Previous studies have shown that Menhaden fish meal, a common ingredient of animal feeds, is frequently contaminated with salmonellae. Animals that eat contaminated feed may become infected. If they, in turn, are eaten by humans, they may be a means by which salmonellae are introduced into the human population. Epidemiological studies of the fish-meal industry were carried out to determine the sources of salmonellae in fish meal and the factors affecting the persistence and survival of salmonellae during the processing of fish meal. Examination of 190 fish immediately after they came from the Gulf of Mexico revealed no salmonellae, but salmonellae were frequently isolated from samples of fish taken from the boats when they arrived at the plants. Salmonellae were also frequently isolated from dockside water at each of the plants. Approximately 50% of the samples taken in the raw fish processing areas were contaminated with salmonellae. The percentage of samples yielding salmonellae decreased progressively through the various sequences of processing, but more than 15% of the samples taken from the finished products were also positive. Salmonellae were isolated from the raw area of the plant most frequently while the plant was operating and less frequently when the plant was idle, whereas in the processing area of the plant the reverse was true. Salmonellae appeared to survive and multiply in the processing area of the plant while the plant was idle, which resulted in contamination of the first portion of each day's production. Salmonellae in the processed fish meal were reduced to nondetectable levels by reprocessing the first 45 min of each day's production.  相似文献

Studies have been conducted over a 2-year period to determine the extent to which a poultry processing plant served as a reservoir of salmonellae reaching the external environment, to examine the question of the importance of salmonellae in the environment, and to consider how best to control the spread of the organisms. The studies have been undertaken at a chicken processing plant handling between 75,000 and 80,000 birds per day. Populations of salmonellae and indicator bacteria were estimated in the raw wastes, through the waste treatment plant, and in the receiving stream waters. The results demonstrate that salmonellae are present in poultry processing wastes in a surprisingly constant relation to fecal coliforms (in excess of 1 Salmonella per 500 fecal coliforms), that serotypes in the environment are constantly changing, and that they may reflect unusual conditions in the processing plant, or a possible source of infection among human and animal residents of the environment. Disinfection of poultry processing wastes is recommended.  相似文献

We describe our experience with the isolation of salmonellae from sewage sludge from four treatment plants in different geographic areas of Ohio. Over 3 years, we isolated salmonellae 50 times from 311 sludge samples. Most isolations were made after enrichment in Selenite broth (BBL Microbiology Systems, Cockeysville, Md.). The largest proportion of isolations came from the plant serving the population of Columbus, a large metropolitan area. A significantly greater number of isolations from this plant were made during the first quarter of the year. Twenty-one different serotypes were isolated, along with five untypable strains. The most frequently isolated serotype was Salmonella infantis. Five of the strains were multiply resistant to antibiotics. We also describe the prevalence of antibodies to salmonellae in members of the families residing on the farms in the study. It was found that antibodies to group C salmonellae predominated.  相似文献

The cultural diffusion of a novel food-finding behaviour, piercing the paper cover of a box containing seed, was experimentally provoked in two flocks of pigeons (Columba livia): a closed aviary flock and an open urban flock. In the urban flock, piercing behaviour spread to 24 previously naive individuals. The rate of cultural diffusion was more rapid in the urban flock than in the aviary. In both situations, many individuals simply fed from the discoveries of birds that pierced, but this effect slowed diffusion less in the open urban flock because experienced piercers left the flock and naive birds entered it. Individual trial-and-error learning and natural shaping can be ruled out as mechanisms of diffusion in the present case.  相似文献

"Further-processed" turkey products, prepared from chilled, eviscerated, and thawed carcasses at two commercial turkey-processing plants, were evaluated, for the presence of salmonellae. These organisms were isolated from swab samples from 12% of chilled, eviscerated turkey carcasses, 27% of finished products, and 24% of processing equipment. The same serotypes as those found throughout a plant on any one visit were recovered from 31% of rinse-samples taken from hands and gloves of processing personnel. Salmonellae were found in samples taken on 37 of 48 visits; a greater number of recoveries were made on days when freshly killed turkeys were processed (87%) than when frozen-defrosted carcasses were processed (59%). The predominant serotype isolated from meat and environment usually changed from visit to visit. Salmonella sandiego and Salmonella anatum were the most frequent among 23 serotypes recovered. Most of the isolated serotypes are commonly associated with turkeys and have been incriminated as causative agents of human salmonellosis. The implication is that further-processed turkey products, if inadequately cooked by the consumer and if improperly refrigerated between the time of manufacture and consumption, could directly transmit salmonellae. These same products might also contaminate other foods by introducing salmonellae into food-preparation areas.  相似文献

Salmonella contamination of swine and morbidity rates among the workers of swine-breeding complexes and the members of their families, as well as among the population inhabiting the zone of possible influence rendered by such complexes on the environment, have been studied as exemplified by 4 complexes for large-scale swine breeding, differing in their technology of swine raising and fattening, their systems of the purification and utilization of manure-containing sewage. Besides, the influence of the raw meat (pork) and meat products, contaminated with salmonellae, sold in town, on salmonellosis morbidity among the urban population has been analyzed. The study has revealed that salmonellae are spread from swine-breeding complexes with the fluid and solid fractions of manure-containing sewage, as well as with the animals supplied to the meat-packing plant; the contamination of the workers of swine-breeding complexes with salmonellae becomes possible due to the low culture of the production and to the breach of sanitary and epidemic-control regulations and the contamination of the population occurs through meat and meat products.  相似文献

A study was conducted to determine the incidence of fecal coliforms, fecal streptococci, Aeromonas hydrophilia, and Salmonella spp. in the waste discharges of seven sewage treatment plants, four fruit and vegetable canneries, a meat packing plant, a poultry processing plant, and a potato processing plant located along the Cornwallis River in Nova Scotia, Canada. Surface water samples were also collected from 13 locations in the river to assess the impact of these waste discharges on the receiving water quality. The results showed that the final effluents from most of the sewage treatment and processing plants were of very poor bacteriological quality, with the number of indicator bacteria comparable with those found in raw sewage. Salmonellae were isolated from the effluents of the meat and poultry plants and five of the seven sewage treatment plants surveyed. No salmonellae were detected in the effluents of the fruit and vegetable canneries. The impact of the discharge of untreated municipal and food processing wastes on the Cornwallis River water quality was evidenced by the recovery from river water of five Salmonella serotypes, and the high fecal coliform counts which exceeded recommended limit for bathing and shellfish harvesting.  相似文献

Immunomagnetic separation with additional enrichment was used in conjunction with improved selective media to improve the isolation of salmonellae from raw sausages. The isolation rate achieved was almost double that of conventional culture with no increase in processing time. The selective media gave an overall specificity of approximately 74%; all false-positive pick-offs being identified as Citrobacter freundii . It is believed that this method represents a significant advance in the isolation of salmonellae from foods, although the ideal media both for enrichment and selection have yet to be found.  相似文献

Investigations of a free-range broiler flock during the rearing period and at the slaughterhouse by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) of the flagellin (flaA) gene (flaA typing) have shown that poultry carcasses are contaminated by Campylobacter spp. strains which were previously present in the poultry faces. Moreover, the investigation of the previous and the following batches in the processing plant using flaA typing have shown that cross-contamination between batches coming from different flocks occurs and is also a risk factor for the presence of Campylobacter spp. on poultry carcasses.  相似文献

Survival of Salmonella senftenberg 775W, S. anatum, and S. typhimurium during exposure to currently practiced, as well as abusive, pecan processing and storage conditions was studied. Thermal treatments normally carried out during the processing of pecans are inadequate to consistently destory salmonellae in highly contaminated inshell nuts. Pecan nut packing tissue was toxic to salmonellae, thus affording some protection against high initial contamination and subsequent survival of the organisms. Examinations of inoculated inshell pecans stored at -18, -7, 5, and 21 C for up to 32 weeks revealed that the extent of survival was inversely correlated to the storage temperature. S. senftenberg 775W and S. anatum were not detectable on inshell nuts after 16 weeks of storage at 21 C. Little decrease in viable population of the three species was noted on inoculated pecan halves stored at -18, -7, and 5 C for 32 weeks. Due to organoleptic quality deterioration in pecan nutmeats at elevated temperatures, sterilization methods other than thermal treatment appear to be required for the elimination of viable salmonellae from pecan nuts.  相似文献

This study evaluated the potential for conversion of Class B to Class A biosolids with respect to salmonellae and fecal coliforms during solar drying in concrete lined drying beds. Anaerobically (8% solids) and aerobically (2% solids) digested Class B biosolids were pumped into field-scale drying beds, and microbial populations and environmental conditions were monitored. Numbers of fecal coliforms and salmonellae decreased as temperature and rate of desiccation increased. After 3 to 4 weeks, Class A requirements were achieved in both biosolids for the pathogens and the indicators. However, following rainfall events, significant increase in numbers was observed for both fecal coliforms and salmonellae. In laboratory studies, regrowth of fecal coliforms was observed in both biosolids and biosolid-amended soil, but the regrowth of salmonellae observed in the concrete-lined drying beds did not occur. These laboratory studies demonstrated that pathogens decreased in numbers when soil was amended with biosolids. Based on serotyping, the increased numbers of salmonellae seen in the concrete lined drying beds following rainfall events was most likely due to recolonization due to contamination from fecal matter introduced by animals and not from regrowth of salmonellae indigenous to biosolids. Overall, we conclude that the use of concrete-lined beds created a situation in which moisture added as rainfall accumulated in the beds, promoting the growth of fecal coliforms and salmonellae added from external sources.  相似文献

Campylobacter isolates from diverse samples within broiler production and processing environments were typed by using flaA short variable region DNA sequence analysis. Sixteen flocks from four different farms representing two broiler producers in Arkansas and California were analyzed. Fourteen of the flocks (87.5%) were Campylobacter-positive; two remained negative throughout the 6-week rearing period. In general, multiple clones were present within a flock. Additionally, clones found within a flock were also present on the final product, although the diversity of Campylobacter spp. on the final product appeared to be reduced relative to that observed within the flock. Comparison of clones between flocks on the same farm revealed that some clones of Campylobacter persisted in multiple flocks. Furthermore, some clones were identified across the two farms that were under the same management. In two sampling periods, environmental isolates were positive for Campylobacter prior to flock shedding. Environmental samples associated with five additional flocks were positive for Campylobacter concomitantly with recovery of Campylobacter from the birds. Analysis of the environmental isolates that were positive prior to flock shedding demonstrated that in some instances the environmental isolates possessed genotypes identical to those of isolates originating from the flock, while in other cases the environmental isolates possessed genotypes that were distantly related to isolates obtained from the flock. Analyses of environmental isolates that tested positive concurrently with the positive isolates from the flocks demonstrated varied results; in some instances the environmental isolates possessed genotypes identical to those of isolates originating from the flock, while in other cases the environmental isolates possessed genotypes that were distantly related to isolates obtained from the flock. These data suggest that the external environment may contribute to Campylobacter contamination during poultry production and processing. However, environmental contamination with Campylobacter does not appear to be the sole contributing factor.  相似文献

AIMS: The objectives of the study were to determine the spread and persistence of Campylobacter in a poultry processing plant and to provide a quantitative estimate of the survival of Campylobacter jejuni on the surface of a cutting board. METHODS AND RESULTS: Several contact surfaces in a poultry processing plant were sampled before the start of processing, after 30 min and after 120 min. Next, the survival of four C. jejuni strains was studied on a beech and polypropylene cutting board during 120 min. CONCLUSIONS: A rapid introduction and spread of Campylobacter in a well cleaned processing plant as well as a significant survival in time on the example of a cutting board is shown. SIGNIFICANCE AND IMPACT OF THE STUDY: The need to prevent cross-contamination in the food processing and preparation area and the importance of an integrated approach throughout the whole food chain to control transmission of Campylobacter is highlighted.  相似文献

A phage-typing technique for salmonellae is described. One battery of phages was used to type three serotypes of Salmonella, namely, S. typhimurium, S.typhimurium var. copenhagen, and S. heidelberg. In all, 443 S. heidelberg cultures were typed into 22 phage types, 185 S. typhimurium cultures into 35 phage type, and 92 S. typhimurium var. copenhagen cultures into 26 phage types. The stability of the phage types was established by retyping 168 cultures belonging to all three serotypes. The epidemiological significance of the phage types demonstrated was evaluated by comparing phage types obtained from the University of Minnesota and those from the National Animal Disease Laboratory. Further investigation of the S. heidelberg phage types has shown that the cultures represented repeated isolates from the same birds or from a group of birds in the same flock.  相似文献

Besides the 'classical' animals known as reservoirs for Salmonella spp., like poultry and cattle, reptiles have emerged as a significant source of human salmonellae infections during the last years. Reptile-associated salmonellae frequently cause severe clinical courses including fatalities due to septicaemia and meningitis. Therefore, it is of major priority to develop measures which may help preventing cases of reptile-associated salmonellae. However, as a first step the epidemiology of salmonellae in reptiles must be understood. Therefore, in this study a population analysis of the salmonellae of two female snakes was performed and the pattern of inheritance of salmonellae to their offspring was investigated. It is demonstrated that adult snakes usually harbour a population of concurrent salmonellae serovars. Colonization of their offspring during pregnancy and birth is a significant way of transmission causing 65% of the newborn to be positive for salmonellae. The effectiveness of the transmission does not seem to be due only to the frequency of a certain serovar, because the most prevailing strain of one female snake was not detectable in any of her offspring.  相似文献

Survival of salmonellae in artificially contaminated beef-pork mixtures (approximately 10(4) salmonellae/g) was studied in pepperoni prepared by either a natural flora or lactic starter culture fermentation or in nonfermented sausages. The pepperoni did not become salmonellae free during the usual commercial 15 to 30-day drying period. Salmonella dublin was present in all products, fermented or unfermented, after 42 to 43 days of drying. At a lower level of contamination, 10(3)/g, S. dublin could not be recovered from starter culture-fermented pepperoni after 14 days of drying but persisted in the natural flora-fermented sausage. S. typhimurium (initial count, 10(4)/g) was absent after 42 days of drying when starter culture was used to ferment the pepperoni, but was still present in the natural flora-fermented and unfermented products. S. dublin, host adapted to cattle, or S. choleraesuis, host adapted to swine, had similar survival patterns in beef pork, or beef-pork pepperoni. Heating salmonellae contaminated beef-pork pepperoni (after fermantation but before drying) to an internal temperature of 60 C (trichinae inactivating) eliminated the food-borne pathogen from the sausage product.  相似文献

S ummary : The direct and indirect fluorescent antibody technique (FAT) were compared with cultural methods for detecting salmonellae in meat products, animal feedingstuffs, poultry carcase swabs, giblets and poultry plant and equipment swabs. Salmonellae were not isolated from meat products and fluorescent cells were not seen on slides prepared by either FAT. The indirect and direct FAT recorded 13% and 9% respectively, false positive results, with samples of animal feedingstuffs, but the direct FAT recorded a single false negative result. Salmonellae were not isolated from poultry carcase swabs but 3% and 4·5% respectively, of false positive results were obtained with the indirect and direct FAT. Salmonellae were isolated from both giblet samples and poultry plant swabs and both gave rise to false negative FAT results. Preliminary studies of the efficacy of the FAT for screening animal faecal material for salmonellae indicated that no single combination of enrichment broth and FAT gives unequivocal results, but the staining of smears from tetrathionate broth by either FAT gives rise to a high percentage of false negative results.  相似文献

Composted sewage sludge may be used to improve soil quality, but there remains some doubt concerning the microbiological safety of the product. Sewage sludge composts from 30 municipalities were sampled, and four samples (12%) contained salmonellae (two contained fewer than 0.3/g, and the other two had 21/g and 1.7 X 10(4)/g). All 30 composts were inoculated with salmonellae; the populations decreased at a specific death rate of about 0.15 h-1 over 24 h at 36 degrees C. In irradiation-sterilized composts inoculated with salmonellae, the salmonellae grew at a rate of 0.65 doublings per h for over 24 h. Growth and death rates were found to be moisture and flora associated. The growth or death rates for antibiotic-resistant salmonellae were not different from those of nonresistant strains. It was concluded that the active indigenous flora of compost establishes a homeostatic barrier to colonization by salmonellae, and in the absence of competing flora, reinoculated salmonellae may grow to potentially hazardous densities. The active microflora of moist composts eliminated contaminating salmonellae (10(5)/g) after 6 weeks.  相似文献
