The level of benzoic acid was measured in Lemna gibba G3 grownon M and E media under inductive and non-inductive daylengths.Benzoic acid was slightly higher in plants grown on M mediumbut there was no difference in the benzoic acid levels in floweringand vegetative plants. When L. gibba G3 was grown under continuouslight on 1/10 M medium or 1/2 H medium there was virtually noflowering, but addition of benzoic acid to either medium ledto a substantial flowering response. In both cases this floweringresponse was inhibited by the plant hormones IAA, GA3, ABA andzeatin, with IAA and GA3 being the least inhibitory and ABAbeing the most inhibitory. This same pattern of inhibition wasseen when L. gibba G3 was grown on M medium under continuouslight, conditions that lead to photoinduction of flowering.These results leave open the possibility that endogenous benzoicacid may interact with other factors to influence the floweringresponse in L. gibba G3. Gibberellin A1, (GA1), GA19, and GA20 in phloem exudates andcotyledons of seedlings of Pharbitis nil cv. Violet, grown underdifferent photoperiodic conditions, were qualitatively and semi-quantitativelyanalyzed by a combination of high performance-liquid chromatography(HPLC) and radioimmunoassays (RIA). The levels of GA19 and GA20were higher in cotyledons from plants grown under dark treatment(DT) conditons of 16 h-light/8 h-dark for 6 days followed by8 h-light/16 h-dark for 3 days than in those grown under continuouslight (CL) for 9 days. This relationship was also observed forthe GAs in phloem exudates, although the levels were much lowerthan in the cotyledons. When GAs were applied to the cotyledons,elongation of the epicotyl was promoted more by GA20 than byGA1 or GA19, especially under the CL treatment. Effects of exogenous growth regulators are related to the histologicalevents in adventitious root initiation in hypocotyls of Pinusradiata D. Don. Indolebutyric acid (IBA) is required both forthe pre-initiative and post-initiative phases in root development.Kinetin has a marked inhibitory effect over the pre-initiativephase, with escape from inhibition following establishment ofmeristemoids. Gibberellic acid (GA3) has three distinct effectsdepending on time of application. While inhibitory during thepre-initiative phase, it strongly enhances rooting if appliedat a time coincident with the first observable stage of rootinitiation. Embryoids obtained from subcultured ovular callus of the Shamoutiorange (Citrus sinensis) were grown on Murashige and Tuckernutrient medium (BM) in the presence and absence of gibberellicacid (GA2), adenine sulphate (ADS) and combinations of these.Both GA2 and ADS stimulated significantly the rooting of smallembryoids with unorganized tissue, and larger embryoids withpartially developed root zones. Only large embryoids with fullydeveloped root zones rooted well on BM but better on BM+GA2+ADS.Rooting was best on media containing 1 to 5 mg per 1 ADS, andthese two combined. The optimum sucrose concentrations were5 to 6 per cent. Shoot formation on embryoids prior to rootingcompletely inhibited rooting which is otherwise enhanced byGA2+ADS. Filter-sterilized and autoclaved GA2, showed similaractivity in rooting.  相似文献

Both IBA and GA3 enhance rooting, IBA being more effective particularlyat a concentration of 100 mg/liter as compared to 10 mg/liter.IBA decreases while GA3 increases the sprouting of buds, withboth effects increasing with concentration. Four 13-hydroxygibberellins, gibberellin A1 (GA1), 3-epi-GA1,GA19 and GA20 were identified by full-scan GC/MS in extractsof lettuce seedlings (Lactuca sativa L. cv. Grand Rapids). Theresults suggest that the early-13-hydroxylation biosyntheticpathway to GA1 functions in the lettuce seedlings. It was alsofound that GA1 is active per se in the control of hypocotylelongation in lettuce seedlings. To investigate the relationshipbetween control by light of hypocotyl elongation and levelsof endogenous GAs in lettuce, endogenous levels of GAs werequantified by radioimmunoassay in seedlings that had been grownfor 5 days in the dark (5D) and in those that had been grownfor 4 days in the dark and then under white light for 1 day(4D1L). The endogenous level of GA1 in the upper and lower partsof hypocotyls in 5D seedlings was about four times higher thanthat in 4D1L seedlings. The response of explants (hypocotylsegments with cotyledons) from dark-grown seedlings to GA1 isknown to be similar in the dark and under white light when theexplants are treated with inhibitors of the biosynthesis ofGA. Therefore, the above information suggests that the highlevel of GA1 in hypocotyls of dark-grown seedlings is responsiblefor the rapid elongation of hypocotyl, while irradiation bywhite light decreases the endogenous level of GA1 in the hypocotylswith a resultant decrease in the rate of hypocotyl elongation. The effects of gibberellin A3(GA3) on chloroplast starch formationfrom exogenous sucrose in epidermal strips of light-grown cucumber(Cucumis sativus L. cv. Aonagajibai) hypocotyls were studied.Chloroplast starch formation was measured by counting microscopicallythe number of I2KI stained chloroplasts. In the presence ofsucrose (1–50 mM) and in light, chloroplast starch formationoccurred rapidly for 5 hr and then the rate declined. GA3 (100µM) simultaneously supplied with sucrose clearly suppressedboth the rate and the degree of starch formation. When the epidermiswas preincubated in the dark with sucrose in the presence orabsence of GA3 and then post-incubated in light with bufferonly in the presence or absence of GA3, starch formation wassuppressed to the same degree by GA3 given either in the pre-or post-incubation period. When the epidermis was preincubatedin the dark with GA3 alone and then transferred to light forpost-incubation with sucrose alone, GA3 suppressed the initialrate of starch formation during the post-incubation period.The degradation of chloroplast starch formed in advance fromexogenous sucrose in light was not significantly affected byGA3 These results are discussed in relation to the mechanismof the GA3 action and also to the GA3-induced decrease in theosmotic potential of the cell. Maize (Zea mays L., hybrid Cargill 147) seedlings, grown inaseptic conditions, were inoculated with three strains of Azospirillumlipoferum (Al op 33, Al iaa 320, and ATCC 29708) or culturedin different concentrations of indol-3-acetic acid (IAA) orgibberellin A3 (GA3). After 48 h, root length, root surfacearea, root dry weight, and shoot dry weight were measured inall treatments. Gibberellin content was evaluated in selectedroots of control and Azospirillum inoculated seedlings by gaschromatography-mass spectrometry-selected ion monitoring withthe use of deuterated gibberellins as internal standards. Thethree strains of A. lipoferum, IAA (2 ng ml–1), and GA3(40 to 400 pg ml–1) significantly enhanced root growth.Improvement of root hair growth and density was obtained mainlywith A. lipoferum strain Al op 33 and GA3 40 pg ml–1.GA3 was identified by gas chromatography-mass spectrometry (byboth, full scan and selected ion monitoring) in a free acidfraction from roots of the seedlings inoculated with A. lipoferum.In the roots of the non inoculated seedlings GA3 was found afterhydrolysis of a fraction expected to contain glucosyl conjugates. We investigated the metabolism and translocation of two gibberellins(GAs), [3H]GA20 and [3H]GA1, which were applied at low concentrationto the cotyledons of Pharbitis nil (cv. Violet). Seedlings weregrown under three different photoperiodic conditions: continuouslight (CL-CL), continous light followed by short day conditions(CL-DT) and long day conditions followed by short day conditions(DT-DT). Translocation of the applied [3H]GAs from cotyledonsto hypocotyls was promoted by DT for all GAs examined. Whilethe conversion of the translocated [3H]GA1 to [3H]GA8 and itsconjugates was rapid in hypocotyl, the conversion of translocated[3H]GA20 to [3H]GA29 was slow. Radioactivity in epicotyls wasdetected much more rapidly on application of [3H]GA20 than of[3H]GA1, [3H]GA8 and [3H]GA29 and their conjugates. The conversionof [3H]GA20 to [3H]GA1 in the epicotyl was more rapid underCL-CL conditions. This result in consistent with the higherlevel of endogenous GA1 existing in epicotyls under CL-DT thanDT-DT conditions. However, when [3H]GA1 was applied to the cotyledon,only small amounts of [3H]GA8 and its conjugates were detectedin the epicotyl regardless of the photoperiodic conditions.This result may suggest that the translocation and metabolismof [3H]GA20 from cotyledons to epicotyl was faster under CL-CLthan DT-DT conditions and may correlate with the increased epicotylelongation of GA20 treated plants under CL-DT than DT-DT conditions. GA3 (1 mM) promotes the germination of dark grown seeds of Lactucasativa variety "Spartan Lake" above levels obtained when theseseeds are germinated in the light for 18 hr. Cyclic-AMP alsopromotes the germination of these seeds in the dark but to amuch smaller extent than that obtainable with 1 mM GA3 and equivalentto levels obtained with 1 µM GA3. Combinations of lowconcentrations of GA3 and cyclic-AMP significantly increasedgermination over levels obtained with either compound alone,promoting germination at times to near optimum levels. GA3 isrequired for less than half the incubation period to bring aboutmaximum germination. ABA was found to inhibit the GA3 and cyclic-AMP induced responseas well as the response obtained with combinations of the nucleotideand hormone. Theophylline and caffeine in combination with lowGA3 concentrations promoted germination over seeds treated withthe hormone alone. Of a variety of adenine containing compoundstested only ADP and ATP gave responses similar to those obtainedwith cyclic-AMP. Some of the implications of these results arediscussed. Darkness and GA3 stimulate the elongation of the first internodebut inhibit the production of roots while IAA inhibits internodalelongation but promotes the production of roots on epiphyllousbuds of Bryophylhum tubiflorum. Cycloheximide inhibits both,implicating the synthesis of proteins in the growth of bothroot and shoot. Even pre-treatment of buds with cycloheximidefor 4 h inhibits rooting as well as internodal elongation whenthese are subsequently transferred to IAA or GA3. On the otherhand, pre-treatment with IAA or GA3 even for 8 h does not alleviatethe inhibitory effect of cycloheximide suggesting that thereis a lag period between the application of these regulatorsand the synthesis of proteins caused by them.  相似文献

Growth in lengths of tomato stems and leaves was acceleratedby 5.0 µg gibberellic acid (GA2) applied to the seed,or by 5.0, 0.5, and 0.05 µg given to the roots. Treatmentwith 5.0 µg also decreased bud number and lengthened thetime between bud appearance and fruit formation on the firsttruss by 1–8 days. Smaller amounts applied to roots shortenedthis time by 1–4 days. Indol-3yl-acetic acid at 0.5 µghad no effect, nor was simultaneous application of GA3 and IAAto the roots more effective than GA3, alone. Single applicationsof very small amounts of GA3 to seeds or seedling roots thusproved capable of changing growth-rates of stems, leaves, andtrusses.The effects of treating tomatoes with GA2 and with culturesof Azotobacter chroococcum, which contain small amounts of GA3,and IAA, are compared.  相似文献

Photoperiod controls the initiation and development of roots on cuttings of Bryophyllum tubiflorum. Root initiation occurred when either the mother plant or the cuttings were exposed subsequently to SD conditions. Cuttings from LD plants exposed to LD did not root at all even after 4 weeks showing that short days are necessary for rooting of cuttings of this plant. The short day requirement can be substituted by IAA or IBA as roots were initiated in cuttings from LD plants when treated with the auxins under LD conditions. GA stimulates rooting of cuttings under SD conditions but does not replace the SD requirement. It failed to induce rooting under LD conditions.  相似文献

GA3 increased the extension growth of Impatiens balsamina L.till 56 days under 8- and 24-h photoperiods. Cycloheximide whichdecreased height slightly under inductive conditions at a laterstage did not affect the GA3-promoted extension growth. BothGA3 and cycloheximide caused enhancement of the rate of differentiation,although this effect was temporary in the case of GA2. The interaction of light, gibberellic acid (GA3), and phlorizinin the growth of lettuce (Lactuca sativa L. cv. ‘GrandRapids’) hypocotyls was investigated. At all concentrationsof GA3, phlorizin enhanced GA3-induced growth at luminous intensitiesabove 50 ft-c (continuous light). Without GA3, phlorizin hadno effect on hypocotyl growth in the light but it inhibitedgrowth in the dark. Both seedlings and hypocotyl sections respondedto phlorizin in the presence of GA3. There was no iteractionbetween phlorizin and KCl. Water-growth was severly inhibitedby light. GA3,-induced growth was slightly inhibited by light,and then only at luminous intensities above 50 ft-c. Thus, relativeto H2O-growth, GA3-induced growth increased with increasingluminous intensity up to 450 ft-c, where it reached saturation.It seems that a synergism may exist between light and GA3 aswell as between phlorizin and GA3. Exogenous gibberellin A3(GA3) reduced the number of leaf nodesat flowering and time to flowering and increased the stem heightat flowering in three genotypes of spring rape (Brassica napusvar.annua L.). The responses to GA3were similar to those forlong days (LD) and low-temperature treatments, suggesting thatthe effect of photoperiod and the vernalization response areprobably mediated through gibberellins. The response to exogenousGA3was greatest in non-cold-treated plants in short days (SD)suggesting that endogenous GAs are limiting in these conditions.CCC, an inhibitor of gibberellin biosynthesis, caused a smallincrease in the number of leaf nodes at flowering and time toflowering and a small decrease in the stem height at flowering,but unexpectedly, its effect was hardly influenced by the applicationof exogenous GA3. Genotypes that showed the clearest responsesto the treatments with regard to the number of leaf nodes atflowering and time to flowering did not show the clearest responseswith regard to the stem height at flowering; the pattern ofresponses of the number of leaf nodes at flowering and timeto flowering was distinct from that of stem height at flowering.This indicates that flower formation and stem elongation areseparable developmental processes which may be controlled bydifferent endogenous gibberellins, different levels of a specificendogenous gibberellin, or different responses to gibberellin.Copyright 1999 Annals of Botany Company Brassica napus var. annua, gibberellin, photoperiod, spring rape, vernalization.  相似文献
