 Biomass, production and heterotrophic activity of bacterioplankton were determined for two weeks in the Great Astrolabe Reef lagoon, Fiji. Bacterial and Bacterial activities were distributed homogeneously throughout the water column (20 to 40 m deep) and varied little from site to site inside the lagoon. Bacterioplankton biomass and production also varied little over a diel period with coefficients of variation of 9 and 22%, respectively. On average, over the whole study, bacterial abundance was 0.77×109 cells l-1 and bacterial production averaged 0.36 μg-at. C l-1 d-1. Bacterial abundance and production were greater in the lagoon than in oceanic waters. Attachment to particles seems to provide an advantage for bacterioplankton growth because specific growth rates for attached bacterioplankton were, on average, significantly greater than that of the free community. Growth efficiency, determined by correlating the net increase of bacterial biomass and the net decrease of dissolved organic carbon (DOC) in dilution cultures, was very low (average 6.6%). Using carbon growth efficiency and bacterial production rates, heterotrophic activity was estimated to average 5.4 μg-at. C l-1 d-1. The turn-over rate of DOC (average 114 μg-at. Phytoplankton production and accumulation of extracellular organic carbon (EOC) was tracked during diel intervals in microcosms by inhibiting bacterioplankton assimilation of EOC with streptomycin and kanamycin. Bacterioplankton production (3H-thymidine incorporation) and metabolism (14C-glucose incorporation) were monitored in samples collected from the Potomac River estuary to determine the effect of the antibiotics. Particulate (i.e., raw water) primary production and EOC (i.e., water passing through 1.0 μm glass fiber filter) production rates were monitored to determine the impact of antibiotics on phytoplankton. In preliminary experiments, neither streptomycin nor kanamycin alone significantly inhibited bacterioplankton activity compared to controls, but when both were present secondary production and metabolism were reduced up to 90%, and remained as such for 45 h. During field evaluations using a streptomycin and kanamycin mixture (50 μM each) particulate primary production and EOC production were not statistically different in control and antibiotic treated samples indicating that the antibiotics did not negatively influence phytoplankton production rates. In the presence of antibiotics dissolved free amino acids (DFAA) and, to a lesser extent, monosaccharides were significantly elevated compared to controls. This study demonstrates that streptomycin and kanamycin are capable of inhibiting bacterioplankton metabolism and uptake of dissolved organic carbon (DOC) in the samples tested so that the contribution of EOC to the DOC pool and to bacterioplankton metabolism could be measured and assessed.  相似文献

The combination of microautoradiography and fluorescence in situ hybridization (MAR-FISH) is a powerful technique for tracking the incorporation of radiolabelled compounds by specific bacterial populations at a single cell resolution. It has been widely applied in aquatic microbial ecology as a tool to unveil key ecophysiological features, shedding light on relevant ecological issues such as bacterial biomass production, the role of different bacterioplankton groups in the global carbon and sulphur cycle, and, at the same time, providing insights into the life styles and niche differentiation of cosmopolitan members of the aquatic microbial communities. Despite its great potential, its application has remained restricted to a few laboratories around the world, in part due to its reputation as a “difficult technique”. Therefore, the objective of this minireview is to highlight the impact of MAR-FISH application on aquatic microbial ecology, and also to provide basic concepts, as well as practical tips, for processing MAR-FISH preparations, thus aiming to contribute to a more widespread application of this powerful method.  相似文献

Relationships between the secondary production of bacterioplankton and the primary production of phytoplankton in the Ezequiel Ramos Mexia Reservoir were examined. Monthly surveys, parallel measurements on consecutive days, and measurements at different times during the same day were undertaken to determine factors that might influence the relationships between the two sets of productivity measurements.The vertical and seasonal distribution of bacterial production appeared to be strongly related to primary production, with temperature as an important component of this trend. The bacterial production in the water column was equivalent to between 2 and 45% of the phytoplankton production over a period of one year.In a diel study, bacterial secondary production rates fluctuated, with maximum values during the night and early morning. There was no temporal coincidence between algal primary production and bacterial secondary production in daily surveys. It was therefore concluded that isolated measurements of productivity reflect events only at the particular time at which they are obtained. The relationship between algae and bacteria is extremely complex and must be carefully evaluated. The nature of the relationship could be underevaluated if the only measurements taken into consideration were made during the daylight period.  相似文献

Environmental factors accountable for bacterioplankton or phytoplankton biomass dominance were analysed in a confined Mediterranean salt marsh (Empordà Wetlands, NE Spain). Two basins located in the same salt marsh, and with differences in size and catchment's area were compared, during four characteristic situations of the hydroperiod. Since bacterio- or phytoplankton relationships may be affected by other factors such as diel variations or vertical differences in nutrient composition and distribution, high frequency fluctuations due to these factors were also taken into account. Differences in catchment area appeared to be the more plausible explanation of differences in nutrient and organic carbon accumulation among basins, since during confinement basins essentially accumulate the allochthonous nutrient and organic matter supplies that previously entered by runoff. DOC (Dissolved Organic Carbon) favoured the bacterioplankton biomass increase, but also was the main variable significantly affecting phytoplankton biomass. Basins showed marked differences in bacterio- and phytoplankton dominances. Relationships between phytoplankton and bacterioplankton were positive, negative or not significant, depending on the basin and on the period of the year. The phytoplankton mixotrophic capabilities, both phagotrophy and osmotrophy, and their production of UV-screening compounds, as sunscreen, may explain the significant correlation between DOC and phytoplankton biomass, and the significant effect of phytoplankton on bacterioplankton found in these ecosystems.  相似文献

Ingestion of fluorescent particles by natural protozoan assemblage was studied in the Řimov Reservoir (Southern Bohemia) from April to October, 1987. Attached and free-living bacterial abundance, proportion of active bacteria, density of suspended particles and biomass of cladocerans were also monitored. Heterotrophic nanoflagellates (HNF; 5–12.8 102ml−1) were the dominant bacterial micrograzers during the spring period and consumed 3 to 9% of the total bacteria per day. After the spring phytoplankton bloom maximum densities of suspended particles and attached bacteria (up to 28% of the total counts) were found. Development of cladocerans in May sharply decreased the proportion of attached bacteria and kept them below 5% of the total counts. All the studied components of plankton except Cladocera decreased during the clearwater phase. The most significant drop was observed in the numbers of protozoans, and they were negligible for bacterial elimination. Bacterial losses during that time apparently were due to cladoceran grazing. During the summer period, ciliates (15–142 ml−1) were mostly dominant micrograzers, and protozoan community grazing increased up to 21% of bacterial standing stock per day. The study on the respiration and production of bacterioplankton in five shrimp cultural enclosure ecosystems showed that the production and respiration fluctuated from 90 to 909 and 248 to 1785 μgC· L-1. d-l, respec-tively. There existed a significant positive relationship between production and respiration. The daily P/B ratio of bacterioplancton averaged 0.93 d-l, and its production efficiency averaged 0.34.  相似文献

 In a first synoptic evaluation, the temporal and spatial distribution of bacterioplankton and chlorophyll-a were determined in the German Wadden Sea. Three surveys were undertaken in winter, spring, and summer of 1994 using up to eight ships simultaneously between the river Ems and Sylt island. Despite intensive hydrodynamic mixing of the Wadden Sea water, spatial gradients were obvious. The abundance of bacterioplankton ranged from 0.4 to 26×105 ml–1 and chlorophyll-a varied between <0.1 and 79 μg l–1. In winter, relatively homogeneous distribution patterns of both parameters with small gradients were found. Highest chlorophyll-a values connected with a highly patchy structure were observed in spring, while in summer both total chlorophyll-a values and the complexity of the distribution pattern had decreased. In contrast, bacterial numbers increased steadily from January to July with the highest bacterial densities and greatest patchiness observed in summer. Moreover, in some regions of the Wadden Sea, a trophic succession of algae as carbon producers and bacteria as consumers was evident. Correlation analysis verified the relationship between bacteria and chlorophyll a, indicating bottom-up control of bacterial abundance in the northern part of the German Wadden Sea. Since the observed regression slope is remarkably low (0.12–0.46) compared to literature values (0.5–0.8), we suggest that the link between phytoplankton and bacteria found here is a special characteristic of the Wadden Sea as a transition zone between the coastal region and the outer North Sea. Four surveys were carried out in August 2001, and in August, September and October 2002 in the southern Yellow Sea, China. Bacterioplankton abundance and biomass were quantified along the transects from Qingdao, China to the Korean island of Jeju. In this paper, the author describes the distribution of bacterioplankton and predatory pressures, and also discusses the impact of Yellow Sea Cold Water Mass (YSCWM) on the distribution of bacterioplankton. The range of Synechococcus biomass was 0.78–33.49 mg C/m3 (on average 6.26 mg C/m3), and heterotrophic bacteria biomass was 1.58–21.25 mg C/m3 (on average 5.79 mg C/m3, n=197). In the vertical direction, the distribution order of Synechococcus biomass was mid-layer>surface>bottom, and the one of heterotrophic bacteria biomass was surface>mid-layer>bottom. The contribution of Synechococcus to the total phytoplankton biomass (SB/PB) ranged from 2% to 99% (on average 42.5%), and the ratio of heterotrophic bacterial biomass to phytoplankton biomass (HBB/PB) was from 5% to 637% (on average 85%). Daily variations of bacterioplankton biomass at anchor stations showed that the maximal Synechococcus biomass was 8.8 times the minimum, and it was 2.8 times in heterotrophic bacterial biomass. The distribution of bacterioplankton accorded with temperature and salinity, and the minimum of bacterioplankton biomass occurred in the YSCWM waters. At the same time, the microzooplankton (20–200 μm) preyed on Synechococcus in these waters, with the ingestion rate of about 0.20–0.42/d.  相似文献

【目的】研究不同浓度石油污染对海水中浮游细菌群落结构及多样性的影响,并尝试阐述造成这种影响的机理。【方法】将采集自海上钻井平台附近的表层海水样品,在实验室条件下用不同浓度的石油(0?10 g/L)处理1周后,用末端限制性片段长度多态性(TRFLP)方法检测其细菌群落结构及多样性变化情况。【结果】得到了一些与以往研究结果不同的新发现,如细菌多样性不是简单地随石油浓度的升高而降低,而是呈现先降低再升高最后又降低的趋势。0.1 g/L石油处理组(M0.1)与0.5 g/L处理组(M0.5)、2.5 g/L石油处理组(M2.5)和10 g/L处理组(M10)的细菌群落结构更相似,且添加石油处理组(M0.1、M0.5、M2.5、M10)与未添加石油的对照组(M0)间细菌群落结构相差较大。与石油降解相关的细菌主要集中于变形菌门(Proteobacteria)、厚壁菌门(Firmicutes)和拟杆菌门(Bacteroidetes)。基于TRFs在不同处理组的相对比例,可将实验得到的52个TRFs划分为6种类型:低浓度(I)、中浓度(II)、高浓度(III)、广浓度(IV)、窄浓度(V)石油适应型和石油敏感型(VI)细菌。另外,首次提出“碳(能)源-毒素”假说来解释石油污染对浮游细菌群落的影响。【结论】石油污染对浮游细菌群落的影响与石油的浓度和海水中原有的历史细菌群落密切相关,可以按照浮游细菌对石油的适应性对其进行分类,并且“碳(能)源-毒素”假说能较好地解释石油污染对浮游细菌群落的影响。  相似文献

Protozoan bacterivory [via uptake of fluorescently labelledbacteria (FLB)] and production of bacteria ([3H]thymidine assay)were simultaneously measured in the mesotrophic ímovReservoir (Southern Bohemia) from April to November, 1988. Heterotrophicnanoflagellates (HNF) were mostly responsible for a greaterfraction of protozoan bacterivory during the spring period.From 10 to 23% of bacterial production was grazed daily withthe only exception of the spring peak of ciliate abundance (upto 60%). Protozoans decreased significantly during the clearwaterphase (ciliates disappeared), and thus their role in bacterivorywas negligible. Through the summer-fall period ciliates, notHNF, were the most important bacterial micrograzers. Protozoancommunity grazing balanced or even exceeded the daily bacterialproduction in August and September. Seasonal and spatial variations in bacterial abundance, biomass and production in a recently flooded reservoir were followed for 2 consecutive years, in conjunction with phytoplankton biomass (chlorophyll a) and activity (primary production). Between the 2 years of the study, the mean value of primary production remained constant, while those of the chlorophyll a concentration, bacterial abundance (BA), bacterial biomass (BB) and bacterial production (BP) decreased. The observed trends of the bacterial variables were linked to changes in the relative importance of allochthonous dissolved organic matter. Moreover, this factor would explain discrepancies observed between the slope of the model II regression equations established from results of the present study and those of the predictive models from the literature, relating to bacterial and phytoplankton variables. An estimate of the carbon budget indicated that 22 and 5% of the ambient primary production in the Sep Reservoir might be channeled through the microbial loop via BP during the 1st and 2nd year of the study, respectively. We conclude that heterotrophic BP in the Sep Reservoir may, on occasion, represent a significant source of carbon for higher order consumers.  相似文献

1. Production of heterotrophic bacterioplankton was estimated monthly by the tritiated thymidine and leucine incorporation methods during the draining and filling of the mesotrophic Lake Pareloup (over a 2.5-years sampling program).
2. Rates of 3H-leucine (leu) and 3H-thymidine (thy DNA) incorporation generally paralleled each other but the ratio of leu/thy DNA incorporation rates was higher for the draining period (34.5 mean) than during and after filling (11.5 mean).
3. After draining, the highest ratios were observed during periods of low temperature and low bacterial specific activity, while DNA labeling by 3H-thymidine was reduced. However, bacterial production estimates obtained by 3H-leucine (BPL) and 3H-thymidine (BPT) incorporation methods were generally well correlated and the average BPL/BPT ratio was equal to 0.78.
4. In addition, both methods were applied during a diel cycle in three lakes of different trophic status. An increase of leu/thy DNA incorporation rates was noted from the oligotrophic to the eutrophic system. In the absence of Cyanobacteria, BPL and BPT values were quite concordant on average.
5. In situations of unbalanced growth, BPL and BPT values can diverge but when considered over a sufficient period of time they were found to be in agreement.  相似文献

Spatial distribution of planktonic viral particles (virioplankton) and mortality of heterotrophic bacteria caused by viral lysis were studied in the eutrophic Ivan’kovskoe and mesotrophic Uglichskoe reservoirs (the Upper Volga). During the summer peak of phytoplankton, the number of viral particles was higher in the Ivan’kovskoe Reservoir ((55.1 ± 9.5) × 106 ml−1 on average) than in the Uglichskoe Reservoir ((42.9 ± 5.1) × 106 ml−1 on average). The ratio of viral to bacterial abundances ranged from 2.5 to 7.0. The average number of mature phages in infected heterotrophic bacteria varied from 17 to 109 particles/cell. Most of the infected bacterial cells in the Ivan’kovskoe Reservoir were rod-shaped, and, in the Uglichskoe Reservoir, they were mainly vibrio-shaped. In the Ivan’kovskoe Reservoir, from 8.3 to 22.4% of planktonic bacteria were infected by phages, suggesting phage-induced mortality of bacterioplankton equal to 10.5–34.8% (19.1% on average) of the daily bacterial production. In the Uglichskoe Reservoir, from 9.4 to 33.5% of bacteria were phage-infected, suggesting phage-induced bacterial mortality of 13.7–40.2% (23.5% on average) of the daily bacterial production. The obtained results testify to an important role of autochthonous viruses in the regulation of bacterioplankton abundance and production in the reservoirs.  相似文献

Abstract Size-selective grazing by Cyclidium sp., isolated as a dominant ciliate bacterivore from the Římov Reservoir (South Bohemia), was examined using fluorescent labelled bacteria (FLB) produced from natural bacterioplankton or pure bacterial cultures. Sizes of ingested bacteria in food vacuoles were measured directly. Three experimental arrangements were used: (1) Ciliates were grown on the pure culture of Alcaligenes xylosoxidans and fed with various proportions of ‘large’ and ‘small’ FLB (mean biovolume, 0.377 and 0.202 μm3, respectively) prepared from the same bacterial species. Results clearly showed significant selection of larger bacteria. (2) Ciliates were grown on natural bacterioplankton from the reservoir and subsequently fed on FLB prepared from the reservoir bacterioplankton (mean biovolume, 0.065 μm3). Independent of either prey or predator abundance, larger FLB (> 0.100 μ m3, and especially those > 0.200 μ m3) were ingested with much higher frequency than their occurrence i the natural assemblage. (3) Ciliates were grown on the reservoir baterioplankton and fed by FLB prepared from the culture of Pseudomonas sp. In contrast with previous results, no size selection of the ciliate was found when FLB were different from the bacterial food used to grow the ciliate. Ecological impacts of size-selective bacterivory are suggested.  相似文献

The goal of this study was to address the top-down and bottom-up controls on different microbial web components (bacterioplankton, picophytoplankton, and >3 μm phytoplankton) in an Antarctic lake. Two experiments using a size fractionation approach and nutrient addition were conducted at microcosm scale (2.5 l). The variation in net growth rates (k′) of bacterioplankton and phytoplankton size fractions was analyzed after 5 days. The results determined significant differences; whereas bacterioplankton and large phytoplankton showed an increase in their k′ when their predators were removed, the picophytoplankton showed a decrease. All the studied plankton components presented the highest k′ when nutrients were added. It is suggested that, in this lake, both the top-down and bottom-up regulations account for the regulation of bacterioplankton and large phytoplankton. As for picophytoplankton, the bottom-up control was evident and grazing did not pose a negative impact and rather, had a positive effect probably due to liberation of nutrients.  相似文献

1. A year-round study was conducted in a mesotrophic reservoir to determine the dynamics of zooplankton populations as a function of food availability (edible phytoplankton), nutrient concentration, temperature and hydraulic regime.
2. Rotifer biomass was correlated with soluble reactive phosphorus (SRP) concentration. The abundance of the rotifers Keratella cochlearis and Anuraeopsis fissa were not correlated with food availability (measured by chlorophyll and cell counts) but showed a strong dependence on P availability. Another rotifer, Synchaeta oblonga , and crustacean species were not related to nutrient availability but seemed to be dependent on food concentrations, especially of some phytoplankton taxa.
3. In this field study, rotifers seemed more susceptible than Daphnia or copepods to P-limitation. Among rotifer species, Keratella seemed to be more susceptible than Anuraeopsis to P limitation. Different susceptibilities of zooplankton species to nutrient limitation may be important in explaining the dynamics of these organisms in natural situations. Further analyses are warranted to clarify the interactions between nutrient limitation and energy limitation among zooplankton.  相似文献
