Nutrient and salt (NaCI) levels were examined in fruits of twostrand species, Cakile maritima and Arctotheca populifolia,collected near Perth, Western Australia. Nutrient levels inseeds of C. maritima and achenes of A. populifolia were relativelyhigh when compared to those in seeds of Australian native speciesor introduced crop plants, despite the very low nutrient statusof the strand habitat. The high levels of organic and essentialinorganic nutrients in seeds would be of especial value in seedlingestablishment. The upper seed of the two-segmented fruit ofCakile was heavier than the lower seed, but both seeds had verysimilar levels of nutrients. Concentrations of N, P, and micronutrientswere very much higher in seeds of Cakile and Arctotheca thanin other parts of the fruit. Cakile leaves had similar levelsof N and P to seeds, but higher levels of micronutrients, especiallyNa and Cl. In Cakile, concentration ratios for Na levels inleaves, silicule and seeds were 359: 74: 1 respectively, thosefor Cl were 200: 30: 1, suggesting very restricted loading ofNa and Cl into the phloem stream destined for fruits, and selectiveuptake by developing embryos. Cakile seeds germinate within their fruit segments, and levelsof NaCl in recently-matured fruits would be high enough to suppressgermination. The top segment of the fruit had a higher concentrationof NaCl than the bottom, and it is likely that this is responsiblefor the lower germination of seeds within top segments. Theliterature on seed dormancy in Cakile is reviewed briefly, andit is concluded that salt levels in the fruit wall and strandsoil are the major determinants of dormancy in Cakile seeds. 选取寄生于竹类植物叶片上的3科6属共8种蚜虫,即居竹舞蚜Astegopteryx bambusifoliae Takahashi、小舞蚜Astegopteryx minuta (van der Goot)、居竹坚蚜Cerataphis bambusifoliae Takahashi、林栖粉角蚜Ceratovacuna silvestrii (Takahashi)、塔毛角蚜Chaitoregma tattakana (Takahashi)、竹色蚜Melanaphis bambusae (Fullaway)、竹纵斑蚜Takecallis arundinariae (Essig)和竹梢凸唇斑蚜 Takecallis taiwanus (Takahashi),在光学显微镜下观察并测量了34个形态特征;统计比较了28个形态特征在种内的变异。通过主成分分析筛选形态特征,每两两特征对应统计作图,标出每种蚜虫的95%椭圆置信区间进行分析。结果表明,体形、腹管和触角的形态在不同的科间有较大差别,喙末端、跗节Ⅰ、跗节Ⅱ及爪的形态在科间有较高的趋同性;在种内各形态特征存在一定变异,其中喙末端 (CV=3.73%~7.59%)、跗节Ⅰ (CV=4.16%~12.05%)、跗节Ⅱ (CV=3.10%~8.39%)和爪(CV=2.60%~11.68%)的变异都很小。主成分分析筛选的第一主成分为喙末端、跗节Ⅰ、跗节Ⅱ和爪,不同的椭圆区间范围提示这些特征组合基本处于稳定的范围内。与蚜虫的取食行为和附着于植物表面相关的形态特征,如喙末端、跗节Ⅰ、跗节Ⅱ和爪等在不同类群间存在显著的相似性,暗示了寄生于竹类植物叶片的蚜虫在这些形态特征上的趋同适应。最后结合蚜虫的生物学信息,初步探讨了形态适应的机制。  相似文献

The relationship between ATPase activity, medium acidificationand auxin-stimulated growth in segments of pea stem (Pisum sativumL., cv. Alaska) and cucumber hypocotyl (Cucumis sativus L.,cv. Long Green Ridge) was investigated using sodium orthovanadate,widely used as a selective inhibitor of plasma membrane-associatedATPase activity. ATPase activity of cucumber microsomal preparationswas about seven times lower than similar preparations from pea(on a mg microsomal protein basis) and was much more effectivelyinhibited by vanadate. Similarly, acidification of the mediumby abraded cucumber segments occurred to a lesser extent thanwith pea and showed a greater inhibition by vanadate. Both growthin controls and auxin-stimulated growth of cucumber segmentswere strongly inhibited by vanadate, whereas in pea auxin-stimulatedgrowth was reduced by only half and controls showed little inhibition.Acidification of the medium by segments of both species wasfound to occur readily even in controls and showed little promotionin the presence of IAA, although growth in both species wasrapidly and significantly promoted by IAA. These results indicatethat acidification is brought about by a plasma membrane-associatedATPase, and suggest that while acidification is an essentialfactor for auxin-stimulated growth it may not be the mechanismby which the growth rate is controlled. By washing out solutes in 0.5 mM CaSO4at 25 °C during aperiod of 5–6 h, the release of sugars by excised stemsegments of Vicia faba L. was measured. The stem parasite Cuscutaeuropaea strongly stimulated the release of sucrose into theefflux medium; this effect was most marked during the last hoursof each experiment but this stimulating effect of the parasitecould not be detected for glucose and fructose. The fact thatparasitized stem segments released higher than normal hexoseamounts during the last hours of several experiments, couldbe explained as the result of extracellular hydrolysis of sucroseby free space invertase. A high free space acid invertase activitywas present in young stem segments of Vicia faba and in tissuesof Cuscuta. The stimulating influence of Cuscuta on sugar releaseby cells of stem segments appears sucrose-specific, supportingthe idea that the stimulating influence of Cuscuta on sugarrelease is restricted to the sieve-tube system. When metabolic inhibitors were added to the washing solutionor when segments were incubated at low temperature, no cleareffect of the parasite could be observed and for all segments(parasitized and non-parasitized) a strongly enhanced releaseof sucrose into the efflux medium was found during the lasthours of an experiment. These data support the idea that anintensive resorption of sucrose occurs within stem segments,after its release into the apoplast. Decapitated segments from maize (Zea mays L.) coleoptiles orientedvertically in an upright position show a strong spontaneousgrowth response (SGR) 3 h after decapitation. The latent periodof the SGR is markedly reduced when these segments are orientedin an inverted position. Coleoptile segments with intact tipsexhibit a weak and transient SGR in the vertical upright orientation.However, in the inverted orientation, these segments show atypical SGR. The data are inconsistent with the current hypothesisthat the SGR is caused by a time-dependent increase in tissuesensitivity to auxin. The parallel increase in membrane potentialdifference and growth rate during the time-course of the SGRindicates a possible role for PM H+-ATPase in the establishmentof the SGR in maize coleoptile segments. RNA干扰(RNA interference, RNAi)是一种调控基因表达的方法, 其通过体外合成一段与内源靶基因同源的双链RNA(dsRNA)或siRNA, 导入生物体内, 使内源靶基因中同源mRNA降解, 从而达到阻抑基因表达的目的。类钙粘蛋白(cadherin-like protein)是位于昆虫中肠刷状缘膜囊(brush border membrane vesicles, BBMV)上与钙粘蛋白(cadherin)结构相似的物质, 是多种昆虫体内Bt杀虫蛋白的受体。本研究利用基因特异引物通过RT-PCR扩增了小菜蛾类钙粘蛋白基因的2个片段(CAD1和CAD2), 合成相对应的双链RNA(double-stranded RNA, dsRNA); 并将dsRNA通过显微注射导入小菜蛾3龄幼虫体内, 测定了不同靶位点、不同剂量、不同检测时间对目的基因mRNA表达量的影响。结果表明: 将70 nL CAD1对应的dsRNA注射到幼虫体内48 h后, 基因表达量显著下降, 72 h后恢复。免疫印迹检测结果表明, 类钙粘蛋白在注射dsRNA 48 h后幼虫BBMV中的含量明显下降。本实验成功实现了小菜蛾类钙粘蛋白基因的沉默, 该体系的成功建立为利用RNAi技术分析小菜蛾及其他鳞翅目昆虫基因的功能提供了参考。  相似文献

Water loss from roots back into drying soil is a problem ofpractical importance in plants growing under conditions of verylow substrate water potential, such as dry or saline areas.Root exodermis is relatively impermeable and has been suggestedto play a protective role against water loss. The relative waterretention ability was compared in root segments from exodermal(maize, onion, sunflower, Rhodes grass and sorghum) and non-exodermalspecies (Pisum sativum,Vicia fabaand wheat). Apical and basalsegments from exodermal roots, with different degrees of exodermisdevelopment, were also compared, as were segments from sorghumroots in which the exodermis thickness had been modified bysubjecting the plants to a 30 d water stress treatment. Waterretention was significantly higher in segments from exodermalroots. In each root, water loss was higher in apical than inbasal segments, regardless of the presence of exodermis. Insorghum, prolonged drought treatment increased exodermis thickeningin nodal roots, however, no differences in rates of water losswere observed in segments obtained from control and droughtedplants. Soil sheaths formed around roots of Rhodes grass growingin very dry soil with the epidermis adhering tightly to thesheath. In plants growing in the field, soil sheaths may bemore effective than the exodermis in preventing root water loss.Copyright1999 Annals of Botany Company. Brassinolide stimulated an increase in fresh weight of segmentsof hypocotyls of etiolated squash (Cucurbita maxima Duch. cv.Houkou-Aokawaamaguri). Brassinolide-treated segments containedhigher levels of IAA than water-treated segments and also showeda tendency towards decreased levels of ABA. Mendel, Z. and Liphschitz, N. 1988. Unseasonable latewood andincrusted pits are the cause of drying in Pinus halepensis andP.eldarica infested with Matsucoccus josephi.—J. exp. Bot.39: 951–959. A study was made of the effects of Matsucoccus josephi (Insecta:Homoptcra: Margarodidae) on (a) water transport in Pinus halepensis,using a basic fuchsin solution and (b) the structural changesof the xylem of Pinus halepensis, P. brutia andP. eldarica usinga scanning electron microscope.Examinations were carried outin artificially and naturally infested plants. Transport ofthe fuchsin dye was arrested below the affected zone of heavilyinfested shoots, but reached the shoot apex through sparselyinfested segments or via segments heavily infested with thefirst larval stage. Heavily infested stem sections of P. halepensisand P.eldarica were characterized by narrow pits and incrustedpit membranes. In non-infested shoots and those infested withfirst or early second-scale larvae in both these species andheavily infested P. brutia shoots, there were typical earlywoodpits with wide pit apertures and thin membranes. It is suggestedthat blocking of water ascent in P. halepensis and P. eldaricastarts when female second-instar larvae complete their developmentand only when population density is relatively high. Therefore,it is considered that the accelerated drying of the growth ofP. halepensis and P. brutia is directly related to the narrowingof the pit apertures, which resembled those of latewood pitsalthough produced early in the season. The absence of changesin the xylem of heavily infested P brutia may explain why thisspecies tolerates a high density of M. josephi. The structuralmodification of the xylem caused by the scale may be relatedto imbalance in plant growth hormones which are either transmittedwith the insect saliva or formed in the plant by salivary enzymes. Rhizomorphs put out by Armillaria mellea from small woody inoculain glass tubes of moist soil were tested for their ability tocolonize segments of willow shoots, previously killed by autoclavingand then buried for various periods in soil before inoculationwith A. mellea. The degree of saprophytic colonization of thesesubstrate segments by A. mellea was assessed by reburying themin fresh tubes of moist soil, and recording weekly growth incrementsof rhizomorphs put out from them over a period of 5 weeks. Aperiod of previous burial in soil up to 3 weeks was found notto diminish availability of the substrate segments to A. mellea,but with longer periods substrate value for A. mellea progressivelydeclined. Segments of living, green shoots of willow provideda consistently better substrate for A. mellea than did deadsegments, and these living segments maintained their substratevalue over periods (up to 7 weeks) of previous burial in soil.These results are interpreted in terms of competition betweenA. mellea and other soil fungi. In the infection of living tissues,A. mellea benefits from the exclusion of competitors by hostresistance. In the saprophytic colonization of dead tissues,A. mellea has the advantage of its capacity to decompose celluloseand lignin, which are substrates restricted to a minority ofsoil fungi.  相似文献

The effects of the morphactin 2-ehloro-9-hydroxyfluorene-9-carboxylicacid methyl ester [CFM] on growth, geotropic curvature and transportand metabolism of indol-3yl-acetic acid [IAA-5-3H] in the coleoptilesof Zea mays and A vena saliva have been investigated. A strongcorrelation has been found to exist between the inhibition ofthe geotropic response and the inhibition of auxin transport.CFM supplied at concentrations sufficient to abolish auxin transporthas been shown to promote the elongation of Zea, but not ofAvena, coleoptile segments. CFM does not change the patternof metabolism of IAA in Zea coleoptile segments. In these segmentsIAA is metabolized when its concentration is high, but the radioactivitytransported basipetally, or laterally in geotropically stimulatedcoleoptiles, is virtually confined to the IAA molecule. Radioactivityexported into the basal receiver blocks is wholly confined toIAA. It is concluded that CFM inhibits the geotropic responsein coleoptiles by suppression of the longitudinal and lateralauxin transport mechanisms. The growth-promoting propertiesof this substance cannot be linked with its effects on eitherauxin metabolism or transport.  相似文献

Structure of the Mutant Fibrillin-1 Gene in the Tight Skin (TSK) Mouse   总被引:1,自引:0,他引:1  
Mice carrying the tight skin (TSK) mutation harbors a 3.0-kbgenomic duplication (exons 17–40) of the fibrillin-1 gene(Fbn-1) locatedon band F of chromosome 2 as TSK mutation. Wecloned and sequenced the mutated Fbn-1 gene, since it is believedto be responsible for TSK syndrome. Sequence analysis showednumerous amino aciddifferences in the 5' and 3' segments betweenthe TSK mutation and wild-type fbn-1 gene, but any amino aciddifference between the TSK mutation and C57BL/6 mice. (TSK andC57Bl/6 mice are genetically similar, differing only by TSKmutation.) Four amino acid differences were observed betweentwo copies of TSK's fbn-1 gene encoded by exons 17–40.Our results suggest that the majority of structural differencesoccurred in the N and C termini segments during strain divergenceand only a few after the duplication event.  相似文献

Stem segments of non-tumorous Nicotiana glauca and N. langsdorffiiplants and of their tumor-producing amphidiploid F1 hybrid weretreated with 6-furfurylaminopurine (kinetin) prior to transporttests with applied labeled indoleacetic acid (IAA-2-14C). Kinetin-treatmentsincreased the uptake of IAA in non-tumorous shoots; the IAAuptake by N. langsdorffii segments was increased up to 3-fold.The auxin uptake in stem-segments of the tumor-forming hybrid,however, could not be increased significantly by kinetin. Theeven distribution of IAA-14C in segments of normal and tumorproneNicotiana shoots is stimulated by kinetin. The effect of stem segment length on conductance measurementswas determined by repeatedly shortening the transport distancein isolated stem segments of Lonicera fragrantissima. If vesselends limit water flow, shorter segment lengths could resultin higher readings of Kh (hydraulic conductance per unit pressuregradient). The mean Kh in the successively shortened segmentsshowed no statistically significant change when shortened from20 to 2 cm, even though maximum vessel lengths ranged from 6to 14 cm. This suggests vessel ends may be less limiting towater flow than are vessel lumens. There was a statisticallysignificant drop in Kh (19%, s.e.=0·29%) when segmentswere shortened from 2 to 1 cm, regardless of the position ofthe segment within the branch. The Kh remained constant withapplied pressure in 2 cm segments while it was more variablewith applied pressure in 1 cm segments. Based on the paint-infusionmethod mean vessel lengths were from 0·6 to 1·2cm. The consistently lower Kh in 1 cm segments might be dueto an end effect in segments with a high percentage of cut openvessels. Within a branch, distal and proximal stem segmentshad lower Kh values than segments from intermediate positions,but nodes had no measurable impact on Kh. Longer segments allowfor more accurate and consistent measurements of Kh since theyintegrate localized variation in Kh while minimizing the segmentend effect. Use of centrifugal force in the study of xylem cavitation   总被引:17,自引:4,他引:13  
Two methods were evaluated for using centrifugal force to measurethe occurrence of cavitation as a function of negative pressuresin xylem. The general protocol was to measure the hydraulicconductivity of xylem segments (stem or root pieces) beforeand after centring them on a centrifuge rotor and spinning themabout their long axis to generate negative xylem pressure. Thepercentage decrease in conductivity from the initial to finalmeasurement was used to quantify the embolism resulting fromcavitation during spinning. In one approach, segments were spunwith their ends exposed to air. This method could only be usedwhen xylem conduits were much shorter than the segment. Resultsfrom an angiosperm (Betula occidentalis) and a gymnosperm (AblesIasiocarpa) corresponded to previous observations of embolismcaused by air dehydration where negative pressure was measuredwith the pressure chamber. Results also agreed with embolismcaused by injection of air into the xylem, in support of theair-seeding hypothesis for cavitation. In a second approach,segments were spun in a rotor designed to keep the segment endsimmersed in water during spinning. This gave the same resultsas for non-immersed segments. Immersing the segment ends allowedmeasurements on any material, regardless of conduit length,as demonstrated for roots of B. occidentalis. The chief advantageof the centrifugal force method is the rapidity and precisionwith which any desired xylem pressure can be imposed. Prat, R. 1985. A comparative study of some methods for measuringcell growth potentials in Vigna radiata hypocotyl, in situ andafter excision.—J. exp. Bot. 36: 1150–1158. Accurate estimations of growth rates are required to understandthe development of physiological and biochemical parametersinvolved in growth processes. Growth measurements can be madeeither ‘in situ’ on whole seedlings by marking successivelevels or ‘in vitro’, after excision of successivehypocotyl segments. These different methods have been comparedin order to determine which measurements allow one to assessaccurately the normal growth characteristics. In vitro measurementsperformed with excised segments required a prolonged incubationperiod before stable growth rates were achieved. The growthpotentials of the successive hypocotyl levels remain unchangedafter excision. These in vitro measurements can therefore beused to assess the growth characteristics of cells in hypocotyls. SYNOPSIS. Amphioxus is widely held to be the closest invertebraterelative of the vertebrates and the best available stand-infor the proximate ancestor of the vertebrates. The spatiotemporalexpression patterns of developmental genes can help suggestbody part homologies between vertebrates and amphioxus. Thisapproach is illustrated using five homeobox genes (AmphiHoxl,AmphiHox2, AmphiOtx, AmphiDll, and AmphiEri) to provide insightsinto the evolutionary origins of three important vertebratefeatures: the major brain regions, the neural crest, and rostrocaudalsegmentation. During amphioxus development, the neural expressionpatterns of these genes are consistent with the presence ofa forebrain (detailed neuroanatomy indicates that the forebrainis all diencephalon without any telencephalon) and an extensivehindbrain; the possible presence of a midbrain requires additionalstudy. Further, during neurulation, the expression pattern ofAmphiDll as well as migratory cell behavior suggest that theepidermal cells bordering the neural plate may represent a phylogeneticprecursor of the vertebrate neural crest. Finally, when theparaxial mesoderm begins to segment, the earliest expressionof AmphiEn is detected in the posterior part of each nascentand newly formed somite. This pattern recalls the expressionof the segment-polarity gene engrailed during establishmentof the segments of metameric protostomes. Thus, during animalevolution, the role of engrailed in establishing and maintainingmetameric body plans may have arisen in a common segmented ancestorof both the protostomes and deuterostomes.  相似文献

A genome project focusingon the nematode Caenorhabditis elegans has demonstrated thepresence of eight cDNAs belonging to the major intrinsic proteinsuperfamily. We functionally characterized one of these cDNAs namedC01G6.1. Injection of C01G6.1 cRNA increased the osmotic waterpermeability (Pf) of Xenopusoocytes 11-fold and the urea permeability 4.5-fold but failed toincrease the glycerol permeability. It has been speculated that the MIPfamily may be separated into two large subfamilies based on thepresence or absence of two segments of extra amino acid residues (~15amino acids) at the second and third extracellular loops. BecauseC01G6.1 (designated AQP-CE1), AQP3, and glycerol facilitator (GlpF) all have these two segments, we replaced the segments of AQP-CE1 with thoseof AQP3 and GlpF to identify their roles. The functional characteristics of these mutants were principally similar to that ofwild-type AQP-CE1, although the values of Pf andurea permeability were decreased by 39-74% and 28-65%,respectively. These results suggest that the two segments of extraamino acid residues may not contribute to channel selectivity orformation of the route for small solutes.
