Isoxaflutole [5-cyclopropyl-4-(2-methylsulphonyl-4-trifluromethylbenzoyl)isoxazole] is a new preemergence herbicide for broad-spectrum weed control in maize. The effect of isoxaflutole on chlorophyll (Chl) and carotenoid (Car) biosynthesis was investigated in cucumber (Cucumis sativus L.) cotyledons. Etiolated tissue was incubated with 5 mM isoxaflutole for 24 h and irradiated (60 mol m-2 s-1). The irradiation for 3 h did not reduce Chl a, Chl b, and Car contents, but after a 28-h irradiation the contents of Chl a and Car decreased by 35 and 15 %, respectively, and the content of Chl b increased by 24 %. Increasing the concentration of isoxaflutole beyond 5 mM resulted in reduction of Chl a (71 %), Chl b (20 %), and Car (31 %) contents. Similarly, increase in irradiance from 60 to 180 mol m-2 s-1 resulted in larger reduction of Chl and Car contents. Exogenously supplied 5-aminolevulinic acid did not reverse the isoxaflutole-inhibited Chl synthesis, whereas an exogenously supplied homogentisic acid lactone reversed the inhibition of pigment synthesis due to isoxaflutole.  相似文献

Long-term (30 d) effects of 100, 200, 300, and 400 mM NaCl on photosystem 2 (PS 2)-mediated electron transport activity and content of D1 protein in the thylakoid membranes of chrysanthemum (Dendranthema grandiflorum) cultured in vitro at low irradiance 20 μmol(photon) m−2 s−1 were investigated. 100 mM NaCl increased contents of chlorophylls (Chl) a and b, carotenoids (Car; xanthophylls + carotenes), and the ratio of Chl a/b, and Car/Chl a+b. However, further increase in NaCl concentration led to the significant reduction in the contents of Chl a, and Chl b, and increase in the ratio of Chl a/b and Car/Chl a+b. NaCl treatment decreased the PS 2-mediated electron transport activity and contents of various thylakoid membrane polypeptides including D1 protein.  相似文献

Changes in various components of photosynthetic apparatus during the 4 d dark incubation at 25°C of detached control and ultraviolet-B (UV-B) treatedVigna unguiculata L. leaves were examined. The photosynthetic apparatus was more degraded in younger control seedlings and for a longer time UV-B treated seedlings than in the older or for a shorter time UV-B treated seedlings. This was shown by determining the losses in chlorophyll (Chl) and protein contents, variable fluorescence yield, photosystem (PS) 2, PS1 and ribulose-1,5-bisphosphate carboxylase (RuBPC) activities, and photosynthetic14CO2 fixation. In contrast, the Car/Chl ratio increased during the dark incubation due to less expressed degradation of Car.  相似文献

Salt Tolerance in Aquatic Macrophytes: Ionic Relation and Interaction   总被引:1,自引:0,他引:1  
Effects of seawater salinity (SWS) and pure NaCl on the intracellular contents of Na+, K+, Mg2+, Ca2+, chlorophylls (Chl) and carotenoids (Car) were studied in three submerged aquatic macrophytes, Hydrilla verticillata, Najas indica and Najas gramenia, which differed in their tolerance to salinity. NaCl resulted in significant increase in Chl/Car ratio in the salt-sensitive H. verticillata and moderately salt-tolerant N. indica, but not in the salt-tolerant N. gramenia. SWS treatment did not result in any significant change in the ratio. The intracellular content of Na+ increased significantly in all the test plants upon exposure to both NaCl and SWS. The content of K+ decreased significantly in these plants upon salinity treatment, except in N. gramenia. The contents of Ca2+ and Mg2+ decreased significantly upon NaCl treatment and remained unchanged or increased upon SWS treatment. No relationship between salt tolerance and K+/Na+ ratio was observed. The maintenance of a minimal level of K+ was observed to be the most probable requirement of salt tolerance in aquatic macrophytes.  相似文献

With the increase in concentration of applied salicylic acid (SA), chlorophyll (Chl) content decreased significantly in both wheat and moong seedlings. Chl a/b ratio decreased significantly only in wheat and remained constant in moong. On the other hand, total carotenoid (Car) content, size of xanthophyll pool, and de-epoxidation rate increased significantly with an increase in SA concentration in both plant species. Hence SA treatment may induce Car biosynthesis in these plant species, but the increase in the xanthophyll pool and de-epoxidation rate indicates that SA may create oxidative stress the degree of which is different in various plants. The photosynthetic pigments of twigs in five tree and shrub species possessing chlorenchyma under a well developed, stomata-less, and highly photon absorptive periderm were analysed and compared to those of the corresponding canopy leaves. We asked whether the unavoidable shade acclimation of corticular chlorenchyma results in photosynthetic pigment complements typically found in shade leaves. As expected, chlorophyll (Chl) a/b ratios in twigs were consistently low. However, carotenoid (Car) analysis did not confirm the initial hypothesis, since twigs generally contained increased Chl-based pool sizes of the xanthophyll cycle components. The contents of photo-selective neoxanthin and lutein were high as well. Yet, -carotene content was extraordinarily low. In addition, twigs retained high pre-dawn ratios of the deepoxidized antheraxanthin and zeaxanthin, although environmental conditions were not pre-disposing for such a state. The unexpected Car composition allows the conclusion that other micro-environmental conditions within twigs (hypoxia, increased red to blue photon ratios, and extremely high CO2 concentrations) are more important than shade in shaping the Car profiles.This revised version was published online in March 2005 with corrections to the page numbers.  相似文献

The effect of aluminium (0.5 –1.0 mM) on contents of phosphorus, pigments, and pigment-protein complexes was studied in soybean (Glycine max Merril.) grown in different nutrient medium with and without P. Increased Al concentrations led to the decrease in the contents of chlorophylls (Chl) a and b, and carotenoids (Car) in soybean leaves, but Chl a/b ratio did not vary significantly. Field experiments with Glycine max (L.) Merr. cv. Ludou 11 and Ludou 4 were conducted to evaluate changes in photosynthetic rate, antioxidative enzyme activity, soluble protein, chlorophyll (Chl) and carotenoid (Car) contents in relation to leaf senescence during seed filling period. Photosynthetic rate, soluble protein content, catalase and peroxidase activities were the highest at 25 days after flowering (DAF). Chl a, Chl b and Car contents reached the maximum at 15 DAF and rapidly decreased after 33 DAF.  相似文献

We examined, under laboratory conditions, the influence of temperature (2 °C vs. 10 °C) on the physiological responses of two aquatic bryophytes from a mountain stream to artificially enhanced UV-B radiation for 82 d. These organisms may be exposed naturally to relatively low temperatures and high levels of UV-B radiation, and this combination is believed to increase the adverse effects of UV-B radiation. In the moss Fontinalis antipyretica, UV-B-treated samples showed severe physiological damages, including significant decreases in chlorophyll (Chl) and carotenoid (Car) contents, Chl a/b and Chl/phaeopigment ratios, Chl a fluorescence parameters Fv/Fm and PS2, electron transport rate (ETRmax), and growth. In the liverwort Jungermannia cordifolia, UV-B radiation hardly caused any physiological change except for growth reduction. Thus, this liverwort seemed to be more tolerant to UV-B radiation than the moss under the specific experimental conditions used, maybe partly due to the accumulation of UV-B absorbing compounds. The influence of temperature on the effects of UV-B radiation depended on the species: the higher the UV-B tolerance, the lower the influence of temperature. Also, different physiological variables showed varied responses to this influence. Particularly, the lower temperature used in our study enhanced the adverse effects of UV-B radiation on important physiological variables such as Fv/Fm, growth, and Chl/phaeopigment ratios in the UV-B-sensitive F. antipyretica, but not in the more UV-B-tolerant J. cordifolia. Thus, the adverse effects of cold and UV-B radiation were apparently additive in the moss, but this additiveness was lacking in the liverwort. The Principal Components Analyses (PCA) conducted for both species with the physiological data obtained after 36 and 82 d of culture confirmed the above results. Under natural conditions, the relatively high water temperatures in summer might facilitate the acclimation of aquatic bryophytes from mountain streams to high levels of UV-B radiation. This may be relevant to predict the consequences of concomitant global warming and increasing UV-B radiation.  相似文献

Gender based differences in response to low temperature stress in leaf chlorophyll (Chl), and carotenoids (Car) contents and chlorophyllase (Chlase) activity were monitored in male (Kapoori Vellaikodi and Madras Pan Kapoori) and female (Bangla Mahoba, Desi Bangla and Kaker) betel vine landraces. Although female plants contained nearly two fold more Chl than male counterparts, the low temperature induced Chl loss was comparable, however, male plants showed higher Chl a/b ratio than females. Chlase activity increased due to cold stress in all the landraces. Male plants always showed higher activities of Chlase, which may be one of the reasons for the rather low Chl contents in male plants.  相似文献

Net photosynthetic rate of yellow upper leaves (UL) of Ligustrum vicaryi was slightly, but not significantly higher than that of green lower leaves (LL). Diurnally, maximum photochemical efficiency of photosystem 2, PS2 (Fv/Fm) of LL did not significantly decline but the UL showed fairly great daily variations. Yield of PS2 of UL showed an enantiomorphous variation to the photosynthetically active radiation and was significantly lower than in the LL. Unlike Fv/Fm, the efficiency of energy conversion in PS2 and both non-photosynthetic and photosynthetic quenching did not differ in UL and LL. Significant differences between UL and LL were found in contents of chlorophyll (Chl) a, b, and carotenoids (Car) and ratios of Chl a/b, Chl b/Chl (a+b), and Car/Chl (a+b). Leaf colour dichotocarpism in L. vicaryi was mainly caused by different photon utilization; sunflecks affected the LL.  相似文献

The rate of accumulation of total chlorophyll (Chl) and carotenoids (Car) of leaves grown under high irradiance, HI (30 and 45 W m–2) was faster than at moderate irradiance, MI (15 W m–2). However, the senescence phase started earlier in the samples and proceeded at a faster rate. Chl a/b and Chl (a+b)/Car values showed faster loss of Chl a (compared to Chl b) and Chl (a+b) (compared to Car) in HI leaves. Protein accumulation and loss were also similar to that of Chl (a+b) content. Increase in Chl fluorescence during the development phase may suggest a gradual change in thylakoid organisation, however, the temporal kinetics were different in HI and MI samples. Increase in fluorescence polarisation during senescence of HI leaves compared to the control (MI) suggests conversion of thylakoid membranes to gel phase. Chloroplasts prepared from HI seedlings showed higher rate of photochemical activities, however, the activity declined earlier and at faster rate compared to the control.  相似文献

Three clones of tobacco transformed with the T-DNA of Agrobacterium rhizogenes Ri plasmid A4b cultivated in vitro on a solid agar medium neither showed pronounced morphological diversity nor significantly differed in chlorophyll (Chl) contents from control plants. However, the transformation caused a 27 to 83 % decay in leaf oxygen evolution and in both ribulose-1,5-bisphosphate carboxylase (RuBPC) and phosphoenolpyruvate carboxylase (PEPC) activities. Therefore, the transformation brought about a reduction of active PEPC as well as activable RuBPC amounts in plant tissues. Individual clones did not mutually differ. In tobacco transformed with A. rhizogenes strain TR101 and grown in soil only the mean leaf area tended to reduce. Chl contents, Chl a/b ratio, oxygen evolution, and activities of both RuBPC and PEPC were insignificantly affected by the transformation.  相似文献

The portable chlorophyll (Chl) meter (CL-01, Hansatech) has been successfully used for a rapid and direct estimation of total Chl content in the leaves of some crops. We compared CL-01 meter readings (Chl value) and Chl contents in leaves of Zea mays, Cucumis sativus, Raphanus sativus, and Ceiba speciosa. At chilling stress, the contents of photosynthetic pigments decreased significantly in maize, but in wheat the contents of chlorophyll (Chl) remained unchanged whereas the contents of total carotenoids (Car) increased. In both species the contents of + carotene and lutein + lutein-5,6-epoxide remained unaffected, but the de-epoxidation state involving the components of the xanthophyll cycle increased. Under chilling stress the photosynthetic electron transport also displayed a general failure in maize but in wheat only photosystem (PS) 2 coupled to the water oxidation complex was inhibited. Moreover, in stressed maize the quinone pool decreased, while the low and high potential forms of cytochrome b 559 increased. In wheat only the contents of cytochrome b 559LP decreased. Peroxidation of acyl lipids in the chloroplast lamellae became more distinct in chilling stressed maize but could also be detected in wheat. Thus in chilling stressed maize prevails an impairment of the acceptor site of PS2 while in wheat photodamage is restricted to the electron donation pathway from water to P680 or to the oxygen evolving complex.  相似文献

The lichens Parmelia quercina, Parmelia sulcata, Evernia prunastri, Hypogymnia physodes, and Anaptychia ciliaris were exposed to ozone (O3) in controlled environment cuvettes designed to maintain the lichens at optimal physiological activity during exposure. Measurements of gas exchange, modulated chlorophyll (Chl) fluorescence, and pigment analysis were conducted before and after exposure to 300 mm3 (O3) m–3, 4 h per d for 14 d. No changes in the efficiency of photosystem 2 (PS2) photochemistry, the reduction state of QA, or the electron flow through PS2, measured by Chl fluorescence, were detected in any of the five lichen species studied. Additionally, neither photosynthetic CO2 assimilation nor xanthophyll cycle activity or photosynthetic pigment concentration were affected by high O3 concentrations. Thus the studied lichen species have significant capacities to withstand oxidative stresses induced by high concentration of O3.  相似文献

Six barley chlorophyll (Chl) mutants, viridis, flavoviridis, chlorina, xanhta, lutea, and albina, differed in the contents of Chl (a+b) and carotenoids (Cars). In accordance with their Chl-deficient phenotype, the Chl a and b and Car contents of mutants decreased from viridis to albina, only xantha had the same or even higher concentration of Cars as the wild type plant. The albina mutant completely lacked and xantha had a significantly reduced photosynthetic activity. We found quantitative differences in protein contents between wild type and mutant plants, with the lowest concentration per fresh mass in the albina mutant. Chl fluorescence analysis revealed that heat-treated barley leaves of both the wild type and Chl mutants had a lower photosystem 2 efficiency than the untreated ones. With 35S-methionine labelling and SDS-PAGE we found that six to nine de novo synthetized proteins appeared after heat shock (2 h, 42 °C) in the wild type and Chl mutants. In albina the expression of heat shock proteins (HSPs) was reduced to 50 % of that in the wild type. Hence mainly albina mutants, with a completely destroyed proteosynthetic apparatus of the chloroplasts, are able to synthesize a small set of HSPs. The albina mutant is a very useful tool for the study of different gene expression of chloroplast and nuclear DNA.  相似文献

为了解竹柏(Podocarpus nagi)的光合特性,以3 a生全绿叶和花叶竹柏为材料,测定其光合色素含量和气体交换参数。结果表明,全绿叶竹柏叶片的叶绿素a、叶绿素b、类胡萝卜素、叶绿素a+b、叶绿素a/b和叶绿素a+b/类胡萝卜素均显著高于花叶竹柏;全绿叶竹柏叶片的初始量子效率、最大光合速率和暗呼吸速率均显著高于花叶,而光饱和点和光补偿点均显著低于花叶;全绿叶竹柏叶片的初始羧化效率、光合速率、CO2饱和点和光呼吸速率均高于花叶,而CO2补偿点低于花叶。2种颜色叶片的气孔导度、蒸腾速率和水分利用效率均随着光合有效辐射的增大而增大,且均表现为全绿叶花叶,而胞间CO2浓度则相反,表现为花叶全绿叶。因此,全绿叶竹柏利用弱光的能力强于花叶竹柏,而花叶竹柏利用强光的能力更强,在园林绿化配置中,可根据2种颜色叶片的光合特性合理配置。  相似文献

Uniform saturating irradiances were created using CuSO4 filtered radiation from stroboscopes. The system was tested using maize leaves treated with diuron. The content of photosynthetic pigments and the ratios between them were studied in 71 species of vascular plants, 17 species of Bryophyta, and 10 species of lichens inhabiting West Spitsbergen. With an increase in the level of organization from lichens to vascular plants, the content of photosynthetic pigments (chlorophylls (Chl) and carotenoids (Car)) increased; the Chl a/b ratios and light-harvesting complex values varied independently of organism taxonomy; the Chl/Car ratio tended to increase. The content of pigments is related to taxon advancement at the level of both divisions and higher plant families. In plants inhabiting Arctic region, the pigment content in primitive species was lower than in more advanced ones. In angiosperms inhabiting Spitsbergen, the amount of Chl was lower than in plants inhabiting other botanicgeographical regions.  相似文献
