Investigations were carried out on smooth muscle cells (SMC) of rat and rabbit anococcygeus by the method of a double "sugar bridge" in the presence of tetraethylammonium (1 mM/1) in the Krebs solution. Stimulation of the muscle strip by the electric current rectangular pulse of the maximal value and of short duration caused the development of excitatory postsynaptic potentials (EP SP) in the rat and rabbit SMC, and of inhibitory postsynaptic potentials in the rabbit SMC. The value of postsynaptic potentials displayed a linear dependence on the level of the membrane potential. Elimination of chlorine ions from the external solution decreased the EP SP of the SMC of rabbit anococcygeus and shifted the reversion potential in the direction of sodium balance potential. Apparently generation of the EP SP of the SMC of rabbit anococcygeus was associated with the increased permeability of the membrane both for sodium and for chlorine ions.  相似文献

Voltage-gated K(+) (Kv) channels are important in the regulation of pulmonary vascular function having both physiological and pathophysiological implications. The pulmonary vasculature is essential for reoxygenation of the blood, supplying oxygen for cellular respiration. Mitochondria have been proposed as the major oxygen-sensing organelles in the pulmonary vasculature. Using electrophysiological techniques and immunofluorescence, an interaction of the mitochondria with Kv channels was investigated. Inhibitors, blocking the mitochondrial electron transport chain at different complexes, were shown to have a dual effect on Kv currents in freshly isolated rat pulmonary arterial smooth muscle cells (PASMCs). These dual effects comprised an enhancement of Kv current in a negative potential range (manifested as a 5- to 14-mV shift in the Kv activation to more negative membrane voltages) with a decrease in current amplitude at positive potentials. Such effects were most prominent as a result of inhibition of Complex III by antimycin A. Investigation of the mechanism of antimycin A-mediated effects on Kv channel currents (I(Kv)) revealed the presence of a mitochondria-mediated Mg(2+) and ATP-dependent regulation of Kv channels in PASMCs, which exists in addition to that currently proposed to be caused by changes in intracellular reactive oxygen species.  相似文献

Circumferential stretch due to increases in pressure induces vascular smooth muscle cell depolarization and contraction known as the myogenic response. The aim of this study was to determine the in vivo effects of axial-longitudinal stretch of the rat saphenous artery (SA) on smooth muscle membrane potential (Em) and on external diameter. Consecutive elongations of the SA were carried out from resting length (L0) in 10% increments up to 140% L0 while changes in membrane potential and diameter were determined in intact and de-endothelized vessels. Axial stretching resulted in a small initial depolarization at 120% of L0 followed by a progressive 20 to 33% hyperpolarizaion of vascular smooth muscle between 130% and 140% of L0. At 140%, an average maximal 10.6 mV reversible hyperpolarization was measured compared to -41.2 +/- 0.49 mV Em at 100% L0. De-endothelialization completely eliminated the hyperpolarization to axial stretching and augmented the reduction of diameter beyond 120% L0. These results indicate that arteries have a mechanism to protect them from vasospasm that could otherwise occur with movements of the extremities.  相似文献

Summary The structure and function of vascular smooth muscle cells have been extensively investigated with the aid of in vitro culture techniques. The majority of studies have utilized aortic tissue as the source of cells. We present here a method for isolating and culturing smooth muscle cells of the rat superior mesenteric artery, an elasto-muscular vessel that is structurally and functionally different from the aorta. Cells were isolated from partially digested explants and characterized by immunochemical and biochemical techniques. Unlike cultured fibroblasts, the cultured cells stained positive for smooth muscle specific actin. The cells also produced laminin and type IV collagen in culture. This method provides a means for the isolation of large numbers of viable smooth muscle cells from the superior mesenteric artery which can be propagated in culture for in vitro study.  相似文献

The hypothesis that protein kinase C (PKC) isable to regulate the whole cell Ca-activated K(KCa) current independently of PKC effects on local Ca release events was tested using the patch-clamp technique and freshly isolated rat tail artery smooth muscle cells dialyzed with a strongly buffered low-Ca solution. The active diacylglycerol analog1,2-dioctanoyl-sn-glycerol (DOG) at 10 µM attenuated the current-voltage(I-V)relationship of the KCa current significantly and reduced the KCacurrent at +70 mV by 70 ± 4% (n = 14). In contrast, 10 µM DOG after pretreatment of the cells with 1 µM calphostin C or 1 µM PKC inhibitor peptide, selective PKCinhibitors, and 10 µM1,3-dioctanoyl-sn-glycerol, aninactive diacylglycerol analog, did not significantly alter theKCa current. Furthermore, thecatalytic subunit of PKC (PKCC)at 0.1 U/ml attenuated theI-Vrelationship of the KCa currentsignificantly, reduced the KCacurrent at +70 mV by 44 ± 3% (n = 17), and inhibited the activity of singleKCa channels at 0 mV by 79 ± 9% (n = 6). In contrast, 0.1 U/mlheat-inactivated PKCC did notsignificantly alter the KCacurrent or the activity of singleKCa channels. Thus these resultssuggest that PKC is able to considerably attenuate theKCa current of freshly isolatedrat tail artery smooth muscle cells independently of effects of PKC onlocal Ca release events, most likely by a direct effect on theKCa channel.  相似文献

Single intramural stimulation of the atropinized muscle strip of the guinea pig stomach by square pulses was accompanied by the formation of inhibitory postsynaptic potentials (IPSPs) in the muscle cells and by relaxation of the strip. The mean latent period of the IPSPs was 150±15 msec, and the time for its amplitude to rise and fall was 150±15 and 375±92 msec respectively. The latent period of the onset of relaxation of the muscle strip was 1850±230 msec. The amplitude and rate of rise of these IPSPs increased with an increase in the strength of stimulation. This shows that the same gastric muscle cell is under the inhibitory influence not of one, but of several nerve fibers. The effectiveness of synaptic transmission was largely dependent on the character of the preceeding stimulation. For instance, in response to repeated stimulation with short intervals facilitation was observed, but after repetitive stimulation had ended posttetanic depression followed.A. A. Background

The extracellular calcium-sensing receptor (CaSR) belongs to family C of the G protein coupled receptors. Whether the CaSR is expressed in the pulmonary artery (PA) is unknown.

Methods

The expression and distribution of CaSR were detected by RT-PCR, Western blotting and immunofluorescence. PA tension was detected by the pulmonary arterial ring technique, and the intracellular calcium concentration ([Ca2+]i) was detected by a laser-scanning confocal microscope.

Results

The expressions of CaSR mRNA and protein were found in both rat pulmonary artery smooth muscle cells (PASMCs) and PAs. Increased levels of [Ca2+]o (extracellular calcium concentration) or Gd3+ (an agonist of CaSR) induced an increase of [Ca2+]i and PAs constriction in a concentration-dependent manner. In addition, the above-mentioned effects of Ca2+ and Gd3+ were inhibited by U73122 (specific inhibitor of PLC), 2-APB (specific antagonist of IP3 receptor), and thapsigargin (blocker of sarcoplasmic reticulum calcium ATPase).

Conclusions

CaSR is expressed in rat PASMCs, and is involved in regulation of PA tension by increasing [Ca2+]i through G-PLC-IP3 pathway.  相似文献

Matrix metalloproteinases (MMPs) and their tissue inhibitors (TIMPs) are involved in tissue remodeling processes. TIMP-1 is the main native inhibitor of MMPs and it contributes to the development of tissue fibrosis. It is known that ANG II plays a fundamental role in vascular remodeling. In this study, we investigated whether ANG II modulates TIMP-1 expression in rat aortic smooth muscle cells. In vitro, ANG II induces TIMP-1 mRNA expression in a dose-dependent manner. The maximal increase in TIMP-1 expression was present after 3 h of ANG II stimulation. The ANG II increase in TIMP-1 expression was mediated by the ANG type 1 receptors because it was blocked by losartan. The increase in TIMP-1 expression was present after the first ANG II treatment, whereas repeated treatments (3 and 5 times) did not modify TIMP-1 expression. In vivo, exogenous ANG II was administered to Sprague-Dawley rats (200 ng. kg(-1). min(-1) sc) for 6 and 25 days. Control rats received physiological saline. After treatment, systolic blood pressure was significantly higher (P < 0.01), whereas plasma renin activity was suppressed (P < 0.01), in ANG II-treated rats. ANG II increased TIMP-1 expression in the aorta of ANG II-treated rats both at the mRNA (P < 0.05) and protein levels as evaluated by Western blotting (P < 0.05) and/or immunohistochemistry. Neither histological modifications at the vascular wall nor differences in collagen content in the tunica media were present in both the ANG II- and saline-treated groups. Our data demonstrate that ANG II increases TIMP-1 expression in rat aortic smooth muscle cells. In vivo, both short- and long-term chronic ANG II treatments increase TIMP-1 expression in the rat aorta. TIMP-1 induction by ANG II in aortic smooth muscle cells occurs in the absence of histological changes at the vascular wall.  相似文献

目的:研究大鼠肺动脉平滑肌细胞钙激活氯通道电流的电生理特性。方法:膜片钳全细胞和膜内向外记录模式检测大鼠肺动脉平滑肌细胞上钙激活氯通道全细胞电流和单通道电流。结果:大鼠肺动脉平滑肌细胞记录到稳定的钙激活氯通道电流(ICl(Ca));ICl(Ca)表现出典型的外向整流特性和电压时间依赖性激活。结论:大鼠肺动脉平滑肌细胞膜上存在电压、时间依赖性氯通道电流,钙激活氯通道通过促进肺动脉平滑肌细胞去极化而成为调节肺动脉特性的关键调节因子。  相似文献

Nitric oxide (NO) released from the endothelium or from NO-donors is a powerful vasodilator. Its effect is mediated partly by vascular smooth muscle high conductance calcium-activated potassium (Kca) channels. Contradictory data exist as to whether NO activated the KCa channel directly or indirectly via protein kinase G (PKG). Thus the hypothesis that NO-donors can activate the KCa directly was investigated using the patch-clamp technique and freshly isolated smooth muscle cells from the rat tail artery. In inside-out experiments, the activity of KCa-channels was increased 1.61 +/- 0.20-fold (n = 10) by 10 microM SNP and 1.45 +/- 0.17-fold (n = 8) by 10 microM SNAP. However, the activity of KCa channels was also increased 1.46 +/- 0.20-fold (n = 8) by addition of the experimental bath solution. Thus these results suggest that NO released from NO-donors cannot activate KCa channel of the rat tail artery smooth muscle cells directly.  相似文献

Exogenous carbon monoxide (CO) can induce pulmonary vasodilation by acting directly on pulmonary artery (PA) smooth muscle cells. We investigated the contribution of K+ channels to the regulation of resistance PA resting membrane potential on control (PAC) rats and rats exposed to CO for 3 wk at 530 parts/million, labeled as PACO rats. Whole cell patch-clamp experiments revealed that the resting membrane potential of PACO cells was more negative than that of PAC cells. This was associated with a decrease of membrane resistance in PACO cells. Additional analysis showed that outward current density in PACO cells was higher (50% at +60 mV) than in PAC cells. This was linked to an increase of iberiotoxin (IbTx)-sensitive current. Chronic CO hyperpolarized membrane of pressurized PA from -46.9 +/- 1.2 to -56.4 +/- 2.6 mV. Additionally, IbTx significantly depolarized membrane of smooth muscle cells from PACO arteries but not from PAC arteries. The present study provides initial evidence of an increase of Ca2+-activated K+ current in smooth muscle cells from PA of rats exposed to chronic CO.  相似文献

Phosphodiesterases (PDE) metabolize cyclic nucleotides limiting the effects of vasodilators such as prostacyclin and nitric oxide (NO). In this study, DNA microarray techniques were used to assess the impact of NO on expression of PDE genes in rat pulmonary arterial smooth muscle cells (rPASMC). Incubation of rPASMC with S-nitroso-l-glutathione (GSNO) increased expression of a PDE isoform that specifically metabolizes cAMP (PDE4B) in a dose- and time-dependent manner. GSNO increased PDE4B protein levels, and rolipram-inhibitable PDE activity was 2.3 +/- 1.0-fold greater in GSNO-treated rPASMC than in untreated cells. The soluble guanylate cyclase (sGC) inhibitor, 1H-[1,2,4]oxadiazolo[4,3,-a]quinoxalin-1-one, and the cAMP-dependent protein kinase inhibitor, H89, prevented induction of PDE4B gene expression by GSNO, but the protein kinase G (PKG) inhibitors, Rp-8-pCPT-cGMPs and KT-5823, did not. Incubation of rPASMC with IL-1beta and tumor necrosis factor-alpha induced PDE4B gene expression, an effect that was inhibited by l-N(6)-(1-iminoethyl)lysine, an antagonist of NO synthase 2 (NOS2). The GSNO-induced increase in PDE4B mRNA levels was blocked by actinomycin D but augmented by cycloheximide. Infection of rPASMC with an adenovirus specifying a dominant negative cAMP response element binding protein (CREB) mutant inhibited the GSNO-induced increase of PDE4B gene expression. These results suggest that exposure of rPASMC to NO induces expression of PDE4B via a mechanism that requires cGMP synthesis by sGC but not PKG. The GSNO-induced increase of PDE4B gene expression is CREB dependent. These findings demonstrate that NO increases expression of a cAMP-specific PDE and provide evidence for a novel "cross talk" mechanism between cGMP and cAMP signaling pathways.  相似文献

Significant Ca(2+) release was previously noted with the activation of L-type Ca(2+) current in rat superior cerebral artery smooth muscle cells. Here we examined whether the P(2X) current that is partly carried by Ca(2+) also triggers Ca(2+) release in this preparation. Application of P(2X) agonists evoked membrane currents and concomitant Ca(2+) transients in whole cell voltage-clamped single cells. The expected increase in intracellular Ca(2+) concentration ([Ca(2+)](i)) was calculated from the time-integrated P(2X) current by assuming Ca(2+) is the only charge carrier. The measured increase in [Ca(2+)](i) was plotted as a function of the expected increase in [Ca(2+)](i), and Ca(2+)-buffering power was obtained as a reciprocal of the linear fit to this relationship. Both ryanodine, a Ca(2+)-induced Ca(2+)-release inhibitor, and cADP ribose, a putative activator of Ca(2+)-induced Ca(2+) release, had no significant effects on Ca(2+)-buffering power. These results suggest that Ca(2+) influx through P(2X) receptors does not trigger significant Ca(2+) release. We then examined whether P(2X) responses influence the subsequent P(2Y) response. P(2Y) responses were characterized by measuring the rate of [Ca(2+)](i) increase obtained as the slope of the linear regression to the rising phase of the Ca(2+) transient. During simultaneous application of the P(2X) and P(2Y) agonist, the rate of [Ca(2+)](i) increase was facilitated or suppressed depending on the size of the P(2X) receptor-mediated [Ca(2+)](i) increase. Membrane depolarization close to the Ca(2+) equilibrium potential significantly promoted the rate of [Ca(2+)](i) increase. Our results suggest that the [Ca(2+)](i) increase and membrane depolarization caused by the P(2X) current may regulate the subsequent P(2Y) response.  相似文献
