Within a joint research project entitled “Analysis and occurrence of importantFusarium toxins (deoxynivalenol and zearalenone) and dietary intake of these toxins by the German consumer”, supported by the German Federal Ministry of Consumer Protection, Food and Agriculture (BMVEL), representative analytical data are generated on the contamination level of foods withFusarium mycotoxins. This paper gives a comprehensive summary concerning the contamination of foods from the German market with deoxynivalenol (DON) in the period from August 2001 to April 2004. More than 4700 food samples (mostly cereals and cereal-containing foods) were purchased from food shops in Germany and analysed for DON by enzyme immunoassay, HPLC, and LC-MS/MS, respectively. All analytical methods were validated through intra- and interlaboratory studies and gave mean recoveries of >80% for each matrix. Although DON was detected with high frequency in all cerealcontaining samples, the mean and median levels were in most products well below the recently established maximum permitted limits in Germany.  相似文献

Growth and toxigenesis by Fusarium graminearum R6576, were compared in four liquid media. Parameters monitored during the fermentation were deoxynivalenol (DON) and 15-acetyl deoxynivalenol (15-ADON) production, fungal mass, carbohydrate utilization, and pH. Factors which were varied included basal medium composition, corn steep liquor (CSL) concentration, sucrose concentration and ammonium tartrate concentration. Growth in modified Fries medium resulted in only low levels of DON (0.25 mg/ L) and 15-ADON (0.25 mg/ L) after 20 days. Addition of 4% CSL to modified Fries medium raised the 20 day DON yield to 16.5 mg/ l. Increasing the sucrose concentration in modified Fries medium amended with 4% CSL resulted in increased mycelial dry weight but decreased levels of DON. Concentrations of ammonium tartrate greater than 1% in modified Fries amended with 4% CSL greatly reduced DON yield. Use of glucose-yeast extract-peptone (GYEP) for toxin production resulted in higher yields of 15-ADON (14.0 mg/ L) than DON (5.5 mg/ L) after 20 days. However, supplementation of GYEP with 4% CSL resulted primarily in DON production (4.5 mg/ L) after 20 days. In general, qualitative and quantitative production of DON and 15-ADON by Fusarium graminearum R6576 were dependent on the composition of the complex liquid medium.  相似文献

A study of experimental contamination of foods with spores of toxigenic micromycetes (Aspergillus flavus andPenicillium verrucosum) was realized. Samples of bread, apricot jam, Edam cheese, processed cheese and half-fat dried milk purchased on the market were used for experimental contamination. Phosphine is a food fumigant, used until now as an insecticide and rodenticide. The present work researches the action of phosphine treatment on growth and aflatoxin production of 23 Aspergillus strains. Production of aflatoxins B1, B2, G1, and G2 decreased in almost all cases by a ratio of 10 to 100. Phosphine treatment therefore seems favorable to prevent growth of various Aspergillus strains, in the context of keeping food safe.  相似文献

The deoxynivalenol and nivalenol contamination in wheat and by-products obtained through milling was analized by Trucksess method slightly modified in the proportion of acetonitrile—water (3:1). Only one sample of wheat showed deoxynivalenol contamination, 1,200μg/kg. No samples obtained in different stages of the milling were contaminated with deoxynivalenol or nivalenol. In the commercial wheat flours the levels found ranged between 400 and 800μg/kg, as follows: 400μ/kg, 5 samples; 800jug/kg, 1 sample.  相似文献

Deoxynivalenol (DON) in Durum Wheat High contamination levels of deoxynivalenol (DON) were found during an investigation of noodles in 2001 and initiated an additional survey of Durum wheat used as raw material for these products. Analyses of 53 samples of Durum wheat by HPLC revealed 89% positives and a median concentration of 790 ug/kg, which clearly exceeded the EU action level of 500 μg/kg. Based on these findings, producers increased quality controls on Durum wheat. In 2002 a total of 60 samples were analysed and 85% positives found. The median, however, lay at 215 μg/kg, below a probable maximum level of 500 μg/kg and demonstrated the positive efforts of food producers to decrease the contamination levels of DON in pasta products.  相似文献

The effects on testicular function of pulse-modulated microwave radiation (PM MWR, 1.3 GHz) and of conventional heating were studied in the rat. Anesthetized adult males (Sprague-Dawley, 400-500 g) were treated then killed at specific intervals with respect to the 13-day cycle of the seminiferous epithelium. PM MWR at 7.7 mW/g (90 min) yielded a modest decline in daily sperm production (DSP) that derived primarily from effects on primary spermatocytes. PM MWR at 4.2 mW/g was ineffective. The mean intratesticular temperature during the former reached 40 degrees C and did not exceed 38 degrees C during the latter. MWR considerably in excess of 7.7 mW/g yielded decrements in virtually all germ cell types, with primary spermatocytes again being most markedly affected. Using conventional heating, intratesticular temperatures in excess of 39 degrees C for 60 min were required for significant decrements in DSP. Levels of circulating follicle-stimulating hormone and of leutinizing hormone were resistant to either treatment. We conclude that the damage threshold and the differential sensitivity of immature germ cells to PM MWR can be adequately explained by the consequent macroscopic heating.  相似文献

The technical feasibility of adopting the fixed-film reactor concept for biogas production from screened dairy manure was investigated. The methane production capability of laboratory-scale 4-L anaerobic reactors (conventional and fixed-film) receiving screened dairy manure and operated at 35 degrees C was compared. Dairy manure filtrate with 4.4% total solids (TS) and 3.4% volatile solids (VS) (average value) was prepared from 1:1 manure-water slurry. The feed material was added intermittently at loading rates ranging from 2.34 to 25 and 2.25 to 785 g VS/L d, respectively, for the conventional and fixed-film reactors. Maximum methane production rate (L CH(4)/L d) for the conventional reactor was 0.63 L CH(4)/L d achieved at a 6-day hydraulic retention time (HRT). For the fixed-film reactor the maximum production rate was 3.53 L CH(4)/L d when operated at a loading rate of 262 g VS/L d (3 h HRT). The fixed-film reactor was capable of sustaining a loading of 785 g VS/L d (1 h HRT). The fixed-film reactor performed much better than the conventional reactors. These results indicate that a large reduction of required reactor volume is possible through application of a fixed-film concept combined with a liquid-solid separation pretreatment of dairy manure.  相似文献
