONs (hPON1, hPON2, and hPON3) genes were respectively cloned into the pMEKm12 shuttle vector and transformed into P. aeruginosa strain PAO1. Expression of the three recombinant proteins was confirmed by Western blotting, and growth of the recombinant strains was not affected by the hPONs gene expression. Biofilm formation and antibiotics resistance of the hPONs recombinant strains were analyzed. Our results showed that biofilm formation was significantly inhibited in all of the three hPONs recombinant strains. Interestingly, this inhibition can be reverted by addition of the corresponding hPONs polyclonal antibodies in the culture media, further indicating that the inhibition of biofilm formation was due to hPONs protein expression. In addition, we also demonstrated that hPONs expression decreased resistance of P. aeruginosa to gentamicin and ceftazidima, two antibiotics clinically used for the treatment of P. aeruginosa infection.  相似文献

Aims: The aim of this study was to determine the prevalence and proportions of opportunistic pathogens harboured on orthodontic retainers. Methods and Results: First, Staphylococcus spp. and Candida spp. were isolated from the retainer’s inner surface and from other mucosal surfaces of the subject’s mouth by routine bacterial culture. The prevalence and proportions of these micro‐organisms on retainers was compared in different areas of the mouth within a group of retainer wearers, and mucosal carriage was compared to a group of nonretainer wearers. Staphylococcus spp. were isolated from 50% of the retainers and comprised on average 8·4% of the viable microbiota. Candida spp. comprised 0·13% of the viable microbiota and were recovered from 66·7% of the retainers. Neither genus was isolated from nonretainer wearers. Second, the two most commonly worn retainers manufactured from different materials were sampled; again Staphylococcus spp. and Candida spp. were recovered; however, no statistical differences were observed between the devices. Conclusions: Opportunistic, nonoral, pathogenic micro‐organisms were recovered from orthodontic retainers. Significance and Impact of the Study: It is possible that an orthodontic retainer could be a reservoir for opportunistic pathogens and act as a source of cross‐, self‐ and re‐infection.  相似文献

Rock-degrading endophytic bacteria in cacti   总被引:1,自引:1,他引:0  
A plant–bacterium association of the cardon cactus (Pachycereus pringlei) and endophytic bacteria promotes establishment of seedlings and growth on igneous rocks without soil. These bacteria weather several rock types and minerals, unbind significant amounts of useful minerals for plants from the rocks, fix in vitro N2, produce volatile and non-volatile organic acids, and reduce rock particle size to form mineral soil. This study revealed the presence of large populations of culturable endophytic bacteria inside the seeds extracted from wild plants, from seeds extracted from the guano of bats feeding on cactus fruit, in seedlings growing from these seeds, in the pulp of fruit, and in small, mature wild plants, and are comparable in size to populations of endophytic populations in some agricultural crops. The dominant culturable endophytes were isolates of the genera Bacillus spp., Klebsiella spp., Staphylococcus spp., and Pseudomonas spp. Based on partial sequencing of the 16s rRNA gene, the isolated strains had low similarity to known strains in these genera. However, these strains have higher molecular similarity among endophytes obtained from seeds, endophytes from roots, and some bacterial strains from the rhizoplane. Seedlings developed from seeds with endophytes contain the similar species of endophytes in their shoots, possibly derived from the seeds. This study shows the involvement of endophytic bacteria in rock weathering by cacti in a hot, subtropical desert and their possible contribution to primary colonization of barren rock. This study proposes that cacti capable of acquiring diverse populations of endophytes may give them an evolutionary advantage to gain a foothold on highly uncompromising terrain.  相似文献

【目的】副溶血性弧菌是水产品中常见的食源性致病菌,生物被膜的形成对副溶血性弧菌的环境生存和传播至关重要。这项工作的目的是评估临床和环境中分离出的44株副溶血性弧菌菌株形成的生物被膜的结构多样性。【方法】该研究基于共聚焦激光扫描显微镜的高通量方法,使用与高分辨率成像兼容的96孔微量滴定板,结合结构分析软件ISA-2来研究生物被膜形成和结构,分析22株食品与22株临床来源的副溶血性弧菌菌株形成的生物被膜结构参数(生物体积、平均厚度、粗糙系数)。【结果】CLSM图像显示,44株副溶血性弧菌菌株在培养48h后能够形成3D结构,进一步比较分析了临床来源菌株与环境来源菌株形成的生物被膜结构异同,发现临床菌株生物被膜的变异系数比环境菌株生物被膜的变异系数小,且同时携带tdh和trh两种毒力因子的菌株生物被膜变异性最小。凝聚层次聚类分析结果显示,副溶血性弧菌生物被膜可以分为致密且表面光滑(39%)、斑驳且表面粗糙(27%)、疏松且表面坑洼(34%),临床菌株易形成致密且表面光滑和斑驳且表面粗糙的生物被膜,而环境菌株易形成致密且表面光滑和疏松且表面坑洼的生物被膜。【结论】该研究深入了解了副溶血性弧菌生物...  相似文献
