The surfaces of the pericarps of wheat and barley grains, 15–30days after anthesis, were examined. Stomata were found in thepericarp epidermis on the ventral side at the apical end inone variety of wheat and four varieties of barley. Layers whichstained red with Sudan IV were observed on the pericarp epidermisand on either side of the testa in immature barley grains. Theultrastructure of these layers was investigated. It was concludedthat the cuticular layer inside the testa is derived from thenucellus. The significance of these cuticular layers in relationto the supply of carbon dioxide to the photosynthesizing cellsof the pericarp is discussed. The embryo envelope tissues in both mature dry seed and duringearly germination of Phacelia tanacetifolia were investigatedby bright-field and fluorescence light microscopy and scanningelectron microscopy. The ruminate seed had an irregularly reticulatesurface owing to the presence of polygonal areas, correspondingto the cells of the seed coat. The raised margins of these cellsjoined at the lobe tips, where radially arranged thickeningsoccurred. The unitegmic seed coat was made up of three distinctlayers: the frayed outer layer, the middle layer with portionsrising outwards to form the radial thickenings, and the innerlayer, the thickness of which was greatest in the micropylarzone. The endosperm tissue had two regions, the micropylar andthe lateral endosperm, which differed in polysaccharide composition,thickness and metachromasy intensity, and presence (in the lateralendosperm) or absence (in the micropylar endosperm) of birefringenceof the cell walls. Moreover, in the micropylar region, wherethe embryo suspensor remnant was found, Ca-oxalate crystalswere scarce or absent. The presence of a partially permeablecuticle covering the seed endosperm was observed. Incubationof seeds in Lucifer Yellow CH indicated that water was ableto penetrate quickly into the seed coat along the pathway formedby the radial thickenings, the raised margins of the polygonalcells and the middle layer. Afterwards, LY-CH readily infiltratedthe apical portions of the seed lobes and then the whole endosperm.Following imbibition, morphological changes were found in themicropylar endosperm, such as the initial digestion of proteinbodies. In addition, both in the seed coat and in the endosperm,a weaker fluorescence, probably due to leaching of polyphenolicsubstances, was observed. Once the seed coat was broken at themicropylar end of the seed, the endosperm cap surrounding theradicle tip had to be punctured by it so that complete germinationcould occur. The seeds of Cuscuta pedicellata have been investigated by transmissionand scanning electron microscopy. Additional observations havebeen made on seeds of C. campestris by SEM only. The seed coatconsists of an outer single epidermis, two different palisadelayers, and an inner multiparenchyma layer. The outer epidermalwall in C. pedicellata has a thick cuticle and zones rich inpectic substances. The thicker ‘U-shaped’ cell wallsin the outer palisade layer are strengthened by a wall layerof hemicellulose. The inner palisade layer has thick walledcells with a ‘light line’. The inner cell wall ofthe compressed multiparenchyma layer has a thin cuticle. A fairlythick cuticle is positioned directly on the endosperm surface.The aleurone cell walls are different from the remaining endospermwalls. The latter are thick and believed to be of galactomannans.There is a ‘clear’ zone between the plasmalemmaand the cell wall in the aleurone cells. The embryo cells arepacked with lipids and proteins. In Cuscuta campestris mostendosperm has been absorbed during the seed development. Theembryo apex has two minute leaf primordia. The features of theCuscuta seeds are discussed in relation to functional and environmentalconditions. The ultrastructure of the nectary spur of Limodorum abortivum(L) Sw. was examined before and after anthesis. In cross sectionthe nectary spur shows an internal epidermal layer of thin-walledcells bordering the secretory cavity and 10–12 layersof parenchyma cells. The ultrastructure of the secretory cellssuggests the involvement of ER, Golgi and plastids in nectarsecretion. The nectar accumulated in the sub-cuticular spaceis released into the nectariferous cavity by rupture of theouter layer of the cuticle. The growth of the fruit of two varieties of almond (Prunus dulcis(Mill.) D. A. Webb) was studid from anthesis (week 0) to maturity(week 32). The dimensions, fresh weight, moisture content, anatomyand chemical composition of the pericarp, testa, embryo, endospermand nucellus are recorded diagrarnmatically, graphically andby micrographs for one variety. Of the two ovules present atflowering only one normally developed further. By 12 weeks afterflowering the whole fruit had reached full size. The space encloscdby the pericarp was filled by nuallus until weck 10, with subsequentenlargement of both endosperm and embryo. From week 16 to week20 the embryo increased to full size with a concumnt decreasein the size of the endosperm. Sixteen weeks after flowering,the embryo began to accumulate protein and lipid, little ofwhich originated from either the nucellus or endosperm. Theembryo contained no starch or reducing sugar but up to 3% sucrosein the early stags which dtcreascd as lipid and protein increased.Starch and sucrose levels were high in the testa at week 16but subsbquently dropped, starch more rapidly than sucrose.The role of the testa in transport of metabolites to the embryois discussed. In a previous paper, an account was given of the ultrastructuralfeatures of the testa and outer cotyledon layers of the cashewnut. Reference was also made to the effect of industrial processingon these structural features. The object of industrial processingis to remove the testa. However, processing is not completelysuccessful and the present paper describes the ultrastructuralfeatures of the areas of the nut where the testa was difficultto remove. The cotyledon cuticle and cuticular layer, also thetesta lining complex, were absent in these areas and severalnew features, including lacunae, were observed.  相似文献

Emmenanthe penduliflora is an obligate fire-recruiter and demonstrates a prolonged seed dormancy followed by germination closely cued to the immediate post-fire environment. This study investigated, at the ultrastructure level, the causal factor(s) associated with seed dormancy and the stimulation of germination after exposure to smoke. The seed coat was responsible for the proximal regulation of dormancy - a waxy cuticular layer situated between the testa and endosperm was the primary barrier to the diffusion of water and small diameter solutes. The sub-testa cuticle in dormant seeds was partially permeable, as indicated by the presence of permeate channels. A short exposure to dry smoke (3 min) promoted a significant increase in seed germination (dormant 80.3% smoke-treated 793%). Exposure to smoke also resulted in two major changes to the morphology of the seed. First, smoke treatment produced an intense chemical scarification at the seed surface; the external cuticle was plasticized to form numerous small spheres on the seed surface. Second, smoke significantly altered the permeability of the internal (sub-testa) cuticle. A significant increase in both the number and size of permeate channels in the sub-testa cuticle indicated that these modifications were directly associated with the breaking of seed dormancy. The observed changes at both the internal (sub-testa) and external cuticles are consistent with the hypothesis that volatiles in smoke exert a surfactant-like reaction to break seed dormancy in E. penduliflora.  相似文献

Fusarium oxysporumf. sp.orthoceras, a potential agent for biologicalcontrol of the root-parasitic weedOrobanche cumanain sunflower,attacks underground developmental stages, such as shoots, tuberclesand seed germ tubes. The germination of inoculated seed wassignificantly reduced, and cytological investigation showedthatF. oxysporumhad penetrated and colonized dormant seeds ofO.cumana. Germ tubes of microconidia penetrated all parts of thethick, complex seed testa, and seed contents were completelydestroyed. Hyphae dissolved the endosperm cell walls and metabolizedthe cytoplasm which was rich in lipid- and protein-bodies. Seed size and shape, seed coat surface pattern, seed coat thickness, and endosperm structure were investigated in Androsace septentrionalis, Cortusa matthioli, Hottonia palustris, Primula elatior, Soldanella carpatica (Primulaceae), Anagallis arvensis, A. minima, Cyclamen purpurascens, Glaux maritima, Lysimachia nemorum, L. vulgaris, Trientalis europaea (Myrsinaceae), and Samolus valerandi (Theophrastaceae). Three seed size categories were distinguished on the basis of biometric measurements. Almost all seeds examined were found to be small with an angular shape classified as sectoroid or polyhedral. A new type of seed shape, suboval, was identified for H. palustris. Cyclamen purpurascens seeds differed from seeds of all other species examined because of their large size, subglobose shape, and concave hilar area. Despite the different shape types, the length/width ratio of the seeds examined was constant, while their hilum length/width ratio was a highly variable feature. Three types of seed surface patterns were observed: (1) reticulate, (2) tuberculate with secondary striations, and (3) poroid-alveolate with the presence of a spongy outer layer. For seeds of Anagallis arvensis, A. minima, Cortusa matthioli, Lysimachia nemorum, and Soldanella carpatica, secondary seed sculpture was described. The seed coats of all species examined were two-layered, and great differences in testa thickness were found (9.9?C128.6???m). In general, seeds of the Myrsinaceae species were more often characterized by thick testa. Different proportions in thickness of the inner and outer testa layers were observed. In seeds with reticulate seed patterns, the inner testa layer was twice to several times thicker than the outer layer, while the opposite proportions were observed in seeds with the tuberculate or poroid-alveolate seed sculpture pattern. In seeds of all species examined, oxalate crystals were present on the surface of the inner testa layer. Thus, the presence or absence of oxalate crystals in testa is not a feature distinguishing species with angular or subglobose seeds. Four types of endosperm structure were identified according to the thickness of the endosperm cell walls and the relief of their inner surface: (1) with evenly thickened and smooth cell walls, (2) with evenly thickened cell walls and circular or helical thickenings on their inside surfaces, (3) with very thick, but not evenly thickened cell walls with constrictions (??pitted??), and (4) with very thin papery and undulate cell walls. There is no rule concerning the seed shape, type of the seed sculpture, testa thickness, or endosperm structure that corresponds to the family affiliation of the species examined.  相似文献

Developmental studies were made on the dewaxed thin-sectionedapple fruit cuticles of 10 Malus sylvestris Mill, cultivarsfrom 4 weeks before anthesis through fruit maturation and harvest.Cuticular development appears to correlate well with the generalgrowth of the fruit. However, no correlation exists betweenfruit size and cuticle thickness. Cuticular pores were evidentas early as 1 week before anthesis and transcuticular canalsbecame evident by 1 week following anthesis. Dewaxed thin-sectioned,as well as isolated, mature fruit cuticles of 16 cultivars andfour crab-apples consistently revealed the distinct presenceof ubiquitous pores and canals. Evidence is provided by lightmicroscopy and scanning electron microscopy (SEM). Measurementswere taken of cuticle thicknesses during development and ofcuticular pore dimensions, and calculations were made of poreand canal numbers. Fruit size alone is not directly indicativeof total pore numbers per surface area. Canal lengths are directlyrelated to the developmental thickness of the cuticle. No correlationwas found between the thickness of the mature cuticle and eitherthe number of pores present or the pore diameters. Endosperm tissue of mature kernels of rye (Secale cereale L.)cv. Dominant was examined by light and transmission electronmicroscopy. It was found that storage protein in sub-aleuronecells occupies up to 35 per cent of the cell volume and formsa continuous matrix in which starch grains and cytoplasmic remnantsare embedded. In the prismatic endosperm, the storage proteinis present as a fine network interspersed between the numeroustype A and B starch grains. Protein bodies are not found inthe prismatic endosperm; only a few, less than 1 µm indiameter, are observed in pockets of disorganized cytoplasmin the sub-aleurone tissue. Cuticle ultrastructure and radiolabelling of isolated cuticlesafter incorporation of [14C] acetate in foliar discs were investigatedwith ivy plants grownin vitrothenex vitro. Results show an increasein thickness, mass and wax content, between young and expandedleaves, for bothin vitroandex vitrocuticles. The cuticle ofinvitrounexpanded leaves was very thin and only constituted alamellate zone. The ultrastructure ofin vitroyoung and expandedleaf cuticles showed characteristics similar toin situcuticles.The thickness of the lamellate zone remained fairly constantand represented 33% of the cuticle thickness in young leaves,but only 11.4% in expanded leaves. The number of lamellar unitsdecreased from 14 to nine between these two growth stages. Themain difference between young leaves developedin vitroorex vitrowasa thinner lamellate zone forex vitrocuticles. However, theselatter cuticles had an intermediary zone between the lamellateand reticulate zones. The cuticle thickness of expanded leaveswas greater forin vitrocuticles suggesting a temporary decreasein cuticle biosynthesis after transfer of the plant fromin vitrotoexvitro.Results from cuticle radiolabelling show higher radioactivityincorporation in cuticles isolated from leaves developedex vitrocomparedtoin vitro. This radiolabelling was particularly marked forexvitroyoung leaf cuticles and depended on the duration of theexvitrogrowth period revealing a progressive activation of cuticlebiosynthesis in response to new environmental conditions. The anatomy of the embryo, endosperm, and testa throughout thedevelopment of charlock seeds is described. In mature seedsthe embryos are morphologically fully differentiated. Each embryoconsists of a shoot meristem with two large cotyledons but noleaf primordia, and a root meristem with a root cap. The embryois surrounded by a single layer of aleurone-like cells derivedfrom the endosperm and enclosed within the testa. Mucilagesand phenols which could greatly retard the diffusion of oxygeninto the tissues of the embryo are found in the testa. Dormancy,which appears to be induced by a shortage of oxygen, is probablyassociated with the presence of these covering layers.  相似文献

Changes in the ultrastructure of the epidermis and cuticle ofCorsican pine (Pinus nigra var. maritima (Aiton) Melville) inrelation to infection of the needle by Lophodermella sulcigena(Rostr.) v Hohn were recorded throughout a growing season. Epidermalcells when young had thin walls and a thin smooth cuticle: bothstructures thickened with age, and the needle surface becamerough. Densely packed wax tubes developed around the stomatalpore and later became scattered over the needle surface. Themicro-flora present on the needles is briefly noted. Anatomical features of seeds of Prosopis farcata (Banks & Solander) Eig, collected at nine localities from various geographical regions and habitats in Israel, were studied. The seeds differ in size, shape and colour, and also in the cracking ability of the surface layer and whether the palisade layer peels off from the rest of the testa upon imbibition or not.
The gum extruded during imbibition is formed in gum cavities extending from the endosperm throughout the testa up to the palisades.
Water penetration is apparently barred by the outermost wall layers of the external cells of the testa (palisade caps).  相似文献

Metabolite deposition during seed development was examined histochemicallyin Trifolium repens by light- and fluorescence microscopy. Allendosperm haustorium at the chalazal pole of the embryo sacand wall protrusions in cell walls of the suspensor and theembryo sac suggest that transfer of metabolites from maternalto offspring tissue takes place primarily at these sites. Thisis further supported by prominent cutinization of the interpolarregion of the embryo sac wall, accumulation of starch in integumentaltissue at the embryo sac poles, and breakdown of interpolarendothelial cells. Decomposition of osteosclereid starch isfollowed by accumulation in the cellular endosperm and subsequentlyin the embryo parallel to endosperm degradation. The starchaccumulates gradually inward from the subepidermal cells ofthe embryo to the stele. Protein bodies are formed in the vacuolesalong the tonoplast, later to be cut off in vesicles releasedinto the cytoplasm. At maturity the embryo is packed with proteinand starch, but without lipid reserves. Phytin is observed inthe protein bodies. 为探明种皮和胚乳是否是限制桃儿七种子萌发的主要因素,利用组织切片和显微技术,对桃儿七种子及其不同萌发期(1、7、14、21、28 d)解剖结构和播种后一定时期内(7~210 d)的植株生长形态进行观察。桃儿七种子由种皮、胚乳和胚构成。种皮包括外种皮和内种皮,外种皮致密规整,由外至内分别为栅状石细胞和表皮层细胞,内种皮由5~6层海绵细胞组成。胚乳占种子体积的绝大部分,包括珠孔胚乳和外胚乳。胚由胚根、胚轴和子叶组成,被致密种皮、多层珠孔胚乳和外胚乳包围。萌发期1~7 d胚根和胚轴开始伸长,7~14 d两片子叶分离,14~21 d胚根突破珠孔胚乳和种皮,21~28 d胚根、胚轴和子叶继续扩张伸长。种子播种210 d后可平均形成3片功能真叶和5条不定根。致密种皮(物理休眠)和多层胚乳(机械休眠)是限制桃儿七种子萌发的两个主要因素。  相似文献

The pistil of Hypericum calycinum has a pentacarpellary, syncarpousovary with five slender styles, each terminating in a smallstigma. The stigma is dry and papillate with a thin lining ofpellicle. The cuticle is thin and continuous around the papillae.A large vacuole filled with tannins occupies the major partof the papillae and the cytoplasm forms a thin lining aroundthe vacuole. The cell wall of the mature papillae show two distinctlayers - an outer layer of loosely woven fibrils and an innerdenser layer with compact fibrils. A large number of small lipoidalbodies accumulate just below the cuticle. The papillae havefewer organelles than those typical of glandular cells. Dictyosomesobserved occasionally are without associated vesicles. The cytoplasmis rich in ribosomes. The basal portions of the papillae mergeinto the transmitting tissue made up of loosely arranged cells.The intercellular matrix of the transmitting tissue is richin lipids. The testa structure and histochemistry of Leucaena leucocephalaLam. (De Wit) seed were investigated by bright-field and fluorescencelight microscopy, and scanning electron microscopy. The testaconsisted of several separate layers. Externally there was anon-cellular layer made up of two parts differing in histochemicalcharacteristics: an "outer strip" rich in hydrophilic substances,and a "thicker part" showing the occurrence of phenolics, involvedin water impermeability control. A second underlying thin layerwas formed by the palisade cell caps, joined one to another.This layer, which we called the "cap film ", became metachromaticreddish-blue with Toluidine O pH 4·4 and reacted positivelywith Alcian blue pH 2·5, revealing the presence of polysaccharidehydrophilic material. The palisade cells consisted of two parts,having different structures and histochemical features. Thefurrowed upper part revealed the simultaneous presence of hydrophilicand hydrophobic substances in the furrows and in the ribs respectively.The corresponding periclinal section showed a central daisy-likepattern (made up of hydrophilic material) with a small chainof tiny spots (made up of hydrophobic material) all around it.The inner part of the palisade cells was also furrowed, butshowed only hydrophobic substances of lipidic nature, detectedby Auramine O and Phosphine 3R. The light line was found tobe rich in callose as shown by the strong fluorescence inducedby Aniline Blue. Studies were carried out on structural and cytochemical aspectsof the stigma and style ofVitis vinifera . The stigma is ofthe wet papillate type with a continuous cuticle and pellicle.During the development of the papillae, the cell walls increasein thickness and produce a secretion product constituted oflipids that pass through the wall forming the exudate. The styleis solid with a central core of transmitting tissue which hasconspicuous intercellular spaces that increase remarkably fromthe periphery to the centre where the cuticle is present. Theintercellular spaces, where the pollen tubes grow, contain amatrix that includes polysaccharides, pectic substances andscattered areas of lipidic nature.
