This experiment was conducted to evaluate the effect of administering increasing doses of royal jelly (RJ) on reproductive parameters in ewes. Additionally, this study compared using RJ vs. equine chorionic gonadotropin (eCG) in estrous cycle control. In May (transitional period between anestrous and the breeding season) 37 multiparous, winter-lambing Awassi ewes 3-6 years of age (average body weight of 53+/-1.2 kg) were fitted with intravaginal flourogestone acetate-impregnated sponges (FGA, 40 mg) for 12 days. Ewes were randomly assigned into five treatment groups to receive no RJ (CON, n=7), 250 mg RJ/d (RJ250, n=8), 500 mg RJ/d (RJ500, n=8), 750 mg RJ/d (RJ750, n=7), or 600IU eCG (eCG, n=7). Royal jelly was administrated orally on daily basis when sponges were in place while eCG was administered on the day of sponge withdrawal (d 0). Behavioral estrus was checked using fertile Awassi rams at 6h intervals for 5 days beginning on d 1. Interval from d 0 to onset of estrus was shorter (P<0.05) in eCG than in CON and RJ250 groups. No differences in the onset of estrus were detected among the RJ-treated groups. The intervals from d 0 to first progesterone rise were shorter (P<0.05) in the eCG-treated compared with RJ-treated and control ewes (100+/-15.3, 138.4+/-14, 135.7+/-15, 155.6+/-15, 154.4+/-15.1h in eCG, CON, RJ250, RJ500, and RJ750, groups, respectively). The overall pregnancy rate from mating at induced estrus was 75.7% (28/37). Of these ewes, 23/37 (64.8%) lambed within 155 days following d 0. Lambing rate was higher (P<0.05) in the RJ500 group compared with controls. Lambing rate from mating at induced estrus was 2/7 (28.5%), 4/8 (50%), 8/8 (100%), 4/7 (57%), and 5/7 (71%) in CON, RJ250, RJ500, RJ750, and eCG groups, respectively. Results of the present study demonstrate that eCG but not RJ was effective in improving estrus expression in ewes during the transition between the non-breeding and breeding seasons. Royal jelly may be effective in improving pregnancy and lambing rates but further studies are required to confirm such findings.  相似文献

The objective was to compare the effects of royal jelly (RJ) and eCG treatments on reproductive performance of ewes synchronized using intravaginal progesterone-releasing devices. Forty-two cycling Awassi ewes were treated intramuscularly (i.m.) with 15 mg PGF2alpha. On the following day, all ewes were administered with CIDR-G for 12 days and were randomly allocated to three (RJ, eCG and control) groups of 14 ewes each. Ewes in the RJ-treated group received daily i.m. treatments of 400mg RJ during the period of CIDR-treatment. Each ewe in the eCG-treated group received an i.m. treatment of 500 IU eCG at the time of CIDR-G removal (day 0) and no further treatment was given to ewes in the control group. Ewes were exposed to four fertile rams for 72 h, from the time of CIDR-G removal, and checked for breeding marks at 6-h intervals. Blood samples were collected from day -13 until day 0 and thereafter until day 19 for progesterone analysis. Royal jelly treatment resulted in a greater rate of decline and lower (P<0.02) progesterone concentrations between days -10 and 0 than eCG-treated and control ewes. Expression of estrus was similar among the three groups and intervals to onset of estrus were shorter (P<0.01) in RJ-treated (31.3h) and eCG-treated (29.8h) than control (41.3h) ewes. First-cycle pregnancy and lambing rates were greater (P<0.05) in RJ-treated (71.4 and 71.4%) and eCG-treated (85.7 and 78.6%) than in control (42.9 and 35.7%) ewes, respectively. Results demonstrate that the treatments of RJ and eCG in conjunction with CIDR-G were similarly effective in induction of estrus and improvement of pregnancy and lambing rates.  相似文献

Investigation were carried out to study the norms of progesterone concentration in the blood serum of buffaloes during various phases of oestrous cycle. Twenty four animals (12 heifers and 12 cows) were used. The blood serum samples were stored at -20 degrees C until processed for progesterone assay. The progesterone concentrations were measured by the radioimmunoassay technique. The progesterone levels were 0.360 +/- 0.062 and 0.334 +/- 0.066 ng/ml on the day of oestrus in buffalo-heifers and buffalo-cows, respectively. The values were around 1 ng/ml till day 6, followed by a gradual increase to a peak average value of 4.888 +/- 0.399 and 5.119 +/- 0.415 ng/ml on day 15 of the cycle in heifers and cows, respectively. Thereafter, the progesterone concentration fell abruptly to a level similar to that at oestrus. The mean progesterone value a day before oestrus was 0.488 +/- 0.067 and 0.577 +/- 0.053 ng/ml in buffalo-heifers and buffalo-cows, respectively. The mean progesterone concentration of different days of the cycle (except day 16) did not differ significantly (P / -0.01) between heifers and cows.  相似文献

Two experiments involving 24 and 54 Australian Merino ewes were conducted in which the establishment of a cervical population of spermatozoa and several endocrinological events were studied after several regimens for the synchronization of oestrus. Intravaginal sponges impregnated with 500 mg (Exp. 1) or 200, 400 or 600 mg (Exp. 2) progesterone resulted in the maintenance of plasma progesterone concentrations of 1.5-4.9 ng/ml over a 12-day insertion period compared with 1.9-6.9 ng/ml during dioestrus in control ewes. In Exp. 1 basal concentrations of less than or equal to 0.25 ng/ml plasma were attained by 4 h after sponge withdrawal and this decline was much more rapid than in normal luteolysis. This was associated with fewer spermatozoa recovered from the cervix 2 h after insemination, and PMSG had no significant effect. In Exp. 2 injection of a supplementary dose of progesterone at sponge withdrawal resulted in a rapid increase in plasma progesterone concentrations followed by an equally rapid decrease and an attenuation of the rise in plasma oestradiol-17 beta, the LH surge, and the onset of oestrus. The numbers of spermatozoa recovered 4 h after insemination were not increased, and PMSG had no significant effect. Two factors were significant, namely the dose of progesterone in the sponge (600 mg greater than 400 or 200 mg, P less than 0.05) and stage of oestrus when inseminated (mid- or late oestrus greater than early). The data demonstrated that an adequate dose of progesterone/progestagen incorporated into intravaginal sponges and accurate timing of insemination relative to the LH surge are the most important factors involved in penetration of the cervix by spermatozoa.  相似文献

Ovariectomized ewes were treated with progesterone and oestradiol to induce oestrus (day of expected oestrus = day 0) and with progesterone on days 1 to 12. The concentrations of endometrial oxytocin receptors and the 13,14-dihydro-15-keto prostaglandin F2 alpha (PGFM) response induced by oxytocin were measured on days 12, 14, 16 and 18 after the cessation of progesterone treatment on day 12, by a receptor binding assay and direct radioimmunoassay, respectively. During the period of treatment, the concentrations of plasma progesterone were high and remained above 2 ng ml-1 until day 13 when they dropped rapidly to less than 0.5 ng ml-1 by day 14. The concentrations of oxytocin receptors in endometrium of control ewes were high (820.7 +/- 91.7 (SEM) fmol mg-1 protein). Treatment with progesterone significantly (P < 0.01) reduced the concentrations of the receptors on days 12 and 14 (144.1 +/- 65.0 and 200.4 +/- 45.4 fmol mg-1 protein, respectively). After lambing forty-five ewes were allocated to three groups, two of sixteen and one of thirteen ewes. The lambs of the two groups of sixteen ewes were weaned on Day 1 after lambing and the ewes were fed a diet of 100% (Group H) or 50% (Group R) of maintenance energy requirements. The thirteen ewes in the third group (Group L) suckled twin lambs and were fed freely. During the first 3 weeks after lambing, oestrus was observed for 11/16 (Group H) and 8/16 (Group R) ewes; of the ewes which had shown oestrus in the two groups, ovulation occurred in 5/8 and 5/7 respectively. Only 1/13 Group-L ewes showed oestrus and ovulated during the same period. The mean plasma concentrations of progesterone and LH were unaffected by the treatments and were around 0-4 and 1-5 ng/ml, respectively. Restricted feeding had no effect on oestrus, ovulation or the hormone levels during the oestrus cycle following synchronization. The onset of oestrus and the start of the preovulatory discharge of LH were 3 and 6 hr later, respectively, in the lactating ewes (Group L) than in those in Groups H and R. Ewes in Group L also had a higher ovulation rate, 2-8 +/- 0-2 versus 2-1 +/- 0-2 (P less than 0-05). Restricted feeding reduced the number of ewes lambing; only 1/11 ewes in Group R, considered to have conceived because of the presence of high progesterone levels 17 days after mating, subsequently lambed compared with 6/12 in Group H and 5/9 in Group L.  相似文献

Twin bearing mature ewes (n=40) were treated with exogenous progesterone (100mg daily in oil) or vehicle (oil control) from Day 143 of gestation until lambing to investigate the effects on gestation length, foetal survival and colostrum yield and composition. Compared to control ewes, progesterone treated ewes had increased (P<0.05) serum progesterone concentrations (by 4.3 ng/ml) before lambing and in the first day post-partum (by 10 ng/ml). Progesterone treatment increased gestation length (150.4+/-0.6 days versus 147.8+/-0.6 days, P<0.05) and colostrum yield at 1h after lambing (P<0.05) but the colostrum had a lower concentration of IgG (P=0.02). In the first 24h after lambing, total colostrum and IgG yields were not different between groups. Four (20%) of the progesterone treated ewes produced either one or two dead lambs, while one ewe died on day 155 without initiating the birth process. We conclude that the daily administration of 100mg progesterone resulted in extended gestation length and reduced lamb survival but did not lower colostrum yield.  相似文献

Simultaneous concentrations of endogenous progesterone and exogenous FGA have been measured in ewes treated with FGA-impregnated intravaginal sponges at several times relative to the expected time of release of LH. First, a direct double antibody radioimmunoassay (RIA) for FGA, with good precision, sensitivity and reproducibility, was developed and validated. An oxime derivative was prepared and then conjugated to human serum albumen at the 3-position to produce the antigen. Antibodies raised in New Zealand White rabbits showed little cross-reactivity with related steroids. FGA was estimated in extracted and unextracted plasma; results were indistinguishable. Second, sponges impregnated with 40 mg FGA were inserted into 20 anoestrous crossbred ewes for 12 days; 500 i.u. pregnant mare serum gonadotrophin (PMSG) was injected at withdrawal. Similar sponges were reintroduced into four ewes at each of the intervals 1, 3, 5, and 7 days later; three ewes served as controls. Plasma concentrations of progesterone and FGA were estimated by RIA daily during treatment and at intervals of 2 h for 12 h and at 18 and 24 h after withdrawal. The plasma profiles of FGA during the two successive periods of insertion were remarkably similar. A concentration of 3.0 ng/ml (s.e.m. +/- 0.22) was attained on day 1, falling to 1.5 ng/ml (+/- 0.15) by day 4. Thereafter, the concentration was maintained at 1.1 ng/ml (+/- 0.08). Plasma progesterone concentrations were at basal levels of less than 0.2 ng/ml during the first (acyclic) period of sponge insertion. During the second (cyclic) period there was a marked difference related to the time of sponge insertion. Insertion on day 1 (before LH release) resulted in complete inhibition of luteal activity; insertion on day 3, 5 or 7 was followed by apparently normal luteal function. There was no evidence of any feedback mechanism of exogenous progestagen on endogenous progesterone and no interaction. It is concluded that a 12-day treatment is needed in cyclic ewes for full synchronization and that sponges impregnated with 40 mg FGA will maintain an effective plasma concentration of greater than 1 ng/ml to the end of this period.  相似文献

The objective of this work was to study the effect of the endogenous opiate peptide (EOP) antagonist, naloxone, on the preovulatory LH surge and on the time of onset and duration of oestrus in the ewe with induced oestrus during the non-breeding season. Forty Suffolk X Hampshire ewes 2-3-years-old and 50+/- 4kg were studied, ewes were divided at random in two groups of 20, housed in open paddocks under natural photoperiod (19 degrees latitude N); were fed with hay and commercial pellets, and provided water ad libitum. Group one received an intravaginal sponge with 45mg of medroxiprogesterone acetate for 14 days, and upon sponge withdrawal 250IU of eCG was administered i.m. Group two received the same treatment as group 1 but in addition they received two i.m. injections of 0.5mg of naloxone, one given on sponge withdrawal and the second 24h later (total dose 1.0mg). Oestrus in naloxone-treated ewes was present 32+/- 2h and in control ewes in 35+/- 3h after sponge withdrawal. Duration of oestrus in control ewes was shorter (27+/- 2.5h), than naloxone-treated ewes (39+/- 6h); (P<0.0001). The LH surge in naloxone-treated ewes was initiated 5h after onset of oestrus, and 8h after onset of oestrus in control ewes, and the difference was significative (P<0.0006). It was concluded that EOP are important modulators of reproductive function in the ewe.  相似文献

The effect of exogenous melatonin on prostaglandin secretion was measured on Rasa Aragonesa ewes. Fourteen ewes received an 18 mg melatonin implant (M+) on 10 April and were compared with 13 control animals (without implants M-). Twenty days later, intravaginal pessaries were inserted in all animals to induce a synchronized oestrus (day 0). On day 14, ewes were injected, i.v., with 0.5 IU oxytocin. Plasma 15-ketodihydro-PGF(2alpha) (PGFM) concentrations were measured to assess uterine secretory responsiveness to oxytocin. After euthanasia, pieces of endometrium were collected to determine progesterone content and PGE(2) and PGF(2alpha) secretion in vitro, in the presence or absence of either 20 microg/ml recombinant ovine interferon-tau (roIFNt) or 1 nmol/l oxytocin in the medium. Endometrial progesterone content was similar in the two treatments (M+: 50.25+/-17.34 ng/mg tissue, M-: 43.08+/-11.21 ng/mg tissue). M+ ewes that responded to oxytocin had significantly higher plasma PGFM concentrations between 10 and 80 min after oxytocin administration, a higher mean PGFM peak (P<0.001), higher plasma PGFM levels after the challenge (P<0.05) and higher plasma progesterone concentrations (P<0.01) than control ewes. In the in vitro experiment, M+ and M- control samples secreted similar amounts of PGE(2). The presence of roIFNtau and oxytocin only stimulated PGE(2) production (P<0.05) in M- tissues. Control M+ tissues secreted higher amounts of PGF(2alpha) (P=0.07) and PGF(2alpha) secretion was significantly (P<0.01) stimulated by roIFNtau. Oxytocin produced this effect only in M- samples (P<0.01). In conclusion, although previous studies have demonstrated a positive effect of melatonin on lamb production, PGF(2alpha) secretion is higher in vitro and the PGE(2):PGF(2alpha) ratio is unfavourable in response to IFNtau, which could affect embryo survival. Whether or not these mechanisms are similar in pregnant ewes remains to be elucidated.  相似文献

This study examined the effects of progesterone and intrauterine injection of ovine conceptus secretory proteins (oCSP) on endometrial responsiveness to oxytocin. Twelve ewes were ovariectomized on day 4 of the cycle (oestrus = day 0) and assigned in a 2 x 2 factorial arrangement, to receive either 1.5 mg ovine serum proteins (SP) or oCSP containing 25 micrograms ovine trophoblast protein 1 (oTP-1) (by radioimmunoassay) in 1.5 mg total protein into each uterine horn, via catheters, twice a day on days 11, 12, 13 and 14. Ewes received 200 mg progesterone per day (i.m.) from day 4 to day 10 or 15. Oxytocin-induced prostaglandin F2 alpha was measured as 13,14-dihydro-15-keto-prostaglandin F2 alpha (PGFM) on days 11, 12, 13 and 14 in plasma from three integrated, 10 min (10 ml) blood samples (0-10, 10-20, 20-30 min) obtained after intravenous injection of 20 iu oxytocin, and in a pre-oxytocin (-10 to 0 min) sample collected via an indwelling jugular catheter. The pre-oxytocin samples were also assayed for progesterone. Oxytocin-induced turnover of inositol phosphate was determined in endometrium on day 15 after hysterectomy. In ewes receiving progesterone to day 10, plasma progesterone decreased from about 12 to 2 ng ml-1 (SEM +/- 2.6) during the treatment period (days 11-14), but remained high (12-20 +/- 2.6 ng ml-1) in ewes that received progesterone to day 15. Intrauterine injection of oCSP resulted in high basal concentrations of PGFM on days 12 and 13 compared with SP-treated ewes (P less than 0.01). Treatments with progesterone did not affect basal PGFM concentrations. Treatment with oCSP abolished oxytocin-induced endometrial secretion of prostaglandin only if progesterone was maintained to day 15 (P less than 0.01); in ewes receiving such treatment, oCSP inhibited (P less than 0.01), but SP did not inhibit, oxytocin-induced endometrial turnover of inositol phosphate (P less than 0.06), which was greater in ewes treated with progesterone to day 10 than in those treated to day 15 (P less than 0.05). Ewes that responded to oxytocin with increased PGFM exhibited increased oxytocin-stimulated turnover of inositol phosphate on day 15. These results indicate that the antiluteolytic action oTP-1 exerts on the endometrium requires progesterone and that this mechanism involves inhibition of oxytocin-stimulated turnover of inositol phosphate.  相似文献

The use of either 1 or 3 controlled internal drug release (CIDR) devices for progesterone priming in ewes (n=11) superovulated with 1500 IU pregnant mare serum gonadotrophin (PMSG) at 28 hours prior to CIDR device withdrawal was investigated in relation to the stages of development and viability of the ova produced. Progesterone levels in the ewes (n=6) treated with 3 CIDR devices were significantly higher (P<0.01) during the 11 days of insertion than in those (n=5) treated with 1 CIDR device (7.3 vs 3.3 ng/ml) over the same period. However, following superovulation, the mean (+/-SEM) ovulation rates were similar for both groups (8.2 +/- 1.7 vs 10.2 +/- 1.5). The number of ova (M+/-SEM) recovered by laparoscopy 5 days after insemination was 4.2 +/- 1.0 for ewes treated with 3 CIDR devices and 7.0 +/- 1.1 for those treated with 1 CIDR device (P<0.10). The respective ovum recovery rates (M+/-SEM) were 55+/-9.8 and 74+/-13.2%. There was no effect of progesterone concentration in the priming phase on either the stages of development of the recovered ova or on their ability to develop during in vitro culture. It was concluded, therefore, that progesterone concentrations within the range 3.3 +/- 0.1 to 7.3 +/- 0.3 ng/ml during the priming phase and 2.4 +/- 0.3 to 6.5 +/- 0.2 ng/ml at the time of PMSG administration did not affect the ovulation rate or the viability of ova recovered from superovulated ewes.  相似文献

The hypothesis that, in the ewe, prostaglandin (PG) F2alpha administration on day 3 after ovulation is followed by luteolysis and ovulation was tested using 24 animals. The ewes were treated with a dose of a PGF2alpha analogue (delprostenate, 160 microg) on days 1 (n=8), 3 (n=8) or 5 (n=8) after ovulation, was established by transrectal ultrasonography. Daily scanning and blood sampling were performed to determine ovarian changes and progesterone serum concentrations by radioinmunoassay. The treatment induced a sharp decrease of progesterone concentrations followed by oestrus and ovulation in all ewes treated on days 3 and 5 and in one ewe treated on day 1 (8/8, 8/8, 1/8; P<0.05). Seven ewes treated on day 1 did not respond to PGF2alpha treatment and had an inter-ovulatory cycle of normal length (17.4 +/- 0.5 days). However, the profile of progesterone concentrations during the cycle of these ewes was delayed 1 day (P<0.05) compared with a control cycle. The overall interval between PGF2alpha and oestrus for the 17 responding ewes was 42.4 +/- 2.3 h. In 15 of these ewes the ovulatory follicle was originated from the first follicular wave and the ovulation occurred at 60.8 +/- 1.8 h after PGF2alpha treatment. The other two responding ewes ovulated an ovulatory follicle originated from the second follicular wave between 72 and 96 h after treatment. These results support the hypothesis and suggest that refractoriness to PGF2alpha of the recently formed corpus luteum (CL) may be restricted to the first 1-2 days post-ovulation.  相似文献

Jugular vein blood was collected daily from four mature ewes throughout anoestrus and the first oestrous cycle of the breeding season until 4 days after the second oestrus. The levels of oestrogen, progesterone and LH were determined by radioimmunoassay. There were fluctuations in the LH level throughout most of the observed anoestrous period with a mean plus or minus S.E. value of 2-3 plus or minus 0-9 ng/ml. High LH values of 20-0, 41-2 and 137-5 ng/ml were observed in three ewes on Day - 24 of anoestrus. A brief minor rise in progesterone level was also observed around this period. Progesterone levels were consistently low (0.11 plus or minus 0-01 ng/ml) before Day - 25 of anoestrus. A major rise occurred on Day - 12 of anoestrous and this was followed by patterns similar to those that have been previously reported for the oestrous cycle of the ewe. Random fluctuations of oestrogens deviating from a mean level of 4-40 plus or minus 0-1 pg/ml were observed during anoestrus and the mean level during the period from the first to the second oestrus was 5-2 plus or minus 0-3 pg/ml. A well-defined peak of 13-3 plus or minus 0-7 pg/ml was seen in all ewes on the day of the second oestrus. Results of the present study suggest that episodic releases of LH occur during anoestrus and periods of low luteal activity. The fluctuations in LH levels, as observed during the period of low luteal activity, i.e. before Day - 25 of anoestrus, were less pronounced during the periods of high luteal activity. The view that luteal activity precedes the first behavioural oestrus of the breeding season is supported.  相似文献

A total of 71 synchronized dairy heifers (Holstein Friesian x German Black Pied) were used as recipients of seven-day old frozen/thawed bovine embryos. Plasma progesterone concentrations and corpus luteum quality on the day of nonsurgical transfer (= day 7) were determined and related to pregnancy rates or estrus intervals in nonpregnant recipients. A total of 32 recipients (45.1 %) maintained pregnancy; 39 recipients (54.9 %) did not. No significant differences could be detected between progesterone levels in recipients that remained pregnant (3.14 +/- 0.24 ng/ml; x +/- SEM ) and those that did not maintain pregnancy (3.23 +/- 0.28 ng/ml). Optimal progesterone levels were between 2 and 5 ng/ml coinciding with a pregnancy rate of 51.1 % (24 47 ). Pregnancy rates apparently were decreased when progesterone levels were below 2 ng/ml (35.3 %; 6 17 ) or above 5 ng/ml (28.6 %; 2 7 ). Hence, optimal progesterone levels were identical to those for freshly collected embryos reported previously by Remsen et al. (1). Bovine corpus luteum quality graded by rectal palpation was related to some extent to progesterone levels but not to pregnancy rates. Out of 39 nonpregnant recipients seven animals (17.9 %) with a mean plasma progesterone level of 3.76 +/- 0.72 ng/ml showed an extended estrus interval of more than 55 days, probably indicating early embryonic mortality. Progesterone levels did not significantly differ between nonpregnant recipients with estrus intervals of various length. Plasma progesterone levels at the time of transfer are of limited diagnostic value for screening recipients prior to transfer of frozen/thawed embryos.  相似文献

The breeding season was 157, 154, <126, 210 and 217 days for Rambouillet, Columbia, Suffolk, Rambouillet x Finnish Landrace and Columbia x Finnish Landrace ewes respectively. Treatment of cyclic ewes with pregnant mare serum gonadotropin (PMSG) (500 IU), following a 12-day treatment with progestin-containing intravaginal sponges, did not affect fertility, but did decrease the time from sponge removal to estrus, (control 48.0 +/- 3.1 hr; PMSG 39.4 +/- 1.8 hr) to the preovulatory surge of LH (control 52.7 +/- 2.8 hr; PMSG 39.0 +/- 1.7 hr) and FSH (control 52.3 +/- 2.9 hr; PMSG 42.8 +/- 1.6 hr) and caused an elevation of serum LH levels prior to the preovulatory surge (control 1.25 +/- 0.18 ng/ml; PMSG 2.31 +/- 0.22 ng/ml). Exposure of the purebred ewes to 18 hours of daylight in January, decreasing by 30 minutes a week subsequently, counteracted the seasonal reduction in the number of ewes lambing following induced breeding under natural daylight in May. The pattern of change in plasma progesterone and LH concentrations was monitored in Clun Forest ewes at a natural oestrus and compared to that observed after removal of progesterone implants. The rate of decline in plasma progesterone concentrations after implant withdrawal (1.8 +/- 0.2 ng/ml h-1) was significantly greater (P less than 0.001) than that observed at natural luteolysis (0.2 +/- 0.1 ng/ml h-1), and this resulted in an abnormal pattern of change in tonic LH secretion up to the time of the preovulatory LH surge. This more rapid rate of progesterone removal was also associated with a shortening of the intervals from the time that progesterone concentrations attained basal values to the onset of oestrus (P less than 0.05) and the onset of the preovulatory LH surge (P less than 0.01). However, there were no significant differences in the duration of the LH peak, preovulatory peak LH concentration, ovulation rate or the pattern of progesterone concentrations in the subsequent cycle. It is suggested that the abnormal patterns of change in progesterone and tonic LH concentrations may be one factor involved in the impairment of sperm transport and abnormal patterns of oestradiol secretion known to occur at a synchronized oestrus.  相似文献

These studies test the hypothesis that pregnancy alters the feedback effects of cortisol on stimulated ACTH secretion. Ewes were sham-operated (Sham), or adrenalectomized (ADX) at approximately 108 days gestation and replaced with aldosterone (3 microg x kg(-1) x day(-1)) and with cortisol at either of two doses (ADX + 0.6 and ADX + 1 mg x kg(-1) x day(-1)); ewes were studied during pregnancy and postpartum. Mean cortisol levels produced in ADX ewes were similar to normal pregnant ewes (ADX+1) or nonpregnant ewes (ADX+0.6), respectively. Plasma ACTH concentrations in response to infusion of nitroprusside were significantly increased in the pregnant ADX+0.6 ewes (1,159 +/- 258 pg/ml) relative to pregnant Sham ewes (461 +/- 117 pg/ml) or the ADX+1 ewes (442 +/- 215 pg/ml) or the same ewes postpartum (151 +/- 69 pg/ml). Plasma ACTH concentrations were not significantly different among the groups postpartum. Increasing plasma cortisol to 20-30 ng/ml for 24 h before hypotension produced similar inhibition of ACTH in all groups. Pregnancy appears to decrease the effectiveness of low concentrations of cortisol to inhibit ACTH responses to hypotension.  相似文献

The daily plasma progesterone (P) concentrations achieved during insertion of P (750 mg) sponges into two groups of ewes were examined. Group I received prostaglandin (PG) treatment, which was required to suppress the P production (to levels of < 0.3 ng hormone/ml plasma) from the corpora lutea (CL) of a previous superovulation treatment, following which these Group I ewes and the anestrous Group II ewes were sponge treated. Radioimmunoassay (RIA) and enzymeimmunoassay (EIA) were used to measure the P levels in both groups. Progesterone (750 mg) sponges with and without citric acid impregnation were inserted into all the ewes for 12 days (d). Citric acid lowered the P levels reaching the plasma from the sponges, but it did not mask the characteristic profile (during the treatments) determined by the states of the ewes (single PG and double PG injected, Group I or in the anestrous Group II). The plasma P levels in Group I and II ewes rose to at least 7.0 ng/ml at intervals during treatment. The duration and magnitude of the P concentrations in the plasma were higher in the single PG compared with the double PG ewes during sponge insertion in Group I. The anestrous Group II ewes showed two major peaks (Day 1, P<0.01 and Days 11 to 12, P<0.05) during sponge treatment. A P level > 2.0 ng/ml was maintained over the entire treatment in the single PG and in the anestrous hormone-treated ewes, and was of shorter duration (7 d) in the double PG-treated animals. These endogenous patterns in P profiles of the ewes indicate that the hormone level during sponge insertion varies in magnitude and duration, parameters determined by the physiological/endocrinological state of the ewes at the start of the treatment. The EIA correlated significantly (P<0.001) with the RIA for the measurement of P concentration, when analyzed daily on an individual animal basis.  相似文献

Two experiments were designed to investigate release patterns of oxytocin into plasma during oestrus and the early luteal phase. In Exp. 1, blood samples were collected from 5 ewes every 30 min for 10 h during 6 days around oestrus and the early luteal phase. During oestrus concentrations of oxytocin were generally low (1.27 +/- 0.54 pg/ml; mean +/- s.d.) but with occasional pulses up to 6 pg/ml. By Day 5 mean basal concentrations had risen to 4.5 +/- 2.1 pg/ml with a fluctuating release pattern. In Exp. 2, a method was developed for continuous blood sampling from conscious, unrestrained ewes. On the predicted day of oestrus following an untreated oestrous cycle, 8-ml blood samples were collected every minute for two 35-min periods (8 ewes: 16 sampling periods). For 6 ewes a ram was introduced to the pen for part of this time, and resulting behaviour was recorded. Additional blood samples were assayed for LH and progesterone to determine the stage of the cycle. Overall mean oxytocin concentrations ranged from 1.5 +/- 0.53 to 6.8 +/- 5.25 pg/ml in different animals. Ewes which were both in oestrus and exposed to the ram showed a pulsatile oxytocin release pattern consisting of low baseline concentrations with short-duration pulses superimposed (duration 1-4 min; amplitude 2.5-31.7 pg/ml; frequency 3.18/h). Coitus was not temporally associated with pulsatile release. However, the importance of the presence of the ram was indicated by total separation of 2 oestrous ewes from the ram until after experimentation. In these animals only 1 pulse of oxytocin was detected in 2.7 h of sampling. It is concluded that, although mean oxytocin concentrations at oestrus were low, short duration pulses were released into the plasma at this time.
