Phosphorus responses of C3 and C4 species   总被引:2,自引:0,他引:2  
An hypothesis was formulated that phosphorus (P) partitioningin tissues of C4 leaves would permit C4 plants to resist P deficiencybetter than C3 plants. To test this hypothesis, 12 C3, C4, andC3–C4 intermediate species were grown at adequate, deficient,and severely deficient P supply in a solid-phase-buffered sandculture system to characterize photosynthetic and growth responses.Species differed considerably in response to P stress. The growthof C3 species was more sensitive to P supply than C4 species,but C3 and C4 species had similar photosynthetic P use efficiency,and C4 species did not have low leaf P content, contrary toour hypothesis. In fact, leaf photosynthetic rates were notcorrelated with growth responses. Moncots had lower leaf P contentand better maintenance of leaf production under P stress thandicots, because of greater inhibition of branching (dicots)than of tillering (monocots). The most P efficient species inthis survey was Brachiaria, a C4 monocot that increased rootbiomass allocation under stress while maintaining P allocationto the shoot. It is concluded that C4 species are not inherentlymore P efficient than C3 species, but that monocots are moreP efficient than dicots, because of contrasting P and biomassallocation under stress. Changes in phloem loading rate were inferred from observationsof 11C export from attached leaves of C3 and C4 monocots anddicots. Loading decreased under anoxia in C3 leaves, but notin general in the leaves of either C4 monocots or dicots inthe light. However, loading rate in the C4 leaves did reduceif the leaf was also darkened or received no CO2. We suggestthat insensitivity to anoxia in C4 leaves is due to oxygen fromphotosynthesis which is retained in the bundle sheath at a concentrationsufficient to energize a phloem loading system. There may alsobe another system which is insensitive to anoxia since the effectsof shade and CO2 deprivation were not always seen, and loadingwas not completely stopped by these treatments. Exposure of the facultative halophyte Mesembryanthemum crystallinumL. to salt stress induces a shift from C3 photosynthesis toCrassulacean acid metabolism (CAM). During induction of CAM,the activity of NADP-malic enzyme (EC 1.1.1.40 [EC] ) increased asmuch as 12-fold in leaves, while the enzymatic activity in rootsfell to half of the original level. These changes in the activityof the enzyme corresponded to changes in levels of the enzymeprotein. NADP-malic enzymes extracted from leaves in the C3and CAM modes could be distinguished by differences in electrophoreticmobility during electrophoresis on a non-denaturing polyacrylamidegel. NADP-malic enzyme extracted from roots in the C3-mode andin the CAM mode migrated as fast as the enzyme extracted fromleaves in the CAM mode on the same gel. Although the patternof peptide fragments from NADP-malic enzyme from CAM-mode leaveswas similar to that from C3-mode leaves, as indicated by peptidemapping, both immunoprecipitation and an enzyme-linked immunosorbentassay revealed some antigenic differences between the enzymesextracted from leaves in the C3 and the CAM modes. These resultssuggest the existence of at least two isoforms of NADPmalicenzyme that differ in their levels of expression during inductionof CAM. Kleczkowski, L. A. and Randall, D. D. 1987. Immunologicallydistinct forms of adenylate kinase in leaves: comparison ofsubunit size of adenylate kinase from C3 and C4 plants.—J.exp. Bot. 38: 1440–1445. Antibodies prepared against maize leaf adenylate kinase (E.C.2.7.4.3 [EC] ) cross-reacted with the enzyme isolated from leavesof both C3 and C4 plants. The immunoreaction was very specificas judged by the presence of a single band on Western immunoblotscontaining proteins from leaf extracts of several species. Themolecular weight (M1) of adenylate kinase determined by meansof the immunoblotting was 29 kD and 27 kD for C4 and C3 species,respectively. For both C3 and C4 plants, the antibodies failedto precipitate all adenylate kinase activity in leaf extracts,while they were 100% effective in pelleting the enzyme frommaize mesophyll chloroplasts. This indicated the presence ofat least two immunologically distinct forms of adenylate kinasein leaves. It is suggested that the observed differences in molecular structure(M1s) of adenylate kinase from C3 and C4 species might be responsiblefor distinct catalytic and functional properties of the enzymein these two groups of plants. The irrununologically-determinedoccurrence of distinct pools of adenylate kinase in leaves supportsprevious evidence obtained by means of subcellular fractionationstudies.  相似文献

Kidney bean plants (Phaseolus vulgaris) were found to have thecapability to produce C6-aldehydes (hexanal and hexenals) fromlinoleic and linolenic acids. The various organs tested hadlipoxygenase and hydroperoxide lyase activities responsiblefor the C6-aldehyde formation. Young leaves showed relativelyhigh activities for C6-aldehyde formation. However, the activitiesof the leaves decreased gradually with leaf expansion. Seedlingsand seeds containing cotyledons showed low activities for C6-aldehydeformation, because of the occurrence of an inhibitory factorin the cotyledons. The substrate specificity of the enzymeswas essentially the same among the various developmental stagesof leaves tested. (Received February 5, 1982; Accepted March 19, 1982)  相似文献

C4 cereals (Zea maya L. and Sorghum bicolor L. Moench) and C3cereals (Triticum aestivum L. and Hordeum vulgare L) were grownin nutrient solutions with constant, interrupted, or absentpotassium supply. The lack of potassium retarded shoot growthand depressed the chlorophyll accumulation in all species ina similar way. After the renewal of potassium, the differencesin the compensation for growth retardation were not correlatedwith the photosynthetic system, but with the recovery of chlorophyllaccumulation in younger leaves. As important for the compensationof shoot growth retardation was a slower senescence of old leavescompared to plants with a constant potassium supply. This wasshown by the chlorophyll content and PEP carboxylase activity.In contrast to C3 cereals, the C4 cereals did not react withhigher chlorophyll contents to the same extent after the renewalof the postassium supply. The PEP carboxylase activity, however,was immediately raised higher than in control leaves. Chlorophylland PEP carboxylase activity increased simultaneously only inless aged leaves.  相似文献

The possible role of photorespiration as a general stress protectionmechanism, and in C4 plant metabolism, is controversial. Inparticular, the potential involvement of photorespiration inthe acquisition of desiccation tolerance in ‘resurrection’plants is unknown. An investigation was carried out into whetherphotorespiration is present in leaves of the C4 resurrectionplant Sporobolus stapfianus Gandoger (Poaceae) and whether itfunctions as a mechanism of stress resistance in the desiccation-tolerantyounger leaves (YL) of this plant. It is shown that the enzymesinvolved in the photorespiratory pathway maintain their activityuntil 88% relative water content (RWC) in both YL and desiccation-sensitiveolder leaves (OL). In subsequent stages of dehydration stress,the enzymatic activity declined similarly in both YL and OL.The content of the phorespiratory metabolite, serine, and ethanolamine,a direct product of serine decarboxylation, is higher in theearly stages of dehydration (88% RWC) in OL, suggesting a transientlyenhanced photorespiratory activity in these leaves. This wasconfirmed by simultaneous gas exchange and fluorescence measurements,showing suppression of the electron transport rate in OL exposedto non-photorespiratory conditions (2% O2) at 85% RWC. It isconcluded that a higher photorespiratory electron transportoccurs in desiccation-sensitive OL, and it is therefore proposedthat the capacity to scavenge excess electrons through photorespirationdoes not contribute to protect leaves of the desiccation-tolerantYL of S. stapfianus during the stress. The initial products of 14CO2 assimilation were determined understeady state illumination of leaves of Flaveria trinervia, aC4 dicot of the NADP-mialic enzyme subgroup. Leaf age influencedthe partitioning of 14CO2 between the C4 cycle and the reductivepentose phosphate (RPP) pathway. An estimated 10 to 12%of theCO2 entered the RPP pathway directly in leaves about 20% fullyexpanded, whereas CO2 was apparently fixed entirely throughthe C4 pathway in leaves 75% or more expanded. This partitioningpattern was attributed to the bundle sheath compartment in youngleaves having a relatively high conductance to CO2 (i.e., beingsomewhat leaky). Of the initially labelled C4 acids, the proportion that wasmalate, relative to aspartate, increased continuously duringleaf expansion (from 60 : 40 to 87 : 13 at full expansion).Concurrently, there was an increase in the whole leaf activityof NADP malate dehydrogenase and a decrease in the activitiesof aspartate and alanine aminotransferases. Low chlorophylla/b values were observed in young leaves, which may coincidewith an enhanced capacity for non-cyclic electron transportin the bundle sheath chloroplasts of such tissue. Both enhancedaspartate metabolism and direct fixation of CO2 in the bundlesheath could provide a greater sink for utilization of photochemicallyderived NADPH in the bundle sheath of young leaves. C6-Aldehydes emitted from intact tea leaves were analyzed quantitatively.Emission of the aldehydes increased temporarily in mid-May whenenzymatic activities involved in aldehyde formation from lipidsbegan to increase. Levels of C6-aldehydes in tea leaves alsoincreased temporarily. However, the accumulated C6-aldehydesdid not always correspond to emitted ones. Leaves of three C4 plants, Setaria italica, Pennisetum typhoides,and Amaranthus paniculatus possessed five- to ten-fold higheractivities of a (Na+-K+)-dependent ATPase than those of twoC3 plants, Oryza sativa and Rumex vesicarius. Na+-K+ ATPasefrom leaves of Amarathus exhibited an optimal pH of 7?5 andan optimal temperature of 35 ?C. It required 40 mM K+ and 80mM Na+ for maximal activity. Ouabain partially inhibited (Na+-K+)-dependentATPase activity in leaves of C4 plants. Ouabain also blockedthe movement of label from initially formed C4 acids into endproducts in leaves of only C4 plants, Setaria and Amaranthusbut not in a C3 plant, Rumex. We propose that Na+-K+ ATPasemay mediate transfer of energy during active transport of C4acids from mesophyll into the bundle sheath.  相似文献

Radioactive starch, glucose and fructose have been preparedfrom tobacco leaves after assimilation of C14O2. The apparatusused for photosynthesis consisted of a shallow Perspex leafchamber connected to a closed gas system, in which C14O2 wasgenerated from BaC14O2. Six leaves, area 14 to 18 sq. dm. whenexposed to bright sunlight with an initial CO2 concentrationof 8 to 10 per cent., assimilated 3.35 g. of C14O2 in 8 to 10hours. At least 80 per cent. of the C14O2 supplied appearedin the leaves as starch and sugar and over 80 per cent. of theradioactivity was accounted for in these carbohydrates. Thespecific activity per m. atom of carbon of the isolated productswas 85 to 90 per cent. of that of the C14O2. Small amounts ofradioactive carbon were also incorporated in the leaf proteinand in the celluose, hemicellulose and polyuronides.  相似文献

NADP-malic enzyme (NADP-ME) and phosphoenolpyruvate carboxykinase(PCK) are specifically expressed in bundle sheath cells (BSCs)in NADP-ME-type and PCK-type C4 plants, respectively. Unlikethe high activities of these enzymes in the green leaves ofC4 plants, their low activities have been detected in the leavesof C3 plants. In order to elucidate the differences in the geneexpression system between C3 and C4 plants, we have producedchimeric constructs with the ß-glucuronidase (GUS)reporter gene under the control of the maize NADP-Me (ZmMe)or Zoysia japonica Pck (ZjPck) promoter and introduced theseconstructs into rice. In leaves of transgenic rice, the ZmMepromoter directed GUS expression not only in mesophyll cells(MCs) but also in BSCs and vascular cells, whereas the ZjPckpromoter directed GUS expression only in BSCs and vascular cells.Neither the ZjPck nor ZmMe promoters induced GUS expressiondue to light. In rice leaves, the endogenous NADP-Me (OsMe1)was expressed in MCs, BSCs and vascular cells, whereas the ricePck (OsPck1) was expressed only in BSCs and vascular cells.Taken together, the results obtained from transgenic rice demonstratethat the expression pattern of ZmMe or ZjPck in transgenic ricewas reflected by that of its counterpart gene in rice. Enzymes of the C4, C3 pathway and photorespiration have beenanalyzed for P. hians and P. milioides, which have chlorenchymatousbundle sheath cells in the leaves. On whole leaf extracts thelevels of PEP carboxylase are relatively low compared to C4species, RuDP carboxylase is typical of C3 species, and enzymesof photorespiratory metabolism appear somewhat intermediatebetween C3 and C4. Substantial levels of PEP carboxylase, RuDPcarboxylase, and photorespiratory enzymes were found in bothmesophyll and bundle sheath cells. Low levels of C4-acid decarboxylatingenzymes may limit the capacity for C4 photosynthesis in P. hiansand P. milioides. We report new information on silica deposition in 15 plant species,including nine grasses, two sedges and four composites. Thesilica depositional patterns found in seven of the grass speciesindicate that they are C4 plants. However the festucoid grassCortaderia selloana is a C3 plant with long leaf trichomes andoval silica structures in the leaves. In contrast the panicoidC4 grasses Chasmathium latifolium, Chasmathium sessiflorum,Imperata cylindrica, Panicum repens, Panicum commutatum andSetaria magna, all produce dumb-bell-shaped silica structuresin the leaves. The chloridoid grasses Spartina patens and Spartinacynosuroides have saddle-shaped structures and no dumb-bellor oval shaped ones. The sedges Rhynchospora plumosa and Scirpuscyperinus were found to have oval phytoliths and may be C3 plants.Our examination of these and other grasses strongly suggeststhat C4 grasses tend to produce the same type of silica cells.Grasses and sedges with C3 type photosynthesis tend to produceoval silica structures. The composite Grindelia squarrosa andsunflowers Helianthus angustifolia, Helianthus atrorubens andHelianthus tuberosus absorb relatively small amounts of siliconand larger amounts of calcium, where both elements deposit inleaf trichomes. We found no clear indicator for the C3 sunflowersor C4 types in the Asteraceae. Helianthus tuberosus leaves havemany trichomes on the adaxial surface. These trichomes havea higher concentration of silica than the surrounding leaf surface.Helianthus tuberosus leaves had much higher ash and silica contentsthan those of Helianthus angustifolia and Helianthus atrorubens.The composite Grindelia squarrosa has a usual deposition ofsilica in the basal cells around the guard cells. Silica depositionoften reflects the surface features of a leaf. An exceptionis Scripus cyperinus where the silica structures are deep inthe tissue and do not reflect the surface configurations. Theinforescence of Setaria magna had a 14.64 silica content. Thetufts of white, silky hairs characteristic of Imperata cylindricainflorescence have no silica. Methionine sulfoximine caused ammonia accumulation and photosyntheticrate inhibition in leaf discs from two C4 species, Zea maysL. cv. F. M. Cross (Hybrid) and Sorghum bicolor L. Moench cv.NC-70X, as well as one C3 plant species, Datura stramonium L.cv. stramonium. Similar results were obtained earlier with theC3 species Spinacia oleracea L. The effect occurred in the absenceof inorganic nitrogen reduction and was light dependent. When tea leaves were homogenized and incubated, the volatileC6-compounds hexanal, cis-3-hexenal, cis-3-hexenol and trans-2-hexenalwere formed much more by summer leaves than by winter leavesof tea plants (Camellia sinensis). The enzymes lipolytic acylhydrolase (LAH), lipoxygenase, fatty acid hydroperoxide lyase(HPO lyase) and alcohol dehydrogenase (ADH) and an isomerizationfactor were responsible for the sequential reactions of C6-compoundformation from linoleic and linolenic acids in tea leaf lipids,and there were seasonal changes in their activities. The tealeaf enzymes were of 3 types: LAH and lipoxygenase, which hadhigh activities in summer leaves and low activities in winterleaves; ADH, which had low activity in summer leaves and highactivity in winter ones; and HPO lyase and the isomerizationfactor, which did not seem to have any effect on the rate ofC6-compound formation throughout the year. Changes in enzymeactivities were induced by shifts in the environmental air temperaturerather than by the age of the leaves. The combined activitiesof these enzymes determined the amounts and compositions ofthe volatile C6-compounds formed, which are the factors thatcontrol the quality of the raw leaves processed for green tea. Excised leaves of a C3-photosynthetic type, Hordeum vulgare,a C4-type, Panicum miliaceum, and an intermediate-type, Panicummilioides, were allowed to take up through their cut ends a1 mM solution of butyl hydroxybutynoate (BHB), an irreversibleinactivator of glycolate oxidase. After 30 to 60 min in BHB,extractable glycolate oxidase activity could not be detectedin the distal quarter of the leaf blades. Following this pretreatment,recovery of 14C-glycolate from 14CO2 incorporated in a 10 minperiod was nearly maximal for each of the three plant types.Labeled glycolate was 51% of the total 14CO2 incorporated forthe C3-species, 36% for the intermediate-species, and 27% forthe C4-species Increased labeling of glycolate was compensatedfor primarily by decreased labeling of the neutral and basicfractions for the C3 and intermediate-type species. In the C4-type,label decreased primarily in the neutral and insoluble fractions,but increased in the basic fraction. A lower rate of glycolatesynthesis is indicative of a lower rate of photorespirationand consistent with a lower O2/CO2 ratio present in the bundle-sheathcells of C4-plants. We conclude that both decreased glycolatesynthesis and the refixation of photorespiratory-released CO2are important in maintaining a lower rate of photorespirationin C4-plants compared to C3 plants. Anatomical and morphological structures of leaf blades werecompared between C3 and C4 species in Panicum. Inter-specificvariation of stomatal density, longitudinal vein density andmesophyll thickness was highly correlative either plus or minuswithin respective groups. The two groups could not be distinguishedby a single character, since the variation ranges overlappedeach other. However, the quantitative relations between veindensity and the other two characters differentiated the twogroups well. In C3, stomatal density seemed to be a primaryfactor for regulating water balance, while in C4 vein systemwas considered to be important for the regulation. The specieswith intermediate photosynthesis behaved similar to the C3 species.In the C3 group, correlative variation was observed betweenleaf width, leaf angle and the three characters mentioned above.Variation of light-receiving area due to the changes of widthand angle of leaf blades was considered to be one of the adaptivestrategies of this group. Increase of light-receiving area wasin connection with the thinning of leaves. On the other hand,in the C4 correlations between length, width and angle of leaveswere low. Such loose character correlation may be achieved byits efficiency of CO2 utilization and its well developed veinsystems. Besides, NAD-me type species tended to have relativelylower stomatal and vein densities as compared with the otherdecarboxylation types in this group. Light and electron microscopic observations of the leaf tissueof Panicum milioides showed that the bundle sheath cells containeda substantial number of chloroplasts and other organelles. Theradial arrangement of chlorenchymatous bundle sheath cells,designated as Kranz leaf anatomy, has been considered to bespecific to C4 plants. However, photosynthetic 14CO2 fixationand 14CO2 pulse-and-chase experiments revealed that the reductivepentosephosphate pathway was the main route operating in leavesof P. milioides. The interveinal distance of the leaves wasintermediate between C3and C4Gramineae species. Alloteropsis semialata (R.Br.) Hitchcock is a widely distributedgrass species known to show marked morphological, anatomicaland physiological variation. An unusual feature of this grassis that it shows both C3, and C4 photosynthetic pathways withthe respective anatomy related to these pathways in one speciesand within a single environment. This study investigates ultrastructuraldifferences between C3 and C4 forms of A. semialata, and showsdistribution of chlorenchyma in the two forms, at light microscopelevel. Quantitative assessment shows marked differences in mesophylland Kranz sheath cells with regard to size and organelle content;also in distribution of starch grains, lipid droplets and organellesin the mesophyll cells of both forms. These differences arediscussed in relation to the C3/C4 syndrome. Measurements were made using a Digiplan electronic planimeteron transmission electron micrographs of mature leaves sectionedtransversely at mid-lamina, and on drawings of leaf sectionsshowing the distribution of chlorenchymatous tissue.
