In recent decades a significant raise in the incidence of myocardial infarction among young women has been recorded. It is presumed that, apart from the classical risk factors, other reasons exist for premature atherosclerosis in young women, related to the homeostasis of gonadal hormones. The aim of the study was to analyze the levels of gonadal hormones (estradiol, progesterone, follicle-stimulating hormone, luteinizing hormone, testosterone and dehydroepiandrosterone) measured in the luteal phase, in 65 normally menstruating women post myocardial infarction (MI) and to investigate a possible relationship between the hormone profile and selected coronary artery disease (CAD) risk factors. The levels of gonadal hormones: estradiol, progesterone, follicle-stimulating hormone, luteinizing hormone, testosterone and dehydroepiandrosterone were measured in the luteal phase. All examined women had normal mean levels of gonadal hormones. In the post MI patients leading a sedentary life style, a significantly lower mean progesterone concentration was observed (16.29 ± 9.11 versus 29.43 ± 21.14 nmol/l, p < 0.05) and significantly higher mean testosterone concentration (2.34 ± 0.98 versus 1.76 ± 1.09 nmol/l, p < 0.05) when compared to patients from the same group, but leading a more active life. In obese post MI women (BMI ≥ 30 kg/m(2)) a lower mean concentration of progesterone was detected (18.02 ± 8.12 versus 26.16 ± 14.72 nmol/l, p < 0.05), than in slimmer patients from the same group. In post MI women with a positive family history for CAD, a significantly higher mean concentration of testosterone was detected (2.31 ± 1.22 versus 1.67 ± 0.74 nmol/l, p < 0.05) than in patients with no family history. The results suggest a correlation between levels of gonadal hormones and classical CAD risk factors.  相似文献

Recently, it has been reported that mating can delay the age-associated decline in reproductive function of female rats. Since circulating progesterone (P) levels are elevated for a 2- to 3-wk interval during pregnancy, the following study was conducted to determine whether intermittent elevation in P levels can alter the rate of reproductive aging in female rats. Beginning at 2 mo of age, 4-day-cycling, virgin rats were divided into two groups. In one group, 3 Silastic capsules containing crystalline P were inserted s.c. into each rat while rats in another group each received 1 empty capsule. After 2 wk, the capsules were removed for 2 wk. Thereafter, implantation and removal of capsules was repeated 5 additional times. Rats receiving P capsules became acyclic 3-4 days after exposure to P and resumed cyclicity 4-7 days after removal of P-capsules. One month after the last series of capsules was removed (rats approximately 8-mo-old), rats exhibiting consecutive 4-day cycles were inserted with indwelling atrial cannulae and bled at 4-h intervals from 1400 h on proestrus (Pr) to 1000 h on estrus (E). At 1600 h E, rats were killed and trunk blood was collected. For comparison, a group of 3-mo-old (young) rats was bled on Pr and E. In 8-mo-old rats that received empty capsules, 27% exhibited 4-day cycles compared to 66% of the young rats. However, in contrast to rats that received empty capsules, 63.1% of P-treated rats exhibited 4-day cycles. This study aims to determine the effects of maternal protein and energy malnutrition during lactation on the linear growth, body weight and onset of puberty of the female offspring. At parturition, dams were randomly assigned to the following groups: (C) control group, with free access to a standard laboratory diet containing 23% protein; (PR) protein-restricted group, with free access to an isoenergy and protein-restricted diet containing 8% protein; and (ER) energy-restricted group, receiving standard laboratory diet in restricted quantities. After weaning, the female pups had free access to standard laboratory diet. From day 30 onwards, the pups were inspected daily for vaginal opening. Cyclic stages of the ovaries were studied by daily vaginal smears after vaginal opening until day 40 when all animals were sacrificed with pentobarbital. From day 4 after birth until day 40, body weight and linear growth in the PR and ER rats were significantly lower than in controls (p < 0.001). In spite of the significant (p<0.05) delayed in the vaginal opening in PR and ER rats, the first estrous cycle occurred at the same time of vaginal opening in all groups. The PR and ER rats exhibited a lower uterine (PR = 42%, ER = 40%, p < 0.001) and ovarian (PR = 26%, ER=19%, p < 0.05) absolute weight and uterus relative weight (PR = 27%, ER = 22%, p < 0.05). Our data showed that maternal protein and energy malnutrition during lactation leads to growth retardation and delayed on the onset of puberty in female pups, with vaginal opening and estrous cycle occurring at the same time.  相似文献

Intact female rats injected daily with 5 mg progesterone beginning at Day 27 of age weighed more than oil-treated controls on Day 39, i.e., prior to puberty; progesterone did not, however, increase food intake until well after puberty (Day 54). Progesterone may affect body weight independently of its interactions with estrogens and via mechanisms that do not involve the central substrate for food intake or energy balance. The ability of progesterone to influence food intake also depends in part on its modulation of estrogen-sensitive mechanisms that do not become functional until after puberty. Progesterone increased food intake of prepubertal overiectomized-adrenalectomized rats but not that of ovariectomized rats of any age. It was concluded that progesterone mimics the actions of adrenal hormones and that it permissively increases food intake of adrenalectomized rats by improving the health of these animals. Progesterone only partially substitutes for adrenal secretions in ovariectomized-adrenalectomized rats and supplementary sodium ingestion may be necessary for progesterone to produce positive energy balance.  相似文献

Proestrous hormonal profiles were characterized in lightly androgenized female rats prior to the onset of the delayed anovulatory syndrome (DAS). In these females, ovulatory failure and persistent vaginal estrus (PVE) occur at a very early age. Female Sprague-Dawley rats were injected with 10 micrograms testosterone propionate (TP) on postnatal Day 5. Control rats were untreated. All animals were weaned at 21 days of age, and following the onset of puberty, estrous cyclicity was monitored by vaginal smear. Rats showing regular 4-day cycles were used. Between 50-70 days of age, intra-atrial cannulae were implanted on a morning of proestrus (0700-0900 h) and blood was sampled at 2-h intervals from 1000 to 2000 h. Additional samples were taken at 0.5-h intervals from 1600 to 1800 h. Plasma was assayed for luteinizing hormone (LH), follicle-stimulating hormone (FSH) and progesterone (P) by radioimmunoassay (RIA). All animals were monitored for the onset of PVE or other alterations in estrous cyclicity. Females treated neonatally with TP that subsequently showed PVE by 150 days of age (PRE DAS) displayed a reduced peak amplitude (P less than 0.01) and delay in onset (1600 vs. 1400 h) of LH but not FSH secretion, when compared to controls. Females treated neonatally with TP that did not enter PVE by 150 days of age (No DAS) also showed a delayed rise in LH when compared to controls. The body growth and the onset of puberty in spontaneously hypertensive rats (SHR) and in normotensive controls (WKY) have been studied. In female rats the onset of puberty was determined by both the age and the body weight at which the vaginal opening and first estrus appeared, as well as the ability of estradiol and progesterone to induce pituitary LH release. For this purpose females were injected with estradiol benzoate (0.1 mg/kg) and progesterone (1 mg/rat). Control animals received only oil vehicle. In male rats, puberty was assessed by studying the age and body weight at the time of balano-preputial separation. In another experiment, SH and WKY rats were decapitated on day 30 to determine FSH, LH, PRL, GH and testosterone plasma levels in males and FSH and LH in females. The results obtained show: a) A greater body weight, at all the ages studied (every 4 days between days 28 and 92) in SHR animals. b) A delay in vaginal opening and first estrus presentation in SHR females. c) Absence of spontaneous LH peaks in WKY females. d) Advancement in balano-preputial separation in SHR males and e) Higher plasma FSH levels in SHR males than in WKY males, without differences in other hormones.  相似文献

Effects of embelin on oestrous cycle, plasma levels of progesterone and oestradiol, and in vitro production of oestradiol and progesterone by mixed ovarian cells was studied. Forty adult (4 months old) regularly cycling female Sprague-Dawley rats were divided into four groups of 10 rats each. Groups I and II (controls) were given 1 ml/kg body weight of physiological saline or corn oil (vehicle). Groups III and IV received 10 mg/kg and 20 mg/kg body weight embelin in corn oil, respectively. Emberlin disrupted the oestrous cycles in Groups III and IV animals, and there was a significant depression in plasma oestradiol (p <0.05) and progesterone (p <0.02) at both 10 and 20 mg/kg body weights, respectively. Isolated mixed ovarian cells from embelin treated rats produced significantly less progesterone and estradiol than controls in vitro. It is concluded that embelin probably interferes with reproductive functions in female rats by suppressing ovarian production of sex steroid hormones.  相似文献

1. Endogenous serotonin (5-HT), 5-hydroxyindol acetic acid (5-HIAA) content and exogenous 5-HT uptake (Km and Vmax) were measured in different brain regions (cerebellum, diencephalon, brain stem and telencephalon) of rats fed with a corn diet and restricted protein (8%) diet during 6 weeks. 2. A reduction of 5-HT levels was found in all regions studied of animals fed a corn diet, whereas, 5-HIAA was only decreased in brain stem and diencephalon. 3. An important increase in Km and Vmax were registered in brain stem and diencephalon of protein restricted animals, whereas, an increase of 5-HT uptake affinity in cerebellum, brain stem and telencephalon (35, 42 and 33% respectively) was observed. Simultaneously, under corn diet conditions, the Vmax decreased 40, 30 and 34% respectively in those regions. 4. It is suggested that the brain stem was the more sensitive area under nutritional restricted conditions and the development of some possible compensatory mechanisms of the 5-HTergic system is discussed.  相似文献

The present study evaluates the participation of the vagus nerve in pre-pubertal rats with unilateral ovariectomy on puberty onset, and on progesterone, testosterone and estradiol serum levels, and the compensatory responses of the ovary. Unilateral vagotomy did not modify the onset of puberty in unilaterally ovariectomized rats. Ovulation rates of animals with the left vagus nerve sectioned and the left ovary in-situ was lower than in rats with only unilateral ovariectomy. Sectioning the left vagus to 32-day old rats with the left ovary in-situ resulted in lower compensatory ovarian hypertrophy than in rats with right unilateral ovariectomy. Twenty-eight or 32-day old animals with sectioning of the right vagus nerve and the right ovary in situ showed higher compensatory ovulation. Twenty-eight -day old rats with the right ovary in situ had higher progesterone and testosterone levels than animals of the same age with the left ovary in-situ. Compared to animals with the right ovary in situ, animals treated at 32-days of age, sectioning the ipsi-lateral vagus nerve resulted in higher progesterone levels. Higher progesterone levels were observed in 28- and 32 days old rats with the left ovary in situ and left vagus nerve sectioned. Thirty-two day old animals with the right ovary in situ and right vagus nerve sectioned had higher progesterone levels than rats of the same age with the left ovary in situ and left vagus nerve sectioned. Left vagotomy to 28-day old rats with the left ovary in situ resulted in higher testosterone levels, a reverse response to that observed in animals with sectioning of the right vagus and the right ovary in situ. Thirty-two day old rats with the left ovary in situ and left vagus nerve sectioned showed lower testosterone levels than animals without vagotomy and with the left ovary in situ.  相似文献

The role of gonadal hormones in the control of platelet serotonin levels was studied by evaluating the effect of sexual maturation in rats of both sexes and the time-course of changes following gonadectomy performed either prepubertally or on sexually mature animals. In males, platelet serotonin levels remained fairly stable during sexual maturation as well as during the whole postgonadectomy period monitored (four months). In females, somewhat higher values of platelet serotonin levels in adult than in sexually immature animals were found (9%, p less than 0.001, N = 34). A slight decrease of platelet serotonin (10-18%, p less than 0.05) was observed following ovariectomy of sexually mature females, but it was of transient nature. When females were ovariectomized prepubertally a tendency towards permanently lower platelet serotonin levels was noticed. These results suggest that gonadal hormones have no major role in the control of platelet serotonin levels in rats, although a subtle hormonal modulation of this platelet variable in females may exist.  相似文献

Long-term (including gestational and lactational) restriction of protein (8% of diet) significantly lowered the absolute and relative consumption of 6% ethanol (EtOH) in a two-bottle, free-choice (H2O vs EtOH) situation during a 76-day test period. This difference in response between rats fed the low protein diet and those fed an isocaloric normal protein (24%) diet became non-significant in two subsequent 100-day test periods. Statistical analysis of observations on individual performance indicated that regularity, cyclicity, and duration of drinking in each animal was random over all three time intervals for both groups. The early, significantly lower EtOH consumption by the protein-restricted group may be due to a paucity of EtOH-metabolizing enzymes in brain and liver, thereby prolonging the CNS effects of lower doses of EtOH consumed. The disappearance of this difference in subsequent test periods may reflect either a behavioral or metabolic adaptation in the developing protein-deficient rat.  相似文献

Mean serum levels of testosterone (T) determined by radioimmunoassay were found to be reliably higher in male than female rats at 21 days of gestation and 6 hr and 3 days after birth. However, serum T concentrations in individual female samples were frequently equal to or greater than those found in male samples. Another study was performed to evaluate whether protection against androgen in females might be associated with mechanisms involving circulating progesterone and cellular adenosine 3′5′-cyclic monophosphate (cAMP). Female rats were injected on the day of birth with 1.0 μg testosterone propionate (TP) or sesame oil in combination with theophylline, antiserum to progesterone or saline. Females injected with TP lost vaginal cyclicity. The duration of time after puberty before reaching the sterility criterion was approximately 86% and 35% shorter for TP-groups receiving theophylline and antiprogesterone, respectively, than for those injected with saline. Thus, progesterone might provide some protection against the defeminizing effects of T and cAMP could be involved in hormone action during the perinatal period.  相似文献

Female gymnasts have a delayed onset and probably retarded progression of puberty. The aim of this study was to test the hypothesis that the delay in onset of puberty in gymnasts as compared to girl swimmers is modulated by a lower estrone level due to a smaller amount of body fat. The sex-hormone and gonadotropin levels of 46 gymnasts and 37 girl swimmers of the same biological maturation (breast development: M = 1 or M = 2) were studied. In each subject the following hormones were measured in plasma: estrone, 17-beta-estradiol, DHEAS, testosterone, androstenedione, LH, and FSH. In prepubertal children (M = 1) the levels of estrone, testosterone, and androstenedione were lower in the gymnastic group as compared to the swimming group. In the early pubertal (M = 2) gymnastic and swimming groups these hormone levels were no longer different. The other hormone levels were not significantly different in either the prepubertal groups or the early pubertal ones. Within the total prepubertal group there is a clear relationship between the estrone levels and the levels of testosterone and androstenedione, but not between estrone and 17-beta-estradiol, nor between the calculated fat mass and any of the hormone levels. It appears that the androstenedione and testosterone levels are responsible for the difference in estrone level, rather than the amount of body fat.  相似文献

Age-related changes in the levels of pituitary, thyroid, and sex hormones and their dependence on the day length were studied in 242 healthy 10-to 16-year-old boys born and permanently living in the southern part of Arkhangel’sk oblast. It was found that the serum levels of thyroid hormones, gonadotropins, and sex steroids underwent circannual changes. The minimum concentrations of thyroxine, testosterone, and estradiol and the maximum concentrations of triiodothyronine, lutcinizing and follicle-stimulating hormones, and progesterone were associated with the shortest days. In contrast, the longest days were associated with the maximum concentrations of thyroxine, testosterone, and estradiol and the minimum concentrations of triiodothyronine, luteinizing and follicle-stimulating hormones, and progesterone. A factor dependence of the thyroxine, triiodothyronine, testosterone, estradiol, and prolactin concentrations on the day length was found in the group examined. This dependence was more pronounced during the second half of adolescence.  相似文献

Exogenous bovine growth hormone-releasing factor (bGRF) at the dose rate of 10 microg/100 kg body weight was administered intravenously (i.v.) to six Murrah buffalo heifers as treatment group, while another six buffalo heifers served as control group which received the vehicle (0.9% NaCl solution) at an interval of 15 days for a period of 9 months to study the effect of bGRF on puberty onset associated with temporal hormonal changes in peri-pubertal buffalo heifers. Blood samples were collected at 3-day interval from all the animals during the experimental period and plasma harvested was assayed for growth hormonal (GH), luteinizing hormone (LH) and progesterone. The day that plasma progesterone was greater than 1.0 ng/ml for three consecutive sampling days was defined as the day of puberty. Exogenous bGRF administration increased (P = 0.02) plasma GH concentration in treatment group over control group during the treatment of bGRF as well as during the peri-pubertal period. Plasma progesterone concentrations increased transiently earlier (P = 0.05) by 58.5 days in bGRF-treated buffaloes than that in the control group. However, plasma LH concentrations were unaffected by the treatment of bGRF (P = 0.48). Both plasma GH and LH in the buffalo heifers increased (P < 0.01) over time preceding puberty and the higher hormonal concentrations were maintained during the onset of puberty, and thereafter, the concentrations of both the hormones declined (P < 0.05) after puberty. GH and LH were positively correlated both before puberty (r = +0.59 and +0.63; P < 0.05 for control and treatment group, respectively) and after puberty (r = +0.42 and +0.46; P < 0.05 for control and treatment group, respectively) indicating the interaction and/or close relationship of GH and LH in the mechanism of puberty in buffalo species.  相似文献

We measured progesterone and estradiol levels from birth to the beginning of adult life in 10 Thoroughbred fillies from the Equilia Stud Farm in Avaré SP, Brasil. The animals were measured and weighed monthly for the determination of body development and of a possible correlation between the rate of weight and height gain and the onset of detectable sex hormone levels. Jugular blood was collected twice a week and stored at -20 degrees C until assay of progesterone by a solid phase RIA with a sensitivity of 0.32 nmol L and of estradiol by liquid phase RIA adapted to low levels (3.67 pmol L ). The fillies were born with high serum levels of both hormones, which fell to undetectable levels by the first week of life. A variation in growth rate was observed, with the highest levels occurring from birth to the 3rd month of life, followed by a reduction until 15 mo of life, when fast growth was resumed. The monthly weight gain was 1.5% when the fillies reached puberty and 5.4% during the next month, (P < 0.05, Friedman test). During this second period of accelerated growth after the beginning of progesterone production at detectable levels (above 0.318 nmol L ), the parameters of skeletal growth did not differ (P > 0.05). The month of onset of puberty was the month of lowest weight gain in the life of the fillies, and it coincided with the highest insolation period. In conclusion, horses, like all other developed vertebrates, have a double pattern of development, with the acceleration observed at puberty depending on sex steroids, which in turn coincides with the highest insolation period. Gonadal activity characterized by serum progesterone levels was low from birth to the onset of puberty. After puberty the progesterone cycles were similar to those of adult animals with a mature hypothalamic-gonadal axis.  相似文献

The long-term consumption of a tryptophan-poor, corn diet by rats decreases electroshock response thresholds. This hyperalgesia appears to be related directly to diet-induced reductions in the brain concentrations of the putative neurotransmitter, serotinin. Rehabilitating corn-fed animals by feeding them the corn diets supplemented with tryptophan restores brain serotonin and pain thresholds to normal; similarly, injecting the tryptophan-deficient, corn-fed animals with fluoxetine, a drug that blocks the uptake of serotonin into brain neurons, also restores the electroshock response thresholds to control levels. The tryptophan hydroxylase inhibitor, p-chlorophenylalanine, increases the hyperalgesia to electroshock in corn-fed rats and further reduces brain serotonin concentrations. Injection of the amino acid valine, on the other hand, produces hyperalgesia and decreases brain serotonin in casein-fed rats but not in animals fed the corn diet. These data lend support to the hypothesis that serotonin neurons may mediate the sensitivity or reactivity to painful stimuli.  相似文献

Inoculation of cyclic female rats with the prolactin (Prl)/growth hormone-secreting pituitary tumor, MtT.W15, resulted in a cessation of estrous cyclicity within 5--10 days. Associated with this acyclicity was a persistently low serum concentration of estradiol and marked increases in both circulating Prl and progesterone. At Day 26 of acyclicity, basal serum luteinizing hormone (LH) values measured in samples taken every 20 min from 0900--1100 h were significantly reduced when compared to cyclic, nontumor animals on diestrus Day 2. There was no difference in basal follicle-stimulating hormone (FSH) concentrations. In a separate group of acyclic, tumor-bearing females 42--56 days after transplantation, a single s.c. injection of 20 micrograms estradiol benzoate (EB) at 1030 h elicited significant increases in both serum LH and FSH values between 1700 and 1830 h on the next day. The magnitude of the LH surge was reduced and that of FSH was increased in tumor-bearing animals when compared to cyclic, nontumor females given a similar EB injection on diestrus Day 1. These results demonstrate that chronic hyperprolactinemia is associated with inhibition of basal LH secretion and ovarian estrogen production and an increase in circulating progesterone concentrations. Nevertheless, the stimulatory feedback effects of estrogen on LH and FSH release are still present and functioning in acyclic female rats under chronically hyperprolactinemic conditions. These data suggest that the cessation of regular ovulatory cycles associated with hyperprolactinemia may be due to a deficiency of LH and/or estrogen secretion, but not to a lack of central nervous system response to the stimulatory feedback action of estrogen.  相似文献

Administration of either progesterone (P) or a dopamine agonist, Legrotrile mesylate (LM), have been shown to induce the ovulatory release of LH in rats. In order to elucidate the mode of action of dopamine agonists we studied the effects of LM on P secretion by the adrenals. A subcutaneous injection of LM, in doses which induce ovulation, stimulated adrenal P secretion in ovariectomized, estrogen-primed rats and in castrate male rats. Peak plasma P concentrations were found at 2 h with a gradual return to pre-injection levels at 6 hrs following LM injection. These results raise the possibility that P increments following LM administration may be responsible for inducing ovulation in young and old rats.There is considerable evidence to show that modification of hypothalamic monoamine metabolism by pharmacologic agents results in markedly altered pituitary gonadotropin secretion [1]. Lergotrile mesylate (LM), a dopamine agonist, has been shown to advance the ovulatory release of LH on proestrus and induce ovulation and cyclicity in anovulatory aged rats [2,3]. Everett [4] observed that progesterone (P) administration induced ovulation in rats rendered anovulatory under constant light and also advanced ovulation by one day if administered to 5-day cycling rats. A similar effect of LM on ovulation advancement has been noted (Clemens, personal communication). These similarities in action of P and LM on ovulation in young and aged rats, led us to speculate that LM may stimulate P secretion which in turn may elicit the discharge of ovulatory hormones. To test this hypothesis we examined the effects of LM on progesterone secretion in gonadectomized rats.  相似文献

This experiment concerned the changing patterns in secretion of luteinizing hormone (LH), follicle-stimulating hormone (FSH), and growth hormone (GH) under conditions of food restriction and subsequent catch-up growth. Weanling male rats were given either restricted (4 g food/day) or unrestricted access to food until 60 days of age. At this age, food-restricted rats weighed only 25% as much as rats fed ad libitum. Food restriction resulted in a dramatic decrease in the frequency of LH and GH pulses, and in the amplitude of GH pulses. It also slightly but significantly decreased mean blood levels of FSH (which was not secreted in a pulsatile manner in 60-day-old controls fed ad libitum). When restricted rats were given unrestricted access to food, frequency of LH and GH pulses and mean levels of FSH increased significantly and simultaneously within 2 days in half of the animals. Only an additional 8-10% of their body weight decrement was recovered at this time. After 10 days of food restoration, when restricted rats still weighed 50% less than controls, their secretory patterns of all three hormones were not significantly different from those of controls. Thus, recovery of gonadotropin and GH secretion was relatively rapid. Except for the quantitatively lesser impact of food restriction on FSH secretion, there was no evidence of any priorities in the secretion of the three hormones. Under conditions of rapid catch-up growth, the secretory patterns of LH, FSH, and GH appeared to develop simultaneously.  相似文献
