The anoxygenic phototrophic bacterial community of the brackish meromictic Lake Shira (Khakassia) was investigated in August 2001, July 2002, and February–March 2003. In all the periods of investigation, the prevailing microorganisms were purple sulfur bacteria similar to Lamprocystis purpurea in morphology and pigment composition. Their highest number (3 × 105 cells/ml) was recorded in July 2002 at the depth of 15 m. According to 16S rRNA gene analysis, the strain of purple sulfur bacteria isolated in 2001 and designated ShAm01 exhibited 98.6% similarity to the type strain of Thiocapsa roseopersicina and 97.1–94.4% similarity to the type strains of Tca. pendens, Tca. litoralis, and Tca. rosea. The minor microorganisms of the anoxygenic phototrophic bacterial community within the period of investigation were nonsulfur purple bacteria phylogenetically close to Rhodovulum strictum (98.3% similarity, strain ShRb01), Ahrensia kielensis (of 93.9% similarity, strain ShRb02), Rhodomicrobium vannieli (of 99.7% similarity, strain ShRmc01), and green sulfur bacteria, phylogenetically close to Chlorobium limicola (of 98.7% similarity, strain ShCl03). 相似文献

The results of the first methodical investigation into the aerobic methanotrophic communities inhabiting the bottom sediments of Lake Baikal are reported. Use of the radioisotopic method revealed methane consumption in 12 10- to 50-cm-long sediment cores. The maximum methane consumption rates (495-737 microl/(dm3 day) were recorded in sediments in the regions of hydrothermal vents and oil and gas occurrence. Methane consumption was most active in the surface layers of the sediments (0-4 cm); it decreased with the sediment depth and became negligible or absent at depths below 20 cm. The number of methanotrophic bacteria usually ranged from 100 to 1000 cells/cm3 of sediment and reached 1 million cells/cm3 in the regions of oil and gas occurrence. The 17 enrichment cultures obtained were represented mainly by morphotype II methanotrophs. Phylogenetic analysis of the enrichment cultures in terms of the amino acid sequence of the alpha subunit of the membrane-bound methane monooxygenase revealed the predominance of methanotrophs of the genus Methylocystis. The results obtained suggest the presence of an active aerobic methanotrophic community in Lake Baikal. 相似文献

The postglacial sediments of Kortowskie Lake contain well preserved chrysophycean cysts. Forty-six morphotypes were identified by scanning electron microscopy (SEM) and light microscopy (LM). Ten new morphotypes of fossil chrysophycean cysts are described. Some of the most abundant taxa show clear trends associated with the increase in lake trophic status. The cyst flora is divided into four groups: eutrophic forms (Cysta globata, C. cortoviensis, C. vermicularis); oligotrophic forms (Cysta curvicollis); cool-water forms (Cysta carinifera, C. crassicollis, C. microspinosa, C. modica and Cysta stellata), and indifferent forms (all other morphotypes which did not show a clear tendency). The percentage abundance of chrysophycean cysts wa related to climate changes, showing a considerable increase during the Subboreal period. Two factors, lake fertility and water level changes affected the abundance of the fossil cyst flora in the sediments of Kortowskie Lake. 相似文献

The specific features of the distribution, abundance, and taxonomic diversity of bacteria oxidizing iron and manganese in
the bottom sediments sampled from different areas of Lake Baikal are studied. These bacteria are widespread in Baikal bottom
sediments. Cultivated iron- and manganese-oxidizing microorganisms are ascribed to six genera: Metallogenium, Leptothrix, Siderocapsa, Naumaniella, Bacillus, and Pseudomonas. The surface layers of the ground and the border between oxidized and reduced sediments serve as ecological niches for iron-
and manganese-oxidizing microorganisms. Redox conditions of the environment and the availability of dissolved forms of iron
and manganese, as well as of organic matter, which are controlled by the conditions of sedimentation in the lake, are the

This paper is an overview of Russian literature dealing with the accumulation, the transformations and the release of phosphate and nitrogen compounds in a great number of Russian lakes and reservoirs. A considerable data bank has been analysed. Special attention is given to the relations of N- and P-accumulation with the input and transformation of organic carbon, as well as to the release mechanisms, often in relation to eutrophication of the lakes and reservoirs. It is shown that the major input of organic matter into the sediments comes from autochthonous material, and is usually > 70 %. The relative importance of phytoplankton and macrophytes as sources of organic matter is discussed; it appears that trophic state, depth and other factors may have a large influence on this ratio. In shallow eutrophic lakes macrophytes may be the source of organic matter, which source can amount to 1.5–2.5 times that of phytoplankton. It is also shown that the C/N ratio is not a good indicator of the source of the organic matter, because their C/N ratios often are not very different. The decomposition rate of organic matter was analysed; it depends on trophic state and other factors. Sediment N accumulation is mostly (> 90%) in organic form, and depends on nitrogen and organic matter inputs coming from phytoplankton or macrophytes. A correlation coefficient of 0.9–0.95 was found in 176 lakes. In 113 lakes the N accumulation was 0.11 x C accumulation, with C/N ratios between 7.4 and 12.9. Ammonification was rather constant in different groups of lakes; values were often about 20–25 mg m−2 d−1. The presence of the different forms of nitrogen in interstitial water and in adsorbed forms is discussed. The N in interstitial water is usually in the form of NH3. Sediment P-accumulation is usually in inorganic form and is related to primary production. Three different groups of sediments could be distinguished with C/P ratios of 31–100, of 101–350 and > 350. In hard water lakes P sedimentation was found to be 0.3–0.5 times that in soft water lakes with comparable primary production. The relative occurrence of apatite, non-apatite and residual P in sediments was calculated. In the interstitial water the P concentration appeared to be controlled by the input and decomposition of organic matter. The concentration of phosphate dissolved in the interstitial water of the top 2 cm layer is often 10–100 times lower than that of the dissolved N. The concentrations of interstitial phosphate are from a few μgl−1 up to 15 mgl−1, but the higher concentrations occur only rarely. Different types of vertical profiles of P compounds in the sediments were shown to be related with the presence of an oxidised zone, the presence of clay etc. Autochthonous apatite and non-apatite phosphates are more mobile than the allochthonous ones and are in equilibrium with interstitial phosphate. Accumulation of autochthonous apatite in sediments is controlled by decomposition of organic matter and accumulation of carbonates. 相似文献

Methods for cultivation of microorganisms oxidizing iron and manganese in bottom sediments of Lake Baikal were tested. Bacteria were grown on selective media with a characteristic accumulation of metal oxides. The morphology and developmental patterns of the isolated microbial group were studied. The studied sediment core was used to isolate 42 strains of bacteria oxidizing iron and manganese. The cultivated bacteria were confined to the upper sediment layers (down to 11 cm) and their abundance depended on sediment type and redox conditions. 相似文献

The spatio-temporal organization of the bacterial community inhabiting the chemocline of the stratified meromictic Lake Shunet (Khakassia, Russia) was investigated from May to September 2005 by means of microscopy, analysis of photosynthetic pigments, and PCR-DGGE with subsequent 16S rDNA analysis. The samples were collected with a multisyringe stratification sampler, sampling being performed every 5 cm. It was demonstrated that, during the period of investigation, there were no large changes in the bacterial community of the chlemocline, at least among the detected forms. During the whole period of study, purple sulfur bacteria related to Lamprocystis purpurea (Chromatiaceae) were predominant in the chemocline. Beneath the layer of purple bacteria, green sulfur bacteria were revealed that were phylogenetically distant from strain ShNPel02, which was previously isolated from this lake. Development of phytoflagellates of the genus Cryptomonas was observed in the upper zone of the chemocline. In the chemocline of Lake Shunet, the numbers of picoplankton cyanobacteria of the genus Synechococcus increased from May to September. It was demonstrated that the application of universal bacterial primers for DGGE resulted in the same qualitative distributional pattern of predominant species as microscopic studies. 相似文献

The remains of all the groups of invertebrates in a 131 cm long sediment core taken from the main basin of the lake and corresponding to about 600 years have been examined and counted. The middle horizons (90–40 cm) are dominated by Testacida. Deeper layers and those closer to the surface are dominated by Cladocera. Among the Cladocera, planktonic forms dominate at all depths of the sediment. The present zoocenosis has existed for about 400 years. A 50 cm core from the lake bay has also been studied. Cladocera dominate at 50–40 cm and near the surface. Porifera spicules are abundant at 20–10 cm while Protozoa are numerous at 30 cm. Among the Cladocera, planktonic forms are less important than in the main lake basin. 相似文献

Massive parallel sequencing (the Roche 454 platform) of the 16S rRNA gene fragments was used to investigate microbial diversity in the sediments of the Posolsk Bank cold methane seep. Bacterial communities from all sediment horizons were found to contain members of the phyla Actinobacteria, Bacteroidetes, Deinococcus-Thermus, Firmicutes, Nitrospirae, Chloroflexi, Proteobacteria, and the candidate phyla Aminicenantes (OP8) and Atribacteria (OP9). Among Bacteria, members of the Chloroflexi and Proteobacteria were the most numerous (42 and 46%, respectively). Among archaea, the Thaumarchaeota predominated in the upper sediment layer (40.1%), while Bathyarchaeota (54.2%) and Euryarchaeota (95%) were predominant at 70 and 140 cm, respectively. Specific migration pathways of fluid flows circulating in the zone of gas hydrate stability (400 m) may be responsible for considerable numbers of the sequences of Chloroflexi, Acidobacteria, and the candidate phyla Aminicenantes and Atribacteria in the upper sediment layers and of the Deinococcus-Thermus phylum in deep bottom sediments. 相似文献

It is known that the conventional microbiological methods of investigation reveal not more than 1% of the species of microorganisms actually inhabiting one biotope or another. The most complete characterization of the microbial community is achieved by investigating DNA isolated from the samples differing from each other in ecological conditions. The sediments of Lake Baikal are characterized by a high content of humic compounds which hinders molecular biological analyses. Because of this, the procedure of DNA isolation is a step o critical importance for the analysis of bottom sediments. In the present work we compare the methods of isolation of the whole bacterial DNA from the bottom sediments of Lake Baikal. Approximately 14 million people get their drinking and industrial water from the most southern part of Lake Biwa. This shallow region, however, has been eutrophicated severely from year to year by nutrients loading from both the surrounding inflowing rivers and the north basin. Sedimented solids of the surface layer of bottom sediments are stirred up very frequently by strong winds and waves in this shallow region. Moreover, the water quality has been deteriorated by the release of nutrients from the surface layer of bottom sediments in which organic matter has accumulated. For the period 1989 to 1990, the suspended solids content had a close correlation with the COD value, and the T-N and T-P concentrations in the water of inflowing rivers to the basin. T-N release rate from the surface layer of bottom sediments ranged from 6.3 to 213.8 mg m-2 d-1 and the release load was 936 kg d-1 for the entire 58 km2 of the south basin. On the other hand, T-P release rate ranted from 0.91 to 3.56 mg m-2 d-1 and the total release load was approximately 69 kg d-1 in the basin. Suction dredging of organic matter rich sediments was carried out in the basin to improve the water quality. Removed sediments were used after processing for various kinds of construction materials, such as bricks for pavement, walls of domestic houses, and ornamental materials of public buildings. 相似文献

The total number and morphological and size composition of viriobenthos, number of bacteria infected by viruses, and burst size, as well as virus-induced mortality, abundance, and production of bacteriobenthos, have been estimated in bottom sediments of the Rybinsk Reservoir. The total number of viriobenthos in the reservoir varies within (1.1–10.9) × 109 (on average, (5.9 ± 0.6) × 109) particles/cm3; the total number of virus-to-bacteria ratio ranges within 0.2–2.1 (on average 0.9 ± 0.3). A weak positive correlation is found between the abundance of benthic viruses and the abundance and production of benthic bacteria. In most surveyed parts of the reservoir, infected benthic bacteria were not found or the portion of visibly infected bacterial cells (FVIC) did not exceed 0.5% of the total abundance of bacteriobenthos (N B). A comparatively high infection of bacteria by viruses was recorded in bottom sediments only at one deep-water station, where FVIC was 2.5 of N B. 相似文献
