The bacterial aetiology of rosy discoloration of ancient wall paintings   总被引:1,自引:0,他引:1  
The inventory of microorganisms responsible for biological deterioration of ancient paintings has become an integral part of restoration activities. Here, the microbial agent of rosy discoloration on medieval frescoes in the Crypt of the Original Sin (Matera, Italy) was investigated by a combination of microscopic, molecular and spectroscopic approaches. The bacterial community from three rosy-discoloured painting sites was characterized by 16S rRNA gene-based techniques. The eubacterial population was prevalently composed of Actinobacteria, among which Rubrobacter radiotolerans-related bacteria accounted for 63-87% of the 16S rRNA gene pool per sampled site. Archaea, with prevalence of Haloarchaea-related species, were detected in one of the three sites where they accounted for < 0.1% of the total 16S rRNA gene pool. Raman spectroscopy confirmed the identity between R. radiotolerans carotenoids (bacterioruberins) and pigments responsible for colour alteration of frescoes. This investigation provides the first evidence of a causal relationship between heavy contamination by Rubrobacter-related bacterioruberin-producing bacteria and rosy discoloration of ancient wall paintings.  相似文献

Knowledge of the bioreceptivity of wall paintings to microbially induced decay processes has increased considerably in the last few years. Indoor wall paintings are widely recognised as a favourable environment for microbial biofilms in general and for microscopic fungi in particular. This environment also provides a suitable substrate for insects, spiders and other arthropodas. A combination of mycological and entomological studies of indoor wall paintings demonstrates an important ecological correlation between fungi and arthropods in the developmental processes of this complex microbial community. The co-existence of microscopic fungi and arthropods on the surface of wall paintings suggests an ecological interaction between them. Faunal impacts on the growth of biofilms due to grazing and propagule dispersal cannot be overlooked. Along with the above-mentioned perturbations, arthropods can directly damage the wall paintings by bioabrasion during grazing on the biofilm, as well as by the production of dwelling cavities. In this publication, we present the ecological connections between the major microscopic inhabitants of these important objects of cultural heritage. The additional role of arthropods as a remarkable source of organic substrate (e.g., chitin, spidersilk and faecal pellets) available for many micromycetes is discussed.  相似文献

古代岩画和壁画是人类文明发展历史进程的重要记录,具有极高的历史价值、艺术价值及科学价值,但他们无时无刻不受到环境因素引起的物理、化学及生物的劣化作用。近年,有关微生物对古代岩/壁画的危害逐渐受到关注。阐述了世界一些著名的岩/壁画文物的微生物的研究进展,如法国拉斯科洞穴岩画(Lascaux Cave)、西班牙阿尔塔米拉洞穴岩画(Altamira Cave)、敦煌石窟壁画、嘉峪关魏晋墓壁画以及我国东北地区公元5世纪的墓葬壁画等。通过对以上所述岩画和壁画微生物群落的分析比较,发现主要的细菌群落是变形菌门和放线菌门,放线菌门以假诺卡氏菌为主。真菌群落主要以虫生真菌为主。进一步分析了岩画和壁画微生物病害的共性和区别,揭示了洞穴、墓室等特殊环境下的微生物群落特点,为防治古代岩/壁画微生物病害提供了以下借鉴。  相似文献

In the present paper, results of studies conducted for the isolation and collection of different microorganisms and insects found at the Ajanta caves during April, July and October 1986 are presented. A number of fungal forms and bacteria were isolated and identified, but only a few were found in all the three trips. Among the insects, silverfish (Lepisma saccharina L.) was noticed during all our collection periods.  相似文献

A new, gentle enzymatic method was developed for a controlled removal of casein layers from medieval wall paintings. These casein layers were applied over the last 60 years on wall paintings in order to decrease substantial damage due to a peeling off of the frescoes from the roughcast surface due to environmental effects. However, due to the aging of the casein layers (at 40-50 years), a more drastic peeling occurred and the danger of total destruction of the wall paintings is severe. Thus, screening was performed to find the most suitable enzyme for casein digestion. Alcalase 2.5 DX L was the most appropriate enzyme for an effective proteolysis reaction. The enzyme was immobilized on functionalized cellulose membrane. A membrane pad system with immobilized enzymes was developed which could be pressed on the casein layers on the wall painting. A controlled removal of the casein layers by proteolytic digestion was observed and it was possible to continuously wash off the hydrolyzed casein fragments from the wall painting surface by an aqueous carbonate buffer flowing through the membrane pad. The removal and the digestion was monitored by reverse HPLC. Additionally, an on-line monitoring system was set up in order to continuously follow the casein layer removal and the digestion procedure directly on the wall painting. This technique is based on noninvasive 2D-fluorescence monitoring. Optical fiber systems were used to continuously monitor the fluorescence intensity of casein-bound tryptophan. The off-line data were verified with the on-line 2D-fluorescence data. Based on the scientific result an appropriate technique for the controlled enzymatic removal of damaging casein layers on the surface of medieval wall paintings using immobilized enzyme is now available. It is now applied to remove such casein layers from medieval wall paintings in the Allerheiligen-Kapelle Cloister, Wienhausen, Germany, and the St. Alexander Kirche, Wildeshausen, Germany.  相似文献

Summary A cooked haemoglobin medium was developed for the detection of hydrogen-peroxide-producing bacteria. The composition and preparation of this medium are described. Nitrogen-fixingEnterobacter agglomerans andEnterobacter spp. were consistently isolated from the bark beetleDendroctonus terebrans. Large populations of nitrogen-fixing bacteria were found with the beetle, although no in situ acetylene reduction was demonstrated. The constant occurrence of nitrogenfixing bacteria withD. terebrans suggests a symbiotic relationship. Nitrogen is generally considered one of the major limiting nutrients in plant growth. The biological process responsible for reduction of molecular nitrogen into ammonia is referred to as nitrogen fixation. A wide diversity of nitrogen-fixing bacterial species belonging to most phyla of the Bacteria domain have the capacity to colonize the rhizosphere and to interact with plants. Leguminous and actinorhizal plants can obtain their nitrogen by association with rhizobia or Frankia via differentiation on their respective host plants of a specialized organ, the root nodule. Other symbiotic associations involve heterocystous cyanobacteria, while increasing numbers of nitrogen-fixing species have been identified as colonizing the root surface and, in some cases, the root interior of a variety of cereal crops and pasture grasses. Phenol forms an insoluble complex with the polysaccharide in the cell wall of M.lysodeicticus, and renders it resistant to the action of lysozyme. The formation of the stable complex in the wall accounts for the bactericidal action of phenol. Metabolism of sulfur in bacteria associated with cheese has long been a topic of interest. Volatile sulfur compounds, specifically methanethiol, are correlated to desirable flavor in Cheddar cheese, but their definitive role remains elusive. Only recently have enzymes been found that produce this compound in bacteria associated with cheese making. Cystathionine - and -lyase are found in lactic acid bacteria and are capable of producing methanethiol from methionine. Their primary function is in the metabolism of cysteine. Methionine -lyase produces methanethiol from methionine at a higher efficiency than the cystathionine enzymes. This enzyme is found in brevibacteria, bacilli, and pseudomonads. Addition of brevibacteria containing this enzyme improves Cheddar cheese flavor. Despite recent progress in sulfur metabolism more information is needed before cheese flavor associated with sulfur can be predicted or controlled.  相似文献

Lactobacilli, streptococci and coliforms of pigeon crop contents, 5th wash, macerate and those of first-day pigeon milk have been studied. Streptococci predominated in all the samples tested. Relatively higher counts of lactobacilli and streptococci in crop macerate than in the 5th wash suggested the adhesion of these bacteria to crop wall. Because of frequent occurrence in crop of partially digested fibrous food, its contents were tested for the presence of cellulolytic bacteria. The results indicated that four isolates were capable of utilizing cellulose with the resultant production of reducing sugars. It is inferred that the crop microflora is involved in the degradation of dietary fibre in the pigeon.  相似文献

Mesosomes are unique membranous structures in bacteria. It is recognized that the mesosomes should be involved in several fundamental processes. The structure and behaviour of mesosomes have been studied and largely identified, while new evidences of mesosome function have been strikingly obtained. Our previous studies confirmed that hydrogen peroxide is involved in mesosomes formation during cell injury and cell division processes. Mesosome formation is always accompanied by excessive H2O2 accumulation. Furthermore, our recent data showed that mesosomes could not only enrich the excess H2O2, but also bring the H2O2 outside of the cells injured by antibiotics. It is a possibility that the enrichment of H2O2 in mesosomes might be a mechanism of drug resistance of bacteria. This article describes the bacterial mesosome and its functions as well as the involvement of hydrogen peroxide in mediating these functions.  相似文献

Bacterial communities associated with the brown alga Laminaria saccharina from the Baltic Sea and from the North Sea were investigated using denaturing gradient gel electrophoresis and 16S rRNA gene clone libraries. The rhizoid, cauloid, meristem and phyloid revealed different 16S rRNA gene denaturing gradient gel electrophoresis banding patterns indicating a specific association of bacterial communities with different parts of the alga. Associations with cauloid and meristem were more specific, while less specific associations were obtained from the old phyloid. In addition, seasonal and geographical differences in the associated communities were observed. Results from 16S rRNA gene libraries supported these findings. Bacterial phylotypes associated with the alga were affiliated with the Alphaproteobacteria (nine phylotypes), Gammaproteobacteria (nine phylotypes) and the Bacteroidetes group (four phylotypes). A number of bacteria associated with other algae and other marine macroorganisms were among the closest relatives of phylotypes associated with L. saccharina.  相似文献

The bacterial communities of aphids were investigated by terminal restriction fragment length polymorphism and denaturing gradient gel electrophoresis analysis of 16S rRNA gene fragments generated by PCR with general eubacterial primers. By both methods, the gamma-proteobacterium Buchnera was detected in laboratory cultures of six parthenogenetic lines of the pea aphid Acyrthosiphon pisum and one line of the black bean aphid Aphis fabae, and one or more of four previously described bacterial taxa were also detected in all aphid lines except one of A. pisum. These latter bacteria, collectively known as secondary symbionts or accessory bacteria, comprised three taxa of gamma-proteobacteria (R-type [PASS], T-type [PABS], and U-type [PAUS]) and a rickettsia (S-type [PAR]). Complementary analysis of aphids from natural populations of four aphid species (A. pisum [n = 74], Amphorophora rubi [n = 109], Aphis sarothamni [n = 42], and Microlophium carnosum [n = 101]) from a single geographical location revealed Buchnera and up to three taxa of accessory bacteria, but no other bacterial taxa, in each aphid. The prevalence of accessory bacterial taxa varied significantly among aphid species but not with the sampling month (between June and August 2000). These results indicate that the accessory bacterial taxa are distributed across multiple aphid species, although with variable prevalence, and that laboratory culture does not generally result in a shift in the bacterial community in aphids. Both the transmission patterns of the accessory bacteria between individual aphids and their impact on aphid fitness are suggested to influence the prevalence of accessory bacterial taxa in natural aphid populations.  相似文献

Scanning electron microscopy revealed that the average cell size of bacteria associated with the digestive tract of soil millipedes was 0.65 μm in diameter, 1.36 μm in length, and 0.60 μm3 in volume. An example of millipedes illustrated that the intestinal tract bacteria of soil invertebrates share the following features: (1) a high density level in this habitat; (2) existence mostly in the form of vegetative cells; (3) a cell size significantly smaller than that of bacteria functioning in soil; (4) a cell size closer to the lower limits of the size range characteristic for bacterial cultures grown in laboratory media. All this suggests that the bacterial community of the digestive tract differs from the typical soil community not only in composition but also in a higher level of physiological activity.  相似文献

Endosymbiosis is a mutualistic, parasitic or commensal symbiosis in which one symbiont is living within the body of another organism. Such symbiotic relationship with free-living amoebae and arthropods has been reported with a large biodiversity of microorganisms, encompassing various bacterial clades and to a lesser extent some fungi and viruses. By contrast, current knowledge on symbionts of nematodes is still mainly restricted to Wolbachia and its interaction with filarial worms that lead to increased pathogenicity of the infected nematode. In this review article, we aim to highlight the main characteristics of symbionts in term of their ecology, host cell interactions, parasitism and co-evolution, in order to stimulate future research in a field that remains largely unexplored despite the availability of modern tools.  相似文献
