Ethylene–vinyl acetate (EVA) copolymer was used to immobilize H2-producing sewage sludge for H2 production in a three-phase fluidized bed reactor (FBR). The FBR with an immobilized cell packing ratio of 10% (v/v) and a liquid recycle rate of 5 l/min (23% bed expansion) was optimal for dark H2 fermentation. The performance of the FBR reactor fed with sucrose-based synthetic medium was examined under various sucrose concentration (Cso) and hydraulic retention time (HRT). The best volumetric H2 production rate of 1.80 ± 0.02 H2 l/h/l occurred at Cso = 40 g COD/l and 2 h HRT, while the optimal H2 yield (4.26 ± 0.04 mol H2/mol sucrose) was obtained at Cso = 20 g COD/l and 6 h HRT. The H2 content in the biogas was stably maintained at 40% or above. The primary soluble metabolites were butyric acid and acetic acid, as both products together accounted for 74–83% of total soluble microbial products formed during dark H2 fermentation.  相似文献

A three-phase bed bioreactor including a mix of immobilized microbes was used to degrade isopropanol (IPA). The immobilization method was studied and cells immobilized with calcium alginate, polyvinyl alcohol, activated carbon, and SiO2 were demonstrated to be the best immobilization method for the degradation of 90% of 2?g/L IPA in just 4 days, 1 day earlier than with free cells. Acetone was monitored as an indicator of microbial IPA utilization as the major intermediate of aerobic IPA biodegradation. The bioreactor was operated at hydraulic retention time (HRT) values of 32, 24, 16, 12, and 10?hr, which correspond to membrane fluxes of 0.03, 0.04, 0.06, 0.08, and 0.10?L/m2/hr, respectively. The chemical oxygen demand (COD) removal efficiencies were maintained at 98.0, 97.8, 89.1, 80.6, and 71.1% at a HRT of 32, 24, 16, 12, and 10?hr, respectively, while the IPA degradations were 98.6, 98.3, 90.3, 81.6, and 73.3%, respectively. With a comprehensive consideration of COD removal and economy, the optimal HRT was 24?hr. The results demonstrate the potential of immobilized mixed bacterial consortium in a three-phase fluidized bed reactor system for the aerobic treatment of wastewater containing IPA.  相似文献

Anaerobic granular sludge bed (AnGSB) bioreactors were supplemented with activated carbon carriers and combined with distributors (e.g., acrylic resin board, stainless steel net and plastic net) installed at different locations to investigate the effect of distributor/carrier on biohydrogen production efficiency. The results show that plastic net stimulated the substrate/microorganisms contact and sludge granulation, thereby leading to a much better H2 production performance when compared with those obtained from traditional CSTR. The highest H2 production rate (7.89 L/h/L) and yield (3.42 mol H2/mol sucrose) were obtained when two pieces of plastic nets were installed at both 4 cm and 16 cm from the bottom of AnGSB without carrier addition and the bioreactor was operated at a HRT of 0.5 h. For the AnGSB installed with two pieces of plastic net distributors, addition of carriers led to significant improvement on the H2 production efficiency at a longer HRT (1–4 h) when compared with the carrier-absent system.  相似文献

Three bioreactor configurations were employed in these investigations, which consisted of working volumes of 10, 1.2 and 1.2 L. Power spectrum diagrams of bed pressure fluctuation were used with hydraulic retention times (HRT) and geometric factors to identify the flow regimes in the bioreactors, where HRT varied from 8 to 1 h. It was found that the flow regimes in the bioreactors changed from a dispersed regime to coalesced and slugging regimes, when the biogas production rate (BPR) increased, as a result of decreasing the operating HRT. The flow regime was a dispersed bubble regime when the HRT was higher than 4 h in the bioreactor, whereas when the HRT was 2 h the coalesced bubble phenomena occurred in the bioreactor. A slugging regime was found when the HRT was lower than 1 h in thinner bioreactor.  相似文献

Cellulase was immobilized in a collagen fibril matrix, and no leakage of cellulase from the collagen fibril matrix was observed. The immobilized cellulase was more stable than the native cellulase. The substrate cellulose was hydrolyzed quantitatively with immobilized cellulase. The final reaction product was identified as glucose. Immobilized cellulase was used in a fluidized bed reactor where the pressure drop of the fluidized bed reactor was low and constant. Cellulose was hydrolyzed to glucose by the cellulase-bead fluidized bed reactor. The minimum flow velocity (Umf) was 0.5 cm/sec and the optimum flow velocity of the cellulose hydrolysis was 1 cm/sec.  相似文献

The aim of this work is to report on the physical characteristics of carrier material (perlite), biomass growth on the carrier material and the biogas production during an apparent steady state period in an inverse anaerobic fluidized bed reactor (IAFBR) for treating high strength organic wastewater. Before starting up the reactor, physical properties of the carrier material were determined. One millimeter diameter perlite particle is found to have a wet specific density of 295 kg/m(3) with specific surface area of 7.010 m(2)/g. This material has provided a good surface for biomass attachment and development. The biofilm concentration (in terms of attached volatile solids (AVS)) attached to carrier material was found to be 0.66 g(AVS)/g(solid). Most particles have been covered with a thin biofilm of uniform thickness. Once the inverse anaerobic fluidized bed system reached the steady state, the organic load was increased step wise by reducing hydraulic retention time (HRT) from 2 days to 0.16 day, while maintaining the constant feed of chemical oxygen demand (COD) concentration. This system has achieved 84% COD removal and reached the biogas production of 13.22 l/l/d at an organic loading rate (OLR) of 35 kgCOD/m(3)/d.  相似文献

A novel bioreactor containing self-flocculated anaerobic granular sludge was developed for high-performance hydrogen production from sucrose-based synthetic wastewater. The reactor achieved an optimal volumetric hydrogen production rate of approximately 7.3 L/h/L (7,150 mmol/d/L) and a maximal hydrogen yield of 3.03 mol H2/mol sucrose when it was operated at a hydraulic retention time (HRT) of 0.5 h with an influent sucrose concentration of 20 g COD/L. The gas-phase hydrogen content and substrate conversion also exceeded 40 and 90%, respectively, under optimal conditions. Packing of a small quantity of carrier matrices on the bottom of the upflow reactor significantly stimulated sludge granulation that can be accomplished within 100 h. Among the four carriers examined, spherical activated carbon was the most effective inducer for granular sludge formation. The carrier-induced granular sludge bed (CIGSB) bioreactor was started up with a low HRT of 4-8 h (corresponding to an organic loading rate of 2.5-5 g COD/h/L) and enabled stable operations at an extremely low HRT (up to 0.5 h) without washout of biomass. The granular sludge was rapidly formed in CIGSB supported with activated carbon and reached a maximal concentration of 26 g/L at HRT = 0.5 h. The ability to maintain high biomass concentration at low HRT (i.e., high organic loading rate) highlights the key factor for the remarkable hydrogen production efficiency of the CIGSB processes.  相似文献

An immobilized growing cell system was applied to the continuous L -isoleucine production by Serratia marcescens. In the new immobilized-cell systems using the carrageenan gel method. S. marcescens cells in the gel required nutrients and oxygen for growth, and the numbers of living cells per milliliter of gel increased to the levels of that of free cells in the liquid medium. This immobilized growing cell system exhibited high and stable activity for isoleucine production under steady-state conditions. Continuous isoleucine production was carried out by feeding the nutrient medium under aeration into a fluidized bed reactor containing the immobilized cells. In the continuous operation, an efficient production was maintained by automatically controlling the pH of the reaction mixture at 7.5. The productivity of isoleucine increased using multibed reactors. In a two-bed reactor system, the effluent L -isoleucine concentration reached 4.5 mg/ml at a retention time of 10 hr, and a steady state was maintained for longer than 30 days.  相似文献

The analysis of a continuous, aerobic, fixed-film bioreactor is performed by simulating the behavior of penicillin production in a three-phase fluidized bed. Rigorous mathematical models are developed for a fluidized-bed fermentor in which bioparticles are fluidized by the liquid medium and air. The steady-state performance of the fluidized-bed reactor is appraised in terms of penicillin productivity and outlet concentration by considering the two extremes in contacting patterns, complete back-mix and plug flow, in the absence of a growing biofilm. The results show that the complete back-mix contacting pattern is preferred over that of plug flow due to the nature of the penicillin kinetic relationships. It is also shown that for the dual-nutrient (glucose and oxygen) penicillin reaction system the optimum biofilm thickness does not equal the penetration depth of a limiting nutrient, but depends upon the total reactor configuration.  相似文献

A novel continuously stirred anaerobic bioreactor (CSABR) seeded with silicone-immobilized sludge was developed for high-rate fermentative H2 production using sucrose as the limiting substrate. The CSABR system was operated at a hydraulic retention time (HRT) of 0.5-6 h and an influent sucrose concentration of 10-40 g COD/L. With a high feeding sucrose concentration (i.e., 30-40 g COD/L) and a short HRT (0.5 h), the CSABR reactor produced H2 more efficiently with the highest volumetric rate (VH2) of 15 L/h/L (i.e., 14.7 mol/d/L) and an optimal yield of ca. 3.5 mol H2/mol sucrose. The maximum VH2 value obtained from this work is much higher than any other VH2 values ever documented. Formation of self-flocculated granular sludge occurred during operation at a short HRT. The granule formation is thought to play a pivotal role in the dramatic enhancement of H2 production rate, because it led to more efficient biomass retention. A high biomass concentration of up to 35.4 g VSS/L was achieved even though the reactor was operated at an extremely low HRT (i.e., 0.5 h). In addition to gaining high biomass concentrations, formation of granular sludge also triggered a transition in bacterial community structure, resulting in a nearly twofold increase in the specific H2 production rate. According to denatured-gradient-gel-electrophoresis analysis, operations at a progressively decreasing HRT resulted in a decrease in bacterial population diversity. The culture with the best H2 production performance (at HRT = 0.5 h and sucrose concentration = 30 g COD/L) was eventually dominated by a presumably excellent H2-producing bacterial species identified as Clostridium pasteurianum.  相似文献

Summary Enzymic conversion of glucose to fructose was carried out in a packed bed and in a fluidized bed reactor. The flow dynamics of these two flow systems, loaded with two different types of immobilized loaded with two different types of immobilized glucose isomerase particles, were studied. The theoretical RTD curve calculated from the axial dispersed plug flow model equation was matched to the experimental RTD curve by an optimization technique. The effect of fluid velocity on the extent of liquid dispersion was established. Theoretical predictions on the conversion of glucose to fructose were calculated using three mathematical models, namely, a plug flow model, a continuous stirred tank reactor (CSTR) model and an axial dispersed plug flow model. A mathematical model is developed to describe the performance of a three-phase fluidized bed reactor utilizing a transverse magnetic field. The model is based on the axially dispersed plug flow model for the bulk of liquid phase and on the Michaelis-Menten kinetics. The model equations are solved by the explicit finite difference method from transient to steady state conditions. The results of the numerical simulation indicate that the magnetic field increases the degree of bioconversion. The mathematical model is experimentally verified in a three-phase fluidized bed reactor with Penicillium chrysogenum immobilized on magnetic beads. The experimental results are well described by the developed model when the reactor operates in the stabilized regime. At low and relatively high magnetic field intensities certain discrepancy in the model solution is observed when the model over estimates the product concentration.  相似文献

A biofilm fluidized sand bed column reactor (14 L) has been operated in the three-phase mode on a soluble glucose-yeast hydrolysate substrate in which the biofilm-sand phase (1-2.5 L) was suspended by direct aeration of the bed. Within two weeks a tight biofilm was formed whose activity resulted in a 90% reduction, with loads of 10.7 kg TC/m(3)day. The residence time was 1 h. The biofilm remained intact during operation with high residence times (up to 23 h) over three weeks. Oxygen transfer coefficients varied with aeration rate and sand quantity between 0.02 and 0.04 s(-1) during non growth conditions; they decreased with increasing amounts of clean sand and were higher and relatively independent of the sand fraction with biofilm-covered sand. Aeration rates used in the 14 L reactor were 23-40 L/min (2.4-4.1 cm/s) and were sufficient to suspend 78-92% f the biofilm-covered sand. Clean sand was 50-75% suspended. Oxygen uptake rates varied between 15.4 and 23.1 mol/m(3) h.  相似文献

The effects of the hydraulic retention time (HRT = 8, 10, 12 or 16.7 h) and glucose concentration (30, 40 or 50 g/L) on the production of hydrogen and butyrate by an immobilized Clostridium tyrobutyricum culture, grown under continuous culturing conditions, were evaluated. With 30 g/L glucose, the higher HRTs tested led to greater butyrate concentrations in the culture, i.e., 9.3 g/L versus 12.9 g/L with HRTs of 8 h and 16.7 h, respectively. In contrast, higher biogas and hydrogen production rates were generally seen when the HRT was lower. Experiments with different glucose concentrations saw a significant amount of glucose washed out when 50 g/L was used, the highest being 22.7 g/L when the HRT was 16.7 h. This study found the best conditions for the continuous production of hydrogen and butyric acid by C. tyrobutyricum to be with an HRT of 12 h and a glucose concentration of 50 g/L, respectively.  相似文献

The concepts of feed pretreatment, phase separation, and whole-cell immobilization technology have been incorporated in this investigation for the development of rational and cost-effective two- and three-stage methane recovery systems from water hyacinth (WH)Analyses of laboratory data reveal that a three-stage system could be designed with an alkali pretreatment stage [3.6% Na(2)CO(3) + 2.5% Ca(OH)(2) W/W, 24 h HRT] followed by an open acid reactor (2.1 days HRT) and closed immobilized methane reactor (12 h HRT), providing steady-state COD conversion of 62-65%, TVA conversion of 91-95%, and gas productivity of 4.08-5.36 L/L reactor volume/day with 82% methane. A gas yield of 50 L/kg WH/day (dry wt basis) at 35-37 degrees C is possible with this system. Insulation bricks, with particle size distribution of 500-3000 mum, were used as support material in the reactors at organic loading rate of 20 kg COD/m(3) day. The reactors matured in 15-18 weeksSubstantial reduction in retention time for the conversion of volatile acids in immobilized methane reactors prompted further research on the combined immobilized reactor to make possible an additional reduction in the cost of a WH-based biogas system. Evaluation of laboratory data reveals that a two-stage system could be designed with an open alkali pretreatment stage and a combined immobilized reactor (12 h HRT), providing steady-state COD conversion of 53% and gas productivity of 3.1 L/L reactor volume/day with 86% methane. A gas yield of 44 L/kg WH/day (dry wt basis) at 35-37 degrees C could be obtained from this system. Insulation bricks, with 500-1000 mum particle size distribution, was used as support material at an organic loading rate of 15 kg COD/m(3) day. Notwithstanding the fact that the technology in this study has been developed with water hyacinth as substrate, the implicit principles could be extended to any other organic substrate.  相似文献

Mixed culture methanotrophic attached biofilms immobilized on diatomite particles in a three-phase fluidized bed reaction system were developed. Methane monooxygenase (MMO) activity on diatomite particles increased as soon as the lag phase ended. More than 90% of the MMO activity in the fluidized bed was attached. A biofilm concentration of 3.3c3.7mg dry weight cell (dwc) per g dry solid (DS) was observed. Batch experiments were performed to explore the possibility of producing epoxypropane by a propene–methane co-oxidation process. The effect of methane on the epoxidation of propene and the effect of propene on the growth of methanotroph was also studied. In continuous experiments, optimum mixed gas containing 35 methane, 20 propene and 45% oxygen were continuously circulated through the fluidized bed reactor to deliver substrates and extract product. Initial epoxypropane productivity was 110–150 μmol/day. The bioreactor operated continuously for 53 days without obvious loss of epoxypropane productivity.  相似文献

Cells of Candida guilliermondii immobilized onto porous glass spheres were cultured batchwise in a fluidized bed bioreactor for xylitol production from sugarcane bagasse hemicellulose hydrolyzate. An aeration rate of only 25 mL/min ensured minimum yields of xylose consumption (0.60) and biomass production (0.14 g(DM)/g(Xyl)), as well as maximum xylitol yield (0.54 g(Xyt)/g(Xyl)) and ratio of immobilized to total cells (0.83). These results suggest that cell metabolism, although slow because of oxygen limitation, was mainly addressed to xylitol production. A progressive increase in the aeration rate up to 140 mL/min accelerated both xylose consumption (from 0.36 to 0.78 g(Xyl)/L.h) and xylitol formation (from 0.19 to 0.28 g(Xyt)/L.h) but caused the fraction of immobilized to total cells and the xylitol yield to decrease up to 0.22 and 0.36 g(Xyt)/g(Xyl), respectively. The highest xylitol concentration (17.0 g(Xyt)/L) was obtained at 70 mL/min, but the specific xylitol productivity and the xylitol yield were 43% and 22% lower than the corresponding values obtained at the lowest air flowrate, respectively. The concentrations of consumed substrates and formed products were used in material balances to evaluate the xylose fractions consumed by C. guilliermondii for xylitol production, complete oxidation through the hexose monophosphate shunt, and cell growth. The experimental data collected at variable oxygen level allowed estimating a P/O ratio of 1.35 mol(ATP)/mol(O) and overall ATP requirements for biomass growth and maintenance of 3.4 mol(ATP)/C-mol(DM).  相似文献

The effect of substrate concentration (sucrose) on the stability and yield of a continuous fermentative process producing hydrogen was studied. High substrate concentrations are attractive from an energy standpoint as they would minimise the energy required for heating. The reactor was a CSTR; temperature was maintained at 35 degrees C; pH was controlled between 5.2 and 5.3, and the hydraulic retention time (HRT) was 12 h. Online measurements were taken for ORP, pH, temperature, %CO2, gas output and %H2, and data logged using a MatLAB data acquisition toolbox. Steady-state operation was obtained at 10, 20 and 40 g/L of sucrose in the influent, but a subsequent step change to 50 g/L was unsustainable. The hydrogen content ranged between 50% and 60%. The yield of hydrogen decreased as the substrate concentration increased from 1.7 +/- 0.2 mol/mol hexose added at 10 g/L, to 0.8 +/- 0.1 mol/mol at 50 g/L. Sparging with nitrogen improved the hydrogen yield by at least 35% at 40 g/L and at least 33% at 50 g/L sucrose. Sparging also enabled steady-state operation at 50 g/L sucrose. Addition of an extra 4 g/L of n-butyric acid to the reactor operating at 40 g/L sucrose increased the butyrate concentration from 9,830 to 18,900 mg/L, immediately stopping gas production and initiating the production of propionate, whilst the addition of 2 g/L taking the butyrate concentration to 12,200 mg/L did not do so. It was shown that operation at 50 g/L sucrose in a CSTR in butyrate fermentation is possible.  相似文献

The treatment of real acid mine drainage water (pH 2.7-4.3) containing sulfate (1.5-3.34 g/L) and various metals was studied in an ethanol-fed sulfate-reducing fluidized bed reactor at 35 °C. The robustness of the process was tested by increasing stepwise sulfate, ethanol and metal loading rates and decreasing feed pH and hydraulic retention time. Highest sulfate reduction rate (4.6 g/L day) was obtained with feed sulfate concentration of 2.5 g/L, COD/sulfate ratio of 0.85 and HRT of 12 h. The corresponding sulfate and COD removal efficiencies were about 90% and 80%, respectively. The alkalinity produced in sulfidogenic ethanol oxidation neutralized the acidic mine water. Highest metal precipitation efficiencies were observed at HRT of 24 h, the percent metal removal being over 99.9% for Al (initial concentration 55 mg/L), Co (9.0 mg/L), Cu (49 mg/L), Fe (435 mg/L), Ni (3.8 mg/L), Pb (7.5 mg/L) and Zn (6.6 mg/L), and 94% for Mn (7.21 mg/L).  相似文献

This paper examines the effect of organic loading rate on the removal efficiency of COD and TOC anaerobic thermophilic fluidized bed reactor (AFBR) in the treatment of cutting-oil wastewater at different hydraulic retention time (HRT) conditions. The essays are development at laboratory scale using a porous support medium. The AFBR reactor was subjected to a programme of steady-state operation over a range of hydraulic retention times, HRTs, in the range 12-2h and organic loading rates, OLRs, between 11.9 and 51.3kgCOD/m(3)d. The highest efficiency was 95.9% for an OLR of 13kgCOD/m(3)d and HRT of 11h. Over an operating period of 92 days, an OLR of 51.3kgCOD/m(3)d was achieved with 67.1% COD removal efficiency (71.3% TOC) in the experimental AFBR reactor. Although the level of biogas generation was not high, the anaerobic fluidized bed technology provided significant advantages over the conventional physico-chemical treatment applied in the factory. The effluent had a better quality (lower organic loading) and it was possible to reuse it in different applications in the factory (e.g., irrigation of gardens). The biological treatment did not lead to the generation of oily sludge, which is considered as hazardous waste by legislation. Furthermore, a continuous stream is produced and this reduced the impact of large flows discharged 4-5 times per week to the urban collector and MWWTP (municipal wastewater treatment plant).  相似文献
