本研究测定了长臀属(MystacoleucusG櫣nther)中的3个种 :月斑长臀(M.chilopterusFowler)、长臀[M.marginatus(CuvieretValenciennes)]和细尾长臀(M.lepturusHuang)共22个个体的线粒体细胞色素b部分基因片段 ,其序列为408bp,并选用亚科(Acheilognathinae)中华(RhodeussinensisG櫣nther)及亚科(Barbinae)中的云南倒刺(Spinibarbus.denticulatusyunnanensisTsu)作为外群。结果表明 :细尾长臀与月斑长臀、长臀间存在遗传分化 ,分化程度与鲤科其它鱼类相似 ;而月斑长臀和长臀却无法分开。结果提示 ,月斑长臀和长臀很可能为同一个种。此外 ,长臀属与云南倒刺的亲缘关系较近 ,表明有争议的长臀属归入亚科是合理的。  相似文献

Variability of the major noncoding mitochondrial DNA region of char (Salvelinus alpinus complex) was analyzed by the median network method on the basis of the literature data on nucleotide sequences of this region. A significant portion (21%) of the polymorphic sites of char mtDNA was shown to be evolutionarily unstable, which results in mutation homoplasia at these sites. Using median-joining analysis the chars were divided into three phylogeographical groups: Arctic, Beringian, and chars occurring in the vast region from Lake Baikal and Taimyr Peninsula to the Atlantic Ocean. The latter group includes three subgroups (Siberian, Atlantic, and Acadian), which diverged from a common ancestor. In the Arctic group, the branch of Taranetz char S. taranetzi is clearly separated. It diverged from the common ancestor earlier than S. boganidae andS. elgyticus occurring on the Chukotka Peninsula species and S. a. erythrinus occurring in Alaska. The Beringian group, which includes dolly varden char from various regions of Asia and America, is an intermediate between the two char groups. The main problems of char systematics inferred from the major noncoding mtDNA region variability arise from the high level of mutation homoplasia.  相似文献

Restriction enzyme analysis was employed in studying the mitochondrial DNA (mtDNA) ATPase6/ND4L region in several Northeast Asian populations of resident lake char of the genus Salvelinus. On evidence of mitotypes, genetic similarity was assumed for populations of neiva (Ueginskoe Lake), lake resident char from the northern coast of the Sea of Okhotsk (Mak-Mak Lake and Elekchanskie lakes), and dolly varden. Mitotype AAAA proved to be common for these populations. Lake char populations of the Juliet and Maxi lakes (the basin of the Kolyma River) had mitotype DBAA, which is similar to mitotype DBAB observed earlier in Taranetz char from Chukotka. The divergence between mitotypes AAAA and DBAA was estimated at 0.3%. Different origins were assumed for the lake resident char populations from the basins of the Sea of Okhotsk and of the Kolyma River, the former originating in the Pacific and the latter, in the Arctic basins.  相似文献

通过对犬科的赤狐、蓝狐、貉和狼4种的线粒体细胞色素b约372bpDNA片段序列分析,结合GenBank中狗、西门豺和非洲野犬3种的该区段DNA序列的比较,共发现113个核苷酸位点存在变异(约30%)。NJ法构建的分子系统树显示,非洲野犬最先从犬科动物中分化出来;犬属的我狼、狗和西门豺等3种为系统树上独立的一支,且其分歧的时间较赤狐、蓝狐和貉早;赤狐和蓝狐具有较近的亲缘关系。上述结果与形态的观点基本  相似文献

On the basis of comparison of the cytochrome b gene nucleotide sequences from genetic databases, the possible phylogenetic relationships of mitochondrial DNA (mtDNA) among all major lineages of Salmoninae (Brachymystax, Parahucho, Salvelinus, Salmo, Parasalmo, and Oncorhynchus) were examined. Three different phylogenetic methods (UPGMA, NJ, and ML) yielded phylogenetic trees of essentially the same topology: (((Brachymystax, Parahucho), Salvelinus, Salmo), (Parasalmo, Oncorhynchus)). The results obtained using the maximum parsimony method were less clear. Apparently, the divergence of the main salmonid lineages occurred during a relatively short time period; hence, the number of synapomorphs marking the order of their divergence was extremely low. This may account for the relative failure to use the maximum parsimony method of phylogenetic reconstruction. The problem of concordance of mtDNA and species phylogenetic schemes is discussed. Their discrepancy in salmonids may be caused by interspecific introgressive hybridization.  相似文献

采用线粒体细胞色素b(cytochrome b)基因,利用DNA测序的方法研究了雀形目18种鸟类的分子系统发育关系。通过序列分析比对,运用邻接法和最大似然法构建了雀形目18种鸟类的系统发育树,结果表明,燕雀类和类的分歧程度达到了科级水平,建议分别独立成科;长尾山雀可从山雀科中分离出来,单列成科;柳莺归入柳莺亚科(Phylloscopinae),树莺归入大苇莺亚科(Acrocephalinae)。Cytb基因的各主要分支中存在着相对恒定的分子钟,根据鸟类mtDNA的cytb进化速度大约是每百万年1.6%,得到18种鸟的科间分歧时间在10.5百万年左右,科内分歧在9.0百万年左右。  相似文献

为了提供我国多刺蚁属(Polyrhachis)昆虫的分子系统学资料,我们对9种多刺蚁的mtDNA的Cytb部分序列进行测定和分析.在所得465个位点的序列中,变异位点152个,占32.7%,碱基T、C、A和G的平均含量分别为42.0%、19.6%、29.2%和9.2%,A+T平均含量71.2%.在氨基酸组成上,共编码155个氨基酸.碱基替换主要发生在密码子第三位点,转换频率大于颠换频率,转换主要发生在T与C之间,颠换主要发生在T与A.以弓背蚁属(Camponotus)的黄斑弓背蚁(C.albosparsus)和浅毛弓背蚁(C.albivillosus)为外群,用NJ、MP及贝叶斯推论法构建系统发育树.本研究结果与经典分类结果相符:在9种多刺蚁中,Myrmhopla亚属的双齿多刺蚁(P.dives)独立成为一支,分化较早,位于系统进化树的最底部,最早分化出来;而Myrma亚属的亚毛多刺蚁(P.subpilosa)、梅氏多刺蚁(P.illaudata)、警觉多刺蚁(P.vigilans)以及拟梅氏多刺蚁(P.proxima)聚合成一支,位于系统进化树的最顶端,它们的亲缘关系相近,属于较进化的类群.但是Polyrhachis亚属的叶形多刺蚁(P.lamellidens)和Cyrtomyma亚属的结多刺蚁(P.rastellata)的进化关系仍需要进一步探讨.  相似文献

以线粒体DNA细胞色素b基因的全序列作为遗传标记 ,探讨红喉姬普通亚种 (Ficedulaparvaal bicilla)和指名亚种 (F .p .parva)的分类地位。应用Kimura 2 parameter法计算出红喉姬普通亚种个体间的遗传距离为 0 1%~ 0 2 % ,而与指名亚种之间的遗传距离为 6 4 % ,两亚种之间的遗传距离远远大于种间的遗传距离。结合形态学的特征我们认为 :红喉姬指名亚种和普通亚种应为两个独立种 ,分别为红喉姬 (F .albicilla)和红胸姬 (F .parva) ,两者的分化时间为 3 15~ 3 2 5百万年  相似文献

基于线粒体Cytb基因部分序列,以波蛱蝶为外群,采用邻接法、最大简约法和贝叶斯法分别构建了中国线蛱蝶亚科10属25种蝶类的系统发育树,探讨了各主要类群间的系统发育关系。其结果表明,所有线蛱蝶亚科聚为两大枝:第一枝包括环蛱蝶属、菲蛱蝶属、蟠蛱蝶属和缕蛱蝶属,其中缕蛱蝶属与环蛱蝶族亲缘关系较近;第二枝包括丽蛱蝶属、穆蛱蝶属、线蛱蝶属、带蛱蝶属、律蛱蝶属和翠蛱蝶属,其中线蛱蝶属为非单系群,翠蛱蝶属和律蛱蝶属则为单系发生,并构成姐妹群。  相似文献

本研究测定了懒猴属(Nycticebus)D环的部分序列和细胞色素b基因的全序列(1140bp),分析了该属物种之间的系统发育进化关系。在DNA水平上,序列分析结果一致地提供了新的分类学证据:支持Rataiszczak和Groves的观点,即N.intermedus只是N.pygmaeus的成体(Ratajszczak,1998;Groves,1971)。对两种序列的数据做了联合及个别分析,获得相似的系统树,支持懒猴属由两个单系群组成:第一群由N.pygmaeus聚成,第二群由N.coucang聚成。该结果也提供了新的分子遗传证据,支持懒猴属由N.coucang和N.pygmaeus两物种组成。  相似文献

通过雉科虹雉属(Lophophorus)、角雉属(Tragopan)、勺鸡属(Pucrasia)和血雉属(Ithaginis)7种鸟类的细胞色素b(cyt b)基因序列比较,构建的虹雉属及其近缘属的分子系统树表明:①3种虹雉构成一个单系群(monophyletic group),虹雉属与角雉属、勺鸡属构成一个单系群;②虹雉属内分为白尾梢虹雉,以及棕尾虹雉和绿尾虹雉两个演化枝。综合分子系统学、地理分布格局和形态学的证据,推测虹雉属鸟类起源于中国的横断山脉,其中繁衍生活在原地的一枝演化为白尾梢虹雉;另一枝则分别进入喜马拉雅山区(西)和中国西南部(东),向西的演化为棕尾虹雉,向东的则为绿尾虹雉。  相似文献

Twelve of 30 species examined in the ant genus Polyrhachis carry single nucleotide insertions at one or two positions within the mitochondrial cytochrome b (cytb) gene. Two of the sites are present in more than one species. Nucleotide substitutions in taxa carrying insertions show the strong codon position bias expected of functional protein coding genes, with substitutions concentrated in the third positions of the original reading frame. This pattern of evolution of the sequences strongly suggests that they are functional cytb sequences. This result is not the first report of +1 frameshift insertions in animal mitochondrial genes. A similar site was discovered in vertebrates, where single nucleotide frameshift insertions in many birds and a turtle were reported by Mindell et al. (Mol Biol Evol 15:1568, 1998). They hypothesized that the genes are correctly decoded by a programmed frameshift during translation. The discovery of four additional sites gives us the opportunity to look for common features that may explain how programmed frameshifts can arise. The common feature appears to be the presence of two consecutive rare codons at the insertion site. We hypothesize that the second of these codons is not efficiently translated, causing a pause in the translation process. During the stall the weak wobble pairing of the tRNA bound in the peptidyl site of the ribosome, together with an exact Watson–Crick codon–anticodon pairing in the +1 position, allows translation to continue in the +1 reading frame. The result of these events is an adequate level of translation of a full-length and fully functional protein. A model is presented for decoding of these mitochondrial genes, consistent with known features of programmed translational frameshifting in the yeast TY1 and TY3 retrotransposons.Reviewing Editor: Dr. W. 采用PCR技术获得中国鲇形目鱼类11科24属27个代表种类细胞色素b基因1138bp全序列,比较分析了来自北美洲、非洲的部分鲇形目鱼类同一基因序列,并选取脂鲤目、鲤形目和鲱形目鱼类作外类群,采用Bayesian方法和最大简约法(MP)构建分子系统树。结果表明:(1)鲇形目鱼类细胞色素b基因序列中,与脂鲤目、鲤形目以及鲱形目鱼类相比存在3bp的缺失;(2)鲇形目鱼类各科代表种类形成一单系群;(3)两种建树方法均支持铫科、粒鲇科和钝头鮠科形成一单系群;而胡子鲇科、刀鲇科、海鲇科、鮰科、长臀鮠科、鲢科、鲇科、棘脂鲿科、鲿科形成一大的单系群;但鳗鲇科的系统位置两种建树方法没有取得一致结果;而其中长臀鲍科与北美的鮰科形成姐妹群,胡子鲇、鮰科、鲇科、鲿科和鮡科是较明显的单系群。  相似文献

A fragment of the mitochondrial cytochrome b (cyt b) gene of 13 agronomically important plant pathogenic Basidiomycetes was sequenced, including several Puccinia spp., Uromyces appendiculatus, Phakopsora pachyrhizi, Hemileia vastatrix and Rhizoctonia solani. The deduced amino acid sequences (residues 142–266) were used to study the relatedness of these pathogens as compared to other species of the Basidiomycetes, Ascomycetes and Oomycetes. The relatedness was also studied at nuclear level using the Internal Transcribed Spacers (ITS) in the ribosomal DNA. Phylogenic trees were constructed with the maximum parsimony (MP) and the neighbour‐joining (NJ) methods. On the basis of both cytochrome b and ITS sequences, the Puccinia species pathogenic to graminaceous crop plants, such as Puccinia recondita f. sp. tritici, Puccinia graminis f. sp. tritici, Puccinia striiformis f. sp. tritici, Puccinia coronata f. sp. avenae, Puccinia hordei, P. recondita f. sp. secalis and Puccinia sorghi, together with Puccinia horiana from Chrysanthemum, were very closely related to each other, whereas Puccinia arachidis (from peanut) was closely related to U. appendiculatus (from beans) but more distant from the other Puccinia species. Both rusts on soybean (P. pachyrhizi) and coffee (H. vastatrix) were outside the Puccinia cluster. All rusts were separated from other Basidiomycetes such as R. solani and the strobilurin producing species Strobilurus tenacellus and Mycena galopoda. Our results demonstrate that the amino acid sequence of the mitochondrial cytochrome b is a valid tool to study phylogenic relatedness among plant pathogenic Basidiomycetes and supports taxonomic grouping based on morphological structures and host specificity. Because of their high variability, ITS sequences were able to discriminate Puccinia species, which were identical on the basis of the cytochrome b amino acid sequence. Thus, ITS sequences could better show differences among species or within a species, whereas cytochrome b is more suitable than ITS for phyologenic inference at family or genus level. In addition, the sequence data obtained during this study represent essential information for easy isolation of the cyt b gene and detection of point mutations conferring resistance to Qo Inhibitor fungicides that eventually may evolve.  相似文献

新疆3种雅罗鱼线粒体DNA细胞色素b序列的差异与系统进化   总被引:3,自引:1,他引:3  
利用鲤科鱼类线粒体DNA细胞色素b通用引物对分布在新疆的准噶尔雅罗鱼(Leuciscusmerzbacheri)、贝加尔雅罗鱼(L .baicalensis)和高体雅罗鱼(L .idus) 3个鱼种共1 5尾个体的线粒体DNACytb部分核苷酸序列进行了测定,获得1 5条长度为41 3bp的同源基因序列。同源基因序列分析显示,在3种雅罗鱼1 5条mtDNACytb基因片段中,A、T、C、G碱基的平均含量分别是:2 7 4%、2 6 7%、1 7 2 %、2 8 7% ,其中A T含量(5 4 1 % )高于G C含量(4 5 9% ) ;共检测到5 2个突变位点;转换比颠换发生频率高,转换颠换比值(R)是3 5。mtDNACytb的进化速率在3种雅罗鱼间表现出不一致性,贝加尔与高体雅罗鱼、贝加尔与准噶尔雅罗鱼种间遗传距离分别是0 1 2 5 1和0 1 2 61 ,保守性较低;高体与准噶尔雅罗鱼种间的遗传距离是0 0 0 0 7,高度保守。mtDNACytb分子系统树显示,在3种雅罗鱼中贝加尔雅罗鱼独立成一枝,高体雅罗鱼和准噶尔雅罗鱼聚成一类。提示了,在mtDNACytb分子水平上,高体雅罗鱼和准噶尔雅罗鱼的亲缘关系十分相近,贝加尔雅罗鱼与准噶尔雅罗鱼的亲缘关系相距较远。我们对准噶尔、贝加尔和高体雅罗鱼mtDNACytb方面的研究与陈星玉对其骨骼类型的研究结果相吻合。  相似文献

本文测定了来自四川和青海岩羊的细胞色素b 基因全序列(1 140 bp) , 结合从GenBank 中检索获得的山羊、北山羊、绵羊和盘羊4 个近缘种细胞色素b 核苷酸同源区序列进行比较, 分析了碱基组成和变异情况以及核苷酸序列差异。分别采用简约法和距离矩阵法构建分子系统树, 得到了基本相同的拓扑结构, 从分子水平初步探讨了岩羊的系统起源问题。岩羊与山羊属的山羊、北山羊有着比绵羊属的绵羊、盘羊更近的亲缘关系。岩羊与山羊、北山羊的分歧时间大约在3~6 百万年, 而与盘羊、绵羊的分歧时间大约在6~8 百万年。  相似文献

The Gobioninae are a group of morphologically and ecologically diverse Eurasian freshwater cyprinid fishes. The intergeneric relationships of this group are unresolved and the possible monophyly of this subfamily remains to be established. We used complete mitochondrial cytochrome b gene sequences from most genera within the gobionine group, in addition to a selection of cyprinid outgroups, to investigate the possible monophyly of this group and resolve the interrelationships within the group. Our results support the monophyly of the Gobioninae and identify four monophyletic groups within the subfamily; the Hemibarbus group, the Sarcocheilichthys group, the Gobio group, and the Pseudogobio group. The morphologically aberrant genera Gobiobotia, Xenophysogobio and Gobiocypris are included in the Gobioninae, with the latter a sister group of Gnathopogon.  相似文献

对红鲫19尾个体的线粒体tRNA-Thr基因、tRNA-Pro基因和部分控制区的核苷酸序列进行了测定.获得19条长度为836~837bp的同源基因序列,共发现18个变异位点.定义了4种单元型.在红鲫4种单元型中确认了DNA复制终止相关的序列TAS、中央保守区序列(CSB-F、CSB-E和CSB-D)和保守序列CSB1.在18个变异位点中,除一个缺失外,序列变异全部为转换,无颠换发生.4种单元型之间的序列差异在0.1%。1.7%之间.结果显示,红鲫群体具有较高的遗传多样性水平,因而暗示了它具有较强的适应外界环境变化的能力,同时说明它是鱼类遗传育种的较好实验材料.  相似文献
