采用不同比例的CO2(20%、40%、60%、80%)对接种一定量的酵母菌的鲜榨圣女果汁进行储藏,对糖、酸、醛、醇等果汁主要成分的变化量进行检测,研究气调储藏对接种酵母菌的鲜榨圣女果汁变质过程的影响。结果显示:随着时间的延长,不同CO2初始浓度影响圣女果汁糖的消耗量及速率,其中20%组>40%组>80%组>60%组;同时酵母的数量和果汁糖降解生成酸的量随CO2浓度的增加而减少,生成醇的量随着CO2浓度的升高而增加。说明高浓度CO2条件抑制果汁中糖降解生成酸类物质,同时促使糖酵解产生醇,因此,CO2浓度较高时会促进鲜榨圣女果汁的保藏作用。  相似文献

寄主植物影响害虫药剂敏感性的研究进展   总被引:14,自引:0,他引:14  
害虫取食不同寄主植物后,对杀虫剂的反应可归为3类:敏感性降低、增高和无明显变化。害虫对药剂的敏感性变化与不同植物中次生物质诱导激活/抑制昆虫体内相关解毒酶活性有关。这种诱导作用可受到植物营养、次生物质种类及其含量分布、害虫种类与发育阶段、以及环境温度等多种因素影响。经诱导的昆虫解毒酶对不同类型杀虫剂的代谢能力并不相同,进而导致对不同药剂的敏感性变化有明显差异。解毒酶系的诱导激活在害虫抗药性形成早期被认为有利于提高隐性抗性基因频率,从而可促进害虫抗药性的发展。最后,就寄主植物影响害虫对药剂敏感性在害虫治理中的应用作了探讨。  相似文献

摘要: 应用植物介导的昆虫RNAi进行害虫防治近10年来受到了广泛的关注,其作用机理包括两个阶段,首先是害虫靶标基因dsRNA在植物体内的表达、运输和贮存,然后是害虫取食该植物后,dsRNA特异性抑制害虫体内靶标基因的表达。目前,植物介导的昆虫RNAi主要针对鳞翅目、鞘翅目和同翅目害虫,可以引起害虫生长发育的异常,导致死亡/繁殖力下降,甚至影响到其子代的生长。影响植物介导昆虫RNAi效率的因素主要包括害虫靶标基因的选择、dsRNA靶定位点及长度、植物表达dsRNA载体的结构和转基因植物的遗传转化方式等。植物介导昆虫RNAi防治害虫的策略也面临着潜在的安全性问题,如转基因植物安全性和RNAi潜在脱靶性等。随着植物介导昆虫RNAi技术的成熟,该方法有望成为害虫防治的新策略。  相似文献

人类的发展面临着许多问题,其中包括环境污染与害虫暴发失控等, 对策应是人类谋求与自然协调共存。人类必须学会既要和众多的非生命物又要和数千万种生物包括100多万种昆虫协调共存。天敌昆虫是大害虫的克星; 人类赖以生存的经济植物是由昆虫传播花粉的; 昆虫是人类的美味佳肴和医病良药; 有些昆虫是重要工业原料。所以人类要锐意合理利用昆虫资源。对于农林害虫要尽可能谋求自然控制; 提倡利用植物的抗害性、利用害虫的天敌、利用昆虫生长调节剂、种间信息物质、昆虫辐射不育技术控制害虫。尽量利用植物性杀虫剂控制害虫。尽量采用不杀伤或少杀伤天敌、不污染环境的化学杀虫剂或方法防治害虫。我国创造了很多行之有效的利用农业技术防治害虫的方法。  相似文献

大气CO2浓度增加已经受到国内外的极大关注。自2002年以来,在自行设计、组装的一系列密闭式动态CO2气室和开顶式CO2浓度控制箱基础上,研究了我国主要类型昆虫对CO2浓度升高响应的特征。结果显示,大气CO2浓度升高降低了棉铃虫Helicoverpa armigera的适合度和对棉花的危害作用,增加了棉花对棉铃虫为害的补偿作用,使以咀嚼式口器昆虫为代表的棉铃虫种群发生与危害下降;但大气CO2浓度升高改变了植物组织营养物质的组成与含量,提高了蚜虫Aphis gosypii对氨基酸营养的利用与补偿效率,降低了3种麦蚜的种间竞争,导致蚜虫种群发生与危害严重;而对烟粉虱Bemisia tabaci的种群特征影响较少。大气CO2浓度升高对天敌昆虫的影响存在种的特异性,表现出种群上升、下降和变化不大等特征。未来大气CO2浓度升高下,由于作物生长发育加快,生物量增加,蚜虫种群增多,导致吡虫啉农药防治效果下降,由此未来大气CO2浓度升高下农民将被迫使用更多的化学农药防治蚜虫类害虫,进而加重环境污染。  相似文献

固醇不但是昆虫细胞膜的重要组成部分,还是昆虫蜕皮激素的前体。尽管胆固醇是昆虫体内最常见的固醇,但不同昆虫的固醇组成有着显著差异,而且其组成随食物中固醇组成的变化而变化;即使同一种昆虫,其体内不同组织间也呈动态不均匀分布。为了从食物中获得固醇,昆虫有一套完整的运输体系,可以将固醇经过消化道运输到细胞内。在此过程中,携脂蛋白和固醇运输蛋白起着重要的作用。此外,国际上非常重视以固醇和昆虫为中心的各级生物的营养联系研究。本文综述了国际上的最新研究进展,系统地总结了固醇物质在昆虫体内的分布及代谢利用、固醇物质在昆虫体内的运输研究成果,提出了固醇类似物的开发、细胞内固醇运输载体抑制剂研制和基于改变固醇的抗虫植物培育等手段,以发展害虫防治新技术。  相似文献

Incidental transport of arthropods on plant material can be a significant mode of pest entry into greenhouses. We evaluated the use of controlled atmosphere treatments as a potential way to eliminate arthropod pests on plant propagules (i.e., cuttings or small rooted plants). Lethal exposures to CO2 or N2 were determined for common greenhouse pests including fungus gnat larvae, Bradysia sp.; green peach aphid, Myzus persicae (Sulzer); sweetpotato whitefly, Bemisia sp.; twospotted spider mite, Tetranychus urticae Koch; and western flower thrips, Frankliniella occidentalis (Pergande). We also studied the effect of pest species, life stage, and presence or absence of plants on efficacy of modified atmosphere treatments. Finally, effects of modified atmospheres on plant quality were evaluated for several bedding plant species including begonia, Begonia semperflorens-cultorum Hort. 'Cocktail Series', chrysanthemum, Dendranthema grandiflora Tzvelev., geranium, Pelargonium X hortorum L.H. Bailey, and impatiens, Impatiens wallerana Hook f., and among cultivars of geranium and chrysanthemum. Exposure for 12-18 h to >99% N2 or CO2 caused complete mortality of aphids, mites, thrips, and whiteflies. Fungus gnat larvae were more tolerant of hypoxic conditions. Adult mites and eggs were equally susceptible. For most pests, there was no difference in response to atmospheres modified by CO2 or N2. However, there was variation in response among plant species and cultivars, with effects ranging from delayed flowering to mortality. Despite the possibility of adverse effects on some plants, this work indicates that use of modified atmospheres has potential to eliminate arthropod pests on plant propagules before they are introduced into greenhouses.  相似文献

Nonchemical, environmentally friendly quarantine treatments are preferred for use in postharvest control of insect pests. Combined high temperature and controlled atmosphere quarantine treatments for phytosanitary fruit pests Macchiademus diplopterus (Distant) (Hemiptera: Lygaeidae) and Phlyctinus callosus (Schoenherr) (Coleoptera: Curculionidae) were investigated to determine the potential of such treatments for quarantine security. Field-collected, aestivating M. diplopterus adults and P. callosus adults were treated using a controlled atmosphere waterbath system. This system simulates the controlled atmosphere temperature treatment system (CATTS) used to control a number of phytosanitary pests in the United States and allows for a rapid assessment of pest response to treatment. Insects were treated under regular air conditions and a controlled atmosphere of 1% oxygen, 15% carbon dioxide in nitrogen, at two ramping heat rates, 12 and 24 degrees C/h. Treatment of both species was more effective under both heating rates when the controlled atmosphere condition was applied. Under these conditions of controlled atmospheres, mortality of P. callosus was greater when the faster heating rate was used, but the opposite was true for M. diplopterus. This could be due to the physiological condition of aestivation contributing to metabolic arrest in response to the stresses being applied during treatment. Results indicate that the potential for the development of CATTS treatments for these phytosanitary pests, particularly P. callosus, is promising.  相似文献

The development of postharvest quarantine treatments can be both expensive and time-consuming. It is necessary to determine the species and stage of the pest most tolerant to the treatment, if more than one species is the target of the treatment. Initial laboratory studies often include infesting the commodity with various egg and larval stages of the pest and performing treatments and evaluations of the fruit. In collaboration with others, I have previously developed combination high temperature under controlled atmosphere treatments against two quarantine pests in apples (Malus spp.) and peaches and nectarines (both Prunus spp.). I decided to develop an artificial system that can be used for these initial tests without the need for infesting large quantities of the fruit. I tested the system on the immature stages of the pests under regular air and controlled atmospheres by using the controlled atmosphere water bath system. This system can be used for rapid assessment of the most tolerant stage and species of a pest to a combination heat and controlled atmosphere treatment without the expense of infesting, treating, and evaluating the commodity.  相似文献

Previous studies have demonstrated that mature green tomatoes can be stored for up to 10 wk at 12. 5°C, 93–95% r.h. in a controlled atmosphere (CA) containing 5% CO 2 , 5% O2 and 90% N 2 , and will then ripen satisfactorily in air at 20°C. The effects of different concentrations of ethylene between <0.1 and 30 μl/litre in this storage atmosphere on ripening changes and fruit quality after 5 wk CA storage and a further 8 or 9 days ripening in air were investigated using cv. Sonatine glasshouse tomatoes. Maintaining ethylene concentrations in the storage atmosphere to.1 plllitre resulted in poor and uneven ripening of the tomatoes after storage, and increased their susceptibility to infection by Botrytis cinerea and Penicillium spp. Fruit previously stored in atmospheres containing 5 to 30 μl/litre ethylene were significantly softer after ripening than tomatoes stored in lower ethylene concentrations. Overall, the best results in terms of fruit quality (colour, firmness) and a low incidence of fungal infection were achieved with 1–3 μl/litre ethylene. The practical problems in achieving and maintaining optimum conditions, including the correct ethylene level, in CA storage of tomatoes are also discussed.  相似文献

Peaches (Prunus persica cv. Hermoza) were stored at 0C in regular air (RA) or in controlled atmosphere (CA 10% CO2, 3% O2) for 4 weeks and then ripened for 4 days at 20 degrees C. Woolliness developed in the regular air stored fruit while the controlled atmosphere stored fruit ripened normally. In the woolly fruit symptoms of the disorder were greater in the inner mesocarp than in the outer. Polygalacturonase (PG) and pectin esterase (PE) activities differed in the outer and inner mesocarp of the affected fruit. PG activity was low and PE activity was high in the inner mesocarp of the woolly fruit during ripening relative to the outer mesocarp, while in the healthy fruit, activities were similar in both areas. Cell wall fractions of water-soluble, CDTA-soluble and carbonate-soluble pectins were prepared from freshly harvested peaches and incubated with PE and PG from ripe peaches at different ratios. Only the CDTA-soluble fraction formed a gel with peach enzymes, and the rate of gelation increased with increasing amounts of PE relative to PG. Both water-soluble and CDTA-soluble pectin fractions formed gels with commercial PE (extracted from orange peel). The PE extracted from peaches was stable when stored at 0 degrees C for 9 days, while PG activity was stable only for 1 day. We suggest that PE, acting on pectins in the cell wall in vivo may cause gel formation and that the CDTA-soluble polymers have the capacity to bind apoplastic water and create the dry appearance observed in woolly fruit.  相似文献

Timing of nuclear maturation of nonstored and stored domestic cat oocytes   总被引:2,自引:0,他引:2  
In this study we compared the effects of preculture storage of ovaries, IVM medium, a reduced O(2) atmosphere and duration of culture on in vitro maturation (IVM) of domestic cat oocytes. One randomly selected ovary of each pair (69 pairs) was stored in PBS at 10 degrees C for 16-24h before oocyte recovery. The second ovary from each pair was used as a nonstored control. In Experiment I, we investigated the effect of culture media (TCM 199 versus SOF) and a reduced O(2) atmosphere (a humidified gas atmosphere of either 5% CO(2) in air or 5% CO(2):5% O(2):90% N(2)) on IVM of both stored and nonstored oocytes. In the second experiment, we compared timing of nuclear maturation of both stored and nonstored oocytes cultured for 17-18, 20-21, 24-26, 28-30, 33-34 or 42-45 h before being evaluated for meiotic status. In both, Experiments I and II, the recovery rate, quality and competence for maturation of oocytes originating from stored ovaries did not differ (P>0.05) compared with nonstored. In Experiment I, neither culture medium (37.5 versus 43.2% of Metaphase II, respectively in TCM 199 versus SOF) or gas atmosphere (40.0 versus 32.5% of Metaphase II, respectively in 5% CO(2) in air versus 5% CO(2):5% O(2):90% N(2)) affected oocyte maturation. In Experiment II, the mean proportion of oocytes achieving Metaphase II within 17-18 h of culture was 36.1% and did not significantly increase (P>0.05) over time up to 28 h. The highest proportion of oocytes (67.3%) reached Metaphase II stage after 42-45 h of culture. Therefore, we conclude that two "waves" of nuclear maturation of cat oocytes can be distinguished. The first wave takes place within 26 h and it is likely that most oocytes of this wave mature by 17-18 h; the second wave occurs after 28-30 h of IVM. It can be assumed that this double wave may reflect the presence of two oocyte populations with two different degrees of "prematuration" which require different lengths of IVM.  相似文献

The effect of atmospheric composition and storage temperature on growth and survival of uninjured and sublethally heat-injured Escherichia coli O157:H7, inoculated onto brain heart infusion agar containing 0.3% beef extract (BEM), was determined. BEM plates were packaged in barrier bags in air, 100% CO2, 100% N2, 20% CO2: 80% N2, and vacuum and were stored at 4, 10, and 37 degrees C for up to 20 days. Package atmosphere and inoculum status (i.e., uninjured or heat-injured) influenced (P < 0.01) growth and survival of E. coli O157:H7 stored at all test temperatures. Growth of heat-injured E. coli O157:H7 was slower (P < 0.01) than uninjured E. coli O157:H7 stored at 37 degrees C. At 37 degrees C, uninjured E. coli O157:H7 reached stationary phase growth earlier than heat-injured populations. Uninjured E. coli O157:H7 grew during 10 days of storage at 10 degrees C, while heat-injured populations declined during 20 days of storage at 10 degrees C. Uninjured E. coli O157:H7 stored at 10 degrees C reached stationary phase growth within approximately 10 days in all packaging atmospheres except CO2. Populations of uninjured and heat-injured E. coli O157:H7 declined throughout storage for 20 days at 4 degrees C. Survival of uninjured populations stored at 4 degrees C, as well as heat-injured populations stored at 4 and 10 degrees C, was enhanced in CO2 atmosphere. Survival of heat-injured E. coli O157:H7 at 4 and 10 degrees C was not different (P > 0.05). Uninjured and heat-injured E. coli O157:H7 are able to survive at low temperatures in the modified atmospheres used in this study.  相似文献

Stored grains are subject to deterioration and losses through various factors, but mainly insects and fungi. Various techniques are employed to detect stored product pests; however, there is an urgent need for an industrial-scale on-line detection technique. Near-infrared hyperspectroscopic imaging and soft X-rays have shown the potential for real-time application. These techniques are particularly effective for detecting internal infestations of stored grains. The digital images of the scanned objects are analyzed for various spectral and image features using statistical techniques such as complex multivariate tools. Classification accuracies as high as 80–100 % have been achieved for various pest and grain combinations. Dual-energy X-rays have been shown to detect the concealed eggs of stored product insect pests. The main threats to stored cereals come from Aspergillus spp., Penicillium spp., and Fusarium spp., which may produce mycotoxins. These imaging techniques have shown good results in the detection of fungal infections of stored grain.  相似文献

主要探讨冬枣(Ziziphus jujuba Mill. cv Dongzao)在-1 C的动态气调(CA-Ⅰ, 70% O2 0% CO2处理7天,然后转入5% O2 0% CO2中)、普通气调(CA-II, 5% O2 0% CO2;CA-III, 10% O2 0% CO2)及普通冷藏和常温(25C)等条件下,果实发病率、色素、可溶性固形物、可滴定酸、乙醇和乙酸乙酯含量等的变化。结果表明:与普通冷藏相比,气调贮藏能减缓果实的腐烂,抑制色素的分解和减少果肉中乙醇、乙酸乙酯的含量。动态高氧处理能有效地保持果实的颜色,抑制色素降解及果皮褐变。CA-Ⅲ (10% O2 0% CO2)能有效地减少果肉乙醇的含量而CA-Ⅱ(5% O2 0% CO2)能有效地减少果肉乙酸乙酯的含量。气调贮藏的果实可滴定酸及可溶性固形物含量与其他处理的果实没有明显的差异。  相似文献
