A new method for the determination of Cu(II) and Pb(II) by flame atomic absorption spectrometry (FAAS) after preconcentrating on a column containing Anoxybacillus sp. SO B1–immobilized Amberlite XAD-16 was developed. The functional groups of Anoxybacillus sp. SO B1 immobilized on Amberlite XAD-16 were characterized in KBr tablets by Fourier transform infrared (FT-IR) spectrometry. Various parameters such as pH, amount of the adsorbent, eluent type and volume, and flow rate of the sample solution were studied. The optimum pH values of quantitative sorption for Cu(II) and Pb(II) were found to be pH 7.0 and 5.0 and Cu(II) and Pb(II) ions could be quantitatively eluted with 5.0 ml of 1.0 mol L?1 HCI and 10.0 ml of 0.25 mol L?1 HNO3, respectively. Recoveries of Cu(II) and Pb(II) were found to be 100.9 ± 1.57% and 100.3 ± 0.49% (N = 5), the limits of detection of Cu(II) and Pb(II) in the determination by FAAS (3 s, N = 10) were found to be 0.8 and 1.6 μg L?1, respectively. The proposed enrichment method was applied for metal ion determination from water samples such as two parts of Tigris River water in Diyarbak?r and Elaz??, Lake of Hazar in Elaz??, and tap water in Diyarbak?r. Furthermore, the accuracy of the proposed method was verified by studying the analytical recovery and by analyzing certified reference material (NCS-DC 73350 leaves of poplar).  相似文献

Mispah form (FAO: Lithosol) soil contaminated with >380 000 mg kg?1 creosote was co-composted with cattle manure and mixed vegetable waste for 19 months. The soil was mixed with wood chips to improve aeration and then mixed with cattle manure or mixed vegetable waste in a ratio of 4:1. Moisture, temperature, pH, ash content, C:N ratios, and the concentrations of creosote in the compost systems were monitored monthly. The concentrations of selected hydrocarbons in the compost systems were determined at the end of composting. Temperature rose to about 45°C in the cattle manure compost within two months of incubation while temperature in the control and vegetable waste remained below 30°C until the fourth month. Creosote concentration was reduced by 17% in the control and by more than 99% in the cattle manure and vegetable waste compost after composting. The rate of reduction in concentration in the mixed vegetable waste compost was initially lower than in the cattle manure compost. The reduction rate became similar in later months with only small differences towards the end of the composting. The concentrations of selected creosote components were reduced by between 96% and 100% after composting. There was no significant difference in reduction in concentration in both compost systems at p 0.05. Microbial activity correlated with reduction in creosote concentration.  相似文献

Summary -Amylase production with Bacillus subtilis was studied in the presence of PEG 600 and PEG 3350 as well as different surfactants: Trixon X-100, Tween 80, CTAB (cetylammonium-bromide) and SDS (sodiumdodecylsulphate) at concentrations resulting in comparable decreases in surface tension. Only PEG 600, at a concentration of 200 g/kg, was found to increase the enzyme production. Cell growth estimated as optical density at 620 nm and viable counts were not influenced by either PEG or surfactants. The results are discussed in relation to -amylase production in aqueous two-phase systems.  相似文献

We have introduced the α-amylase gene from Bacillus licheniformis (amy gene) in a non-replicative plasmid which can be conveniently integrated and amplified at a specific site of the B. subtilis chromosome. Although we were able to select spontaneous and stable gene amplification of about 20 integrated copies, the amylase secretion remained very low. A DNA fragment presenting a high promoter activity in B. subtilis was therefore inserted upstream from the amy gene coding sequence, leading to a significant increase of amylase production. However, the amplified structures obtained with this construction were found to contain no more than 12 copies of the amy gene and to be rather unstable when cells were grown under non-selective conditions.  相似文献

In Bacillus subtilis cells, we identified a new partner of HPr, an enzyme of the glycolysis pathway, the glyceraldehyde-3-phosphate dehydrogenase GapA. We showed that, in vitro, phosphorylated and unphosphorylated forms of HPr and its homologue, Crh, could interact with GapA, but only their seryl-phosphorylated forms were able to inhibit its activity.  相似文献

A temperature-inducible mutant of temperate Bacillus bacteriophage phi105 was isolated and used to lysogenize a thymine-requiring strain of Bacillus subtilis 168. Synthesis of phage and bacterial deoxyribonucleic acid (DNA) was studied by sucrose gradient centrifugation and density equilibrium centrifugation of DNA extracted from induced bacteria. The distribution of DNA in the gradients was measured by differential isotope and density labeling of DNA before and after induction and by measuring the biological activity of the DNA in genetic transformation, in rescue of phage markers, and in infectivity assays. At early times after induction, but after at least one round of replication, phage DNA remains associated with high-molecular-weight DNA, whereas, later in the infection, phage DNA is associated with material of decreasing molecular weight. Genetic linkage between phage and bacterial markers can be demonstrated in replicated DNA from induced cells. Prophage induction is shown to affect replication of the bacterial chromosome. The overall rate of replication of prelabeled bacterial DNA is identical in temperature-induced lysogenics and in "mock-induced" wild-type phi105 lysogenics. The rate of replication of the bacterial marker phe-1 (and also of nia-38), located close to the prophage in direction of the terminus of the bacterial chromosome, is increased in induced cells, however, relative to other bacterial markers tested. In temperature-inducible lysogenics, where the prophage also carries a ts mutation which blocks phage DNA synthesis, replication of both phage and bacterial DNA stops after about 50% of the phage DNA has replicated once. The results of these experiments suggest that the prophage is not initially excised in induced cells, but rather it is specifically replicated in situ together with adjacent parts of the bacterial chromosome.  相似文献

Mannanase, an extracellular enzyme that catalyzes the hydrolysis of hemicelluloses to produce oligosaccharides, has potential to be applied in food industries. In this study a mannanase gene from B. subtilis Z-2 was isolated through PCR screening of a genomic DNA library. The nucleotide sequence of the mannanase gene, man, contained an open reading frame of 1080 bp, which codes for a deduced 26 amino-acid signal peptide and a mature protein with a deduced molecular mass of 38 kDa. The man gene can both be expressed heterologously into the periplasm from the plasmid pET22b(+) containing an intact signal peptide (pET-NdeI18) or the pelB signal peptide of the pET22b(+)vector (pET-NcoI3). Escherichia coli BL21 (DE3) containing pET-NcoI3 secreted about twice as much mannanase as that harboring pET-NdeI18. The E. coli DH5α expression of man was under the control of the lac promoter in the pRK415 vector; it was much more effective when the Shine Dalgarno (SD) sequence was changed from GGGGAG to AAGGAG and the start codon was changed from TTG to ATG, respectively. These results suggest that genetic modification of the SD sequence and start codon is practical for a high-level mannanase expression in different bacterial strains. The causes of night blindness in children are multifactorial, and particular consideration has been given to childhood nutritional deficiency, which is the most common problem found in underdeveloped countries. Such deficiency can result in physiological and pathological processes that in turn influence hair composition. This study was designed to compare the levels of zinc (Zn), copper (Cu), and iron (Fe) in scalp hair, blood, and urine of both genders of children with night blindness with age range of 3–7 and 8–12 years, comparing them to sex- and age-matched controls. A microwave-assisted wet acid digestion procedure was developed as a sample pretreatment, for the determination of zinc, copper, and iron in biological samples of children with night blindness. The proposed method was validated by using conventional wet digestion and certified reference samples of hair, blood, and urine. The digests of all biological samples were analyzed for Cu, Fe, and Zn by flame atomic absorption spectrometry using an air/acetylene flame. The results indicated significantly lower levels of Fe, Cu, and Zn in the biological samples (blood and scalp hair) of male and female children with night blindness, compared with control subjects of both genders. These data present guidance to clinicians and other professionals investigating the deficiency of essential trace metals in biological samples (scalp hair and blood) of children with night blindness.  相似文献

Rainfall erosivity (RE) is the ability of rainfall to cause soil or regolith erosion. Understanding the spatial distribution and temporal trends of RE is critical for assessing soil erosion risk and improving upon soil conservation planning. The aim of this paper is to study the temporal and spatial changes in RE in the Huaihe River Basin, China. This will be based on daily precipitation data from 67 meteorological stations in the Huaihe River Basin for the period 1971 to 2016. The assessment of the resulting RE values involved inverse distance-weighted (IDW) interpolation, Sen's slope estimation, and the Mann-Kendall nonparametric test. In addition, the possible influence of the El Niño – Southern Oscillation (ENSO) on RE in the Huaihe River Basin will be examined by the use of Cross Wavelet Transform (XWT) and Wavelet Coherence (WTC), where the relationship between RE and the multivariate ENSO index (MEI) are analyzed. The results showed that the spatial distribution of RE is characterized by it increasing from west to east and north to south. In most areas, the annual RE has increased slightly, while this upward trend in the southeastern part of the study area was more significant. The rainfall erosivity of rainstorms (RE-SM) in the Huaihe River Basin played a leading role in the annual RE, and 97% of the stations displayed the order of importance of the different categories of rainfall as rainfall erosivity of rainstorms (RE-SM) > rainfall erosivity of heavy rain (RE-HR) > rainfall erosivity of moderate rain (RE-MR). Over monthly time scales, the RE was the highest in July, while it was lowest in December, with the monthly differences being apparent. The RE in the Huaihe River Basin were relatively large during non-El Niño/La Niña periods and relatively small during El Niño/La Niña periods. Finally, correlations between RE and the MEI in various parts of the basin showed different characteristics over time and space, with both displaying similarities and differences.  相似文献

Toxic levels of heavy metals and low levels of essential minerals have been suggested to play a critical role in the pathogenesis of autism spectrum disorders (ASD). This study documents the levels of heavy metals and essential minerals in hair samples of children with ASD in Muscat, the urbanized capital of Oman, Muscat. The study included 27 children with ASD and 27 matched non-ASD controls. Parental interviews were held and dietary intake questionnaires completed in conjunction with the collection of hair samples. Analysis of heavy metals and essential minerals was carried out by inductively coupled plasma mass spectrometry. Chi-square analysis and non-parametric Fisher’s exact tests were used to assess statistical significance. Children with ASD had significantly higher levels of all 11 analyzed heavy metals in their hair samples (P?<?0.05), ranging from 150 to 365 % of control levels. ASD children also had significantly higher levels of essential minerals sulfur, sodium, magnesium, potassium, zinc, and iron, but lower levels of calcium and copper in their hair samples. This study corroborates data from previous studies in different parts of the world indicating the presence of elevated levels of heavy metals and selective depletion of essential minerals in the hair of children with ASD.  相似文献

This study describes the development by response surface methodology (RSM) of a procedure for copper determination by inductively coupled plasma optical emission spectrometry (ICP-OES) in water and biological samples after extraction by magnetic nanoparticles. Four variables such as, pH of solution, amount of extractant, amount of nanoparticles, and time were regarded as factors in the optimization study. Results of the two-level full factorial design (24) based on an analysis of variance demonstrated that only the pH, amount of extractant (E), and amount of nanoparticles (N) were statistically significant. Optimal conditions for the extraction of copper samples were obtained by using Box–Behnken design. Optimum conditions were 5.1, 7.2 mg, and 9.6 mg, for pH of solution, amount of nanoparticles, and amount of extractant, respectively. Under the optimized experimental conditions, the detection limit of the proposed method followed by ICP-OES was found to be 0.9?µg L?1. The method was applied to the determination of copper in water and biological samples.  相似文献

Summary Analysis of a large number of experimental data from the cultivation of Bacillus subtilis formed the basis for a kinetic model of the process explaining the effect of composition of the culture medium and of the growth rate on the rate of enzyme production. The resulting rate of formation of -amylase (EC 3.2.1.1) reflects the sum of the rate of enzyme production and the rate of its degradation as affected by the environment. The kinetic dependence confirms the previously described mechanism of regulation of enzyme biosynthesis. The mathematical model of the process served here to determine the optimal conditions for enzyme biosynthesis which were then verified in a fed-batch cultivation. An extracellular amylase (AmyKS) produced by a newly isolated Bacillus subtilis strain US572 was purified and characterized. AmyKS showed maximal activity at pH 6 and 60°C with a half-life of 10 min at 70°C. It is a Ca2+ independent enzyme and able to hydrolyze soluble starch into oligosaccharides consisting mainly of maltose and maltotriose. When compared to the studied α-amylases, AmyKS presents a high affinity toward soluble starch with a Km value of 0.252 mg ml−1. Coupled with the size-exclusion chromatography data, MALDI–TOF/MS analysis indicated that the purified amylase is a dimer with a molecular mass of 136,938.18 Da. It is an unusual feature of a non-maltogenic α-amylase. A 3D model and a dimeric model of AmyKS were generated showing the presence of an additional domain suspected to be involved in the dimerization process. This dimer arrangement could explain the high substrate affinity and catalytic efficiency of this enzyme.  相似文献

Radiolabeled phage SPO2 fragments were tested for the abiity to form interspecies deoxyribonucleic acid (DNA) duplexes with DNA from Bacillus subtilis and from phages beta3 and beta22. No reassociation, above control values, occurred between the DNA of phage SPO2 and that of its host or either of the virulent B. subtilis phages. It appears that extensive nucleotide sequence similarities between portions of host and phage DNA species are not necessary for the formation of the lysogenic state. The thermal elution profile of reassociated SPO2 DNA exhibited a normal distribution, indicating no heterogeneity in base composition. To produce PreS2 antigen of human hepatitis B virus extra-cellularly in Bacillus subtilis, it was fused with the COOH-termini of B. subtilis -amylases of 522 (Amy+), 467 (Amy+) and 443 (Amy) amino acids, Among them, -amylase-A467, which has 467 amino acids, was a relatively stable carrier when the cells were cultured in Luria-Bertani (LB) medium. The -amylase-A443-PreS2 hybrid protein (Amy) was quickly degraded. The -amylase-A522-PreS2 hybrid was most efficiently produced when a B. subtilis transformant of a protease-super-deficient mutant was cultured in the presence of 0.5 m sodium sulphate. The production of A522-PreS2 hybrid protein under such conditions reached 5–10 mg/l and was eight, and 200–500 times higher than those obtained by the transformants of an alkaline/neutral protease-deficient mutant of B. subtilis, and a wild-type strain, in LB medium, respectively. Background

The amyloid-β (Aβ) peptide is the primary component of the extracellular senile plaques characteristic of Alzheimer''s disease (AD). The metals hypothesis implicates redox-active copper ions in the pathogenesis of AD and the Cu2+ coordination of various Aβ peptides has been widely studied. A number of disease-associated modifications involving the first 3 residues are known, including isomerisation, mutation, truncation and cyclisation, but are yet to be characterised in detail. In particular, Aβ in plaques contain a significant amount of truncated pyroglutamate species, which appear to correlate with disease progression.

Methodology/Principal Findings

We previously characterised three Cu2+/Aβ1–16 coordination modes in the physiological pH range that involve the first two residues. Based upon our finding that the carbonyl of Ala2 is a Cu2+ ligand, here we speculate on a hypothetical Cu2+-mediated intramolecular cleavage mechanism as a source of truncations beginning at residue 3. Using EPR spectroscopy and site-specific isotopic labelling, we have also examined four Aβ peptides with biologically relevant N-terminal modifications, Aβ1[isoAsp]–16, Aβ1–16(A2V), Aβ3–16 and Aβ3[pE]–16. The recessive A2V mutation preserved the first coordination sphere of Cu2+/Aβ, but altered the outer coordination sphere. Isomerisation of Asp1 produced a single dominant species involving a stable 5-membered Cu2+ chelate at the amino terminus. The Aβ3–16 and Aβ3[pE]–16 peptides both exhibited an equilibrium between two Cu2+ coordination modes between pH 6–9 with nominally the same first coordination sphere, but with a dramatically different pH dependence arising from differences in H-bonding interactions at the N-terminus.

Conclusions/Significance

N-terminal modifications significantly influence the Cu2+ coordination of Aβ, which may be critical for alterations in aggregation propensity, redox-activity, resistance to degradation and the generation of the Aβ3–× (× = 40/42) precursor of disease-associated Aβ3[pE]–x species.  相似文献
