Transfer of male golden (Syrian) hamsters from a 14L:10D (light:dark) to a 5L:19D photoperiod induced significant changes in pituitary function tested in vitro. Within 27 days after transfer to a 5L:19D photoperiod, basal prolactin (Prl) release was significantly depressed and response to dopamine (DA) was significantly enhanced as compared to Prl release by pituitaries from 14L: 10D hamsters. Follicle-stimulating hormone (FSH) release tended to be depressed after 9 or 27 days of 5L:19D exposure, but the effect was not significant. After 77 days of 5L:19D exposure, Prl release was further suppressed, while FSH release surpassed that seen in 14L:10D pituitaries. In vitro FSH response to luteinizing hormone releasing hormone (LHRH) was also enhanced at this time. After 15 weeks of exposure to a short photoperiod, FSH secretion was still elevated above control levels, but Prl release and Prl response to DA were no longer different from that of 14L: 10D controls. Secretion of luteinizing hormone (LH) in vitro, either basal or LHRH stimulated, was not affected by photoperiod at any time tested. From these results, we conclude that short photoperiod exposure does not reduce the pituitary's ability to secrete LH or FSH, although secretion of Prl is severely attenuated.  相似文献

The present studies examine the effects of neonatal treatment with monosodium glutamate (MSG) on dopamine (DA), 5-hydroxytryptamine (5-HT) and norepinephrine (NE) metabolism in discrete brain regions and correlate them with steroid receptor kinetics in the anterior pituitary (PIT), preoptic hypothalamus (POA) and caudal hypothalamus (HYP), and with steroid negative and positive feedback effects on luteinizing hormone (LH) secretion. Substantial decreases in the neuronal activity of all three amines in the arcuate nucleus, decreased DA and 5-HT metabolism in the suprachiasmatic nucleus and, surprisingly, increased metabolism of 5-HT and NE in the median eminence was observed in adult ovariectomized (OVX), MSG-treated versus OVX, vehicle-treated litter mate controls. Measurement of estradiol receptors in the nuclear and cytosolic fractions of the POA, HYP and PIT from MSG- and vehicle-treated rats killed during diestrus or 2 weeks after OVX revealed no differences. Similarly, no differences in cytosolic progestin receptors between control and MSG unprimed or estradiol-primed, OVX rats or on progestin receptor translocation induced by progesterone in Eb-primed rats were observed. Negative and positive feedback effects of estradiol or the positive feedback of progesterone on LH secretion were not significantly impaired in MSG rats, and indeed, MSG animals actually were hyper-responsive to the administration of the steroids or of luteinizing hormone-releasing hormone. These results indicate that the MSG-induced damage to DA, 5-HT and NE elements observed within several preoptic and hypothalamic nuclei does not impair estrogen and progestin receptor kinetics, nor does it prevent adequate negative or positive steroid feedback responses, if appropriate steroid regimens are employed, and that the impaired gonadal function reported in these animals does not result primarily from inadequate steroid feedback mechanisms.  相似文献

Effects of age and castration on secretion of luteinizing hormone (LH) and metabolism of hypothalamic monoamines were determined in Holstein bulls. Calves were assigned to be intact or castrated and killed at 8, 12, or 24 wk of age. Animals were castrated and bled every 10 min for 6 h at 96 and 24 h prior to slaughter, respectively. The stalk median eminence (SME), medial basal (MBH), and anterior-preoptic (AHA-POA) hypothalamic regions were obtained at slaughter and assayed for norepinephrine (NE), dopamine (DA), dihydroxy-phenylacetic acid (DOPAC), homovanillic acid (HVA), serotonin (5-HT), and 5-hydroxyindole-acetic acid (5-HIAA) using high performance liquid chromatography with electrochemical detection (HPLC-EC). Concentrations of LH and testosterone in plasma were determined by radioimmunoassay (RIA). In intact calves, LH pulse frequency (pulses/6 h) increased between 8 and 12 wk (1.4 vs. 3.4) and then declined (1.6 at 24 wk of age). Frequency of LH discharges did not change during the first 72 h post-castration in calves 8 (1.4 vs 1.0) and 12 (3.4 vs. 3.8) wk of age, but increased in 24-wk-old calves during this time (1.6 vs. 6.4). The amplitude of LH pulses increased with age (p less than 0.05) and after castration (p less than 0.05). There were marked regional differences in concentrations of monoamines. However, effects of age and castration on concentrations of monoamines were observed only within the SME where DA, DOPAC and NE increased significantly with age. Plasma concentrations of testosterone were correlated with concentrations of NE and DOPAC within the SME. Changes in 5-HT with age were biphasic; at each age, 5-HT increased after castration. From these data, it is concluded that 1) different mechanisms regulate LH pulse frequency and amplitude in calves as early as 8 wk of age, and 2) differences in hypothalamic metabolism of monoamines may be related to maturational changes in secretion of LH in bull calves.  相似文献

Binding of 125I-prolactin (Prl) to hamster ovarian homogenates was found to decrease markedly at the time of the preovulatory gonadotropin surge (PGS). Saturation analysis revealed that the decrease was due to a reduction in the number of available Prl receptors and not due to a change in binding affinity. Loss of Prl receptors following the PGS was not affected by treatment with ergocryptine to block the release of pituitary Prl, indicating that the reduction in the number of available Prl receptors was not due to increased occupancy by endogenous Prl. Loss of Prl receptors was prevented by treatment with phenobarbital (Phen) to block the normal luteinizing hormone (LH)/follicle-stimulating hormone (FSH) surge; whereas, an injection of 50 micrograms of LH or 50 micrograms FSH (but not 100 micrograms Prl) induced a marked decrease in Prl receptors in Phen-treated hamsters. To determine whether Prl receptor loss induced by 50 micrograms FSH might be due to LH contamination, Phen-treated hamsters were injected with minimal ovulatory doses of LH and FSH. Injection of 5 micrograms or 2.5 micrograms LH induced a loss of Prl receptors in 90% and 70% of Phen-treated hamsters, respectively. In contrast, injection of 5 micrograms or 2.5 micrograms FSH induced a loss of Prl receptors in 0% and 20% of Phen-treated hamsters, respectively. These results indicate that the PGS causes an acute heterologous down regulation of ovarian Prl receptors and suggest that this down regulation may be due principally to the action of LH.  相似文献

1. The turnovers of hypothalamic 5-hydroxytryptamine (5HT), dopamine (DA) and noradrenaline (NE) were measured in male and female ring doves (Streptopelia risoria) at three stages of the breeding cycle: courtship, 3 days after pairing; early incubation, 1–2 days after egg laying; and brooding, 1–3 days after the squabs had hatched.2. In both sexes plasma LH decreased progressively from courtship through incubation to brooding young. Crop sacs were fully developed in doves brooding young but not at other stages of the reproductive cycle, indicating increased concentrations of plasma prolactin.3. The turnovers of 5HT and DA in both sexes were significantly higher (P<0.001) in doves brooding young than in birds incubating eggs or nest building. The turnover of DA was higher in females than in males at the onset of incubation. The turnover of NE was lower (P > 0.01) in females at the onset of incubation than during courtship or brooding.4. Increased turnover of hypothalamic DA may be more closely related to brooding behaviour than to changes in prolactin or LH secretion.5. Increased hypothalamic 5HT turnover in brooding doves appears to be more directly related to crop sac development, and by inference increased prolactin secretion, than to depressed plasma LH concentrations.  相似文献

Passive immunization of male lambs against oestradiol-17 beta from 2 to 16 weeks of age significantly elevated androgen concentrations in plasma and depressed the median eminence content of dopamine. Removal of endogenous oestrogens had no significant effects on plasma FSH, LH or prolactin concentrations or on testicular growth and hypothalamic content of GnRH. These results suggest that endogenous oestrogens may indirectly suppress testicular androgen secretion by exerting a stimulatory influence on hypothalamic dopaminergic neurones, which in turn may inhibit GnRH secretion by the median eminence.  相似文献

Autoradiographic histochemistry was employed to examine changes in the binding of 125I-labeled prolactin (Prl) to ovaries from proestrous hamsters before (at 1200 h), during (at 1600 h), and after (at 2000 h) the preovulatory gonadotropin surge. In untreated control hamsters, there was a marked and progressive loss of Prl binding, first in the interstitial cells and follicular thecae by 1600 h, and then in the granulosa cells of the preovulatory follicles by 2000 h. When proestrous hamsters were treated with ergocryptine to significantly lower serum Prl, or injected with exogenous Prl, Prl binding to their ovaries did not differ from controls, suggesting that decreased Prl binding was due to neither increased occupancy of binding sites by endogenous Prl nor down regulation of Prl receptors by Prl itself. Conversely, when proestrous hamsters were treated with phenobarbital to block the luteinizing hormone (LH)/follicle-stimulating hormone (FSH) surge, the loss of Prl binding sites in the ovaries was prevented, suggesting that the LH/FSH surge might initiate a down regulation of Prl receptors in the ovary. Such a down regulation of Prl receptors may serve as a mechanism by which the ability of Prl to affect periovulatory events in the ovary might be regulated.  相似文献

FSH levels begin to rise 3-5 days after male Siberian hamsters are transferred from inhibitory short photoperiods to stimulatory long photoperiods. In contrast, LH levels do not increase for several weeks. This differential pattern of FSH and LH secretion represents one of the most profound in vivo examples of differential regulation of the gonadotropins. The present study was undertaken to characterize the molecular mechanisms controlling differential FSH and LH synthesis and secretion in photostimulated Siberian hamsters. First, we cloned species-specific cDNAs for the three gonadotropin subunits: the common alpha subunit and the unique FSHbeta and LHbeta subunits. All three subunits share high nucleotide and predicted amino acid sequence identity with the orthologous cDNAs from rats. We then used these new molecular probes to examine the gonadotropin subunit mRNA levels from pituitaries of short-day male hamsters transferred to long days for 2, 5, 7, 10, 15, or 20 days. Short-day (SD) and long-day (LD) controls remained in short and long days, respectively, from the time of weaning. We measured serum FSH and LH levels by RIA. FSHbeta, LHbeta, and alpha subunit mRNA levels were measured from individual pituitaries using a microlysate ribonuclease protection assay. Serum FSH and pituitary FSHbeta mRNA levels changed similarly following long-day transfer. Both were significantly elevated after five long days (2.3- and 3.6-fold, respectively; P < 0.02) and declined thereafter, but they remained above SD control values through 20 long days. Alpha subunit mRNA levels also increased significantly relative to SD control values (maximum 2-fold increase after seven long days; P < 0.03), although to a lesser extent than FSHbeta. Neither serum LH nor pituitary LHbeta mRNA levels changed significantly following long-day transfer. The results indicate that long-day-associated increases in serum FSH levels in Siberian hamsters reflect an underlying increase in pituitary FSHbeta and alpha subunit mRNA accumulation.  相似文献

Adult male Syrian hamsters of the inbred LSH/Ss Lak strain were maintained under a 14L:10D light cycle until 13 weeks of age. At this point, they were implanted s.c. with elastomer capsules that were either empty or packed with 30-40 mg of 6-methoxybenzoxazolinone (6-MBOA), a compound found naturally in some monocotyledonous plants; half of the animals from each treatment group were then kept in long days (14L:10D) or transferred to short days (9L:15D). Testicular size was measured and blood samples collected from each hamster immediately before capsule implantation and again 2, 4, 6 and 8 weeks later. Within just 2 weeks of exposure to short days the mean plasma levels of LH and FSH had significantly declined, in both the control and 6-MBOA-treated animals, and were basal within 4 weeks. Testicular size closely followed these gonadotrophin changes; within 4-6 weeks the testes from all of the short-day hamsters had completely regressed to a prepubertal size. At the end of the experiment, at Week 8, the animals were killed and various components of the hypothalamo-pituitary-testicular axis were compared between the treatment groups. The pituitary content of FSH and LH, testicular weight, mean serum level of testosterone, but not hypothalamic LHRH content or pituitary gland weight, were considerably lower in the short-day than in the long-day hamsters, regardless of whether or not they had been chronically treated with 6-MBOA.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The central effects of both an antagonist and an antiserum to substance P (SP) on gonadotropin and prolactin (Prl) secretion were studied in castrated male rats. The lateral ventricular injection (20 micrograms) of an analogue to SP possessing antagonistic properties resulted in significantly suppressed serum LH levels without altering serum FSH and Prl levels when compared with saline-injected control animals. Similarly, the lateral ventricular injection of an antiserum to SP also resulted in significantly suppressed LH levels when compared to control animals injected with normal rabbit serum. Additionally, no changes were observed in the levels of serum FSH and Prl as a result of the anti-SP injection. Thus, although indirect, these results support the hypothesis that SP may have a central stimulatory action on LH secretion, but not FSH and Prl secretion.  相似文献

The effects of artificial photoperiod, temperature, and long-term testosterone treatment on testicular luteinizing hormone (LH) binding were studied in adult male Djungarian hamsters. In hamsters transferred to long-day (LD; 16 hr light, 8 hr dark) photoperiod 8 weeks after adaptation in short-day (SD; 8 hr light, 16 hr dark) photoperiod of 25 degrees C, testicular growth was associated with an increase in the total LH binding per two testes and a decrease in LH binding per unit testicular weight. Plasma testosterone levels reached a peak 47 days after transfer to LD and tended to decrease thereafter, while the testes continued growing. In contrast, when hamsters reared under LD conditions at 25 degrees C for 12 weeks were transferred to SD, testicular regression was associated with a decrease in plasma testosterone and the total LH binding per two testes and an increase in LH binding per unit testicular weight. A significant decrease in LH binding per unit weight compared to SD controls was observed in those hamsters exposed to SD with continuous testosterone treatment. The testosterone treatment tended to induce decrease in the total LH binding. Scatchard plot analyses of the binding suggested that changes in LH binding were due to changes in the number of binding sites. When sexually mature male hamsters were subjected for 8 weeks to two different ambient temperatures (7 degrees C and 25 degrees C) and photoperiods (LD and SD), the difference between the two temperature groups was statistically not significant regarding the weights of testes, epididymides, and prostates; plasma testosterone levels; and LH binding in either LD or SD group. These results suggest that photoperiod is a more important environmental factor than temperature for the regulation of testicular activity and LH receptors and that testosterone reduces the number of LH receptors per unit testicular weight in adult male Djungarian hamsters.  相似文献

—Alterations in whole-brain and hypothalamic levels of serotonin (5-HT), 5-hydroxyindoleacetic acid (5-HIAA), norepinephrine (NE), dopamine (DA) as well as the turnover rates of NE and DA of adult male rats were analysed fluorometrically at either 3 weeks or 6 weeks following castration. Significant increases were observed in whole-brain (minus hypothalamus) 5-HIAA levels and hypothalamic DA levels, fractional rate constants and utilization rates at the 3 but not the 6 week intervals. Elevated levels of 5-HT were observed at both time intervals while an increase in whole-brain DA was seen only at the 6 week interval. Whole brain NE turnover rates of castrated animals did not differ significantly from those of sham-castrate control animals at either test interval. However, a tendency toward increased hypothalamic NE turnover rates was seen in the castrated animals. These biochemical changes resulted in decreased NE/5-HT and DA/5-HT ratios for the castrate rats as compared to controls. The results are discussed in relation to emotional and aggressive behavior and are interpreted as being consistent with the hypothesis purporting an inhibitory role for 5-HT and excitatory role for NE and DA in sex-specific behavior patterns including aggression.  相似文献

Norepinephrine (NE) and dopamine (DA) actively inhibit the release of LH in anestrous ewes. This can be detected as an increase in LH pulse frequency following i.v. injection of NE and DA antagonists. The objective of this study was to determine the sites of these inhibitory actions in the ovine hypothalamus by using local administrations of the NE antagonist, phenoxybenzamine (PBZ), or the DA antagonist, pimozide (PIM), into specific hypothalamic areas. Each neurotransmitter antagonist was administered via a chronically implanted steel guide tube into either the preoptic area (POA), retrochiasmatic area (RCh), or the median eminence region (ME). Blood samples were taken every 15 min for 2 h before and 4 h during implantation of these drugs and were analyzed for LH and prolactin by RIA. Control (no treatment) samples were obtained similarly from the same animals on another day. Placement of PBZ into the POA significantly increased LH pulse frequency and mean LH concentrations over control values whereas PIM did not. In contrast, PIM significantly increased LH pulse frequency and mean LH concentrations when placed in the ME or in the RCh, but PBZ did not. No effects of PIM on prolactin concentrations were detected. These results suggest that an NE neural system operates in the POA and that a DA system acts in the medial basal hypothalamus (RCh or ME) to suppress GnRH pulse frequency in the ovary-intact anestrous ewe.  相似文献

Exposure to short days (SD) induces profound changes in the physiology and behaviour of Siberian hamsters, including gonadal regression and up to 30% loss in body weight. In a continuous SD environment after approximately 20 weeks, Siberian hamsters spontaneously revert to a long day (LD) phenotype, a phenomenon referred to as the photorefractory response. Previously we have identified a number of genes that are regulated by short photoperiod in the neuropil and ventricular ependymal (VE) cells of the hypothalamus, although their importance and contribution to photoperiod induced physiology is unclear. In this refractory model we hypothesised that the return to LD physiology involves reversal of SD expression levels of key hypothalamic genes to their LD values and thereby implicate genes required for LD physiology. Male Siberian hamsters were kept in either LD or SD for up to 39 weeks during which time SD hamster body weight decreased before increasing, after more than 20 weeks, back to LD values. Brain tissue was collected between 14 and 39 weeks for in situ hybridization to determine hypothalamic gene expression. In VE cells lining the third ventricle, expression of nestin, vimentin, Crbp1 and Gpr50 were down-regulated at 18 weeks in SD photoperiod, but expression was not restored to the LD level in photorefractory hamsters. Dio2, Mct8 and Tsh-r expression were altered by SD photoperiod and were fully restored, or even exceeded values found in LD hamsters in the refractory state. In hypothalamic nuclei, expression of Srif and Mc3r mRNAs was altered at 18 weeks in SD, but were similar to LD expression values in photorefractory hamsters. We conclude that in refractory hamsters not all VE cell functions are required to establish LD physiology. However, thyroid hormone signalling from ependymal cells and reversal of neuronal gene expression appear to be essential for the SD refractory response.  相似文献

This study analyzes cadmium effects at the hypothalamic-pituitary-testicular axis. Male rats were given cadmium during puberty or adulthood. Cadmium exposure through puberty increased norepinephrine content in all hypothalamic areas studied, but not in the median eminence. Metal exposure increased serotonin turnover in median eminence and the anterior hypothalamus, while decreased it in mediobasal hypothalamus. Also, decreased plasma levels of testosterone were found. Cadmium exposure during adulthood increased norepinephrine content in posterior hypothalamus and decreased the neuro-transmitter content in anterior and mediobasal hypothalamus. Decreased circulating levels of luteinizing hormone (LH) and testosterone and increased plasma follicle stimulating hormone (FSH) levels were also observed. Cadmium accumulated in all analyzed tissues. Various parameters showed age-dependent changes. These data suggest that cadmium globally effects hypothalamic-pituitary-testicular axis function by acting at the three levels analyzed and that an interaction between cadmium exposure and age emerge.  相似文献

Transfer of gonadally regressed male golden hamsters from a short (5 L:19 D) to a stimulatory (14 L:10 D) photoperiod elicits, within 24 hr, significant changes in hypothalamic dopamine, serotonin, and possibly norepinephrine metabolism. Hypothalamic LHRH content was significantly elevated in short-photoperiod animals, but within 24 hr of transfer to a 14:10 photoperiod, LHRH declined to levels not different from those in hamsters maintained continuously in a long photoperiod. Plasma FSH levels were also significantly elevated within 24 hr of transfer, but increases in plasma LH were somewhat slower. Chronic treatment with the tyrosine hydroxylase inhibitor, alpha-methyl tyrosine (alpha MPT), which inhibits catecholamine synthesis, blocked the effect of a stimulatory photoperiod on plasma FSH levels, while treatment of 5:19 hamsters with the catecholamine precursor, L-dopa, mimicked the effects of photostimulation on plasma FSH levels. Testicular weights were not affected by alpha MPT or L-dopa treatment for 1 week. From these data, it appears that endocrine events associated with photoperiod-induced testicular recrudescence are under the control of hypothalamic neurotransmitters.  相似文献

Siberian hamsters (Phodopus sungorus) exhibit reproductive and immunological responses to photoperiod. Short (<10-h light/day) days induce gonadal atrophy, increase leukocyte concentrations, and attenuate thermoregulatory and behavioral responses to infection. Whereas hamster reproductive responses to photoperiod are dependent on pineal melatonin secretion, the role of the pineal in short-day induced changes in immune function is not fully understood. To examine this, adult hamsters were pinealectomized (PINx) or sham-PINx, and transferred to short days (9-h light/day; SD) or kept in their natal long-day (15-h light/day; LD) photoperiod. Intact and PINx hamsters housed in LD maintained large testes over the next 12 weeks; sham-PINx hamsters exhibited gonadal regression in SD, and PINx abolished this effect. Among pineal-intact hamsters, blood samples revealed increases in leukocyte, lymphocyte, CD62L+ lymphocyte, and T cell counts in SD relative to LD; PINx did not affect leukocyte numbers in LD hamsters, but abolished the SD increase in these measures. Hamsters were then treated with bacterial lipopolysaccharide (LPS), which induced thermoregulatory (fever), behavioral (anorexia, reductions in nest building), and somatic (weight loss) sickness responses in all groups. Among pineal-intact hamsters, febrile and behavioral responses to LPS were attenuated in SD relative to LD. PINx did not affect sickness responses to LPS in LD hamsters, but abolished the ameliorating effects of SD on behavioral responses to LPS. Surprisingly, PINx failed to abolish the effect of SD on fever. In common with the reproductive system, PINx induces the LD phenotype in most aspects of the immune system. The pineal gland is required for photoperiodic regulation of circulating leukocytes and neural-immune interactions that mediate select aspects of sickness behaviors.  相似文献

This study was undertaken to analyze if the effects of subchronic alternating cadmium exposure on pituitary hormone secretion are mediated by changes in dopamine turnover in an age dependent way or are directly correlated to cadmium accumulation at the hypothalamic-pituitary axis. Male rats were treated sc. from day 30 to 60 (prepubertal period) or from day 60 to 90 (adult age) of life, with cadmium chloride (CdCl2) at a dose of 0.5 and 1.0 mg kg–1 bw, every 4th day in an alternate schedule, starting with the smaller dose. Dopamine (DA) turnover, expressed as the ratio of acid 3,3-dihidroxifenil acetic (DOPAC)/DA in various hypothalamic areas, the plasma levels of prolactin, growth hormone (GH) and adrenocorticotropic hormone (ACTH), and cadmium accumulation in the hypothalamus and pituitary were studied. Prepubertal cadmium exposure decreased DA content in all hypothalamic areas studied, although its turnover was not modified. A decrease in plasma ACTH levels with no changes in plasma prolactin and GH levels were found. Cadmium did not accumulate in pituitary while it increased in the hypothalamus. Metal exposure during adulthood decreased DA content in mediobasal and posterior hypothalamus, and its turnover in posterior hypothalamus and median eminence. It decreased plasma prolactin and ACTH levels but not those of GH. Cadmium concentration increased in both hypothalamus and pituitary. These results suggest that cadmium exposure produces age dependent changes on the secretory mechanisms of the pituitary hormones studied, related to the selective accumulation of the metal at both hypothalamic and hypophyseal level changes. However the effects of the metal are not mediated by dopamine.  相似文献

Repeated oral administration of the non-psychoactive cannabinol (CBN; 5 or 50 mg/kg) significantly reduced the concentration of norepinephrine (NE) in median eminence and greatly reduced NE levels 1 and 2 hrs after administration of alpha-methylparatyrosine (alpha-MPT). The levels of dopamine (DA) in median eminence were significantly different, as indicated by the differences in slopes obtained in CBN- treated and control mice before and after alpha-MPT. Plasma levels of luteinizing hormone (LH) were significantly reduced in CBN-exposed mice before alpha-MPT, elevated at 1 hr post-injection, but were also reduced 2 hrs post-injection at 50 mg/kg CBN. Follicle-stimulating hormone (FSH) levels were increased at 1 hr post-alpha-MPT in mice receiving 50 mg/kg CBN. Oral administration of CBN at 50 mg/kg for 4 days enhanced testicular testosterone (T) production in response to intratesticular in vivo injection of 2.5 or 25 mIU human chorionic gonadotropin (hCG). A single oral dose of the psychoactive delta 9-tetrahydrocannabinol (THC) enhanced the production of T 15 min after intratesticular LH (10 ng) injection. However, at 45 or 60 min post-THC treatment, the response to LH was significantly attenuated. These studies demonstrate that both psychoactive and non-psychoactive components of marihuana alter testicular responsiveness to gonadotropins in vivo. These effects may be biphasic, involving stimulation and inhibition of responsiveness, and appear to be correlated with alterations in plasma LH levels. Alterations in plasma gonadotropins may be mediated by cannabinoid effects on catecholamine concentrations in median eminence and THC-induced alterations in testicular responsiveness to gonadotropin probably also involve direct effects of THC at the gonadal level.  相似文献

Axons arising from the dorsolateral pontine tegmentum of the rat were traced in various hypothalamic and limbic nuclei by the electron microscopic degeneration method (0.5-8 day survival times) and by measuring regional norepinephrine (NE) concentrations after 12 days of survival using a radioenzymatic method. Significant reductions (41-85%) in NE contents were observed in the supraoptic, arcuate, basal and lateral amygdaloid nuclei and in the hippocampus 12 days after the bilateral electrolytic lesions of the locus coeruleus. No changes in NE concentrations were observed in the ventromedial, septal, central amygdaloid nuclei, in the median eminence and olfactory tubercle. Parabrachial lesions resulted in a decrease of NE content only in the olfactory tubercle. By means of electron microscopy terminal degeneration was found in the hypothalamic paraventricular, dorsomedial nuclei, in the median eminence, in the bed nucleus of the stria terminalis, in the central, lateral and basal amygdaloid nuclei, in the hippocampus and in the anterior ventral thalamic nucleus.  相似文献
