We investigated different concentrations of auxins (1AA, IBA, NAA), the strength of the MS medium, sucrose and ammonium/nitrate contents, initial medium pH, and inoculum size to determine their effects on biomass increase and the accumulation of total phenols and flavonoids in adventitious roots ofEchinacea angustifolia. These roots were cultured under darkness in shake flasks for 4 weeks. IBA proved the best auxin for inducing root proliferation. Root growth was inhibited when the initial pH was maintained below 5.0 or above 6.0. Nitrate, rather than ammonium, was more necessary for root growth and phenolics accumulations. Overall, biomass increased and total phenol and flavonoid contents were maximized under the following conditions: half-strength MS medium supplemented with 2 mg L-1 IBA, 5% (w/v) sucrose, 5:25 (mM) ammonium/nitrate ratio, pH adjusted to 6.0 before autoclaving, and an inoculum size of 10 g L-1 FW. These results indicate that the type ofin vitro environment strongly affects growth and the accumulation of phenolics from adventitious root cultures ofE. angustifolia. Such optimization is beneficial to large-scale production of biomass and secondary metabolites in that species.  相似文献

以自选育的白花泡桐优树茎段为外植体,进行种苗组培快繁技术研究。结果表明:其最佳的外植体灭菌方法是以0.1%升汞处理7 min;合适的初代诱导培养基为MS+6-BA 2.0 mg·L~(-1)+IBA 0.2 mg·L~(-1)+糖30 g·L~(-1)+琼脂3.5 g·L~(-1)(pH 5.8),培养30 d,芽诱导率70%;合适的继代增殖方法为在高浓度植物生长物质培养基MS+6-BA 4.0 mg·L~(-1)+IBA 0.4 mg·L~(-1)+蔗糖30 g·L~(-1)+琼脂3.5 g·L~(-1)(pH 5.8)和低浓度植物生长物质培养基MS+6-BA 0.4 mg·L~(-1)+IBA 0.04 mg·L~(-1)+蔗糖30 g·L~(-1)+琼脂3.5 g·L~(-1)(pH 5.8)中交替培养,获得的丛生芽长势良好,玻璃化率低于5%,增殖系数大于6.0/25 d;最适的生根培养基为1/2MS+NAA0.2 mg·L~(-1)+蔗糖20 g·L~(-1)+卡拉胶3.4 g·L~(-1)(pH 5.8),培养14 d,得到白花泡桐生根苗,每株长根5~10条,根长3~5 cm,生根率98%,根系洁白、根毛少而短,易于清洗。将生根苗按照常规方法炼苗后移栽于温室大棚中,50 d后即可出圃,此时平均苗高1.0 m、地径1.0~2.0 cm,成活率在90%以上。  相似文献

Hairy root cultures have demonstrated great promise in terms of their biosynthetic capability toward the production of secondary metabolites, but continue to constitute a major challenge with regard to large-scale cultures. In order to assess the possibility of conducting mass production of biomass, and the extraction of useful metabolites fromPanax ginseng. P. ginseng hairy roots, transformed byRhizobium rhizogenes KCTC 2744, were used in bioreactors of different types and sizes. The most effective mass production of hairy roots was achieved in several differently sized air bubble bioreactors compared to all other bioreactor types. Hairy root growth was enhanced by aeration, and the production increased with increasing aeration rate in a 1 L bioreactor culture. It was determined that the hairy root growth rate could be substantially enhanced by increases in the aeration rate upto 0.5 wm, but at aeration rates above 0.5 wm, only slight promotions in growth rates were observed. In 20 L air bubble bioreactors, with a variety of inoculum sizes, the hairy roots exhibited the most robust growth rates with an inoculum size of 0.1% (w/v), within the range 0.1 to 0.7% (w/v). The specific growth rates of the hairy roots decreased with increases in the inoculum size.  相似文献

Summary A method for the micropropagation of Spathiphyllum cannifolium is presented using shoot tip proliferation onto Murashige and Skoog (MS) medium supplemented with different plant growth regulator concentrations and combinations. The proliferation responses were significantly influenced by the cytokinin type and concentrations. Supplementation of the medium with benzyladenine (BA; 4.44–13.32 μM) increased the shoot proliferation rate significantly as compared to other treatments. When cytokinins were used with auxin (indole-3-butyric acid, IBA and naphthalene acetic acid. NAA), the number of shoots per explant increased in comparison with treatments with BA alone. The largest number of shoots, 9.3 per explant, was obtained with 13.32 μM BA and 4.9 μM IBA. Different MS medium strengths and sucrose concentrations were used with the aim to stimulate in vitro shoot proliferation. Full MS medium with 30 gl−1 sucrose was found to be suitable for shoot tip culture of Spathiphyllum. Comparative studies between gelled medium and bioreactor culture [continuous immersion (with or without net) and temporary immersion in liquid media using ebb and flood] revealed that shoot multiplication and growth were more efficient in continuous immersion (with net) bioreactor with low cytokinin-supplemented media. Plantlets from the bioreactor were cultured hydroponically for 30 d and 100% of plants were rooted and acelimatized successfully. Rapid and efficient multiplication rate in bioreactor and successful transfer to greenhouse makes this protocol suitable for large-scale multiplication of this important foliage plant.  相似文献

采用正交试验设计法,研究了NAA、KT、2,4-D、蔗糖浓度和不同外植体类型等因素对银杏愈伤组织诱导的影响。结果表明:不同外植体类型对银杏愈伤组织诱导率影响最大,KT和NAA其次,2,4-D和蔗糖浓度最小。银杏愈伤组织诱导最佳培养基为MS+NAA 0.5 mg·L-1+KT 0.5 mg·L-1+蔗糖40 g·L-1,最佳外植体为茎段,其愈伤组织诱导率可达100%。  相似文献

The effect of the root-inoculum size and axuin concentration on growth of adventitious roots and accumulation of ginsenosides were studied during suspension cultures of ginseng (Panax ginseng C.A. Meyer). Of the various concentrations of indole-3-butyric acid (IBA) and γ-naphthaleneacetic acid (NAA) used as supplementary growth regulators along with Murashige and Skoog medium, 25 μM IBA was found suitable for lateral root induction and growth, as well as accumulation of ginsenosides. Inoculum size of 5 g L−1 was found suitable for optimal biomass (10.5 g L−1 dry biomass) and ginsenosides (5.4 mg g−1 DW) accumulation. Of the various length of root inocula tested (chopped to 1–3, 4–6, 7–10 mm and un-chopped), root inocula of 7–10 mm was found suitable for biomass and ginsenoside accumulation.  相似文献

The effects of carbon sources (sucrose, glucose, fructose and mannitol) and auxins [indolebutyric acid (IBA) and α-naphthaleneacetic acid (NAA)] on in vitro propagation of banana (Musa spp. AAA) were studied. Over all carbon sources tested, sucrose induced highest frequency of shoot proliferation. Optimal shoot proliferation rates were achieved on the Murashige and Skoog (MS) medium supplemented with sucrose and glucose combination (1:1) at the concentration of 30 g dm−3. Similarly, higher frequency of root induction was obtained at IBA and NAA combination (1:1; concentration of 2 mg dm−3) than at other concentrations of IBA or NAA alone or their combinations.  相似文献

Summary Friable calli were obtained fromAchillea millefolium L. hypocotyls, in Gamborg B5 medium, supplemented with 1.5mg.1–1 2,4-D / 0.1mg.1–1 Kin, and used for the production of cell suspension cultures in the same liquid medium. The growth pattern of the cultures was determined in permanent light or dark conditions and with different inoculum densities, basal media, growth regulators and sucrose concentrations. Different sources and nitrogen amounts were assayed to study the effect on yarrow cell growth. The conditions found to be optimal for growth of yarrow cell suspension cultures were: 70g (f.w.).1–1 of initial inoculum in Gamborg B5 medium, supplemented with 1.5mg. 1–1 2,4-D / 0.1mg.1–1 Kin, NO3 /NH4 + (30/lmM), and 2% sucrose, in darkness. 为探究裸果木再生体系建立的影响因素,确定其不定芽发生的起源,该研究以裸果木健壮植株的茎段为外植体,采用6 BA和IBA不同浓度组合,筛选愈伤增殖及不定芽再生的最佳浓度组合,确定生根诱导的关键影响因素,建立再生体系,并对其不定芽分化进程进行解剖结构分析,以确认其起源。结果表明:(1)裸果木茎段的最佳愈伤增殖培养基为MS+1 mg·L-1 IBA+1 mg·L-1 6 BA+30 g·L-1蔗糖+7 g·L-1琼脂,主体间效应分析表明IBA为关键影响因素;愈伤大小随IBA浓度增加呈现先升高后下降的趋势。(2)最佳不定芽诱导培养基为MS+0.5 mg·L-1 6 BA+30 g·L-1蔗糖+7 g·L-1琼脂,诱导不定芽数量为4.9个/块,生芽率达92.3%。(3)生根诱导中,SH基本培养基和蔗糖浓度为关键因素,最佳生根培养基为SH+0~10 g·L-1蔗糖+7 g·L-1琼脂,生根率达91.3%。(4)解剖结构观察发现,不定芽起源于愈伤表层的分生细胞,为外起源。该研究通过器官发生途径建立了裸果木的再生体系,确定了不定芽为外起源,为裸果木这一珍稀濒危的林木种质资源保护及可持续利用奠定了研究基础,并为其未来的发展利用提供了有效途径。  相似文献

Summary Explants from petioles, folioles or hypocotyls ofOnonis natrix have been used for calli initiation. Hypocotyls inoculated on MS medium supplemented with 2% sucrose and 0.5 mg.1–1 2,4-D / 1 mg.1–1 Kin showed to be the best primary explant. Cell suspension cultures were established in MS basal medium supplemented with 2% sucrose, 0.5 mg.1–1 NAA or 2,4-D and 1 mg.1–1 Kin. Different subculturing periods, inoculum density, hormonal supplementation and sucrose concentration were assayed in order to obtain the best culture growth conditions. The optimal conditions were achieved with cultures initiated with 40 g.1–1 of initial inoculum, growing in MS basal medium supplemented with 4% sucrose, 0.5 mg.1–1 NAA and 1 mg.1–1 Kin subcultured every twelve days. Under these experimental conditions, the cultures showed a doubling time of 36.3 hours.  相似文献

SummarySelf-directing optimization was successfully employed to determine the optimal combination of engineering parameters, viz., pH, aeration rate and agitation rate, for extracellular ribonuclease production by Aspergillus niger SA-13-20 in a batch bioreactor. Maximal RNase production of 5.38 IU ml–1 was obtained at controlled pH of 2.33, aeration rate of 1.67 v/v/m and agitation rate of 850 rev/min. The effect of oxygen on the fermentation was also investigated. With increase in volumetric oxygen transfer coefficients (KLa), cell growth and RNase production first increased and then decreased. RNase production was further increased to 7.10 IU ml–1 and the fermentation time was shortened from 96 to 72 h by controlling dissolved oxygen concentration at 10% saturation by aerating oxygen after about 28 h of fermentation under the above optimal condition. The kinetic model showed that RNase production by A. niger SA-13-20 was growth-associated.  相似文献

以红花草莓叶片为外植体,通过筛选诱导愈伤组织、不定芽及壮苗、生根的培养基,建立一套实用且易推广的红花草莓组培快繁技术体系。结果表明:在愈伤组织的诱导过程中TDZ的诱导效果优于6-BA,TDZ与NAA配合使用效果优于与IBA的组合。6-BA浓度为0.5 mg·L~(-1)时不定芽诱导率高达86.6%。低浓度的6-BA和8 g·L~(-1)的琼脂更有利于壮苗培养,NAA比IBA更有利诱导生根。综上述,最适红花草莓愈伤组织的诱导培养基为MS+1.0 mg·L~(-1)TDZ+0.5 mg·L~(-1)NAA+30 g·L~(-1)蔗糖+7 g·L~(-1)琼脂;最适不定芽分化的培养基为MS+0.5 mg·L~(-1)6-BA+0.1 mg·L~(-1)NAA+30 g·L~(-1)蔗糖+7 g·L~(-1)琼脂;最适壮苗培养基为MS+0.1 mg·L~(-1)6-BA+0.1 mg·L~(-1)NAA+30 g·L~(-1)蔗糖+8g·L~(-1)琼脂;最适生根培养基为MS+0.5mg·L~(-1)NAA+30 g·L~(-1)蔗糖+8 g·L~(-1)琼脂。试管苗移栽生长20 d后,成活率高达93%,且后期草莓苗生长壮健。此体系的建立为优质红花草莓种苗大规模生产提供了科学依据和技术支持。  相似文献

Liquid cultures were successfully generated from cotyledons of two Sonneratia species, S. alba and S. caseolaris in Murashige and Skoog (MS) medium containing 0.1 μmol L−1 2,4-dichlorophenoxyacetic acid (2,4-D). Adventitious roots differentiated from cotyledons of S. alba. Proliferated cells were subcultured and a large volume of suspension cells was subsequently established in 100-mL flasks. All the cytokinins tested inhibited cell proliferation. After three years of culture, the potential to differentiate was tested as indicated by greening of the cells. Greening occurred when suspension cells were transferred to solid MS medium with and without 0.1 μmol L−1 2,4-D. Greening was stimulated by low concentrations of the weak auxins indolebutyric acid (IBA) and naphthaleneacetic acid (NAA) while 2,4-D stimulated late-stage greening. Abscisic acid (ABA) inhibited greening. Gibberellic acid (GA3) at 1.0 μmol L−1 stimulated callus greening and was not inhibitory even when tested at high concentrations. Cytokinins were inhibitory in combination with 0.1 μmol L−1 of either IBA or NAA. The cause of different effects of plant hormones on growth and differentiation was discussed. Small-scale liquid media and 24-well culture plates of solid media methods developed in this paper are suitable for the optimization of hormonal conditions for cell proliferation and differentiation.  相似文献

Summary Mutant lines of Nicotiana plumbaginifolia resistant to the synthetic auxins 1-naphthaleneacetic acid (NAA) and indole-3-butyric acid (IBA) were isolated as germinating seedlings on selective medium. In each case, resistance was conferred by a single recessive nuclear mutation at one of 3 loci designated iba1, iba2 and iba3. Labelling studies with 14C NAA suggest that resistance was not due to changes in the uptake or metabolism of NAA. Plants homozygous for the iba1 mutation exhibit a syndrome of atypical germination and growth suggestive of a defect in the biosynthesis, metabolism or localization of abscisic acid. Wild-type seeds treated with gibberellin exhibit the same syndrome, including resistance to NAA and IBA. Arthrospira (Spirulina) platensis M2 was grown outdoors in 50-mm diameter tubular reactors under the climatic conditions of central Italy (Florence) from September to December 1995 and in March 1996. Except for September, the cultures temperature was regulated. Mean productivities of 0.83, 0.44 and 0.61 g dry wt L–1 d–1 were achieved in autumn (September–October), winter (November–December) and March, respectively. In autumn and winter, the photosynthetic efficiency of the cultures and the degree of correlation between productivity and solar irradiance were significantly greater than in summer. The effect of cell density and aeration rate on productivity was evaluated in September. The productivity of cultures operated at high supra-optimal population density was about 30% less at high aeration rate (1.0 LL–1 min–1), and 50% less at standard aeration rate (0.17 LL–1 min–1), than that of control cultures kept at optimal population density and standard aeration rate. The reduction of productivity in high-density cultures was due to lower daylight output rates and higher night biomass losses (the latter were particularly relevant under standard aeration conditions). The main factor limiting productivity in closed reactors during autumn was the night temperature. Heating the cultures during daylight hours on sunny days did not cause any significant increase of the yields, since under sunlight the unheated cultures also reached the optimal temperature for growth early in the morning. On cloudy days, the day-time temperature of the unheated cultures remained well below the optimum, however this had only a limited effect on productivity since algal growth was mainly light-limited.  相似文献

Somatic embryogenesis and further plant regeneration were observed using zygotic embryos, young inflorescences and young leaves ofEuterpe edulis (Palmae) as explants. Both for the cultures of zygotic embryos and inflorescences, activated charcoal in the medium was essential for the establishment of viable cultures. Embryogenesis was induced by using a gelled basal medium with MS or Euwens salts supplemented by high 2, 4-D levels (50–100 mg L−1). The embryogenic process was direct without a callus stage. For further development, cultures with globular or post-globular embryos were transferred to the basal medium with 2-iP (2.5 mg L−1) and NAA (0.1 mg L−1). To convert embryos to plantlets, cultures were transferred to a third medium in which sucrose and salts were reduced to the half-strenght of the basal medium, without growth regulators. In the case of liquid medium, with either 2, 4-D or NAA (10–20 mg L−1). The developmental stage of each explant was critical for the induction of embryogenesis. The histological study of embryogenic cultures revealed that in the case of zygotic embryos, somatic embryos arise directly from the surface of the cotyledonar node, or from subepidermal tissues. In the inflorescences, a pro-embryogenic tissue is formed at the floral primordium region; in the leaves, the first morphogenic event is cell proliferation in the vascular parenchyma.  相似文献

Astragalus membranaceus is one of the most widely used traditional medicinal herbs in China, but the time required to generate a useful product in the field production is long. The growth of adventitious root cultures was compared between cultures grown in solid, liquid, or a 5-L balloon-type bubble bioreactor. The maximum growth ratio (final dry weight/initial dry weight) was determined for adventitious roots grown in the bioreactor. Studies carried out to optimize biomass production of adventitious roots compared adventitious root growth from various inoculum root lengths, inoculum densities, and aeration volume in the bioreactors. The maximum growth ratio occurred in treatments with a 1.5-cm inoculum root length, with 30 g (fresh weight) of inoculum per bioreactor or with an aeration volume of 0.1 vvm (air volume/culture medium volume per min). The polysaccharide, saponin, and flavonoid content of roots from bioreactor-grown cultures were compared to roots from field-grown plants grown for 1 and 3 yr. Total polysaccharide content of adventitious roots in the bioreactor (30.0 mg g−1 dry weight (DW)) was higher than the roots of 1-yr-old (13.8 mg g−1 DW) and 3-yr-old (21.1 mg g−1 DW) plants in the field. Total saponin (3.4 mg g−1 DW) and flavonoid (6.4 mg g−1 DW) contents were nearly identical to 3-yr-old roots and higher than that of 1-yr-old roots under field cultivation.  相似文献

A bioreactor system for biotoxin production was appraised against traditional methods of growing dinoflagellate cultures. In an optimised bioreactor culture (5.4?L) operated in batch mode, growth of Karenia selliformis was more efficient than in 15-L bulk carboy culture in terms of growth rate (μ?=?0.07?day?1 versus 0.05?day?1) and growth maximum (G max, 169.106 versus 41.106 cells L?1). Maximal gymnodimine concentration (1200?μg L?1) in bioreactor culture was 8-fold higher than in bulk carboy culture, and the yield per cell (pg cell?1) was 2-fold higher. Similarly the bioreactor batch culture of Alexandrium ostenfeldii performed more efficiently than carboy cultures in terms of growth rate (1.6-fold higher), growth maximum (15-fold higher) and desmethyl C spirolide (SPX-desMe-C) yield (5-fold higher [μg L?1], though the yield [pg cell?1basis] was lower). When bioreactor cultures of K. selliformis were operated in continuous mode, the yield of gymnodimine was substantially higher than a carboy or the bioreactor run in batch mode to growth max (793?μg day?1 over 58?days in continuous culture was achieved versus an average of 60?μg day?1 [carboy over 40?days] or 249?μg day?1 [batch mode] over 26?days). Likewise in continuous bioreactor cultures of A. ostenfeldii run over 25?days, the yield of SPX-desMe-C (29?μg day?1) was substantially higher than in same cultures run in batch mode or carboys (10.2 day?1 and 7.7?μg day?1 respectively). Similarly 5.4?L bioreactor batch cultures of K. brevisulcata reached 3.8-fold higher cell densities than carboy cultures, and when operated in continuous mode, the brevisulcatic acids were more efficiently produced than in batch culture (12?μg day?1 versus 7?μg day?1). When the bioreactor system was upscaled to 52?L, the maximum cell densities and toxin yields of K. brevisulcata cultures were somewhat less than those achieved in the smaller reactor, which was attributed to reduced light penetration.  相似文献

The effect of agitation and aeration on the growth and antibiotic production by Xenorhabdus nematophila YL001 grown in batch cultures were investigated. Efficiency of aeration and agitation was evaluated through the oxygen mass transfer coefficient (K L a). With increase in K L a, the biomass and antibiotic activity increased. Activity units of antibiotic and dry cell weight were increased to 232 U ml−1 and 19.58 g l−1, respectively, productivity in cell and antibiotic was up more than 30% when K L a increased from 115.9 h−1 to 185.7 h−1. During the exponential growth phase, DO concentration was zero, the oxygen supply was not sufficient. So, based on process analysis, a three-stage oxygen supply control strategy was used to improved the DO concentration above 30% by controlling the agitation speed and aeration rate. The dry cell weight and activity units of antibiotic were further increased to 24.22 g l−1 and 249 U ml−1, and were improved by 24.0% and 7.0%, compared with fermentation at a constant agitation speed and a constant aeration rate (300 rev min−1, 2.5 l min−1).  相似文献

该文以广西野生金线莲无菌播种离体茎段为材料,采用单因素对比试验,研究了植物激素(NAA、IBA、6-BA、GA_3、KT、ZT、TDZ、2-IP)以及接种方式(竖直接种和水平接种)对壮苗生根培养的影响。结果表明:与对照相比,生长素有利于壮苗生根,NAA的效果优于IBA;细胞分裂素对壮苗生根的效果依次为6-BATDZKT=ZT 2-IPCK,其中6-BA诱导平均株高8.4 cm,3.6条根,茎粗为2.84 mm,植株生长健壮,诱导效果最好;赤霉素GA_3诱导出的植株高且直,但植株细弱,且抑制根系生长,不利于壮苗生根培养;在激素组合6-BA 0.5 mg·L~(-1)、NAA1.0 mg·L~(-1)处理中,组培苗生长健壮且根数量较多,效果最佳;水平接种能诱导出更多的根系,且便于接种操作,可以节省接种时间。因此,确定广西金线莲最适宜的壮苗生根培养基配方为1/2MS+6-BA 0.5 mg·L~(-1)+NAA 1.0 mg·L~(-1)+香蕉汁100 g·L~(-1)+AC(活性炭) 1.0 g·L~(-1)+蔗糖20 g·L~(-1),最佳接种方式为水平接种。  相似文献
