ABSTRACT. The spectral sensitivity and flicker fusion frequency (FFF) of wild-type and salmon Glossina morsitans morsitans Westwood (Diptera, Glossinidae) were compared electroretinographically (ERG). Spectral sensitivity curves were similar in shape for dark-adapted wild-type and salmon flies, but salmon flies were over 100 times as sensitive as wild-type flies over much of their sensitivity range. Estimation of the spectral absorption curve (from the differences in ERG sensitivities) for the pigment absent from (or present in low concentration in) the salmon eye suggests that the pigment is an ommochrome. FFF at threshold light intensities was similar in wild-type and salmon flies, but at higher light intensity (1.3 °W/cm2) the FFF of salmon flies increased c. 200–300%, due to the capacity of the salmon eye to adapt rapidly to the flicker stimulus. Body weight had little effect upon spectral sensitivity and FFF. Wild-type males were more sensitive to yellow-green light and had higher FFF than did wild-type females. Salmon males and females did not differ in spectral sensitivity, but females had higher FFF (when tested with 520-nm light) than did males. Old wild-type females did not differ from young females in either spectral sensitivity or FFF. However, old salmon females were more sensitive but had lower FFF than young salmon females. Food deprivation reduced spectral sensitivity and FFF in wild-type males but not in salmon males. Irradiation (10.5 krad) reduced spectral sensitivity ( c. 75–375%) and FFF ( c. 30%) in wild-type males. The greatly increased spectral sensitivity and FFF in salmon flies indicate that these flies may behave differently from wild-type flies in the field. Differences in the way spectral sensitivity and flicker discrimination are affected by dark and light adaptation, and by such factors as age and sex, indicate that these measurements are of two independent phenomena.  相似文献

BACKGROUND AND PURPOSE: A novel light-emitting diode (LED) light source for use in animal-habitat lighting was evaluated. METHODS: The LED was evaluated by comparing its effectiveness with that of cool white fluorescent light (CWF) in suppressing pineal gland melatonin content and maintaining normal retinal physiology, as evaluated by use of electroretinography (ERG), and morphology. RESULTS: Pineal melatonin concentration was equally suppressed by LED and CWF light at five light illuminances (100, 40, 10, 1, and 0.1 lux). There were no significant differences in melatonin suppression between LED and CWF light, compared with values for unexposed controls. There were no differences in ERG a-wave implicit times and amplitudes or b-wave implicit times and amplitudes between 100-lux LED-exposed rats and 100-lux CWF-exposed rats. Results of retinal histologic examination indicated no differences in retinal thickness, rod outer segment length, and number of rod nuclei between rats exposed to 100-lux LED and 100-lux CWF for 14 days. Furthermore, in all eyes, the retinal pigmented epithelium was intact and not vacuolated, whereas rod outer segments were of normal thickness. CONCLUSION: LED light does not cause retinal damage and can suppress pineal melatonin content at intensities similar to CWF light intensities.  相似文献

Box jellyfish (Cubomedusae) are visually orientating animals which posses a total of 24 eyes of 4 morphological types; 2 pigment cup eyes (pit eye and slit eye) and 2 lens eyes [upper lens-eye (ule) and lower lens-eye (lle)]. In this study, we use electroretinograms (ERGs) to explore temporal properties of the two lens eyes. We find that the ERG of both lens eyes are complex and using sinusoidal flicker stimuli we find that both lens eyes have slow temporal resolution. The average flicker fusion frequency (FFF) was found to be approximately 10 Hz for the ule and 8 Hz for the lle. Differences in the FFF and response patterns between the two lens eyes suggest that the ule and lle filter information differently in the temporal domain and thus are tuned to perform different visual tasks. The data collected in this study support the idea that the visual system of box jellyfish is a collection of special purpose eyes.  相似文献

Spectral responses of male Glossina morsitans morsitans Westw, were investigated using behavioural and electrophysiological techniques. Phototactic responses to monochromatic lights of different wavelengths but equivalent intensities (measured either in energy or quantal units) were tested in an apparatus which permitted the simultaneous presentation of pairs of lights to groups of flies. Near ultraviolet light was the most attractive, followed by blue and red, with green and far red light giving no significant responses above control levels. The spectral sensitivity of the compound eye was also determined from the electroretinogram (ERG) recorded from electrodes implanted in the eye which was stimulated with flashes of monochromatic light. Three areas of maximum sensitivity were found by this method in the UV, blue/green and red. The relationship between the behavioural and electrophysiological sensitivity characteristics is explained with reference to visual systems of other Diptera which have previously been described.  相似文献

The Siberian sturgeon ( Acipenser baeri ) and the sterlet ( A. ruthenus ) were injected with dried sturgeon pituitary (2 mg kg−1), yielding 24 h later respectively 1.71 ± 0.5 and 1.65 ± 0.5 1011 spermatozoa kg−1 body weight. Spermatozoa were best activated with a solution of Tris HC1 50 mM, pH 8.0. The percentage of activated cells was 88 ± 4.4 in A. baeri (n = 5) and 68 ± 19 in A. ruthenus. (n = 5). In A. baeri , immediately after activation, the beat frequency of the flagellum and the mean velocity were in the range of 48–52 Hz and 100–300 μm−1s, respectively. The beat frequency declined to 15 Hz at 30–40 and velocity to 100 μm s−1 at 60 s post-activation. Only a small percentage of the spermatozoa remained motile after 3–4 min. In all cases spermatozoa showed mostly quasi-linear trajectories. Sperm was frozen in liquid nitrogen vapor immediately after dilution 1 v: 1 v in a cryopreservation medium (23.4 mM saccharose, 118 mM Tris-HCl pH 8.0, 20% egg yolk to which 15% DMSO were added). After fast-thawing procedure (25 s at 40°C), the percentage of motile spermatozoa (once activated in 118 mM Tris-HCl, pH 8.0) decreased to 23 ± 8.8 in A. baeri and to 15 ± 11 in A. ruthenus. The fertilizing capacity also decreased: 53 ± 8.3% vs. 89 ± 7.6% in control (P < 0.05) in A. baeri and 23 ± 11% vs 53 ± 8.3% in control (P < 0.05) in A. ruthenus. The motility pattern of the surviving frozen/thawed spermatozoa was the same as in fresh spermatozoa.  相似文献

ABSTRACT Studies were conducted in Zimbabwe of the responses of Glossina morsitans morsitans Westwood and Glossina pallidipes Austen resting in a refuge to various host stimuli. Tsetse took off in response to 100% ox odour, 0.08% carbon dioxide or a visual stimulus consisting of a 0.75 × 0.75 m black target placed c . 5 m from the refuge moving at 4o s-1, but the level of response was low with only 35%, 19% and 29% responding, respectively. Tsetse did not take off in response to any one of 25% ox odour, 0.8% carbon dioxide, acetone (3 μg 1-1) or octenol (0.03 μg 1-1). In the absence of any host stimuli, flies emerged from the refuge later on hotter days (35–37oC) than on cooler days (32–34.5oC). Male G.pallidipes emerging later in the afternoon contained significantly more haematin than those emerging relatively earlier. There were no significant differences between the responses of G.m. morsitans and G.pallidipes. It is suggested that the initial activation of resting flies is primarily mediated through endogenous, rather than host, stimuli.  相似文献

The digital fluorescent imaging for documentation and analysis of gel electrophoretic separations of nucleic acids and proteins is widely used in quantitative biology. Most fluorescent stains used in postelectrophoretic analysis of proteins and nucleic acids have significant excitation peaks with UV light (300-365 nm), making midrange UV (UV-B) as the excitation source of choice. However, coupling quantitative CCD imaging with UV is difficult due to lack of uniformity found in typical UV transilluminators. The apparent amount of those macromolecules depends on the position of the gel band on the imaging surface of the transilluminator. Here, we report the development and validation of a highly uniform UV transillumination system. Using a novel high density lighting system containing a single lamp formed into a high density grid, an electronic ballast, a phosphor coating, and a bandpass filter to convert 254 nm light produced to 300-340 nm, uniformity of 80% CV observed in typical UV transilluminators. The visual response of laboratory-reared wild-type and yellow-eyed mutant (lacking pigments) cabbage loopers, Trichoplusia ni, was studied by standard electroretinogram (ERG) techniques. The ERG waveform, time to dark-adapt, maximum sensitivity, and flicker fusion frequency (FFF) of both strains and sexes were compared with those of moths treated with 15 and 30 krad of γ-radiation. From the rate of dark adaptation, yellow-eyed moths were initially more sensitive than wild-type moths, but no significant differences were apparent in the ERG waveform, total time to dark-adapt, and FFF between strains, sexes, or treatment groups. However, both sexes of both strains exhibited increased sensitivity (required less light to evoke a threshold response when fully dark-adapted) to light stimulus when they had been treated with 15 krad of γ-radiation.A comparison of the effects of irradiation on the fertility and fecundity of the two strains showed no significant differences between the number of eggs oviposited by inseminated females. However, the lower egg hatch by yellow-eyed females at most dose levels indicated reduced egg viability for the yellow-eyed genotype. Thus, a significant dose × strain interaction was found when the percentage of egg hatch of the two strains was compared.  相似文献

Studies in Polar Base stations, where personnel have no access to sunlight during winter, have reported circadian misalignment, free-running of the sleep-wake rhythm, and sleep problems. Here we tested light as a countermeasure to circadian misalignment in personnel of the Concordia Polar Base station during the polar winter. We hypothesized that entrainment of the circadian pacemaker to a 24-h light-dark schedule would not occur in all crew members (n = 10) exposed to 100–300 lux of standard fluorescent white (SW) light during the daytime, and that chronic non-time restricted daytime exposure to melanopsin-optimized blue-enriched white (BE) light would establish an a stable circadian phase, in participants, together with increased cognitive performance and mood levels. The lighting schedule consisted of an alternation between SW lighting (2 weeks), followed by a BE lighting (2 weeks) for a total of 9 weeks. Rest-activity cycles assessed by actigraphy showed a stable rest-activity pattern under both SW and BE light. No difference was found between light conditions on the intra-daily stability, variability and amplitude of activity, as assessed by non-parametric circadian analysis. As hypothesized, a significant delay of about 30 minutes in the onset of melatonin secretion occurred with SW, but not with BE light. BE light significantly enhanced well being and alertness compared to SW light. We propose that the superior efficacy of blue-enriched white light versus standard white light involves melanopsin-based mechanisms in the activation of the non-visual functions studied, and that their responses do not dampen with time (over 9-weeks). This work could lead to practical applications of light exposure in working environment where background light intensity is chronically low to moderate (polar base stations, power plants, space missions, etc.), and may help design lighting strategies to maintain health, productivity, and personnel safety.  相似文献

The electroretinogram (ERG) of Rana ridibunda was recorded at different times of the year. The frogs exhibited distinct seasonal changes in various parameters of the ERG.The amplitudes of both the a- and the b-waves were greater in summer than in winter. With a luminance of 3000 cd/m2 the mean amplitude of the a-wave ranged from minimal 30 μ V (February) to maximal 60 μ V (July), and that of the b-wave from 190 μ V (February) to maximal 550 μ V (June). Average latency was about 28 ms in winter and 14 ms in summer. The greatest difference was found in flicker-fusion frequency which averaged 29 Hz in July and 9 Hz in January. There was also a significant change in the average threshold for response to white light (0.011 cd/m2 in March).  相似文献

Various drosophila mutants were used to dissect the electroretinogram (ERG) frequency response into components of different origins. The ommochrome granules in the receptor cell body are known to migrate in response to light, limiting the amount of light entering the rhabdomere. Comparison between the ERG frequency responses of the wild type and the mutant lacking the ommochrome granules indicates that the pigment migration reduces the amplitude gain at frequencies below 0.5 Hz. The ERG of drosophila compound eyes consists of contributions from receptor cells and the second-order cells in the lamina. Mutants with defective laminae showed a high-frequency cutoff with a corner frequency of about 20 Hz, while in wild type the response peaked in that frequency region. These results suggest that the lamina contributes mainly to the high-frequency components of the ERG transfer function. The shot noise model (Dodge et al., 1968) has been tested in drosophila by comparing the frequency response of the superimposed on the intracellular receptor potential. The results are consistent with the hypothesis that the receptor potential consists of a summation of small discrete potentials (bumps). In a mutant in which the bumps exhibit latency dispersion in response to a dim flash, the receptor showed a poor high-frequency response, the corner frequency being lowered to about 1-2 Hz. The slope of the cutoff was approximately 20 dB/dec indicating that the latency dispersion in this mutant is the major limiting factor in temporal resolution. Light-evoked high frequency oscillations have been observed in the ERG of another mutant. The oscillation was found sharply turned to light flickering at about 55 Hz.  相似文献

The long-term responses of the green microalga, Scenedesmusbicellularis, to a 100 Hz quasi-square wave (intermittent light,IL) were assessed after a 4 week acclimatization period. Atthe end of this period, the photosynthetic pigments, carboxylatingenzyme activities and biochemical composition of algae grownunder IL were compared to those of algae acclimatized to a lightflux without fluctuations (continuous light, CL). Differencesbetween IL and CL treatments were small. IL cells grown underlimiting irradiance had characteristics close to CL cells grownat the same daily irradiance. At saturating irradiance, thecharacteristics of IL cells resembled those of CL cells at thesame instantaneous irradiance, mostly because of the flattenedresponse of microalgae to increased irradiance. The flickeringof any artificial lighting system (100–120 Hz) can thusbe neglected when such light is used to grow algae.  相似文献

Ripples are sharp-wave-associated field oscillations (100-300 Hz) recorded in the hippocampus during behavioral immobility and slow-wave sleep. In epileptic rats and humans, a different and faster oscillation (200-600 Hz), termed fast ripples, has been described. However, the basic mechanisms are unknown. Here, we propose that fast ripples emerge from a disorganized ripple pattern caused by unreliable firing in the epileptic hippocampus. Enhanced synaptic activity is responsible for the irregular bursting of CA3 pyramidal cells due to large membrane potential fluctuations. Lower field interactions and a reduced spike-timing reliability concur with decreased spatial synchronization and the emergence of fast ripples. Reducing synaptically driven membrane potential fluctuations improves both spike-timing reliability and spatial synchronization and restores ripples in the epileptic hippocampus. Conversely, a lower spike-timing reliability, with reduced potassium currents, is associated with ripple shuffling in normal hippocampus. Therefore, fast ripples may reflect a pathological desynchronization of the normal ripple pattern.  相似文献

Ontogenesis of the electroretinogram, the mass electrical response of the retina to flash light stimuli, was studied in the guinea pig (Cavia porcellus), a precocial species with visual function at birth. 2. a-Wave components, b-wave, oscillatory potentials, slow PIII, and c-wave responses to flash stimuli developed between 55 and 64 days of gestation (full term is 68-69 days). 3. a-Waves attributable to photoreceptor functions were fully mature at 60 days. 4. A drought event can cause yield loss or entire crops to fail. In order to study the effects of continuous drought on physiological characteristics, yield, and water use efficiency (WUE) of winter wheat (Triticum aestivum L.), the variety “Zhoumai 22” was grown in controlled water conditions of the pot-planted winter wheat under a mobile rainout shelter. Foot planting and safe wintering were used to evaluate, winter wheat under different drought conditions, including light, moderate and severe drought at the jointing, heading, and filling stages. The soil water content was controlled at 60–70%, 50–60%, or 40–50% of field capacity. Experimental trials included 3 pre-anthesis drought hardening, 3 three-stage continuous drought, and 1 test control conditions. Under drought stress conditions, winter wheat leaf water potential, soil plant analysis development (SPAD), photosynthesis parameters, and yield declined due to pre-anthesis drought hardening. And the degree of decline: continuous drought > pre-anthesis drought hardening. Changes in the post-anthesis photosynthetic capacity of winter wheat were mainly related to the pre-anthesis drought level, rather than post-anthesis rehydration. The threshold of non-stomata limiting factors caused by photosynthesis at the filling stage is 40–50%FC, while comprehensive yield and WUE affected, the yield in severe drought treatments was the most significant, B3C3 and B3C3G3 decreased by 55.68% and 55.88%, respectively. Pre-anthesis drought was the main reason for the decreased crop yield. Thus, severe drought should be avoided during planting, while pre-anthesis light drought is a suitable choice for water-saving and crop production, as proper pre-anthesis drought hardening (60–70% FC) is feasible and effective.  相似文献

Daily counts of Southern sea lions ( Otaria byronia ) made at Punta Norte, Argentina, during four consecutive breeding seasons (1980–1984) yielded similar results in the time of the reproductive events. Males and females began to arrive during the second week of December. Males reached peak numbers (100–110) between 15 and 21 January. Females reached maximum numbers (300–350) at the end of January. About 380–430 pups were born between December 20 and February 2, and 70–80 percent of births occurred between 10 and 25 January. Copulations peaked during the third week of January. The sex ratio of adult males to adult females at peak season fluctuated from 1:3.7 to 1:4.2. By the first week of February, coinciding with the maximum number of young males (25–50), 90 percent of the adult males had abandoned the area and the number of females fluctuated greatly. Since 1980 the number of females and pups has shown a slight increase, particularly during the 1983–1984 breeding season.  相似文献

The lighting industry currently accounts for a significant proportion of all energy demand. Luminescent white lighting is often impure, inefficient, expensive, and detrimentally emits as a point source, meaning the light is emitted from a focused point. A luminescent light diffuser offers the potential to create a spatially broad lighting fixture. We developed a luminescent light diffuser consisting of three commercially available luminescent dye species (rhodamine 6G, fluorescein, 7-diethylamino-4-methylcoumarin) dispersed within a polymer matrix (polyvinyl alcohol), or commercial paint, and coated on a planar waveguide. A Light-emitting diode (LED) (385 nm) is directed into the waveguide which excites the luminescent species, coating the panel, creating a device that emits spatially broad pure white light. As the emission depends on escape cone emission from the waveguide, the device’s emission was found to depend highly on the coating film quality and components. We present two systems: a small 40 mm × 40 mm prototype, made using standard water-soluble polymer (polyvinyl alcohol), to study the underlying operational principles, and a 100 mm × 100 mm device with optimized efficiency fabricated with a clear commercial paint. By doping the polymer matrix with scattering silica microparticles we achieved a maximum photon outcoupling efficiency of 78%, whilst maintaining colour purity with an increased device size of more than 300 times (compared with the input LED). This work shows that it is possible to construct an inexpensive and spatially broad lighting source, whilst maintaining colour purity at a low cost.  相似文献

Light intensity, spectrum and pattern may affect laying hen behaviors and production performance. However, requirements of these lighting parameters from the hens’ standpoint are not fully understood. This study was conducted to investigate hens’ needs for light intensity and circadian rhythm using a light tunnel with five identical compartments each at a different fluorescent light intensity of <1, 5, 15, 30 or 100 lux. The hens were able to move freely among the respective compartments. A group of four W-36 laying hens (23 to 30 weeks of age) were tested each time, and six groups or replicates were conducted. Behaviors of the hens were continuously recorded, yielding data on daily time spent, daily feed intake, daily feeding time, and eggs laid under each light intensity and daily inter-compartment movement. The results show that the hens generally spent more time in lower light intensities. Specifically, the hens spent 6.4 h (45.4%) at 5 lux, 3.0 h (22.1%) at 15 lux, 3.1 h (22.2%) at 30 lux and 1.5 h (10.3%) at 100 lux under light condition; and an accumulation of 10.0 h in darkness (<1 lux) per day. The 10-h dark period was distributed intermittently throughout the day, averaging 25.0±0.4 min per hour. This hourly light-dark rhythm differs from the typical commercial practice of providing continuous dark period for certain part of the day (e.g. 8 h at night). Distributions of daily feed intake (87.3 g/hen) among the different light conditions mirrored the trend of time spent in the respective light intensity, that is, highest at 5 lux (28.4 g/hen, 32.5% daily total) and lowest at 100 lux (5.8 g/hen, 6.7%). Hen-day egg production rate was 96.0%. Most of the eggs were laid in <1 lux (61.9% of total) which was significantly higher than under other light intensities (P<0.05). Findings from this study offer insights into preference of fluorescent light intensity by the laying hens. Further studies to assess or verify welfare and performance responses of the hens to the preferred lighting conditions and rhythm over extended periods are recommended.  相似文献

Using excised fruit/shoot systems of pea ( Pisum sativum L.), the effect of the 10-day-old (post anthesis) fruit (sink) on the translocation of 14C-sucrose applied to the stipule (source) was investigated. We also examined the influence of various growth regulators on the source-sink relationships using this system. Indole-3-yl-acetic acid (100 or 200 ppm), gibberellic acid (25-100 ppm) increased 14C-sucrose translocation into the growing fruit by about 13% and 22–29%, respectively. 2-chloroethyl phosphonic acid, 6-benzyladenine and 2-methyl-4-chlorophenoxyacetic acid also increased 14C-sucrose translocation into the fruit by about 11–26%, 11–23% and 24–37%, respectively. Elevated fruit 14C-sucrose levels found in other treatments include, methyl-2-chloro-9-hydroxyfluorene-9-carboxylate (300 or 1000 ppm), 2,3,5-triiodobenzoic acid (100 and 300 ppm) and butanedioic acid mono-(2,2-dimethylhydrazide) (30-300 ppm) by about 34%, 39–52% and 7–13%, respectively.  相似文献

Microsupercapacitors (MSCs) with high energy densities offer viable miniaturized alternatives to bulky electrolytic capacitors if the former can respond at the kilo Hertz (kHz) or higher frequencies. Moreover, MSCs fabricated on a chip can be integrated into thin‐film electronics in a compatible manner, serving the function of ripple filtering units or harvesters of energy from high‐frequency sources. In this work, wafer‐scale fabrication is demonstrated of MXene microsupercapacitors with controlled flake sizes and engineered device designs to achieve excellent frequency filtering performance. Specifically, the devices (100 nm thick electrodes and 10 µm interspace) deliver high volumetric capacitance (30 F cm?3 at 120 Hz), high rate capability (300 V s?1), and a very short relaxation time constant (τ0 = 0.45 ms), surpassing conventional electrolytic capacitors (τ0 = 0.8 ms). As a result, the devices are capable of filtering 120 Hz ripples produced by AC line power at a frequency of 60 Hz. This study opens new avenues for exploring miniaturized MXene MSCs as replacements for bulky electrolytic capacitors.  相似文献
