The morphological and biological characteristics of ectothermic vertebrates are known to be strongly influenced by environmental conditions, particularly temperature. Epigenetic mechanisms such as DNA methylation have been reported to contribute to the phenotypic plasticity observed in vertebrates in response to environmental changes. Additionally, DNA methylation is a dynamic process that occurs throughout vertebrate ontogeny and it has been associated with the activation and silencing of gene expression during post-embryonic development and metamorphosis. In this study, we investigated genome-wide DNA methylation profiles during turbot metamorphosis, as well as the epigenetic effects of temperature on turbot post-embryonic development. Fish growth and rates of development were greatly affected by rearing temperature. Thus, turbot raised at ambient temperature (18 °C) achieved greater body weights and progressed through development more quickly than those reared at a colder temperature (14 °C). Genome-wide DNA methylation dynamics analyzed via a methylation-sensitive amplified polymorphism (MSAP) technique were not significantly different between animals reared within the two different thermal environments. Furthermore, comparisons between phenotypically similar fish revealed that genome-wide DNA methylation profiles do not necessarily correlate with specific developmental stages in turbot.  相似文献

DNA profiling in forensic casework is based on comparison of the results of biological evidence with direct reference samples of the individual concerned or with indirect references of his close blood relatives. The selection of reference samples for analysis is crucial to the success of a case; it not only depends on the authenticity of the reference samples, but also on the authenticity of the biological relation of the donors with the person in question. There are situations when the social or legal relationship is not the biological one and there is a need to educate investigating officers, forensic analysts, and the judiciary about the associated problems.  相似文献

A high-resolution plotter of differential melting profiles of DNA, RNA, or related biopolymers with an on-line mini-computer is described. With this device, more than 15 transition steps were identified in the thermal melting profile of DNA from bacteriophage lambda. These fine structures were found to be reproducible, and some of them disappear in the deletion mutant. To Examine the melting profile, computer simulations for several hypothetical polynucleotide sequences were performed, and compared with experimental data. The sharp peaks that appeared in the differential melting profile of λ DNA may come from some homogeneous sequences of 500 bases or longer.  相似文献

Ever since methylcytosine was found in genomic DNA, this epigenetic alteration has become a center of scientific attraction, especially because of its relation to gene silencing in disease. There is currently a wide range of methods designed to yield quantitative and qualitative information on genomic DNA methylation. The earliest approaches were concentrated on the study of overall levels of methylcytosine, but more recent efforts havefocused on the study ofthe methylation status of specific DNA sequences. Particularly, optimization of the methods based on bisulfite modification of DNA permits the analysis of limited CpGs in restriction enzyme sites (e.g., combined bisulfite restriction analyses and methylation-sensitive single nucleotide primer extension) and the overall characterization based on differential methylation states (e.g., methylation-specific PCR, MethyLight, and methylation-sensitive single-stranded conformational polymorphism) and allows very specific patterns of methylation to be revealed (bisulfite DNA sequencing). In addition, novel methods designed to search for new methylcytosine hot spots have yielded further data without requiring prior knowledge of the DNA sequence. We hope this review will be a valuable tool in selecting the best techniques to address particular questions concerning the cytosine methylation status of genomic DNA.  相似文献

Invasive alien species (IAS) pose serious threats to native ecosystems worldwide. In some regions, laws and/or lists related to alien species have been made, but their effects on actual measures against alien species have been little studied. In Japan the IAS act, which came into force in 2005, lists many harmful species under the heading “Designated IAS”. The importation, domestication, sale or release of these species is tightly regulated. In addition, other species are named as alerted alien species (AASs) on the “alien species alert list” to raise public awareness. In this paper we examined the situation in Japan as case study that is exposed to threats from alien species under such laws in recent year to evaluate the effects of that for public interesting. We conducted a survey of the effects of the legal designation and other classification of alien plant species, examining a number of aspects, including research publications, eradications and public awareness. In our analysis, the degree of harmfulness of IASs was considered on the basis of weed risk assessment (WRA). The results show that legal classification of IASs has led to increases in research focused on the designated species, eradication efforts and public awareness. Similar results were also apparent for some AASs, but there was a great deal of variability among the many sub-categories of AASs. This may be partly because AAS sub-categories are based on different premises, and partly because the identification of AASs is not necessarily linked to legal controls that affect the behavior of individuals.  相似文献
