A study dealing with the marine fungi associated with decaying wood samples in the brackish water mangrove ecosystem and shoreline ecosystem was carried out in south India. A total of 19 marine fungi were isolated from the brackish water mangrove ecosystem. They included 13 Ascomycetes, one Basidiomycete and five Mitosporic fungi. In terms of percent frequency of occurrence, the most frequent species obtained from the brackishwater were the Lignincola longirostris (16.60%) and Savoryella lignicola (12.09%). Nine species were found frequently. Five species were occasionally encountered. Aigialus mangrovei, Aniptodera mangrovei and Halosarpheia marina were the rare species recorded. The average number of isolates per wood sample was 1.53. A total of 27 marine fungi including 15 ascomycetes, one basidiomycete and ten mitosporic fungi were recorded from the shoreline ecosystem. In terms of percent frequency of occurrence, the most frequent species obtained from Kanyakumari were the Arenariomyces trifurcates (13.66%), Corollospora maritima (12.44%), and Cirrenalia pygmea (10.98%). Seven species were found frequently. Fourteen species were occasionally encountered. Three species were found to be rare in occurrence. The average number of isolates per wood sample was 1.21.  相似文献

Preirradiation with 0.05 Gy of X rays 2 months before a second exposure to a mid-lethal dose significantly enhanced the survival rate in both female and male ICR strain mice. The radioresistance was observed between 2–2.5 months after exposure to 0.05 Gy. It did not appear within 1.5 months, and disappeared after 3 months. This radioresistance was induced only by whole-body preirradiation (not by partial irradiation of the head or the trunk). On the other hand, preirradiation with 0.30 Gy as well as 0.50 Gy resulted in radioresistance 2 weeks later, but not 2 months later. The radioresistance was induced by whole-body preirradiation or partial preirradiation of the trunk. No radioresistance was evident after exposure of intermediate preirradiation doses of 0.15 and 0.20 Gy administered before 2 months and 2–5 weeks, respectively. The present and previous results show that the biological effects of ionizing radiation may be distinguished with the following four radiation dose ranges; (1) below 0.025 Gy: no radioresistance after 2 months; (2) 0.05–0.10 Gy: significant radioresistance after 2–2.5 months; (3) 0.20 Gy: no radioresistance after 2–5 weeks; and (4) 0.30–0.50 Gy or more: significant radioresistance after 2 weeks. These results conflict with previous findings of the biological effects of ionizing radiation in which the radiation hazard increases in relation to increasing accumulated doses. Some stimulation, in addition to adaptation, by low dose irradiation may have occurred.  相似文献

The recently discovered hyperthermophilic and radioresistant archaeon Thermococcus gammatolerans is of great interest to compare and contrast the impact of its physiology on radioresistance and its ability to repair damaged chromosomes after exposure to gamma irradiation with radioresistant bacteria. We showed that, in contrast to other organisms, cell survival was not modified by the cellular growth phase under optimal growth conditions but nutrient-limited conditions did affect the T. gammatolerans radioresistance. We determined the first kinetics of damaged DNA recovery in an archaeon after exposure to massive doses of gamma irradiation and compared the efficiency of chromosomal DNA repair according to the cellular growth phase, nutrient availability and culture conditions. Chromosomal DNA repair kinetics showed that stationary phase cells reconstitute disrupted chromosomes more rapidly than exponential phase cells. Our data also revealed that this radioresistant archaeon was proficient to reconstitute shattered chromosomes either slowly or rapidly without any loss of viability. These results suggest that rapid DNA repair is not required for the extreme radioresistance of T. gammatolerans. Angels Tapias and Christophe Leplat contributed equally to this work.  相似文献

Nematode-trapping fungi are ubiquitous in terrestrial habitats in dung, soils, litter and woody debris and they also occur in freshwater, but only one species has been found in marine habitats. The purpose of this study was therefore to investigate whether nematode-trapping fungi occurred in mangrove habitats. To achieve this we assessed the diversity of nematode-trapping fungi on decaying litter from mangroves, freshwater and terrestrial habitats (22 sites) in Hong Kong. Composite samples (n = 1,320) of decaying litter (wood and leaves) were examined and a total of 31 species of nematode-trapping fungi belonging to four genera, Arthrobotrys, Monacrosporium, and Dactylella were recorded. Twenty-nine species reported in this study are new records for Hong Kong and 16 species are new records from mangrove habitats worldwide. Nematode trapping fungi are therefore present in marine environments. Commonly encountered taxa were Arthrobotrys oligospora and Monacrosporium thaumasium which are abundant in all habitats. A. oligospora, M. thaumasium and Arthrobotrys musiformis were frequent (> 10%). Twenty-six species were rare (0.16–9.32%). Species richness and diversity was higher in terrestrial than in freshwater and mangrove habitats (ANOVA, < 0.001). A higher mean diversity was observed on decaying leaves as compared to decaying wood in all habitats (< 0.001). Based on Shannon diversity index, it was also observed that taxa characterized by adhesive nets were more frequent in all habitats. This can be explained by the fact that these taxa may have a better competitive saprotrophic ability which would allow them to compete favourably in nutrient limited environments. Abiotic factors that could be linked to differences in species diversity between decaying wood and leaves are also discussed.  相似文献

Submerged wood, wood trapped amongst rocks, sand-buried wood and exposed roots and branches of shoreline trees were collected from three intertidal beach sites in the Seychelles. These were examined for the presence of marine fungi, the species present identified and their frequency of occurrence noted. Leaves, seaweeds and seagrasses washed up on the seashore, and seafoam, were also examined for filamentous marine fungi. Species present are listed with their frequency of occurrence. Differences in species composition from one substratum to the next were noted, and the results discussed. Sixty-three species were collected: 35 Ascomycotina, 2 Basidiomycotina and 26 Deuteromycotina, all are new records for the Seychelles. The mycogeography and ecological niche of the fungi encountered are discussed.  相似文献

Cryptosporidium parvum is known as one of the most highly resistant parasites to gamma irradiation. To morphologically have an insight on the radioresistance of this parasite, ultrastructural changes in C. parvum sporozoites were observed after gamma irradiation using various doses (1, 5, 10, and 25 kGy) following a range of post-irradiation incubation times (10 kGy for 6, 12, 24, 48, 72, and 96 hr). The ultrastructures of C. parvum oocysts changed remarkably after a 10-kGy irradiation. Nuclear membrane changes and degranulation of dense granules were observed with high doses over 10 kGy, and morphological changes in micronemes and rhoptries were observed with very high doses over 25 kGy. Oocyst walls were not affected by irradiation, whereas the internal structures of sporozoites degenerated completely 96 hr post-irradiation using a dose of 10 kGy. From this study, morphological evidence of radioresistance of C. parvum has been supplemented.  相似文献

Gamma irradiation was investigated as a possible method for disinfestation of huhu beetle larvae, Prionoplus reticularis White, in Pinus radiataD. Don. Larvae of four representative size classes were irradiated at six doses, and the lethal dose (LD99) calculated from mortality data 3 days and 10 days post treatment. All larval size classes showed a similar sensitivity to gamma irradiation and required 3677 Gray (Gy) and 2476 Gy for a LD99 3 and 10 days post-treatment, respectively. The penetration of gamma irradiation into pine wood was found to be lowest in freshly cut logs, and decreased linearly at a rate of 0.698 Gy mm–1 of wood. The penetration was greatest in wood that had been stored for 2 years, and decreased 0.512 Gy mm–1 of wood. These results are likely to be correlated with wood moisture content. Gamma irradiation appears to be a potential alternative method to fumigation for quarantine treatment of P. reticularis.  相似文献

Natural decay resistance of teak wood grown in home-garden forestry and the factors influencing decay resistance were determined in comparison with that of a typical forest plantation. Accelerated laboratory tests were conducted on 1800 wood samples drawn from 15 trees of three planted sites. Analysis of variance based on a univariate mixed model showed that planted site, fungal species, and their interaction terms were important sources of variation in decay resistance. With increasing decay resistance from centre to periphery of the heartwood, radial position was a critical factor and the interaction effect of fungal species × radial position was significant in influencing the durability. No significant differences were found in decay resistance either between the opposite radii or due to the various possible interaction terms of radii with the site, fungal species and radial position. There were significant differences in decay resistance against brown-rot fungi between wet and dry sites of home-garden teak although differences against white-rot fungi were non-significant among the three planted sites. Polyporus palustris was the more aggressive brown-rot fungus than Gloeophyllum trabeum. The higher susceptibility of wet site home-garden teak to brown-rot decay was associated with a paler colour of the wood and lower extractive content.  相似文献

Waterlogged archaeological wood suffers from microbial degradation. Sections from 92 archaeological samples from seven marine and terrestrial sites in Sweden were investigated using light- and electron microscopy to determine the major forms of microbial decay. Decay was related to sample age, wood species and environmental factors. The results confirmed earlier observations that erosion bacteria were the main degraders of archaeological waterlogged soft- and hardwoods, whereas soft rot and tunnelling bacterial decay were less frequently observed. Characteristic slime material, derived from attack by erosion bacteria and left behind in the cell lumen, may affect the penetration of consolidants used to conserve waterlogged archaeological wood. Advanced decay by erosion bacteria had only a slight effect on the original colour, form and appearance of the wood. Different oxygen levels influence the type of microbes and the extent of decay. Examining the decay can reveal a great deal about the past history of a site and provide valuable information to archaeologists.  相似文献

High-resolution nuclear magnetic resonance of carbon 13 was employed to characterize archaeological wood samples from Portuguese medieval dugout canoes. Structural and chemical modifications were assessed by comparing the structural features of archaeological wood samples with modern wood. The main results concern the strong decrease of sugar moiety with a complete disappearance of hemicelluloses. During ageing in water environment, the β-O-4 inter-unit linkages of lignins have not been degraded. The features of the 13C NMR spectra clearly reveal that the products of degradation of the carbohydrates occurring during depletion of sugars were not recombined inside the lignocellulosic matrix. Finally, this article gives a clear illustration that although the water contents of archaeological wood can certainly be used as a good criterion concerning their state of degradation, solid-state 13C NMR lead to unambiguously data as far the structural properties of archaeological waterlogged woods are concerned.  相似文献

Black fungi are among the most resistant organisms to ionizing radiation on Earth. However, our current knowledge is based on studies on a few isolates, while the overall radioresistance limits across this microbial group and the relationship with local environmental conditions remain largely undetermined. To address this knowledge gap, we assessed the survival of 101 strains of black fungi isolated across a worldwide spatial distribution to gamma radiation doses up to 100 kGy. We found that intra and inter-specific taxonomy, UV radiation, and precipitation levels primarily influence the radioresistance in black fungi. Altogether, this study provides insights into the adaptive mechanisms of black fungi to extreme environments and highlights the role of local adaptation in shaping the survival capabilities of these extreme-tolerant organisms.  相似文献

Polymeric alkylpyridinium salts (poly-APS) isolated from the marine sponge Reniera sarai act as antifouling and anticholinesterase agents. They also show moderate haemolytic and cytotoxic activities against different cell lines. The haemolytic activity of poly-APS is due to their detergent-like structure and behaviour in aqueous solutions. In this work, the lytic activity of poly-APS against freshwater and marine algae, and inhibitory effects on wood decay fungi, were investigated. The results show that poly-APS inhibit the proliferation and movements of susceptible algae. Effects of poly-APS were time- and concentration-dependent and differed between various algal species. No growth inhibition effects were observed towards the examined wood fungi.  相似文献

Mineralization of polymeric wood lignin and its substructures is a result of complex reactions involving oxidizing and reducing enzymes and radicals. The degradation of methoxyl groups is an essential part of this process. The presence of wood greatly stimulates the demethoxylation of a non-phenolic lignin model compound (a [O14CH3]-labeled β-O-4 dimer) by the lignin-degrading white-rot fungi Phlebia radiata and Phanerochaete chrysosporium. When grown on wood, both fungi produced up to 47 and 40% 14CO2 of the applied 14C activity, respectively, under air and oxygen in 8 weeks. Without wood, the demethoxylation of the dimer by both fungi was lower, varying between 0.5 and 35%. Addition of nutrient nitrogen together with glucose decreased demethoxylation when the fungi were grown on spruce wood under air. Because the evolution of 14CO2 in the absence of wood was poor, the fungi may have preferably used wood as a carbon and nitrogen source. The amount of fungal mycelium, as determined by the ergosterol assay, did not show connection to demethoxylation. P. radiata also showed a high demethoxylation of [O14CH3]-labeled vanillic acid in the presence of birch wood. The degradation of lignin and lignin-related substances should be studied in the presence of wood, the natural substrate for white-rot fungi.  相似文献

The climbing orchid Erythrorchis altissima is the largest mycoheterotroph in the world. Although previous in vitro work suggests that E. altissima has a unique symbiosis with wood‐decaying fungi, little is known about how this giant orchid meets its carbon and nutrient demands exclusively via mycorrhizal fungi. In this study, the mycorrhizal fungi of E. altissima were molecularly identified using root samples from 26 individuals. Furthermore, in vitro symbiotic germination with five fungi and stable isotope compositions in five E. altissima at one site were examined. In total, 37 fungal operational taxonomic units (OTUs) belonging to nine orders in Basidiomycota were identified from the orchid roots. Most of the fungal OTUs were wood‐decaying fungi, but underground roots had ectomycorrhizal Russula. Two fungal isolates from mycorrhizal roots induced seed germination and subsequent seedling development in vitro. Measurement of carbon and nitrogen stable isotope abundances revealed that E. altissima is a full mycoheterotroph whose carbon originates mainly from wood‐decaying fungi. All of the results show that E. altissima is associated with a wide range of wood‐ and soil‐inhabiting fungi, the majority of which are wood‐decaying taxa. This generalist association enables E. altissima to access a large carbon pool in woody debris and has been key to the evolution of such a large mycoheterotroph.  相似文献

As a part of our ongoing efforts towards finding novel antimycotic agents from marine microflora of the Red Sea, vanillin, 5,7-dimethoxy-4-p-methoxylphenylcoumarin and the new antimycotic compound saadamycin were isolated from endophytic Streptomyces sp. Hedaya48. The producing strain was isolated from the Egyptian sponge Aplysina fistularis and subjected to different UV irradiation doses. A mutant strain Ah22 with 10.5-fold (420 mg/l as compared to 40 mg/l produced by the parental strain) improved saadamycin production was isolated. Production of saadamycin from mutant Ah22 was enhanced to 2.26-fold (950 mg/l) and 2.38-fold (1000 mg/l) under optimized culture conditions in batch culture and bioreactors, respectively. Both saadamycin and 5,7-dimethoxy-4-p-methoxylphenylcoumarin exhibited significant antimycotic activity against dermatophytes and other clinical fungi.  相似文献

The most abundant marine fungi encountered in various regions of the Sea of Japan belong to the generaPenicillium, Aspergillus, Wardomyces, Trichoderma, Chrysosporium, andChaetomium. Facultative marine fungi of the generaScytalidium, Verticillium, andOidiodendron and obligate marine fungi of the genusDendryphiella are much less abundant. The composition of marine sediments and the anthropogenic load on them were found to influence the abundance and species diversity of fungi, as well as the occurrence of fungal strains producing hemolytically active substances. The biodiversity of mycobiota and the abundance of hemotoxin-producing fungi in marine sediments may be used to evaluate the anthropogenic load on marine biocenoses. Hemolytic compounds were produced by 57% of the fungi isolated from marine sediments. The hemolytic activity ofChaetomium spiculipilium was revealed in the fraction of the culture liquid containing extracellular fatty acids and pigments. The fatty acid composition of this marine fungus was determined.  相似文献

Abstract

Charcoal analysis was carried out in two archaeological sites on the north slope of the Somma-Vesuvius volcano, not far from Naples. Both sites were inhabited between the 2nd century AD and AD 472, when a great Vesuvius eruption (so called Pollena eruption) buried them. In both sites, Castanea sativa wood was largely used for architectural structures as well as firewood. Ten 14C dates, spanning between the 1st and the 5th century AD, testify to a continuative use of this wood throughout the life of the sites. The comparison between archaeobotanical and pollen data from other sites in southern Italy suggests that chestnut woods were close to the sites studied and had a restricted spreading, possibly in a refugium area on Vesuvius' foothills. The match between archaeological charcoals and pollen data suggests that the Romans did not spread chestnut cultivation in the entire Campania region and that chestnut probably became a common timber in Campania starting from the 5th–6th century AD.  相似文献
