On the basis of differences in crossing-over frequency and in radiation sensitivity, two stocks of Phryne cincta can be distinguished. The higher sensitivity of the Alpine stock (as), compared with the Berlin stock (bs), seemed to be due to its higher site number in tightly spiralized pronucleus chromosomes (Israelewski, 1979). A mathematical model was tested to arrive at an estimate of the maximal site number in Phryne. Deducing from this model, repair in a 2-break site would result with the probability of 1/6 in restitution, 1/6 in balanced chromosome aberration and 4/6 in unbalanced chromosome aberration. The maximal site number available is 13 in bs and 20 in as. It is suggested that a high proportion of 2-break sites are repaired after replication of the paternal pronucleus chromosomes. The proportion of prae- and postreplication repair is estimated to be 50% each, possibly in agreement with the distribution of nucleosomal and internucleosomal DNA in eukaryotic chromatin.  相似文献

After treatment of mature spermatozoa of Phryne cincta egg mortality rates were determined and chromosomal mutation rates by examination of the polytene chromosomes of the F1 progeny.--A comparison of the results with similar studies made on Drosophila melanogaster yielded a two- to threefold higher sensitivity in Phryne cincta. Evaluation of data concerning DNA amounts, heterochromatin contents, distribution of breaks, repair capacities in two-break sites and oxygen concentrations in mature spermatozoa revealed no contradiction to the hypothesis that the greater sensitivity of Phryne cincta is mainly due to a high degree of spiralization of the paternal pronucleus chromosomes.  相似文献

Summary The isolation and characterization of a new mutation conferring radiation sensitivity in Escherichia coli is described. This mutation is located close to the gene coding for deoxycytidine deaminase, in the chromosomal region of the gat operon. It is very sensitive to gamma rays and exhibits a decrease in recombination ability. The expression of radiation sensitivity seems to result from the additive effect of the dcd mutation and another mutation of unknown function.  相似文献

Karyotype and genetic variation of the common shrew (Sorex araneus) from Mt. Pelister in southern Macedonia has been studied. Whereas all autosomes in the chromosomal set (2 na, = 28, the only present metacentrics being af, bc, jl, and tu) were of the standard type as well as the sex chromosomes X and Y2 in males, the Y1 chromosome was a small metacentric. This chromosomal feature is unique among the common shrew populations studied cytogenetically so far. Three out of 33 loci analysed (Sdh, 6Pgd, Mdh-1) were discriminant between the Mt. Pelister population and Sorex araneus from Slovenia and two loci (Est-3, Ada) were partially discriminant. A relatively high value of Nei's genetic distance (D = 0.137) confirms unique character of the Pelister population.  相似文献

Reciprocal translocations between chromosomes XVI and VIII were revealed in eight Saccharomyces cerevisiae strains (mostly wine ones) using pulse-field electrophoresis of native chromosomal DNAs and their hybridizations with the CUP1 and GAL4 probes. New and reciprocal translocations of at least the gene CUP1 occur at the expense of crossing-over in the hybrids of such strains with the genetic lines of normal karyotype during meiosis. Relationship between these reciprocal translocations and the sulfite (Na2SO3) resistance gene SSU1-R is discussed.  相似文献

We used genetic methods to estimate the effective number of breeders (N b) in a population of Rana pretiosa, an imperiled amphibian in western North America. Microsatellite data was gathered from large samples of adults, eggs, and juveniles collected in 2006. We wished to determine where in the life cycle the greatest reductions in N b occur, and to compare genetic estimates of N b to an egg mass count estimate of the number of breeding adults. We predicted that N b estimated at the metamorph stage would be reduced by increased variance in family size due to egg mass mortality. Contrary to our prediction, estimates of N b at the egg and metamorph stages were similar. Thus, we found no evidence of inflated variance in family size between the two stages. If our results for this population are typical for R. pretiosa, then increased variance in family size during the egg to metamorph stage may not be a strong factor in reducing the effective population sizes (N e) relative to the census sizes (N) in this species.  相似文献

 Use of resistant host genotypes is an important component of an integrated approach to control black sigatoka, a disease caused by the fungus Mycosphaerella fijiensis Morelet. The objective of the present research was to determine the role of the major gene for black sigatoka resistance (bs 1 ) in the host response to this disease. Euploid hybrids with a known genotype for the bs 1 locus were derived from triploid-diploid crosses of two French plantains and a diploid wild banana, and were assessed for their host response to black sigatoka in plant and ratoon crops in the humid forest zone of Nigeria. Host response was determined at flowering by recording the number of standing leaves, the youngest leaf with symptoms, the youngest leaf spotted, the total leaf area attacked by black sigatoka, and an index of the leaves spotted. An analysis of frequency distribution in each segregating population showed that almost all the traits displayed a normal distribution across ploidy level. This suggests that additive gene action plays an important role in the host-plant response to the fungus. Heritability, repeatibility, and intraclass correlations were calculated. The environment and the genotype-by-environment interaction significantly affected the host response to black sigatoka, which explains the low repeatibility of all traits. The intrafamily variation was larger than the interfamily variation, and most of the genetic variation in each family depended on the individual genotypes, regardless of their ploidy. The additive effect of, and the intralocus interaction at, the bs 1 locus on host response to black sigatoka were established by a one-way analysis of variance and regression analyses. Intralocus interaction in the bs 1 locus apparently regulates the appearance of symptoms on the leaf surface, whereas the additive effect and the intralocus interaction of the bs 1 locus affect disease development in the host plant. Therefore, the gene action(s) at the bs 1 locus may provide durable resistance to black sigatoka by slowing down disease development in the host plant. The race IE-1k of Magnaporthe oryzae recovered from the Southern US overcomes the resistance (R) gene Pita. The objectives of the present study were to identify and tag R genes to IE-1k for rice breeding. TM2, S1, 94071, and B isolates of the race IE-1k were used to identify and map R genes from a resistant indica rice cultivar Zhe733 using a recombinant inbred line population from a cross of the genetic stock KBNTlpa1-1 and Zhe733. The ratio of 3 resistant:1 susceptible in 162 RIL of an F10-11 KBNTlpa1-1/Zhe733 (K/Z) population indicated that two major R genes in Zhe733 confer resistance to IE-1k. A total of 118 polymorphic simple sequence repeat markers were analyzed in 162 F10-11 individuals of the K/Z population to determine chromosomal locations of the loci conferring resistance to race IE-1k using composite interval mapping. Two major R genes temporarily designated as Pi42(t) and Pi43(t) each providing complete resistance to IE-1k were identified on chromosomes 8 and 11, respectively. RILs containing Pi42(t) and Pi43(t) were also resistant to other US races IB-1, IB-45, IB-49, IB-54, IC-17, IE-1, IG-1, and IH-1. The Pi42(t) gene was mapped between RM310 and RM72, and the location of Pi43(t) was closely associated with two flanking SSR markers RM1233 and RM224 on chromosome 11 in a chromosomal region carrying the resistance gene Pi1. Two molecular markers RM72 and RM1233 identified in this study should be useful for fine mapping and for facilitating incorporation of Pi42(t) and Pi43(t) into advanced breeding lines by marker-assisted selection. The chromosomal changes occurred in two independent cell lines (GM2 and GM3) of Drosophila melanogaster maintained in medium supplemented with serum and in serum-free medium were compared. In both culture conditions and in both lines a chromosomal evolution was revealed. Structural and numerical variations were analysed. The breaks giving rise to rearrangements were at heterochromatic level. Moreover, a tetraploidisation followed by loss of chromosomes or of portions of chromosomes recalls an analogous cycle observed in human cells.This work was supported by a grant of the Consiglio Nazionale delle Ricerche, Roma.  相似文献

Analysis of maize F2 plants revealed a decrease in the recombination fraction (rf) between the marker loci in case of heterozygosity of adjacent chromosomal regions. The relative decrease in rf was positively associated with the length of the heterozygous region. Heterozygosity had no effect onrfin other chromosomes. These results can be explained by defective pairing of heterozygous chromosomal regions in meiosis.  相似文献

The efficiency of C4 photosynthesis in Zea mays, Miscanthus x giganteus and Flaveria bidentis in response to light was determined using measurements of gas exchange, 13CO2 photosynthetic discrimination, metabolite pools and spectroscopic assays, with models of C4 photosynthesis and leaf 13CO2 discrimination. Spectroscopic and metabolite assays suggested constant energy partitioning between the C4 and C3 cycles across photosynthetically active radiation (PAR). Leakiness (φ), modelled using C4 light‐limited photosynthesis equations (φmod), matched values from the isotope method without simplifications (φis) and increased slightly from high to low PAR in all species. However, simplifications of bundle‐sheath [CO2] and respiratory fractionation lead to large overestimations of φ at low PAR with the isotope method. These species used different strategies to maintain similar φ. For example, Z. mays had large rates of the C4 cycle and low bundle‐sheath cells CO2 conductance (gbs). While F. bidentis had larger gbs but lower respiration rates and M. giganteus had less C4 cycle capacity but low gbs, which resulted in similar φ. This demonstrates that low gbs is important for efficient C4 photosynthesis but it is not the only factor determining φ. Additionally, these C4 species are able to optimize photosynthesis and minimize φ over a range of PARs, including low light.  相似文献

When diploid cells of Saccharomyces cerevisiae homozygous for the temperature-sensitive cell division cycle mutation cdc6-1 are grown at a semipermissive temperature they exhibit elevated genomic instability, as indicated by enhanced mitotic gene conversion, mitotic intergenic recombination, chromosomal loss, chromosomal gain, and chromosomal rearrangements. Employing quantitative Southern analysis of chromosomes separated by transverse alternating field gel electrophoresis (TAFE), we have demonstrated that 2N-1 cells monosomic for chromosome VII, owing to the cdc6-1 defect, show slow growth and subsequently yield 2N variants that grow at a normal rate in association with restitution of disomy for chromosome VII. Analysis of TAFE gels also demonstrates that cdc6-1/cdc6-1 diploids give rise to aberrant chromosomes of novel lengths. We propose an explanation for the genomic instability induced by the cdc6-1 mutation, which suggests that hyper-recombination, chromosomal loss, chromosomal gain and chromosomal rearrangements reflect aberrant mitotic division by cdc6-1/cdc6-1 cells containing chromosomes that have not replicated fully.  相似文献

The present study was carried out in vitro to determine the efficacy of indigenous fungi isolated from egg masses of root-knot nematode, Meloidogyne incognita on egg parasitism, egg hatching, mobility and mortality against root-knot nematode, M. incognita. The tested fungi were Acremonium strictum, Aspergillus terreus, A. nidulans, A. niger, Chetomium aubense, Chladosporium oxysporum, Fusarium chlamydosporium, F. dimarum, F. oxysporum, F. solani, Paecilomyces lilacinus, Pochonia chlamydosporia, Trichoderma viride and T. harzianum. All tested fungi showed varied effects against the nematodes. Culture filtrates of A. strictum was very effective against the nematode in regards to egg parasitism (53%), egg hatching inhibition (86%) and mortality (68%) compared to controls. A. strictum was found to have an advantage over P. lilacinus, P. chlamydosporia, T. viride and T. harzianum in that it caused greater mortality of the second stage juveniles (J2). A. terreus did not show egg parasitism but was found to be highly toxic against second stage juveniles (J2) causing high mortality (around 68%). Thus, A. strictum and A. terreus showed good biocontrol potential against root-knot nematode, M. incognita under in vitro conditions.  相似文献

A population of lady-bird beetles at the head of the Crowsnest River valley on the immediate east side of the Rocky Mountains has been sampled regularly over the years 1961 to 1964. The 389 individuals studied had chromosome numbers ranging from 2n = 14 (58%) to 2n = 20 (3%). The former is the diploid number of C. hexacyclus, the latter that of C. tricyclus, species otherwise known respectively only in the Prairie Provinces and the interior of British Columbia plus the adjoining part of the State of Washington. About 19% of the beetles had the intermediate number of 17 chromosomes, males possessing either a hexacyclus-type or a morphologically distinct tricyclus-type Y chromosome. — In all numerical categories meiotic pairing was virtually perfect but the trivalents in heterozygotes were frequently linearly aligned. This assures production of aneuploid (duplicationdeficiency) gametes, only complementary recombinations of which are viable. Segregational sterility resulting from triple (92%), double (84%) and single (50%) heterozygotes is estimated to be ca 40%. Of four neo-diploids, with zero chromosomal sterility, predicted in the F2 only one materialized with certainty, the one with 13 + tricyclus-type Y. This is interpreted to mean that F2 progeny are rare; rare because (1) random mating, complete introgression, is the rule and (2) Segregational sterility is severest in triple heterozygotes. — Despite the population-depressing effect of the inviable aneuploid zygotes, it is calculated that segregation and crossing-over generate a minimum of 148 × 109 new recombinants (a minimum were crossing-over strictly localized); those acquiring beneficial attributes will be favoured by natural selection. — Topographically, it is evident that tricyclus adults invade the resident hexacyclus community year after year, borne on the prevailing westerly winds that funnel through Crowsnest Pass (elevation only 4463): hexacyclus is thereby constrained to the east of the Continental Divide and consequently does not penetrate into tricyclus territory.Dedicated to Prof. J. Cells from Rana pipiens embryos were incubated in 3H-thymidine for the duration of the last quarter of the S period plus the G2 period of the cell cycle. Chromosomes of animal hemisphere cells of stage 9 embryos showed uniform labeling, whereas chromosomes of endodermal cells of stage 17 embryos showed terminal labeling. We tested whether egg cytoplasm would alter an established temporal pattern of chromosomal DNA replication. Nuclei from disaggregated endodermal cells of stage 17 embryos were transplanted into activated and enucleated eggs. The eggs were then allowed to develop to the blastula stage. Animal hemisphere explants of these blastulae were incubated in 3H-thymidine. Radioautographic localization of silver halide grains demonstrated a chromosomal DNA replication pattern that was uniform over the the metaphase chromosomes. The egg cytoplasm had evidently altered an established temporal pattern of chromosomal DNA replication.  相似文献

The effects of the previously identified mutations in nuclear genes SRM8, SRM12, SRM15, and SRM17on the maintenance of chromosomes and recombinant plasmids in Saccharomyces cerevisiaecells and on cell sensitivity to ionizing radiation were studied. The srm8mutation caused an increase in spontaneous chromosome loss in diploid cells. In yeast cells with the intact mitochondrial genome, all examined srmmutations decreased the mitotic stability of a centromeric recombinant plasmid with the chromosomal ARS element. Mutations srm12, srm15, and srm17also decreased the mitotic stability of a centromereless plasmid containing the same ARS element, whereas the srm8mutation did not markedly affect the maintenance of this plasmid. Mutations srm8, srm12, and srm17were shown to increase cell sensitivity to -rays. The SRM8gene was mapped, cloned, and found to correspond to the open reading frame YJLO76w in chromosome X.  相似文献

Drought‐ and insect‐associated tree mortality at low‐elevation ecotones is a widespread phenomenon but the underlying mechanisms are uncertain. Enhanced growth sensitivity to climate is widely observed among trees that die, indicating that a predisposing physiological mechanism(s) underlies tree mortality. We tested three, linked hypotheses regarding mortality using a ponderosa pine (Pinus ponderosa) elevation transect that experienced low‐elevation mortality following prolonged drought. The hypotheses were: (1) mortality was associated with greater growth sensitivity to climate, (2) mortality was associated with greater sensitivity of gas exchange to climate, and (3) growth and gas exchange were correlated. Support for all three hypotheses would indicate that mortality results at least in part from gas exchange constraints. We assessed growth using basal area increment normalized by tree basal area [basal area increment (BAI)/basal area (BA)] to account for differences in tree size. Whole‐crown gas exchange was indexed via estimates of the CO2 partial pressure difference between leaf and atmosphere (pa?pc) derived from tree ring carbon isotope ratios (δ13C), corrected for temporal trends in atmospheric CO2 and δ13C and elevation trends in pressure. Trees that survived the drought exhibited strong correlations among and between BAI, BAI/BA, pa?pc, and climate. In contrast, trees that died exhibited greater growth sensitivity to climate than trees that survived, no sensitivity of pa?pc to climate, and a steep relationship between pa?pc and BAI/BA. The pa?pc results are consistent with predictions from a theoretical hydraulic model, suggesting trees that died had a limited buffer between mean water availability during their lifespan and water availability during drought – i.e., chronic water stress. It appears that chronic water stress predisposed low‐elevation trees to mortality during drought via constrained gas exchange. Continued intensification of drought in mid‐latitude regions may drive increased mortality and ecotone shifts in temperate forests and woodlands.  相似文献

Biological parameters such as age, growth and age (or size) at maturity are vital for stock assessment and management. Aging is essential in yielding such information. However, limited aging studies have been conducted for large tropical pelagic species in the eastern and central tropical Pacific Ocean. The objective of this study is to conduct a length frequency analysis for estimating growth and mortality of bigeye tuna in the eastern and central tropical Pacific Ocean using samples from the Chinese longline fishery during February to November 2006. The von Bertalanffy growth parameters of asymptotic fork length L and growth coefficient k were estimated at L = 207.4 cm fork length, k = 0.23 year-1, and theoretical age at zero length t 0 = −0.40 year. The total mortality rate (Z) was estimated to be 0.60; the fishing mortality rate (F) and the natural mortality rate (M) were 0.25 year-1 and 0.35 year-1, respectively. The exploitation rate (E) was 0.16. This study provides the estimates of growth and mortality rate for bigeye tuna in the eastern and central tropical Pacific Ocean, which can be used as biological input parameters in further stock evaluations in this region. However, age analysis, further validation of the age composition and stock structure are needed for future studies.  相似文献

Comparative mapping studies of X-linked genes in mammals have provided insights into the evolution of the X chromosome. Many reptiles including the American alligator, Alligator mississippiensis, do not appear to possess heteromorphic sex chromosomes, and sex is determined by the incubation temperature of the egg during embryonic development. Mapping of homologues of mammalian X-linked genes in reptiles could lead to a greater understanding of the evolution of vertebrate sex chromosomes. One of the genes used in the mammalian mapping studies was ZFX, an X-linked copy of the human ZFY gene which was originally isolated as a candidate for the mammalian testis-determining factor (TDF). ZFX is X-linked in eutherians, but maps to two autosomal locations in marsupials and monotremes, close to other genes associated with the eutherian X. The alligator homologue of the ZFY/ZFX genes, Zfc, has been isolated and described previously. A detailed karyotype of A. mississippiensis is presented, together with chromosomal in situ hybridisation data localising the Zfc gene to chromosome 3. Further chromosomal mapping studies using eutherian X-linked genes may reveal conserved chromosomal regions in the alligator that have become part of the eutherian X chromosome during evolution.  相似文献

Unequal crossing-over between sister chromosomes in the process of DNA replication in Escherichia coli leads to the formation of tandem duplications, thus enhancing the activity of certain genes. In conjugational matings between genetically marked E. coli strains, unequal crossing-over leads to the formation of heterozygous tandem duplications. Studying these duplications as model systems allowed the conclusion that unequal crossing-over between direct DNA repeats of sister chromosomes is the main pathway of the formation of selected recombinants in E. coli strains carrying duplications. This was inferred from the data on the segregation of homozygous diploid recombinants by heterozygous duplications. Unequal crossing-over between sister chromosomes occurs as adaptive exchange providing the survival of the greater part of bacterial cells on a selective medium. The known phenomenon of adaptive mutagenesis may also be a consequence of unequal exchanges at the level of DNA mononucleotide repeats.  相似文献
