The impulse background activity (BA) of neurons of the rat medial vestibular nucleus (MVN) was subjected to computer analysis, and its modifications related to long-term vibration were studied. It was shown that following 5 days of 2-h-long vibration sessions, statistically significant changes in some basic characteristics of BA generated by MVN neurons were observed. More than a twofold increase in the mean BA frequency and substantial shifts practically of all statistical BA parameters were found after 10 days of vibration. Following a 15-day-long vibration, MVN neurons showed a clear-cut tendency to restore control values of the BA indices, which probably was related to adaptation processes.  相似文献

The background impulse activity (BIA) generated by neurons of the right lateral vestibular nucleus (LVN) of rats in the norm and under conditions of long-lasting general vibrational stimulation was subjected to computer analysis. Statistically significant changes in intragroup values of the mean BIA frequency were observed after 5 and 10 days with 2-h-long sessions of vibrational stimulation. Significant shifts in the distributions of LVN neurons by the level of regularity and dynamic types of BIA were observed 10 and 15 days with vibrational influences. Trends toward return of the intragroup mean value of the BIA frequency to the initial level were noticeable at the end of the stimulation period (15 days). We studied in rats changes in the impulse background activity (BA) of locus coeruleus (LC) neurons after short- and long-term immobilization stress; distributions of LC neurons by the level of regularity of their BA, dynamics of spike trains, and pattern of histograms of interspike intervals (ISI) were taken into account. We also calculated the means of the main BA statistical indices. Both short- and long-lasting immobilizations resulted in drops in the mean frequency of background discharges of LC neurons to about half of the initial value. Two-hour-long immobilization evoked statistically significant shifts in the distribution of LC neurons by the level of regularity of their BA, while after longer (15 h) immobilization this distribution nearly returned to the initial pattern. Short-lasting immobilization exerted no significant effect on the dynamic characteristics of BA; statistically significant changes in this respect developed only after longer stress. After 15-h-long immobilization, we also observed a noticeable increase in the number of neurons with polymodal ISI distributions. Therefore, stress results in significant modifications of the temporal parameters of the BA of LC neurons; characteristics of the BA of these neurons should be considered neuronal correlates of the stress state.  相似文献

We analyzed the impulse background activity (BA) of neurons of the inferior vestibular nucleus (IVN) of rats during exposure to long-lasting vibration (daily 2-h-long sessions). It was demonstrated that 5 days after the beginning of vibration stimulation, practically all main characteristics of the BA of IVN neurons changed significantly. Using several techniques of statistical analysis, we studied in detail the extracellularly recorded background impulse activity (BA) of neuronal elements of the rat locus coeruleus (LC). Impulse trains generated by most LC neurons were stationary and demonstrated different levels of regularity; a nonstationary type of BA was observed in 17% of the neurons under study. Statistical parameters of the BA generated by LC neurons showed a wide variability. Distributions of the BA interspike intervals (ISI) of most LC neurons were characterized by more or less expressed bimodality or polymodality. Serial correlation analysis of the ISI durations both in stationary and nonstationary spike trains allowed us to differentiate five main types of the dynamics of ISI successions in the BA of LC neurons.  相似文献

Distributions of neurons by the level of regularity, dynamics of discharge trains, form of histograms of interspike intervals (ISIs), as well as distributions of neurons by the BIA frequency ranges, were studied. We also calculated the mean frequency of impulsation of the neurons under study and the coefficient of variation of ISIs. After vibrational influences, we found modifications of both the internal structure of the recorded spike trains and the mean frequency of impulsation within the entire studied group and different frequency subgroups. We studied the voltage- and use-dependent action of pyrrolo-imidazole derivative, D57, on sodium currents in different dorsal root ganglion neurons of rats. At the level of 50% of maximum tonic block, which corresponded to a concentration of 0.44 mM, the use-dependent block of tetrodotoxin resistant (TTXr) sodium currents reached 59 ± 12% of the remaining currents when neurons were stimulated by 6-msec-long impulses up to -10 mV with a 20 sec-1 frequency, whereas for TTX sensitive (TTXs) currents this value was equal to 38 ± 9%. This block was dependent on the holding potential, and for cells with only TTXr currents the dependence was shifted to more positive potentials compared with that for neurons with only TTXs currents or with both of them.  相似文献

Correlation of impulse activity of visual and sensorimotor neurons of both hemispheres in 10-s interval after self-stimulation of the right and left lateral hypothalamus was studied by plotting cross- and autocorrelation histograms. The number of cell pairs, in which sensorimotor neurons discharged after visual ones, increased after self-stimulation (from 24 +/- 6 to 44 +/- 7%). Frequencies of the alpha- and theta-range in crosscorrelograms increased; the alpha frequency increased mainly in the right hemisphere, while the theta frequency increased in the left hemisphere. The interhemispheric asymmetry in the interaction between cortical neurons was not associated with the side of hypothalamic stimulation.  相似文献

We analyzed background impulse activity of neurons of the supraoptic nucleus of the rat hypothalamus in the course of 15-day-long isolated action of generalized vibrational stimulation and combination of such stimulation with irradiation of the animal’s head with low-intensity extrahigh-frequency (EHF, millimeter-range) electromagmetic waves. The distributions of the neurons by the level of regularity and dynamics of spike trains, separate frequency ranges of impulsation, and pattern of interspike interval (ISI) histograms were estimated. We also calculated the mean frequency of discharges and coefficient of variation of ISIs. A trend toward decreases in the deviations of some parameters of neuronal spike activity generated by supraoptic neurons, which were evident within early time intervals of isolated action of vibration (5 to 10 days), was observed under the influence of EHF electromagnetic irradiation; thus, the latter factor probably exerts a sedative effect. Sustained agonist stimulation induces an asymmetric down-regulation of brain muscarinic acetylcholine receptor (mAChR): 43±2% in the right and 26±2% in the left cerebral hemisphere, respectively (Ref. 1). In order to determine the possible involvement of endogenous diacylglycerols produced under muscarinic stimulation in the down-regulation phenomenon, here we have studied the effects of synthetic diacylglycerols and a phorbol ester on cells dissociated from rat cerebral cortex. Oleoylacetylglycerol decreased the amount of cell-surface mAChR by 37±2% and 25±2% in right and left cerebral cortex, respectively. Long-term treatment with phorbol dibutyrate also produced internalization of the mAChR (25±1.5% and 33±2% in right and left cortical cells, respectively). These changes occurred without modification of the Kdapp for the selective antagonist pirenzepine. The action of calcium ions was also studied using incubation of cells with the ionophore A23187. No changes were observed in the amount of mAChR detected at the plasma membrane with the ionophore alone, but when used in combination with phorbol dibutyrate and the agonist carbamylcholine a sinergistic decrease in mAChR was apparent. Experiments were performed to determine optimal conditions for callus induction and plantlet regeneration from damiana (Turnera diffusa, Willd.) leaf explants. MS media [7] and B5 media [4] were used; both of them were supplemented with different combinations of 2,4-dichlorophenoxyacetic, acid (2,4-D) and 6-benzyl-adenine (BA). In all cases the samples were incubated under continous fluorescent light, 100 mol.m-2.sec-1, at 25±2°C for 60 days. Highest callus percentage was obtained in MS media supplemented with 2,4-D 10–5 M plus BA 10-8M. Maximum percentage and multiple shoots and leaves were produced in MS media with BA 10-5M and BA 10-5M plus 2,4-D 10-7M. These explants were transferred for rooting to MS medium without plant growth regulators. Later, rooted plantlets were successfully established in soil.  相似文献

In acute experiments on albino rats anesthetized with Nembutal (40 mg/kg, i.p.), we recorded the background impulse activity (BIA) generated by neurons of the inferior olive in the norm and after 5-, 10-, and 15-daylong vibrational influence (60 Hz, 2 h, daily). We characterized the distributions of neurons according to the regularity of impulse successions, their dynamics, and pattern of histograms of interspike intervals (ISIs); we also calculated the mean frequency of impulsation and the coefficient of variation of ISIs. It was demonstrated that the most significant shifts of the characteristics of BIA generated by neurons of the inferior olive were formed within the first 10 days of the vibrational influence. These shifts were observed mainly in the mean discharge frequency (increased within the initial period) and, to a lesser extent, in the intrinsic structure of impulse trains. Such shifts in the background activity of the inferior olive caused by long-lasting vibrational influence result, perhaps, from intensification of the influences of excitatory cerebellar/mesodiencephalic inputs to olivary neurons within the early periods of action of the above factor and prevalence of GABAergic influences within the later periods. It seems possible that, under such conditions, the characteristics of electrical synapses of the olivary neurons are also subjected to modification. Summary Na+ and sugar permeabilities of egg lecithin bilayers were measured using curved bilayers and planar bilayers as represented by single-bilayer vesicles and black lipid films, respectively. The Na+ permeability coefficient measured with single-bilayer vesicles at 25°C is (2.1±0.6)×10–13 cm sec–1. Because of technical difficulties it has been impossible to measure ionic permeabilities of values lower than about 10–10 cm sec–1 in planar (black) lipid bilayers using tracer methods. Thed-glucose andd-fructose permeabilities were measured with both curved and planar bilayers. The permeability coefficients measured with vesicles at 25°C are (0.3±0.2)×10–10 cm sec–1 for glucose and (4±1)×10–10 cm sec–1 ford-fructose; these are in reasonable agreement with the corresponding values obtained for planar (black) lipid bilayers which are (1.1±0.3)×10–10 cm sec–1 ford-glucose and (9.3±0.3)×10–10 cm sec–1 ford-fructose, respectively.This paper is dedicated to the memory of Walther Wilbrandt,cuius nomini nullum par elogium.  相似文献

Excited-State Lifetimes of Far-Infrared Collective Modes in Proteins   总被引:1,自引:0,他引:1  
Vibrational excitations of low frequency collective modes are essential for functionally important conformational transitions in proteins. Here we report the first direct measurement on the lifetime of vibrational excitations of the collective modes at 87 pm (115 cm-1) in bacteriorhodopsin, a transmembrane protein. The data show that these modes have extremely long lifetime of vibrational excitations, over 500 picoseconds, accommodating 1500vibrations. We suggest that there is a connection between this relativelyslow anharmonic relaxation rate of approximately 10 g sec-1 and thesimilar observed rate of conformational transitions in proteins, which require require multi-level vibrational excitations and energy exchanges with othervibrational modes and collisional motions of solvent molecules.  相似文献

Summary Retrograde diffusion and precipitation of Co2+ reveals in the ipsilateral pars lateralis (PL) and contralateral pars intercerebralis (PI) of the brain neurons that enter the corpus cardiacum (CC), and, possibly, the corpus allatum (CA) on each side. The PL group consists of 29.6±8.4 somata that fill. Of these, 5.6±0.6 exceed 25 m in diameter, 14.3±2.7 range from 15–25 m, and 9.6±7.6 are smaller than 15 m. After CoCl2 was applied to the right CC-CA of two males, 239 and 265 somata in the left PI stained. Except for 16 ranging from 30–45 m and chiefly located anteriorly, a majority of these somata measured 10–25 m.The only somata revealed by staining whole brains with the performic acid-resorcin fuchsin method are neurosecretory cells 10–20 m in diameter located within the PI. In starved adult males there are 92.4±8.1 on the right, and 93.2±6.9 on the left. The largest somata in the PL group contain numerous granules that stain with paraldehyde fuchsin. These somata also fill with Co2+, and belong to neurosecretory cells that extend into the CC-CA.The cerebral distribution of branches from the PL group, and the relationship of these to the corpora pedunculata, central body, and arborizations from the PI decussation are described.  相似文献

Statistical analysis of the low-frequency (1.0 sec-1 and lower) neuronal impulse activity (IA) meets a few fundamental difficulties. Among them, the most significant is the small number of measurements (interspike intervals) recorded within an acceptable analysis epoch. In our study, we examined the possibility of using the normalized (by its maximum value) informational entropy (Hn) for estimation of the significance of changes in the IA generated by low-frequency neurons of the rostral hypothalamus after electrical stimulation of the prefrontal cortex. We compared the efficiencies of using the U-test (Kolmogorov–Mann–Whitney) and Hn estimate for the analysis of the same samples of neuronal responses. The results allow us to conclude that Hn is a significantly more acceptable estimate for detection of stimulation-induced modifications of the IA generated by low-frequency neurons, as compared with the U-test. The direction of shifts in the Hn value makes it possible to estimate the pattern of neuronal response. This value reflects the state of the neuron and correlates with the type of neuronal responses.  相似文献

Summary Volume-dependent changes in light scatter have been shown to be a linear function of the osmotic gradient imposed upon gastric vesicles purified from hog gastric mucosa. Observation of the light scattered 90° to incident, using the Durrum stop flow system D-110, indicates that the vesicles exposed to hypertonic medium undergo rapid shrinkage due to water loss from the vesicle interior. The rate constant for this water movement is 1.1±0.09 sec–1 (n=10) and is linearly dependent on temperature between 16 and 36°C. The activation energy of 13.93±0.60 kcal mole–1 (n=3), calculated from an Arrhenius plot, is inconsistent with water movement facilitated by a large-pore aqueous channel. A slower reswell phase, dependent on solute entry into the intravesicular space, follows the water-dependent shrink phase. KCl entry, studied because of the intravesicular requirement for active K+/H+ transport, exhibits two entry stages. The faster, described by a single exponential imposed upon a constantly sloping background, has a rate constant of 7.75±0.48×10–3 sec–1 (n=15). The slower phase, which typically accounts for 90% of the reswell process, demonstrates a rate constant of 1.94±0.23×10–4 sec–1 (n=15). In the presence of valinomycin or nigericin, two fast rate constants and one slow rate constant of swelling are observed. The rate constant of the faster reswell phase is increased from 7.75±0.48×10–3 sec–1 (n=15) to 15.74±3.7×10–3 sec–1 (n=5) and 17.23±3.4×10–3 (n=3) by the addition of nigericin (1 g ml–1) and valinomycin (4.5 m), respectively. The second part of the faster reswell phase is approximately that seen in the control population. Transport-dependent volume changes of significant magnitude can be demonstrated following the addition of ATP to vesicles equilibrated with 150mm KCl. The volume change is a function of HCl leak rate and is abolished by ionophores which eliminate the transport-dependent pH gradient. So 4 –- substitution, which eliminates the overshoot phenomena observed in KCl medium, also eliminates the shrinkage resulting from ATP addition.  相似文献

Summary The kinetics of the separate processes of Fe2(III)-transferrin binding to the transferrin receptor, transferrin-receptor internalization, iron dissociation from transferrin, iron passage through the membrane, and iron mobilization into the cytoplasm were studied by pulse-chase experiments using rabbit reticulocytes and59Fe,125I-labeled rabbit transferrin. The binding of59Fe-transferrin to transferrin receptors was rapid with an apparent rate constant of 2×105 m –1 sec–1. The rate of internalization of59Fe-transferrin was directly measured at 520±100 molecules of Fe2(III)-transferrin internalized/sec/cell with 250±43 sec needed to internalize the entire complement of reticulocyte transferrin receptors. Subsequent to Fe2(III)-transferrin internalization the flux of59Fe was followed through three compartments: internalized transferrin, membrane, and cytosol.A process preceding iron dissociation from transferrin and a reaction involving membrane-associated iron required 17±2 sec and 34±5 sec, respectively. Apparent rate constants of 0.0075±0.002 sec–1 and 0.0343±0.0118 sec–1 were obtained for iron dissociation from transferrin and iron mobilization into the cytosol, respectively. Iron dissociation from transferrin is the rate-limiting step. An apparent rate constant of 0.0112±0.0025 sec–1 was obtained for processes involving iron transport through the membrane although at least two reactions are likely to be involved. Based on mechanistic considerations, iron transport through the membrane may be attributed to an iron reduction step followed by a translocation step. These data indicate that the uptake of iron in reticulocytes is a sequential process, with steps after the internalization of Fe2(III)-transferrin that are distinct from the handling of transferrin.  相似文献

The soundness of hypotheses interpreting the mechanism of formation of background activity (BA) generated by the neurons of the rabbit superior cervical ganglion was analyzed, taking into account the statistical characteristics of this activity. These neurons are supplied with a multiple synapic input. According to the random hypothesis, the BA action potentials in the ganglion neurons result from occassional coincidence of the spikes arriving through independent presynaptic inputs. Summary Carrot suspension cells contain one class of high affinity binding sites for cytokinin in an 80,000 × g particulate fraction. Binding of [8-14C]-benzylaminopurine (BA) to this fraction assayed by a sedimentation method was found to be optimal at pH 6.0 and thermolabile. Specific binding was proved in competition experiments in which labelled BA was displaced by increasing concentrations of unlabelled BA. Scatchard plots of these results displayed a dissociation constant (Kd) of 33 ± 6 nM. The number of binding sites found was 1,100 ± 120 fmol g–1 fresh weight which is equivalent to a frequency of 23,000 binding sites per cell. The specificity of the binding sites to cytokinins and their analogues followed the sequence BA with highest affinity, kinetin, zeatin, iP and adenine. The cytokinin ribosides generally had a lower affinity than their cytokinin bases, and the affinity decreased in the order [9 R] BA, [9 R] iP, [9 R] Z, [9 R] A.  相似文献
