培养条件对钝顶螺旋藻(Sp)NS-90020脂肪酸组成和含量的影响   总被引:1,自引:0,他引:1  
研究了不同培养条件对钝顶螺旋藻(Sp)NS-90020脂肪酸合成的影响。随着温度升高,其不饱和脂肪酸,γ一亚麻酸(GLA)相对含量降低,总脂肪酸含量升高,当温度为40℃时总脂肪酸和γ-亚麻酸绝对含量都达到最大值,分别为73.4mg/g干重和11.9mg/g干重;当培养基中NaCl浓度高于0.017mol/L时,其GLA相对含量降低,但低于0.0017mol/L时,对其脂肪酸组成无显著影响;氨水使其脂肪酸和GLA绝对含量升高,并在50mgN(NH3·H2O)时达到最大值,分别为67.96mg/g干重和13.63mg/g千重;暗处理92h使其总脂肪酸和GLA绝对含量升高;缺乏Fe2 或Mg2 或Mo2 时,其总脂肪酸和GLA绝对含量降低,而缺乏PO43-时,其总脂肪酸和GLA绝对含量略有升高。  相似文献

红花(Carthamus tinctorious)的子叶能诱志出愈伤组织,并且该愈伤组织能合成生育酚。正相高效液相色谱测定的结果表明,其主要成分是α-生育酚。在MS培养基基础上添加0.1%酪蛋白,并将生长调节剂浓度调整为2,4-D0.30mg/L,6-BA 1.80mg/L,槿使α,β-γ生育酚含量分别达4.1706,0.3120,0.1650μg/g鲜重。  相似文献

重组大肠杆菌生产谷胱甘肽发酵条件的研究   总被引:6,自引:0,他引:6  
研究了重组E.Coli产GSH的发酵条件,重点考察了添加酵母膏、前体氨基酸和ATP的影响。结果发现,前体氨基酸和ATP均能促进胞内GSH的积累,若在发酵0h和12h分别加入20g/LATP和9mmol/L前体氨基酸,则细胞干重和胞内GSH含量可分别比对照提高24%和14倍。应用正交试验得出的针对细胞干重和GSH总量的最佳组合,最大细胞干重和GSH总量比原试验中的最好结果分别提高了10%和26%。在分析了该菌对葡萄糖利用情况的基础上,对该菌进行了指数流加培养,25h细胞干重与发酵液内GSH总量分别达到80g/L和880mg/L,比摇瓶最好结果分别提高了83和46倍。  相似文献

大瓶螺蛋白腺经磷酸盐缓冲液抽提、硫酸铵分级沉淀、SephadexG-100和Sepharose4B凝胶过滤,可获得在不连续PAGE(pH4.3和pH8.9)上显示单一蛋白质染色带的大瓶螺凝集素(AGL).该凝集素对人血红细胞无血型专一性,但对A型血红细胞的凝集作用最强.AGL的血凝活力可被乳糖或半乳糖所抑制.AGL分子中的中性糖含量为0.24mg/mg蛋白质.用SDS-PAGE法测得其亚基分子量为15000,且只有一种亚基.AGL中Cys和Phe的含量较高,并较耐热.  相似文献

本文用中麻黄无菌苗为外植体,其切段培养在附加2mg/L2,4-D和0.5mg/L 6 BA的MS培养基上,全部脱分化形成白色疏松愈伤组织。愈伤组织继代培养于MS+0.5mg/L2,4-D+0.2mg/L6BA+0.2mg/L NAA+4%蔗糖的培养某上。以继代培养愈伤组织为材料进行悬浮培养,培养基为附加0.2mg/L2,4-D+0.1mg/L6BA+0.1mg/LNAA+2%蔗糖的MS液体培养基,得到分散性好,细胞形状接近圆形,细胞大小均一,细胞团多由2-30个细胞组成的悬浮培养体系。第三代悬浮培养细胞增长率为0.35g·fw/20ml·d,细胞有丝分裂指数为11.2%。条件培养和高密度接种可缩短延迟期,条件培养不能提高分裂指数,1g/10ml接种密度可使分裂指数提高至21.2%。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
